Innovative catalytic synthesis of 5-unsubstituted 3,4-dihydropyrimidine-2-ones: Use of CoCa3(HPO4)(PO4)2(OH) as a novel bimetallic catalyst in multicomponent condensation of Biginelli reaction

Abstract:An innovative and easy multicomponent synthesis of important biologically active molecules, 5-unsubstituted 3,4-dihydropyrimidine-2-ones, via the Biginelli reaction between aromatic aldehydes, acetophenone derivatives and urea catalyzed by a novel bimetallic phosphate catalyst CoCa3(HPO4)(PO4)2(OH) was developed. The catalyst support, prepared by the coprecipitation method using phosphoric acid, calcium nitrate, and cobalt nitrate, was characterized for its chemical structure and composition through various spectroscopy analyses such as X-ray diffraction (XRD), infrared spectroscopy (FTIR), scanning electron microscopy (SEM), and elemental analysis ICP-MS. The developed catalyst showed high efficiency in organic synthesis, allowing us to prepare various 3,4-dihydropyrimidine-2-ones derivatives in excellent yields of up to 97% under mild reaction conditions and in a short reaction time of 10 min. This new approach offers many advantages over conventional systems, notably in terms of simplicity, ease of handling, production cost and the possibility of reusing the catalyst up to seven times. This work provides a novel alternative method for the synthesis of 5-unsubstituted 3,4-dihydropyrimidine-2-ones derivatives.
