Abstract:Narrow dispersity organically modified silica nanoparticles (SiNPs), diameter ~30 nm, entrapping a hydrophobic two-photon absorbing fluorenyl dye, were synthesized by hydrolysis of triethoxyvinylsilane and (3-aminopropyl)triethoxysilane in the nonpolar core of Aerosol-OT micelles. The surface of the SiNPs were functionalized with folic acid, to specifically deliver the probe to folate receptor (FR) over-expressing Hela cells, making these folate two-photon dye-doped SiNPs potential candidates as probes for two-photon fluorescence microscopy (2PFM) bioimaging. In vitro studies using FR over-expressing Hela cells and low FR expressing MG63 cells demonstrated specific cellular uptake of the functionalized nanoparticles. One-photon fluorescence microscopy (1PFM) imaging, 2PFM imaging, and two-photon fluorescence lifetime microscopy (2P-FLIM) imaging of Hela cells incubated with folate-modified two-photon dye-doped SiNPs were demonstrated.

Abstract:Folate functionalized nanoparticles (NPs) that contain fluorogens with aggregation-induced emission (AIE) characteristics are fabricated to show bright far-red/near-infrared fluorescence, a large two-photon absorption cross section and low cytotoxicity, which are internalized into MCF-7 cancer cells mainly through caveolae-mediated endocytosis. One-photon excited in vivo fluorescence imaging illustrates that these AIE NPs can accumulate in a tumor and two-photon excited ex vivo tumor tissue imaging reveals that they can be easily detected in the tumor mass at a depth of 400 mu m. These studies indicate that AIE NPs are promising alternatives to conventional TPA probes for biological imaging.

Abstract:Highly emissive inorganic-organic nanoparticles with core-shell structures are fabricated by a one-pot, surfactant-free hybridization process. The surfactant-free sol-gel reactions of tetraphenylethene- (TPE) and silole-functionalized siloxanes followed by reactions with tetraethoxysilane afford fluorescent silica nanoparticles FSNP-1 and FSNP-2, respectively. The FSNPs are uniformly sized, surface-charged and colloidally stable. The diameters of the FSNPs are tunable in the range of 45-295 nm by changing the reaction conditions. Whereas their TPE and silole precursors are non-emissive, the FSNPs strongly emit in the visible vision, as a result of the novel aggregation-induced emission (AIE) characteristics of the TPE and silole aggregates in the hybrid nanoparticles. The FSNPs pose no toxicity to living cells and can be utilized to selectively image cytoplasm of HeLa cells.

Surface modification of SiNPs with the FA derivative was supported by zeta potential variation and H-1 NMR spectral characterization of the SiNPs as a function of surface modification. In vitro studies using HeLa cells expressing a folate receptor (FR) indicated specific cellular uptake of the functionalized nanopartides. The nanoprobe was demonstrated for FR-targeted one-photon in vivo imaging of HeLa tumor xenograft in mice upon intravenous injection of the probe. The FR-targeting nanoprobe not only exhibited highly selective tumor targeting but also readily extravasated from tumor vessels, penetrated into the tumor parenchyma, and was internalized by the tumor cells. Two-photon fluorescence microscopy bioimaging provided three-dimensional (3D) cellular-level resolution imaging up to 350 mu m deep in the HeLa tumor.

Abstract:In this study, a targeting drug delivery system based on mesoporous silica nanoparticle (MSN) was successfully developed for anti-cancer drug delivery and bioimaging. Carboxyl functionalized MSN (MSN/COOH) was firstly prepared and then modified with folate as the cancer targeting moiety and a near infrared fluorescent dye as labeling segment. Folate was conjugated to MSN/COOH via functional polyethyleneglycol (PEG), constructing the vector MSN/COOH-PEG-FA. The functionalization with carboxyl caused the pore surface of the nanocarrier more negative than native MSN, which could provide attractive forces between the nanoparticles and positively charged doxorubicin hydrochloride (DOX). Meanwhile, the folate modification significantly enhanced the cellular uptake of the delivery system compared to unmodified counterparts. Furthermore, the introduction of PEG increased the water dispersibility. Besides, the modification with the near infrared fluorescent dye Cy5 made the system effective for live cell and in vivo imaging. Therefore, the Cy5-MSN/COOH-PEG-FA system could be a promising nanocarrier for simultaneous diagnosis and treatment of diseases. (c) 2013 Elsevier B.V. All rights reserved.

Abstract:The present work describes the folate receptor targeted chitosan and polydopamine coated mesoporous silica nanoparticles (CS-FA@PDA@MSN) for photothermal therapy and drug delivery. The developed therapeutic diagnostic nanocarrier is used to combat multidrug resistance of cancer in combination with photothermal effect. The nanocarrier exhibited high ability to transport quercetin (Q) into A549 cells through specific targeting ability via recognition through folate receptors expressed on cancer cells and stimulated drug release in its specific acidic environment. In addition, CS-FA@PDA@MSN-Q showed exceptional photothermal effect and as a consequence high cytotoxicity on A549 cells when stimulated by 808 nm laser. Abstract:Dispersed incorporation of functional nanoparticles into ordered mesopores to exert their synergistic properties has shown a promising application, but proved to be a challenging task. Herein, a facile strategy based on solid-phase transformation of the template into polymer-coated carbonaceous nanodots (pCNDs) is used for direct synthesis of multifunctional mesoporous silica nanospheres (MFMSNs) incorporated with dispersed fluorescent carbon nanodots. The homogeneous pCNDs incorporated into mesoporous silica nanospheres (MSNs) can simultaneously act as the nontoxic fluorescence probe for highly efficient fluorescence imaging and also anchoring sites for anticancer drug loading. On the basis of the special hydrophobic π–π/electrostatic interaction between the confined pCNDs in MFMSNs and the aromatic drug molecules, MFMSNs could also function as an advanced drug carrier for pH-triggered drug release. The very simple strategy suggests new insights for synthesizing multifunctional mesoporous nanomaterials and extends the biomedical functions of traditional MSN-based nanovehicles for future biomedical applications.

Abstract:In this study, we introduced,a targeting polymer poly(ethylene glycol) folic acid (PEG FA) on the surface of polyclopatnine (PDA)modified mesoporous silica nanoparticles (MSNs) to develop the novel nanoparticles (NPs) MSNs@PDA-PEGHFA, which were employed as a drug delivery system loaded with doxorubicin (DOX) as a model drug for cervical cancer therapy. The chemical structure and properties of these NPs were characterized by transmission electron microscopy, X-ray photoelectron spectroscopy) N-2 adsorption/desorption, dynamic light scattering-autosizer, thermogravimetric analysis, and Fourier transform infrared spectroscopy. The pH-sensitive PDA coating served as a gatekeeper. The in vitro drug release experiments showed pH-dependent and 'sustained drug,release profiles that could enhance the therapeutic anticancer effect and minimize potential damage to normal cells due to the acidic microenvironment of theitumor. These MSN5@PDA-PEG-FA achieved significantly high targeting efficiency, which was demonstrated by the in vitro Cellular uptake and cellular targeting assay. Compared with that of free DOX and DOX, loaded NPs without the folic targeting ligakl, the FA-targeted NPs exhibited higher antitumor efficky in vivo) implying that they are a highly promising potential carrier for cancer treatments.

Abstract:Liquid crystalline nanoparticles (LCNPs) have gained much attention in cancer nanomedicines due to their unique features such as high surface area, storage stability, and sustained-release profile. In the current study, a novel LCNP for co-encapsulation of Bi2O3 and hydrophilic doxorubicin (DOX) was fabricated and functionalized with folic acid (FA) to achieve efficient tumor targeting toward CT-scan imaging and chemotherapy of melanoma in vitro and in vivo. LCNPs Bi2O3 NPs were prepared using glycerol monooleate-pluronic F-127 (GMO/PF127/water). Firstly, GMO/water were homogenized to prepare LC gel. Then, the stabilizer aqueous solution (PF127/Bi2O3/DOX) was added to the prepared LC gel and homogenized using homogenization and ultrasonication. The formulated NPs exhibited superior stability with encapsulation efficiency. High cytotoxicity and cellular internalization of the FA-Bi2O3-DOX-NPs were observed in comparison with Bi2O3-DOX-NPs and the free DOX in folate-receptor (FR) overexpressing cells (B16F10) in vitro. Moreover, ideal tumor suppression with increased survival rate were observed in tumorized mice treated with FA-Bi2O3-DOX-NPs compared to those treated with non-targeted one. On the other hand, the CT-imaging ability of the Bi2O3-DOX-NPs was tested inB16F10 tumor-bearing mice. The obtained data indicated a high potential of the developed targeted theranostic FA-Bi2O3-DOX-NPs for diagnostics and treatment of melanoma.

Abstract:A smart mesoporous silica nanocarrier with intracellular controlled release is fabricated, with folic acid as dual-functional targeting and capping agent. The folate not only improves the efficiency of the nanocarrier internalized by the cancer cells, but also blocks the pores of the mesoporous silica to eliminate premature leakage of the drug. With disulfide bonds as linkers to attach the dual-functional folate within the surface of mesoporous silica, the controlled release can be triggered in the presence of reductant dithiothreitol (DTT) or glutathione (GSH). The cellular internalization via folate-receptor-mediated endocytosis and the intracellular controlled release of highly toxic anticancer drug DOX were demonstrated with an in vitro HeLa cell culture, indicating an efficient cancer-targeted drug delivery.

Abstract:Mesoporous silica nanoparticles are promising candidates to deliver proteins, antibodies, and anticancer drugs and to be served as an adsorbent of various types of substrate. Herein, a theranostic approach, in the preparation of fluorescence-type mesoporous silica for pH-responsive delivery of anticancer drug, doxorubicin, is introduced. By this mean, three types of mesoporous silica were synthesized. The ordinary type of mesoporous silica nanoparticles containing propylamine functional groups was synthesized by a controlled-size technique. Two types of fluorescence mesoporous silica were prepared, doped with gold or quantum dots nanoparticles. The fluorescence imaging has shown that the optical properties of the gold or carbon quantum dots have been transferred to the mesoporous matrix in doping processes. The as-prepared mesoporous materials were conjugated to doxorubicin via Schiff-base linkage. Several characterization techniques were utilized to detect the structure of the particles including FT-IR spectroscopy, XRD, N2 adsorption/desorption, transition, and scanning electron microscopy. The amount of conjugated doxorubicin was analyzed by UV–Vis spectroscopy. The pH-responsive character was proved assessing the release profile in different pH ranges including physiological pH. The safety of the particles was evaluated by cell cytotoxicity assay on breast cancer cell line, MCF-7. As-prepared particles showed no cytotoxic behavior, but when they were loaded by doxorubicin, the appreciable release of the drug and subsequent cytotoxicity resulted against MCF-7 cells. The hemolytic study was conducted to prove the biocompatibility of these particles in the blood. Altogether, the proposed materials are suitable to be considered as safe and to be used as a theranostic platform for therapeutic and imaging purposes.

DTRF: A dynamic-trust-based recruitment framework for Mobile Crowd Sensing system

Abstract:Mobile Crowd Sensing (MCS) is a promising paradigm in which mobile users collect and share sensor data from their local environment using wireless mobile devices. The inherent openness of this platform and the selfishness of individuals make it easy to contribute low-quality sensor data, so the recruitment of suitable participants who are trustable and contribute high-quality sensor data, becomes a fundamental requirement for MCS system. In this paper, we propose a dynamic-trust-based recruitment framework (DTRF) for MCS system. Real-time direct trust and lightweight feedback aggregation trust are combined to select the well-suited participants. In addition, we adopt an adaptive weight allocation approach to calculate the overall trust degree of the participants. Theoretical analysis and extensive simulation confirm that DTRF can efficiently select the trustworthy participants and effectively stimulate the participants to contribute high-quality sensor data and thus get high task completion rate and data quality.

Abstract:We have constructed a theranostic drug delivery system, composed of a carrier and a drug. Europium (Eu3+) and gadolinium (Gd3+) were doped into mesoporous silica nanoparticles (MSNs) to form the carrier (EuGd-MSNs). The surface of EuGd-MSNs was then grafted by disulfide bonds with L-cysteine (Cys) and folic acid (FA), respectively. This theranostic drug delivery system, FA-EuGd-MSNs-SS-Cys, is capable of serving as a both diagnostic and therapeutic tool for imaging-tracing and providing disease treatment. FA targeting achieved with selectivity and phagocytosis specificity more efficiently. The evaluation of the cellular uptake shows that FA-EuGd-MSNs traps more cancer cells than EuGd-MSNs. The cell material shows that the intense intracellular red fluorescence emitted from the Eu3+ reveals that a significant amount of FA-EuGd-MSNs is ingested by Hela cells, and the paramagnetic functionality of the Gd3+ enhances the photoluminscence sensitivity through MRI, which shows EuGd-MSNs to be an efficient dual imaging agent. In a cytotoxicity evaluation of FA-EuGd-MSNs-SS-Cys by MTT, the Cys serves as an anticancer reagent. Cancer treatment can use drug therapies with higher concentration glutathione (GSH) to cut disulfide bonds intracellularly and allow the Cys to treat the disease more effectively. The EuGd-MSNs also presents excellent properties, including a lack of cyto-toxicity, good biocompatibility, and stability when it is cultivated with normal L929 cells. The FA-EuGd-MSNs-SS-Cys presents a useful theranostic nano-platform, with four primary functionalities: imaging probes, tracing, delivering therapeutic drugs, and a targeting ligand that can serve as an all-in-one diagnostic and therapeutic tool for cancer and aging-related diseases.

Abstract:Herein, we fabricated a new yolk-shell-structured mesoporous silica nanoparticle (YMSN) with multifunctionalities of fluorescence imaging, photothermal therapy (PTT), and drug delivery by using a fluorescein isothiocyanate-doped silica nanoparticle partially covered by patchy gold as the core. Different from the conventional selective etching procedure, the multifunctional silica core is left intact, and the alkali etching mainly occurs in a hexadecyl trimethyl ammonium bromide/silica hybrid layer, which leads to the formation of the void space in YMSNs. In addition, the utilization of patchy gold as the PTT agent can avoid the shield against the outer irradiation on the core. Results show that the as-prepared YMSNs have a good biocompatibility in the concentration of 0-1000 μg·mL-1 and a high doxorubicin-loading capability (8.04 wt %). In vitro and in vivo antitumor experiments reveal that the resulting YMSNs can be utilized for chemo- and photothermic combination therapy as well as optical imaging.

Abstract:Targeted nanocarrier can selectively deliver anti-tumor drugs to cancer sites, improving drug efficiency. Accordingly, a targeted nanocarrier (MSN-FA) was synthesized based on folic acid (FA) modified mesoporous silica nanoparticles (MSNs). These were loaded with 10-hydroxycamptothecin (HCPT) to obtain the nano-drug MSN-FA@HCPT. These nanocarriers were characterized by transmission electron microscopy (TEM), zeta potential, ultraviolet–visible spectroscopy (UV–Vis), Fourier transform infrared spectroscopy (FT-IR) and thermogravimetric analysis (TGA). Notably, the nanocarriers were nearly spherical before and after loading HCPT and exhibited good dispersibility. Also, folate receptor (FR) over-expressing HeLa cells and FR deficient HepG2 cells were used to evaluate in vitro cellular uptake and cytotoxicity of MSN-FA@HCPT and MSN@HCPT. Interestingly, FA-modified nanocarriers enhanced the cytotoxicity of HCPT by improving drug targeting to tumor cells. Also, apoptotic and mitochondrial membrane potential (MMP) reducing effects of MSN-FA@HCPT were more prominent than the MSNs without FA modification. The MSN-FA-loaded HCPT reduced MMP, increased intracellular ROS and induced cell apoptosis. MSN-FA@HCPT can be excellent drug carriers with profound biomedical applications.

Evaluation of novel platinum(ii) based AIE compound-encapsulated mesoporous silica nanoparticles for cancer theranostic application

Abstract:Advanced biomedical research has established that cancer is a multifactorial disorder which is highly heterogeneous in nature and responds differently to different treatment modalities, due to which constant monitoring of therapy response is becoming extremely important. To accomplish this, different theranostic formulations have been evaluated. However, most of them are found to suffer from several limitations extending from poor resolution, radiation damage, to high costs. In order to develop a better theranostic modality, we have designed and synthesized a novel platinum(ii)-based 'aggregation induced emission' (AIE) molecule (named BMPP-Pt) which showed strong intra-cellular fluorescence and also simultaneously exhibited potent cytotoxic activity. Due to this dual functionality, we wanted to explore the possibility of using this compound as a single molecule based theranostic modality. This compound was characterized using elemental analysis, NMR and IR spectroscopy, mass spectrometry and single crystal X-ray structure determination. BMPP-Pt was found to exhibit a high AIE property with emission maxima at 497 nm. For more efficient cancer cell targeting, BMPP-Pt was encapsulated into mesoporous silica nanoparticles (Pt-MSNPs) and the MSNPs were further surface modified with an anti-EpCAM aptamer (Pt-MSNP-E). Pt-MSNPs exhibited higher intracellular fluorescence compared to free BMPP-Pt, though both of them induced a similar degree of cell death via the apoptosis pathway, possibly via cell cycle arrest in the G1 phase. Anti-EpCAM aptamer modification was found to increase both cytotoxicity and intracellular fluorescence compared to unmodified MSNPs. Our study showed that EpCAM functionalized BMPP-Pt loaded MSNPs can efficiently internalize and induce apoptosis of cancer cells as well as show strong intracellular fluorescence. This study provides clues towards the development of a potential single compound based theranostic modality in future.

Abstract:The development of a drug delivery system with fluorescent biolabels is important in anti-cancer drug delivery application due to the potential for simultaneous diagnosis and treatment of diseases. Here, we reported the synthesis and multiple functionalization of mesoporous silica nanoparticle (MSN) for bioimaging and controlled drug release. After the functionalization with carboxyl group, the nanoparticles exhibited much better dispersity and stability in aqueous solution than MSN. Furthermore, a substantial doxorubicin (DOX) loading level was achieved and DOX-loaded nanoparticles exhibited noticeable pH-sensitive behavior with accelerated release of DOX in acidic environment. Compared with native DOX–MSN, DOX–MSN/COOH–Cy5 exhibited enhanced intracellular uptake efficacy and stronger effect on killing tumor cells. Meanwhile, it was observed that the MSN/COOH–Cy5 was able to locate in the cytoplasm of MCF-7 cells and could accumulate in tumor tissues for a long period of time. Overall, the functional nanoparticle could potentially be used for simultaneous controlled drug release and near-infrared fluorescent bioimaging.

Abstract:Highlights • Nanoparticle-based drug delivery devices may be useful for cancer treatment due to passive drug transfer. • Mesoporous silica and poly amid amine dendrimer nanoparticles are attractive for cancer treatment. • Mesoporous silica nanoparticles sustained the release of the 5-fluorouracil in both acidic and neutral conditions. • The release of 5-fluorouracil nanoparticles is controlled and influenced by pH changes. Modern nanomedicine delivers drugs to malfunctioning cells. Several synthetic processes can change the shape, particle size, polydispersity index, surface area, and porosity of mesoporous silica nanoparticles (MSNs). Polyamidoamine (PAMAM) dendrimers are attractive polymers with circular forms, uniform distribution, well-structured branching, internal cavities, and surface terminal groups. The objective of this study is to deliver 5-fluorouracil (5-FU) as passive targeting to carcinogen cells via MSN-PAMAM-G3 nanoparticles. This study involved the direct and adaptable synthesis of MSN as the core vehicle, together with third-generation PAMAM dendrimers as the vehicle shell, with the goal of delivering the 5-FU in a targeted and controllable manner. Tetraethyl orthosilicate (TEOS) and cetyltrimethylammonium bromide (CTAB) are used as templates to make the center nanoparticle. Methyl acrylate (MA) and ethylenediamine (EDA) are monomers that cause the formation of branches in PAMAM. The obtained particles were fully characterized in terms of particle size, polydispersity index, zeta potential, thermal behavior, morphology, pore diameter, pore volume, and specific surface area. The particle size of MSN-PAMAM-G3s showed around 187.71 ± 3.97 nm, with a polydispersity index of 0.17 ± 0.01 and zeta potential of 32.17 ± 1.30 mV. By reducing the ratio of drug to vehicle from 1 to 0.25, a significant increase in the EE% of 5-FU from 21.36 ± 0.70 to 43.12 ± 0.67 was observed. The release of 5-FU from the vehicle (5-FU@MSN and 5-FU@MSN-PAMAM-G3) was shown to be considerably higher in an acidic medium (pH = 5.5) in comparison to a neutral medium (pH = 7.4) (P-value<0.05). Furthermore, in both acidic and neutral conditions, 5-FU release was sustained from the vehicle rather than the 5-FU solution. Our research revealed that the MSN-PAMAM-G3 nanoparticles effectively regulated the encapsulation and release of 5-FU, providing pH-sensitive and sustained medication delivery.
