Abstract:The application of immobilized lipase in the enzymatic production of biodiesel has shown numerous advantages. In this study, surface of Polyacrylonitrile (PAN) hollow membrane was first modified using nitrile-click chemistry in order to fit for interaction with enzyme proteins. Then sodium alginate (SA) was introduced and the membrane was post-treated by CaCl2. When the prepared PAN-PEI-SA-CaCl2 was used for lipase immobilization, the protein loading was 36.90 mg/g, and the enzyme activity reached up to 54.47 U/g, which was 2.5 times as much as that of Novozym® 435. As a result, the constructed immobilized lipase obtained a maximum biodiesel yield of 78.5%, which was 2.4 times that of the Novozym® 435 in transesterification reactions. Moreover, the biodiesel yield decreased by only 11% after the immobilized enzyme was continuously used for 20 times. This study exhibits that this technic has broad application prospects in the field of conversion of biomass resources.
Effect of Immobilization by Modified Ultrastable-Y Molecular Sieve on the Resolution of (R,s)-2-octanol in Nonaqueous Media by Lipase

Abstract:The lipase from Penicillium expansum PED-03 (PEL) was immobilized into a modified ultrastable-Y molecular sieve and the resolution of (R,S)-2-octanol was carried out in a bioreactor in nonaqueous media by the immobilized lipase to investigate the effect of immobilization on the catalytic property of PEL duing the enzymatic reaction. It was found that the conversion (c), enantiomeric excess (ee), and enantioselectivity (E) of the resolution catalyzed by the PEL immobilized on the modified ultrastable-Y molecular sieve were much higher than those of the reaction catalyzed by free PEL and PEL immobilized on other supports. Being immobilized on the modified Ultrastable-Y molecular sieve, the lipase exhibited obvious activity in a wider pH range and at a much higher temperature than the free PEL, and showed a marked enhancement of stability against thermal inactivation, by which the suitable pH value of the buffer used for immobilization could be "memorized". The conversion of the reaction catalyzed by PEL immobilized on the modified ultrastable-Y molecular sieve reached 97.68% of the theoretical value with excellent enantioselectivity (Average E of 8 batches >460) in nonaqueous media at "memorial" pH 9.5, 50 degrees C for 24 h, which showed a good application potential to the production of optically pure (R,S)-2-octanol.
Effect of the Presence of Surfactants and Immobilization Conditions on Catalysts’ Properties of Rhizomucor miehei Lipase onto Chitosan

Abstract:Lipase from Rhizomucor miehei (RML) was immobilized onto chitosan support in the presence of some surfactants added at low levels using two different strategies. In the first approach, the enzyme was immobilized in the presence of surfactants on chitosan supports previously functionalized with glutaraldehyde. In the second one, after prior enzyme adsorption on chitosan beads in the presence of surfactants, the complex chitosan beads-enzyme was then cross-linked with glutaraldehyde. The effects of surfactant concentrations on the activities of free and immobilized RML were evaluated. Hexadecyltrimethylammonium bromide (CTAB) promoted an inhibition of enzyme activity while the nonionic surfactant Triton X-100 caused a slight increase in the catalytic activity of the free enzyme and the derivatives produced in both methods of immobilization. The best derivatives were achieved when the lipase was firstly adsorbed on chitosan beads at 4 °C for 1 h, 220 rpm followed by cross-link the complex chitosan beads-enzyme with glutaraldehyde 0.6% v.v−1 at pH 7. The derivatives obtained under these conditions showed high catalytic activity and excellent thermal stability at 60° and 37 °C. The best derivative was also evaluated in the synthesis of two flavor esters namely methyl and ethyl butyrate. At non-optimized conditions, the maximum conversion yield for methyl butyrate was 89%, and for ethyl butyrate, the esterification yield was 92%. The results for both esterifications were similar to those obtained when the commercial enzyme Lipozyme® and free enzyme were used in the same reaction conditions and higher than the one achieved in the absence of the selected surfactant.

Multipoint covalent immobilization of lipase on chitosan hybrid hydrogels: influence of the polyelectrolyte complex type and chemical modification on the catalytic properties of the biocatalysts

Abstract:This work aimed at the production of stabilized derivatives of Thermomyces lanuginosus lipase (TLL) by multipoint covalent immobilization of the enzyme on chitosan-based matrices. The resulting biocatalysts were tested for synthesis of biodiesel by ethanolysis of palm oil. Different hydrogels were prepared: chitosan alone and in polyelectrolyte complexes (PEC) with κ-carrageenan, gelatin, alginate, and polyvinyl alcohol (PVA). The obtained supports were chemically modified with 2,4,6-trinitrobenzene sulfonic acid (TNBS) to increase support hydrophobicity, followed by activation with different agents such as glycidol (GLY), epichlorohydrin (EPI), and glutaraldehyde (GLU). The chitosan-alginate hydrogel, chemically modified with TNBS, provided derivatives with higher apparent hydrolytic activity (HA(app)) and thermal stability, being up to 45-fold more stable than soluble lipase. The maximum load of immobilized enzyme was 17.5 mg g(-1) of gel for GLU, 7.76 mg g(-1) of gel for GLY, and 7.65 mg g(-1) of gel for EPI derivatives, the latter presenting the maximum apparent hydrolytic activity (364.8 IU g(-1) of gel). The three derivatives catalyzed conversion of palm oil to biodiesel, but chitosan-alginate-TNBS activated via GLY and EPI led to higher recovered activities of the enzyme. Thus, this is a more attractive option for both hydrolysis and transesterification of vegetable oils using immobilized TLL, although industrial application of this biocatalyst still demands further improvements in its half-life to make the enzymatic process economically attractive.
Protection of Opening Lids: Very High Catalytic Activity of Lipase Immobilized on Core-Shell Nanoparticles.

Abstract:Various hydrophobic supports have been used for lipase immobilization since the active site of lipase can be opened in a hydrophobic environment. Nevertheless, the increase of lipase activity is still limited. This study demonstrates a hyperactivation-protection strategy of lipase after immobilization on poly(n-butyl acrylate)-polyaldehyde dextran (PBA-PAD) core-shell nanoparticles. The inner hydrophobic PBA domain helps to rearrange lipase conformation to a more active form after immobilization into the PAD shell. More importantly, the outer PAD shell with dense polysaccharide chains prevents the immobilized lipase from contact with outside aqueous medium and revert its conformation back to an inactive form. As a result, under optimal conditions the activity of lipase immobilized in PBA-PAD nanoparticles was enhanced 40 times over the free one, much higher than in any previous report. Furthermore, the immobilized lipase retained more than 80 % of its activity after 10 reaction cycles.
Stabilization of Immobilized Lipases by Intense Intramolecular Cross-Linking of Their Surfaces by Using Aldehyde-Dextran Polymers

Abstract:Immobilized enzymes have a very large region that is not in contact with the support surface and this region could be the target of new stabilization strategies. The chemical amination of these regions plus further cross-linking with aldehyde-dextran polymers is proposed here as a strategy to increase the stability of immobilized enzymes. Aldehyde-dextran is not able to react with single amino groups but it reacts very rapidly with polyaminated surfaces. Three lipases-from Thermomyces lanuginosus (TLL), Rhizomucor miehiei (RML), and Candida antarctica B (CALB)-were immobilized using interfacial adsorption on the hydrophobic octyl-Sepharose support, chemically aminated, and cross-linked. Catalytic activities remained higher than 70% with regard to unmodified conjugates. The increase in the amination degree of the lipases together with the increase in the density of aldehyde groups in the dextran-aldehyde polymer promoted a higher number of cross-links. The sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) analysis of those conjugates demonstrates the major role of the intramolecular cross-linking on the stabilization of the enzymes. The highest stabilization was achieved by the modified RML immobilized on octyl-Sepharose, which was 250-fold more stable than the unmodified conjugate. The TLL and the CALB were 40-fold and 4-fold more stable than the unmodified conjugate.

Abstract:Efforts have recently been made to improve the biocompatibility of support surface for enzyme immobilization, which could create a specific microenvironment for the enzymes and thus benefit the enzyme activity. In this work, one natural macromolecule, chitosan, was tethered on the surface of poly(acrylonitrile-co-maleic acid) (PANCMA) membrane to prepare a dual-layer biomimetic support for enzyme immobilization. Lipase from Candida rugosa was immobilized on this dual-layer biomimetic support by adsorption. The properties of the immobilized enzyme were assayed and compared with those of the free one. It was found that the adsorption capacity of lipase on the chitosan-tethered PANCMA membrane increases with the decrease of ionic strength and there is an optimum pH value for the adsorption. The activity retention of the immobilized lipase on the chitosan-tethered membrane by adsorption (54.1%) is higher than that by chemical bonding (44.5%). In comparison with the immobilized lipase by chemical bonding, there is a decrease of the Km value and an increase of the Vmax value for the immobilized lipase by adsorption. Additionally, the experimental results of thermal stabilities indicate that the residual activity of the immobilized lipase at 50°C is 38% by adsorption and 65% by chemical bonding.
Immobilization of Thermomyces lanuginosus lipase by different techniques on Immobead 150 support: characterization and applications

Abstract:Thermomyces lanuginosus lipase (TLL) was immobilized on native and modified Immobead 150, with epoxy groups removed by hydrolysis and oxidized to add aldehyde on its surface. Immobilizations on both supports were performed by adsorption, adsorption and cross-linking, covalent attachment, multipoint covalent attachment, and, for the modified support, multipoint covalent attachment using ethylenediamine. Biocatalysts were evaluated for thermal and solvent stabilities, and the best biocatalyst was also tested after incubation in ionic liquids and used in the synthesis of butyl butyrate and isoamyl butyrate. Multipoint covalent immobilized TLL on the native Immobead 150 (Emulti) showed a half-life of 5.32 h at 70 °C, being approximately 30 times more stable than its soluble form; it showed high stability in acetone, hexane, and isooctane. Its enzymatic activity was up to 40% when incubated in ionic liquids. Ester synthesis produced yields of esterification above 60% in 24 h. Of all immobilization protocols, the Emulti performed best concerning the thermal, solvent, and ionic liquids stabilities. Emulti was successfully applied to the synthesis of butyl butyrate and isoamyl butyrate, which are very important products for the food and beverage industries.
Preparation and characterization of stable chitosan nanofibrous membrane for lipase immobilization

Abstract:Nanofibrous membrane with a fiber diameter of 80–150nm was fabricated from mixed chitosan/poly(vinyl alcohol) (PVA) solution by an electrospinning process. Field emission scanning electron microscope and transmission electron microscope were used to characterize the morphology of the nanofibrous membrane. It was found that chitosan nanofibrous membrane with stabilized morphology could be prepared through removing most of PVA from the nascent one with 0.5M NaOH aqueous solution. This treatment also resulted in an obvious decrease in fiber diameter. The stabilized chitosan nanofibrous membrane was explored as support for enzyme immobilization due to the characteristics of excellent biocompatibility, high surface/volume ratio, and large porosity. Lipase from Candida rugosa was immobilized on the nanofibrous membrane using glutaraldehyde (GA) as coupling reagent. The properties of the immobilized lipase were assayed and compared with the free one. Results showed that, the observed lipase loading on this nanofibrous membrane was up to 63.6mg/g and the activity retention of the immobilized lipase was 49.8% under the optimum condition. The pH and thermal stabilities of lipase were improved after it was immobilized on the chitosan nanofibrous membrane. In addition, the experimental results of reusability and storage stability indicated that the residual activities of the immobilized lipase were 46% after 10 cycles and 56.2% after 30 days, which were obviously higher than that of the free one.

Abstract:In this work, we report the preparation of bacterial cellulose (BC)-chitosan composite hydrogel beads by co-dissolution of BC and chitosan in 1-ethyl-3-methylimidazolium acetate and subsequent reconstitution with distilled water. The BC-chitosan hydrogel beads were used as enzyme supports for immobilizing Candida rugosa lipase by physical adsorption and covalent cross-linking. BC-chitosan hydrogel beads immobilized lipase more efficiently than microcrystalline cellulose (MCC)-chitosan hydrogel beads. The amount of protein adsorbed onto BCchitosan beads was 3.9 times higher than that adsorbed onto MCC-chitosan beads, and the catalytic activity of lipase was 1.9 times higher on the BC-chitosan beads. The lipase showed the highest thermal and operational stability when covalently cross-linked on BC-chitosan hydrogel beads. The half-life time of the lipase cross-linked on BC-chitosan bead at 60°C was 22.7 times higher than that of free lipase. Owing to their inherent biocompatibility and biodegradability, the BC-chitosan composite hydrogel beads described here could be used to immobilize proteins for various biomedical, environmental, and biocatalytic applications.

Abstract:Delirium is a common neuropsychiatric condition that affects 15% to 70% of elderly medical and surgical patients. It tends to be a transient disorder, although long-term complications are not uncommon. Medical comorbidity is the rule, and predisposing, as well as precipitating, factors are important to consider in its management. Major risk factors for delirium include advanced age, cognitive impairment, and chronic medical illness. Delirium is associated with several adverse outcomes including mortality, increased length of hospital stay, increased risk of dementia, and high rates of institutional placement. Delirium is distressing for patients, families, and staff. Nonpharmacologic-integrated intervention programs may improve outcome and may be incorporated into the overall medical management.

Abstract:Lipase from Thermomyces lanuginosus (TLL) has been immobilized on a methacrylate macroporous resin coated with octadecyl groups (Purolite Lifetech®® ECR8806F). This immobilization protocol gave a biocatalyst with significantly higher stability than that obtained using octyl agarose. To further improve the biocatalyst features, we tried to covalently immobilize the enzyme using this support. For this purpose, the support was activated with divinyl sulfone. The results showed that at least 1/3 of the immobilized enzyme molecules were not covalently immobilized. To solve the problem, we produced an aminated support and then activated it with divinyl sulfone. This permitted the full covalent immobilization of the previously immobilized TLL. The use of different blocking agents as the reaction endpoint (using ethylenediamine, Asp, Gly, and Cys) greatly altered the biocatalyst functional features (activity, specificity, or stability). For example, the blocking with ethylenediamine increased the ratio of the activity versus R- and S-methyl mandelate by a three-fold factor. The blocking with Cys produced the most stable biocatalyst, maintaining close to 90% of the activity under conditions where the just adsorbed enzyme maintained less than 55%. That way, this strategy to modify the support has permitted obtaining an enzyme interfacially activated versus the octadecyl layer and, later, covalently immobilized by reaction with the vinyl sulfone groups.

[Rifampicin in the treatment of Staphylococcus arthritis].

Abstract:Rifampicin is used for treating 20 Staphylococcus septic arthritis, in combination with another antibiotic. The former is chosen for its striking bone diffusion. The average duration of the whole antibiotic treatment is 3 months. Rifampicin is usually prescribed, in daily doses of 1 200 mg for 60 days, after one month of parenteral antibiotherapy. The bactericidal activity is controlled regularly by measuring serum inhibitory titre. For 2 patients, the administration of rifampicin is suspended after 12 and 25 days respectively, due to digestive disorders. Results are evaluated at the end of a period averaging 18 months. Articular infection has been cured in all patients. Taking these results into account, we are able to emphasize that the inclusion of rifampicin in the antibiotic scheme is of great value in treating bone and articular infections.

Abstract:Immobilized lipases are biotechnological spotlights as tools to improve and enhance enzyme applicability. Here, lipase from Rhizomucor miehei (RML) was immobilized onto core/shell supports, named polymethylmethacrylate (PMMA/PMMA); polymethylmethacrylate copolymerized with divinylbenzene (PMMA- co -DVB/PMMA- co -DVB); and polystyrene copolymerized with divinylbenzene (PS- co -DVB/PS- co -DVB), and on a commercial non-core/shell porous support (polypropylene - Accurel MP 1000). Different enzyme loadings were evaluated (400 U.g -1 , 500 U.g -1 , 600 U.g -1 , 4000 U.g -1 , and 6000 U.g -1 ), and the immobilized biocatalysts were used in hydrolysis (using p -nitrophenyl laurate ( p -NPL) and p -nitrophenyl butyrate ( p -NPB) as substrates) and in esterification reactions (using oleic acid and ethanol as substrates). The influence of pH, temperature, and buffer concentration in the hydrolysis of p -NPL was also investigated. The immobilized biocatalysts were compared to free RML and/or to commercially immobilized RML (RM-IM) and/or to Novozym 435. RML interacted differently with each support, being RML-PMMA- co -DVB/PMMA- co -DVB and RML-PMMA/PMMA the biocatalysts with the highest enzymatic activities (hydrolytic activity: >400 U.g -1 , using p -NPL; esterification activity: 25,532 U.g -1 ). Immobilization improved the thermal and pH tolerance and buffer concentration tolerance of RML. Both free RML and the immobilized biocatalysts exhibited their maximum activity at pH 7, with the exception of RML-Accurel MP 1000, which displayed optimal performance at pH 9.0. In addition, RML-PS- co -DVB/PS- co -DVB demonstrated increased activity at 70 °C. Notably, RML-PMMA/PMMA and RML-PMMA- co -DVB/PMMA- co -DVB showcased a maximum activity of 120 U/g, and this was achieved at different temperatures, 30°C and 50°C, respectively. This diverse library of immobilized biocatalysts shows promising potential for application in various industrial processes.

Immobilization of Lipases on Heterofunctional Octyl-Glyoxyl Agarose Supports: Improved Stability and Prevention of the Enzyme Desorption

Abstract:Lipases are among the most widely used enzymes in industry. Here, a novel method is described to rationally design the support matrix to retain the enzyme on the support matrix without leaching and also activate the enzyme for full activity retention. Lipases are interesting biocatalysts because they show the so-called interfacial activation, a mechanism of action that has been used to immobilize lipases on hydrophobic supports such as octyl-agarose. Thus, adsorption of lipases on hydrophobic surfaces is very useful for one step purification, immobilization, hyperactivation, and stabilization of most lipases. However, lipase molecules may be released from the support under certain conditions (high temperature, organic solvents), as there are no covalent links between the enzyme and the support matrix. A heterofunctional support has been proposed in this study to overcome this problem, such as the heterofunctional glyoxyl-octyl agarose beads. It couples the numerous advantages of the octyl-agarose support to covalent immobilization and creates the possibility of using the biocatalyst under any experimental conditions without risk of enzyme desorption and leaching. This modified support may be easily prepared from the commercially available octyl-agarose. Preparation of this useful support and enzyme immobilization on it via covalent linking is described here. The conditions are described to increase the possibility of achieving at least one covalent attachment between each enzyme molecule and the support matrix.
Audit of major gastrointestinal surgery in patients aged 80 years or over.

Abstract:The value of preoperative assessment in relation to outcome and long-term survival was studied in 171 octogenarians undergoing major gastrointestinal surgery at Dumfries and Galloway Royal Infirmary between 1985 and 1990. The 30-day mortality rates for elective and emergency operations were 11% and 25% respectively. The American Society of Anesthesiologists (ASA) physical status scale was found to be a useful predictor of morbidity and mortality. ASA class 2 patients had a postoperative complication rate of 36% and a mortality rate of 13% while the figures for ASA class 4 patients were 85% and 54% respectively. Cox proportional hazards survival analysis using the variables age group, sex, ASA class, and type of surgery showed that the only statistically significant differences were between ASA classes. The relative 5-year survival of 63% indicates that these patients compare favourably with a population of the same age and sex not undergoing an operation. These data suggest that in the absence of severe concomitant medical disease, gastrointestinal surgery can be carried out reasonably safely in the elderly.

Use of hydroxyapatite as a support in the immobilization of Thermomyces lanuginosus lipase for application in the production of biodiesel using a by-product as lipid raw material

Abstract:The use of new materials in the field of biofuel production has been represented as a step in the development of remarkable catalysts. The use of lipases in the production of biodiesel is often seen as a cost-limiting step, as the operating expenses in recovering such catalysts can lead to unfeasible market expectations. In this study, hydroxyapatite (HAp) particles were evaluated as a support to immobilize commercial lipase, following application in ethyl ester synthesis. First, hydroxyapatite was synthesized through the co-precipitation method at constant pH and selected as a support to be used in enzyme immobilization. The characterization of the biocatalyst support materials produced was carried out using DRX, BET, FTIR, TGA, and SEM analysis. The lipase from Thermomyces lanuginosus was then immobilized in the matrices, and, subsequently, there was transesterification of the vegetable oil deodorization distillate (VODD). The biodiesel samples generated showed that they were within commercial standards, achieving ester conversion greater than 96.5%. Other properties such as density (0.87 g.cm −3 ) and viscosity (4.36 mm 2 .s −1 ) meet the specifications required by ASTM to be used as a biofuel. In the experiment planning technique, the results revealed an experimental trend and a defined behavior: a higher lipase loading in the immobilization and the use of temperatures in the range of 40–50 °C favor high conversions of ethyl esters. Thus, this confirms that the enzymatic chemical catalyst was able to form the main fatty acid esters even using a residual lipid raw material.

Abstract:The bactericidal capacity of neutrophils from healthy adults, male children with chronic granulomatous disease, and obligate heterozygote mothers for type Ic group B streptococci was assessed, using a fluorochrome microassay. Neutrophils from patients with chronic granulomatous disease had impaired bactericidal capacity for group B streptococci when compared with adults (P less than 0.01). Carriers demonstrated intermediate killing capacity.
