Abstract:Diuron, a phenylurea herbicide, has been extensively applied in controlling a wide range of weeds in several crops. In the current study, a mixed culture of three bacterial strains, i.e., Bacillus subtilis DU1, Acinetobacter baumannii DU, and Pseudomonas sp. DUK, isolated from sugarcane soil, completely degraded diuron and 3,4-DCA in liquid media at 20 mg L-1 within 48 h. During diuron degradation, a few metabolites (DCPMU, DCPU, and 3,4-DCA) were produced. Further determination of ring-cleavage pathways demonstrated that Acinetobacter baumannii DU and Pseudomonas fluorescens DUK degraded diuron and 3,4-DCA via ortho-cleavage. In contrast, Bacillus subtilis DU transformed these compounds via meta-cleavage pathways. Moreover, diuron caused a significant shift in the bacterial community in soil without diuron history. The augmentation of mountain soil with the isolated bacteria resulted in nearly three times higher degradation rate of diuron than the degradation by indigenous microorganisms. This study provides important information on in situ diuron bioremediation from contaminated sites by bioaugmentation with a mixed bacterial culture.
Degradation of thiobencarb and propanil in an herbicide and their effects on bacterial community in soil

Abstract:Diuron has been widely applied to control weeds causing serious environmental pollution. DUK, was investigated for diuron degradation in a packed reactor and soil. The immobilization of the bacteria mixture in rice straw increased the degradation. The specific degradation rates of diuron by bacteria immobilized in rice straw in batch cultures increased from 0.38 ± 0.03 mg/L at the first cycle to 0.98 ± 0.10 mg/L at the fourth cycle. The degradation using the reactor was also carried out in a continuous operation. Moreover, the introduction of bacteria into soil increased diuron degradation. This study shows the potential of diuron degradation in the reactor and in soil using the mixed bacterial culture immobilized in rice straw.
Enhanced Biodegradation of Phenylurea Herbicides by Ochrobactrum anthrophi CD3 Assessment of Its Feasibility in Diuron-Contaminated Soils

Abstract:The phenylurea herbicides are persistent in soil and water, making necessary the de-velopment of techniques for their removal from the environment. To identify new options in this regard, bacterial strains were isolated from a soil historically managed with pesticides. Ochrobactrum anthropi CD3 showed the ability to remove completely herbicides such as diuron, linuron, chlorotoluron and fluometuron from aqueous solution, and up to 89% of isoproturon. In the case of diuron and linuron, their main metabolite, 3,4-dichloroaniline (3,4-DCA), which has a higher toxicity than the parent compounds, was formed, but remained in solution without further degradation. O. anthropi CD3 was also tested for bioremediation of two different agricultural soils artificially contaminated with diuron, employing bioremediation techniques: (i) biostimulation, using a nutrient solution (NS), (ii) bioaugmentation, using O. anthropi CD3, and iii) bioavailability enhancement using 2-hydroxypropyl-β-cyclodextrin (HPBCD). When bioaugmentation and HPBCD were jointly applied, 50% of the diuron initially added to the soil was biodegraded in a range from 4.7 to 0.7 d. Also, 3,4-DCA was degraded in soil after the strain was inoculated. At the end of the soil biodegradation assay an ecotoxicity test confirmed that after inoculating O. anthropi CD3 the toxicity was drastically reduced.

Abstract:Diuron (DUR) is a phenylurea herbicide widely used for the effective control of most annual and perennial weeds in farming areas. The extensive use of DUR has led to its widespread presence in soil, sediment, and aquatic environments, which poses a threat to non-target crops, animals, humans, and ecosystems. Therefore, the removal of DUR from contaminated environments has been a hot topic for researchers in recent decades. Bioremediation seldom leaves harmful intermediate metabolites and is emerging as the most effective and eco-friendly strategy for removing DUR from the environment. Microorganisms, such as bacteria, fungi, and actinomycetes, can use DUR as their sole source of carbon. Some of them have been isolated, including organisms from the bacterial genera Arthrobacter, Bacillus, Vagococcus, Burkholderia, Micrococcus, Stenotrophomonas, and Pseudomonas and fungal genera Aspergillus, Pycnoporus, Pluteus, Trametes, Neurospora, Cunninghamella, and Mortierella. A number of studies have investigated the toxicity and fate of DUR, its degradation pathways and metabolites, and DUR-degrading hydrolases and related genes. However, few reviews have focused on the microbial degradation and biochemical mechanisms of DUR. The common microbial degradation pathway for DUR is via transformation to 3,4-dichloroaniline, which is then metabolized through two different metabolic pathways: dehalogenation and hydroxylation, the products of which are further degraded via cooperative metabolism. Microbial degradation hydrolases, including PuhA, PuhB, LibA, HylA, Phh, Mhh, and LahB, provide new knowledge about the underlying pathways governing DUR metabolism. The present review summarizes the state-of-the-art knowledge regarding (1) the environmental occurrence and toxicity of DUR, (2) newly isolated and identified DUR-degrading microbes and their enzymes/genes, and (3) the bioremediation of DUR in soil and water environments. This review further updates the recent knowledge on bioremediation strategies with a focus on the metabolic pathways and molecular mechanisms involved in the bioremediation of DUR.

Abstract:Biodegradation was considered a promising and environmentally friendly method for treating environmental pollution caused by diuron. However, the mechanisms of biodegradation of diuron required further research. In this study, the degradation process of diuron by Achromobacter xylosoxidans SL-6 was systematically investigated. The results suggested that the antioxidant system of strain SL-6 was activated by adding diuron, thereby alleviating their oxidative stress response. In addition, degradation product analysis showed that diuron in strain SL-6 was mainly degraded by urea bridge cleavage, dehalogenation, deamination, and ring opening, and finally cis, cis -muconic acid was generated. The combined analysis of metabolomics and transcriptomics revealed the biodegradation and adaptation mechanism of strain SL-6 to diuron. Metabolomics analysis showed that after the strain SL-6 was exposed to diuron, metabolic pathways such as tricarboxylic acid cycle ( cis, cis -muconic acid), glutathione metabolism (oxidized glutathione), and urea cycle (arginine) were reprogrammed in the cells. Furthermore, diuron could induce the production of membrane transport proteins in strain SL-6 cells and overexpress antioxidant enzyme genes, finally ultimately promoting the up-regulation of genes encoding amide hydrolases and dioxygenases, which was revealed by transcriptomics studies. This work enriched the biodegradation mechanism of phenylurea herbicides and provided guidance for the removal of diuron residues in the environment and promoting agriculture sustainable development. Abstract:From agricultural soils, where the herbicide Diuron has been frequently applied, a microbial community capable of degrading Diuron and 3,4-dichloroaniline was obtained. The volumetric rates and degradation efficiencies of Diuron and 3,4-DCA were evaluated in two distinct biofilm reactors, which differ in their operating conditions. One is a horizontal fixed bed reactor; plug-flow operated (PF-PBC) with severe limitation of oxygen. In this reactor, the air was supplied to an equalizer reservoir at the start of the PF-PBC reactor. The other is a compartmentalized aerobic biobarrier with internal recirculation of liquid aerated through airlift devices (ALB), continuously or intermittently operated. Both reactors were inoculated with a microbial community capable of degrading Diuron, isolated from a sugarcane field. In the oxygen-limited PF-PBC reactor, 3,4-DCA accumulation was detected, mainly in the middle zone of the packed channel. On the contrary, in the fully aerobic ALB reactor, minimal accumulation of catabolic byproducts was detected, and high Diuron removal efficiencies and removal rates were obtained when it was continuously operated in steady-state conditions. Additionally, the influence of oxygen limitation on the kinetic behavior of the PF-PBC reactor was determined, and a method to estimate the local removal rates of Diuron RV,CD along the plug-flow channel is described. It was observed that the local values of the instantaneous removal rate of Diuron dCD/dt are high in the aerobic region of the PF-PBC reactor; but, suddenly decay in the reactor zones limited by dissolved oxygen.
Fungal bioremediation of diuron-contaminated waters: evaluation of its degradation and the effect of amendable factors on its removal in a trickle-bed reactor under non-sterile conditions

Abstract:The occurrence of the extensively used herbicide diuron in the environment poses a severe threat to the ecosystem and human health. Four different ligninolytic fungi were studied as biodegradation candidates for the removal of diuron. Among them, T. versicolor was the most effective species, degrading rapidly not only diuron (83%) but also the major metabolite 3,4-dichloroaniline (100%), after 7-day incubation. During diuron degradation, five transformation products (TPs) were found to be formed and the structures for three of them are tentatively proposed. According to the identified TPs, a hydroxylated intermediate 3-(3,4-dichlorophenyl)-1-hydroxymethyl-1-methylurea (DCPHMU) was further metabolized into the N-dealkylated compounds 3-(3,4-dichlorophenyl)-1-methylurea (DCPMU) and 3,4-dichlorophenylurea (DCPU). The discovery of DCPHMU suggests a relevant role of hydroxylation for subsequent N-demethylation, helping to better understand the main reaction mechanisms of diuron detoxification. Experiments also evidenced that degradation reactions may occur intracellularly and be catalyzed by the cytochrome P450 system. A response surface method, established by central composite design, assisted in evaluating the effect of operational variables in a trickle-bed bioreactor immobilized with T. versicolor on diuron removal. The best performance was obtained at low recycling ratios and influent flow rates. Furthermore, results indicate that the contact time between the contaminant and immobilized fungi plays a crucial role in diuron removal. This study represents a pioneering step forward amid techniques for bioremediation of pesticides-contaminated waters using fungal reactors at a real scale.
Degradation of isoproturon in vitro by a mix of bacterial strains isolated from arable soil

Abstract:Isoproturon (IPU) is widely used to control annual grasses and broad leaf weeds in cereal crops. In this study, four IPU-degrading bacterial strains, i.e., Sphingomonas sp. ISP1, Arthrobacter sp. ISP2, Acinetobacter baumannii 4IA, and Pseudomonas sp. ISP3, were isolated from agricultural soil. The mixed culture of four isolates completely degraded the herbicide at 100 mg/L within 10 days. During IPU degradation, several transient accumulations of the metabolites, including 3-(4-isopropylphenyl)-1-methylurea, 3-(4-isopropylphenyl)-urea, 4-isopropylaniline, and 4-toluidine, were also identified. Moreover, the inoculation of the isolated mixed culture into the soil from a mountain with no previous herbicide application increased the degradation rate by 51% of the herbicide on average. Furthermore, bioaugmentation with isolated bacteria in the soil resulted in short-term variations in bacterial structure compared to the unaugmented soil. The findings of this study were instrumental in understanding the mechanisms of pesticide breakdown and bioremediation in liquid media and soil.
Biochemical insights into the biodegradation mechanism of typical sulfonylureas herbicides and association with active enzymes and physiological response of fungal microbes: A multi-omics approach

Abstract:The extensive use of sulfonylurea herbicides has raised major concerns regarding their long-term soil residues and agroecological risks despite their role in agricultural protection. Microbial degradation is an important approach to remove sulfonylureas, whereas understanding the associated biodegradation mechanisms, enzymes, and physiological responses remains incomplete. Based on the rapid biodegradation of nicosulfuron by typical fungal isolate Talaromyces flavus LZM1, the dependency on cellular accumulation and environmental conditions, e.g. pH and nutrient supplies, was shown in the study. The biodegradation of nicosulfuron occurred intracellularly and followed the cascade of reactions including hydrolysis, Smile contraction rearrangement, hydroxylation, and opening of the pyrimidine ring. Besides 2-amino-4,6-dimethoxypyrimidine (ADMP) and 2-aminosulfonyl-N,N-dimethylnicotinamide (ASDM), numerous products and intermediates were newly identified and the structural forms of methoxypyrimidine and sulfonylurea bridge contraction rearrangement are predicted to be more toxic than nicosulfuron. The biodegradation should be enzymatically regulated by glycosylphosphatidylinositol transaminase (GPI-T) and P450s, which were manifested with the significant upregulation in proteomics. It is the first time that the hydrolysis of nicosulfuron into ADMP and ASDM have been associated with GPI-T. The integrated pathways of biodegradation were further elucidated through the involvement of various active enzymes. Except for the enzymatic catalysis, the physiological responses verified by metabolo-proteomics were critical not only to regulate material synthesis, uptake, utilization, and energy transfer but also to maintain antioxidant homeostasis, biodegradability, and tolerance of nicosulfuron by the differentially expressed metabolites, such as acetolactate synthase and 3-isopropylmalate dehydratase. The obtained results would help understand the biodegradation mechanism of sulfonylurea from chemicobiology and enzymology and promote the use of fungal biodegradation in pollution rehabilitation.
A novel metabolic pathway for biodegradation of DDT by the white rot fungi, Phlebia lindtneri and Phlebia brevispora

Abstract:Abstract1,1,1-trichloro-2,2-bis(4-chlorophenyl)ethane (DDT) was used as the substrate for a degradation experiment with the white rot fungi Phlebia lindtneri GB-1027 and Phlebia brevispora TMIC34596, which are capable of degrading polychlorinated dibenzo-p-dioxin (PCDD) and polychlorinated biphenyls (PCBs). Pure culture of P. lindtneri and P. brevispora with DDT (25 μmol l−1) showed that 70 and 30% of DDT, respectively, disappeared in a low-nitrogen medium after a 21-day incubation period. The metabolites were analyzed using gas chromatography/mass spectrometry (GC/MS). Both fungi metabolized DDT to 1,1-dichloro-2,2-bis(4-chlorophenyl)ethane (DDD), 2,2-bis(4-chlorophenyl)acetic acid (DDA) and 4,4-dichlorobenzophenone (DBP). Additionally, DDD was converted to DDA and DBP. DDA was converted to DBP and 4,4-dichlorobenzhydrol (DBH). While DBP was treated as substrate, DBH and three hydroxylated metabolites, including one dihydroxylated DBP and two different isomers of monohydroxylated DBH, were produced from fungal cultures, and these hydroxylated metabolites were efficiently inhibited by the addition of a cytochrome P-450 inhibitor, piperonyl butoxide. These results indicate that the white rot fungi P. lindtneri and P. brevispora can degrade DBP/DBH through hydroxylation of the aromatic ring. Moreover, the single-ring aromatic metabolites, such as 4-chlorobenzaldehyde, 4-chlorobenzyl alcohol and 4-chlorobenzoic acid, were found as metabolic products of all substrate, demonstrating that the cleavage reaction of the aliphatic-aryl carbon bond occurs in the biodegradation process of DDT by white rot fungi.

Abstract:Diuron is a broad-spectrum herbicide used to control weeds and algae in the agriculture sector. Diuron as a water pollutant has been detected in various water bodies due to its slow biodegradation. The adsorption methods offer a promising solution because of their low costs and efficiencies. This article discusses the presence, fate, ecotoxicology, and removal of diuron from aqueous media by adsorption. It was observed that the most effective adsorbents are employed at dosages below 1.0 g L −1 in slightly acidic to neutral solutions (pH 6–7). Pseudo-second-order model describes the best fitting of the adsorption kinetics, while Langmuir isotherm is used for the determination of adsorption capacity. Activated carbon derived from argan nutshells exhibited exceptional adsorption capacity up to 780 mg g −1 , followed by activated carbon obtained from cassava biomass with 122 mg g −1 adsorption capacity. It is also observed that increased dissolved oxygen in water enhances diuron removal with carbonaceous adsorbents (above 60% removal); even in the presence of other common pesticides. In fixed-bed systems, rice husk ash shows an adsorption capacity of 9.5 mg g −1 . Finally, theoretical results suggest that diuron adsorption occurs through various physical interactions (π-π interactions, H-bonding, dipole-dipole, and van der Waals forces), depending on the adsorbent surface. Diuron removal is well established at the laboratory scale, but further research is needed to bridge the gap between successful lab-scale applications and continuous large-scale applications.

Abstract:The present study evaluated the performance of the fungus Trichoderma reesei to tolerate and biodegrade the herbicide diuron in its agrochemical presentation in agar plates, liquid culture, and solid-state fermentation. The tolerance of T. reesei to diuron was characterized through a non-competitive inhibition model of the fungal radial growth on the PDA agar plate and growth in liquid culture with glucose and ammonium nitrate, showing a higher tolerance to diuron on the PDA agar plate (inhibition constant 98.63 mg L −1 ) than in liquid culture (inhibition constant 39.4 mg L −1 ). Diuron biodegradation by T. reesei was characterized through model inhibition by the substrate on agar plate and liquid culture. In liquid culture, the fungus biotransformed diuron into 3,4-dichloroaniline using the amide group from the diuron structure as a carbon and nitrogen source, yielding 0.154 mg of biomass per mg of diuron. A mixture of barley straw and agrolite was used as the support and substrate for solid-state fermentation. The diuron removal percentage in solid-state fermentation was fitted by non-multiple linear regression to a parabolic surface response model and reached the higher removal (97.26%) with a specific aeration rate of 1.0 vkgm and inoculum of 2.6 × 10 8 spores g −1 . The diuron removal in solid-state fermentation by sorption on barley straw and agrolite was discarded compared to the removal magnitude of the biosorption and biodegradation mechanisms of Trichoderma reesei . The findings in this work about the tolerance and capability of Trichoderma reesei to remove diuron in liquid and solid culture media demonstrate the potential of the fungus to be implemented in bioremediation technologies of herbicide-polluted sites.

Abstract:The spread and environmental accumulation of DDT over the years represent not only a threat to human health and ecological security but also a major challenge because of the complex chemical processes and technologies required for remediation. Saprotrophic fungi, isolated from contaminated sites, hold promise for their bioremediation potential toward toxic organic compounds, since they might provide an environment-friendly solution to contamination. Once we verified the high tolerance of autochthonous fungal strains to high concentrations of DDT, we showed how fungi from different phyla demonstrate a high metabolic versatility in the presence of DDT. The isolates showed the singular ability to keep their functionality, despite the DDT-induced production of reactive oxygen species. ABSTRACT DDT (dichlorodiphenyltrichloroethane) was used worldwide as an organochlorine insecticide to control agricultural pests and vectors of several insect-borne human diseases. It was banned in most industrialized countries; however, due to its persistence in the environment, DDT residues remain in environmental compartments, becoming long-term sources of exposure. To identify and select fungal species suitable for bioremediation of DDT-contaminated sites, soil samples were collected from DDT-contaminated agricultural soils in Poland, and 38 fungal taxa among 18 genera were isolated. Two of them, Trichoderma hamatum FBL 587 and Rhizopus arrhizus FBL 578, were tested for tolerance in the presence of 1-mg liter−1 DDT concentration by using two indices based on fungal growth rate and biomass production (the tolerance indices Rt:Rc and TI), showing a clear tolerance to DDT. The two selected strains were studied to evaluate catabolic versatility on 95 carbon sources with or without DDT by using the Phenotype MicroArray system and to investigate the induced oxidative stress responses. The two strains were able to use most of the substrates provided, resulting in both high metabolic versatility and ecological functionality in the use of carbon sources, despite the presence of DDT. The activation of specific metabolic responses with species-dependent antioxidant enzymes to cope with the induced chemical stress has been hypothesized, since the presence of DDT promoted a higher formation of reactive oxygen species in fungal cells than the controls. The tested fungi represent attractive potential candidates for bioremediation of DDT-contaminated soil and are worthy of further investigations. IMPORTANCE The spread and environmental accumulation of DDT over the years represent not only a threat to human health and ecological security but also a major challenge because of the complex chemical processes and technologies required for remediation. Saprotrophic fungi, isolated from contaminated sites, hold promise for their bioremediation potential toward toxic organic compounds, since they might provide an environment-friendly solution to contamination. Abstract:A bacterial strain YN-8, capable o f utilizing nicosulfuron as sole carbon, was isolated from activated sludge collected from wastewater-treating system of a pesticide manufacturer. This strain was identified as Pseudomonas sp. according to its morphological, physiological and biochemical analysis and the analysis of its 16S rRNA gene sequence. This strain could degrade nicosulfuron in its 100 mg/L liquor medium, with degradation efficiency reaching 65.4%in 7 days. The optimum pH, inoculation and concentration for nicosulfuron degradation by strain YN-8 were 7.0, 5% and 100 mg/L, respectively. In the tested metal ions, Co2+and Ni2+could inhibit the nicosulfuron degradation rate. According to the analysis of the metabolites detected by liquid chromatography-mass spectrometry, we proposed the following pathway of nicusulfuron biodegradation by strain: cleavage of the sulfonylurea bridge and generate 3-(dimethylcarbamoyl) pyridin-2-ylsulfonylcarbamic acid and 2-Amino-4, 6-dimethoxypyrimidine.
Biodegradation of Carbofuran and Paraquat by Indigenous Soil Microorganisms

Abstract:Aim: It aimed at the biodegradation of Carbofuran and Paraquat using the active microbial population resident in a farmyard soil in the Federal University of Petroleum Resources (FUPRE), Ugbomoro Community, Delta State. Study Design: Microcosms were set-up in triplicates; sampling for pesticides’ loss and microbial counts were done bi-weekly. Place and Duration of Study: Study was done in Environmental Management and Toxicology Department, FUPRE and Chemistry Department, University of Benin between June 2019 and July 2019. Methodology: Carbofuran and Paraquat were applied to farmyard soil containing active microbial population at recommended and four times the recommended doses. A sterile control was equally setup. Biodegradation was monitored with the High Performance Liquid Chromatography (HPLC). Microbial counts were monitored using standard methods. Results: Carbofuran decreased from 108.6 ± 0.69 μg/kg (day 0) to 39.2 ± 3.8 μg/kg (day 28) and 301.4 ± 1.29 μg/kg (day 0) to 241.4 ± 2.83 μg/kg (day 28) for recommended and four times recommended rates, respectively. Also, there were complete removal of Paraquat in treatments with the recommended rate while it decreased from 268.3 ± 5.21μg/kg (day 0) to 144.4 ± 2.38 μg/kg (day 28) at four times the recommended rate. In the abiotic control, were little losses of both pesticides. Total heterotrophic bacterial, fungal and actinomycetes counts increased (day 14 to 21) in contaminated soils. Significant differences in degradation with respect to pesticides treatment and time were observed. Conclusion: The microorganisms grew at different concentrations reducing both Carbofuran and Paraquat in contaminated soils. Their presence and high numbers confirmed that they are ubiquitous, diverse and can adjust to any harsh environment. Increased microbial counts showed that they grew in presence of the chemicals and degraded them. Thus, these indigenous microbial population can be used for the clean-up of these pesticides pollution in farms to improve such environment.

Abstract:Parallel to the important use of pesticides in conventional agriculture there is a growing interest for green technologies to clear contaminated soil from pesticides and their degradation products. One specific technique, the inoculation of degrading micro-organisms in polluted soil, known as bioaugmentation, is a promising method still in needs of further developments. Specifically, improvements in the understanding of how degrading microorganisms must overcome abiotic filters and interact with the autochthonous microbial communities are needed in order to efficiently design bioremediation strategies. Here we designed a protocol aiming at studying the degradation of two herbicides, glyphosate (GLY) and isoproturon (IPU), experimental modifications of two source bacterial communities. We used statistical methods stemming from genomic prediction to link community composition to herbicides degradation potentials. Our approach proved to be efficient with correlation estimates over 0.8 - between model predictions and measured pesticide degradation values. OTUs significantly associated with the degradation ability, and therefore identified as relevant by the models were confronted to the literature. Next, multi-degrading bacterial communities were obtained by coalescing bacterial communities with high GLY or IPU degradation ability based on their community-level properties. Finally, we evaluated the efficiency of constructed multi-degrading communities to remove pesticide contamination in a different soil. While results are less clear in the case of GLY, we showed an efficient transfer of degrading capacities towards the receiving soil even at relatively low inoculation levels in the case of IPU. Altogether, we developed an innovative protocol for building multi-degrading simplified bacterial communities with the help of genomic prediction tools and coalescence, and proved their efficiency in a contaminated soil.

Abstract:Nicosulfuron is among the sulfonylurea herbicides that are widely used to control annual and perennial grass weeds in cornfields. However, nicosulfuron residues in the environment are likely to cause long-lasting harmful environmental and biological effects. Nicosulfuron degrades via photo-degradation, chemical hydrolysis, and microbial degradation. The latter is crucial for pesticide degradation and has become an essential strategy to remove nicosulfuron residues from the environment. Most previous studies have focused on the screening, degradation characteristics, and degradation pathways of biodegrader microorganisms. The isolated nicosulfuron-degrading strains include Bacillus, Pseudomonas, Klebsiella, Alcaligenes, Rhodopseudomonas, Ochrobactrum, Micrococcus, Serratia, Penicillium, Aspergillus, among others, all of which have good degradation efficiency. Two main intermediates, 2-amino-4,6-dimethoxypyrimidine (ADMP) and 2-aminosulfonyl-N,N-dimethylnicotinamide (ASDM), are produced during microbial degradation and are derived from the C-N, C-S, and S-N bond breaks on the sulfonylurea bridge, covering almost every bacterial degradation pathway. In addition, enzymes related to the degradation of nicosulfuron have been identified successively, including the manganese ABC transporter (hydrolase), Flavin-containing monooxygenase (oxidase), and E3 (esterase). Further in-depth studies based on molecular biology and genetics are needed to elaborate on their role in the evolution of novel catabolic pathways and the microbial degradation of nicosulfuron. To date, few reviews have focused on the microbial degradation and degradation mechanisms of nicosulfuron. This review summarizes recent advances in nicosulfuron degradation and comprehensively discusses the potential of nicosulfuron-degrading microorganisms for bioremediating contaminated environments, providing a reference for further research development on nicosulfuron biodegradation in the future.
Soil Actinobacteria Exhibit Metabolic Capabilities for Degrading the Toxic and Persistent Herbicide Metribuzin

Abstract:Metribuzin, a widely used triazine herbicide, persists in agricultural soils and poses significant environmental pollution threats globally. The aim of this study was to investigate the biodegradation of metribuzin by actinobacterial strains in vitro at different environmental conditions. From an initial screen of 12 actinobacterial strains, four bacteria exhibited robust growth in the presence of the metribuzin as the sole carbon source at 50 mg/L concentration. The optimization of metribuzin biodegradation under different conditions (pH, temperature and inoculum size) using a spectrophotometric method revealed that maximum degradation of metribuzin occurred at a pH of 7.2, a temperature 30 °C, and at an inoculum volume of 4%. Subsequent GC-MS validation confirmed the remarkable biodegradation capabilities of the actinobacterial isolates, where the strain C1 showed the highest rate of metribuzin degradation of 83.12%. Detailed phylogenetic identified the active strains as Streptomyces toxytricini (CH), Streptomyces stelliscabiei (B2), and two Streptomyces heliomycini (C1, C3). Structural analysis by ATR-FTIR spectroscopy confirmed the extensive biotransformation of the herbicide molecule. Our findings highlight the immense untapped potential of soil actinobacteria, particularly the Streptomyces heliomycini C1 strain, as versatile bioremediation agents for removing persistent agrochemical pollutants.

Abstract:Antibiotics can contaminate the environment as a result of improper disposal. The induction of doxycycline in the environment could give rise to lineages of the cucurbit beetle Diabrotica speciosa containing resistant endosymbiotic microorganisms, giving rise to bacterial strains capable of biodegrading these antibiotic residues. This work determined the relationship between the intestinal microbiota of D. speciosa and its ability to biodegrade an antibiotic. Bacterial strains of Enterobacter cloacae and Stenotrophomonas maltophila isolated from the gastrointestinal tract of D. speciosa were evaluated for the biodegradation of doxycycline. An analytical method for the extraction and quantification of doxycycline by high-efficiency liquid chromatography-ultraviolet detection (HPLC-UV) was developed and applied to monitor the biodegradation rates during bacterial growth for nutrient and nutrient-deficient media, both supplemented with the antibiotic. Degradation kinetics showed a decrease in antibiotic concentration after 96 h in saline medium, with recoveries of 63.1 and 87.7% for E. cloacae and S. maltophilia, respectively. Two degradation products were then identified and characterized from the liquid chromatography-high resolution mass spectrometry data after bioassays with E. cloacae. The gastrointestinal tract of insects such as D. speciosa can be a source of useful microbes for biotechnological processes such as the biodegradation of exogenous organic compounds in the environment.
