Abstract:In Arabidopsis, there is a family of receptor-like protein kinases (RLKs) containing novel cysteine-rich repeats in their extracellular domains. Genes encoding many of these cysteine-rich RLKs (CRKs) are induced by pathogen infection, suggesting a possible role in plant defense responses. We have previously generated Arabidopsis plants expressing four pathogen-regulated CRK genes (CRK5, 6, 10 and 11) under control of a steroid-inducible promoter and found that induced expression of CRK5, but not the other three CRK genes, triggered hypersensitive response-like cell death in transgenic plants. In the present study, we have analyzed the structural relationship of the CRK family and identified three CRKs (CRK4, 19 and 20) that are structurally closely related to CRK5. Genes encoding these three CRKs are all induced by salicylic acid and pathogen infection. Furthermore, induced expression of CRK4, 19and 20 all activates rapid cell death in transgenic plants. Thus, the activity of inducing rapid cell death is shared by these structurally closely related CRKs. We have also performed yeast two-hybrid screens and identified proteins that interact with the kinase domains of CRKs. One of the identified CRK-interacting proteins is the kinase-associated type 2C protein phospohatase known to interact with a number of other RLKs through its kinase-interacting FHA domain. Other CRK-interacting proteins include a second protein with a FHA domain and another type 2C protein phosphatase. Interactions of CRKs with these three proteins in vivo were demonstrated through co-immunoprecipitation. These CRK-interacting proteins may play roles in the regulation and signaling of CRKs.
Functional Characterization of GhAKT1, a Novel Shaker-like K+ Channel Gene Involved in K+ Uptake from Cotton (gossypium Hirsutum)

Abstract:Shaker-like potassium (K+) channels in plants play an important role in K+ absorption and transport. In this study, we characterized a Shaker-like K+ channel gene GhAKT1 from the roots of Gossypium hirsutum cv. Liaomian17. Phylogenetic analysis showed that the GhAKT1 belongs to the AKT1-subfamily in the Shaker-like K+ channel family. Confocal imaging of a GhAKT1-green fluorescent fusion protein (GFP) in transgenic Arabidopsis plants indicated that GhAKT1 is localized in the plasma membrane. Transcript analysis located GhAKT1 predominantly in cotton leaves with low abundance in roots, stem and shoot apex. Similarly, β-glucuronidase (GUS) activity was detected in both leaves and roots of PGhAKT1::GUS transgenic Arabidopsis plants. In roots, the GUS signals appeared in the epidermis, cortex and endodermis and root hairs, suggesting the contribution of GhAKT1 to K+ uptake. In leaves, GhAKT1 was expressed in differentiated leaf primordial as well as mesophyll cells and veins of expanded leaves, pointing to its involvement in cell elongation and K+ transport and distribution in leaves. Severe K+ deficiency did not affect the expression of GhAKT1 gene. GhAKT1-overexpression in either the Arabidopsis wild-type or akt1 mutant enhanced the growth of transgenic seedlings under low K+ deficiency and raised the net K+ influx in roots at 100 μM external K+ concentration, within the range of operation of the high-affinity K+ uptake system. The application of 2 mM BaCl2 resulted in net K+ efflux in roots, and eliminated the differences between GhAKT1-overexpression lines and their acceptors indicating that the K+ uptake mediated by GhAKT1 is also as Ba2 +-sensitive as AtAKT1.

Abstract:Neutron-rich matter at subnuclear densities may involve complex structures displaying a variety of shapes, such as spherical, slablike, and∕or rodlike shapes. These phases of the nuclear pasta are expected to exist in the crust of neutron stars and in core-collapse supernovae. The dynamics of core-collapse supernovae is very sensitive to the interactions between neutrinos and nucleons∕nuclei. Indeed, neutrino excitation of the low-energy modes of the pasta may allow for a significant energy transfer to the nuclear medium, thereby reviving the stalled supernovae shock. The linear response of the nuclear pasta to neutrinos is modeled via a simple semiclassical simulation. The transport mean free path for $\ensuremath{\mu}$ and $\ensuremath{\tau}$ neutrinos (and antineutrinos) is expressed in terms of the static structure factor of the pasta, which is evaluated using Metropolis Monte Carlo simulations.

Abstract:SHORT-ROOT (SHR) is a key regulator of root growth and development in Arabidopsis (Arabidopsis thaliana). Made in the stele, the SHR protein moves into an adjacent cell layer, where it specifies endodermal cell fate; it is also essential for apical meristem maintenance, ground tissue patterning, vascular differentiation, and lateral root formation. Much has been learned about the mechanism by which SHR controls radial patterning, but how it regulates other aspects of root morphogenesis is still unclear. To dissect the SHR developmental pathway, we have determined the genome-wide locations of SHR direct targets using a chromatin immunoprecipitation followed by microarray analysis method. K-means clustering analysis not only identified additional quiescent center-specific SHR targets but also revealed a direct role for SHR in gene regulation in the pericycle and xylem. Using cell type-specific markers, we showed that in shr, the phloem and the phloem-associated pericycle expanded, whereas the xylem and xylem-associated pericycle diminished. Interestingly, we found that cytokinin level was elevated in shr and that exogenous cytokinin conferred a shr-like vascular patterning phenotype in wild-type root. By chromatin immunoprecipitation-polymerase chain reaction and reverse transcription-polymerase chain reaction assays, we showed that SHR regulates cytokinin homeostasis by directly controlling the transcription of cytokinin oxidase 3, a cytokinin catabolism enzyme preferentially expressed in the stele. Finally, overexpression of a cytokinin oxidase in shr alleviated its vascular patterning defect. On the basis of these results, we suggest that one mechanism by which SHR controls vascular patterning is the regulation of cytokinin homeostasis.
Importin-β Directly Regulates the Motor Activity and Turnover of a Kinesin-4

Abstract:Spatiotemporal regulation of kinesins is essential for microtubule-dependent intracellular transport. In plants, cell wall deposition depends on the FRA1 kinesin, whose abundance and motility are tightly controlled to match cellular growth rate. Here, we show that an importin-β, IMB4, regulates FRA1 activity in a developmental manner. IMB4 physically interacts with a PY motif in the FRA1 motor domain and inhibits its motility by preventing microtubule binding, while also protecting FRA1 against proteasome-mediated degradation, thus providing a mechanism to couple the motility and stability of FRA1. This regulatory mechanism is likely to be broadly applicable, based on the conservation of the PY motif in the motor domains of plant and animal kinesins and the direct interaction of multiple plant kinesins with IMB4. Together, our data establish IMB4 as a multi-functional regulator of FRA1 and reveal a mechanism for how plants control the magnitude of cargo transport needed for cell wall assembly.

Abstract:Cytokinesis is a key process in the development of multicellular organisms, through both formative and proliferative divisions. In land plants, successful cytokinesis requires precise targeting of Golgi-derived vesicles to the future division plane by the phragmoplast, a cytoskeletal structure that undergoes continuous remodelling. Endomembrane trafficking and phragmoplast remodelling are tightly coupled, indicating the existence of active crosstalk. However, although many molecular regulators of membrane trafficking and cytoskeletal organisation are known, it remains poorly understood how these processes are co-ordinated. Here, we describe a regulatory module consisting of the membrane-associated GTPase RAB-A2a, cytoskeleton-associated Class II Kinesin-12s, and the Fused kinase orthologue TIO. We provide evidence that the interaction between these molecules at the midzone is essential for cytokinesis in Arabidopsis through simultaneously targeting to the midzone and coupling phragmoplast remodelling to vesicle delivery.

Abstract:Here we describe the establishment of microscope-based functional screening assays in intact cells that allow us to systematically identify new proteins involved in secretory membrane traffic, and proteins that can influence the integrity of the Golgi complex. We were able to identify 20 new proteins that affected either secretory transport, Golgi morphology, or both, when overexpressed in cells. Control experiments with human orthologs to yeast proteins with a role in membrane traffic, or already well characterized mammalian regulators of the secretory pathway, confirmed the specificity and significance of our results. Proteins localized to the Golgi complex or endoplasmic reticulum (ER) showed preferential interference in our assays. Bioinformatic analysis of the new proteins interfering with membrane traffic and/or Golgi integrity revealed broad functional variety, but demonstrated a bias towards proteins with predicted coiled-coil domains and repeat structures. Extending our approach to a much larger set of novel proteins in the future will be an important step toward a more comprehensive understanding of the molecular basis of the secretory pathway. It will also serve as an example for similar microscope-based screens addressing different biological questions.

Abstract:The plant SNF1-related kinase (SnRK1) is the a-subunit of the SnRK1 heterotrimeric compleses. Although SnRK1 is widely known as a key regulator of plant response to various physiological processes including nutrient-and energy-sensing, regulation of global metabolism, and control of cell cycle, development, as well as abiotics stress, less is known about the function of SnRK1 during pathogen infection. Our previous work has demonstrated that a tomato SNF1-related kinase (SlSnRK1) can interact with and phosphorylate beta C1, a pathogenesis protein encoded by tomato yellow leaf curl China betasatellite. Our results also showed that the plant SnRK1 can affect genimivirus infection in plant and reduce viral DNA accumulation. Phosphorylation of beta C1 protein negatively impacts its function as a pathogenicity determinant. Here we provide more information on interaction between beta C1 and SlSnRK1 and propose a mechanistic model for the SlSnRK1mediated defense responses against geminiviruses and the potential role of SnRK1 in plant resistance to geminivirus.

Abstract:In the year 2000, new international guidelines for cardiopulmonary resuscitation (CPR) were published by the American Heart Association, and the European Resuscitation Council. These guidelines are evidence-based, indicating that these recommendations are based primarily on interpretation of data from clinical studies. Levels of recommendation range from class I (proven safe and useful), class IIa (intervention of choice), IIb (alternative intervention), indeterminate (research stage), and class III (unacceptable, no benefit). Administration of drugs during CPR should be performed intravenously or intraosseously (class IIa) or, as a second-line approach, endotracheally (class IIb). Due to lack of evidence, the standard dose of 1 mg epinephrine to treat ventricular fibrillation, pulseless electrical activity, or asystole was categorized as class indeterminate; while a single dose of 40 units vasopressin to treat adults with shock-refractory ventricular fibrillation received a IIb recommendation. Owing to a lack of clinical data, the use of vasopressin was neither recommended to treat adults with pulseless electrical activity or asystole, nor for the use in children. Both endothelin and calcium were not recommended for routine use (class indeterminate). Careful titration of acid-base status with 1 mL/kg 8.4% sodium bicarbonate should only be administered if indicated by blood gas analysis (class indeterminate). If 1 mg epinephrine fails to be effective in adult patients with pulseless electrical activity or asystole, 1 mg atropine can be administered (class indeterminate). Regarding antiarrhythmic drugs, 300 mg amiodarone (class IIb) showed the best results in shock-refractory ventricular fibrillation. The postresuscitation phase has the goal to achieve the best possible neurological performance after return of spontaneous circulation, which requires careful optimization of organ functions.
KIPK and KIPK-LIKE1 suppress overbending during negative hypocotyl gravitropic growth

Abstract:Plants use environmental cues, such as the direction of gravity or the direction, quantity and quality of light, to orientate organ and plant growth. During germination of angiosperm seeds in the soil, hypocotyl elongation is directed by negative gravitropism responses such that the seedling can reach the light for photosynthesis and autotrophic growth. Hypocotyl elongation in the soil, however, also requires mechanisms to efficiently grow around obstacles such as soil particles. Here, we identify KIPK (KINESIN-LIKE CALMODULIN-BINDING PROTEIN INTERACTING PROTEIN KINASE) and the paralogous KIPKL1 (KIPK-LIKE1) as genetically redundant regulators of hypocotyl bending, in that KIPK and KIPKL1 are required to efficiently align hypocotyl growth with the gravity vector after obstacle avoidance. At the same time, we find that the highly homologous KIPKL2 (KIPK-LIKE2) must be functionally distinct. We further find that KIPK, and likely also KIPKL1, phosphorylate BRXL2 (BREVIS RADIX LIKE2) and ARKs (ARMADILLO REPEAT KINESINs), that mutants of both KIPK phosphorylation substrates share the overbending phenotype with kipk kipkl1 mutants, and that KIPK and KIPKL1 act synergistically with the ARK-regulatory NEK6 (NIMA-RELATED PROTEIN KINASE6). We propose that KIPK and KIPKL1 regulate ARK kinesins and thereby cortical microtubules for efficient gravitropic hypocotyl bending.

Microtubule Motor Proteins in the Eukaryotic Green Lineage: Functions and Regulation

Abstract:Kinesins and dyneins are two superfamilies of microtubule motor proteins that regulate many diverse fundamental cellular and developmental processes including cell shape, cell division and intracellular transport as well as spatial and temporal organization of molecules and organelles within the eukaryotic cells. These motor proteins use chemical energy from ATP to move unidirectionally using microtubules as roadways or to regulate microtubule dynamics. This review focuses on a comparative analysis of kinesins in eukaryotes, especially in the green lineage and their roles in plants. Comprehensive comparative analysis of kinesins among completed genome sequences of animal and several photosynthetic eukaryotes ranging from algae to monocots revealed considerable expansion of kinesins in flowering plants. Much of this expansion is due to an increase in members of two families (Kinesin-7 and Kinesin-14). Of the fourteen recognized families of kinesins in eukaryotes, members of four families are not found in flowering plants. However, a group of plant-specific kinesins does not fall into any of the recognized families, and some plant kinesins form plant-specific clades inside of their respective families. Some known domains are found exclusively either in plant and animal lineages, suggesting their functional specialization. Arabidopsis has the highest number of kinesins of any known multicellular eukaryotes, including humans, with a total of 61 kinesins. Although the processes regulated by many plant kinesins are yet to be discovered, functions of some kinesins have been elucidated in recent years using cell biological, molecular and genetic approaches and these are discussed briefly here. In addition, insights into regulatory mechanisms of a unique plant Ca2+/CaM-interacting motor called kinesin-like calmodulin-binding protein (KCBP) obtained through biochemical assays and crystal structure studies of its motor domain alone and as a complex with a calcium-binding protein are presented.
Kinesins and Myosins: Molecular Motors that Coordinate Cellular Functions in Plants

Abstract:Kinesins and myosins are motor proteins that can move actively along microtubules and actin filaments, respectively. Plants have evolved a unique set of motors that function as regulators and organizers of the cytoskeleton and as drivers of long-distance transport of various cellular components. Recent progress has established the full complement of motors encoded in plant genomes and has revealed valuable insights into the cellular functions of many kinesin and myosin isoforms. Interestingly, several of the motors were found to functionally connect the two cytoskeletal systems and thereby to coordinate their activities. In this review, we discuss the available genetic, cell biological, and biochemical data for each of the plant kinesin and myosin families from the context of their subcellular mechanism of action as well as their physiological function in the whole plant. We particularly emphasize work that illustrates mechanisms by which kinesins and myosins coordinate the activities of the cytoskeletal system.
A kinetochore-associated kinesin-7 motor cooperates with BUB3.3 to regulate mitotic chromosome congression in Arabidopsis thaliana

Abstract:Faithful genome partition during cell division relies on proper congression of chromosomes to the spindle equator before sister chromatid segregation. Here we uncover a kinesin-7 motor, kinetochore-associated kinesin 1 (KAK1), that is required for mitotic chromosome congression in Arabidopsis . KAK1 associates dynamically with kinetochores throughout mitosis. Loss of KAK1 results in severe defects in chromosome congression at metaphase, yet segregation errors at anaphase are rarely observed. KAK1 specifically interacts with the spindle assembly checkpoint protein BUB3.3 and both proteins show interdependent kinetochore localization. Chromosome misalignment in BUB3.3-depleted plants can be rescued by artificial tethering of KAK1 to kinetochores but not vice versa, demonstrating that KAK1 acts downstream of BUB3.3 to orchestrate microtubule-based chromosome transport at kinetochores. Moreover, we show that KAK1's motor activity is essential for driving chromosome congression to the metaphase plate. Thus, our findings reveal that plants have repurposed BUB3.3 to interface with a specialized kinesin adapted to integrate proper chromosome congression and checkpoint control through a distinct kinetochore design.

Abstract:Kinesin-1, also known as conventional kinesin, is widely used for microtubule plus-end-directed (anterograde) transport of various cargos in animal cells. However, a motor functionally equivalent to the conventional kinesin has not been identified in plants, which lack the kinesin-1 genes. Here we show that plant-specific armadillo repeat-containing kinesin (ARK) is the long sought-after versatile anterograde transporter in plants. In ARK mutants of the moss Physcomitrium patens, the anterograde motility of nuclei, chloroplasts, mitochondria and secretory vesicles was suppressed. Ectopic expression of non-motile or tail-deleted ARK did not restore organelle distribution. Another prominent macroscopic phenotype of ARK mutants was the suppression of cell tip growth. We showed that this defect was attributed to the mislocalization of actin regulators, including RopGEFs; expression and forced apical localization of RopGEF3 partially rescued the growth phenotype of the ARK mutant. The mutant phenotypes were partially rescued by ARK homologues in Arabidopsis thaliana, suggesting the conservation of ARK functions in plants.

Abstract:Symbioses between legumes and rhizobia require establishment of the plant-derived symbiosome membrane, which surrounds the rhizobia and accommodates the symbionts by providing an interface for nutrient and signal exchange. The host cytoskeleton and endomembrane trafficking systems play central roles in the formation of a functional symbiotic interface for rhizobia endosymbiosis; however, the underlying mechanisms remain largely unknown. Here we demonstrate that the nodulation-specific kinesin-like calmodulin-binding protein (nKCBP), a plant-specific microtubule-based kinesin motor, controls central vacuole morphogenesis in symbiotic cells in Medicago truncatula . Phylogenetic analysis further indicated that nKCBP duplication occurs solely in legumes of the clade that form symbiosomes. Knockout of nKCBP results in central vacuole deficiency, defective symbiosomes and abolished nitrogen fixation. nKCBP decorates linear particles along microtubules, and crosslinks microtubules with the actin cytoskeleton, to control central vacuole formation by modulating vacuolar vesicle fusion in symbiotic cells. Together, our findings reveal that rhizobia co-opted nKCBP to achieve symbiotic interface formation by regulating cytoskeletal assembly and central vacuole morphogenesis during nodule development.

Abstract:SNF1-related kinase (SnRK1) in plants belongs to a conserved family that includes sucrose non-fermenting 1 kinase (SNF1) in yeast and AMP-activated protein kinase (AMPK) in animals. These kinases play important roles in the regulation of cellular energy homeostasis and in response to stresses that deplete ATP, they inhibit energy consuming anabolic pathways and promote catabolism. Energy stress is sensed by increased AMP:ATP ratios and in plants, 5'-AMP inhibits inactivation of phosphorylated SnRK1 by phosphatase. In previous studies, we showed that geminivirus pathogenicity proteins interact with both SnRK1 and adenosine kinase (ADK), which phosphorylates adenosine to generate 5'-AMP. This suggested a relationship between SnRK1 and ADK, which we investigate in the studies described here. We demonstrate that SnRK1 and ADK physically associate in the cytoplasm, and that SnRK1 stimulates ADK in vitro by an unknown, non-enzymatic mechanism. Further, altering SnRK1 or ADK activity in transgenic plants altered the activity of the other kinase, providing evidence for in vivo linkage but also revealing that in vivo regulation of these activities is complex. This study establishes the existence of SnRK1-ADK complexes that may play important roles in energy homeostasis and cellular responses to biotic and abiotic stress.

Abstract:ABSTRACT Kinesins are microtubule-based motor proteins that play diverse cellular functions by regulating microtubule dynamics and intracellular transport in eukaryotes. The early branching kinetoplastid protozoan Trypanosoma brucei has an expanded repertoire of kinetoplastid-specific kinesins and orphan kinesins, many of which have unknown functions. We report here the identification of an orphan kinesin named KIN-G that plays an essential role in maintaining hook complex integrity and promoting Golgi biogenesis in T. brucei . KIN-G localizes to the distal portion of the centrin arm of the flagellum-associated hook complex through association with the centrin arm protein TbCentrin4. Knockdown of KIN-G in T. brucei disrupts the integrity of the hook complex by reducing the length of the centrin arm and eliminating the shank part of the hook complex, thereby impairing flagellum attachment zone elongation and flagellum positioning, which leads to unequal cytokinesis. KIN-G associates with Golgi through a centrin arm-localized Golgi peripheral protein named CAAP1, which maintains Golgi-centrin arm association to facilitate Golgi biogenesis. Knockdown of KIN-G impairs Golgi biogenesis by disrupting CAAP1 at the centrin arm, thereby impairing the maturation of centrin arm-associated Golgi. In vitro microtubule gliding assays demonstrate that KIN-G is a plus end-directed motor protein, and its motor activity is required for hook complex assembly and Golgi biogenesis. Together, these results identify a kinesin motor protein for promoting hook complex assembly and uncover a control mechanism for Golgi biogenesis through KIN-G-mediated maintenance of Golgi-hook complex association. IMPORTANCE Trypanosoma brucei has a motile flagellum, which controls cell motility, cell morphogenesis, cell division, and cell-cell communication, and a set of cytoskeletal structures, including the hook complex and the centrin arm, associates with the flagellum. Despite the essentiality of these flagellum-associated cytoskeletal structures, their mechanistic roles and the function of their associated proteins remain poorly understood. Here, we demonstrate that the orphan kinesin KIN-G functions to promote the biogenesis of the hook complex and the Golgi apparatus. KIN-G exerts this function by mediating the association between centrin arm and Golgi through the centrin arm protein TbCentrin4 and a novel Golgi scaffold protein named CAAP1, thereby bridging the two structures and maintaining their close association to facilitate the assembly of the two structures. These findings uncover the essential involvement of a kinesin motor protein in regulating the biogenesis of the hook complex and the Golgi in trypanosomes.

Trajectory Data Mining via Cluster Analyses for Tropical Cyclones That Affect the South China Sea

Abstract:The equal division of tropical cyclone (TC) trajectory method, the mass moment of the TC trajectory method, and the mixed regression model method are clustering algorithms that use space and shape information from complete TC trajectories. In this article, these three clustering algorithms were applied in a TC trajectory clustering analysis to identify the TCs that affected the South China Sea (SCS) from 1949 to 2014. According to their spatial position and shape similarity, these TC trajectories were classified into five trajectory classes, including three westward straight-line movement trajectory clusters and two northward re-curving trajectory clusters. These clusters show different characteristics in their genesis position, heading, landfall location, TC intensity, lifetime and seasonality distribution. The clustering results indicate that these algorithms have different characteristics. The equal division of the trajectory method provides better clustering result generally. The approach is simple and direct, and trajectories in the same class were consistent in shape and heading. The regression mixture model algorithm has a solid theoretical mathematical foundation, and it can maintain good spatial consistency among trajectories in the class. The mass moment of the trajectory method shows overall consistency with the equal division of the trajectory method.

Abstract:Plant cell fate is primarily the result of cell position. Cellular proteins, including transcription factors, can be transported from one cell into another through regulated pores (plasmodesmata) that connect adjacent cells (see the Perspective and associated animations by Jorgensen ) and may be one mechanism by which information about cell location is transmitted. Nakajima et al. determined that restricted expression of and the movement of the transcription factor shoot-root (SHR) was essential for proper cell fate specification and division in Arabidopsis thaliana roots. SHR transcripts are normally only expressed in the stele cells of the root (see Hake). Expression of a green fluorescent protein (GFP)-SHR fusion protein demonstrated that SHR protein appeared in the cells of the adjacent layer (quiescent center, the cortex-endodermis precursors, and the endodermis), where it was localized to the nucleus, supporting a role for SHR that is not cell autonomous. If SHR gene expression was forced by fusion to the SCARECROW ( SCR ) promoter in the cells of the adjacent layer, then there was an expansion of the root layers with an increase in the number of endodermal cells and quiescent center cells. SHR stimulates the transcription of SCR , thus SHR expressed from the SCR promoter establishes a positive feedback system. The ability of ectopically expressed SHR to stimulate increased cell numbers and endodermal cell fates in the adjacent layers was lost in SCR mutant plants, consistent with SHR acting through activation of SCR. Thus, restricted expression and subsequent movement into adjacent cells appears to be essential for proper cell fate specification by SHR. R. A.
