Abstract:The South American tomato leafminer, Tuta absoluta Meyrick (Lepidoptera: Gelechiidae), is a major pest that has recently invaded Afro-Eurasia. Biological control, especially by Trichogramma parasitoids, is considered to be promising as a management tool for this pest. However, further development of Trichogramma-based biocontrol strategies would benefit from assessing the impact of released parasitoid offspring on the pest. Under laboratory conditions, we 1) compared the parasitism of five Trichogramma species-strains on the pest-plant system T. absoluta-tomato, and 2) assessed various biological traits of parasitoids, mass-reared on a factitious host (Ephestia kuehniella Zeller), when developing on T. absoluta. In addition, we evaluated the overall efficiency of two specific Trichogramma species when released under greenhouse conditions in combination with a common natural enemy in tomato crop, the predator Macrolophus pygmaeus Rambur. Parasitoids emerging from T. absoluta on tomato showed lower parasitism rates and poor biological traits, for example, wing deformations, reduced longevity, when compared with the control reared on the factitious host. Under greenhouse conditions, the parasitoids that developed on T. absoluta after initial releases contributed little to biological control of T. absoluta, and parasitism tended to be lower when the predator was present. However, a slightly higher T. absoluta control level was achieved by combining the predator and release of the parasitoid Trichogramma achaeae Nagaraja and Nagarkatti. This study shows that Trichogramma parasitoids may not build up populations on the T. absoluta-tomato system, but that Trichogramma parasitoids can be used in combination with M. pygmaeus to enhance biological control of the pest in tomato crops.

Abstract:New strategies are urgently needed to control Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae), one of the most destructive pests of tomato crops. Originating from South America, it is spreading rapidly and has evolved resistance to most common insecticides making it highly challenging to control. Regarding biological control, the parasitoid Necremnus tutae Ribes & Bernardo (Hymenoptera: Eulophidae) is widely present around the Mediterranean basin and has adapted to the invader playing a substantial role in its control. In addition, the neotropical parasitoid Dolichogenidea gelechiidivoris Marsh (Hymenoptera: Braconidae) has established accidentally following its host in Spain and Algeria and a classical biological control program using this species is ongoing in Sub-Saharan Africa. Understanding the trophic connections between D.gelechiidivoris, N.tutae, the targeted host T.absoluta and potential non-target hosts is essential to assess their value as augmentative or classical biological control agents and to promote their establishment in conservation biocontrol. We conducted host-specificity tests in the laboratory using four European leafminer species to evaluate the specificity of these two parasitoids to T.absoluta. We then assessed their affinity for T.absoluta by exposing sentinel plants with the alternative host Phthorimaea operculella (Zeller) (Lepidoptera: Gelechiidae) inside and around greenhouses and measuring each parasitoid species' field parasitism rate. Our results show that of the four non-target species tested in the laboratory, N.tutae attacked three, while D.gelechiidivoris attacked only P. operculella. In the greenhouse, N.tutae did not prefer P.operculella or T.absoluta, whereas D.gelechiidivoris preferred the latter. The mean parasitism rate of T.absoluta over the three-month monitoring period in the greenhouses reached 27% for N.tutae and 35% for D.gelechiidivoris. We discuss the importance of host affinity for understanding potential non-target effects and the value a natural enemy can have as biological control.

Abstract:Abstract During the last few years following the invasion of European tomato crops by the invasive South American tomato leafminer, Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae), several indigenous larval parasitoid species were recorded as natural enemies of this pest. Necremnus cf. artynes (Walker) and to a lesser extent Stenomesius sp. nr. japonicus (Ashmead) (Hymenoptera: Eulophidae) have been frequently reported attacking T. absoluta in tomato greenhouses in Europe and they are considered as possible biocontrol agents. Few biological data are available on these two species, notably when they parasitize T. absoluta. Therefore, the present study documents their biology when parasitizing T. absoluta, and provides scientific bases for potential development of biological control programs relying on larval ectoparasitoids in invaded areas. We demonstrated the ability of the two parasitoid species to reproduce on T. absoluta as host on tomato plants. On average, N. cf. artynes female parasitized 28.5 larvae and killed 26.2 larvae during its life time whereas S. sp. nr. japonicus female was more long-lived and consequently more efficient; it parasitized 144.3 larvae and killed 90.1 larvae. In a second step, we also explored the effect of T. absoluta larvae instar on parasitism by S. sp. nr. japonicus and subsequent development of the parasitoid offspring. Best results were obtained with the third larvae instar with a higher number of offspring produced, bigger individuals and a higher proportion of females. Our results support the hypothesis that the two species are good candidates for biological control programs targeting T. absoluta in Europe.

Baclofen in the management of inhalant withdrawal: a case series.

Abstract:INTRODUCTION Abuse of inhalants and solvents is a significant public health problem. There is no specific treatment for inhalant withdrawal. OBJECTIVE To study the effect of baclofen in treating craving and withdrawal symptoms in patients with inhalant dependence. CASE REPORTS Case studies of 3 young male patients with DSM-IV diagnoses of inhalant dependence treated in an inpatient setting with baclofen are presented. All patients had nonspecific withdrawal symptoms in the form of irritability, insomnia, and craving. Baclofen was given in doses up to 50 mg/day and was continued throughout the period of hospitalization. DISCUSSION All patients reported significant reduction in withdrawal symptoms within 48 hours of treatment and were free of symptoms for the duration of their hospital stay. One patient continued the medication as an outpatient and has remained abstinent to date. Baclofen was well tolerated by all patients. Our results suggest that baclofen may be an effective treatment modality in this patient population. These effects are possibly due to the agonistic action of baclofen at gamma-aminobutyric acid B receptors in the ventral tegmental area.
Exploring the effects of entomopathogenic nematode symbiotic bacteria and their cell free filtrates on the tomato leafminer Tuta absoluta and its predator Nesidiocoris tenuis

Abstract:The use of biocontrol agents, such as predators and entomopathogenic nematodes, is a promising approach for the effective control of the tomato leafminer Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidaean), an oligophagous insect feeding mainly on Solanaceae species and a major pest of field- and greenhouse-grown tomatoes globally. In this context, the effects of two entomopathogenic nematode species Steinernema carpocapsae (Weiser) (Rhabditida: Steinernematidae) and Heterorhabditis bacteriophora (Poinar) (Rhabditida: Heterorhabditidae), as well as their respective bacterial symbionts, Xenorhabdus nematophila and Photorhabdus luminescens (Enterobacterales: Morganelaceae), which were applied as bacterial cell suspensions and as crude cell-free liquid filtrates on T. absoluta larvae, were investigated. The results showed that of all treatments, the nematodes S. carpocapsae and H. bacteriophora were the most effective, causing up to 98 % mortality of T. absoluta larvae. Regarding bacteria and their filtrates, the bacterium X. nematophila was the most effective (69 % mortality in young larvae), while P. luminescens and both bacterial filtrates showed similar potency (ca. 48-55 % mortality in young larvae). To achieve a holistic approach of controlling this important pest, the impact of these factors on the beneficial predator Nesidiocoris tenuis (Reuter) (Hemiptera: Miridae) was also studied. The results demonstrated that although nematodes and especially S. carpocapsae, caused significant mortality on N. tenuis (87 %), the bacterial cell suspensions of X. nematophila and P. luminescens and crude cell-free liquid filtrates had minimum impact on this beneficial predator (∼11-30 % mortality).

Abstract:The tomato leafminer Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae) represents a global threat to commercial tomato (Solanum lycopersicum L.) production, both in open field and greenhouse. Native to South America, it spread over the Mediterranean Basin, Europe, Africa and part of Asia in only 12 years, and currently it is reported in over 80 countries. Biological control is one of the options for its control and a large number of natural enemies has been reported in association with the pest, both in the areas of origin and of introduction. The egg parasitoid Trichogramma pretiosum, in South America, and the mirid predators Macrolophus pygmaeus and Nesidiocoris tenuis, in Europe and the Mediterranean basin, are used as commercial biocontrol agents. Even if several natural enemies might be promising candidates for biocontrol, their potential role in quantitative pest reduction has been seldom established under practical tomato production conditions. Since climatic suitability indices predict a high probability for continued invasion by T. absoluta, mainly in China and the USA, there is an urgent need for new control options. In order to minimise the use of broad spectrum insecticides, biocontrol techniques should be considered. As tomato is produced seasonally, augmentative biocontrol seems to be the most effective control option, but pest reduction might be optimised by adding conservation biocontrol, and by combining biocontrol within IPM programmes. Here, an overview of predators and parasitoids of T. absoluta in South American and Euro-Mediterranean regions, and their biological control efficacy under laboratory, semi-field and field conditions is provided.

Abstract:The tomato leaf miner, Tuta absoluta is a destructive pest for Solanaceae crops. While pheromone traps are widely used to monitor this pest, the efficiency of this technique remains to be improved. To improve the efficacy of the pheromone traps, a field study was performed in Southwest China where there is a current outbreak of this pest. Different trap heights (0.5, 1.5 and 2.5 m), hanging methods (ladder and vertical), and concentrations of pheromone compounds (0, 1, 2 and 3 mg) were investigated, and their trapping efficiencies were assessed. The results showed that the traps at a height of 0.5 m achieved the best trapping efficiencies among all the tested heights, with trapping effect values ranging between 40.19% and 48.59%. Further, the performance of the ladder hanging method was superior to that of the vertical hanging method. The maximum trapping amount of T. absoluta was achieved with the use of 2.8 mg of pheromone compound mixture. The dose was extrapolated from calculation. This study provides a theoretical basis for the monitoring and trapping assessment of T. absoluta.

Abstract:The tomato leafminer Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae) has recently invaded Mediterranean countries and is a major pest in tomato crops. Trichogrammatid oophagous parasitoids have shown promising potential for controlling the pest before the yield decreases in the greenhouse. In protected tomato crops (greenhouse), mirid predators are commonly used for biological control of whiteflies and they also prey on T. absoluta. These predators do not attack Trichogramma adults but they may partially decrease the overall impact of parasitoids on T. absoluta if intraguild predation (IGP) occurs on parasitized eggs. Under laboratory conditions, we tested if the mirid predator Macrolophus pygmaeus shows preference between parasitized and unparasitized T. absoluta eggs. We also tested if the predator reduces the number of parasitized eggs on caged tomato plants (microcosms) and assessed the efficacy of natural enemies used alone or together in limiting T. absoluta populations. We found that IGP is inflicted on the parasitoid Trichogramma achaeae by M. pygmaeus and that the risk of IGP depends on the developmental stage of T. achaeae inside the egg because the larva is at risk mostly early in its development (when parasitized egg is still yellow). In addition, we observed that non-consumptive events, likely probing of the predator on parasitized eggs, may induce mortality in parasitoid offspring without actual feeding on parasitized eggs. However, both IGP and non-consumptive events decreased when the predator was not confined with parasitized eggs in small arenas. Despite negative effects of the predator to the parasitoid, results demonstrate that adding Trichogramma parasitoids may significantly increase the level of control of the pest over what could be attained when only the mirid predator M. pygmaeus is present on tomato. Implications of results are discussed regarding potential of these natural enemies for biological control of T. absoluta in greenhouse tomato crops.

Abstract:Tuta absoluta, an invasive pinworm characterized by high reproductive potential and adaptation to different agroecological conditions, cause serious damage to tomato crops. Chemical control with synthetic insecticides is widely used to control this pest, although pesticides exhibit side effects on non-target organisms and negatively impact the environment, with the occurrence of resistance to some active substances in the target pest. The use of essential oils (EOs) from aromatic or officinal plants could represent an environmentally safe control method, alternative to synthetic insecticide application. In this work we investigated the effect of EOs from Spanish oregano, laurel, basil, garlic, peppermint, cypress, and eucalyptus EOs on T. absoluta oviposition deterrence and larval repellency. Results showed a remarkable oviposition deterrence using laurel, Spanish oregano, basil, and garlic EOs in both laboratory and greenhouse experiments. In no-choice repellence tests carried 1, 2, 6, and 22 h after treatment application, peppermint and Spanish oregano EOs showed the most lasting effects while cypress and eucalyptus EOs were not effective after 6 and 22 h, respectively. In choice tests, at either 5 or 22 h after treatment application, low preference for the test over control by larvae was observed in all treatments, except for basil EO. The most promising effects were observed in Spanish oregano and laurel treatments where no larvae were detected. Repellent effectiveness of EOs does not seem to be influenced when larvae did not get a chance to do a choice between treated and untreated leaves. This study highlights the effectiveness of EOs of aromatic or officinal plants as an alternative to synthetic insecticides for the control of T. absoluta.

Field assessment of some new insecticides classes and their binary mixtures against Bemisia tabaci and Tuta absoluta and their correlations with tomato productivity

Abstract:Three selected insecticides; chlorantraniliprole, emamectin benzoate and imidacloprid as well as their binary mixtures were assessed against the whitefly, Bemisia tabaci Gennadius and the tomato leafminer, Tuta absoluta Meyrick throughout two growing seasons under field conditions. In addition, the correlation between infestation reduction and tomato yield was also examined. The results indicated that B. tabaci was efficiently controlled by imidacloprid (p < 0.05) followed by chlorantraniliprole and emamectin benzoate. Where adult's reduction percentages were 69.1, 49.1 and 50.7 at 2017 season and 67.5, 52.2 and 55.4 at 2018 season, respectively. The corresponding values for nymphs were 71.3, 49.5 and 50.5 at 2017 season and 66.4, 57.4 and 52.0 at 2018 season. On the other hand, chlorantraniliprole and emamectin benzoate were significantly (p < 0.05) more effective in controlling T. absoluta than imidacloprid with reduction percentages of 84.2, 88.9 and 69.8 at 2017 season and 87.8, 84.6 and 57.9 at 2018 season, consecutively. Indeed, the recommended field rate of the tested insecticides was found to be more effective than the half of the recommended rate. In all treated plots, higher healthy tomato yield production compared to the control plots was achieved. Interestingly, the potential tested compounds which had presented high activity alone didn't show synergism in blends. Simple linear regression analysis of tomato production-infestation reduction relationship showed that total tomato productivity had a positive correlation with T. absoluta and B. tabaci infestation reduction. The correlation was highly significant with T. absoluta (r = 0.895) more than that of B. tabaci, adults and nymphs (r = 0.669 and 0.691, respectively). While the healthy tomato productivity had only a highly positive significant correlation with T. absoluta infestation reduction (r = 0.832). This may suggest that T. absoluta has a serious direct impact on the tomato productivity compared to B. tabaci.

Population Growth Parameters of Tuta absoluta (Lepidoptera: Gelechiidae) on Tomato Plant Using Organic Substrate and Biofertilizers

Abstract:The tomato leafminer, Tuta absoluta (Meyrick) is a devastating pest associated with tomato. In this study, effects of tomato plants treated with vermicompost (20, 40, and 60%), humic fertilizer (2, 4 and 6 g/kg soil) and plant growth promoting rhizobacteria (Pseudomonas fluorescens and Bacillus subtilis) were investigated on the life table parameters of T. absoluta in a growth chamber at 25 ± 2 °C, 65 ± 5% RH, and 16:8 (L:D) h. Significant differences were found for the total developmental time, fecundity, and oviposition period of T. absoluta on the treatments tested. The net reproductive rate (R0), intrinsic rate of natural increase (rm), finite rate of increase (λ), mean generation time (T), and doubling time (DT) of T. absoluta were significantly different among treatments tested. We found that in all vermicompost, humic fertilizer and plant growth promoting rhizobacteria treatments, values of R0, rm, and λ were lower than control treatment. However, the lowest values of these parameters were obtained on 2 g/kg humic fertilizer and 40% vermicompost. Furthermore, T. absoluta had longest T and DT values on 2 g/kg humic fertilizer treatment. Data obtained showed that the addition of 2 g/kg humic fertilizer and 40% vermicompost to the growing soil reduced T. absoluta populations in tomato cultures. In addition, these levels of fertilizers improved growth parameters of tomato seedlings (plant height, wet weight, and dry weight) compared with other treatments. These results could be useful in improving the sustainable management of the moth.

Demography of Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae) reared on elicitor-treated tomato plants with an innovative comparison of projected population sizes and application of the multinomial theorem for population survival

Abstract:Background: Because of the considerable damage caused by tomato leafminer in tomato crops, the use of integrated methods is recommended. In this study, the effect of three different elicitors, methyl jasmonate, salicylic acid and ascorbic acid, on the life table parameters of Tuta absoluta was evaluated. A paired bootstrap test and different bootstrap percentiles were used to compare projected population sizes on specific dates. Survival probabilities were calculated by innovatively linking life tables and multinomial theorem. Results: Preadult duration and mortality significantly increased, and the net reproductive rate (R0 ), intrinsic rate of increase (r), and finite rate of increase (λ) significantly decreased in all elicitor treatments. The lowest fecundity (F = 71.89 eggs/female) was observed in the salicylic acid treatment, with an R0 value of 13.48 offspring/individual, r of 0.0932 d-1 , and λ of 1.0977 d-1 . The population projection revealed the stage structure of T. absoluta during population growth, which was significantly reduced by the elicitor treatments. The survival probability of bootstrap samples was significantly lowered, whereas the extinction probabilities increased in elicitor treatments compared with control when the survival criterion was set to two fertile pairs. Conclusion: Our results demonstrated that the application of elicitors could reduce the risk of T. absoluta damage. Furthermore, population projection based on life tables is applicable to obtain the frequency distribution of the population size at different times. Combined application of life tables and multinomial theorem can be used to calculate the risk of pest emergence. © 2023 Society of Chemical Industry.

Abstract:Background The tomato leafminer , Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae), is one of the most important tomato pests that causes irreversible economic losses. Due to lack of effective control methods, chemical control is still the most preferred approach for management of the pest all over the world. Therefore, researches are mainly focused on implementing a sustainable pest control program against this pest through finding out effective natural enemies. For this purpose, a survey study was conducted to determine the larval parasitoids of T . absoluta in field-grown tomatoes in Adana, Turkey, in 2021–2022. Results Two larval parasitoids, Bracon ( Habrobracon ) didemie Beyarslan and Dolichogenidea appellator (Telenga) (Hymenoptera: Braconidae), were identified morphologically and molecularly. The first parasitoid populations were observed in tomato fields at the end of June and were maintained until the tomato plants were cleared (Middle of July). Throughout the survey, B. didemie was determined to be the more prevalent species than D. appellator . The highest percentage of D . appellator was found in the last week of June in 2021, at 42.8%. When the parasitoid species densities were compared, B . didemie accounted for 93.2% of the total parasitoids. Despite the intensive use of insecticides, the highest parasitoid density was achieved by B . didemie as 28 individuals per 100 leaves. Conclusions Based on these results, it was concluded that B . didemie can be candidate as a successful biological control agent against T . absoluta in tomato field as a part of IPM programs.

Abstract:Background: The tomato, Solanum lycopersicum L., is one of the most important horticultural crops that can be consumed fresh or after being processed worldwide. The tomato leaf miner (Tuta absoluta) is one of the most devastating pest to tomato plants due to its mine-feeding nature in the mesophyll tissue of the plant. Fungal entomopathogens can exist naturally in plants as an asymptote. This study aimed to detect the endophytic colonization of Beauveria bassiana and Metarhizium robertsii within tomato plants via artificial inoculation and their virulence effects on Tuta absoluta. Methods: Isolates with the highest percent germination and virulence against T. absoluta were selected for endophytic evaluation within tomato plants by different artificial inoculation techniques. Results: This study revealed that, isolates with the highest percent germination and virulent to Tuta absoluta had the potential to colonize tomato plants. The result showed that, the maximum mortality rate (97.5%) of Tuta absoluta larvae was achieved by Metarhizium robertsii isolate K-61 at a concentration of 1x10 8conidial/ml at 7 days post inoculated. However, the highest cumulative mortality (100%) was recorded by Beauveria bassiana isolate APPRC-27 at 10 days post inoculated through the direct contact method. The highest endophytic colonization was registered by isolate APPRC-27 (76.67%) at 7 days post-inoculated using the leaf spray technique, but it declined to 11.67% after 28 days of inoculated. In the case of the seedling inoculation technique, the highest endophytic colonization was obtained in the root tissues of tomatoes at 28 days of inoculated by isolate K-61. Conclusions: This study revealed that the leaf spray inoculation technique was the most effective method, followed by seedling inoculation, for the deployment of Beauveria bassiana and Metarhizium robertsii endophytes in tomato plant tissues. Therefore, virulent Beauveria bassiana and Metarhizium robertsii, are promising bioagents for the control of Tuta absoluta if deployed as endophytes.
Efficacy of endophytic wild strains of entomopathogenic fungi against the tomato leafminer Tuta absoluta Meyrick) (Lepidoptera: Gelechiidae) in tomato plants

Abstract:Background Tomato ( Solanum lycopersicum L.) is a vegetable of great economic value. The tomato leafminer Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae) is a serious pest of tomato. Its control is difficult due to the protection of the larvae in the galleries they create its fast resistance development too many active ingredients and due to its many generations per year. Many entomopathogenic fungi (EPF) have been reported as endophytes for various plant pathogens and pests. In this study, the endophyticity of Beauveria bassiana Balsamo (Vuillemin ) (Hypocreales: Cordycipitaceae) and Metarhizium anisopliae (Metchnikoff) Sorokin (Hypocreales: Clavicipitaceae) isolated from the soils of Crete against larvae of T. absoluta in tomato plants was examined. Results As a result, the EPF isolates of Crete decreased both the distance of larval travel from the egg to the opening of the gallery and the weight of the T. absoluta larvae, but also colonized the tomato plants. The entomopathogenic fungi were isolated from tomatoes again. Mycelium began to appear 7 days later and had developed completely after 12 days at 25 ± 2 °C and in 95% RH. Interestingly, the period between the hatched egg and the emerged first instar T. absoluta larvae was affected significantly by the different treatments, but not by "repetition" or "plant". Interestingly, the distance was further significantly shortened after treatment with M. anisopliae isolate Crete and B. bassiana isolate Crete and the commercial strain. The development period of T. absoluta larvae varied significantly among the treatments, but not by repetition in time or plant. The treatments caused a significant variation in the weight of T. absoluta pupae. The lowest weight was recorded in the plants colonized by M. anisopliae isolate Crete. Conclusions The study demonstrated the ability of the B. bassiana and M. anisopliae from Crete to colonize the tomato plants and the wild strains of Crete affect the distance of larvae. With the presence of endophytic EPF, the period between the hatched egg and the emerged first instar T. absoluta larvae was decreased, as does the weight of the T. absoluta pupae.

Abstract:The tomato leafminer, Tuta absoluta (Meyrick), had established in Israel by 2010, attacking both open-field tomatoes and greenhouse crops.We searched for its natural enemies in open-field tomatoes, and tried to determine their potential for controlling this pest. We surveyed the local natural enemies in open tomato fields and measured their impact on pest populations in an unsprayed field. We assessed the suppressive ability of the dominant hemipteran predator, Nesidiocoris tenuis Reuter, against T. absoluta under controlled laboratory conditions and evaluated the impact of its augmentation on T. absoluta control in open-field tomatoes. We found five natural enemy species:the predator, N. tenuis, two braconids, and two eulophids. Predation accounted for 64.5±9.2% (mean ± SE) of T. absoluta larval mortality, whereas parasitism accounted for 20.96±7.5%. Together, they eliminated the pest population at tomato harvest time. Under controlled conditions, predation by N. tenuis rose from 58 to 72% with increased density of T. absoluta, suggesting positive density dependence. The reduction of T. absoluta (83%) by N. tenuis was higher than that of Bemisia tabaci (32%), suggesting a preference of N. tenuis for T. absoluta. Augmentation of N.tenuis was as effective as conventional treatment insecticide treatment, and plant damage was low and did not seem to affect yield. Results indicate that reduced pesticide use enables indigenous natural enemies, particularly N.tenuis, to successfully control T. absoluta and prevent crop damage in open-field tomatoes.
Interactions Between the Nematode Heterorhabditis amazonensis JPM4 and the Predator Macrolophus basicornis: Two Natural Enemies of Tuta absoluta Native to South America

Abstract:Entomopathogenic nematodes (EPNs) and predatory mirids are among the most effective natural enemies for the control of Tuta absoluta, a key pest of tomato worldwide. In the case of South America, the native EPN Heterorhabditis amazonensis and the predator Macrolophus basicornis (Stal) coexist in tomato. The intraguild interactions of two ENPs, H. amazonensis JPM4 and the widely used Steinernema carpocapsae, with the predator M. basicornis, were studied in laboratory setups with the objective of evaluating the potential interactions between them. We hypothesized that the two natural enemies do not interact negatively. The first nymphal stage of the predator was not affected when exposed to tomato leaves directly sprayed with both EPNs, which we attributed to the small size of its stylet. Individuals treated with H. amazonensis JPM4 had low mortality rates in all developmental stages with a maximum of 1.3%. Mortality was higher when the predator was exposed to S. carpocapsae (28% for adults) and correlated positively with increasing developmental stages. In prey choice tests between larvae infected with H. amazonensis and uninfected larvae, M. basicornis fed indistinctively showing no clear preference. When presented with choice larvae infected with S. carpocapsae, the consumption of healthy larvae was higher. In a no-choice prey acceptance experiment, M. basicornis preyed on both healthy and infected larvae with no difference in larvae consumed. Predators that had fed from infected larvae did not exhibit signs of nematode infection and survived. Our results indicate that the South American native H. amazonensis JPM4 and M. basicornis do not show a negative interaction and are promising candidates for combined use in T. absoluta biological control.

Abstract:Introduction: Tuta absoluta is currently considered one of the most devastating invasive pests of solanaceous plants worldwide, causing severe damage to the tomato industry. Insects use volatile organic compounds (VOCs) to locate host plant for feeding and oviposition. Those VOCs could be developed as lures for pest monitoring and control. Methods: In this study, the differentially accumulated VOCs between the preferred host (tomato) and non-preferred host (eggplant) were analyzed by GC–MS method, and their roles on female T. absoluta host selection and egg laying behaviors were investigated using electroantennography (EAG), olfactometer and cage experiments. Results: A total of 39 differentially accumulated VOCs were identified in tomato and eggplant. Strong EAG signals were obtained in 9 VOCs, including 5 VOCs highly accumulated in tomato and 4 VOCs highly accumulated in eggplant. Further olfactometer bioassays showed that 4 compounds (1-nonanol, ethyl heptanoate, ethyl octanoate and o-nitrophenol) were attractive to T. absoluta females, while 5 compounds (2-phenylethanol, 2-pentylfuran, trans,trans-2,4-nonadienal, 2-ethyl-5-methylpyrazine and trans-2-nonenal) were repellent, indicating that VOCs from host plants play important roles in host plant preferences. The attractive activities of 1-nonanol and ethyl octanoate, as well as the repellent activities of trans,trans-2,4-nonadienal and trans-2-nonenal, were further confirmed in cage experiments. Discussion: In this study, two attractants and two repellents for T. absoluta were developed from plant released VOCs. Our results could be useful to enhance the development of eco-friendly and sustainable pest management strategies for T. absoluta .
