Abstract:The significance of protein coronas on the biological fates of nanoparticles has been widely recognized. Therefore, the alterations on biological effects caused by protein coronas need systemic study and interpretation in order to design novel safe and efficient nanomedicines. In the present study, we present a comprehensive quantitative analysis of the protein coronas on gold nanorods coated with various surface ligands of different chemical composition and charge. The design of surface ligands is the most importance for the functionization of nanoparticles and further the ligand-induced biological identity determines the fate of nanoparticles in human body. We found that the surface chemistry influences the composition of the protein corona more profoundly than surface charge. Since the first and most important challenge for administrated nanomedicines is navigating the interaction with macrophages, we further investigated how the surface chemistry-induced specific biomolecular corona affects the phagocytosis and immune responses of macrophages exposed to the corona-nanoparticle complexes. Our results reveal that the protein corona alters the internalization pathways of nanorods by macrophages via the interactions of the predominant coronal proteins with specific receptors on the cell membrane. The cytokine secretion profile of macrophages is also highly dependent on the adsorption pattern of the protein corona. The more abundant proteins involved in immune responses, such as acute phase, complement, and tissue leakage proteins, present in the acquired nanoparticle corona, the more macrophage IL-1β released is stimulated. The ligand-protein corona composition-immune response coefficient analysis may serve next-generation nanomedicines with high-efficiency and good-safety for better clinical translation.
Protein Corona Formation on Colloidal Polymeric Nanoparticles and Polymeric Nanogels: Impact on Cellular Uptake, Toxicity, Immunogenicity, and Drug Release Properties

Abstract:The adsorption of biomolecules to the surface of nanoparticles (NPs) following administration into biological environments is widely recognized. In particular, the "protein corona" is well understood in terms of formation kinetics and impact upon the biological interactions of NPs. Its presence is an essential consideration in the design of therapeutic NPs. In the present study, the protein coronas of six polymeric nanoparticles of prospective therapeutic use were investigated. These included three colloidal NPs-soft core-multishell (CMS) NPs, plus solid cationic Eudragit RS (EGRS), and anionic ethyl cellulose (EC) nanoparticles-and three nanogels (NGs)-thermoresponsive dendritic-polyglycerol (dPG) nanogels (NGs) and two amino-functionalized dPG-NGs. Following incubation with human plasma, protein coronas were characterized and their biological interactions compared with pristine NPs. All NPs demonstrated protein adsorption and increased hydrodynamic diameters, although the solid EGRS and EC NPs bound notably more protein than the other tested particles. Shifts toward moderately negative surface charges were also observed for all corona bearing NPs, despite varied zeta potentials in their pristine states. While the uptake and cellular adhesion of the colloidal NPs in primary human keratinocytes and human umbilical vein endothelial cells were significantly decreased when bearing the protein corona, no obvious impact was seen in the NGs. By contrast, corona bearing NGs induced marked increases in cytokine release from primary human macrophages not seen with corona bearing colloidal NPs. Despite this, no apparent enhancement to in vitro toxicity was noted. Finally, drug release from EGRS and EC NPs was assessed, where a decrease was seen in the EGRS NPs alone. Together these results provide a direct comparison of the physical and biological impact the protein corona has on NPs of widely varied character and in particular highlights a distinction between the corona's effects on NGs and colloidal NPs.
Protein corona modulation of hepatocyte uptake and molecular mechanisms of gold nanoparticle toxicity

Abstract:Protein corona formation over gold nanoparticles (AuNP) can modulate cellular responses by altering AuNP physicochemical properties. The liver plays an essential role in metabolism, detoxification and elimination of xenobiotics and drugs as well as circulating NP clearance. We investigated human hepatic uptake of 40 and 80 nm AuNP with branched polyethylenimine (BPEI), lipoic acid (LA) and polyethylene glycol (PEG) coatings as well as human plasma protein (HP) and human serum albumin (HSA) coronas. AuNP-mediated cytotoxicity, reactive oxygen/reactive nitrogen species (ROS/RNS), and CYP activity in human hepatocytes as well as molecular mechanisms with 40 nm bare and HP BPEI-AuNP were investigated. Time-dependent increase in uptake occurred for all bare AuNP but HP and HSA decreased uptake except for 40 nm HP PEG-AuNP. BPEI-AuNP showed time-and concentration-dependent increase in ROS/RNS which correlated with cytotoxicity at 24 h. HP corona substantially reduced ROS/RNS. The 40 and 80 nm bare, HP or HSA LA- and PEG-AuNP were not toxic but HP was as cytotoxic as bare BPEI-AuNP. All bare and HP BPEI-AuNP, except for HP 80 nm BPEI-AuNP toward CYP1A2, inhibited CYP1A2, CYP2C9 and CYP3A4 activity. Transcriptional profiling was dose-dependent with 40 nm bare BPEI-AuNP (1.9% genes at IC10 and 18.9% at LC50) and HP (23.5% at LC50). Differentially expressed genes at LC50 were mainly involved in phase I metabolism and phospholipidosis pathways. Cytotoxicity of bare BPEI-AuNP caused an upregulation of antioxidant and pro-apoptotic genes. These studies contribute to a better understanding of the dramatic effect of protein coronas (PC) on AuNP cellular uptake, cytotoxicity and their underlying molecular mechanisms of action.

Abstract:With the rapid developments of nanotechnology, chances of exposing nanoscale particles to humans (e.g., workers and consumers) also increase correspondingly, which raises serious concerns on their biosafety. Entrance of nanoparticles into diverse biological environment endows them with new and dynamic biological identities as the so-called nanoparticle–protein corona. Therefore, understanding the role of these nanoparticle–protein coronas and resulting biological responses is crucial, as it helps to clarify the biological mechanism and prevent the potential adverse effects of nanoparticles. In this review, we summarize the latest developments relating to the nanoparticle–protein interaction and corresponding biological responses, with an emphasis on the characterization methods, induced biological effects and possible molecular mechanisms. In addition, we overview both the challenges and opportunities (particularly in nanomedicine) raised by this entrance of nanoparticles into the living creatures, especially human beings, with some future perspectives based on our understanding.

Abstract:Regarding the importance of the biological effects of nanomaterials, there is still limited knowledge about the binding structure and stability of the protein corona on nanomaterials and the subsequent impacts. Here we designed a hard serum albumin protein corona (BSA) on CTAB-coated gold nanorods (AuNRs) and captured the structure of protein adsorption using synchrotron radiation X-ray absorption spectroscopy, microbeam X-ray fluorescent spectroscopy, and circular dichroism in combination with molecular dynamics simulations. The protein adsorption is attributed to at least 12 Au-S bonds and the stable corona reduced the cytotoxicity of CTAB/AuNRs. These combined strategies using physical, chemical, and biological approaches will improve our understanding of the protective effects of protein coronas against the toxicity of nanomaterials. These findings have shed light on a new strategy for studying interactions between proteins and nanomaterials, and this information will help further guide the rational design of nanomaterials for safe and effective biomedical applications.

Abstract:Nanoparticles in a biological fluid (plasma, or otherwise) associate with a range of biopolymers, especially proteins, organized into the “protein corona” that is associated with the nanoparticle and continuously exchanging with the proteins in the environment. Methodologies to determine the corona and to understand its dependence on nanomaterial properties are likely to become important in bionanoscience. Here, we study the long-lived (“hard”) protein corona formed from human plasma for a range of nanoparticles that differ in surface properties and size. Six different polystyrene nanoparticles were studied: three different surface chemistries (plain PS, carboxyl-modified, and amine-modified) and two sizes of each (50 and 100 nm), enabling us to perform systematic studies of the effect of surface properties and size on the detailed protein coronas. Proteins in the corona that are conserved and unique across the nanoparticle types were identified and classified according to the protein functional properties. Remarkably, both size and surface properties were found to play a very significant role in determining the nanoparticle coronas on the different particles of identical materials. We comment on the future need for scientific understanding, characterization, and possibly some additional emphasis on standards for the surfaces of nanoparticles.

Abstract:Abstract To study the influence of colloidal stability on protein corona formation, gold nanoparticles are synthesized with five distinct surface modifications: coating with citric acid, bis( p ‐sulfonatophenyl)phenylphosphine dihydrate dipotassium salt, thiol‐terminated methoxy‐polyethylene glycol, dodecylamine‐grafted poly(isobutylene‐ alt ‐maleic anhydride), and dodecylamine‐grafted poly(isobutylene‐ alt ‐maleic anhydride) conjugated with polyethylene glycol. The nanoparticles are incubated with serum or bronchoalveolar lavage fluid from C57BL/6 mice (15 min or 24 h) to assess the effect of differential nanoparticle surface presentation on protein corona formation in the air–blood barrier exposure pathway. Proteomic quantification and nanoparticle size measurements are used to assess protein corona formation. We show that surface modification has a clear effect on the size and the composition of the protein corona that is related to the colloidal stability of the studied nanoparticles. Additionally, differences in the composition and size of the protein corona are shown between biological media and duration of exposure, indicating evolution of the corona through this exposure pathway. Consequently, a major determinant of protein corona formation is the colloidal stability of nanoparticles in biological media and chemical or environmental modification of the nanoparticles alters the surface presentation of the functional epitope in vivo. Therefore, the colloidal stability of nanoparticles has a decisive influence on nano–bio interactions.

Abstract:Nanoparticles (NPs) that are exposed to blood are coated with an assortment of proteins that establish their biological identity by forming the interface between the NP and the cells and tissues of the body. The biological relevance of this protein corona is often overlooked during toxicological assessments of NPs. However, accurate interpretation of biological outcomes following exposure to NPs, including activation of coagulation, opsonization of pathogens, and cellular phagocytosis, must take this adsorbed proteome into account. In this study, we examined protein coronas on the surface of five poly(acrylic acid) (PAA) metal-oxide NPs (TiO2, CeO2, Fe2O3, ZnO, and PAA-capsules) following exposure to human plasma for key markers of various host response pathways, including humoral immunity and coagulation. We also evaluated the impacts of pre-exposing serum proteins to PAA-NPs on the opsonization and phagocytosis of bacteria by two immune cell lines. Results demonstrated that each PAA-NP type adsorbed a unique profile of blood proteins and that protein-coated PAA-NPs significantly inhibited human plasma coagulation with PAA-zinc oxide NPs and their associated proteome fully abrogating clotting. Protein-coated PAA-NPs also resulted in a 50% increase in phagocytic activity of RBL-2H3 cells and a 12.5% increase in phagocytic activity in the RAW 264.7 cell line. We also identified numerous structural, coagulation, and immune-activating proteins in the adsorbed protein corona, which resulted in altered biological function. Overall, our findings demonstrate that the formation of protein coronas on the surface of NPs plays an important role in directing the biological outcomes of opsonization, cell phagocytosis, and blood coagulation.

Abstract:Neuronanomedicine is an emerging field bridging the gap between neuromedicine and nanoparticle-based drug delivery systems. Despite promise, clinical translation of neuronanomedicines remains elusive, possibly due to a dearth of information regarding the effect of the protein corona on these neuronanomedicines. The protein corona, a layer of proteins adsorbed to nanoparticles following exposure to biological fluids, ultimately determines the fate of nanoparticles in biological systems, dictating nanoparticle-cell interactions. To date, few studies have investigated the effect of the protein corona on interactions with brain-derived cells, an important consideration for the development of neuronanomedicines. Here, two polymeric nanoparticles, PLGA and PLGA-PEG, were used to obtain serum-derived protein coronas. Protein corona characterization and LC-MS analysis revealed distinct differences in biophysical properties and protein composition. PLGA protein coronas contained high abundance of globins (60%) and apolipoproteins (21%), while PLGA-PEG protein coronas contained fewer globins (42%) and high abundance of protease inhibitors (28%). Corona coated PLGA nanoparticles were readily internalized into microglia and neuronal cells, but not into astrocytes. Internalization of nanoparticles was associated with pro-inflammatory cytokine release and decreased neuronal cell viability, however, viability was rescued in cells treated with corona coated nanoparticles. These results showcase the importance of the protein corona in mediating nanoparticle-cell interactions.

Abstract:When inhaled nanoparticles (NPs) come into the deep lung, they develop a biomolecular corona by interacting with the pulmonary surfactant. The adsorption of the phospholipids and proteins gives a new biological identity to the NPs, which may alter their subsequent interactions with cells and other biological entities. Investigations of the interaction between the cell membrane and NPs coated with such a biomolecular corona are important in understanding the role of the biofluids on cellular uptake and estimating the dosing capacity and the nanotoxicology of NPs. In this paper, using dissipative particle dynamics, we investigate how the physicochemical properties of the coating pulmonary surfactant lipids and proteins affect the membrane response for inhaled NPs. We pinpoint several key factors in the endocytosis of lipid NPs, including the deformation of the coating lipids, coating lipid density, and ligand-receptor binding strength. Further studies reveal that the deformation of the coating lipids consumes energy but on the other hand promotes the coating ligands to bind with receptors more tightly. The coating lipid density controls the amount of the ligands as well as the hydrophobicity of the lipid NPs, thus affecting the endocytosis kinetics through the specific and nonspecific interactions. It is also found that the hydrophobic surfactant proteins associated with lipids can accelerate the endocytosis process of the NPs, but the endocytosis efficiency mainly depends on the density of the coating surfactant lipids. These findings can help understand how the pulmonary surfactant alters the biocompatibility of the inhaled NPs and provide some guidelines in designing an NP complex for efficient pulmonary drug delivery.

Abstract:The development of nanotechnology will inevitably lead to the release of consistent amounts of nanoparticles (NPs) in the environment. Invertebrates, that represent >90% of animal species, widespread in different ecosystems, are emerging both as suitable target organisms and as models for evaluating the environmental impact of NPs. Once released in different ecosystems, both NP intrinsic properties and those of the receiving medium will affect particle behavior. In particular, interactions with different biomolecules will lead to the formation of ‘eco-coronas’, that will influence NP bioavailability/uptake/toxicity in different environments, depending on the organisms present and the surrounding conditions. However, as shown in mammalian studies, the evaluation of the biological effects of NPs requires additional understanding of how, once within the organism, NPs interact at the molecular level with cells in a physiological environment, i.e. in biological fluids. Different types of NPs associate with serum soluble components, organized into a ‘protein corona’, which confers a biological identity to NPs and affects their interactions with target cells. In comparison, the study on NP-protein coronas in invertebrates is still at its infancy, since the protein composition of their extracellular fluids is largely unknown, given the large diversity of phyla and species. From the first demonstration of an AgNP protein corona in the body fluid of earthworms, NP-protein coronas are being characterized in marine invertebrates. The identification of protein coronas formed with different types of NPs (amino modified polystyrene- PS NH2 and nano-oxides, n-CeO2 n-TiO2) in hemolymph serum of the marine bivalve Mytilus galloprovincialis is presented as a case study. The results indicate that, in Mytilus hemolymph, the formation of a biomolecular corona is partly NP-specific. The results obtained so far in terrestrial and marine invertebrates indicate that in each model, endowed with a peculiar protein repertoire, NP-coronas are characterized by unique protein components. The role of NP-protein coronas formed by lower organisms, and their possible contribution in evaluating the environmental impact of NPs are discussed.

Abstract:Neuronanomedicine harnesses nanoparticle technology for the treatment of neurological disorders. An unavoidable consequence of nanoparticle delivery to biological systems is the formation of a protein corona on the nanoparticle surface. Despite the well-established influence of the protein corona on nanoparticle behavior and fate, as well as FDA approval of neuro-targeted nanotherapeutics, the effect of a physiologically relevant protein corona on nanoparticle-brain cell interactions is insufficiently explored. Indeed, less than 1 percent of protein corona studies have investigated protein coronas formed in cerebrospinal fluid (CSF) the fluid surrounding the brain. Herein we utilize two clinically relevant polymeric nanoparticles (PLGA and PLGA-PEG) to evaluate the formation of serum and CSF protein coronas. LCMS analysis revealed distinct protein compositions with selective enrichment/depletion profiles. Following incubation with brain cells, serum and CSF coronas on PLGA particles showed enhanced associations with all cell types as compared to their corresponding corona on PLGAPEG particles. CSF-derived protein coronas on PLGA nanoparticles specifically showed the greatest nanoparticle-brain cell interactions with Pearsons correlation analysis revealing that proteins associated with enhanced nanoparticle-cell interactions were exclusively enriched in this protein corona. This study demonstrates the importance of correct choice of physiologically relevant biological fluids and its influence on the formation of the protein corona subsequent nanoparticle-cell interactions.

Abstract:When a pristine nanoparticle (NP) encounters a biological fluid, biomolecules spontaneously form adsorption layers around the NP, called "protein corona". The corona composition depends on the time-dependent environmental conditions and determines the NP's fate within living organisms. Understanding how the corona evolves is fundamental in nanotoxicology as well as medical applications. However, the process of corona formation is challenging due to the large number of molecules involved and to the large span of relevant time scales ranging from 100 μs, hard to probe in experiments, to hours, out of reach of all-atoms simulations. Here we combine experiments, simulations, and theory to study (i) the corona kinetics (over 10-3-103 s) and (ii) its final composition for silica NPs in a model plasma made of three blood proteins (human serum albumin, transferrin, and fibrinogen). When computer simulations are calibrated by experimental protein-NP binding affinities measured in single-protein solutions, the theoretical model correctly reproduces competitive protein replacement as proven by independent experiments. When we change the order of administration of the three proteins, we observe a memory effect in the final corona composition that we can explain within our model. Our combined experimental and computational approach is a step toward the development of systematic prediction and control of protein-NP corona composition based on a hierarchy of equilibrium protein binding constants.

Abstract:Protein coronas provide the biological identity of nanomaterials in vivo . Here we have used dynamic light scattering (DLS) and transmission electron microscopy (TEM) to investigate the adsorption of serum proteins, including bovine serum albumin (BSA), transferrin (TRF) and fibrinogen (FIB), on gold nanoparticles (AuNPs) with different surface modifications (citrate, thioglycolic acid, cysteine, polyethylene glycol (PEG, M w = 2 k and 5 k)). AuNPs with PEG(5 k) surface modification showed no protein adsorption. AuNPs with non-PEG surface modifications showed aggregation with FIB. AuNPs with citrate and thioglycolic acid surface modifications showed 6–8 nm thick BSA and TRF coronas (corresponding to monolayer or bilayer proteins), in which the microscopic dissociation constants of BSA and TRF protein coronas are in the range of 10 −8 to 10 −6 M.

Abstract:Understanding nanoparticle-protein interactions is a crucial issue in the development of targeted nanomaterial delivery. Besides unraveling the composition of the nanoparticle's protein coronas, distinct proteins thereof could control nanoparticle uptake into specific cell types. Here we differentially analyzed the protein corona composition on four polymeric differently functionalized nanoparticles by label-free quantitative mass spectrometry. Next, we correlated the relative abundance of identified proteins in the corona with enhanced or decreased cellular uptake of nanoparticles into human cancer and bone marrow stem cells to identify key candidates. Finally, we verified these candidate proteins by artificially decorating nanoparticles with individual proteins showing that nanoparticles precoated with the apolipoproteins ApoA4 or ApoC3 significantly decreased the cellular uptake, whereas precoating with ApoH increased the cellular uptake.

Abstract:As nanobiotechnologies have become more ubiquitously used in biotechnology, characterizing the protein corona has been critical to predict their in vivo outcomes. In this paper, the current protein corona literature is analyzed and several key advances and shortcomings are unearthed which are opined important to bring to light on various factors that affect the decoration of protein corona at the surface of nanoparticles. The protein corona forms spontaneously on nanoparticle surfaces when nanomaterials are introduced into any biological system/fluid. Reliable characterization of the protein corona is, therefore, a vital step in the development of safe and efficient diagnostic and therapeutic nanomedicine products. 2134 published manuscripts on the protein corona are reviewed and a down‐selection of 470 papers spanning 2000–2021, comprising 1702 nanoparticle (NP) systems is analyzed. This analysis reveals: i) most corona studies have been conducted on metal and metal oxide nanoparticles; ii) despite their overwhelming presence in clinical practice, lipid‐based NPs are underrepresented in protein corona research, iii) studies use new methods to improve reliability and reproducibility in protein corona research; iv) studies use more specific protein sources toward personalized medicine; and v) careful characterization of nanoparticles after corona formation is imperative to minimize the role of aggregation and protein contamination on corona outcomes. As nanoparticles used in biomedicine become increasingly prevalent and biochemically complex, the field of protein corona research will need to focus on developing analytical approaches and characterization techniques appropriate for each unique nanoparticle formulation. Achieving such characterization of the nano‐bio interface of nanobiotechnologies will enable more seamless development and safe implementation of nanoparticles in medicine.

Same Genes, Different Brains: Neuroanatomical Differences Between Monozygotic Twins Discordant for Musical Training

Abstract:Numerous cross-sectional and observational longitudinal studies show associations between expertise and regional brain anatomy. However, since these designs confound training with genetic predisposition, the causal role of training remains unclear. Here, we use a discordant monozygotic (identical) twin design to study expertise-dependent effects on neuroanatomy using musical training as model behavior, while essentially controlling for genetic factors and shared environment of upbringing. From a larger cohort of monozygotic twins, we were able to recruit 18 individuals (9 pairs) that were highly discordant for piano practice. We used structural and diffusion magnetic resonance imaging to analyze the auditory-motor network and within-pair differences in cortical thickness, cerebellar regional volumes and white-matter microstructure/fractional anisotropy. The analyses revealed that the musically active twins had greater cortical thickness in the auditory-motor network of the left hemisphere and more developed white matter microstructure in relevant tracts in both hemispheres and the corpus callosum. Furthermore, the volume of gray matter in the left cerebellar region of interest comprising lobules I-IV + V, was greater in the playing group. These findings provide the first clear support for that a significant portion of the differences in brain anatomy between experts and nonexperts depend on causal effects of training.

Abstract:Nanoparticles (NPs) interact with biological fluids after being injected into the bloodstream. The interactions between NPs and plasma proteins at the nano-bio interface affect their biopharmaceutical properties and distribution in the organ and tissues due to protein corona (PrC) composition, and in turn, modification of the resulting targeting capability. Moreover, lipid and polymer NPs, at their interface, affect the composition of PrC and the relative adsorption and abundance of specific proteins. To investigate this latter aspect, we synthesized and characterized different liposomal formulations (LFs) with lipids and polymer-conjugated lipids at different molar ratios, having different sizes, size distributions and surface charges. The PrC composition of various designed LFs was evaluated ex vivo in human plasma by label-free quantitative proteomics. We also correlated the relative abundance of identified specific proteins in the coronas of the different LFs with their physicochemical properties (size, PDI, zeta potential). The evaluation of outputs from different bioinformatic tools discovered protein clusters allowing to highlight: (i) common as well as the unique species for the various formulations; (ii) correlation between each identified PrC and the physicochemical properties of LFs; (iii) some preferential binding determined by physicochemical properties of LFs; (iv) occurrence of formulation-specific protein patterns in PrC. Investigating specific clusters in PrC will help decode the multivalent roles of the protein pattern components in the drug delivery process, taking advantage of the bio-nanoscale recognition and identification for significant advances in nanomedicine.

Abstract:In most cases, once nanoparticles (NPs) enter the blood, their surface is covered by biological molecules, especially proteins, forming a so-called protein corona (PC). As a result, what the cells of the body "see" is not the NPs as formulated by the chemists, but the PC. In this way, the PC can influence the effects of the NPs and even mask the desired effects of the NP components. While this can argue for trying to inhibit protein-nanomaterial interactions, encapsulating NPs in an endogenous PC may increase their clinical usefulness. In this review, we briefly introduce the concept of the PC, its formation and its effects on the behavior of NPs. We also discuss how to reduce the formation of PCs or exploit them to enhance NP functions. Studying the interactions between proteins and NPs will provide insights into their clinical activity in health and disease.<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Statement of Significance</h3>: The formation of protein corona (PC) will affect the operation of nanoparticles (NPs) in vivo. Since there are many proteins in the blood, it is impossible to completely overcome the formation of PC. Therefore, the use of PCs to deliver drug is the best choice. De-opsonins adsorbed on NPs can reduce macrophage phagocytosis and cytotoxicity of NPs, and prolong their circulation in blood. Albumin, apolipoprotein and transferrin are typical de-opsonins. In present review, we mainly discuss how to optimize the delivery of nanoparticles through the formation of albumin corona, transferrin corona and apolipoprotein corona in vivo or in vitro.

Abstract:Nanoparticle (NP)-protein complexes exhibit the correct identity of NP in biological media. Therefore, protein-NP interactions should be closely explored to understand and to modulate the nature of NPs in medical implementations. This review focuses mainly on the physicochemical parameters such as dimension, surface chemistry, the morphology of NPs and influence of medium pH on the formation of protein corona and conformational changes of adsorbed proteins by different kinds of methods. Also, the impact of protein corona on the colloidal stability of NPs is discussed. Uncontrolled protein attachment on NPs may bring unwanted impacts such as protein denaturation and aggregation. In contrast, controlled protein adsorption by optimal concentration, size, pH and surface modification of NPs may result in potential implementation of NPs as therapeutic agents especially for disaggregation of amyloid fibrils. Also, the effect of NPs-protein corona on reducing the cytotoxicity and clinical implications such as drug delivery, cancer therapy, imaging and diagnosis will be discussed. Validated correlative physicochemical parameters for NP-protein corona formation frequently derived from protein corona fingerprints of NPs which are more valid than the parameters obtained only on the base of NP features. This review may provide useful information regarding the potency as well as the adverse effects of NPs to predict their behavior in the in vivo experiments.
