Abstract:NO 2 is a well-known indoor and outdoor pollutant that may cause adverse health problems. Recently, accumulating but extremely limited evidences show that NO 2 possibly express neurotoxicity and is responsible for various neurological disorders. In the present study, neurological hazard of NO 2 and possible mechanisms were determined in rat pallium following a repetitive inhalation exposure with various concentrations (5, 10 and 20mg/m 3). After 7-day exposure (6h/day), observable adverse effects were induced encompassing decreased ratio of brain to body weight, mild brain pathology, increased neuronal apoptosis, altered antioxidants (Cu/Zn-SOD, Mn-SOD, GPx and NO) activity and increasing formation of PCO. NO 2 inhalation also induced augment of oncogenes (c-fos, c-jun) levels, and deregulation of apoptosis-related genes (p53, bax and bcl-2) expression. With all above data, the present report provided essential information for the characterization of the neurotoxic hazard of NO 2 and related mechanisms, which is required in response to the general concern about the vulnerability of the neurological system to it. © 2012 Elsevier B.V.

Abstract:Objective] To explore the combined neurotoxicity of volatile organic compounds(VOCs) exposure in mice. [Methods] The gaseous mixture of several main compounds of VOCs and formaldehyde were prepared using their pure chemical reagents.The Kunming mice were sub-acutely exposed to the prepared gaseous mixture(three groups of formaldehyde at a dose of 1.5 mg/m3,4.5 mg/m3 or 13.5 mg/m3;three groups of VOCs at a dose of 20 mg/m3,60 mg/m3 or 180 mg/m3).Neurobehavioral function tests include spatial water maze,step-down test,step-through test and pontaneous activity measurement. [Results] Significant differences were observed in all tests between exposed groups and the control group. [Conclusion] It is suggested that volatile organic compounds exposure could damage the ability of learning and memory in mice.

Abstract:Objective:To explore central nervous system damage to astrocyte and possible involved machenisms of volatile organic compounds in vitro.Methods:Astrocytes cell from neonate rats were exposed to VOCs at concentrations of 0、1、3、9mmol/L,respectively.Cell viability,LDH were examined.Result:The result showed that there were morphological changes and reduction of viability in the cells exposed to VOCs compared with non-exposed cells.Conclusion:It was indicated that the injury of astrocyte could have an important effect for the toxicity of central nervous system induced by VOCs.

Abstract:Organophosphate flame retardants (OPFRs) have become a growing concern due to their potential environmental and health risk. However, limited studies have described the toxicity, particularly neurotoxicity of alkyl and aromatic OPFRs. This study investigated the neurotoxicity of alkyl tri‐n‐butyl phosphate (TnBP) and aromatic tricresyl phosphate (TCP) to rat adrenal pheochromocytoma (PC12) cells for 24 h. Viability detection showed dose‐response toxicity effect of TCP and TnBP to PC12 cells. The half‐maximal inhibitory concentration of 24 h (24 h‐IC50) of TCP and TnBP were 2415.61 and 338.09 μM, respectively. Both TnBP and TCP significantly changed the acetylcholinesterase (AChE) activity, and TnBP is more likely to cause neurotoxicity to PC12 cells compared to TCP. Also, The results of LDH and caspase‐3 activity detection as well as Hoechst staining suggested that cell apoptosis induced by TCP and TnBP may be the primary pathway. These findings provide a toxicity data of aromatic and alkyl‐substituted OPFRs to PC12 cells, and a new insight into the toxicity of OPFRs on health risk assessment.

Abstract:Exposure to traffic-related air pollution and ultrafine particles (<100 μm; UFP) is linked with neurodegeneration. However, the impact of the aromatic content in fuels and the contribution of different fractions of UFP, i.e. , solid UFP vs SVOC UFP, on neuronal function is unknown. We therefore studied effects on neuronal activity and viability in rat primary cortical cells exposed for up to 120 h to copper oxide particles (CuO) or UFP (solid and SVOC) emitted from a heavy-duty diesel engine fueled with petroleum diesel (A20; 20 % aromatics) or Hydrotreated Vegetable Oil-type fuel (A0; 0.1 % aromatics), or solid UFP emitted from a non-road Kubota engine fueled with A20. Moreover, effects of UFP and CuO upon simulated inhalation exposure were studied by exposing an lung model (Calu-3 and THP-1 cells) for 48 h and subsequently exposing the cortical cells to the medium collected from the basal compartment of the lung model. Additionally, cell viability, cytotoxicity, barrier function, inflammation, and oxidative and cell stress were studied in the lung model after 48 h exposure to UFP and CuO. Compared to control, direct exposure to CuO and SVOC UFP decreased neuronal activity, which was partly associated with cytotoxicity. Effects on neuronal activity upon direct exposure to solid UFP were limited. A20-derived UFP (solid and SVOC) were more potent in altering neuronal function and viability than A0 counterparts. Effects on neuronal activity from simulated inhalation exposure were minor compared to direct exposures. In the lung model, CuO and A20-derived UFP increased cytokine release compared to control, whereas CuO and SVOC A20 altered gene expression indicative for oxidative stress. Our data indicate that SVOC UFP exhibit higher (neuro)toxic potency for altering neuronal activity in rat primary cortical cells than the solid fraction. Moreover, our data suggest that reducing the aromatic content in fuel decreases the (neuro)toxic potency of emitted UFP.

Abstract:Newly synthesized chemicals are being introduced into the environment without undergoing proper toxicological evaluation, particularly in terms of their effects on the vulnerable neurodevelopment. Thus, it is important to carefully assess the developmental neurotoxicity of these novel environmental contaminants using methods that are closely relevant to human physiology. This study comparatively evaluated the potential developmental neurotoxicity of 19 prevalent environmental chemicals including neonicotinoids (NEOs), organophosphate esters (OPEs), and synthetic phenolic antioxidants (SPAs) at environment-relevant doses (100 nM and 1 μM), using three commonly employed in vitro neurotoxicity models: human neural stem cells (NSCs), as well as the SK-N-SH and PC12 cell lines. Our results showed that NSCs were more sensitive than SK-N-SH and PC12 cell lines. Among all the chemicals tested, the two NEOs imidaclothiz (IMZ) and cycloxaprid (CYC), as well as the OPE tris(1,3-dichloro-2-propyl) phosphate (TDCIPP), generated the most noticeable perturbation by impairing NSC maintenance and neuronal differentiation, as well as promoting the epithelial-mesenchymal transition process, likely via activating NF-κB signaling. Our data indicate that novel NEOs and OPEs, particularly IMZ, CYC, and TDCIPP, may not be safe alternatives as they can affect NSC maintenance and differentiation, potentially leading to neural tube defects and neuronal differentiation dysplasia in fetuses.

Abstract:The neurotoxicity of n-pentane, n-hexane, and n-heptane have been studied in Wistar strain male rats after exposure to 3000 ppm of n-pentane, n-hexane, or n-heptane for 12 hours a day for 16 weeks. The nerve conduction velocity and the distal latency were measured before the beginning of the exposure and after exposure for four, eight, 12, and 16 weeks. The experiment showed that n-hexane disturbed the conduction velocity of the motor nerve and the mixed nerve and prolonged the distal latency in the rat's tail, but that n-pentane and n-heptane did not. The light and electron microscopic examination showed that the peripheral nerve, the neuromuscular junction, and the muscle fibre of the rats exposed to n-hexane were severely impaired, but those of the rats exposed to n-pentane or n-heptane showed no particular changes even after 16 weeks of exposure. These results show that n-hexane is far more toxic to the peripheral nerve of the rat than n-pentane or n-heptane. It is necessary to study the neurotoxicity of other petroleum hydrocarbons, since some reports suggest that petroleum solvents might possibly contain neurotoxic hydrocarbons other than n-hexane.

Abstract:Objective This study aimed to investigate the associations between exposure to blood volatile organic compounds (VOCs) and the level of serum neurofilament light chain (NfL) in adults. Methods We analyzed data from the 2013–2014 National Health and Nutrition Examination Survey (NHANES), including 2008 participants aged 20 to 75 years old. Multiple linear regression models were used to examine the associations between 28 VOCs and NfL after adjusting for multiple potential confounders. Restricted cubic spline (RCS) was used to examine the potential non-linear associations. Results The linear regression models showed that higher levels of 2,5-dimethylfuran (β = 0.042, 95 % confidence interval [CI]: 0.001, 0.096), ethyl acetate (β = 0.118, 95 % CI = 0.008, 0.304), and m-/p-xylene (β = 0.043, 95%CI = 0.012, 0.074) were associated with higher NfL levels. These estimates were largely consistent after adjusting for multiple confounders. Conclusion The findings of our study suggest a potential association between certain volatile organic compounds (2,5-dimethylfuran, ethyl acetate, and m-/p-xylene) and blood NfL levels, implying that they may have a role in revealing neurodegeneration and influencing neurological health.

Abstract:Plants produce a wide variety of natural volatile organic compounds (NVOCs), many of which are unique to each species. These compounds serve many purposes, such as fending off herbivores and adapting to changes in temperature and water supply. Interestingly, although NVOCs are synthesized to deter herbivores, many of these compounds have been found to possess several therapeutic qualities, such as promoting nerve stability, enhancing sleep, and suppressing hyperresponsiveness, in addition to acting as antioxidants and anti-inflammatory agents. Therefore, many NVOCs are promising drug candidates for disease treatment and prevention. Given their volatile nature, these compounds can be administered to patients through inhalation, which is often more comfortable and convenient than other administration routes. However, the development of NVOC-based drug candidates requires a careful evaluation of the molecular mechanisms that drive their therapeutic properties to avoid potential adverse effects. Furthermore, even compounds that appear generally safe might have toxic effects depending on their dose, and therefore their toxicological assessment is also critical. In order to enhance the usage of NVOCs this short review focuses not only on the biological activities and therapeutic mode of action of representative NVOCs but also their toxic effects.

Abstract:Nitric oxide (NO) induces apoptosis in neuronal cells, and has been implicating in a variety of neuronal pathological process. Thus, there is much interest in identifying natural substances which have protective effects against damage induced by nitrosative stress. The roots of Vitis amurensis have been used as traditional medicine and contain structurally diverse resveratrol oligomers with various biological activities. However, there have been few studies on the protective effect of resveratrol oligomers against neurotoxic reactive nitrogen species. In this study, we evaluated the protective effects of two resveratrol oligomers from V. amurensis, vitisin A and heyneanol A, against NO-induced toxicity. Additionally, their antioxidant activities were determined by measuring NO and hydroxyl radical scavenging ability. Both vitisin A and heyneanol A reduced cell death and DNA fragmentation induced by sodium nitroprusside in SH-SY5Y cells. The present study indicates that radical scavenging activities of vitisin A and heyneanol A contribute to protecting neuronal cells against nitrosative stress.

Abstract:Volatile anesthetics are one class of the most commonly used drugs. However, the mechanisms for these drugs to induce anesthesia are not fully understood and have been under intensive investigation. Two other effects of these anesthetics on the central nervous system, volatile anesthetics-induced neuroprotection and neurotoxicity, currently are hot research fields. Although data from animal studies for these two effects are extensive and convincing, clinical data for volatile anesthetics-induced neuroprotection are relatively weak. There is essentially lack of evidence to suggest volatile anesthetics-induced neurotoxicity in humans. In this regard, the contribution of general anesthesia/anesthetics to postoperative cognitive decline, a clinical entity whose existence has been supported by substantial evidence, also has not been established. This paper will be focused on reviewing the evidence, especially the clinical evidence, for volatile anesthetics-induced neuroprotection and neurotoxicity. Efforts will be devoted to facilitating the understanding of the two seemingly contradictory effects of these important drugs on the brain.

Abstract:OBJECTIVE:Study adverse effects of volatile organic compounds volatile organic compounds (VOCs) on brain amino acid neurotransmitter in mice.METHODS:Four groups of mouse were respectively exposed to 0, low, middle and high concentration of VOCs for two hours every day for 30 days. The content of brain amino acid neurotransmitter was determined.RESULTS:The result showed that the content of Glu and ASP in brain of VOCs -exposed mouse decreased and GABA and Gly increased compared with that of control.CONCLUSION:The memory impair in mice induced by exposing VOCs may relate to the chang of content of amino acid neurotransmitter.
A brief exposure to toluene vapor alters the intrinsic excitability of D2 medium spiny neurons in the rat ventral striatum

Abstract:Although volatile organic solvents such as toluene are used for commercial and industrial uses, they are often voluntarily inhaled for their intoxicating and euphoric effects. Research into the effects of inhalants such as toluene on brain function have revealed actions on a variety of ligand-gated and voltage-activated ion channels involved in regulating neuronal excitability. Previous work from this laboratory has also shown that brief exposures to toluene vapor induce changes in the intrinsic excitability and synaptic transmission of neurons within the medial prefrontal cortex and ventral tegmental area that vary depending on projection target. In the present study, we recorded current-evoked spiking of medium spiny neurons (MSNs) in the nucleus accumbens (NAc) core and shell in adolescent rats exposed to an intoxicating concentration of toluene vapor. Compared to air controls, firing of NAc core MSNs in Sprague–Dawley rats was not altered 24 h after exposure to 10,500 ppm toluene vapor while spiking of NAc shell MSNs was enhanced at low current steps but reduced at higher current steps. When the rheobase current was used to putatively identify MSN subtypes, both "D1-like" and "D2-like" MSNs within the NAc shell but not core showed toluene-induced changes in firing. As toluene may itself have altered the rheobase resulting in misclassification of neuron subtype, we conducted additional studies using adolescent D2-Cre rats infused with a Cre-dependent mCherry reporter virus. Following toluene vapor exposure, spiking of NAc shell D2+ MSNs was enhanced at low current steps but inhibited at higher currents as compared to air controls while there were no differences in the firing of NAc shell D2- MSNs. The toluene-induced change in NAc D2+ shell MSN firing was accompanied by alterations in membrane resistance, rheobase, action potential rise time and height with no changes noted in D2- MSNs. Overall, these data add to a growing literature showing that brief exposures to intoxicating concentrations of toluene vapor causes selective alterations in the excitability of neurons within the addiction neurocircuitry that vary depending on sub-region, cell-type and projection target.

Abstract:In recent years, the nail industry has been widely popular in China, and the use of nail care products has also significantly increased. Due to its high content of volatile solvents, the released VOCs not only have a negative impact on indoor air quality but also pose a health threat to nail salon workers who are highly exposed to such environments. The objectives of this research were to characterize VOCs emissions from detailed manicure processes and to evaluate the impact on the environment and health risks. Results showed that the VOCs concentration in the anti-warping treatment process was much higher than that in other manicure steps, at 360.69 mg/m 3 , making its contributions of OFP and SOA equally prominent. The trend of concentration contribution was similar to that of OFP, and OVOCs were the most significant contributor to the VOCs components in the whole manicure process. Since organic solvent nail gels were also frequently used in most steps, the main VOCs were methanol, ethanol and ethyl acetate. Aromatics were the component that contributed the most to SOA, and its contributions in all processes were >85 %. Health risk assessments performed in our study indicated that acrolein was the main non-carcinogen, and the carcinogenic risk of this study could be ignored. The results of this study can be used as a basis for controlling VOCs emission and reducing exposure to VOCs in nail salons.

Observation of general and specific adverse effects of organic solvents on by toxicity testing using behavioral analysis

Abstract:Novel chemical compounds are continuously being developed and are used in various industries as well as in our daily lives. Workers in industries estimate and avoid chemical hazardous risks based on published information on chemical toxicities. However, it is challenging to avoid hazards caused by chemicals with unknown toxicity. Therefore, it is necessary to understand the toxicities of chemicals in high-throughput and multifaceted ways. In this study, I develop a high-throughput method to assess chemical toxicities through the quantitative measurement of behavior in . I determine the acute toxicity of 30 organic solvents that are widely used in industries with motility as an endpoint. The adverse effects of organic solvents on motility were proportional to the lipid solubility of chemicals, which is similar to the positive relationship between the anesthetic effects of volatile organic chemicals and their lipid solubility in organisms, including humans. In addition to the general toxicity on motility, organic solvents caused adverse effects on posture during locomotion in different ways depending on the chemical’s functional group. Alcohols and cellosolves, both of which have a hydroxyl group, reduced the amplitude of body bending, whereas ketones and acetate esters, both of which have a ketone group, increased it during undulatory locomotion. Furthermore, organic solvents caused defects in behavioral plasticity based on the association of starvation and chemical signals at concentrations lower than those that affect locomotion, which suggests that sensory processing is more susceptible to organic solvents than motor function. This study provides a high-throughput method for acute chemical toxicity testing and new insights into the behavioral toxicity of organic solvents based on comprehensive behavioral analysis.

Abstract:The influence of volatile organic compounds (VOCs) on cognitive performance has been widely recognized but is still imperfectly understood. This article hypothesized that the influence of low concentrations of VOCs on cognitive function would occur without triggering subjective awareness. Toluene is a representative VOC commonly found in Chinese dwellings. A within-subject experiment was conducted to compare working memory performance and electroencephalographic patterns of participants exposed to toluene at 70 ppb (Tol group), a WHO-recommended toluene exposure limit, and at 0 ppb (Ctrl group) in a controlled environment chamber. Results demonstrated that toluene exposure at the ppb level could change neural arousal patterns during working memory tasks in healthy subjects, although there was no significant difference in task performance. Compared with the Ctrl group, power spectral density analysis showed that the alpha and theta bands in the frontal region of the Tol group increased considerably in both the encoding and maintenance periods (P < 0.01 for theta, P < 0.05 for alpha). The direct transfer function indicated that the functional connectivity of the frontal network in the theta band, gamma band, and alpha band significantly decreased (P < 0.01 for all bands above). In contrast, the functional connectivity of the frontoparietal network increased under toluene exposure in the theta and alpha bands during the maintenance period (P < 0.01 for both bands). This study proved the feasibility of a potential evaluation method using neurobehavioral tests with electroencephalography (EEG) analysis.

Abstract:Background: Neonicotinoid insecticides are used worldwide for crop protection. They act as agonists at postsynaptic nicotinic acetylcholine receptors (nAChRs), disrupting normal neurotransmission in target insects. Human exposure is high due to the widespread use of neonicotinoids and their residues in food. This study aimed to evaluate the in vitro neurotoxicity of three neonicotinoid commercial formulations Much 600 FS® (imidacloprid 600 g L-1), Evidence 700 WG® (imidacloprid 700 g kg-1), and Actara 250 WG® (thiamethoxam 250 g kg-1) in differentiated human neuroblastoma SH-SY5Y cell line. Methods: Cells were incubated with the pesticides for 96 h, and the cytotoxicity was evaluated through the 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium·bromide (MTT) reduction and neutral red (NR) uptake assays. Toxicological pathways such as reactive oxygen (ROS) and nitrogen species (RNS) production, mitochondrial membrane potential, cell death mode, and the expression of the pro-apoptotic protein Bax were also evaluated. Results: EC50 values of 266.4, 4,175, and 653.2 mg L-1 were found for Much®, Evidence® and Actara®, respectively. Significant increases in ROS and RNS generation were observed for all pesticides, while mitochondrial membrane potential and Bax protein expression showed no significant changes. Analysis of cell death mode revealed an increase in early apoptotic cells. Conclusion: Therefore, neonicotinoid insecticides are potentially neurotoxic, reinforcing concerns about human exposure to these commercial formulations.
A comparison study of volatile organic compounds in hospital and laboratory environments.

Abstract:Air samples were collected at hospitals and laboratories, using SUMMA canisters. 116 kinds of VOCs were identified from indoor and outdoor air of hospitals and laboratories by the pre-concentrator GC/MS system. Alkanes such as ethane, propane and n-butane, alkenes such as ethene, propene and 1-butene, aromatics such as benzene, toluene and ethylbenzene, halohydrocarbons such as chloromethane and dichloromethane, and OVOCs such as acetone and acetaldehyde, were detected frequently from indoor air samples. Those common used reagents, n-hexane, toluene, ethanol and acetone, showed higher concentrations in laboratories air, about 10~1000 times higher than outdoors. High levels of toluene, ethylbenzene and xylenes were found in some waiting areas of hospitals, which would draw more attention.

Abstract:Although many organochlorine pesticides (OCPs) have been banned or restricted because of their persistence and linkage to neurodegenerative diseases, there is evidence of continued human exposure. In contrast, registered herbicides are reported to have a moderate to low level of toxicity; however, there is little information regarding their toxicity to humans or their combined effects with OCPs. This study aimed to characterize the mechanism of toxicity of banned OCP insecticides (aldrin, dieldrin, heptachlor, and lindane) and registered herbicides (trifluralin, triallate, and clopyralid) detected at a legacy contaminated pesticide manufacturing and packing site using SH-SY5Y cells. Cell viability, LDH release, production of reactive oxygen species (ROS), and caspase 3/7 activity were evaluated following 24 h of exposure to the biocides. In addition, RNASeq was conducted at sublethal concentrations to investigate potential mechanisms involved in cellular toxicity. Our findings suggested that aldrin and heptachlor were the most toxic, while dieldrin, lindane, trifluralin, and triallate exhibited moderate toxicity, and clopyralid was not toxic to SH-SY5Y cells. While aldrin and heptachlor induced their toxicity through damage to the cell membrane, the toxicity of dieldrin was partially attributed to necrosis and apoptosis. Moreover, toxic effects of lindane, trifluralin, and triallate, at least partially, were associated with ROS generation. Gene expression profiles suggested that decreased cell viability induced by most of the tested biocides was related to inhibited cell proliferation. The dysregulation of genes encoding for proteins with anti-apoptotic properties also supported the absence of caspase activation. Identified enriched terms showed that OCP toxicity in SH-SY5Y cells was mediated through pathways associated with the pathogenesis of neurodegenerative diseases. In conclusion, this study provides a basis for elucidating the molecular mechanisms of pesticide-induced neurotoxicity. Moreover, it introduced SH-SY5Y cells as a relevant in vitro model for investigating the neurotoxicity of pesticides in humans.

Abstract:The principal health outcome of exposure to toluene is dysfunction of the central nervous system. Effects range from fatalities and severe neurological disorders in toluene abuse situations to deficits in neurobehavioral function in occupational populations. An Inhalation Reference Concentration (RfC) of 0.4 mg toluene/m3 or 0.1 ppm was developed by the U.S. EPA to protect general populations chronically exposed to toluene. The RfC was derived from results of an occupational study involving Asian workers who developed neurobehavioral deficits at a mean toluene exposure level at the time of the study of 88 ppm. The derivation incorporated several uncertainty factors, one of which was a factor of 10 to account for sensitive subpopulations. Recent evidence indicates that some Japanese and possibly other Asian populations harbor a defective gene for aldehyde dehydrogenase, and thus exhibit a decreased rate of toluene metabolism. Although it is not known if reduced metabolism by aldehyde dehydrogenase also was a factor in the occupational study, preshift blood levels of toluene were considerably higher than preshift levels from non-Asian workers exposed to similar air levels of toluene. The elevated blood levels are consistent with defective metabolism but remain to be confirmed. Inasmuch as air levels of toluene in urban environments are about 10-fold lower than the RfC, an adequate measure of protection is afforded by the RfC with or without an uncertainty factor for sensitive subgroups. However, the uncertainty factor for sensitive subgroups should be retained because there is no information regarding toluene metabolism in children.
