Abstract:The black soldier fly (BSF) Hermetia illucens has a strong tolerance to cadmium stress. This helps to use BSF in entomoremediation of heavy metal pollution. Rich metallothionein (MT) proteins were thought to be important for some insects to endure the toxicity of heavy metal. We identified and characterized three MTs genes in BSF (BSFMTs), including BSFMT1, BSFMT2A, and BSFMT2B. Molecular modeling was used to predict metal binding sites. Phylogenetic analysis was used to identify gene families. Overexpression of the recombinant black soldier fly metallothioneins was found to confer Cd tolerance in Escherichia coli. Finally, functions of BSFMTs in BSF were explored through RNA interference (RNAi). RNAi results of BSFMT2B showed that the larval fresh weight decreased significantly, and the larvae mortality increased significantly. This study suggests that BSFMTs have important properties in Cd detoxification and tolerance in BSF. Further characterization analyses of physiological function about metallothioneins are necessary in BSF and other insects.

Abstract:The metallothioneins (MT) in Boettcherisca peregrina larvae were induced and produced after their feeding on the dietary containing 800 μg cadmium chlorides per gram diet for 48 hours. Two forms of MTs (MT-Ⅰ and MT-Ⅱ) were isolated and purified from the supernatant of the whole body homogenates from the larvae after exposure to cadmium chlorides, using gel chromatography on Sephadex G-50, ion-exchange chromatography on UNO TM Q1, and desalination on Bio-Gel P-6. Molecular weight for both MT-Ⅰ and MT-Ⅱ was 9 kD. Both MT-Ⅰ and MT-Ⅱ contained 7 cadmium atoms and 20 sulfhydryl groups per molecule, and showed the spectral characteristic of cadmium-thionein, i.e. a broad shoulder at 254 nm. As to amino acid composition, the content of cysteine residues was the highest, accounting for 36.6% and 31.8% of all amino acids for MT-Ⅰ and MT-Ⅱ respectively; however, the contents of aromatic amino acid and histidine were very low, just sharing 1%-2% of all amino acids.
Influences of chromium and cadmium on the development of black soldier fly larvae

Abstract:The black soldier fly Hermetia illucens is a good candidate for waste management. The harvested insects are rich in protein and have the potential to be used in animal feed. However, people are wary about heavy metals in waste. Therefore, it is necessary to understand how the uptake of heavy metals could affect H. illucens and where and to what extent metals are accumulated by the black soldier fly. Based on these considerations, developmental parameters were investigated in the different life stages of H. illucens fed an increasing concentration gradient of cadmium (Cd) and chromium (Cr); additionally, Cd and Cr distribution in the body parts of H. illucens at the different life stages was monitored. We found that Cd and Cr have no effects on larvae survival and eclosion rate, but they do have effects on larvae duration and pupation rate. Both Cd and Cr were transferred into larvae, prepupae, and pupae. While the concentrations of Cd in larvae and prepupae were much higher than that in their diets, the opposite case was observed with Cr. The concentrations of Cd and Cr in H. illucens decreased in later development stages. In individual larva and prepupa, Cd and Cr were mainly included in the body and not in the integument. In the pupa, the puparium contained higher Cd and Cr concentrations than the pupa body. The distribution of Cd and Cr in the different life stages and body parts may present a potential strategy for how H. illucens tolerate and remove heavy metal stress.
Effects of heavy metals on the bioaccumulation, excretion and gut microbiome of black soldier fly larvae (Hermetia illucens)

Abstract:The black soldier fly larvae (BSFL) have become a promising candidate for waste disposal and are an ideal feed source for animal nutrition. The uptake of heavy metals could influence the growth of BSFL, but the effects of heavy metal pressures on the gut microbiota of BSFL are largely uncharacterized. Here, we examine the influences of Cu and Cd on the growth and gut microbiota of BSFL as well as the distribution of accumulated heavy metals in the larvae and their feces. Exposure to Cu (from 100 to 800 mg/kg) and Cd (from 10 to 80 mg/kg) did not significantly inhibit the weight gain of BSFL. With elevated exposure doses, the contents of both Cu and Cd accumulated in the bodies and feces of BSFL were remarkably increased. In the BSFL feces, Cu mainly existed as residues, while Cd mainly existed as either water-soluble states (in the low-exposure groups) or residues (in the high-exposure groups). Cd was more readily enriched (47.1%-91.3%) than Cu (<30%) in vivo. More importantly, exposure to Cu and Cd remarkably altered the gut microbiota of BSFL, particularly in the phyla Proteobacteria, Firmicutes and Bacteroidetes. High exposure to the metals (i.e., Cu-800 and Cd-80 groups) substantially decreased the abundances of most of the dominant families, but significantly stimulated the enrichment of Brucellaceae, Enterobacteriaceae, Alcaligenaceae, Campylobacteraceae, and Enterococcaceae. Moreover, the bacterial diversity in the BSFL gut was significantly reduced following high exposure to the metals. These results may fill a gap in our knowledge of the effects of heavy metals on the intestinal microbiome of BSFL.

Abstract:The risk posed by heavy metals makes it difficult to dispose of sediment contaminants from dredging lakes in China. Black soldier fly ( Hermetia illucens ) can convert organic waste, such as restaurant waste and lake sediment, to high-value-added protein feed and fertilizer. Experimental groups were formed in this study to explore the conversion of heavy metals present in the mixture of restaurant waste and lake sediment by black soldier fly larvae (BSFL). The results demonstrated that BSFL could survive in pure sediment with an 84.76% survival rate. Relative to the substrate, BSFL could accumulate 70-90% zinc (Zn), chromium (Cr), copper (Cu), and 20-40% cadmium (Cd) and lead (Pb). The experimental group 2:3, with 40% lake sediment and 60% restaurant waste, was the best group after conversion for 15 days, which showed a 95.24% survival rate of BFSL, 82.20 mg average weight of BFSL, 8.92 mm average length of BFSL, with varying content of heavy metals such as Cu (43.22 mg/kg), Zn (193.31 mg/kg), Cd (1.58 mg/kg), Cr (25.30 mg/kg) Cr, and Pb (38.59 mg/kg) in BSFL. Furthermore, the conversion residue conforms to the relevant standards of organic fertilizer in China and can be used as organic fertilizer. Overall, the present study shows that black soldier flies can improve the resource utilization of lake sediment, especially by reducing the effect of heavy metals.

Abstract:Abstract Background The black soldier fly ( Hermetia illucens ) has significant economic potential. The larvae can be used in financially viable waste management systems, as they are voracious feeders able to efficiently convert low-quality waste into valuable biomass. However, most studies on H. illucens in recent decades have focused on optimizing their breeding and bioconversion conditions, while information on their biology is limited. Methods About 200 fifth instar well-fed larvae were sacrificed in this work. The liquid chromatography-tandem mass spectrometry and scanning electron microscopy were employed in this study to perform a proteomic and ultrastructural analysis of the peritrophic matrix (PM) of H. illucens larvae. Results A total of 565 proteins were identified in the PM samples of H. illucen , of which 177 proteins were predicted to contain signal peptides, bioinformatics analysis and manual curation determined 88 proteins may be associated with the PM, with functions in digestion, immunity, PM modulation, and others. The ultrastructure of the H. illucens larval PM observed by scanning electron microscopy shows a unique diamond-shaped chitin grid texture. Conclusions It is the first and most comprehensive proteomics research about the PM of H. illucens larvae to date. All the proteins identified in this work has been discussed in details, except several unnamed or uncharacterized proteins, which should not be ignored and need further study. A comparison of the ultrastructure between H. illucens larval PM and those of other insects as observed by SEM indicates that the PM displays diverse textures on an ultra-micro scale and we suscept a unique diamond-shaped chitin grid texture may help H. illucens larval to hold more food. This work deepens our understanding of the molecular architecture and ultrastructure of the H. illucens larval PM.

Abstract:Metallothioneins, metal-inducible proteins, are being characterized from different organisms and shown as potential biomarkers of exposure to pollution by certain heavy metals. Here we report the identification of a new metallothionein cDNA (433bp) from the shrimp Macrobrachium rosenbergii, putatively encoding a 61 residue polypeptide. Tissue specific analysis indicated that Mar-MT-I (M. rosenbergii Metallothionein Gene-1) is expressed with the highest levels in the hepatopancreas and lowest in the thoracic ganglia, and none in the gills or muscles. In addition, our data showed that Mar-MT-I is differentially regulated in the hepatopancreas by certain heavy metals and thermal stress: Cd and Cu produce somewhat similar expression profile patterns, Zn has a reductional effect and thermal stress alone entirely stops its expression. These results show that Mar-MT-I mRNA levels can potentially be used as biomarkers for Cd, Cu or Zn pollution individually. However, in the case of combined metal treatment, different combinations of these metals have quite different effect on Mar-MT-I expression. Therefore, factors of such differential behaviors should be kept as a priority for further biomonitoring studies.
Proteomic Study Reveals Major Pathways Regulating the Development of Black Soldier Fly

Abstract:Nowadays, biodegrading organic waste, as a solution to confront environmental challenges, has attracted wide attention. A dipteran insect, black soldier fly (BSF), exhibits outstanding capability to convert organic waste into proteins and lipid resources, and thus, much interest has been shown in it. However, information of fundamental biology of BSF is still limited besides its recycling efficiency. In this work, we present a complete proteomic database of BSF at all instars (before prepupa). We further formulated the pathways corresponding to BSF development and built a relationship with the current genetic database. To achieve this, we investigated the proteomics of BSF during different periods. We identified 5036 proteins, and among them, 3905 proteins were annotated in the protein function database. illustrated three pathways related to major physiological processes including the insulin signaling pathway for feeding and growth, fatty acid biosynthesis pathway for fatty acid using, and toll/immune deficiency pathway for immune behavior. The proteins in these three pathways were matched with a published genetic database, and this reference library could be used for future BSF genetic engineering. In conclusion, this work provided a comprehensive protein library of BSF and expands the basic knowledge of BSF for future research.

Abstract:A critique of HC(76)38 In July 1976 the Department of Health and Social Security (DHSS) published HC(76)38 entitled 'Pollution in operating departments, etc. by waste anaesthetic gases'. It purports to inform staff as to the current evidence of the hazards of pollution and instructs hospitals to reduce the level of pollution. It must be a matter of regret that the document does not warn staff of all the current evidence of the hazards or potential hazards. There is now ample data, based not only on retrospective studies but also ongoing measurements and experiments that there are quite a number of potential hazards resulting from pollution. These are abortion, teratogenicity,14 p r e m a t ~ r i t y , ~ , ~ cancer not only to ~ t a f f , 4 * ~ l ~ but also to children of staff," biochemical changes in staff suggestive of organ dysfunction,'* and possible loss of intellectual performance or even suicide. 2-14 Experimentally it has been shown that anaesthetics interfere with cell division,ls,l6 and this effect is dose related.17 The teratogenic studies have used anaesthetics in the clinical dose range and conse quently the yield of congenital abnormalities has been high, whereas the evidence from the surveys has shown that the congenital abnormality, abortion and possibly the cancer hazard are all between x 2 and x 3 that expected by chance. Cancer, particularly reticulo-endothelial cancer, leukaemia and cancer of the breast, has been reported. The only published prospective study has failed to justify 'the withdrawal of the previous claim' that there is an excess cancer incidence among those occupationally exposed to the anaesthetic gases. However, it is the observation of four primary cancersl0 arising in three children from a total of 695 children born to female anaesthetists that is particularly worrying. Cancer in children is a very rare phenomenon and the expected number should, at the very most, have been one. A number of inhalational anaesthetics are Mown carcinogen^.^.^ It is a major weakness of HC(76)38 that no guidance is given so that doctors responsible for the health of staff, or who are asked for advice at least know what to look for or what counsel to give. The only hazard that the document accepts is that of abortion. Surely it would have been more responsible for the Department at least to alert staff to the possibility that these other identified hazards could exist, even if subsequent investigation should later disprove them, rather than to allow staff to remain in ignorance only perhaps later to suffer unnecessarily. Not everyone accepts that pollution by waste anaesthetic gases creates a health hazard but 'the onus of proof lies in demonstrating its innocence'.lp Strikingly, evidence of innocence has been lacking. Criticisms may be made that much of the human evidence is based on retrospective studies-as many of the survey authors acknowledge. As far as man is concerned this is the best evidence we have or are likely to have and one normally makes a judgement of the evidence available. To undertake a prospective study and perhaps deliberately risk inflicting ill health on members of staff without any compensatory benefit to justify the risk would seem somewhat unethical. In this connection the earliest reports of smoking and lung cancer were based on retrospective surveys and who would doubt the validity of that association. It is apparent that a rq jor consideration in the preparation of HC(76)38 has been to avoid creating alarm among staff. This paternalistic approach would imply that staff are not capable of making their own judgement and ignores the fact that staff have ready access to experts who can apprise them in detail of the relative strength of the various parts of the evidence. Senior anaesthetists given a bibliography would know how to find the data and are presumably capable of evaluating that data and so giving an informed opinion of this if asked. Offering staff the evidence does not necessarily imply that all the evidence is indisputable proven fact but at least would allow staff to decide for themselves whether they want to wait for uncontrovertible proof before taking action to protect themselves. Other Governments appear to take the pollution hazard more seriously. In East Germany staff are permitted to work only 25 hours per week in the operating departments-even when pollution control system are installed. They have a scheme of maximum permitted exposure loads and staff receive financial compensation. In Russiathere has also been a suggestion of financial compensation.20

Abstract:Binding of cadmium (Cd) to metallothionein (MT) and non-MT proteins with low contents of cysteine has been observed in terrestrial arthropods. We recently isolated a Cd-binding protein with no cysteine that was induced in Cd-exposed larvae of the beetle Tenebrio molitor. In this study we have examined the molecular distribution of Cd within extracts of different tissues and compartments of Cd-exposed T. molitor larvae. A Cd-peak consistent with the low cysteine Cd-binding protein was induced within the gut content where it could be detected after 4–8 days of exposure. Examination of gut wall tissue revealed no increase in Cd-binding capacity, indicating that no accumulation of MTs was taking place in this tissue. Incorporation of Cd in the gut wall tissue stabilized after 8 days of Cd-exposure at a rather low level compared to the other organs. There was a statistical trend towards Cd being incorporated in the gut content in a manner that was disproportionally high compared to the amount of Cd in the gut wall tissue. The possible role of the low cysteine Cd-binding protein in reducing the uptake of Cd in the tissues is discussed.
Biogas Digestate and Sewage Sludge as Suitable Feeds for Black Soldier Fly (Hermetia illucens) Larvae

Abstract:Hermetia illucens larvae can use organic wastes as a substrate, which makes them an interesting potential feed. However, waste may contain heavy metals, which are limited in feed. Here, we investigated the ability of H. illucens to grow on organic wastes and measured their heavy metal bioaccumulation. The larvae were fed with food waste, biogas digestates, and sewage sludge. When the first adult fly was visible, the tests were stopped and the larvae immediately processed. The samples (wastes before use, larvae after feeding) were analysed for mineral nutrient and heavy metal content using AAS and ICP-OES, respectively. The results show that the weight of the larvae fed with food waste increased sevenfold, which was broadly in line with expectations. Those fed with sewage sludge and digestate from biogas station increased threefold. While the larvae fed with sewage sludge exceeded the limits for heavy metals, particularly Cd and Pb, in feedstock, those fed with biogas digestate and food waste did not. These findings add to the literature showing the suitability of different wastes as H. illucens feed, and the importance of excluding waste contaminated with heavy metals from larvae intended for use as animal feed, or else diverting these larvae to non-feed uses.

Abstract:BACKGROUND The black soldier fly (Hermetia illucens) is one of the most promising insect species for use in animal feed. However, studies investigating feed and food safety aspects of using black soldier fly as feed are scarce. In this study, we fed black soldier fly larvae feeding media enriched with seaweed, which contains naturally high concentrations of heavy metals and arsenic. The aim of this study was to investigate the potential transfer of such undesirable substances from the feeding media to the larvae. RESULTS The larvae accumulated cadmium, lead, mercury and arsenic. Concentrations of these elements in the larvae increased when more seaweed was added to the feeding media. The highest retention was seen for cadmium (up to 93%) and the lowest for total arsenic (up to 22%). When seaweed inclusion exceeded 20% in the media, this resulted in larval concentrations of cadmium and total arsenic above the current European Union maximum levels for these elements in complete feed. CONCLUSION Our results confirm that insect larvae can accumulate heavy metals and arsenic when present in the feeding media. A broader understanding of the occurrence of these undesirable substances in processed larvae products is needed to assess feed and food safety. © 2017 Society of Chemical Industry.

Reducing the mass and decreasing the bioavailability of heavy mental from organic wastes treated by black soldier fly larvae

Abstract:Black soldier fly larvae (BSFL), Hermetia illucens L., are widely used to reduce the mass of various wastes. However, the potential metal tolerance mechanisms during periods of waste bioconversion by BSFL remain largely unknown. To further reveal the mechanisms, BSFL were used to treat the agricultural organic wastes, including pig manure (PM), cow manure (COM), spent mushroom substrate (SMS), and wet distiller grains (WDG). After these individual and combined waste(s) were treated by BSFL, we investigated the waste reduction rates and evaluated the responses of BSFL gut microbes to heavy metals of agricultural organic wastes. Additionally, the colloidal particles of residual wastes were characterized by combing energy dispersive X-ray (EDX) spectroscopy, Size potential, Zeta potential, and excitation-emission matrix (EEM) spectroscopy. Results indicated that the waste reduction rates were up to 74% in COM+WDG and 69% in WDG, most of heavy metals (e.g., Zn and Co) from organic wastes were not accumulated in the bodies of mature larvae after treatment. Further, results obtained from the prediction of gene function on the basis of 16 S rRNA data revealed that the presence of multi-resistance genes in the gut of BSFL can help the larvae resist Zn and/or Co stress. In addition, the drug sensitivity test implied that BSFL5_L and BSFL6_L from BSFL gut bacterial strains have multi-resistance to Co and Zn. Additionally, EDX results revealed that the colloidal particles in five waste residues after BSFL treatment are mainly consisted of Fe, Ca and Si, which can capture heavy metals (e.g., Cu, Mn). Results from EEM spectroscopy and PARAFAC showed that tryptophan-like and humic-like accumulatively account for 56%- 68% of all components. Importantly, these two components could strongly bind the metal elements and form colloidal particles with high stability, and therefore reduce the heavy metal pollution of agricultural organic wastes. Our findings offered an environment-friendly method to treat agricultural organic wastes, which would be far-reaching influence to our environment.

Abstract:This study investigated the use of Hermetia illucens (black soldier fly - BSF) larvae as a new species for use in entomoremediation. The H. illucens larvae effectively reduced the dry mass of polluted corn leaves by an average of 49% after 36days for both Cd and Zn (50mg·kg-1), which is a better result than that, which can be obtained by one of the standard proposed pretreatments for biomass polluted after phytoextraction: composting. The presence of heavy metals did not significantly affect the dry mass utilization efficiency. Based on this, we proposed the use of H. illucens as a new post-harvest management method of phytoextraction-polluted biomass. Cadmium mostly accumulates in the puparium, while Zn accumulates in the adults. The high Cd content in the puparium further creates possibility of its application in the metal recovery process. It is also proposed that the general definition of entomoremediation be expanded.

Abstract:Aquatic Dipterans are a diverse assemblage of insects that make up important components of aquatic ecosystems. Because of their importance in these systems, aquatic dipterans are ideal candidates to monitor water heavy metal pollution. This review summarizes the species diversity of these insects and their potential use for monitoring heavy metal pollution. We describe the influence of heavy metal pollution on the morphological and internal structural changes of aquatic Dipteral insects. Finally, we highlight research into the biochemical and molecular markers that provide sensitive information necessary to detect the very early stages of heavy metal pollution.

Abstract:The increased human activities and the worldwide population growth are constantly increasing the production of solid wastes. Over the years, waste management has thus become a prominent issue for several companies and municipalities, and several engineering techniques have been developed over the years in order to convert wastes into other solid materials or fuels. Yet, several techniques are important contributors to environmental pollution, and biological-based solutions have thus become progressively very popular. In particular, insect-based conversion of organic wastes represent eco-friendly tools, and the growth and development of insect species such as the black soldier fly have been tested and improved for a large diversity of organic wastes. However, organic wastes, including food wastes, may contain several pollutants such as heavy metals and catechol which could affect the bioconversion efficiency by incurring physiological costs that would be undetectable at the organismal level, i.e. have null to little effects on the life cycle of Hermetia illucens. In this context, assessments of antioxidant capacities can provide a rapid and low-cost evaluation of the capability of insects to handle exposure to heavy metals and catechol. Here, we aimed at measuring the physiological responses of the black soldier fly H. illucens grown on food wastes (kitchen, fruit or vegetable wastes) contaminated by cadmium, iron, lead or catechol. Biomarkers of oxidative stress (concentrations of hydrogen peroxide and protein carbonyls), non-enzymatic total antioxidant capacity (ascorbic acid amounts) and activity of enzymatic antioxidants (activities of superoxide dismutase and polyphenoloxidase) were measured from the gut of the larvae. We found no evidence of deleterious impacts of food waste contamination by catechol or heavy metals on H. illucens. In most experimental treatments, the array of physiological endpoints we measured for evaluating the degree of oxidative stress experienced by the larvae remained similar to controls. Possible physiological effects were reported for cadmium and catechol only, which tended to increase the oxidation of proteins and hydrogen peroxide in the larvae. Finally, our results suggested that the nature of the food waste could equally affect the physiological responses of the insect.
Growth performance and dynamic copper accumulation in tissues of black soldier fly (Hermetia illucens) larvae under copper exposure

Abstract:Abstract To figure out the copper metabolism features in vivo and evaluate the potential risk of copper residue in tissues of black soldier fly larvae (BSFL) and their by-products, the effects of oral copper exposure of 0, 50, 500 and 1000 mg/kg (Control, Cu50, Cu500 and Cu1000) on growth performance and dynamic copper accumulation in the fat body, cuticle of BSFL and the rearing residue were investigated in a 12-days rearing trial. The results showed that, the body weight and length, and body protein content of BSFL were improved by exposure to 50 mg/kg copper, but reduced by exposure to 500 and 1000 mg/kg copper, as well as that of the pre-pupa rate. Under the copper exposure of 500 and 1000 mg/kg, the copper concentrations in the larval fat body were high at the early developmental stage, but sharply reduced to be 8.1 ± 0.3 mg/kg within 12 days with no significant differences among groups. Similarly, the copper concentrations in the cuticle also decreased rapidly to be 46.3 ± 2.9 and 91.3 ± 9.4 mg/kg in the Cu500 and Cu1000 groups within 12 days, but they were significantly higher than those of the control (8.1 ± 0.3 mg/kg) and Cu50 (10.1 ± 1.1 mg/kg) groups. The linear regression analysis between copper exposure doses and copper concentrations in the rearing residues indicated that the latter are dose- and time-dependent. Overall, the growth performance and body protein content of BSFL were improved by low copper exposure, but reduced by high copper exposure. The oral copper could be excluded from larval fat body and cuticle rapidly, and be largely accumulated in the rearing residues. Therefore, little risk of copper residue in the main tissues of post larvae or pre-pupae of black soldier fly (BSF), but higher risk of it in the rearing residues could be anticipated. Our results provide valuable information for safety assessment of copper residues in BSF products and by-products.

Enrichment and speciation changes of Cu and Cd in black soldier fly (Hermetia illucens) larval compost and their effects on larval growth performance

Abstract:Municipal sludge (MS), rainwater sludge (RS), and kitchen waste (KW) were used as nutritional supplements for black soldier fly larvae (BSFL). Cd (52.3 %) was more easily assimilated in the BSFL body than Cu (34.8 %). After biotransformation in BSFL, the weak acid-soluble fraction (F1) of Cu and Cd increased by an average of 29.0 % and 42.7 %, respectively, whereas the reducible fraction (F2) of Cu and Cd decreased by an average of 13.8 % and 56.4 %, respectively, in the BSFL sand (BSFL feces and waste residues). A significant correlation (P < 0.01) was found between pH and the speciation of Cu and Cd. The abundance of Bacteroides had a positive correlation (P < 0.05) with the F1 of Cu, an extremely significant negative correlation (P < 0.001) with the F2 of Cd, and an extremely significant positive correlation with the F1 of Cd (P < 0.001). In addition, Cu and Cd exposures significantly (P < 0.01) reduced larval weight by 67.7 % and 45.3 %, respectively, pupation rate by 46.3 % and 26.5 %, respectively, and eclosion rate by 35.5 % and 33.4 %, respectively. Exposure to high concentrations of Cu and Cd also prolonged the development cycle (1-12 days) of BSFL and led to the failure of BSFL to complete their metamorphosis.

Abstract:The black soldier fly (BSF) is well known for its ability to biologically convert organic waste into insect biomass, including protein and oil, which can be utilised as animal feed. Since raw BSF products, such as BSF powder, are difficult to differentiate from other biological raw materials, therefore new analytical approaches are required. In this study, we have developed a new and fast method based on loop-mediated isothermal AMPlification (LAMP) reaction that can diagnose black soldier fly larvae and BSF byproducts with high accuracy, specificity and sensitivity. Species-specific primers for BSF were designed based on targeting the mitochondrial cytochrome C oxidase I (COI) gene. The assay was able to detect as low as 820 fg/L of BSF DNA in 60 min at 65 °C, which was a hundredfold higher than the detection limit of classical polymerase chain reaction and did not show cross-reactivity. In conclusion, the LAMP assay demonstrated excellent sensitivity and specificity to detect BSF and BSF byproducts, with a sampling-to-result identification time of 60 min.

Abstract:A new branch of the insect-based food and feed industry is intensively developing all over the world, the best proof of which is the recent change in legislation at the European level allowing the use of insect protein in the production of feed for aquaculture animals. Previous publications have proven that some heavy metals can be bioaccumulated in fly H. illucens (black soldier fly), even when the insect is raised on optimal feed with an acceptable heavy metal content. The purpose of this study was to determine the bioaccumulation potential of H. illucens in relation to micro- and macroelements, toxic elements and for the first time, selected non-essential elements from optimal feed. Our results showed that bioaccumulation of Ba, Bi, Cu, Fe, Hg, Mg, Mo, Se and Zn occurred in all stages of insect development and in puparia, while bioaccumulation of Al, As, Co, K, Pb and Si was not found. The highest bioaccumulation factors were obtained for Ca and Mn in puparia - 38 and 21 respectively. In addition, Ca, Cd, Ga, Mn, P and S were bioaccumulated only in some developmental stages of the insect. The results are discussed in the context of the safety of feed and food production from H. illucens.
