Abstract:The results presented here show that the tumor incidence in T-cell depressed SHR after injection of MCA was higher than that in WKA rats with normal T-cell function. The depression of T-cell functions in SHR was almost completely restored by allogeneic thymus grafts or injection of NSP. At the same time, the generation of Killer T-cells was induced, and the activity of NK cells against K-562 cells was enhanced. The tumor incidence was suppressed and average latency periods were also prolonged in SHR grafted with WKA thymuses. The administration of NSP was effective in prolonging latency periods for the development of tumors. From these results, we suggest that the SHR is a suitable animal model for the investigation of the role of cell-mediated immunity in carcinogenesis.
Immunologic Suppression of Carcinogenesis in Spontaneously Hypertensive Rats (SHR) with T Cell Depression.

Abstract:A strain of spontaneously hypertensive rats (SHR) showed a selective depression of T cell functions brought about by aging. Conversely, this strain had a high NK cell activity as compared to other normal rat strains. This SHR strain was found to be much more sensitive to the carcinogenic activity of low doses of MCA than were WKA rats with normal T cell functions. Allogeneic thymus grafts almost completely restored the T cell functions of SHR, whereas injection of an immunopotentiator, NSP, enhanced NK cell activity and also caused a partial recovering of T cell functions. When immunologic restoration was achieved, generation of killer T cells to syngeneic SMT-5 tumor cells was induced and the cytotoxic activity of NK cells to K-562 cells was also enhanced. But the cytotoxic activity to the SMT-5 cells of NK cells and macrophages from the treated or untreated SHR was not detected. Allogeneic thymus grafts induced a significant transplantation resistance against a syngeneic SMT-5 tumor and injection of NSP enhanced only the survival days of the rats. Allogeneic thymus grafts also significantly suppressed the incidence of tumors induced by MCA, whereas the injection of NSP was not effective in the prevention of tumor development but was effective in prolongation of latency periods. These results support the hypothesis that immune surveillance mediated by T cells is an important mechanism for the control of tumor development.
Cocultivation of Umbilical Cord Blood CD34+cells with Retro-Transduced Hmscs Leads to Effective Amplification of Long-Term Culture-Initiating Cells

Abstract:AIM: To establish a novel coculture system for ex vivo expansion of umbilical cord blood(UCB) hematopoietic progenitors using thrombopoietin (TPO)/Flt-3 ligand (FL)-transduced human marrow-derived mesenchymal stem cells (tfhMSCs) as feeder.METHODS: UCB CD34(+) cells were isolated and cultured using four culture systems in serum-containing or serum-free medium. Suitable aliquots of cultured cells were used to monitor cell production, clonogenic activity, and long-term culture-initiating culture (LTC-IC) output. Finally, the severe-combined immunodeficient (SCID) mouse-repopulating cell (SRC) assay was performed to confirm ability of the cultured cells to reconstitute long-term hematopoiesis.RESULTS: There were no significant differences in the number of total nucleated cells among different culture systems in serum-containing medium during 21-d culture. However, on d 14, the outputs of CD34(+) cells, CFU-C and CFU-GEMM in tfhMSCs coculture system were significantly enhanced. LTC-IC assay demonstrated that the tfhMSCs coculture system had the most powerful activity. The severe-combined immunodeficient (SCID) mouse repopulating cell (SRC) assay confirmed extensive ability of the expanded cells to reconstitute long-term hematopoiesis. Furthermore, PCR analysis demonstrated the presence of human hematopoietic cells in the bone marrow and peripheral blood cells of NOD/SCID mice.CONCLUSION: The TPO/FL-transduced hMSCs, in combination with additive cytokines, can effectively expand hematopoietic progenitors from UCB in vitro and the tfhMSCs coculture system may be a suitable system for ex vivo manipulation of primitive progenitor cells under contact culture conditions. Abstract:Objective To assess the immune tolerance of allogeneic T cells induced by dendritic cells genetically engineered to express IκBα mutant. Methods DCs were prepared from WF rat bone marrow cells and modified by IκBαM gene wit h adenovirus vector, and then the expression of IκBα and IκBαM was detected by Western-blot. The cell-surface expression of costimulatory molecules (CD80, CD86 and CD40) was detected by flow cytometry, and the production of IL-12 in DCs culture supermatant was determined by ELISA. The ability to stimulate the pr oliferation of Lewis rat T cells was analyzed by mixed leukocyte reactions (MLR) . The antigen-specific T cell hyporesponsiveness was tested by secondary MLR. Results IκBαM suppressed the cell-surface expression of costimulatory molecules and i nhibited the production of IL-12. IκBαM-DC showed reduced ability to stimula te T cells proliferation, and potential to induce antigen-specific T cell hypo responsiveness. Conclusion Immune tolerance of T cells can be induced by dendritic cells genetically engine ered to express IκBα mutant.
Mechanism of Enhancement of the CTL Activity in Mice Co-Expressing CD80, CD86 and CD137Lgenes

Abstract:OBJECTIVE:To study the mechanism of enhancement of the CTL activity in mice co-expressing of CD80, CD86 and CD137L genes.METHODS:The mice were randomly divided into five groups, named A, B, C, D and E. The group A and B were control groups (CG). H22-BAL B/c HCC mouse model was established by subcutaneous injection with hepatocellular carcinoma cells of cell line H22-Wt (group A), H22-neo (group B), H22-CD80/CD86(+) (group C), H22-CD137L(+) (group D) and H22-CD80/CD86/CD137L(+) (group E), respectively. On the 14th, 35th, 56th and 84th day after the first inoculation of tumor cells, TUNEL staining and DNA ladder examination were used to detect apoptosis of splenic T lymphocytes in all groups at each post-inoculation time point. Electrophoretic mobility-shift assay (EMSA) method was used to detect the activity of nuclear factor kappaB (NF-kappaB) in splenic T lymphocytes in each group at each time point post-inoculation.RESULTS:Apoptosis was found in a great number of T lymphocytes in CG on the 14th day, much more than that in group C and E. The number of apoptotic T cells in group C had a significant difference compared with that in the group E from 14th to 84th day (P = 0.003). DNA ladder analysis showed typical positive results in group C and E. The significant apoptosis fragments were found in group C on 21st, 35th and 84th days. NF-kappaB activity of T cells in groups C and E was remarkably higher than that of groups CG and D, with higher in group D than that of CG (P = 0.002), and with no significant difference between group C and E on 14th day. The activity in group E was stable and remarkably higher than that of group C on 56th and 84th days after the first inoculation.CONCLUSION:H22-CD80/CD86/CD137L(+) induces higher NF-kappaB activity of the host T cells by synergistic action of CD28 and CD137, which may be one of the mechanisms of enhancement of the host CTL activity induced by co-expression of CD80, CD86 and CD137L genes.
[Establishment of Long-Term Culture System for Human B Cells Derived from Peripheral Blood Mononuclear Cells Activated Via Soluble CD40 Ligand].

Abstract:OBJECTIVE:To develop a method for long-term culture of human B cells from peripheral blood mononuclear cells (PBMCs) by means of human soluble CD40 ligand (sCD40L), and to investigate the proliferation and antigen-presenting function of the CD40-activated B cells.METHOD:Peripheral blood sample of 30 ml was collected from a healthy person. PBMCs were isolated and cultured in the presence of sCD40L. Flow cytometry was used to examine the proliferation, DNA cycle, and cell surface markers: CD86, CD80, major histocompatibility complex (MHC) class II, and MHC Class I of the B cells. The cells cultured for 45 days were divided into 2 groups: Group 1 incubated with the fluorescein isothiocyanate (FITC)-conjugated hepatitis-B core antigen (HBcAg-FITC) and Group 2 used as control group. Eighteen hours later cytometry and fluorescence microscopy were used to detect the peptide pulsing.RESULTS:The B cells could be cultured up to 50 days in the sCD40L culture system. The ratio of B cells in the PBMCs was 8.21% at the beginning, and increased to 70.67% by day 47, and the B cell absolute count was 6.56 x 10(5) at the beginning and increased to 8.61 x 10(6) at day 50, about 10 times higher. sCD40L promoted a strong up-regulation of cell surface markers. The expression rates of CD80, CD86, MHC-II, and MHC-I of the sCD40L-activated B cells were 83.97%, 84.73%, 99.59%, and 98.70% respectively. Flow cytometry showed that 98.10% of the B cells co-incubated with HBcAg-FITC were loaded with HBcAg. Fluorescence microscopy showed that the HBcAg was in the cytoplasm of the B cells.CONCLUSION:A human sCD40L culture system has been developed with the ability to culture human peripheral blood B cells, thus realizing the long-term proliferation and activation of human peripheral blood B cells that function as antigen-presenting cells.

Abstract:Chronic graft-versus-host disease (cGVHD) is a major complication in long-term survivors of allogeneic hematopoietic stem cell transplantation (allo-HSCT). Graft-derived T cells (TG) have been implicated in the induction of cGVHD; however, the extent of their contribution to the pathogenesis of cGVHD remains unclear. Using a mouse model of cGVHD, we demonstrate that TG predominate over hematopoietic stem cell-derived T cells generated de novo (THSC) in cGVHD-affected organs such as the liver and lung even at day 63 after allo-HSCT. Persisting TG, in particular CD8+ TG, not only displayed an exhausted or senescent phenotype but also contained a substantial proportion of cells that had the potential to proliferate and produce inflammatory cytokines. Host antigens indirectly presented by donor HSC-derived hematopoietic cells were involved in the maintenance of TG in the reconstituted host. Selective depletion of TG in the chronic phase of disease resulted in the expansion of THSC and thus neither the survival nor histopathology of cGVHD was ameliorated. On the other hand, THSC depletion caused activation of TG and resulted in a lethal TG-mediated exacerbation of GVHD. The findings presented here clarify the pathological role of long-lasting TG in cGVHD.

Differentiation of Mouse Spermatocytes to Spermatids during in vitro Culture

Abstract:Objective: To investigated the differentiation of immature mouse spermatogenic cells during in vitro culture. Methods: Testicular cells of 14-day-old BALB/c mice were dispersed, and incubated at 34℃ with 5% CO2 in TM4 conditioned medium. In control group, dispersed testicular cells were cultured in MEM medium supplemented with 1% FBS in order to keep cells alive. Cells were harvested at different culture days, and real time RT-PCR was applied to analyze the expression levels of the spermatid specific gene Tnp2. Results: Real time RT-PCR showed, Tnp2 expression increased when the age of mouse increased, which synchronized the differentiation of spermatids in vivo. During in vitro culture, Tnp2 expression inceased significantly in TM4 conditioned medium at d 5 (P<0.05), while in control group, Tnp2 expression didn’t increase all the time. Conclusion: Mouse spermatocytes differentiated to spermatids during in vitro culture in TM4 conditioned medium.
Culture and Identification of Mouse Bone Marrow-Derived Dendritic Cells and Their Capability to Induce T Lymphocyte Proliferation

Abstract:BACKGROUND: The aim of this study was to establish a culture method for mouse dendritic cells (DCs) in vitro and observe their morphology at different growth stages and their ability to induce the proliferation of T lymphocytes.MATERIAL/METHODS: Granulocyte-macrophage colony stimulating factor (GM-CSF) and interleukin-4 (IL-4) were used in combination to induce differentiation of mouse bone marrow (BM) mononucleocytes into DCs. The derived DCs were then assessed for morphology, phenotype, and function.RESULTS: The mouse BM-derived mononucleocytes had altered cell morphology 3 days after induction by GM-CSF and IL-4 and grew into colonies. Typical dendrites appeared 8 days after induction. Many mature DCs were generated, with typical dendritic morphology observed under scanning electron microscopy. Expression levels of CD11c, a specific marker of BM-derived DCs, and of co-stimulatory molecules such as CD40, CD80, CD86, and MHC-II were elevated in the mature DCs. Furthermore, the mature DCs displayed a strong potency in stimulating the proliferation of syngenic or allogenic T lymphocytes.CONCLUSIONS: Mouse BM-derived mononucleocytes cultured in vitro can produce a large number of DCs, as well as immature DCs, in high purity. The described in vitro culture method lays a foundation for further investigations of anti-tumor vaccines.

Selective expansion and engraftment of human CD16+ NK cells in NOD/SCID mice

Abstract:NK cells are large granular lymphocytes that represent a critical component of the innate immunity. Investigations of human NK cell function are largely based on in vitro assays because of the lack of suitable animal models. Here we have established conditions leading to the development of human NK cells in NOD/SCID (severe combined immunodeficiency) mice receiving grafts of cord blood mononuclear cells (CBMC), and GFP-transduced HFWT inducing NK cells (GHINK-1), which have been shown to support the selective expansion of NK cells from human PBMC and CBMC in vitro. Significant numbers of CD56dimCD16+ cytotoxic and CD56-CD16+ immature NK cells appeared in peripheral blood (PB), peritoneal cavity, spleen, bone marrow and liver of the mice. The newly generated NK cells did not express activation markers such as CD25, CD69 and NKp44, the expression of which was augmented by IL-2 in vitro. The NOD/SCID mice engrafted with human NK cells exhibited antitumor activity against K562 erythroleukemia in vitro and in vivo. Thus, we succeeded in developing a CD56dimCD16+ cytotoxic NK cell populations in NOD/SCID mice closely resembling the main NK fraction in human PB and CD56-CD16+ immature NK cells. Our model provides not only information about the development and dynamics of physiological human NK cells but also an important pre-clinical system for immunotherapeutic strategies.

Abstract:We generated an NOD/Shi-scid-IL2Rγ null (NOG) mouse deficient for the Fcer1g and Fcgr2b genes (NOG-FcγR-/- mice), in which monocytes/macrophages do not express activating (FcγRI, III, and IV) or inhibitory (FcγRIIB) Fcγ receptors. Antibody-dependent cellular cytotoxicity (ADCC) by innate immune cells was strongly reduced in this strain. Thus, while the growth of xenogeneic human tumors engrafted in conventional NOG mice was suppressed by innate cells upon specific antibody treatment, such growth inhibition was abrogated in NOG-FcγR-/- mice. Using this novel strain, we further produced NOG-FcγR-/--mice expressing human IL-15 (NOG-FcγR-/--hIL-15 Tg). The mice inherited unique features from each strain, i.e., the long-term sustenance of human natural killer (NK) cells, and the elimination of mouse innate cell-mediated ADCC. As a result, segregation of human NK cell-mediated ADCC from mouse cell-mediated ADCC was possible in the NOG-FcγR-/--hIL-15 Tg mice. Our results suggest that NOG-FcγR-/--hIL-15 Tg mice are useful for validating the in vivo function of antibody drug candidates.

Immune regulation of in vitro murine megakaryocyte development. Role of T lymphocytes and Ia antigen expression

Abstract:Mitogen-activated murine T lymphocytes or T cell hybridomas produce an activity (megakaryocyte [Mk] potentiator activity) that enhances the in vitro growth and development of Mk colonies. This activity was found in optimal concentrations (2.5%) in T cell hybridoma-conditioned medium, and was also produced by feeder layers of concanavalin A-activated T cells. A subpopulation of murine Mk progenitor cells (colony-forming units; CFU-Mk) bears the Ia antigen. Separate experiments indicated that T cell products stimulate CFU-Mk by increasing their basal levels of Ia expression as well as the frequency of cells actively synthesizing DNA. The hypothesis that the expression of this antigen was related to the cell cycle status of these progenitor cells was confirmed in studies that indicated that ablation of actively cycling cells in vivo abrogated the cytotoxic effects of anti-Ia monoclonal antibodies. The interdependence of T cell lymphokine regulation of both Ia expression and cell cycle status was also seen in in vitro experiments in which Ia+ progenitor cells were eliminated by complement-dependent cytotoxicity. The removal of Ia+ cells prevented 5-hydroxyurea-mediated inhibition of cells in S phase. We hypothesize that immune modulation of megakaryocytopoiesis occurs via soluble T cell products that augment Mk differentiation. Further, the mechanism of immune recognition/modulation may occur via Ia antigens present on the surface of these progenitor cells.
Anti-CD28 McAb Enhance Anti-tumor Activity of Tumor Specific Cytotoxic T Lymphocyte in Vitro and in Vivo

Abstract:Purpose To enhance anti tumor activity of α?CD3 McAb activated tumor specific cytotoxic T lymphocyte(CTL) by α?CD28 McAb in vitro and vivo for tumor adoptive immunotherapy. Methods Two different modles were applied to culture splenocytes from FBL?3 immunized mice:(1) activated by α?CD3 McAb for 48 h and then cultured in α?CD3 McAb plus 20 U/ml rIL?2(α?CD3+IL?2 group);(2) activated by α?CD3 McAb,α?CD28 McAb for 48 h and then cultured in α?CD3 McAb,α?CD28 McAb plus 20 U/ml rIL?2(α?CD3+α?CD28+IL?2 group).The proliferation ( 3H?TdR incorporation assay)and anti tumor activity(standard 4 h? 51 Cr release assay) of FBL?3 specific CTL were studied in both groups.On day 12,each bearing tumor mouse was transferred with 1×10 7 CTL by ip. Results 3H?TdR incorporation (cpm) in α?CD3+IL?2 group and α?CD3+α?CD28+IL?2 group were 8.36×10 4,1.31×10 5 respectively.On day 12,the anti tumor activities of both groups were 35.6% and 49.5% respectively (E/T=12.5/1).The CTL of α?CD3+IL?2 group and α?CD3+α?CD28+IL?2 group were transferred to bearing tumor mouse respectively.The lifespans of bearing tumor mouse were 16.7 d and 20.8 d respectively. Conclusions α?CD28 McAb can enhance antitumor activity of tumor specific CTL activated by α?CD3 McAb.
Selective Support of a Mouse Thymus Epithelial Cell Line (MTEC1) to the Viability and Proliferation of CD4+ CD8−, CD4− CD8−, and CD4+CD8+ Thymocytes in Vitro

Abstract:The MTEC1 cell line, established in our laboratory, is a normal epithelial cell line derived from thymus medulla of Balb/c mice and these cells constitutively produce multiple cytokines. The selection of thymic microenvironment on developing T cells was investigated in an in vitro system. Unseparated fresh thymocytes from Balb/c mice were cocultured with MTEC1 cells or/and MTEC1- SN,then,the viability, proliferation and phenotypes of cultured thymocytes were assessed. Without any exogenous stimulus, both MTEC1 cells and MTEC1–SN were able to maintain the viability of thymocytes, while only the MTEC1 cells, not the MTEC1–SN, could directly activate thymocytes to exhibit moderate proliferation, indicating that the proliferative signal is delivered through cell surface interactions of MTEC1 cells and thymocytes. Phenotype analysis on FACS of viable thymocytes after coculture revealed that MTEC1 cells preferentially activate the subsets of CD4+ CD8−, CD4+ CD8+ and CD4− CD8− thymocytes; whereas MTEC1 - SN preferentially maintained the viability of CD4+ CD8− and CD4− CD8+ thymocyte subsets. For the Con A-activated thymocytes, both MTEC1 cells and MTEC1-SN provided accessory signal (s) to significantly increase the number of viable cells and to markedly enhance the proliferation of thymocytes with virtually equal potency, phenotyped as CD4+CD8−, CD4− CD8+, and CD4− CD8− subsets. In summary, MTEC1 cells displayed selective support to the different thymocyte subsets, and the selectivity is dependent on the status of thymocytes.
T-lymphocyte Mediated Tumor Cell Destruction in Vivo Associating with a Specific Feature of Apoptosis

Abstract:The immunological responsiveness of the T-cell immunodificient NC nude mice tumor models was reconstructed by thymic transplantation of the semidominance NC mice. After immune reconstruction (IR) the tumor continued to grow until 2 to 3 weeks, then the volume of the tumor reduced gradually and disappeared at the 9th week. In both H901 and SW1116 solid tumor nodules was found by light microscopic study (IM), after IR, tumor cells gradually replaced by lymphocytes and fibroblasts, shrinked till only isolated cell groups, then totally disappeared. The whole processes like that the tumor cells were nibbled. It was found that the main tumor cell death related with a specific feature of apoptosis, which had typical dense chromatin distributed along the inner surface of the nuclear membrane by transmission electron microscopic study (TEM). The IR model could be useful for further mechanical research of immune system.

Abstract:Generation and adoptive transfusion of alloreactive cytotoxic T lymphocytes (CTLs) specific for tumor are expected to circumvent tumor tolerance. Here we describe a novel protocol to raise allo-restricted, peptide-specific CTLs by co-culture of murine splenocytes and autologous macrophage bearing an allogeneic H-2K molecule associated with its restricted peptide (peptide/allo-MHC). The extracellular domains of H-2Kd were fused with constant domains of murine IgG2a heavy chain to generate a fusion protein (peptide/H-2Kd/IgG2aFc, the dimer) consisting of divalent TCR-ligands and an IgG Fc receptor type I (FcγRI)-reactive moiety. The dimer is able to bind to macrophage (Mϕ) of H-2Kk via the interaction of the Fc part with FcγRI, and cause the H-2Kk Mϕ to be coated with the peptide/H-2Kd complex. The results show that proliferation of CD8+ cells is enhanced and that the specific-tetramer stained CD8+ cells appear more frequently by co-culture of H-2Kk splenocytes with the autologous Mϕ loaded with the dimer. Furthermore, the CD8+ T cells from the co-cultural bulk exhibit an elevated cytotoxicity against a specific target (H-2Kd-restricted, peptide-specific cytotoxicity), compared with that against the irrelevant targets. This study provides a strategy for preparation of allo-restricted, peptide-specific CTLs, which may add to our arsenal for adoptive immunotherapy to eliminate chronic virally infected or tumor cells.
Anti-ulcer and antioxidant activity of Normacid, a herbomineral formulation.

Abstract:Effect of various doses (125, 250, 500 and 1000 mg/kg, po) of Normacid was studied on gastric secretion and gastric ulcers in pylorus-ligation and on ethanol-induced gastric mucosal injury in rats. The reduction in ulcer index in both the models along with the reduction in total acidity and an increase in the pH of gastric fluid in pylorus-ligated rats proved the anti-ulcer activity of Normacid. The increase in the levels of superoxide dismutase, catalase, reduced glutathione and membrane bound enzymes like Ca2+ ATPase, Mg2+ ATPase and Na+K+ ATPase and decrease in lipid peroxidation in both the models showed the antioxidant activity of the formulation. Thus it can be concluded that the anti-ulcer activity shown by Normacid may be due to the modulation of defensive factors by improvenent in gastric cytoprotection and partly due to antioxidant property.

Abstract:The MTEC1 cell line, established in our laboratory, is a normal epithelial cell line derived from thymus medulla of Balb/c mice and these cells constitutively produce multiple cytokines. The selection of thymic microenvironment on developing T cells was investigated in an in vitro system. Unseparated fresh thymocytes from Balb/c mice were cocultured with MTEC1 cells or/and MTEC1- SN,then,the viability, proliferation and phenotypes of cultured thymocytes were assessed. Without any exogenous stimulus, both MTEC1 cells and MTEC1–SN were able to maintain the viability of thymocytes, while only the MTEC1 cells, not the MTEC1–SN, could directly activate thymocytes to exhibit moderate proliferation, indicating that the proliferative signal is delivered through cell surface interactions of MTEC1 cells and thymocytes. Phenotype analysis on FACS of viable thymocytes after coculture revealed that MTEC1 cells preferentially activate the subsets of CD4+ CD8−, CD4+ CD8+ and CD4− CD8− thymocytes; whereas MTEC1 - SN preferentially maintained the viability of CD4+ CD8− and CD4− CD8+ thymocyte subsets.
Effect of anti-CD3 monoclonal antibody on expansion and anti-tumor activity of tumor specific T cells

Abstract:Purpose: The changes on cell expansion and anti-tumor activity of mouse tumor-specific T cells activated by anti-CD3 monoclonal antibody (McAb) were investigated. The goal of this study is to find out more efficient method for tumor adoptive immunotherapy. Methods: Tumor-specific T cells from tumor-immunized mice were obtained by in vitro activation of anti-CD3 monoclonal antibody. The changes on cell expansion and anti-tumor activity as well as phenotype were studied. Four different culture modes were applied in this experiment: (1) cultured in anti-CD3 McAb alone (anti-CD3 group); (2) cultured in 20000 u/L rIL-2 alone (IL-2 group); (3) activated by anti-CD3 McAb for 48 hr and then cultured in 20000 u/L rIL-2 alone (CD3 -IL-2 group); (4) activated by anti-CD3 McAb for 48 hr and then cultured in CD3 McAb plus 20000 u/L rIL-2 (CD3 +IL-2 group). Results: The cells in CD3 -IL-2 group and CD3 +IL-2 group increased and expanded 780 folds and 610 folds respectively within 20 day. The maximum anti-tumor activities of both groups were 41.8% and 91.3% respectively on day 12. However, the continuous use of anti-CD3 McAb alone had neither proliferative effect nor anti-tumor activity. The IL-2 group maintained a low level of proliferation within 20 days and showed its maximum anti-tumor activity of 30.2% on day 12. The study of cell phenotype by FACS analysis on day 6 indicated that >98% effective cells of both the CD3 -IL-2 group and CD3 +IL-2 group were Thy1.2+ cells. On day 20 Thy1.2+ cells of CD3 +IL-2 group was still 98.8% while Thy1.2+ cell of CD3 -IL-2 group was only 44.4%. There were also differences in CD4 + and CD8 + between the 2 groups. Conclusions: The tumor-specific T cells cultured in the presence of the combination of anti-CD3 McAb and low dosed rIL-2 may facilitate a rapid expansion and long term maintenance of higher cytotoxicity. In general, this is a promising approach in tumor adoptive immunotherapy.

Abstract:A murine CD4 + thymocyte subset with phenotype of TCRαβ + 3G11 − 6C10 − CD4 + CD8 − CD69 +/- HSA med/lo contains the cells in relatively functional matured status. The functional property of the cells in this subset is characterized by the unique pattern of cytokine production at transitional stage from Th0 to Th2 type with the latter being the dominant type. After being co-cultured with murine thymic medullary epithelial cell line (MTEC1) cells, a murine thymic medullary type epithelial cell line, the TCRαβ + 3G11 − 6C10 − CD4 + CD8 − CD69 +/- HSA med/lo thymocytes, has exhibited significantly higher levels of proliferation capability and IL-6 production, whereas the production of IL-4 and IL-10 is suppressed after co-culturing with MTECl. By contrast, MTECl could not induce thymocytes to secrete Thl type of cytokines. The results suggest that MTECl can regulate functional status of this thymocyte subset and induce them to develop into a specialized Th2 subset.
