Abstract:In this study, soil-grown pakchoi after 2 weeks seedling cultivation were exposed to CeO 2 nanoparticles (CeO 2 NPs) at 0.7, 7, 70, and 350 mg kg −1 for 30 days. Results showed that chlorophyll content and photosynthetic assimilation rate were decreased significantly under all treatments with the largest decrease of 34.16% (0.7 mg kg −1 CeO 2 NPs), however, sub-stomatal CO 2 was increased dramatically under low dose of CeO 2 NPs (0.7 mg kg −1 ). There were 4576, 3548, 2787, and 2514 genes up/down regulated significantly by 0.7, 7, 70, and 350 mg kg −1 CeO 2 NPs, respectively, and 767 genes affected under all treatments. In addition, 0.7 mg kg −1 CeO 2 NPs up-regulated 10 chlorophyll synthesis genes, 20 photosynthesis genes, and 10 carbon fixation enzyme genes; while 350 mg kg −1 CeO 2 NPs down-regulated 5 photosynthesis genes and 28 auxin-activated genes. Among the key genes of photosynthesis, Ferredoxin-NADP reductase (PetH) was upregulated in 0.7, 7 and 70 mg kg −1 treatments, while Photosystem II lipoprotein (Psb27) was downregulated under 7, 70 and 350 mg kg −1 treatments. Top 20 metabolic pathways affected by CeO 2 NPs including plant hormone, amino acids, and glutathione , and carbon metabolism These results provide information about utilizing CeO 2 NPs more safely and effectively in the future.

Abstract:We investigated the frequency and subset origin of circulating factor VIII (FVIII)-specific CD4 T cells in healthy donors. Total CD4 T cells and purified CD4 T-cell subsets were stimulated with FVIII-loaded autologous dendritic cells and challenged for specificity in interferon-γ enzyme-linked immunospots. The number of specific T-cell lines allowed estimation of the frequency of T cells circulating in the blood of the donors. All the 16 healthy donors generated strong in vitro T-cell responses, leading to the generation of 154 FVIII-specific T-cell lines. The mean frequency of FVIII-specific CD4 T cells in healthy donors was similar to that of T cells specific for foreign antigens and greater than that of T cells specific for known immunogenic therapeutic proteins. Normal levels of endogenous FVIII in healthy donors therefore do not prevent a significant escape of FVIII-specific CD4 T cells from negative thymic selection. FVIII-specific T cells mainly originated from both the naïve and central memory cell subsets, but their frequencies remained low as compared with those of cells specific for foreign antigens in immunized donors. The observation of a spontaneous generation of FVIII-specific memory T cells without a global expansion suggests peculiar peripheral tolerance mechanisms to FVIII in healthy donors.
A meta-analysis of elevated O3 effects on herbaceous plants antioxidant oxidase activity

Abstract:Increases in near-surface ozone (O 3 ) concentrations is a global environmental problem. High-concentration O 3 induces stress in plants, which can lead to visible damage to plants, reduced photosynthesis, accelerated aging, inhibited growth, and can even plant death. However, its impact has not been comprehensively evaluated because of the response differences between individual plant species, environmental O 3 concentration, and duration of O 3 stress in plants. We used a meta-analysis approach based on 31 studies 343 observations) to examine the effects of elevated O 3 on malondialdehyde (MDA), superoxide dismutase (SOD), and peroxidase (POD) activities in herbaceous plants. Globally, important as they constitute the majority of the world’s food crops. We partitioned the variation in effect size found in the meta-analysis according to the presence of plant species (ornamental herb, rice, and wheat), O 3 concentration, and duration of O 3 stress in plants. Our results showed that the effects of elevated O 3 on plant membrane lipid peroxidation depending on plant species, O 3 concentration, and duration of O 3 stress in plants. The wheat SOD and POD activity was significantly lower compared to the herbs and rice ( P60 days, may result in antioxidant SOD lose its regulatory ability, and the antioxidant component POD in the symplast is mainly used to resist O 3 damage, and (3) the important factors affected the activity of SOD and POD in plants were not consistent: the duration of O 3 stress in plants was more important than plant species and O 3 concentration for SOD activity. However, for POD activity, plant species was the most important factor.

Central Metabolic Responses to Ozone and Herbivory Affect Photosynthesis and Stomatal Closure

Abstract:Plants have evolved adaptive mechanisms that allow them to tolerate a continuous range of abiotic and biotic stressors. Tropospheric ozone (O3), a global anthropogenic pollutant, directly affects living organisms and ecosystems, including plant-herbivore interactions. In this study, we investigate the stress responses of Brassica nigra (wild black mustard) exposed consecutively to O3 and the specialist herbivore Pieris brassicae Transcriptomics and metabolomics data were evaluated using multivariate, correlation, and network analyses for the O3 and herbivory responses. O3 stress symptoms resembled those of senescence and phosphate starvation, while a sequential shift from O3 to herbivory induced characteristic plant defense responses, including a decrease in central metabolism, induction of the jasmonic acid/ethylene pathways, and emission of volatiles. Omics network and pathway analyses predicted a link between glycerol and central energy metabolism that influences the osmotic stress response and stomatal closure. Further physiological measurements confirmed that while O3 stress inhibited photosynthesis and carbon assimilation, sequential herbivory counteracted the initial responses induced by O3, resulting in a phenotype similar to that observed after herbivory alone. This study clarifies the consequences of multiple stress interactions on a plant metabolic system and also illustrates how omics data can be integrated to generate new hypotheses in ecology and plant physiology.
Transcriptome and Physiological Analysis of Heat and Chilling Stress Responses in Chinese Cabbage (Brassica rapa L. var. pekinensis)

Abstract:Temperature stress impacts the quality and growth of Chinese cabbage. To evaluate resistance mechanisms of temperature stress in Chinese cabbage, the levels of oxidative stress response enzymes such as superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT), as well as metabolites including proline, malondialdehyde (MDA), chlorophylls a and b were estimated. Under temperature stress, the contents of proline and MDA, along with the activities of SOD, POD and CAT increased significantly. Through transcriptome analysis, we found that 11 759 differentially expressed genes were highly enriched in metabolic pathways, which were concentrated on 'starch and sucrose metabolism' and 'plant hormone signal transduction'. Besides, spliceosome pathway was upregulated for heat stress adaptation, but no upregulation of the spliceosome pathway was observed during cold treatment. qRT-PCR validation and transcriptome analysis indicated that the genes of reactive oxygen species scavenging enzymes have different expression patterns under heat and cold stresses. BraA08g030930 (SOD-related), BraA01g040660 (POD-related), and BraA01g003280 (CAT-related) were significantly upregulated under heat stress, while BraA08g015510 (CAT-related) downregulated under cold stress. We found that the transcription factors of MYB s were the main family involved in heat and chilling stress response, and NAC s (NAM, ATAF, and CUC transcription factors) actively responded to chilling stress. Our results reveal the changes in physiological parameters and gene expression patterns under heat and chilling stress, thereby providing valuable insights into elucidating the regulatory mechanisms of cold or heat tolerance in Chinese cabbage.

Abstract:Oxybenzone (OBZ), one of a broad spectrum of ultraviolet (UV) absorbents, has been proven to be harmful to both plants and animals, while omics analysis of big data at the molecular level is still lacking. Lysine succinylation (Ksuc) is an important posttranslational modification of proteins that plays a crucial role in regulating the metabolic network in organisms under stress. Here, we report the changes in intracellular Ksuc modification in plants under OBZ stress. A total of 1276 succinylated sites on 507 proteins were identified. Among these sites, 181 modified proteins were hypersulfinylated/succinylated in OBZ-stressed pakchoi leaves. Differentially succinylated proteins (DSPs) are distributed mainly in the chloroplast, cytoplasm, and mitochondria and are distributed mainly in primary metabolic pathways, such as reactive oxygen species (ROS) scavenging, stress resistance, energy generation and transfer, photosynthetic carbon fixation, glycolysis, and the tricarboxylic acid (TCA) cycle. Comprehensive analysis shows that Ksuc mainly changes the carbon flow distribution, enhances the activity of the antioxidant system, affects the biosynthesis of amino acids, and increases the modification of histones. The results of this study first showed the profiling of the Kusc map under OBZ treatment and proposed the adaptive mechanism of pakchoi in response to pollutants and other abiotic stresses at the posttranslational level, which revealed the importance of Ksuc in the regulation of various life activities and provides a reference dataset for future research on molecular function.

The phototoxicity of sulfamethoxazole stress on pakchoi cabbage (Brassica rapa var. chinensis) seedlings: From the perspective of photoreaction and omics analysis

Abstract:The increasing use of antibiotics has attracted widespread attention to their environmental risks. However, the phototoxicity of sulfonamide antibiotics to plants remain unclear. In this study, the mechanism of the effect of sulfamethoxazole on photosynthesis of pakchoi cabbage (Brassica rapa var. chinensis) was investigated. The results showed that sulfamethoxazole inhibited the growth of pakchoi cabbage and produced photosynthetic toxicity. The growth inhibition rates increased with concentration, the root and shoot weight were 76.02 % and 47.04 % of the control, respectively, with stay-greens phenomenon in 4 mg·L-1 sulfamethoxazole treatment. Chlorophyll precursors (protoporphyrin IX (Proto IX), Mg-proto IX, and protochlorophyllide (Pchlide), 5-aminolevulinic acid (ALA), and porphobilinogen (PBG)) were 1.38-, 1.26-, 1.12-, 1.71-, and 0.96-fold of the control, respectively; photosynthetic pigments (chlorophyll a, chlorophyll b, and carotenoids) were 1.26-, 1.39-, and 1.03-fold of the control, respectively. Respiration rate was 271.42 % of the control, whereas the net photosynthetic rate was 50.50 % of the control. The maximum photochemical quantum yield of PSII (Fv/Fm), the actual photosynthetic efficiency (Y(II)), the quantum yield of non-regulated energy dissipation (Y(NO)), the apparent electron transfer efficiency of PSII (ETR) under actual light intensity were affected, and chloroplast swelling was observed. Proteomic analysis showed that photosynthesis-related pathways were significantly up-regulated, biological processes such as light response, carbohydrates, and reactive oxygen species were activated. Metabolomic analysis revealed that the tricarboxylic acid cycle (TCA cycle) and carbohydrate catabolism were stimulated significantly (p < 0.05), sugars and amino acids were increased to regulate and enhance the resilience of photosynthesis. While folate biosynthesis and ribosomal pathways were significantly down-regulated, the synthesis and translation processes of amino acids and nucleotides were inhibited.
Metabolite Profiling for Model Cultivars of Wheat and Rice under Ozone Pollution

Abstract:As essential source for human consumption, plants of wheat, rice and soybean are highly sensitive to ozone (O3), resulting in significant agricultural losses under O3 pollution. However, little is known about the effects of elevated O3 on their metabolite profiling. In this study, three model cultivars were used for the metabolome analysis under elevated O3 and charcoal filtered air. Our study revealed that wheat and rice differed significantly from soybean in metabolic number and certain pathways. Metabolites response to elevated O3 were less in soybean, whilst those in wheat and rice were considerably larger. Under O3 stress, tricarboxylic acid cycle (TCA) was impaired in three crop plants. Methylerythritol 4-phosphate pathway and glycerol phosphate pathway were altered in wheat and rice with reduced terpene accumulation and high level of phospholipids. However, these pathways were not affected in soybean. Meanwhile, O3 suppressed the generation of flavonoid via benzoic acid pathway in three crop plants. Accordingly, the expressional level of genes coding key enzymes which catalyzed the synthesis or degradation of these metabolites. These findings provide valuable information for understanding of ozone’s effects on the metabolite profiling of crop plants, exploring the metabolite differences of three crop plants under elevated O3.
Lead exposure dose-dependently affects oxidative stress, AsA-GSH, photosynthesis, and mineral content in pakchoi (Brassica chinensis L.)

Abstract:Lead (Pb) is a heavy metal pollutant and negatively affects agriculture and ecosystems. Pb can cause oxidative stress and abnormal plant growth. The ascorbic acid-glutathione (AsA-GSH) cycle mainly exists in chloroplasts and resists oxidative stress, scavenges reactive oxygen radicals, and maintains normal photosynthesis. However, the dosage related effects of Pb on pakchoi photosynthesis, via oxidative stress and the AsA-GSH system, remains unclear. In this study, various Pb dosage stress models were tested (low: 300 mg/kg; medium: 600 mg/kg; high: 900 mg/kg). Pb stress induced a dose-dependent increase in Pb content in pakchoi leaves (P < 0.05). Principal component analysis showed that Se, B, and Pb were significantly and negatively correlated. Pb stress also increased MDA content and decreased antioxidant enzymes SOD, GSH-Px, and T-AOC activities (P < 0.05). We also found that Vc content, as well as the GSH/GSSG ratio, decreased. Additionally, Pb stress destroyed chloroplast structure, decreased photosynthesis indicators Pn, Tr, Gs, Ci and VPD, and attenuated Fv/Fm and Fv/Fo (P < 0.05). In the high-dose group, the contents of chlorophyll a, chlorophyll b, and carotenoids decreased significantly, while the expression of chloroplast development genes (GLK, GLN2) decreased (P < 0.05). Our data suggest that Pb stress leads to dosage-dependent, aberrant photosynthesis by inhibiting the AsA-GSH system in pakchoi. This study expands the Pb toxicology research field and provides indications for screening antagonists.

Abstract:Glucosinolates and their breakdown products are well-known for their cancer-chemoprotective functions and biocidal activities against pathogens and generalist herbivores. Insecticides are commonly used in the production of pak choi (Brassica rapa L. ssp. chinensis var. communis [N. Tsen & S.H. Lee] Hanelt). We studied the effects of four commonly used insecticides, namely, beta-cypermethrin, acephate, pymetrozine, and imidacloprid, on glucosinolate metabolism in pak choi. All insecticides significantly increased both the transcription of glucosinolate biosynthetic genes and the aliphatic and total glucosinolate accumulations in pak choi. beta-Cypermethrin and acephate caused gradual and continuous up-regulation of gene expression from 0.5 to 24 h after treatment, whereas pymetrozine and imidacloprid did so more rapidly, reaching a peak at 1 h and returning to normal at 3 h. Our findings indicate that the four insecticides affect glucosinolate metabolism in pak choi plants to various degrees and suggest that glucosinolates may be involved in plant insecticide metabolism.

Abstract:Plants live in a world of changing environments, where they are continuously challenged by alternating biotic and abiotic stresses. To transfer information from the environment to appropriate protective responses, plants use many different signaling molecules and pathways. Reactive oxygen species (ROS) are critical signaling molecules in the regulation of plant stress responses, both inside and between cells. In natural environments, plants can experience multiple stresses simultaneously. Laboratory studies on stress interaction and crosstalk at regulation of gene expression, imply that plant responses to multiple stresses are distinctly different from single treatments. We analyzed the expression of selected marker genes and reassessed publicly available datasets to find signaling pathways regulated by ozone, which produces apoplastic ROS, and high light treatment, which produces chloroplastic ROS. Genes related to cell death regulation were differentially regulated by ozone versus high light. In a combined ozone + high light treatment, the light treatment enhanced ozone-induced cell death in leaves. The distinct responses from ozone versus high light treatments show that plants can activate stress signaling pathways in a highly precise manner.

Response of carbon, nitrogen and phosphorus concentration and stoichiometry of plants and soils during a soybean growth season to O3 stress and straw return in Northeast China

Abstract:Carbon (C), nitrogen (N) and phosphorus (P) concentrations and stoichiometry play important roles in biogeochemical cycles of the ecosystems, yet it is still unclear how the allocations of C, N and P concentrations and stoichiometry among plant organs and soils related to O3 stress and straw return. Here, a pot experiment was conducted in open top chambers to monitor the response of C, N and P concentrations and stoichiometry of leaves, stems, roots and soils during a growing season (branching, flowering and podding stages) of soybean (Glycine max; a species highly sensitive to O3) to background O3 concentration (44.8 ± 5.6 ppb), O3 stress (79.7 ± 5.4 ppb) and straw treatment (no straw return and straw return). O3 stress significantly decreased root biomass. Straw return significantly increased root biomass under O3 stress at branching and flowering stages. Generally, O3 stress and straw return showed significant effects on the C, N and P concentrations of leaves and soils, and stoichiometric ratios of leaves, stems and microbial biomass. The C, N and P concentrations and stoichiometry of leaves, stems, roots and soils in response to O3 stress and straw return at the branching stage were inconsistent with the changes observed at the flowering and podding stages. The P conversion efficiency showed significant relationship with root P concentration under the combined effects of O3 stress and straw return. Altogether, the present study indicated that C, N and P concentrations of soybean might be more important than stoichiometric ratios as a driver of root defence against O3 stress in the case of straw return.

Transcriptomics Underlying Pulmonary Ozone Pathogenesis Regulated by Inflammatory Mediators in Mice

Abstract:Ozone (O3) is the predominant oxidant air pollutant associated with airway inflammation, lung dysfunction, and the worsening of preexisting respiratory diseases. We previously demonstrated the injurious roles of pulmonary immune receptors, tumor necrosis factor receptor (TNFR), and toll-like receptor 4, as well as a transcription factor NF-κB, in response to O3 in mice. In the current study, we profiled time-dependent and TNFR- and NF-κB-regulated lung transcriptome changes by subacute O3 to illuminate the underlying molecular events and downstream targets. Mice lacking Tnfr1/Tnfr2 (Tnfr-/-) or Nfkb1 (Nfkb1-/-) were exposed to air or O3. Lung RNAs were prepared for cDNA microarray analyses, and downstream and upstream mechanisms were predicted by pathway analyses of the enriched genes. O3 significantly altered the genes involved in inflammation and redox (24 h), cholesterol biosynthesis and vaso-occlusion (48 h), and cell cycle and DNA repair (48–72 h). Transforming growth factor-β1 was a predicted upstream regulator. Lack of Tnfr suppressed the immune cell proliferation and lipid-related processes and heightened epithelial cell integrity, and Nfkb1 deficiency markedly suppressed lung cell cycle progress during O3 exposure. Common differentially regulated genes by TNFR and NF-κB1 (e.g., Casp8, Il6, and Edn1) were predicted to protect the lungs from cell death, connective tissue injury, and inflammation. Il6-deficient mice were susceptible to O3-induced protein hyperpermeability, indicating its defensive role, while Tnf-deficient mice were resistant to overall lung injury caused by O3. The results elucidated transcriptome dynamics and provided new insights into the molecular mechanisms regulated by TNFR and NF-κB1 in pulmonary subacute O3 pathogenesis.

Abstract:Carotenoid oxygenase is a key enzyme involved in the metabolism of carotenoids, which are important quality factors affecting the pigmentation and aroma of flowers and fruits, as well as main regulators of plant phytohormones, such as abscisic acid (ABA) and strigolactone (SL). To investigate the regulation of carotenoid metabolism, including ABA and SL biosynthesis by carotenoid oxygenase genes, in B. rapa and B. oleracea, we studied the expression of these genes in response to salt, drought, cold, ABA, and SL treatments. In the Brassica database, we identified genes related to carotenoid cleavage dioxygenase (CCD) 1, CCD4, CCC7, CCD8, NCED (9-cis-epoxycarotenoid dioxygenase) 2, NCED3, NCED5, NCED 6, and NCED9. We classified 15 and 14 carotenoid oxygenase candidate genes in B. rapa and B. oleracea, respectively. CCD1 and CCD4 showed similar transcription trends under abiotic stress treatment conditions in both B. rapa and B. oleracea, with expression peaking at 3–6 h post treatment before decreasing at 12 h. NCED2, NCED6, and NCED9 were responsive to ABA and SL treatments. Our results show that the phytohormone biosynthetic enzymes involved in carotenoid oxygenase pathways may be activated as multifunctional stress signaling factors with a variety of transcriptional responses via their interactions under abiotic stress treatment conditions in B. rapa and B. oleracea.

Transcriptome Analysis of Ethylene-Related Genes in Chlorine Dioxide-Treated Fresh-Cut Cauliflower

Abstract:Chlorine dioxide (ClO2) is widely used for the quality preservation of postharvest horticultural plants. However, the molecular mechanism of how ClO2 works is not clear. The purpose of this study was to understand ethylene-related molecular signaling in ClO2-treated fresh-cut cauliflower florets. Transcriptome analysis was used to investigate ethylene-related gene regulation. A total of 182.83 Gb clean data were acquired, and the reads of each sample to the unique mapped position of the reference genome could reach more than 85.51%. A sum of 2875, 3500, 4582 and 1906 differential expressed genes (DEGs) were identified at 0 d, 4 d, 8 d and 16 d between the control group and ClO2-treated group, respectively. DEGs were enriched in functions such as 'response to oxygen-containing compounds' and 'phosphorylation', as well as MAPK signaling pathway, plant hormone transduction pathway and so on. Genes, including OXI1, MPK3, WRKY22 and ERF1, which are located at the junction of wounding, pathogen attack, pathogen infection or ethylene signal transduction pathways, were up-regulated in response to stress. ETR and CTR1 (both up-regulated), as well as three down-regulated genes, including BolC5t34953H (a probable NAC), BolC1t05767H (a probable NAC) and BolC2t06548H (a probable ERF13), might work as negative regulators for ethylene signal transduction. In conclusion, ethylene-related genes and pathways are involved in ClO2 treatment, which might enhance stress resistance and have a negative feedback mechanism.

Comparative Transcriptomic Analysis Reveals the Involvement of Auxin Signaling in the Heat Tolerance of Pakchoi under High-Temperature Stress

Abstract:Pakchoi is a kind of nonheading Chinese cabbage being widely cultivated not only in China but also all over Asia. High temperature is a major limiting factor influencing the yield and quality of pakchoi, while the mechanism of pakchoi dealing with high-temperature challenges remains largely elusive. In the present study, we conducted a comparative transcriptomic analysis, which was also validated by qPCR, of the heat-tolerant Xinxiaqing (XXQ) variant and Suzhouqing (SZQ) variant, which are heat-sensitive under high-temperature treatment. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses suggest that high-temperature-induced phytohormones signal transduction, especially auxin signal transduction, regulates the heat responses of pakchoi. Our further investigations imply that high-temperature-activated auxin signal plays a positive role in helping pakchoi deal with high-temperature challenge; IAA-pretreated pakchoi plants exhibited greater resistance to the high-temperature treatment, probably due to the induction of antioxidant activity. In addition, our study also identified six heat shock proteins/factors (HSPs/HSFs) whose up-regulation correlates with the elevated heat tolerance of pakchoi. Notably, among these high-temperature-induced heat-responsive factors, HSP20 and HSP26.5 are under the regulation of auxin signal, and this signal cascade contributes to enhancing the thermostability of pakchoi. In the present study, we identified crucial high-temperature-responsive factors and signaling pathways in pakchoi, which help in understanding the mechanism of pakchoi coping with high-temperature challenge.

The Physiological and Molecular Responses of Arabidopsis Thaliana to the Stress of Oxalic Acid

Abstract:Many fungal phytopathogens can secrete oxalic acid (OA), which is the crucial pathogenic determinant and plays important roles in pathogenicity and virulence of pathogen during infection process. However, how plants respond to OA stress still needs further characterization. In this study, we observed the physiological and molecular responses of Arabidopsis thaliana to OA stress. The leaves of 6-wk-old A. thaliana were sprayed with OA and distilled water respectively, and 0, 2, 4, 8, 12, and 24 h later, the leaves were collected and the contents of MDA, H2O2, and GSH, and the activities of CAT, SOD, and POD were determined and the expressions of PR1 and PDF1.2 were also studied. Under the stress of 30 mmol L−1 OA, SOD activity was first enhanced to reduce the accumulation of O2−. But immediately, POD, CAT, and GSH all decreased extremely resulting in the accumulation of H2O2, and the MDA content increased 24 h later. GSH activity was enhanced significantly at 24 h after OA used. However, H2O2 wasn't eliminated at the same time, suggesting that the activity inhibitions of POD and CAT might be the reasons that caused Arabidopsis cells' impairment under OA stress. RT-PCR results indicated that PDF1.2, a marker gene of the JA/ET signaling was significantly induced; PR1, an indicator gene in SA signaling, was slighlty induced from 8 to 12 h after OA stress. In conclusion, Arabidopsis may recruit metabolism of reactive oxygen, both JA/ET and SA signaling pathways to respond to OA stress. These results will facilitate our further understanding the mechanisms of plant response to OA and OA-dependent fungal infection.
Characterization and co-expression analysis of WRKY orthologs involved in responses to multiple abiotic stresses in Pak-choi (Brassica campestris ssp. chinensis)

Abstract:Background: The WRKY transcription factor is an important member of the stress-related transcription factors, which mediate diverse abiotic stresses in many plants. However, up until now, the number of WRKY members, and the regulatory mechanisms involved in abiotic stress responses in Pak-choi (Brassica campestris ssp. chinensis), remained unknown. Results: We isolated and identified 56 full-length WRKY cDNAs from a Pak-choi stress-induced cDNA library. The 56 putative BcWRKY proteins were divided into three groups based on structural and phylogenetic analyses. A subcellular localization prediction indicated that the putative BcWRKY proteins were enriched in the nuclear region. Experiments involving BcWRKY25 and BcWRKY40 confirmed the prediction. A total of 22 BcWRKYs were differentially expressed in response to at least one stress condition (abscisic acid, cold, salinity, heat, or osmosis) tested on Pak-choi leaves, and a co-expression analysis indicated stress-inducible BcWRKYs co-regulated multiple abiotic stresses. BcWRKY33, BcWRKY40, BcWRKY53, and BcWRKY70 acted as key regulators and played dominant roles within co-regulatory networks of stress-inducible BcWRKYs. Conclusions: We first isolated and characterized the 56 stress-inducible WRKY transcription factor family members. A total of 22 stress-inducible BcWRKYs found in leaves can co-regulate multiple environmental stresses by integrating the potential mutual interactions of WRKYs in Pak-choi. This information will be valuable when exploring the molecular mechanisms of WRKYs in response to abiotic stresses in plants.
Ozone stress-induced DNA methylation variations and their transgenerational inheritance in foxtail millet

Abstract:Elevated near-surface ozone (O3) concentrations have surpassed the tolerance limits of plants, significantly impacting crop growth and yield. To mitigate ozone pollution, plants must evolve a rapid and effective defense mechanism to alleviate ozone-induced damage. DNA methylation, as one of the most crucial epigenetic modifications, plays a pivotal role in maintaining gene stability, regulating gene expression, and enhancing plant resilience to environmental stressors. However, the epigenetic response of plants to O3 stress, particularly DNA methylation variations and their intergenerational transmission, remains poorly understood. This study aims to explore the epigenetic mechanisms underlying plant responses to ozone stress across generations and to identify potential epigenetic modification sites or genes crucial in response to ozone stress. Using Open Top Chambers (OTCs), we simulated ozone conditions and subjected foxtail millet to continuous ozone stress at 200 nmol mol-1 for two consecutive generations (S0 and S1). Results revealed that under high-concentration ozone stress, foxtail millet leaves exhibited symptoms ranging from yellowing and curling to desiccation, but the damage in the S1 generation was not more severe than that in the S0 generation. Methylation Sensitive Amplified Polymorphism (MSAP) analysis of the two generations indicated that ozone stress-induced methylation variations ranging from 10.82% to 13.59%, with demethylation events ranged from 0.52% to 5.58%, while hypermethylation occurred between 0.35% and 2.76%. Reproductive growth stages were more sensitive to ozone than vegetative stages. Notably, the S1 generation exhibited widespread demethylation variations, primarily at CNG sites, compared to S0 under similar stress conditions. The inheritance pattern between S0 and S1 generations was mainly of the A-A-B-A type. By recovering and sequencing methylation variant bands, we identified six stress-related differential amplification sequences, implicating these variants in various biological processes. These findings underscore the potential significance of DNA methylation variations as a critical mechanism in plants' response to ozone stress, providing theoretical insights and references for a comprehensive understanding of plant adaptation mechanisms to ozone stress and the epigenetic role of DNA methylation in abiotic stress regulation.
RNA-Seq Reveals Transcription Factors Involved in Temperature-mediated Anthocyanin Accumulation and Biosynthesis in Purple Pakchoi ( Brassica campestris ssp. chinensis Makino.)

Abstract:transcription factors involved in temperature-mediated anthocyanin accumulation and in ( Molecular Abstract Temperature is a main environmental factor that affects anthocyanin biosynthesis and accumulation in purple pakchoi ( Brassica campestris ssp . Chinensis Makino. ). Purple pakchoi is one of the most popular vegetables with high content of anthocyanin in China. Recently, we found that the purple color of purple pakchoi cultivar, "ziyi", deepened after 10-day low temperature(5°C, LT) treatment with increased anthocyanin content compared to plants after 20°C (normal temperature, NT, control) treatment. Contrarily, the color of pakchoi treated with 10-day heat temperature (35°C, HT) became lighter with decreased anthocyanin content than that of control. The transcriptom analysis revealed a total of 51 008 unigenes from plants treated with NT ， LT, and HT by RNA-seq. A total of 4 321 and 8 455 differentially expressed genes (DEGs) were identified from HT and LT compared to NT, respectively. Among these DEGs, 173 unigenes were downregulated in LT and upregulated in HT compared to NT. 218 unigenes were upregulated in LT and downregulated in HT. Further Gene Ontology enrichment analysis revealed a series of candidate genes that may be involve in temperature-mediated anthocyanin accumulation, including structural genes and 20 transcription factors. Collectively, our study provides a global view of transcriptiomic resources in response to temperature-induced anthocyanin accumulation in purple pakchoi. Among these DEG, a total of 391 unigens responded to LT and HT which 173 unigenes were upregulated in HT and downregulated in LT whereas 218 were downregulated in HT and upregulated in LT. These data indicated that the 391 unigenes might involve in temperature-associated anthocyanin biosynthesis and accumulation.
