Abstract:AIM We aimed to assess the effect of autophagy and stromal interaction molecule 1 (STIM1) on podocyte epithelial-mesenchymal transition in diabetic nephropathy. METHODS The sera of 8-week-old db/db and C57BL/KsJ rats were used to culture MPC5 cells. The experiment was divided into 4 groups: MPC5 + siRNA-Scr + 10% C57BL/KsJ (Group A), MPC5 + siRNA-STIM1 + 10% C57BL/KsJ (Group B), MPC5 + siRNA-Scr + 10% db/db (Group C), and MPC5 + siRNA-STIM1 + 10% db/db (Group D). Podocyte autophagy was evaluated via immunofluorescence staining for LC3II and P62, and via Western blotting for P62 and LC3 (LC3II/LC3I). Western blotting was also used to assess the expression of TRPC6, Orai1, Beclin-1, Bcl-2, Caspase3, E-cadherin, fibronectin, and α-SMA protein. Furthermore, podocyte apoptosis was assessed via flow cytometry. RESULTS We found that, in podocytes cultured in the serum of diabetic nephrotic rats, the autophagy level decreased, whereas the apoptosis level increased, and EMT can be advanced. However, after silencing STIM1 with siRNA, a converse outcome was noted. Furthermore, in diabetic nephropathy rats, the up-regulated expression of podocyte STIM1 can activate TRPC6 and Orai1 channels, which results in Ca2+ entry. CONCLUSIONS We found that, in podocytes cultured in the serum of diabetic nephrotic rats, the autophagy level increased, whereas the apoptosis level decreased, and EMT can be inhibited by silencing STIM1 with siRNA.
TNF-? and IL-1? Promote Renal Podocyte Injury in T2DM Rats by Decreasing Glomerular VEGF/eNOS Expression Levels and Altering Hemodynamic Parameters

Abstract:Purpose: Diabetic nephropathy (DN) is a serious microvascular complication in those with type 2 diabetes mellitus (T2DM). Evidence confirms that serum tumor necrosis factor-alpha (TNF-alpha) and interleukin-1 beta (IL-1 beta) in the T2DM stage are proposed as prognostic markers for DN development, but it is unclear how they affect renal podocyte-associated nephrin and WT-1 expression. In the presence of podocyte injury, glomerular vascular endothelial growth factor (VEGF), endothelial nitric oxide synthase (eNOS) and hemodynamic parameters are dysregulated. The current research aimed to clarify the relationship of TNF-alpha and IL-1 beta with podocyte injury by altering VEGF/eNOS expression and hemodynamic parameters.Methods: A high-fat diet/streptozotocin-induced DN rat model was established. Serum TNF-alpha and IL-1 beta levels were tracked in the pre-T2DM, T2DM and DN stages. In the DN stage, the mRNA and protein expression levels of renal TNF-alpha, IL-1 beta, VEGF, eNOS, nephrin and WT-1 were studied. Renal hemodynamic parameters, including peak systolic velocity, end-diastolic flow velocity and mean velocity were measured with a color Doppler ultrasound technique.Results: Compared to those in the normal control (CTL) group, serum TNF-alpha and IL-1 beta levels increased significantly in the preT2DM stage (obesity, insulin resistance and hyperlipidemia), T2DM stage (hyperglycemia) and DN stage (abnormal renal functions) (all: P < 0.05) in the DN group. Serum TNF-alpha and IL-1 beta levels in the T2DM stage were significantly higher than those in the preT2DM stage (two: P < 0.05). Compared to the CTL group, renal nephrin, WT-1, TNF-alpha, IL-1 beta, eNOS and VEGF expression and hemodynamic parameters in the DN stage all showed significant differences separately (all: P < 0.05).Conclusion: Increased serum and renal TNF-alpha and IL-1 beta levels played important roles in reducing renal nephrin and WT-1 expression levels, which may be related to the fact that the former affected renal VEGF/eNOS expression and blood flow parameters in the DN rats.
Changes of TGF - β1 , Vimentin and Desmin Gene Expression in the Renal Cortex of Streptozotocin - induced Diabetic Rats

Abstract:Objective To study the expression of TGF-β_1 , vimentin and desmin gene in the renal cortex of STZ induced diabetic rats. Methods Diabetes was induced in 32 male SD rats by single intraperitoneal injection of 1.0%STZ (70mg/kg). 36 age and weight, sex matched SD rats used as controls. The expression of TGF-β_1 , vimentin and desmin mRNA in the renal cortex were detected by RT-PCR on the 3~ rd , the 7~ th , the 14~ th , the 30~ th , the 60~ th and the 90~ th days after the DM rat model established. Results The expression of TGF-β_1 , vimentin mRNA in the renal cortex of diabetic rats gradually increased respectively from the 7~ th day and the 14~ th day after the model established, and the expressive intensity was significantly greater than that in controls (P<0.05 or P<0.01). However, the expression of desmin mRNA in the renal cortex of diabetic rats gradually decreased from the 14~ th day after the model established, and the expressive intensity was significantly less than that in controls (P<0.05 or P<0.01). ② The expression level of TGF-β_1 mRNA correlated positively to that of vimentin mRNA in the renal cortex of diabetic rats, while the expression level of desmin mRNA correlated negatively to that of TGF-β_1 mRNA and to that of vimentin mRNA in the renal cortex of diabetic rats. Conclusions There may be partial transformation of renal tubular epithelial into fibroblast and TGF-β may play a role in this transformation during the progressive interstitial fibrosis of diabetic nephropathy.
Angiotensin II Type 1 Receptor Activation Mediates High Glucose-Induced Rat Proximal Tubule Epithelial Mesenchymal Transition

Activation of the renin-angiotensin system (RAS) is involved in diabetic nephropathy. The present study investigated the positive role of angiotensin AT(1) receptors in high glucose-induced EMT in cultured tubular epithelial cells. 2. A rat kidney proximal tubular epithelial cell line (NRK-52E) was used in the present study. Levels of EMT makers, namely E-cadherin and vimentin, were estimated using fluorescence immunocytochemistry, mRNA levels of angiotensinogen (AGT), angiotensin-converting enzyme (ACE) and AT(1) receptors were determined by real-time polymerase chain reaction, protein levels of E-cadherin, vimentin, fibronectin, matrix metallopeptidase (MMP)-9 and phosphorylated extracellular signal-regulated kinase (ERK) 1/2 were analysed by western blotting and the concentrations of angiotensin (Ang) II and transforming growth factor (TGF)-beta 1 in the culture medium were determined by enzyme immunoassay and ELISA. 3. High glucose (30 mmol/L) induced EMT and increased the synthesis of fibronectin and MMP-9. Furthermore, high glucose increased AGT, ACE and AT(1) receptor mRNA levels, as well as AngII and TGF-beta 1 concentrations in the culture medium and ERK1/2 phosphorylation. Pretreatment of cells for 15 min with the AT(1) receptor antagonist losartan (10-5 mol/L) attenuated high glucose-induced increases in TGF-beta 1 and ERK1/2 phosphorylation and reduced EMT, as well as the consequent synthesis of fibronectin and MMP-9. 4. The results of the present study suggest that the activated local RAS mediates high glucose-induced EMT. By activating AT(1) receptors and stimulating TGF-beta 1 synthesis, the elevated local RAS participates in high glucose-induced EMT and increased extracellular matrix secretion.

Abstract:Bone morphogenetic protein-7 (BMP-7) expression is known to be protective for renal damage during diabetic nephropathy and disappears early during the progression of diabetic nephropathy. However, changes in expression of BMP-7 and BMP-7 type II receptor (BMP-RII) during kidney nephropathy response to high glucose-induced oxidative stress remain unclear. In this study, we used streptozotocin-induced diabetic rats with diabetic nephropathy and treated them with insulin, phloridzin, or antioxidant tiron. The insulin, phloridzin, or tiron treatment improved the renal function and decreased fibronectin expression in the streptozotocin-induced diabetic rats. Both insulin and phloridzin could reverse the attenuation effects of hyperglycemia on BMP-7 and BMP-RII expressions in the kidneys of streptozotocin-induced diabetic rats through the correction of hyperglycemia. However, the decrease of BMP-7 and BMP-RII expressions in kidney of streptozotocin-induced diabetic rats could be reversed by tiron through decreasing the high glucose-induced oxidative stress but not through changing the levels of glucose. We further confirmed the effect on reversing the BMP-7 and BMP-RII expressions through decreasing oxidative stress by tiron treatment in high glucose exposed mesangial cells. Thus, we suggest that a decrease in oxidative stress is responsible for the improvement of renal function and recovery of renal BMP-7 and BMP-RII expression in streptozotocin-induced diabetic rats.

Abstract:In streptozotocin (STZ)-induced Type-1 diabetic (T1D) rats, the collecting duct is the main source of intrarenal prorenin. The fact that binding of prorenin to the PRR triggers intracellular signals linked to tissue fibrosis has raised the expectation that prorenin might be directly responsible for diabetic nephropathy (DN). We showed that STZ-induced T1D rats exhibit greater apical distribution of PRR in the collecting duct. Therefore, we hypothesize that high glucose increases PRR abundance at the cell plasma membrane (PM) of collecting duct cells, which contributes to the stimulation of downstream fibrotic factors. To test this, we used male Sprague-Dawley rats (14w of age) with STZ-induced hyperglycemia for 7d (ip, single injection; 60 ng/kg), and cultures of collecting duct M-1 cells treated for 0, 5 min, and 1, 6, 12 and 24h with normal glucose (NG; 5mM glucose + 20mM mannitol) and high glucose (HG; 25mM). After 7d, STZ-induced rats (N=9) showed plasma levels of glucose as 428±13 vs.138±9 gr/dL and insulin as 0.07±0.02 vs. 2.4±0.01 ng/mL; compared to controls (N=7). Although PRR mRNA levels did not differ between groups; PRR protein levels and its downstream target TGF-β mRNA levels were augmented in the renal medulla of STZ-induced rats (0.55±0.03 vs.0.44±0.02 PRR/β-actin protein ratio; P<0.01; 1.22±0.06 vs.0.97±0.03 TGF-β/β-actin mRNA ratio; P<0.01). Interestingly, PRR protein levels were maximum elevated at 1h in PM extracts from M-1 cells treated with HG (0.95±0.04 vs. 0.33±0.2 PRR/E-cadherin protein ratio) but not in NG-treated cells. HG also unregulated downstream fibrotic factor fibronectin (1.9±0.2 vs.0.46±0.05 fold change) compared with NG-treated cells. Immunofluorescence studies of M-1 cells treated with NG for 1h showed that PRR is mainly localized in the perinuclear areas, whereas in HG-treated cells it was predominantly localized toward the cell surface, as it was more clearly observed in de-convoluted images. These data indicate that hyperglycemia leads to PRR trafficking alterations by inducing PRR translocation towards PM in the collecting duct cells and suggest that the activation of PRR during hyperglycemia conditions might be a novel mechanism underlying the development of DN, particularly tubulointerstitial fibrosis in DM.

Angiotensin II Type 2 Receptor Mediated Angiotensin II and High Glucose Induced Decrease in Renal Prorenin/renin Receptor Expression.

Abstract:Activation of prorenin/renin receptor [(P)RR] mediates non-enzymatic pathway for the physiological function of prorenin/renin. It also plays a role in the pathogenesis of diabetic nephropathy. However, the mechanisms of regulating (P)RR expression are only partially understood. In the present study, we examine the change of (P)RR under hyperglycemic conditions in the kidneys of streptozotocin (STZ)-induced diabetic (DM) rats and cultured rat renal glomerular mesangial cells (MCs). The (P)RR mRNA level was significantly lower in the kidney of (DM) rats than that of untreated control animals, meanwhile the plasma and renal angiotensin II (Ang II) levels and Ang II type 2 receptor (AT(2)R) mRNA level, but not Ang II type 1 receptor (AT(1)R) mRNA level were increased in (DM) rats. In cultured MCs, Ang II reduced the (P)RR expression and this inhibitory effect was blocked by the AT(2)R antagonist, PD123319, but not the AT(1)R antagonist, losartan. The AT(2)R agonist CGP42112A produced a similar effect as Ang II. Exposure to high glucose (30mM) resulted in a decrease in the (P)RR expression. PD123319, but not losartan, reversed high glucose induced (P)RR expression reduction. The present results indicate that activation of AT(2)R, but not AT(1)R, is most likely responsible for the reduced (P)RR expression in response to Ang II and high glucose in glomerular MCs and the renal tissue of STZ-treated rats.
Urotensin II Induces ER Stress and EMT and Increase Extracellular Matrix Production in Renal Tubular Epithelial Cell in Early Diabetic Mice

Abstract:Background/Aims: Urotensin II (UII) and its receptor are highly expressed in the kidney tissue of patients with diabetic nephropathy (DN). The aim of this study is to examine the roles of UII in the induction of endoplasmic reticulum stress (ER stress) and Epithelial-mesenchymal transition (EMT) in DN in vivo and in vitro. Methods: Kidney tissues were collected from patients with DN. C57BL/6 mice and mice with UII receptor knock out were injected with two consecutive doses of streptozotocin to induce diabetes and were sacrificed at 3th week for in vivo study. HK-2 cells in vitro were cultured and treated with UII. Markers of ER stress and EMT, fibronectin and type IV collagen were detected by immunohistochemistry, real time PCR and western blot. Results: We found that the expressions of protein of UII, GRP78, CHOP, ALPHA-SMA, fibronectin and type IV collagen were upregulated while E-cadherin protein was downregulated as shown by immunohistochemistry or western blot analysis in kidney of diabetic mice in comparison to normal control; moreover expressions of GRP78, CHOP, ALPHA-SMA, fibronectin and type IV collagen were inhibited while E-caherin expression was enhanced in kidney in diabetic mice with UII receptor knock out in comparison to C57BL/6 diabetic mice. In HK-2 cells, UII induced upregulation of GRP78, CHOP, ALPHA-SMA, fibroblast-specifc protein 1(FSP-1), fibronectin and type collagen and downregulation of E-cadherin. UII receptor antagonist can block UII-induced ER stress and EMT; moreover, 4-PBA can inhibit the mRNA expression of ALPHA-SMA and FSP1 induced by UII in HK-2 cells. Conclusions: We are the first to verify UII induces ER stress and EMT and increase extracellular matrix production in renal tubular epithelial cell in early diabetic mice. Moreover, UII may induce renal tubular epithelial EMT via triggering ER stress pathway in vitro, which might be the new pathogenic pathway for the development of renal fibrosis in DN.
Gene expression profiling in glomeruli of diabetic nephropathy rat

Abstract:Diabetic nephropathy (DN) remains the most common cause of end-stage renal disease (ESRD) as the burden of diabetes increases worldwide. To find improved intervention strategies for this disease, it is necessary to investigate the molecular mechanisms involved. To obtain more insight into processes that lead to DN, mRNA expression profiles of diabetic and normal glomeruli from rat kidneys were compared. Rats were divided into a control group and a DN group randomly. The DN group was injected with streptozotocin. Fasting blood glucose (FBG) and weight were measured monthly. On the 12th week, blood samples were collected and analyzed for plasma creatinine and blood urea nitrogen (BUN). Glomeruli were isolated and Illumina Rat Ref-12 V1.0 Expression Beadchip gene array was performed. Quantitative realtime polymerase chain reaction (Q-RT-PCR) was used to confirm the results of gene array for a selected number of genes. We found FBG, 24-h urinary albumin, serum creatinine and BUN were significantly increased, while urinary creatinine and body weight were significantly decreased in the DN group. Glomeruli from the DN group had 624 genes with differential expression. DAVID (Database for Annotation, Visualization and integrated Discovery) analysis showed that the three most enriched terms were ‘cytosol’ (GO:0005829), ‘translational elongation’ (GO:0006414) and ‘mitochondion’ (GO:0005739). Those genes could be mapped to eight pathways. The most common type of enriched pathway was related to ‘extracellular matrix (ECM)-receptor interaction’. Other pathways included those for ‘ribosome’, ‘focal adhesion’, ‘oxidative phosphorylation’, ‘transforming growth factor (TGF)-beta signaling pathway’, ‘Parkinson's disease’, ‘Alzheimer's disease’ and ‘renin–angiotensin system’. Q-RT-PCR verified that Atp5b (F1-ATPase beta subunit), Col1a1 (collagen type 1 alpha 1), Cox6c (cytochrome c oxidase subunit VIc), Ndufs3 (NADH dehydrogenase [ubiquinone] Fe-S protein 3) and Tgfb1 (transforming growth factor β1) were significantly up-regulated in the DN group. The expressions of NDUFS3 and TGF- β1 in DN rats were increased. Our findings suggested that the oxidative phosphorylation pathway, ECM-receptor interaction, TGF- β pathway and renin–angiotensin system may be involved in the development of DN.

Inhibition of STAT3 in tubular epithelial cells prevents kidney fibrosis and nephropathy in STZ-induced diabetic mice

Abstract:Abstract Recent evidences indicate that signal transducer and activator of transcription 3 (STAT3) is one of the crucial signaling pathways in the progression of diabetic nephropathy (DN). Here, we investigated the hypothesis that pharmacological blockade of STAT3 limits the progression of DN. Treatment with selective STAT3 inhibitor, S3I-201 for 16 weeks significantly attenuated kidney injuries in streptozotocin (STZ) induced diabetic mice, associated with downregulated expression of TGF-β1, ACE/AT1, and VEGF in diabetic mouse kidneys. Similar results were confirmed using genetic knockdown of STAT3 in mouse kidneys by injections of AAV2 expressing STAT3 shRNA in diabetic mouse. Further, STAT3 localization in kidney tissue was evaluated using immunofluorescent double-staining analysis, which indicated that STAT3 expression was mainly in the tubular epithelial cells. As expected, in renal tubular epithelial NRK-52E cells, high glucose (HG)-induced overexpression of TGF-β1, ACE/AT1, and VEGF were abrogated by S3I-201 pretreatment, as well as by genetic knockdown of STAT3 using specific siRNA sequence. This study found that renal tubular epithelial cells contributed to STAT3-mediated progression of DN and provided the first evidence that pharmacological inhibition of STAT3 attenuates DN.

Identification of nuclear receptor mRNAs by RT-PCR amplification of conserved zinc-finger motif sequences.

Abstract:Highly degenerate PCR primers were designed based on the amino acid sequence of the zinc finger motifs of known nuclear receptor members. Reverse transcription (RT)-PCR was performed using these degenerate primers starting with total RNA isolated from human umbilical vein endothelial cells (HUVEC). We identified 13 nuclear receptors of which three were novel. Of the novel orphans two-RZR alpha and PHR-1-were further characterised. Cloning of their cDNAs and sequence comparisons revealed that RZR alpha is related to Drosophila DHR3, RAR and RXR. PHR-1 turned out to be the human homologue of the recently published murine LRH-1. By Northern blot analyses we found RZR alpha mRNA expressed in a variety of organs, whereas a high amount of PHR-1 mRNA was found in pancreas and less in liver. Overexpression of Nrf2 in Renal Proximal Tubular Cells Stimulates Sodium–Glucose Cotransporter 2 Expression and Exacerbates Dysglycemia and Kidney Injury in Diabetic Mice

Abstract:We investigated the impact of nuclear factor erythroid 2–related factor 2 (Nrf2) overexpression in renal proximal tubular cells (RPTCs) on blood glucose, kidney injury, and sodium–glucose cotransporter 2 (Sglt2) expression in diabetic Akita Nrf2−/−/Nrf2RPTC transgenic (Tg) mice. Immortalized human RPTCs (HK2) stably transfected with plasmid containing the SGLT2 promoter and human kidneys from patients with diabetes were also studied. Nrf2 overexpression was associated with increased blood glucose, glomerular filtration rate, urinary albumin-to-creatinine ratio, tubulointerstitial fibrosis, and Sglt2 expression in Akita Nrf2−/−/Nrf2RPTC Tg mice compared with their Akita Nrf2−/− littermates. In vitro, oltipraz or transfection of NRF2 cDNA stimulated SGLT2 expression and SGLT2 promoter activity in HK2, and these effects were inhibited by trigonelline or NRF2 siRNA. The deletion of the NRF2-responsive element (NRF2-RE) in the SGLT2 promoter abolished the stimulatory effect of oltipraz on SGLT2 promoter activity. NRF2 binding to the NRF2-RE of the SGLT2 promoter was confirmed by gel mobility shift assay and chromatin immunoprecipitation assays. Kidneys from patients with diabetes exhibited higher levels of NRF2 and SGLT2 in the RPTCs than kidneys from patients without diabetes. These results suggest a link by which NRF2 mediates hyperglycemia stimulation of SGLT2 expression and exacerbates blood glucose and kidney injury in diabetes.

Abstract:Erythropoietin (EPO) has beneficial effects on glucose metabolism and insulin resistance. However, the mechanism underlying these effects has not yet been elucidated. This study aimed to investigate how EPO affects hepatic glucose metabolism. Here, we report that EPO administration promoted phosphatidylinositol 3-kinase (PI3K)/AKT pathway activation in palmitic acid (PA)-treated HepG2 cells and in the liver of high-fat diet (HFD)-fed mice, whereas adenovirus-mediated silencing of the erythropoietin receptor (EPOR) blocked EPO-induced AKT signalling in HepG2 cells. Importantly, a peroxisome proliferator-activated receptor γ (PPARγ) antagonist and PPARγ small interfering RNA (siRNA) abrogated the EPO-induced increase in p-AKT in HepG2 cells. Lentiviral vector-mediated hepatic PPARγ silencing in HFD-fed C57BL/6 mice impaired EPO-mediated increases in glucose tolerance, insulin sensitivity and hepatic AKT activation. Furthermore, EPO activated the AMP-activated protein kinase (AMPK)/sirtuin 1 (SIRT1) signalling pathway and AMPKα and SIRT1 knockdown each attenuated the EPO-induced PPARγ expression and deacetylation and PPARγ-dependent AKT activation in HepG2 cells. In summary, these findings suggest that PPARγ is involved in EPO/EPOR-induced AKT activation and targeting the PPARγ/AKT pathway via EPO may have therapeutic implications for hepatic insulin resistance and type 2 diabetes.
Glomerular 20-HETE, EETs, and TGF-beta1 in diabetic nephropathy

Abstract:The early stage of diabetic nephropathy (DN) is linked to proteinuria. Transforming growth factor (TGF)-beta1 increases glomerular permeability to albumin (P(alb)), whereas 20-HETE and EETs reduce P(alb). To investigate the impact of hyperglycemia and hyperlipidemia on 20-HETE, EETs, and TGF-beta1 in the glomeruli, rats were divided into four groups: ND rats were fed a normal diet, HF rats were fed a high-fat diet, STZ rats were treated with 35 mg/kg of streptozotocin, and HF/STZ rats were fed a HF diet and treated with STZ. After 10 wk on these regimens, blood glucose, urinary albumin, serum cholesterol, serum triglyceride levels, and the kidney-to-body weight ratio were significantly elevated in STZ and HF/STZ rats compared with HF and ND rats. STZ and HF/STZ rats had histopathologic changes and abnormal renal hemodynamics. Expression of glomerular CYP4A, enzymes for 20-HETE production, was significantly decreased in STZ rats, whereas expression of glomerular CYP2C and CYP2J, enzymes for EETs production, was significantly decreased in both STZ and HF/STZ rats. Moreover, glomerular TGF-beta1 levels were significantly greater in STZ and HF/STZ rats than in HF and ND rats. Five-week treatment of STZ rats with clofibrate induced glomerular CYP4A expression and 20-HETE production, but reduced glomerular TGF-beta1 and urinary protein excretion. These results demonstrate that hyperglycemia increases TGF-beta1 but decreases 20-HETE and EETs production in the glomeruli, changes that may be important in causing glomerular damage in the early stage of DN.

Abstract:The association between type 2 diabetes and obesity is very strong, and cardiovascular complications are the major cause of morbidity and mortality in diabetic patients. The aim of this study was to investigate early changes in the pattern of genes encoding cardiac muscle regulatory proteins and associated changes in ventricular myocyte contraction and Ca2+ transport in young (9‐ to 13‐week‐old) type 2 Zucker diabetic fatty (ZDF) rats. The amplitude of myocyte shortening was unaltered; however, time‐to‐peak shortening and time to half‐relaxation of shortening were prolonged in ZDF myocytes (163 ± 5 and 127 ± 7 ms, respectively) compared with age‐matched control rats (136 ± 5 and 103 ± 4 ms, respectively). The amplitude of the Ca2+ transient was unaltered; however, time‐to‐peak Ca2+ transient was prolonged in ZDF myocytes (66.9 ± 2.6 ms) compared with control myocytes (57.6 ± 2.3 ms). The L‐type Ca2+ current was reduced, and inactivation was prolonged over a range of test potentials in ZDF myocytes. At 0 mV, the density of L‐type Ca2+ current was 1.19 ± 0.28 pA pF−1 in ZDF myocytes compared with 2.42 ± 0.40 pA pF−1 in control myocytes. Sarcoplasmic reticulum Ca2+ content, release and uptake and myofilament sensitivity to Ca2+ were unaltered in ZDF myocytes compared with control myocytes. Expression of genes encoding various L‐type Ca2+ channel proteins (Cacna1c, Cacna1g, Cacna1h and Cacna2d1) and cardiac muscle proteins (Myh7) were upregulated, and genes encoding intracellular Ca2+ transport regulatory proteins (Atp2a2 and Calm1) and some cardiac muscle proteins (Myh6, Myl2, Actc1, Tnni3, Tnn2, and Tnnc1) were downregulated in ZDF heart compared with control heart. A change in the expression of genes encoding myosin heavy chain and L‐type Ca2+ channel proteins might partly underlie alterations in the time course of contraction and Ca2+ transients in ventricular myocytes from ZDF rats.

Abstract:Background. Renal ischemia/reperfusion (I/R) injury (RI/RI) is a common complication of diabetes mellitus (DM) in surgical practice. Oxidative stress plays a crucial role in this process. Recombinant human erythropoietin (rhEPO) is usually used to treat anemia resulting from several diseases. However, the functional involvement of rhEPO in diabetic RI/RI remains unclear. The present study was intended to investigate the antioxidant role of rhEPO on RI/RI in DM rats. Methods. The bilateral renal arteries and veins of streptozotocin-induced diabetic rats were subjected to 45 minutes of ischemia followed by 1, 6, and 24 hours of reperfusion with or without rhEPO pretreatment at the beginning of an I/R procedure. The renal tissue pathomorphology, renal function, oxidative stress, and inflammatory response were evaluated by detection of a series of indices by hematoxylin-eosin staining, commercial kits, enzyme-linked immunosorbent assay, and spectrophotofluorometry, respectively. Results. Compared to the I/R group, renal function was significantly advanced in the erythropoietin group, whose subjects were also subjected to renal tissue injury, oxidative stress, and inflammation. Conclusion. These results suggest that rhEPO preconditioning can attenuate diabetic RI/RI through regulating endogenous antioxidant activity.
Insulin deficiency induces rat renal mesangial cell dysfunction via activation of IGF-1/IGF-1R pathway

Abstract:Aim:Diabetic nephropathy is one of the major complications of diabetes and the major cause of end-stage renal disease. In this study we investigated the insulin deficiency (ID) induced changes in renal mesangial cells (MCs) and in the kidney of STZ-induced diabetic rats.Methods:Cultured rat renal MCs were incubated in ID media. Cell proliferation was analyzed using BrdU incorporation assay. The expression of insulin receptor (IR), insulin-like growth factor-1 receptor (IGF-1R), phosphorylated IGF-1R, fibronectin, and collagen IV was determined with Western blot analysis. STZ-induced diabetic rats were treated with an IGF-1R antagonist picropodophyllin (PPP, 20 mg·kg−1·d−1, po) for 8 weeks. After the rats were euthanized, plasma and kidneys were collected. IGF-1 levels in renal cortex were measured with RT-PCR or ELISA. The morphological changes in the kidneys were also examined.Results:Incubation in ID media significantly increased cell proliferation, the synthesis of fibronectin and collagen IV, and the expression of IGF-1 and IGF-1R and phosphorylated IGF-1R in renal MCs. Pretreatment of the cells with PPP (50 nmol/L) blocked ID-induced increases in cell proliferation and the synthesis of fibronectin and collagen IV; knockdown of IGF-1R showed a similar effect as PPP did. In contrast, treatment of the cells with IGF-1 (50 ng/mL) exacerbated ID-induced increases in cell proliferation. In the kidneys of diabetic rats, the expression of IGF-1, IGF-1R and phosphorylated IGF-1R were significantly elevated. Treatment of diabetic rats with PPP did not lower the blood glucose levels, but significantly suppressed the expression of TGF-β, fibronectin and collagen IV in the kidneys, the plasma levels of urinary nitrogen and creatinine, and the urinary protein excretion.Conclusion:Insulin deficiency increases the expression of IGF-1 and IGF-1R in renal MCs and the kidney of diabetic rats, which contributes to the development of diabetic nephropathy.

Abstract:BACKGROUND:Diabetic nephropathy (DN), a complication of diabetes, is the result of high glucose-induced pathological changes in podocytes, such as epithelial-mesenchymal transition (EMT). Autophagy is an important mechanism of podocyte repair. Ginsenoside Rg1, the active ingredient of ginseng extract, has antifibrotic and proautophagic effects. Therefore, we hypothesized that ginsenoside Rg1 can reverse podocyte EMT via autophagy and alleviate DN.AIM:This study aimed to investigate the effect of ginsenoside Rg1 on DN rats and high glucose-induced podocyte EMT by regulating the AKT/GSK3β/β/.METHODS:Diabetic rats induced by STZ injection were treated with 50 mg/kg ginsenoside Rg1 for 8 weeks, and the renal functional, metabolic, and histopathological indices were evaluated. DN was simulated in vitro by exposing podocytes to high glucose levels and treated with ginsenoside Rg1. The expression of EMT and autophagy-related markers was analyzed in vivo and in vitro by exposing podocytes to high glucose levels and treated with ginsenoside Rg1. The expression of EMT and autophagy-related markers was analyzed.RESULTS:Ginsenoside Rg1 significantly alleviated renal fibrosis and podocyte EMT in diabetic rats, and podocytes exposed to high glucose levels, which was abolished by the autophagy inhibitor 3-MA. Furthermore, ginsenoside Rg1 regulated the AKT/GSK3 β/β/.CONCLUSION:Ginsenoside Rg1 alleviated podocyte EMT by enhancing AKT/GSK3β/β-catenin pathway-mediated autophagy, indicating its therapeutic potential for DN and other glomerular diseases.β/β/.

Abstract:P102 is a multifunctional transcriptional co-activator. This experiment is designed to investigate the role of p102 in the activation of renin–angiotensin system (RAS) and sequentially extracellular matrix (ECM) over synthesis in diabetic nephropathy. Rat glomerular mesangial cells (MCs) or isolated glomeruli were cultured in normal glucose (NG, 5.5mM) or high glucose (HG, 25mM) DMEM. The generation of reactive oxygen species was measured by 2′,7′-dichlorodihydrofluorescein diacetate (DCFH-DA) fluorescent probe assay. The protein levels were analyzed by Western blot and the mRNA levels were evaluated by real-time PCR. HG treatment induced an increase in reactive oxygen species production. Culturing the cells in HG for 48h, p102 mRNA and protein, angiotensin II type 1 receptor (AT1 receptor) mRNA, transforming growth factor-β1 (TGF-β1) and fibronectin proteins were significantly increased. NADPH oxidase inhibitor DPI blocked the HG-induced p102, TGF-β1 and fibronetcin elevations. Knockdown on p102 expression by siRNA depressed the HG-induced AT1 receptor up-regulation as well as the increases in TGF-β1 and fibronectin. In contrast, AT1 receptor antagonist candesartan did not influence p102 levels under either NG or HG condition, but blocked the HG-induced TGF-β1 and fibronectin increases. The results from isolated glomeruli were consistent with that of MCs, which showed that HG exposure stimulated the expression of p102. These results suggest that the overproduction of reactive oxygen species at the early stage of HG incubation stimulates p102 synthesis, which in turn up-regulates AT1 receptor expression. The activation of RAS stimulates TGF-β1 and fibronectin production, which further results in ECM accumulation.
