Abstract:The present study developed an improved analytical method for the simultaneous quantification of 17 kinds of Aspergillus, Fusarium and Penicillium mycotoxin contaminants in foods and feeds by ultra-performance liquid chromatography combined with electrospray ionization triple quadrupole tandem mass spectrometry (UPLC–MS/MS) under the multiple reaction monitoring (MRM) mode, and especially focused on the optimization of extraction, clean-up, UPLC separation and MS/MS parameters of analytes. Homogenized samples were sequentially extracted by 84% (v/v) of acetonitrile aqueous solution with the selected internal standard (zearalanone) spiking, SPE clean-up with Mycosep 226 Aflazon+ Multifunctional cartridges, filtration, concentration and secondary filtration. Using double sample injection method, the analytes were separated by UPLC BEH C18 column (100mm×2.1mm I.D., 1.7μm), and eluted with ammonium acetate/methanol and aqueous ammonia/methanol for the ESI+ and ESI− analysis, respectively. The 10 positive ions and 7 negative ions of mycotoxins were separated by gradient elution with the retention time of 6.5 and 4min, respectively. The LOQ of selected analytes ranged from 0.01 to 0.70μgkg−1, which was lower than the criteria of EU, USA and other countries on the determination of the minimum limiting level of various mycotoxins in foods including baby foods and feed stuffs. Meanwhile, high correlation coefficients (r2>0.99) of 17 mycotoxins were obtained within their respective linear ranges (0.05–20μgkg−1 for 10 positive ions and 0.5–50μgkg−1 for 7 negative ions) and reasonable recoveries (70.6–119.0%) of them were also demonstrated in different spiked levels. This quantitative method has many advantages including simple pretreatment, rapid determination and high sensitivity, which could be applied to the determination and quantification of multi-component mycotoxin contaminants in complex matrixes. Meanwhile, the method successfully fulfilled the minimum limiting level requests from various countries.
A dilute-and shoot approach using ultra high-performance liquid chromatograph-mass/mass spectrometry for 16 mycotoxins analysis in cereals and products

Abstract:Objective To develop and validate the ultra high-performance liquid chromatograph coupled with mass spectrometry (UPLC-MS/MS) method for simultaneous determination of 16 mycotoxins,including aflatoxins,ochratoxins,trichothecenes,fumonisins and so on.Methods Five grams of the sample was extracted with acetonitrile/water/formic acid (79 ∶ 20 ∶ 1,V/V).After centrifuged and diluted,the extraction was separated by Cortecs C18 (100 mm × 3.0 mm,1.6 μm) analytical column under acetonitrile-0.1％ formic acid gradient eluting,then was determined by UPLC-MS/MS.Dilution isotope internal calibration was used for qualification.Results The result showed good linearities (0.05-200 ng/ml) for 16 mycotoxins in the certain correlation ranges with the coefficients all above 0.99.The limits of quantification ranged from 0.1 to 50 μg/kg.The average recoveries of 16 mycotoxins at three spiked levels ranged in 72.67％-126.92％ with relative standard deviations of 0.15％-16.83％.The accuracies were acceptable by detecting natural matrix quality control samples.Conclusion The method is rapid,simple,sensitive,reproducible and accurate for simultaneous determination of multiple mycotoxins in cereals and products in routine laboratories.It meets the the requirement of mycotoxins monitoring in relative food.
Establishment of an Isotope Dilution Lc-Ms/Ms Method Revealing Kinetics and Distribution of Co-Occurring Mycotoxins in Rats

Abstract:An isotope dilution liquid chromatography-tandem mass spectrometry (LC-MS/MS) method with a fast sample preparation using homemade clean-up cartridges was developed for simultaneous determination of co-occurring mycotoxins exemplified with aflatoxin B1 (AFB1) and T-2 toxin (T-2) in representative biomatrices of rat plasma, heart, liver, kidney, spleen, lung and brain in a total run time of 7 min. The established approach using stable internal standards of [C-13(17)]-AFB1 and [C-13(24)]-T-2 was extensively validated by determining the specificity, linearity (R-2 >= 0.9990), sensitivity (lower limit of quantitation at 0.05 ng mL(-1)), accuracy (70.9-107.7%), precision (RSD <= 14.2%) and stability (>= 70.8%). Based on this methodological advance, the subsequent kinetics and tissue distribution after oral administration of 0.5 mg kg(-1) b.w. of both AFB1 and T-2 in rats were thoroughly studied. As revealed, both AFB1 and T-2 were rapidly eliminated with the half-life time (t(1/2)) in plasma of 8.44 +/- 4.02 h and 8.12 +/- 4.05 h, respectively. Moreover, AFB1 accumulated in all organs where the highest concentration was observed in liver (1.34 mu g kg(-1)), followed by kidney (0.76 mu g kg(-1)). Notably, only low levels of T-2 were observed in spleen (0.70 mu g kg(-1)) and in liver (0.15 mu g kg(-1)). The achieved data as supporting evidence would substantially promote the practical application of the proposed LC-MS/MS method for in vivo toxicokinetics and toxicity studies of co-occurring mycotoxins imitating natural incidence in rat system.

Abstract:Accurate quantification of mycotoxin in cereals is crucial for ensuring food safety and human health. However, the preparation of traditional multisample external calibration curves (MSCCs) is labor-intensive and error-prone. Here, a multiple isotopologue reaction-monitoring (MIRM)-LC-MS/MS method for accurate quantitation of ten major mycotoxins in cereals was successfully developed and validated, where a novel one-sample multipoint calibration curve (OSCC) strategy is used instead of MSCCs. The OSCC can be established by examining the correlation between the calculated theoretical isotopic abundances and the measured abundance across various MIRM channels. In comparison to the MSCC, the OSCC strategy exhibits outstanding performance including superior selectivity, accuracy (78.4-108.6%), and precision (<12.5%). Furthermore, the proposed OSCC-MIRM-LC-MS/MS method was successfully applied to investigate mycotoxin contamination in cereal samples in China. Considering the advantages of simplified workflows and improved throughput, the OSCC-MIRM-LC-MS/MS methodology holds great promise for accurately quantifying chemical contaminants in foods.

Abstract:Mycotoxins are frequently found in food samples and have become a major concern in the maintenance of food safety and human health. In this study, we developed an isotope-coded derivatization (ICD) method for the quantification of six mycotoxins in various complex matrices by using ultra-high-performance liquid chromatography–tandem mass spectrometry (UHPLC-MS/MS). The targeted mycotoxins were ochratoxin A, ochratoxin B, fumonisin B1, fumonisin B2, zearalenone, and zearalanone. The mycotoxins in the actual samples and standards were derivatized with d0-dansyl chloride and its heavy form, d4-dansyl chloride. A mixture of light-isotope- and heavy-isotope-labeled derivatives was subjected to UHPLC-MS/MS, and isotopic internal calibration quantification of all targeted mycotoxins was achieved. The retention time difference between light-isotope- and heavy-isotope-labeled derivatives was less than 0.02 min, and the analysis could be completed within 7.5 min. The developed method was validated in terms of its sensitivity, precision, and accuracy. The obtained limits of detection were in range of 31.03–38.15 ng/L, and the accuracy was 97.13–104.1% with a precision of less than 3.42%. This method was also successfully applied to analyze mycotoxins in three selected cereal samples (corn, wheat, and sorghum), achieving recoveries of 71.3–92.6%. The ICD strategy provides an alternative assay to isotopic internal standards, thereby helping to effectively minimize matrix effects during mass spectrometry analysis. The proposed ICD-based UHPLC-MS/MS method has high potential for the determination of amino- and phenol-containing mycotoxins in various food samples.

Abstract:A high performance liquid chromatography/electrospray ionization tandem mass spectrometric method has been developed for the simultaneous determination of nine carbamate pesticide residues in grains. Carbamate pesticides were extracted with acetonitrile, cleaned up with neutral-alumina column, and then analyzed by high performance liquid chromatography-electrospray (HPLC-ESI) (+) -tandem mass spectrometry (MS/MS). Sample pre-treatment and LC-MS/MS analytical conditions were optimized. The linear range was 0. 1 - 100 g/L, the correlation coefficients were 0. 9986 - 0. 9998. The average recovenies of nine carbamate pesticides in spiked grains (from 0. 001 to 0. 05 mg/Kg) ranged from 73. 40% to 102. 0%, and the relative standard deviation were between 1. 3% and 9. 9%. The results indicate that the method developed is easier, faster, more sensitive and has a better purification effect. It also demonstrats that this multiresidue analysis method can meet the requirements for simultaneous determination of many carbamate pesticides in grains for import & export inspection.

Abstract:Cell‐mediated contraction of tissues containing fibrillar collagene can lead to organ compromise and loss of function. The same process that is biologically advantageous during the contraction phase of wound healing can be subverted in diseases such as hepatic cirrhosis, pulmonary fibrosis, and scleroderma, although the cellular and molecular mechanism of matrix tissue contraction is difficult to study in such chronic diseases. However, certain human eye diseases that result in tractional detachment of the retina and loss of vision are characterized by acute cell‐mediated contraction of collagenous tissue in the vitreous cavity. In this study, we demonstrate that human cells can contract vitreous, a complex biological gel containing type II collagen, in vitro. This cell‐mediated contraction can be blocked by antibodies and peptides that antagonize the function of α2β1 integrin, and the potential for contraction can be conferred upon noncontracting cells by stable transfection of cells with α2 cDNA. We also show that this contractile process, if focally resisted, can result in remodeling vitreous from a gel to a structure that resembles a planar membrane, and that substantial isometric forces can be measured across this tissue. We propose that in diseases such as proliferative diabetic retinopathy and proliferative vitreoretinopathy, α2β1 integrin‐mediated contraction of the vitreous and tension at the site of vitreoretinal attachments contribute to the terminal event of tractional retinal detachment. By extension, we propose that α2β1 integrin is a centrally important molecule in human diseases characterized by remodeling and contraction of collagenous tissue (i.e., fibrocontractive diseases).—Kupper, T. The method involves addition of fumonisins isotope internal standards, extraction with a mixture of acetonitrile and water and clean-up with solid-phase extraction (SPE) cartridges before UHPLC-MS/MS analysis. A single-laboratory method validation was conducted by testing three different spiking levels for repeatability and recovery according to International Union of Pure and Applied Chemistry (IUPAC) guidelines. The LOQ of FB(1), FB(2) and FB(3) were 1.50, 1.65 and 0.4 μg kg(-1), respectively, which were lower than the criteria of EU, USA and other countries regarding minimum residue limits of fumonisins in foods including baby foods and feedstuffs. Recoveries of three fumonisins ranged from 80.9% to 97.0% with RSD values of 2.4-11.1%.The advantages of this method include simple pretreatment, rapid determination and high sensitivity, and it fulfills the requirements for food analysis with respect to minimum residue limits of fumonisins in various countries.

Abstract:A generic procedure, which involved accelerated solvent extraction and homemade cleanup cartridges, has been developed for the extraction and purification of 35 mycotoxins in various traditional Chinese medicine (TCM) matrixes, i.e., rhizomes and roots, seeds, flowers, and grasses and leaves, for subsequent analysis by ultra-high-performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS). All target analytes could be simultaneously quantitated in less than 17 min per run, showing narrow symmetrical peaks. The developed method was also successfully applied in routine monitoring programs, which implied a significant reduction of both effort and time, to investigate the contamination of TCMs. Among 60 commercial TCMs analyzed, 50 were positive. The achieved data underpin the practical application of the UHPLC-MS/MS method as a valuable tool for the trace analysis of multiple mycotoxins in TCMs.

Abstract:Abstract Mycotoxins and pesticides regularly co-occur in agricultural products worldwide. Thus, humans can be exposed to both toxic contaminants and pesticides simultaneously, and multi-methods assessing the occurrence of various food contaminants and residues in a single method are necessary. A two-dimensional high performance liquid chromatography tandem mass spectrometry method for the analysis of 40 (modified) mycotoxins, two plant growth regulators, two tropane alkaloids, and 334 pesticides in cereals was developed. After an acetonitrile/water/formic acid (79:20:1, v/v/v) multi-analyte extraction procedure, extracts were injected into the two-dimensional setup, and an online clean-up was performed. The method was validated according to Commission Decision (EC) no. 657/2002 and document N° SANTE/12682/2019. Good linearity ( R 2 > 0.96), recovery data between 70-120%, repeatability and reproducibility values < 20%, and expanded measurement uncertainties < 50% were obtained for a wide range of analytes, including very polar substances like deoxynivalenol-3-glucoside and methamidophos. However, results for fumonisins, zearalenone-14,16-disulfate, acid-labile pesticides, and carbamates were unsatisfying. Limits of quantification meeting maximum (residue) limits were achieved for most analytes. Matrix effects varied highly (−85 to +1574%) and were mainly observed for analytes eluting in the first dimension and early-eluting analytes in the second dimension. The application of the method demonstrated the co-occurrence of different types of cereals with 28 toxins and pesticides. Overall, 86% of the samples showed positive findings with at least one mycotoxin, plant growth regulator, or pesticide. Graphical abstract

Abstract:A novel pre-treatment was proposed for the simultaneous determination of aflatoxins, ochratoxin A and zearalenone in foodstuffs using high-performance liquid chromatography with fluorescence detection. The analytical procedure was based on a first step using a quick, easy, cheap, effective, rugged, and safe based extraction procedure, followed by salting out and purification with a C18 solid-phase extraction column as interference removal clean-up. Subsequently, collected supernatant was subjected to dispersive liquid-liquid microextraction. Response surface methodology based on central composite design was employed to optimize conditions in the microextraction procedure. Under the optimum conditions, satisfactory analytical performance with recoveries ranging from 63.22 to 107.6% were achieved in different types of cereals and beans, as well as desirable precisions (0.81-8.13%). Limits of detections and quantifications for these six mycotoxins ranging from 0.03 to 13 mu g/ kg and 0.22 to 44 mu g/ kg, respectively, were obtained. Finally, the established method was successfully validated by four certified reference materials (P = 0.897 > 0.05) and applied to 79 samples from local markets.
Antibody Microarray Immunoassay for Simultaneous Quantification of Multiple Mycotoxins in Corn Samples.

Abstract:We developed and tested a prototype of an antibody microarray immunoassay for simultaneous quantitative detection of four typical mycotoxins (aflatoxin B1, ochratoxin A, zearalenone, and fumonisin B1) in corn samples. The test kit consisted of a nitrocellulose membrane layered with immobilized monoclonal antibodies against mycotoxins. During the assay, the mycotoxin-protein conjugates were biotinylated. The signal detection was enhanced by a combination of the biotin-streptavidin system and enhanced chemiluminescence (ECL). This improved the sensitivity of the assay. Under the optimized conditions, four calibration curves with goodness of fit (R2 > 0.98) were plotted. The results showed that the detection limits for aflatoxin B1, ochratoxin A, zearalenone, and fumonisin B1 were 0.21, 0.19, 0.09, and 0.24 ng/mL, with detection ranges of 0.47–55.69, 0.48–127.11, 0.22–31.36, and 0.56–92.57 ng/mL, respectively. The limit of detection (LOD) of this antibody microarray for aflatoxin B1, ochratoxin A, zearalenone, and fumonisin B1 in corn was 5.25, 4.75, 2.25, and 6 μg/kg, respectively. The recovery rates from the spiked samples were between 79.2% and 113.4%, with coefficient of variation <10%. The results of the analysis of commercial samples for mycotoxins using this new assay and the liquid chromatography-tandem mass spectrometry (LC-MS/MS) were comparable and in good agreement. This assay could also be modified for the simultaneous detection of other multiple mycotoxins, as well as low-weight analytes, hazardous to human health.

Abstract:An analytical method for the simultaneous determination of 13 mycotoxins in feed by magnetic dispersive solid-phase extraction combined with ultra-high performance liquid chromatography and tandem mass spectrometry was developed. The samples were extracted with acetonitrile/water (80:20, v/v, containing 3% acetic acid), and separated by centrifugation after salting-out, and then treated with magnetic adsorbents to remove interferences. The separation of target mycotoxins was performed on an ACQUITY UPLC HSS T3 column using a mobile phase consisting of 1 mmol/L ammonium acetate with 0.1% formic acid and methanol by gradient elution. Good linearities for the 13 mycotoxins were achieved with correlation coefficients over 0.99, and the recoveries of mycotoxins were in the range of 89.3-112.6% at spiking at levels of 5, 20, and 100 mu g/kg, with relative standard deviations of 0.9-10.4%. Based on the functional magnetic materials (MDN@Fe3O4, PSA@Fe3O4, ZrO2@Fe3O4) applied in dispersive solid-phase extraction, the pretreatment process is more convenient and it is beneficial to reduce the experimental cost by reusing the recycled magnetic materials. It is a simple, rapid, and environmentally friendly analytical method for the determination of mycotoxins in feed.

Abstract:A speedy and selective ultra-HPLC-MS/MS method for simultaneous determination of deoxynivalenol (DON), 3-acetyldeoxynivalenol (3-ADON), 15-ADON. nivalenol and fusarenon X in traditional Chinese medicines (TCMs) was developed. The method was based on one-step sample cleanup using reliable homemade cleanup cartridges. A linear gradient mobile-phase system, consisting of water containing 0.2% aqueous ammonia and acetonitrile/methanol (90:10, v/v) at a flow rate of 0.4 mL/min, and an Acquity UPLC HSS T3 column (100 mm x 2.1 mm, 1.8 mu m) were employed to obtain the best resolution of the target analytes. [C-13(15)]-DON was used as the internal standard to accomplish as accurate as possible quantitation. The established method was further validated by determining the linearity (R-2 >= 0.9990), sensitivity (LOQ, 0.29-0.99 mu g/kg), recovery (88.5-119.5%) and precision (RSD <= 15.8%). It was shown to be a suitable method for simultaneous determination of DON, 3-ADON, 15-ADON, nivalenol and fusarenon X in various TCM matrices. The utility and practical impact of the method was demonstrated using different TCM samples.

Abstract:An ultra-high-performance liquid chromatography-tandem mass spectrometry (UHPLC–MS/MS) method for simultaneous determination of aflatoxins B1, B2, G1, G2, M1 and M2 in traditional Chinese medicines (TCMs) was developed. The approach was characterized in details and a special focus was placed on the recovery rates of isolation procedure in different TCM matrices, i.e. rhizomes and roots, seeds, flowers, grasses and leaves. For this purpose, [13C17]-aflatoxinB1 was employed as the internal standard and a reliable solid phase extraction-based clean-up method was developed. The observed recovery rates of the six aflatoxins ranged from 85.6% to 117.6% in different matrices. Then, the established method was successfully applied to the determination of the six aflatoxins in various TCMs. For 30 commercial samples analyzed, 16 were contaminated with aflatoxins. The mean levels (incidence) of aflatoxins B1, B2, G1 and G2 in positive samples were 1.40 (68.8%), 1.27 (50.0%), 0.50 (43.8%) and 0.94 (43.8%) μgkg−1, respectively. Interestingly, aflatoxin M1 was detected in two samples with the maximal content of 0.70μgkg−1. No sample was contaminated with aflatoxin M2. Meanwhile, a possible association between the contamination levels and the selected herbs was clarified in the present study.

Abstract:Due to the challenge of hundreds of potential mycotoxins that may be present in cereals, a rapid and reliable ultra-high performance liquid chromatography coupled with quadrupole-time-of-flight tandem mass spectrometry (UHPLC-Q-TOF MS) technique was developed for universal screening of 200 mycotoxins (prototype, emerging and related derivatives) in cereals. With satisfactory sensitivity of accurate mass full-spectrum acquisition, it is feasible to preliminarily identify tentative untargeted mycotoxins with a large range of polarity without reference materials. The current screening method was also validated by the determination of 33 typical mycotoxins, and the screening detection limits in the range of 0.5-100 mu g kg(-1) were established in cereals. In total, 138 stored samples were contaminated by 46 mycotoxins and their metabolites, some of which were firstly reported in cereals, posing emerging potential health risks to humans and animals. Furthermore, the accumulation, transformation and degradation mechanisms of typical mycotoxins in cereals were investigated under real storage conditions.

Abstract:A novel and rapid method for the determination of ochratoxin A (OTA), funmonisin B1 (FB1) and zearalenone (ZEN) in legumes samples by high throughput flow injection analysis-tandem mass spectrometry (FIA-MS/MS) in conjunction with isotope-coded labeling (ICD) coupled with dispersive liquid–liquid microextraction (DLLME) was developed. The samples were extracted by a known QuEChERS method (Quick, Easy, Cheap, Effective, Rugged, Safe). The extracted components and mycotoxin standards were, respectively, labeled by d 0 -10-methyl-acridone-2-sulfonyl chloride and its deuterated counter part (d 3 -MASC). Subsequently, the mixed light-/heavy-labeled solutions were enriched and purified by DLLME method and corresponding extracts were directly used for FIA-MS/MS analysis. The FIA-MS/MS method coupled with ICD-coded labeling technique for the determination of mycotoxins in legume samples could be achieved within 2 min, which was much suitable for high throughput screening analysis. Meanwhile, the labeled mycotoxins with d 3 -MASC as internal standards could effectively eliminate matrix interference and ensure the accuracy and precision of mass spectrometry analysis. The limits of detection (LODs) and the limits of quantification (LOQs) ranged from 0.29 to 0.42 μg/L and 0.85 to 1.30 μg/L, respectively. The recoveries were 66.9 %–85.2 %, and the daily and inter day RSDs were within the range of 2.0 %–4.9 % and 2.4 %–5.0 %, respectively. The established method could be feasibility applied to the determination of mycotoxins in legume samples with satisfactory results.

Abstract:Mycotoxins have the potential to enter the human food chain through carry-over of contaminants from feed into animal-derived products. The objective of the study was to develop a reliable and sensitive method for the analysis of 30 mycotoxins in animal feed and animal-derived food (meat, edible animal tissues, and milk) using liquid chromatography–tandem mass spectrometry (LC–MS/MS). In the study, three extraction procedures, as well as various cleanup procedures, were evaluated to select the most suitable sample preparation procedure for different sample matrices. In addition, timed and highly selective reaction monitoring on LC–MS/MS was used to filter out isobaric matrix interferences. The performance characteristics (linearity, sensitivity, recovery, precision, and specificity) of the method were determined according to Commission Decision 2002/657/EC and 401/2006/EC. The established method was successfully applied to screening of mycotoxins in animal feed and animal-derived food.
