Abstract:Biofortification – the process of increasing the concentrations of essential nutrients in staple crops – is a means of addressing the burden of micronutrient deficiencies at a population level via existing food systems, such as smallholder farms. To realise its potential for global impact, we need to understand the factors that are associated with decisions to adopt biofortified crops and food products. We searched the literature to identify adoption determinants, i.e. barriers to (factors negatively associated) or facilitators of (factors positively associated) adoption, of biofortified crops and food products. We found 41 studies reporting facilitator(s) and/or barrier(s) of adoption. We categorised the factors using the Consolidated Framework of Implementation Research 2.0, resulting in a set of factors that enable or constrain adoption of biofortified foods across twenty-four constructs and five domains of this meta-theoretical determinant framework from implementation science. Facilitators of orange sweet potato adoption included knowledge about importance, relative advantage, efficient production and management practices; barriers included lacking timely access to quality vines and market remoteness (28 studies total). Facilitators of vitamin A cassava adoption included awareness of its benefits and access to information; barriers included poor road networks and scarcity of improved technology including inadequate processing/storage facilities (8). Facilitators of high-iron bean adoption included farmers' networking and high farming experience; barriers included low knowledge of bean biofortification (8). Barriers to vitamin A maize adoption included low awareness and concerns regarding yield, texture and aflatoxin contamination (1). These barriers and facilitators may be a starting point for researchers to move towards testing implementation strategies and/or for policymakers to consider before planning scale-up and continuous optimisation of ongoing projects promoting adoption of biofortified crops and food products.

Abstract:Biofortification (increasing the micronutrient content of food before harvest) has been successfully used to nutritionally improve staple foods in low- and middle-income countries. This approach could also help address micronutrient shortfalls in at-risk populations in high-income countries (HICs), however, the potential of biofortification interventions in this context is not well understood. The aim of this scoping review is to assess the nature and extent of available research evidence on biofortified foods in relation to human consumption in HICs. Literature searches were conducted in MEDLINE, WoS, ProQuest, CINAHL, AGRIS and Epistemonikos. Forty-six peer-reviewed articles were included. Most research was conducted in the USA ( n = 15) and Italy ( n = 11), on cereal crops ( n = 14) and vegetables ( n = 11), and on selenium ( n = 12) and provitamin A ( n = 11). Seven research domains were identified in the literature: bioavailability ( n = 17); nutrient stability ( n = 11); opinions and attitudes ( n = 9); functionality ( n = 9); sensory properties ( n = 2); safety ( n = 1); and modeling ( n = 1). Evidence from HICs in each domain is limited. There is a need for more research particularly in areas sensitive to the cultural and socio-economic context.

Availability, production, and consumption of crops biofortified by plant breeding: current evidence and future potential

Abstract:Biofortification is the process of increasing the density of vitamins and minerals in a crop through plant breeding-using either conventional methods or genetic engineering-or through agronomic practices. Over the past 15 years, conventional breeding efforts have resulted in the development of varieties of several staple food crops with significant levels of the three micronutrients most limiting in diets: zinc, iron, and vitamin A. More than 15 million people in developing countries now grow and consume biofortified crops. Evidence from nutrition research shows that biofortified varieties provide considerable amounts of bioavailable micronutrients, and consumption of these varieties can improve micronutrient deficiency status among target populations. Farmer adoption and consumer acceptance research shows that farmers and consumers like the various production and consumption characteristics of biofortified varieties, as much as (if not more than) popular conventional varieties, even in the absence of nutritional information. Further development and delivery of these micronutrient-rich varieties can potentially reduce hidden hunger, especially in rural populations whose diets rely on staple food crops. Future work includes strengthening the supply of and the demand for biofortified staple food crops and facilitating targeted investment to those crop-country combinations that have the highest potential nutritional impact.

The socioeconomics of genetically modified biofortified crops: a systematic review and meta-analysis

Abstract:Building upon the growing interest and research on genetically modified (GM) biofortification, its socioeconomic potential has been increasingly examined. We conducted two systematic reviews and meta-analyses to provide comprehensive evidence of consumers' willingness to pay (11 economic valuation studies, 64 estimates) and cost-effectiveness/benefits (five economic evaluation studies, 30 estimates). Worldwide, consumers were willing to pay 23.9% more for GM biofortified food crops. Aside from crop and design-related differences, information provision was deemed crucial. Positive information (nutrition and GM benefits) is associated with the highest consumer willingness to pay, compared with negative, objective, and conflicting GM information, especially when negative information was mentioned last. This health intervention would reduce the aggregated micronutrient deficiency burden in Asia (15.6 million disability-adjusted life years (DALYs)) by 12.5-51.4%, at a low cost of USD 7.9-27.8 per DALY in a pessimistic and optimistic scenario, respectively. Given that GM biofortified crops could tackle hidden hunger in a cost-effective and well-accepted way, its implementation is worth pursuing. A case study on folate biofortification further elaborates on the importance of socioeconomic research and the determinants of their market potential.
The research and implementation continuum of biofortified sweet potato and maize in Africa

Abstract:The enhancement of sweet potato and maize with provitamin A carotenoids has been part of HarvestPlus's research continuum since the formation of the biofortification project. This review includes case studies of biofortification strategies used for sweet potato in Uganda and orange maize in Zambia. The current status of the science and release of biofortified varieties was reviewed by three scientists who were part of the HarvestPlus program for more than a decade with input from a scientist who experienced orange maize dissemination in Zambia. High β-carotene varieties of sweet potato were introduced into South Africa and Mozambique, and efficacy and effectiveness studies, respectively, showed promise to improve vitamin A status, followed by dissemination efforts in Uganda. A randomized, controlled effectiveness trial tested extension models to promote sweet potato and assessed vitamin A intake among Ugandans. Orange maize breeding was initially a challenge, but considering that the carotenoid biosynthetic pathway was present in maize germplasm, breeders quickly bred higher amounts of provitamin A into the maize that was ultimately released in Zambia. Initial resistance occurred because orange maize was associated with yellow maize, which had negative connotations associated with food aid and animal feed, and consumers preferred white maize. Currently, both orange crops are available on the market.

Abstract:Biofortification is an upcoming, promising, cost-effective, and sustainable technique of delivering micronutrients to a population that has limited access to diverse diets and other micronutrient interventions. Unfortunately, major food crops are poor sources of micronutrients required for normal human growth. The manuscript deals in all aspects of crop biofortification which includes-breeding, agronomy, and genetic modification. It tries to summarize all the biofortification research that has been conducted on different crops. Success stories of biofortification include lysine and tryptophan rich quality protein maize (World food prize 2000), Vitamin A rich orange sweet potato (World food prize 2016); generated by crop breeding, oleic acid, and stearidonic acid soybean enrichment; through genetic transformation and selenium, iodine, and zinc supplementation. The biofortified food crops, especially cereals, legumes, vegetables, and fruits, are providing sufficient levels of micronutrients to targeted populations. Although a greater emphasis is being laid on transgenic research, the success rate and acceptability of breeding is much higher. Besides the challenges biofortified crops hold a bright future to address the malnutrition challenge.

Abstract:Nutritious foods are essential for human health and development. However, malnutrition and hidden hunger continue to be a challenge globally. In most developing countries, access to adequate and nutritious food continues to be a challenge. Although hidden hunger is less prevalent in developed countries compared to developing countries where iron (Fe) and zinc (Zn) deficiencies are common. The United Nations (UN) 2nd Sustainable Development Goal was set to eradicate malnutrition and hidden hunger. Hidden hunger has led to numerous cases of infant and maternal mortalities, and has greatly impacted growth, development, cognitive ability, and physical working capacity. This has influenced several countries to develop interventions that could help combat malnutrition and hidden hunger. Interventions such as dietary diversification and food supplementation are being adopted. However, fortification but mainly biofortification has been projected to be the most sustainable solution to malnutrition and hidden hunger. Plant-based foods (PBFs) form a greater proportion of diets in certain populations, hence, fortification of PBFs is relevant in combating malnutrition and hidden hunger. Agronomic biofortification, plant breeding, and transgenic approaches are some currently used strategies in food crops. Crops such as cereals, legumes, oilseeds, vegetables, and fruits have been biofortified through all these three strategies. The transgenic approach is sustainable, efficient, and rapid, making it suitable for biofortification programs. Omics technology has also been introduced to improve the efficiency of the transgenic approach.

Abstract:Background: Food fortification and biofortification are well-established strategies to address micronutrient deficiencies in vulnerable populations. However, the effectiveness of fortification programs is not only determined by the biological efficacy of the fortified foods but also by effective and sustainable implementation, which requires continual monitoring, quality assurance and control, and corrective measures to ensure high compliance. Objective: To provide an overview of efficacy, effectiveness, economics of food fortification and biofortification, and status of and challenges faced by large-scale food fortification programs in low- and middle-income countries (LMIC). Methods: A literature review of PubMed publications in English from 2000 to 2017, as well as gray literature, targeting nongovernmental organizations whose work focuses on this topic, complemented by national reports and a “snowball” process of citation searching. The article describes remaining technical challenges, barriers, and evidence gap and prioritizes recommendations and next steps to further accelerate progress and potential of impact. Results: The review identifies and highlights essential components of successful programs. It also points out issues that determine poor program performance, including lack of adequate monitoring and enforcement and poor compliance with standards by industry. Conclusions: In the last 17 years, large-scale food fortification initiatives have been reaching increasingly larger segments of populations in LMIC. Large-scale food fortification and biofortification should be part of other nutrition-specific and nutrition-sensitive efforts to prevent and control micronutrient deficiencies. There are remaining technical and food system challenges, especially in relation to improving coverage and quality of delivery and measuring progress of national programs.

Abstract:Around 2 billion people are suffering from chronic malnutrition or “hidden hunger”, which is the result of many diseases and disorders, including cognitive degeneration, stunting growth, and mortality. Thus, biofortification of staple food crops enriched with micronutrients is a more sustainable option for providing nutritional supplements and managing malnutrition in a society. Since 2001, when the concept of biofortification came to light, different research activities have been carried out, like the development of target populations, breeding or genetic engineering, and the release of biofortified cultivars, in addition to conducting nutritional efficacy trials and delivery plan development. Although, being a cost-effective intervention, it still faces many challenges, like easy accessibility of biofortified cultivars, stakeholders’ acceptance, and the availability of biofortified germplasm in the public domain, which varies from region to region. Hence, this review is focused on the recent potential, efforts made to crop biofortification, impacts analysis on human health, cost-effectiveness, and future perspectives to further strengthen biofortification programs. Through regular interventions of sustainable techniques and methodologies, biofortification holds huge potential to solve the malnutrition problem through regular interventions of nutrient-enriched staple food options for billions of people globally.

Abstract:Biofortification of staple crops has been proposed as a strategy to address micronutrient malnutrition, particularly with respect to insufficient intake of vitamin A, iron, zinc, and folate. The World Health Organization, in collaboration with the Food and Agriculture Organization of the United Nations and the Sackler Institute for Nutrition Science at the New York Academy of Sciences, convened a technical consultation entitled "Staple Crops Biofortified with Vitamins and Minerals: Considerations for a Public Health Strategy" in April 2016. Participants of the consultation reviewed the definition of biofortification of staple crops, patterns of crops production, processing, consumption, seed varieties, and micronutrient stability and bioavailability, as well as farmers' adoption and acceptability of the modified crops. Also discussed were economic, environmental, safety, and equity aspects of biofortified crops, as well as legal, policy, regulatory, and ethical issues for the implementation of biofortification strategies in agriculture and nutrition. Consultation working groups identified important and emerging technical issues, lessons learned, and research priorities to better support the evidence of improved nutrition and unintended adverse effects of biofortification. This paper provides the background and rationale of the technical consultation, synopsizes the presentations, and provides a summary of the main considerations proposed by the working groups.

Abstract:Micronutrient deficiencies, particularly in essential vitamins and minerals, pose a significant public health challenge, affecting over two billion people worldwide. These deficiencies contribute to various health issues, impaired cognitive development, and reduced productivity, ultimately hindering social and economic progress. Biofortification, a process of enhancing the nutritional content of staple crops through conventional breeding or genetic engineering, has emerged as a promising and sustainable approach to combat micronutrient deficiencies and ensure global food security. This review explores the potential of Biofortification as a cost-effective and sustainable solution to address hidden hunger and improve the nutritional status of vulnerable populations. Biofortification offers several advantages over traditional interventions, such as supplementation and food fortification. By targeting staple crops consumed by the majority of the population, Biofortification ensures a wide reach and sustained nutrient intake without requiring significant changes in dietary habits. Moreover, biofortified crops can be grown locally, reducing the reliance on external interventions and empowering farmers to improve their nutritional status and livelihoods. Numerous studies have demonstrated the efficacy of bio fortified crops in increasing micronutrient intake and improving health outcomes. For instance, iron-biofortified pearl millet has been shown to increase iron absorption and reduce anemia prevalence in children, while zinc-biofortified wheat has improved zinc status and reduced stunting. Additionally, vitamin A-biofortified sweet potato and cassava have significantly increased vitamin A intake and reduced vitamin A deficiency in various populations. Despite the promising results, the success of Biofortification relies on several factors, including the development of nutrient-dense varieties, consumer acceptance, and effective dissemination strategies. Collaboration among researchers, policymakers, and stakeholders is essential to scale up Biofortification efforts and ensure their long-term sustainability. By prioritizing Biofortification as a key strategy in combating micronutrient deficiencies, we can work towards a more nourished and food-secure world.

Abstract:Abstract The deficiencies of micronutrients known as hidden hunger are severely affecting more than one-half of the world’s population, which is highly related to low bioavailability of micronutrients, poor quality diets, and consumption of cereal-based foods in developing countries. Although numerous experiments proved biofortification as a paramount approach for improving hidden hunger around the world, its effectiveness is highly related to various soil factors, climate conditions, and the adoption rates of biofortified crops. Furthermore, agronomic biofortification may result in the sedimentation of heavy metals in the soil that pose another detrimental effect on plants and human health. In response to these challenges, several studies suggested intercropping as one of the feasible, eco-friendly, low-cost, and short-term approaches for improving the nutritional quality and yield of crops sustainable way. Besides, it is the cornerstone of climate-smart agriculture and the holistic solution for the most vulnerable area to solve malnutrition that disturbs human healthy catastrophically. Nevertheless, there is meager information on mechanisms and processes related to soil-plant interspecific interactions that lead to an increment of nutrients bioavailability to tackle the crisis of micronutrient deficiency in a nature-based solution. In this regard, this review tempted to (1) explore mechanisms and processes that can favor the bioavailability of Zn, Fe, P, etc. in soil and edible parts of crops, (2) synthesize available information on the benefits and synergic role of the intercropping system in food and nutritional security, and (3) outline the bottlenecks influencing the effectiveness of biofortification for promoting sustainable agriculture in sub-Saharan Africa (SSA). Based on this review SSA countries are malnourished due to limited access to diverse diets, supplementation, and commercially fortified food; hence, I suggest integrated research by agronomists, plant nutritionists, and agroecologist to intensify and utilize intercropping systems as biofortification sustainably alleviating micronutrient deficiencies. Cytogenetic analysis of the pathology of frozen shoulder

Abstract:Background: Frozen shoulder (FS) is a debilitating musculoskeletal condition with an uncertain etiology and pathogenic mechanism. The aim of this study was to investigate the hypothesis that an alteration in the level of cytokines may disrupt the normal inflammatory and tissue healing process in the shoulder, leading to the development of FS. Materials and Methods: A prospective case–control study was undertaken, analyzing patients undergoing arthroscopic treatment of FS and control patients being treated for subacromial bursitis. Synovial biopsies were taken from all subjects. Synovial RNA levels were analyzed using quantitative polymerase chain reaction (qPCR) Results: Thirteen patients with FS were recruited, four of whom were diagnosed with diabetes mellitus, along with 10 control patients. Cytogenetic analysis using qPCR revealed both fibrogenic cytokine matrix metalloproteinase 3 (MMP 3) (1.98×10 5 vs. 755.0, P=0.068) and inflammatory cytokine interleukin 6 (IL 6) (1679.2 vs. 372.8, P=0.062) to be elevated in FS cases as compared to controls. Comparison between diabetic and non-diabetic patients revealed a decrease in the level of expression of inflammatory cytokine, monocyte colony stimulating factor (M-CSF) (12,496 vs. 305.1, P=0.04) in diabetic FS patients. Conclusions: The results demonstrate that levels of inflammatory and fibrogenic cytokines are elevated in the synovium of patients with FS compared with controls. This indicates that altered levels of inflammatory cytokines may be associated with the pathogenesis of inflammation evolving into fibrosis, which is the characteristic feature of FS. We have also shown the opposite to be the case in patients with diabetic FS.

Abstract:Microorganisms play a fundamental role in sustainable agriculture, and their importance in common bean (Phaseolus vulgaris) cultivation cannot be underestimated. This review article aims to comprehensively explore the diverse roles of microorganisms in sustainable biofortified common bean cultivation. Biofortification refers to the process of increasing the nutrient content in crops, which helps combat deficiencies in iron, zinc, and vitamins in the human body. Biofortified beans have better agronomic characteristics and offer higher micronutrient content compared to conventional crops. We examine the contribution of various microbial communities in nitrogen fixation, soil structure improvement, nutrient recycling, and disease suppression. Understanding the interaction between beneficial microorganisms and biofortified common bean plants enables us to develop ecologically sound and sustainable approaches to optimize crop productivity and improve nutrition and livelihoods for millions of people worldwide while reducing the environmental impact of agricultural practices.

Abstract:The relationship between malnutrition and climate change are still poorly understood but a comprehensive knowledge on their interactions is needed to address the global public health agenda. Limited studies have been conducted to propose robust and economic-friendly strategies to augment the food basket with underutilized species and biofortify the staples for nutritional security. Sea-buckthorn is a known "superfood" rich in vitamin C and iron content. It is found naturally in northern hemispherical temperate Eurasia and can be utilized as model species for genetic biofortification in cash crops like wheat. This review focuses on the impacts of climate change on inorganic (iron, zinc) and organic (vitamin C) micronutrient malnutrition employing wheat as highly domesticated crop and processed food commodity. As iron and zinc are particularly stored in the outer aleurone and endosperm layers, they are prone to processing losses. Moreover, only 5% Fe and 25% Zn are bioavailable once consumed calling to enhance the bioavailability of these micronutrients. Vitamin C converts non-available iron (Fe 3+ ) to available form (Fe 2+ ) and helps in synthesis of ferritin while protecting it from degradation at the same time. Similarly, reduced phytic acid content also enhances its bioavailability. This relation urges scientists to look for a common mechanism and genes underlying biosynthesis of vitamin C and uptake of Fe/Zn to biofortify these micronutrients concurrently. The study proposes to scale up the biofortification breeding strategies by focusing on all dimensions i.e., increasing micronutrient content and boosters (vitamin C) and simultaneously reducing anti-nutritional compounds (phytic acid). Mutually, this review identified that genes from the Aldo-keto reductase family are involved both in Fe/Zn uptake and vitamin C biosynthesis and can potentially be targeted for genetic biofortification in crop plants.

Abstract:Micronutrient malnutrition, the so‐called hidden hunger, affects more than one‐half of the world's population, especially women and preschool children in developing countries. Despite past progress in controlling micronutrient decencies through supplementation and food fortification, new approaches are needed to expand the reach of food‐based interventions. Biofortification, a new approach that relies on conventional plant breeding and modern biotechnology to increase the micronutrient density of staple crops, holds great promise for improving the nutritional status and health of poor populations in both rural and urban areas of the developing world. HarvestPlus, a research program implemented with the international research institutes of the CGIAR, targets a multitude of crops that are a regular part of the staple‐based diets of the poor and breeds them to be rich in iron, zinc, and provitamin A. This paper emphasizes the need for interdisciplinary research and addresses the key research issues and methodological considerations for success. The major activities to be undertaken are broadly grouped into research related to nutrition research and impact analysis, and research considerations for delivering biofortified crops to end‐users effectively. The paper places particular emphasis on the activities of the plant breeding and genetics component of this multidisciplinary program. The authors argue that for biofortification to succeed, product profiles developed by plant breeders must be driven by nutrition research and impact objectives and that nutrition research must understand that the probability of success for biofortified crops increases substantially when product concepts consider farmer adoption and, hence, agronomic superiority.

[Methodology for selecting areas for biofortified crop intervention]

Abstract:Objective: To identify geographical areas in Latin America and the Caribbean where biofortification of staple crops, such as beans, corn, rice, cassava, and sweet potatoes, might help reduce nutritional deficiencies in the Region. Methods: A geographic information system (GIS) was produced with records on nutritional risks, crop production, food consumption, and demographic and socioeconomic data, for 11 countries in the Region. Four case studies were conducted (in Bolivia, Colombia, Guatemala, and Mexico) using exploratory and descriptive analysis of thematic maps that were superimposed and compared to reveal overlapping and spatial patterns, thereby identifying areas suited to intervention. Results: In Guatemala, the highest rates of nutritional risk, bean production, and population density overlapped in the northeast and southeast areas. In Mexico, spatial distribution of the highest risk levels for nutrition, poverty, and corn production were concentrated in the central and southern municipalities. In Bolivia, bean production tended to be in the eastern part of the country, and nutritional risk, in the west. In Colombia, both nutritional risk and cassava production showed wide geographic dispersion. Conclusions: For Guatemala, we propose iron biofortification of beans in the southern parts of the northeast and southeast; for Mexico, amino-acid biofortification of corn in the central and southern municipalities that produce it; for Bolivia, iron and zinc biofortification of beans in the bean-producing areas of Santa Cruz, Chuquisaca, and Tarija; and for Colombia, beta-carotene biofortification of cassava in the Cordoba and Cundinamarca departments.

Abstract:Micronutrient deficiency poses a significant and challenging threat to over one-third of the global population. To deal with this issue, nutrient management via biofortification is considered one of the effective and sustainable approaches. Biofortification involves multifaceted strategies such as agronomic practices—mineral and microbial interventions, conventional plant breeding, molecular and genetic engineering techniques. All these strategies fortify crops with essential elements like pro-vitamin A, vitamin C, iron, zinc, calcium, iodine, and selenium while reducing anti-nutritional factors and trypsin inhibitors in staple foods. However, most of these techniques are considered expensive and require more time than routine practices. Therefore, this review focused on the interaction among soil–plant-nutrient and their management through the introduction of plant growth-promoting bacteria, fungi, and endophytes. The aim was to enrich our understanding of how these interventions facilitate nutrient acquisition, translocation, and uptake within both plant shoots and grains. We emphasize on the use of beneficial microbes in biofortification of staple crops for sustaining human nutritional requirement. Microbes-mediated biofortification exhibits tremendous potential in enhancing the bioavailability of nutrients in staple crops, providing solution to widespread micronutrient deficiencies and nutritional security. Diversification of crop biofortification in different cropping systems is the demand of the hour to mitigate the challenges of hidden hunger. Understanding and harnessing this synergistic relationship can offer promising avenues for sustainable agriculture, addressing nutritional deficiencies, improving crop resilience, and ensuring food security in different croping systems.

Abstract:Cereals and pulses are consumed as a staple food in low-income countries for the fulfillment of daily dietary requirements and as a source of micronutrients. However, they are failing to offer balanced nutrition due to deficiencies of some essential compounds, macronutrients, and micronutrients, i.e., cereals are deficient in iron, zinc, some essential amino acids, and quality proteins. Meanwhile, the pulses are rich in anti-nutrient compounds that restrict the bioavailability of micronutrients. As a result, the population is suffering from malnutrition and resultantly different diseases, i.e., anemia, beriberi, pellagra, night blindness, rickets, and scurvy are common in the society. These facts highlight the need for the biofortification of cereals and pulses for the provision of balanced diets to masses and reduction of malnutrition. Biofortification of crops may be achieved through conventional approaches or new breeding techniques (NBTs). Conventional approaches for biofortification cover mineral fertilization through foliar or soil application, microbe-mediated enhanced uptake of nutrients, and conventional crossing of plants to obtain the desired combination of genes for balanced nutrient uptake and bioavailability. Whereas, NBTs rely on gene silencing, gene editing, overexpression, and gene transfer from other species for the acquisition of balanced nutritional profiles in mutant plants. Thus, we have highlighted the significance of conventional and NBTs for the biofortification of cereals and pulses. Current and future perspectives and opportunities are also discussed. Further, the regulatory aspects of newly developed biofortified transgenic and/or non-transgenic crop varieties via NBTs are also presented.
