Abstract:Abstract Perception of visual motion is important for a range of ethological behaviors in mammals. In primates, specific visual cortical regions are specialized for processing of coherent visual motion. However, whether mouse visual cortex has a similar organization remains unclear, despite powerful genetic tools available for measuring population neural activity. Here, we use widefield and 2-photon calcium imaging of transgenic mice to measure mesoscale and cellular responses to coherent motion. Imaging of primary visual cortex (V1) and higher visual areas (HVAs) during presentation of natural movies and random dot kinematograms (RDKs) reveals varied responsiveness to coherent motion, with stronger responses in dorsal stream areas compared to ventral stream areas. Moreover, there is considerable anisotropy within visual areas, such that neurons representing the lower visual field are more responsive to coherent motion. These results indicate that processing of visual motion in mouse cortex is distributed heterogeneously both across and within visual areas.

Abstract:During sensory experience, the retina transmits a diverse array of signals to the brain, which must be parsed to generate meaningful percepts that can guide decisions and actions. Decades of anatomical and physiological studies in primates and carnivores have revealed a complex parallel and hierarchical organization by which distinct visual features are distributed to, and processed by, different brain regions. However, these studies have been limited in their ability to dissect the circuit mechanisms involved in the transformation of sensory inputs into complex cortical representations and action patterns. Multiple groups have therefore pushed to explore the organization and function of higher visual areas in the mouse. Here we review the anatomical and physiological findings of these recent explorations in mouse visual cortex. These studies find that sensory input is processed in a diverse set of higher areas that are each interconnected with specific limbic and motor systems. This hierarchical and parallel organization is consistent with the multiple streams that have been found in the higher visual areas of primates. We therefore propose that the mouse visual system is a useful model to explore the circuits underlying the transformation of sensory inputs into goal-directed perceptions and actions.

Abstract:The brain monitors the sensory environment via signals from the sensory periphery, such as the olfactory epithelium, the inner ear, and the retina. Understanding how sensory stimuli are processed throughout the sensory hierarchy, and how this relates to behavior, is a central outstanding question in the field of neuroscience. The processing of visual motion in mice offers unique opportunities for addressing these questions thanks to a rich literature on the anatomical and physiological properties of motion-sensitive neurons across the visual system, paired with recent developments of cutting-edge genetic and imaging approaches. A visual scene typically contains motion originating from either moving objects or optic flow caused by self-generated movements. Neurons encoding the direction of visual motion are said to be 'direction-selective'. It was historically believed the circuits responsible for creating direction selectivity <em>de novo</em> exist within the visual cortex. Yet, in mice, direction-selective responses can be found already in the retina, suggesting in this model organism visual motion analysis starts at the earliest stage of the visual hierarchy. This minireview presents emerging literature demonstrating how retinal direction-selective cells make causal contributions to central visual motion processing and visually guided behaviors in mice, and their potential clinical relevance, and outlines experiments for testing remaining questions. Research in this field will undoubtedly continue to advance our understanding of the basic principles of the visual system and how sensory neurons extract fundamental features of the world.

Abstract:The brain has evolved to transform sensory information in the environment into neural representations that can be used for perception and action. Higher-order sensory cortical areas, with their increasingly complex receptive fields and integrative properties, are thought to be critical nodes for this function. This is especially true in the primate visual cortex, in which functionally specialized areas are engaged in parallel streams to support diverse computations. Recent anatomical and physiological studies of the mouse visual cortex have revealed a similarly complex network of specialized higher-order areas. This structure provides a useful model for determining the synaptic and circuit mechanisms through which information is transformed across distinct processing stages. In this review, we summarize the current knowledge on the layout, connectivity, and functional properties of the higher visual areas in the mouse. In addition, we speculate on the contribution of these areas to perception and action, and how knowledge of the mouse visual system can inform us about the principles that govern information processing in integrated networks.

Highly Selective Receptive Fields in Mouse Visual Cortex

Abstract:Genetic methods available in mice are likely to be powerful tools in dissecting cortical circuits. However, the visual cortex, in which sensory coding has been most thoroughly studied in other species, has essentially been neglected in mice perhaps because of their poor spatial acuity and the lack of columnar organization such as orientation maps. We have now applied quantitative methods to characterize visual receptive fields in mouse primary visual cortex V1 by making extracellular recordings with silicon electrode arrays in anesthetized mice. We used current source density analysis to determine laminar location and spike waveforms to discriminate putative excitatory and inhibitory units. We find that, although the spatial scale of mouse receptive fields is up to one or two orders of magnitude larger, neurons show selectivity for stimulus parameters such as orientation and spatial frequency that is near to that found in other species. Furthermore, typical response properties such as linear versus nonlinear spatial summation (i.e., simple and complex cells) and contrast-invariant tuning are also present in mouse V1 and correlate with laminar position and cell type. Interestingly, we find that putative inhibitory neurons generally have less selective, and nonlinear, responses. This quantitative description of receptive field properties should facilitate the use of mouse visual cortex as a system to address longstanding questions of visual neuroscience and cortical processing.

Abstract:The cerebral cortex contains cells which respond to movement of the head, and these cells are thought to be involved in the perception of self-motion. In particular, studies in the primary visual cortex of mice show that both running speed and passive whole-body rotation modulates neuronal activity, and modern genetically targeted viral tracing approaches have begun to identify previously unknown circuits that underlie these responses. Here we review recent experimental findings and provide a road map for future work in mice to elucidate the functional architecture and emergent properties of a cortical network potentially involved in the generation of egocentric-based visual representations for navigation.

Abstract:The cerebral cortex of the mouse has become one of the most important systems for studying information processing and the neural correlates of behavior. Multiple studies have examined the first stages of visual cortical processing: primary visual cortex (V1) and its thalamic inputs from the dorsal lateral geniculate nucleus (dLGN), but more rarely in the lateral posterior nucleus (LP) in mice. Multiple single-unit surveys of dLGN and V1, both with electrophysiology and two-photon calcium imaging, have described receptive fields in anesthetized animals. Increasingly, awake animals are being used in physiological studies, so it is important to compare neuronal responses between awake and anesthetized state. We have performed a comprehensive survey of spatial and temporal response properties in V1, dLGN, and lateral posterior nucleus of both anesthetized and awake animals, using a common set of stimuli: drifting sine-wave gratings spanning a broad range of spatial and temporal parameters, and sparse noise stimuli consisting of flashed light and dark squares. Most qualitative receptive field parameters were found to be unchanged between the two states, such as most aspects of spatial processing, but there were significant differences in several parameters, most notably in temporal processing. Compared with anesthetized animals, the temporal frequency that evoked the peak response was shifted toward higher values in the dLGN of awake mice and responses were more sustained. Further, the peak response to a flashed stimulus was earlier in all three areas. Overall, however, receptive field properties in the anesthetized animal remain a good model for those in the awake animal.SIGNIFICANCE STATEMENT: The primary visual cortex (V1) of the mouse and its inputs from visual thalamus (dLGN), have become a dominant model for studying information processing in the brain. Early surveys of visual response properties (receptive fields) were performed in anesthetized animals. Although most recent studies of V1 have been performed in awake animals to examine links between vision and behavior, there have been few comprehensive studies of receptive field properties in the awake mouse, especially in dLGN and lateral posterior nucleus. We have performed a comparative survey of receptive fields in dLGN, lateral posterior nucleus, and V1 in anesthetized and awake mice. We found multiple differences in processing of time-varying stimuli, whereas the spatial aspects of receptive fields remain comparatively unchanged.

Contralateral Bias of High Spatial Frequency Tuning and Cardinal Direction Selectivity in Mouse Visual Cortex

Abstract:Binocular mechanisms for visual processing are thought to enhance spatial acuity by combining matched input from the two eyes. Studies in the primary visual cortex of carnivores and primates have confirmed that eye-specific neuronal response properties are largely matched. In recent years, the mouse has emerged as a prominent model for binocular visual processing, yet little is known about the spatial frequency tuning of binocular responses in mouse visual cortex. Using calcium imaging in awake mice of both sexes, we show that the spatial frequency preference of cortical responses to the contralateral eye is ∼35% higher than responses to the ipsilateral eye. Furthermore, we find that neurons in binocular visual cortex that respond only to the contralateral eye are tuned to higher spatial frequencies. Binocular neurons that are well matched in spatial frequency preference are also matched in orientation preference. In contrast, we observe that binocularly mismatched cells are more mismatched in orientation tuning. Furthermore, we find that contralateral responses are more direction-selective than ipsilateral responses and are strongly biased to the cardinal directions. The contralateral bias of high spatial frequency tuning was found in both awake and anesthetized recordings. The distinct properties of contralateral cortical responses may reflect the functional segregation of direction-selective, high spatial frequency-preferring neurons in earlier stages of the central visual pathway. Moreover, these results suggest that the development of binocularity and visual acuity may engage distinct circuits in the mouse visual system.<b>SIGNIFICANCE STATEMENT</b> Seeing through two eyes is thought to improve visual acuity by enhancing sensitivity to fine edges. Using calcium imaging of cellular responses in awake mice, we find surprising asymmetries in the spatial processing of eye-specific visual input in binocular primary visual cortex. The contralateral visual pathway is tuned to higher spatial frequencies than the ipsilateral pathway. At the highest spatial frequencies, the contralateral pathway strongly prefers to respond to visual stimuli along the cardinal (horizontal and vertical) axes. These results suggest that monocular, and not binocular, mechanisms set the limit of spatial acuity in mice. Furthermore, they suggest that the development of visual acuity and binocularity in mice involves different circuits.

Abstract:The mouse is emerging as an important model for understanding how sensory neocortex extracts cues to guide behavior, yet little is known about how these cues are processed beyond primary cortical areas. Here, we used two-photon calcium imaging in awake mice to compare visual responses in primary visual cortex (V1) and in two downstream target areas, AL and PM. Neighboring V1 neurons had diverse stimulus preferences spanning five octaves in spatial and temporal frequency. By contrast, AL and PM neurons responded best to distinct ranges of stimulus parameters. Most strikingly, AL neurons preferred fast-moving stimuli while PM neurons preferred slow-moving stimuli. By contrast, neurons in V1, AL, and PM demonstrated similar selectivity for stimulus orientation but not for stimulus direction. Based on these findings, we predict that area AL helps guide behaviors involving fast-moving stimuli (e.g., optic flow), while area PM helps guide behaviors involving slow-moving objects.

Abstract:Vision is the sense humans rely on most to navigate the world, make decisions, and perform complex tasks. Understanding how humans see thus represents one of the most fundamental and important goals of neuroscience. The use of the mouse as a model for parsing how vision works at a fundamental level started approximately a decade ago, ushered in by the mouse's convenient size, relatively low cost, and, above all, amenability to genetic perturbations. In the course of that effort, a large cadre of new and powerful tools for in vivo labeling, monitoring, and manipulation of neurons were applied to this species. As a consequence, a significant body of work now exists on the architecture, function, and development of mouse central visual pathways. Excitingly, much of that work includes causal testing of the role of specific cell types and circuits in visual perception and behavior—something rare to find in studies of the visual system of other species. Indeed, one could argue that more information is now available about the mouse visual system than any other sensory system, in any species, including humans. As such, the mouse visual system has become a platform for multilevel analysis of the mammalian central nervous system generally. Here we review the mouse visual system structure, function, and development literature and comment on the similarities and differences between the visual system of this and other model species. We also make it a point to highlight the aspects of mouse visual circuitry that remain opaque and that are in need of additional experimentation to enrich our understanding of how vision works on a broad scale.

Experience-dependent predictions of feedforward and contextual information in mouse visual cortex

Abstract:Neurons in primary visual cortex are driven by feedforward visual inputs and top-down contextual inputs. The nature of this contextual information is difficult to study, as responses to feedforward and top-down inputs overlap in time and are difficult to disentangle experimentally. To address this issue, we measured responses to natural images and partially occluded versions of these images in the visual cortex of mice. Assessing neuronal responses before and after familiarizing mice with the non-occluded images allowed us to study experience-dependent and stimulus-specific contextual responses in pyramidal cells (PyCs) in cortical layers 2/3 and 5 in the absence of feedforward input. Surprisingly, in the same retinotopic region of cortex, we found that separate populations of PyCs in layer 2/3 responded to occluded and non-occluded images. Responses of PyCs selective for occluded images were strengthened upon familiarization and decoding analysis revealed they contained image-specific information, suggesting that they signaled the absence of predicted visual stimuli. Responses of PyCs selective for non-occluded scenes were weaker for familiarized images but stronger for unfamiliar images, suggesting that these neurons signaled the presence of unpredicted visual stimuli. Layer 5 also contained PyCs preferring either feedforward or contextual inputs, but their responses were more complex and strengthening of responses to occluded images required task engagement. The results show that visual experience decreases the activity of neurons responding to known feedforward inputs but increases the activity of neurons responding to contextual inputs tied to expected stimuli.

Abstract:Classic ON-OFF direction-selective ganglion cells (DSGCs) that encode the four cardinal directions were recently shown to also be orientation-selective. To clarify the mechanisms underlying orientation selectivity, we employed a variety of electrophysiological, optogenetic, and gene knock-out strategies to test the relative contributions of glutamate, GABA, and acetylcholine (ACh) input that are known to drive DSGCs, in male and female mouse retinas. Extracellular spike recordings revealed that DSGCs respond preferentially to either vertical or horizontal bars, those that are perpendicular to their preferred-null motion axes. By contrast, the glutamate input to all four DSGC types measured using whole-cell patch-clamp techniques was found to be tuned along the vertical axis. Tuned glutamatergic excitation was heavily reliant on type 5A bipolar cells, which appear to be electrically coupled via connexin 36 containing gap junctions to the vertically oriented processes of wide-field amacrine cells. Vertically tuned inputs are transformed by the GABAergic/cholinergic "starburst" amacrine cells (SACs), which are critical components of the direction-selective circuit, into distinct patterns of inhibition and excitation. Feed-forward SAC inhibition appears to "veto" preferred orientation glutamate excitation in dorsal/ventral (but not nasal/temporal) coding DSGCs "flipping" their orientation tuning by 90° and accounts for the apparent mismatch between glutamate input tuning and the DSGC's spiking response. Together, these results reveal how two distinct synaptic motifs interact to generate complex feature selectivity, shedding light on the intricate circuitry that underlies visual processing in the retina.

Abstract:Brains have evolved to optimize sensory processing. In primates, complex cognitive tasks must be executed and evolution led to the development of large brains with many cortical areas. Rodents do not accomplish cognitive tasks of the same level of complexity as primates and remain with small brains both in relative and absolute terms. But is a small brain necessarily a simple brain? In this review, several aspects of the visual cortical networks have been compared between rodents and primates. The visual system has been used as a model to evaluate the level of complexity of the cortical circuits at the anatomical and functional levels. The evolutionary constraints are first presented in order to appreciate the rules for the development of the brain and its underlying circuits. The organization of sensory pathways, with their parallel and cross-modal circuits, is also examined. Other features of brain networks, often considered as imposing constraints on the development of underlying circuitry, are also discussed and their effect on the complexity of the mouse and primate brain are inspected. In this review, we discuss the common features of cortical circuits in mice and primates and see how these can be useful in understanding visual processing in these animals.

Abstract:A highly important question in visual neuroscience is to identify where in the visual system information from different processing channels is integrated to form the complex scenery we perceive. A common view to this question is that information is processed hierarchically because small and selective receptive fields in lower visual areas melt into larger receptive fields in specialized higher visual areas. However, a higher order area in which all incoming signals ultimately converge has not yet been identified. Rather, modulation of subthreshold influences from outside the classical receptive field related to contextual integration occurs already in early visual areas. So far it is unclear how these influences are mediated (Gilbert, 1998; Angelucci and Bullier, 2003; Gilbert and Sigman, 2007). In the present study, we show that feedback connections from a higher motion processing area critically influence the integration of subthreshold global motion cues in early visual areas. Global motion cues are theoretically not discernible for a local motion detector in V1, however, imprints of pattern motion have been observed in this area (Guo et al., 2004; Schmidt et al., 2006). By combining reversible thermal deactivation and optical imaging of intrinsic signals we demonstrate that feedback signals from the posteromedial suprasylvian sulcus are critical for the discrimination between global and local motions already in early visual areas. These results suggest that global features of the visual scenery are fed back to lower visual processing units in order to facilitate the integration of local cues into a global construct.
A Circuit for Integration of Head- and Visual-Motion Signals in Layer 6 of Mouse Primary Visual Cortex

Abstract:To interpret visual-motion events, the underlying computation must involve internal reference to the motion status of the observer's head. We show here that layer 6 (L6) principal neurons in mouse primary visual cortex (V1) receive a diffuse, vestibular-mediated synaptic input that signals the angular velocity of horizontal rotation. Behavioral and theoretical experiments indicate that these inputs, distributed over a network of 100 L6 neurons, provide both a reliable estimate and, therefore, physiological separation of head-velocity signals. During head rotation in the presence of visual stimuli, L6 neurons exhibit postsynaptic responses that approximate the arithmetic sum of the vestibular and visual-motion response. Functional input mapping reveals that these internal motion signals arrive into L6 via a direct projection from the retrosplenial cortex. We therefore propose that visual-motion processing in V1 L6 is multisensory and contextually dependent on the motion status of the animal's head.

Abstract:In an ever-changing visual world, animals' survival depends on their ability to perceive and respond to rapidly changing motion cues. The primary visual cortex (V1) is at the forefront of this sensory processing, orchestrating neural responses to perturbations in visual flow. However, the underlying neural mechanisms that lead to distinct cortical responses to such perturbations remain enigmatic. In this study, our objective was to uncover the neural dynamics that govern V1 neurons' responses to visual flow perturbations using a biologically realistic computational model. By subjecting the model to sudden changes in visual input, we observed opposing cortical responses in excitatory layer 2/3 (L2/3) neurons, namely, depolarizing and hyperpolarizing responses. We found that this segregation was primarily driven by the competition between external visual input and recurrent inhibition, particularly within L2/3 and L4. This division was not observed in excitatory L5/6 neurons, suggesting a more prominent role for inhibitory mechanisms in the visual processing of the upper cortical layers. Our findings share similarities with recent experimental studies focusing on the opposing influence of top-down and bottom-up inputs in the mouse primary visual cortex during visual flow perturbations. This study aims to shed light on the intricate dynamics of neural responses within the mouse primary visual cortex (V1) subjected to visual flow perturbations, unraveling the emergence of distinct functional classes of excitatory L2/3 neurons, namely depolarizing (dVf) and hyperpolarizing (hVf) neurons. Through the implementation of a biologically realistic computational model, the investigation highlights the profound impact of synaptic connectivity, inhibitory circuits, and dynamic inputs on shaping these responses. The identified competition by common inhibition mechanism between dVf and hVf neurons, driven not only by long-range thalamic inputs but also by local connectivity, provides new insights into the underlying neural circuitry. This study opens avenues for further exploration into the role of locomotion-related inputs in modulating neural responses, offering a comprehensive framework for future investigations into sensory perception and neural coding.

Abstract:The superior colliculus (SC) is a prominent and conserved visual center in all vertebrates. In mice, the most superficial lamina of the SC is enriched with neurons that are selective for the moving direction of visual stimuli. Here we study how these direction selective neurons respond to complex motion patterns known as plaids, using two-photon calcium imaging in awake male and female mice. The plaid pattern consists of two superimposed sinusoidal gratings moving in different directions, giving an apparent pattern direction that lies between the directions of the two component gratings. Most direction selective neurons in the mouse SC respond robustly to the plaids and show a high selectivity for the moving direction of the plaid pattern but not of its components. Pattern motion selectivity is seen in both excitatory and inhibitory SC neurons and is especially prevalent in response to plaids with large cross angles between the two component gratings. However, retinal inputs to the SC are ambiguous in their selectivity to pattern versus component motion. Modeling suggests that pattern motion selectivity in the SC can arise from a nonlinear transformation of converging retinal inputs. In contrast, the prevalence of pattern motion selective neurons is not seen in the primary visual cortex (V1). These results demonstrate an interesting difference between the SC and V1 in motion processing and reveal the SC as an important site for encoding pattern motion. Significance Statement An important function of the visual system is to encode the direction of complex motion patterns in the environment. Studies using the plaid stimulus have revealed neurons in different cortical areas that are tuned to either pattern motion or component motion, but how neurons in the SC respond to plaids has not been studied. Here we show that direction selective neurons in the mouse SC respond to plaids with a clear pattern motion selectivity, at a level not seen in the retina or V1. Our results thus provide new information regarding the function and organization of the early visual system and highlight the importance of SC circuits in computing complex motion.

Abstract:Adapting goal-directed behaviors to changing sensory conditions is a fundamental aspect of intelligence. The brain uses abstract representations of the environment to generalize learned associations across sensory modalities with shared properties. The circuit organization that mediates such cross-modal generalizations remains, however, unknown. Here, we demonstrate that mice can bidirectionally generalize sensorimotor task rules between touch and vision by using abstract representations of peri-personal space within the cortex. Using large-scale mapping in the dorsal cortex at single-cell resolution, we discovered multimodal neurons with congruent spatial representations within multiple associative areas of the dorsal and ventral stream. Optogenetic sensory substitution and systematic silencing of these associative areas revealed that a single area in the dorsal stream is necessary and sufficient for cross-modal generalization. Our results identify and comprehensively describe a cortical circuit organization that underlies an essential cognitive function, providing a structural and functional basis for abstract reasoning in the mammalian brain.

Abstract:Conscious reportability of visual input is associated with a bimodal neural response in primary visual cortex (V1): an early-latency response coupled to stimulus features and a late-latency response coupled to stimulus report or detection. This late wave of activity, central to major theories of consciousness, is thought to be driven by prefrontal cortex (PFC), responsible for “igniting” it. Here we analyzed two electrophysiological studies in mice performing different stimulus detection tasks, and characterize neural activity profiles in three key cortical regions: V1, posterior parietal cortex (PPC) and PFC. We then developed a minimal network model, constrained by known connectivity between these regions, reproducing the spatio-temporal propagation of visual-and report-related activity. Remarkably, while PFC was indeed necessary to generate report-related activity in V1, this occurred only through the mediation of PPC. PPC, and not PFC, had the final veto in enabling the report-related late wave of V1 activity.

Specific excitatory connectivity for feature integration in mouse primary visual cortex

Abstract:Abstract Local excitatory connections in mouse primary visual cortex (V1) are stronger and more prevalent between neurons that share similar functional response features. However, the details of how functional rules for local connectivity shape neuronal responses in V1 remain unknown. We hypothesised that complex responses to visual stimuli may arise as a consequence of rules for selective excitatory connectivity within the local network in the superficial layers of mouse V1. In mouse V1 many neurons respond to overlapping grating stimuli (plaid stimuli) with highly selective and facilitatory responses, which are not simply predicted by responses to single gratings presented alone. This complexity is surprising, since excitatory neurons in V1 are considered to be mainly tuned to single preferred orientations. Here we examined the consequences for visual processing of two alternative connectivity schemes: in the first case, local connections are aligned with visual properties inherited from feedforward input (a ‘like-to-like’ scheme specifically connecting neurons that share similar preferred orientations); in the second case, local connections group neurons into excitatory subnetworks that combine and amplify multiple feedforward visual properties (a ‘feature binding’ scheme) . By comparing predictions from large scale computational models with in vivo recordings of visual representations in mouse V1, we found that responses to plaid stimuli were best explained by a assuming ‘feature binding’ connectivity. Unlike under the ‘like-to-like’ scheme, selective amplification within feature-binding excitatory subnetworks replicated experimentally observed facilitatory responses to plaid stimuli; explained selective plaid responses not predicted by grating selectivity; and was consistent with broad anatomical selectivity observed in mouse V1. Our results show that visual feature binding can occur through local recurrent mechanisms without requiring feedforward convergence, and that such a mechanism is consistent with visual responses and cortical anatomy in mouse V1. Author summary The brain is a highly complex structure, with abundant connectivity between nearby neurons in the neocortex, the outermost and evolutionarily most recent part of the brain. Although the network architecture of the neocortex can appear disordered, connections between neurons seem to follow certain rules. These rules most likely determine how information flows through the neural circuits of the brain, but the relationship between particular connectivity rules and the function of the cortical network is not known. We built models of visual cortex in the mouse, assuming distinct rules for connectivity, and examined how the various rules changed the way the models responded to visual stimuli. We also recorded responses to visual stimuli of populations of neurons in anaesthetised mice, and compared these responses with our model predictions. We found that connections in neocortex probably follow a connectivity rule that groups together neurons that differ in simple visual properties, to build more complex representations of visual stimuli. This finding is surprising because primary visual cortex is assumed to support mainly simple visual representations. We show that including specific rules for non-random connectivity in cortical models, and precisely measuring those rules in cortical tissue, is essential to understanding how information is processed by the brain.
