Abstract:The chemical senses and pharmaceuticals fundamentally depend on similar biological processes, but novel molecule discovery has classically been approached from vastly different vantage points. From the perspective of ingredient and flavor companies, there are countless ingredients that act via largely unknown mechanisms, whereas the pharmaceutical industry has numerous mechanisms in search of novel compounds. Mixtures of agonists can result in synergistic (superadditive) responses, which can be quantified via isobole analysis, a well-proven clinical approach in pharmacology. For the food and beverage industries, bulk (caloric) sweeteners like sugars are a key ingredient in sweetened foods and beverages, but consumers also desire products with fewer calories, which has led to the development of sweet enhancers and sweetener blends intended to achieve synergy or superadditivity. Synergistic mixtures are highly attractive targets commercially as they enable lower usage levels and enhanced efficacy. Although the psychophysical literature contains numerous prior reports of sweetener synergy, others have also noted that classical additive models fail to account for nonlinear dose-response functions. To address this shortcoming, here we systematically apply the isobole method from pharmacology to quantify the presence or absence of psychophysical synergy for binary pairs of sweeteners in a series of 15 separate experiments, each with ~100 adult volunteers (total n = 1576). Generally, these data support the hypothesis that structurally similar sweeteners acting as agonists will not synergize, whereas structurally dissimilar sweeteners binding to overlapping or distal sites can act as allosteric agonists or agonist-antagonists, respectively.

Abstract:This study investigated the effects of various binary sweetener mixtures on sweetness enhancement and their interactions with sweet or bitter taste receptors, focusing on sensory perception and receptor activity. Acesulfame K or saccharin was mixed with allulose, aspartame, erythritol, fructose, glucose, or sucrose to match a target sucrose sweetness. The effects of the mixtures on sweet and bitter taste receptors (in the human embryonic kidney -293 cells) and sensory taste intensities were evaluated. Sweetness enhancement at the sweet taste receptor level was observed in some cases, with several monosaccharides reducing the acesulfame K- or saccharin-induced bitter taste receptor activity. Combining acesulfame K or saccharin with any of the six sweeteners perceptually enhanced sweetness (60% ∼ 100% in 50:50 ratio), correlating with a reduction in inherent bitterness (-35% ∼ -63% in 50:50 ratio). This finding suggests that sweetness perception likely increased because the monosaccharides mitigate the activation of bitter receptors caused by high-potency sweeteners.

Abstract:The sweet taste is elicited by activation of the TAS1R2/1R3 heterodimer G protein-coupled receptor. This is a therapeutic target for treatment of obesity and metabolic dysfunctions. Sweetener blends provide attractive strategies to lower the sugar level while preserving the attractive taste of food. To understand the synergic effect of various sweetener blend combinations of artificial and natural sweeteners, we carried out our molecular dynamics studies using predicted structures of the TAS1R2/1R3 heterodimer and predicted structures for the sweeteners. We used as a measure of activation the intracellular ionic lock distance between transmembrane helices 3 and 6 of TAS1R3. We find that full synergic combinations [rebaudioside A (Reb-A)/acesulfame K and Reb-A/sucralose] and partial synergic combinations (sucralose/acesulfame K) show significantly more negative changes in the free energy compared to single-ligand cases, while a pair known to be suppressive (saccharin and acesulfame K) shows significantly less changes than for the single-ligand case. This study provides an atomistic understanding of the mechanism for synergy and identifies new combinations of sweeteners to reduce the caloric content for treating diseases.

Abstract:Bitter taste perception is initiated by TAS2R receptors, which respond to agonists by triggering depolarization of taste bud cells. Mutations in TAS2Rs are known to affect taste phenotypes by altering receptor function. Evidence that TAS2Rs overlap in ligand specificity suggests that they may also contribute joint effects. To explore this aspect of gustation, we examined bitter perception of saccharin and acesulfame K, widely used artificial sweeteners with aversive aftertastes. Both substances are agonists of TAS2R31 and -43, which belong to a five-member subfamily (TAS2R30-46) responsive to a diverse constellation of compounds. We analyzed sequence variation and linkage structure in the ∼140 kb genomic region encoding TAS2R30-46, taste responses to the two sweeteners in subjects, and functional characteristics of receptor alleles. Whole-gene sequences from TAS2R30-46 in 60 Caucasian subjects revealed extensive diversity including 34 missense mutations, two nonsense mutations and high-frequency copy-number variants. Thirty markers, including non-synonymous variants in all five genes, were associated (P< 0.001) with responses to saccharin and acesulfame K. However, linkage disequilibrium (LD) in the region was high (D', r(2) > 0.95). Haplotype analyses revealed that most associations were spurious, arising from LD with variants in TAS2R31. In vitro assays confirmed the functional importance of four TAS2R31 mutations, which had independent effects on receptor response. The existence of high LD spanning functionally distinct TAS2R loci predicts that bitter taste responses to many compounds will be strongly correlated even when they are mediated by different genes. Integrative approaches combining phenotypic, genetic and functional analysis will be essential in dissecting these complex relationships.

Abstract:In our previous study, phloridzin, sucrose, l-alanine, and dulcitol presented synergistic effects in Camellia nanchuanica black tea (NCBT). This study aims to verify the synergistic effects of the aforementioned sweet taste compounds and the mechanism involved. By conducting σ-τ plot analysis, phloridzin at the recognition threshold concentration (phl) exhibited synergistic effects with different concentrations of sucrose (Lsuc-6suc). Various concentrations of sucrose, phloridzin, and their combinations were selected to investigate the impact on sweet taste receptor cells. The results revealed that sucrose/phloridzin significantly increased the calcium signal compared to phloridzin and sucrose alone, attributed to the greater stability of the sucrose/phloridzin combination when binding to Taste 1 Receptor Member 3 (TAS1R3; one subunit of sweet taste receptor proteins). Ultimately, the sweet taste signal of sucrose/phloridzin was transmitted to the brain, triggering the activation of more brain regions associated with sweet taste perception (right insular, postcentral, and amygdala).

Abstract:Sweeteners are commonly used as food additives in our daily life, which, however, have been causing a number of undesirable diseases since the last century. Therefore, the detection and quantification of sweeteners are of great value for food safety. In this study, we used a taste biosensor to measure and analyze different sweeteners, both natural and artificial sweeteners included. Electrophysiological activities from taste epithelium were detected by the multi-channel biosensors and analyzed with spatiotemporal methods. The longtime signal result showed different temporal-frequency properties with stimulations of individual sweeteners such as glucose, sucrose, saccharin, and cyclamate, while the multi-channel results in our study revealed the spatial expression of taste epithelium to sweet stimuli. Furthermore, in the analysis of sweetener with different concentrations, the result showed obvious dose-dependent increases in signal responses of the taste epithelium, which indicated promising applications in sweetness evaluation. Besides, the mixture experiment of two natural sweeteners with a similar functional unit (glucose and sucrose) presented two signal patterns, which turned out to be similar with responses of each individual stimulus involved. The biosensor analysis of common sweeteners provided new approaches for both natural and artificial sweeteners evaluation.
An in Vivo Bioelectronic Tongue for Sweetness Detection Using Rat Gustatory Perception Based on Brain-Computer Interface

Abstract:The detection of sweetness is of great significance due to flavor and health concerns. In this study, an in vivo bioelectronic tongue for sweetness detection is constructed using rat gustatory system and implantable microelectrode array. By recording extracellular potentials in “sweet hotspot” within rat gustatory cortex, sweetener-evoked response patterns are generated by neural decoding algorithms. The results show that sweetener stimulation can cause a significant increase in the power spectral density of the local field potentials signal at 50-80 Hz, and its dynamic response in the time domain can be used for identification and classification of sweetness. Meanwhile, quantitative study shows that power of response signal is dose-dependent and the detection range covers sugar contents of daily beverages. Comparisons with results of human taste panelists also confirm that this in vivo bioelectronic tongue can work across species and has the potential to mimic human taste and evaluate sweetness in practical applications.
Sensory sweetness and sourness interactive response of sucrose-citric acid mixture based on synergy and antagonism

Abstract:The clarity of taste sensation interaction is a key basis for promoting the food sensory science research and its application to the beverage and food additive industries. This study explored the synergy and antagonism effect of sucrose-citric acid mixture and established an optimized method to determine the human sweetness and sourness interactive response. Sucrose-citric acid mixtures were evaluated by the "close type" question. According to the sensory difference strength curves and Weber-Fechner law, citric acid increased the sucrose's absolute threshold (0.424-0.624%) and weber fraction (20.5-33.0%). Meanwhile, sucrose increased citric acid's absolute threshold (0.0057-0.0082%) and decreased its weber fraction (17.96-9.53%). By fitting absolute threshold and weber fraction variation equations, the sweet-sour taste sensory strength variation models (SSTVM) were derived, which could be used to explain the synergy and antagonism effect of sweet-sour taste. According to the SSTVM, the interactive response to sweet-sour taste could be quantitatively calculated. The high coincidence between SSTVM and human evaluation (1.02% of relative error) indicated that it could be applied in the food industry, health management, and intelligent sensory science.

Abstract:The sweetness of sugar is regarded by humans as the optimal sensation; that of alternative non-sugar sweeteners possesses an inferior sweet quality, which limits their use in low-calorie foods. These sweeteners may produce bitter, metallic or cooling sensations, as well as lingering sweet aftertaste. The molecular basis for these undesirable sensations is poorly understood. Although various sweeteners may stimulate the same G-protein-coupled taste receptors (GPCRs), inferior sweet-taste quality is uniquely related to some non-sugar sweeteners. This chapter presents data indicating that such tastants may rapidly permeate taste cells under physiological conditions in vivo and interact with downstream signaling components such as signal-termination-related kinases in vitro. Abstract:A microfluidic tongue-on-a-chip platform has been evaluated relative to the known sensory properties of various sweeteners. Analogous metrics of typical sensory features reported by human panels such as sweet taste thresholds, onset, and lingering, as well as bitter off-flavor and blocking interactions were deduced from the taste receptor activation curves and then compared. To this end, a flow cell containing a receptor cell array bearing the sweet and six bitter taste receptors was transiently exposed to pure and mixed sweetener samples. The sample concentration gradient across time was separately characterized by the injection of fluorescein dye. Subsequently, cellular calcium responses to different doses of advantame, aspartame, saccharine, and sucrose were overlaid with the concentration gradient. Parameters describing the response kinetics compared to the gradient were quantified. Advantame at 15 μM recorded a significantly faster sweetness onset of 5 ± 2 s and a longer lingering time of 39 s relative to sucrose at 100 mM with an onset of 13 ± 2 s and a lingering time of 6 s. Saccharine was shown to activate the bitter receptors TAS2R8, TAS2R31, and TAS2R43, confirming its known off-flavor, whereas addition of cyclamate reduced or blocked this saccharine bitter response. The potential of using this tongue-on-a-chip to bridge the gap with in vitro assays and taste panels is discussed.

Abstract:The past decade has witnessed tremendous progress achieved in taste research, while few studies focus on interactions among taste compounds. Indeed, sweeteners and acidulants are commonly used food additives, and sweet-sour mixtures always provide improved tastes. For example, sensory studies have shown that sourness suppresses sweetness. However, the degree of sweetness suppression by sourness is difficult to evaluate quantitatively and objectively. Therefore, we propose a biohybrid tongue that is constructed by integrating mammalian gustatory epithelium with a microelectrode array chip. The taste quality and intensity information is coded in time-frequency patterns of local field potential. Different response patterns evoked by sweet and sour stimuli are observed, and the response is dose-dependent. Then, interaction effects of sourness against sweetness are quantified. The results indicate that suppression of sweetness by sourness occurs by increasing sourness concentrations. In summary, this study provides a powerful new tool for quantitative evaluation of sweet, sour, and their binary taste interactions that mimic the mammalian taste system.

Abstract:Bitter is generally undesirable, although it is an important part of flavor. Bitter substances exhibit diverse health-promoting activities, which is in line with the famous Chinese saying ‘a good medicine tastes bitter’. Naringin (NAG) and neohesperidin (NHP), two important flavanones that give bitterness to citrus fruits, show various pharmacological activities. Interestingly, their hydrogenation products, i.e. naringin dihydrochalcone (NDC) and neohesperidin dihydrochalcone (NHDC), undergo a dramatic taste shift from bitter to intensely sweet, which can be 300 and 1000 times sweeter than sucrose, respectively. Such sweeteners not only provide a sweet taste without the burden of increased calorie intake and glycemia, but also may exert multiple bioactivities. This review summarizes common dietary bitter and sweet compounds with sensory scores. Taste conversions induced by structural changes from bitter NAG and NHP to sweet NDC and NHDC are particularly discussed. In addition, the taste-sensing mechanisms, pharmacological characteristics, dietary distribution, synthesis, and food industry applications of these bitter–sweet interchangeable compounds are outlined. In conclusion, the bitter NAG and NHP are promising therapeutic candidates for management of diverse etiologically complex diseases while their corresponding dihydrochalcones NDC and NHDC are promising sweeteners, which might be a blessing for those who need to control sugar intake.

Abstract:Sweet taste perception starts from the interaction of a sweet molecule with residues of the sweet taste receptor (STR). Yet, how the perception of sweet taste emerges from this interaction is still not clear. Here, we reveal a potential mechanism in the initial stages of this cascade. In our previous studies, we observed a 0.6 Pearson correlation coefficient between binding affinity and sweetness intensity. We conducted a series of MD simulations by YASARA Structure, for a total of 1.8 microseconds, to further investigate the mechanism of action of the molecules which we observed to induce considerable sweet taste perception despite having low binding affinity. We observed a gradual expansion in the hydrogen bond (HBond) network around each sweetener in the binding site which involved the water molecules and the residues of the binding pocket. Important HBond and hydrophobic interactions for sweet taste were detected. Some of the residues which we identified to be involved in the HBond network, and also to cause pattern changes in the dynamic cross-correlation matrix, have been known to be parts of the sweet taste signaling pathway. An analysis of the sweeteners' pharmacophore during the MD experiments demonstrated a gradual emergence of the Nofre-Tinti conformation, a proposed determinant in the sweetness intensity, in the ligands despite its initial absence in the ligand–protein complex. In conclusion, these results reveal a novel mechanism for inducing sweet taste and open a new avenue for designing artificial sweeteners, besides the classical hunt for high binding affinity.

Abstract:The last decade witnessed remarkable advances in our knowledge of the gustatory system. Application of molecular biology techniques not only determined the identity of the membrane receptors and downstream effectors that mediate sweetness, but also uncovered the overall logic of gustatory coding in the periphery. However, while the ability to taste sweet may offer the obvious advantage of eliciting rapid and robust intake of sugars, a number of recent studies demonstrate that sweetness is neither necessary nor sufficient for the formation of long-lasting preferences for stimuli associated with sugar intake. Furthermore, uncoupling sweet taste from ensuing energy utilization may disrupt body weight control. This minireview examines recent experiments performed in both rodents and Drosophila revealing the taste-independent rewarding properties of metabolizable sugars. Taken together, these experiments demonstrate the reinforcing actions of sugars in the absence of sweet taste signaling and point to a critical role played by dopamine systems in translating metabolic sensing into behavioral action. From a mechanistic viewpoint, current evidence favors the concept that gastrointestinal and post-absorptive signals contribute in parallel to sweet-independent sugar acceptance and dopamine release.

Abstract:Growing demand for the tasty and healthy food has driven the development of low-calorie sweeteners, sweet taste modulators, and bitter masking compounds originated from natural sources. With the discovery of human taste receptors, increasing numbers of sweet taste modulators have been identified through human taste response and molecular docking techniques. However, the discovery of novel taste-active molecules in nature can be accelerated by using advanced spectrometry technologies based on structure-activity relationships (SARs). SARs explain why structurally similar compounds can elicit similar taste qualities. Given the characterization of structural information from reported data, strategies employing SAR techniques to find structurally similar compounds become an innovative approach to expand knowledge of sweeteners. This review aims to summarize the structural patterns of known natural non-nutritive sweeteners, sweet taste enhancers, and bitter masking compounds. Innovative SAR-based approaches to explore sweetener derivatives are also discussed. Most sweet-tasting flavonoids belong to either the flavanonols or the dihydrochalcones and known bitter masking molecules are flavanones. Based on SAR findings that structural similarities are related to the sensory properties, innovative methodologies described in this paper can be applied to screen and discover the derivatives of taste-active compounds or potential taste modulators.

Abstract:Covering: 2000 to 2016On the molecular level humans sense food by a variety of specialized tissues which express sensory receptors to handle nutritive value. In general, this means the interplay of gustatory, olfactory, trigeminal and haptic sensation is translated into perception and leads, in terms of taste, to descriptions like sweet, bitter, salty, sour and umami. Further perceptions include astringent, cool, hot, prickle, lingering, kokumi and fatty to name predominant characterizations. It is still not fully understood how this plethora of impressions can be perceived by quite a limited number of receptors obviously being the initial compilers to judge palatability. However, since the discovery of mammalian taste receptors (TASRs) almost 30 years ago the use of taste receptors in cell-based screening campaigns is advancing in industrial approaches. The article will highlight the impacts and the limits of cell-based guided identification of taste modulators for food applications with an emphasis on sweet, bitter and savory taste as well as implications emerging from natural products.

Glia-like taste cells mediate an intercellular mode of peripheral sweet adaptation

Abstract:The sense of taste generally shows diminishing sensitivity to prolonged sweet stimuli, referred to as sweet adaptation. Yet, its mechanistic landscape remains incomplete. Here, we report that glia-like type I cells provide a distinct mode of sweet adaptation via intercellular crosstalk with chemosensory type II cells. Using the microfluidic-based intravital tongue imaging system, we found that sweet adaptation is facilitated along the synaptic transduction from type II cells to gustatory afferent nerves, while type I cells display temporally delayed and prolonged activities. We identified that type I cells receive purinergic input from adjacent type II cells via P2RY2 and provide inhibitory feedback to the synaptic transduction of sweet taste. Aligning with our cellular-level findings, purinergic activation of type I cells attenuated sweet licking behavior, and P2RY2 knockout mice showed decelerated adaptation behavior. Our study highlights a veiled intercellular mode of sweet adaptation, potentially contributing to the efficient encoding of prolonged sweetness.
