Abstract:Hair samples and paired serum samples were collected from e-waste and urban areas in Wenling of Zhejiang Province, China. The PBDE and DBDPE concentrations in hair and serum samples from e-waste workers were significantly higher than those of non-occupational residents and urban residents. BDE209 was the dominating BFRs in hair and serum samples from the e-waste area, while DBDPE was the major BFRs from the urban area. Statistically significant correlations were observed between hair level and serum level for some substances (BDE209, DBDPE, BDE99, BDE47, BDE28, and BDE17), although the PBDE congener profiles in hair were different from those in the serum. A statistically significant positive correlation between the PBDE concentrations and the working age, as well as gender difference, was observed in e-waste workers. Different sources of PBDEs and DBDPE in three groups were identified by principal component analysis and spearman correlation coefficient. Hair is suggested to be a useful matrix for biomonitoring the PBDE exposure in humans.

Abstract:We detected the polybrominated diphenyl ethers (PBDEs) or decabromodiphenyl ethane (DBDPE) in paired hair-serum and nail-serum samples collected from the corresponding chemical manufacturing workers. The levels of decabrominated diphenyl ether (BDE-209) or DBDPE in the serum, hair and nail samples were all significantly higher than those reported in other studies, and the "work place" (pretreatment or posttreatment workshop) was the primary influencing factor that affected the levels of specific BFRs in vivo. For BDE-209 workers, the BDE-209 in both the hair and nail samples were significantly and positively related to the BDE-209 in the serum, indicating that both hair and nails can be used as noninvasive biomatrices to reflect internal exposure to BDE-209. In DBDPE workers, hair rather than nails was more suitable for use as a noninvasive biomatrix to infer the DBDPE exposure level. A series of serum biomarkers reflecting thyroid hormones and liver and kidney injuries were tested to calculate the correlations between hair or nail BFR levels and the levels of the biomatrices. The BDE-209 in the hair samples was significantly and positively correlated with the total protein (TP), and the nail BDE-209 level was significantly and positively related to the total bilirubin (TBIL), indirect bilirubin (IDBIL) and uric acid (UA). The DBDPE in hair was significantly and positively correlated with the thyroid hormones free triiodothyronine (fT3) and total triiodothyronine (tT3) and kidney injury markers, including blood urea nitrogen (BUN), creatinine (CRE) and cystatin C (Cys-C). In addition, the nail DBDPE levels were significantly and positively correlated with the albumin/globulin (A/G), BUN, CRE and Cys-C but negatively correlated with the TP and globulin (GLO). Our findings provide preliminary evidence that hair and nails can be used as noninvasive biomatrices for assessing internal BFR exposure and health damage in occupational workers.

Abstract:The large usage of Tetrabromobisphenol A (TBBPA) in consumer products leads to ubiquitous distribution globally, however, studies on the occurrence of their debromination compounds were rather scarce. Also, though many studies illustrate the effectiveness of hair analysis to assess human exposure to organic pollutants, evidence on the associations with health implications is still fairly limited. Herein, 598 participants from across China were employed to investigate chronic, low-level exposure to TBBPA and debromination products by hair analysis. The geomean concentrations of TBBPA, 2,2′,6-tribromobisphenol A (Tri-BBPA), 2,2′- and 2,6-dibromobisphenol A (Di-BBPA), and 2-monobromobisphenol A (Mo-BBPA) were 1.07, 0.145, 0.135, and 0.894 ng/g, respectively, indicating nonnegligible health risks of debromination products. Hair analyte levels correlated with population age and population density among sampling regions. Sexual- and spatial-variations were observed with higher concentrations in females and in E -waste recycling sites. Logistic regression models showed that TBBPA exposure (adjusted odds ratio (OR): 1.02, 95 % confidential interval (CI): 1.01–1.05) was positively associated with risk of metabolic syndrome by adjusting for various covariates. These findings imply usefulness of hair as an alternative biomonitoring tool to assess human exposure to TBBPA and relative health effects, which highlights public concerns on co-exposure to these chemicals.

Abstract:Dechlorane plus (DP) has been detected in a variety of environmental media and in human. Measurement of DPs in hair, urine, and house dust across different habitats allows for the assessment of short-term spatial changes in human exposure to DPs, as well as their excretion in urine. This offers a significant reference point for further research on the behavior of persistent pollutants within organisms. We measured and analyzed the concentrations of DP in the hair and urine of 32 students from a university in Beijing during school and home phases, and in indoor dust from dormitories and some home environments. The results indicated that the concentrations of DP in three types of samples were higher during the home phase compared to the school phase. We compared the fanti values and identified selective enrichment of syn-DP in hair, along with selective excretion of syn-DP in urine. Utilizing molecular docking technique, we simulated the binding effect between DP and the Megalin protein. The results demonstrated that the binding energy of anti-DP to Megalin was higher than that of syn-DP, suggesting that anti-DP has a greater propensity to bind to Megalin and be reabsorbed. This results in higher levels of syn-DP excretion in urine. Finally, we categorized students based on their participation in the organic exposure experiment and their BMI. The results indicated that the concentrations of DP in hair and urine were higher in the exposed group compared to the non-exposed group during the school year. After excluding the effect of exposure, habitat changes were more likely to affect the accumulation and excretion of DP in normal-weight students (BMI ≤ 24 kg/m2, n=28), while overweight students (BMI > 24 kg/m2, n=4) were less affected by the effect of habitat because of their higher body fat percentage and their greater ability to accumulate DP.

Abstract:Polybrominated diphenyl ethers (PBDEs) are becoming a public health concern because of their potential toxicity, from endocrine disruption system to neurodevelopmental impairments. Nonetheless, information on their levels in human blood is scarce. In this study, human serum samples collected in Shanghai, China, were analyzed for the concentrations of PBDEs and their hydroxylated metabolites (OH-PBDEs). Eight PBDE congeners and six OH-PBDE congeners were quantified in serum samples by gas chromatography with mass spectrometry (GC-MS) and high-performance liquid chromatography with tandem mass spectrometry (HPLC-MS/MS). As a result, total PBDE concentration ranged from 0.280 to 12.330 ng g−1 on a lipid weight basis lw (median: 1.100 ng g−1 lw) and the total OH-PBDE level ranged from 0.045 to 0.363 ng g−1 (lw) (median: 0.187 ng g−1 lw). Among them, BDE-47 and 6-OH-BDE-47 were the predominant PBDEs and OH-PBDEs, respectively. In addition, based on the results of the Bartelett X2 test, BDE-47 significantly (p < 0.05) correlated with BDE-28, BDE-100, BDE-85, and BDE-154, whereas 3′-OH-BDE-7 significantly (p < 0.01) correlated with 3-OH-BDE-47, 2-OH-BDE-68, and 6′-OH-BDE-99. Among all donors, no significant association between age and PBDEs (or OH-PBDEs) was found. Further research on the exposure routes in the environment and metabolic processing of PBDEs in human blood is necessary.

Abstract:Humans are exposed to various anthropogenic chemicals in daily life, including endocrine-disrupting chemicals (EDCs). However, there are limited data on chronic, low-level exposure to such contaminants among the general population. Here hair analysis was used to investigate the occurrence of four polychlorinated biphenyls (PCBs), seven polybrominated diphenyl ethers (PBDEs) and two bisphenols (BPs) in 204 Chinese women living in the urban areas of Baoding and Dalian and 311 pregnant French women. All the PCBs and PBDEs tested here were more frequently detected in the hair samples of the French women than in those of the Chinese women. In both cohorts, PCB 180 and BDE 47 were the dominant PCB and PBDE congener, respectively. PCB 180 was found in 82% of the French women and 44% of the Chinese women, while the corresponding values of BDE 47 were 54% and 11%, respectively. A discriminant analysis further demonstrated the difference in PCBs and PBDEs exposure profile between the two cohorts. These results demonstrate that hair analysis is sufficiently sensitive to detect exposure to these pollutants and highlight differences in exposure between populations even at environmental levels. Although BPA and BPS were found in 100% of the hair samples in both cohorts, the French women had significantly higher levels of BPA and BPS than the Chinese women. The median concentrations of BPA were one order of magnitude higher than BPS in both the Chinese (34.9 versus 2.84 pg/mg) and the French women (118 versus 8.01 pg/mg) respectively. Our results suggest that both French and Chinese populations were extensively exposed to BPA and BPS.

Abstract:Human hair has been identified as a non-invasive alternative matrix for assessing the human exposure to specific organic contaminants. In the present study, a solvent-saving analytical method for the simultaneous determination of 8 polybrominated diphenyl ethers (PBDEs), 3 hexabromocyclododecanes (HBCDDs), 12 phosphorus flame retardants (PFRs), and 4 emerging PFRs (ePFRs) has been developed and validated for the first time. Hair sample preparation protocols include precleaning with Milli-Q water, digestion with HNO3/H2O2 (1:1, v/v), liquid–liquid extraction with hexane:dichloromethane (4:1, v/v), and fractionation and cleanup on a Florisil cartridge. The method was validated by using two levels of spiked hair samples of 3 replicates for each spiking group. Limits of quantification (LOQs) were 0.12−22.4 ng/g for all analytes, average values of accuracies were ranging between 88−115%, 82−117%, 81−128%, and 81−95% for PBDEs, HBCDDs, PFRs, and ePFRs, respectively; and precision was also acceptable (RSD < 20%) for all analytes. Eventually, this method was applied to measure the levels of the targeted analytes in hair samples of e-waste dismantling workers (n = 14) from Qingyuan, South China. Median values ranged between 3.00−18.1 ng/g for PBDEs, 0.84−4.04 ng/g for HBCDDs, 2.13−131 ng/g PFRs, and 1.49−29.4 ng/g for ePFRs, respectively. PFRs/ePFRs constitute the major compounds in human hair samples, implying the wide use of PFRs/ePFRs as replacements of PBDEs and HBCDDs, as well the potential high human exposure risks of PFRs/ePFRs. Overall, this work will allow to a comprehensive assessment of human exposure to multiple groups of FRs using hair as a non-invasive bioindicator.

Abstract:Polybrominated diphenyl ethers (PBDEs) have been used as brominated flame retardants worldwide and are correlated with extensive environmental pollution and human health concerns. This study is aimed at analyzing the concentrations of PBDEs and at evaluating their temporal trends among a population of blood donors ( n = 33) over a 4-year period. A total of 132 serum samples were used for PBDE detection. Nine PBDE congeners were quantified in serum samples by gas chromatography with mass spectrometry (GC–MS). The median concentrations of Σ 9 PBDEs in each year were 33.46, 29.75, 30.85, and 35.02 ng/g lipid, respectively. Most of the PBDE congeners showed a downward trend from 2013 to 2014 and then increased after 2014. No correlations between age and PBDE congener concentrations were observed, while concentrations of each congener and Σ 9 PBDE were nearly always lower in females than in males, especially in BDE-66, BDE-153, BDE-183, BDE-190, and Σ 9 PBDE. We also found that the intake of fish, fruit, and eggs in the daily diet was related to the exposure level of PBDEs. Our results suggest that, as deca-BDE is still produced and used in China, diet is an important exposure pathway for PBDEs, and follow-up studies will be required to improve our understanding of the behaviors of PBDE isomers in humans and the exposure levels.

Abstract:One possible source of urinary bromophenol (BP) glucuronide and sulfate conjugates in mammalian animal models and humans is polybromodiphenyl ethers (PBDEs), a group of additive flame-retardants found ubiquitously in the environment. In order to study the correlation between levels of PBDEs in human blood plasma and those of the corresponding BP-conjugates in human urine, concentrations of 17 BDE congeners, 22 OH-BDE and 13 MeO-BDE metabolites, and 3 BPs in plasma collected from 100 voluntary donors in Hong Kong were measured by gas chromatograph tandem mass spectrometry (GC-MS). Geometric mean concentration of ΣPBDEs, ΣOH-BDEs, ΣMeO-BDEs and ΣBPs in human plasma were 4.45 ng g(-1) lw, 1.88 ng g(-1) lw, 0.42 ng g(-1) lw and 1.59 ng g(-1) lw respectively. Concentrations of glucuronide and sulfate conjugates of 2,4-dibromophenol (2,4-DBP) and 2,4,6-tribromophenol (2,4,6-TBP) in paired samples of urine were determined by liquid chromatography tandem triple quadrupole mass spectrometry (LC-MS/MS). BP-conjugates were found in all of the parallel urine samples, in the range of 0.08-106.49 μg g(-1)-creatinine. Correlations among plasma concentrations of ΣPBDEs/ΣOH-BDEs/ΣMeO-BDEs/ΣBPs and BP-conjugates in urine were evaluated by multivariate regression and Pearson product correlation analyses. These urinary BP-conjugates were positively correlated with ΣPBDEs in blood plasma, but were either not or negatively correlated with other organobromine compounds in blood plasma. Stronger correlations (Pearson's r as great as 0.881) were observed between concentrations of BDE congeners having the same number and pattern of bromine substitution on their phenyl rings in blood plasma and their corresponding BP-conjugates in urine.
Levels of Dechlorane Plus and Polybrominated Diphenylethers in Human Milk in Two Canadian Cities

Abstract:Flame retardant dechlorane plus (DP) and several polybrominated diphenylether (PBDE) congeners have been measured in 87 human milk samples collected in two Canadian cities: Kingston and Sherbrooke. The levels of PBDEs in human milk (mean (median), ngg−1 lipid weight=10 (5.9), 4.1 (2.8), 3.0 (1.6), 5.12 (1.6), and 15 (ND) for BDE-47, BDE-99, BDE-100, BDE-153, and BDE-209, respectively) were comparable to those reported in Europe, U.S.A. and China. The levels of DP, with a mean value of 0.98ngg−1 and a median value of 0.60ngg−1 (lipid weight), were two to ten times lower than those of concurrently measured major PBDEs including BDE-209. While there is little difference in the levels of measured contaminants in milk samples collected from the two cities, the contaminants levels in human milk show, indicated by Principal Components Analysis, that DP, deca-BDE, and penta-BDE come from three distinct sources. The mean and median isomer ratio values of DP in milk were 0.67 and 0.69, respectively, very similar to that of DP commercial products.

Abstract:The global presence of Dechlorane Plus (DP) in environmental and human samples highlights the need for further research into its ecological risks and health effects. This study determined DP concentrations in soil and air samples from an old electronic waste dismantling area (Fengjiang) and a new electronic waste dismantling area (Binhai) in Taizhou City, China. Both syn -DP and anti -DP isomers were found in all soil and air samples. The median concentrations of DP in soil and air were 90.5 ng/g and 48.4 pg/m 3 in Fengjiang, and 6.69 ng/g and 28.8 pg/m 3 in Binhai. DP were widespread in the local environment, and in general at moderate levels compared with reports from other electronic waste dismantling areas. The analysis of DP isomer profiles in different seasons and the calculation of related fugacity fraction showed that anti -DP perhaps easier than syn -DP to migrate from soil to air in summer. DP concentrations in serum of residents from Fengjiang (median 9.74 ng/g) were significantly higher than those from Huangyan (common control area, median 2.77 ng/g; p < 0.01), while the f anti value showed that Fengjiang was significantly lower than Huangyan ( p < 0.05). This revealed that DP exposure levels in serum perhaps one of the crucial factors influencing f anti value. Moreover, DP concentrations in serum showed an increasing trend with increasing body mass index (BMI) or age. However, the relationship between DP and sex was not observed. The f anti values of serum samples were higher than those in soil and air samples, suggested that DP perhaps stereo-selectively absorbed into the body or stereo-selectively metabolised.

Human Biomonitoring of Serum Polybrominated Diphenyl Ethers by Supported Liquid Extraction and Gas Chromatography Coupled With Tandem Mass Spectrometry

Abstract:This study aims to develop and validate a robust analytical method for the quantification of polybrominated diphenyl ethers (PBDEs) in human serum using gas chromatography–tandem mass spectrometry. We compared procedural blanks, recoveries, and operational convenience of liquid–liquid extraction and supported liquid extraction for the determination of serum PBDEs. We evaluated different extraction solvents for their effect on PBDE recoveries. Supported liquid extraction was selected for method validation due to its operational convenience. The method demonstrated satisfactory linearity, sensitivity, and reproducibility, with the range of 0.10–5.00 μg/L for most PBDE congeners and 0.20–10.0 μg/L for PBDE‐154 and PBDE‐183, with limits of detection ranging from 2 to 48 ng/L, and with matrix effects ranging from 94% to 113%. Quality control assessments indicated that recoveries ranged from 85% to 110% and relative standard deviations of less than 11%. The proposed method was applied to biomonitoring of 111 healthy adults, revealing detectable levels of PBDEs in over 90% of the samples. BDE‐47 and BDE‐183 were the most prevalent, with mean concentrations of 4.13 and 22.1 ng/L, respectively. Detection frequencies ranged from 0.90% for BDE‐17 and BDE‐85 to 25.2% for BDE‐47. Males had higher mean concentrations of BDE‐183 than females.

Abstract:Commonly, analytical methods measuring brominated flame retardants (BFRs) of different chemical polarities in human serum are labor consuming and tedious. Our study used acidified diatomaceous earth as solid-phase extraction (SPE) adsorbent and defatting material to simultaneously determine the most abundant BFRs and their metabolites with different polarities in human serum samples. The analytes include three types of commercial BFRs, tetrabromobisphenol A (TBBPA), hexabromocyclododecane (HBCD) isomers, and polybrominated biphenyl ethers (PBDEs), and dominant hydroxylated BDE (OH-PBDE) and methoxylated BDE (MeO-PBDE) metabolites of PBDEs. The sample eluents were sequentially analyzed for PBDEs and MeO-BDEs on online gel permeation chromatography/gas chromatography-electron capture-negative ionization mass spectrometry (online GPC GC-ECNI-MS) and for TBBPA, HBCD, and OH-BDEs on liquid chromatography-tandem mass spectrometry (LC-MS/MS). Method recoveries were 67–134% with a relative standard deviation (RSD) of less than 20%. Method detection limits (MDLs) were 0.30–4.20 pg/mL fresh weight (f.w.) for all analytes, except for BDE-209 of 16 pg/mL f.w. The methodology was also applied in a pilot study, which analyzed ten real samples from healthy donors in China, and the majority of target analytes were detected with a detection rate of more than 80%. To our knowledge, it is the first time for us in effectively determining BFRs of most types in one aliquot of human serum samples. This new analytical method is more specific, sensitive, accurate, and time saving for routine biomonitoring of these BFRs and for integrated assessment of health risk of BFR exposure.

Abstract:A method for the determination of 14 polybrominated diphenyl ethers (PBDEs) in human serum using isotope internal standard-gas chromatography-high resolution dual-focus magnetic mass spectrometry (GC-HRMS) was developed. After thawed to room temperature, 0.5 mL serum samples were mixed with 13C-labeled isotopic internal standard. Subsequently, methanol was added to precipitate the proteins in the samples. The effects of three kinds of acids on the removal of cellulite from the serum samples and the corresponding recoveries were compared, and the results revealed that sulfuric acid was the most optimal. The target compounds were extracted by liquid-liquid extraction (LLE), and the effects of different extraction solvents on recoveries were compared. The results indicated that n-hexane (6 mL)-methyl tert-butyl ether (6 mL) was the best extraction solvent. The extracts were cleaned and eluted using solid phase extraction cartridges. Furthermore, the factors that influenced the cleanup effects and recoveries, including the solid phase extraction columns and elution solvents, were investigated in detail. The results indicated that the optimal conditions were cleanup with a silica gel column and elution with hexane-dichloromethane (1∶1, v/v). The eluate was re-dissolved in hexane after being blown to near dryness using nitrogen. The detection of PBDEs was performed using GC-HRMS. The instrument conditions were optimized, and the capillary column used was an Rtx-1614 column (30 m×0.25 mm×0.1 μm). Helium was used as the carrier gas at a flow rate of 1.0 mL/min. The injector temperature was 290 ℃, and the oven temperature was programmed as follows: 150 ℃ for 2 min, 150 ℃ to 250 ℃ at 15 ℃/min, held for 1 min, 250 ℃ to 290 ℃ at 25 ℃/min, held for 3 min, and 290 ℃ to 320 ℃ at 25 ℃/min, held for 12.5 min. The injection volume was 1 μL in splitless mode. The samples were ionized in the positive electron ionization (EI) mode at 35 eV. Precursor ions and the production of each compound were identified using a voltage-selective ion detection (VSIR) program with a resolution of 10000. The ionization temperature was set at 280 ℃, and the transmission line temperature was set at 320 ℃. To ensure the integrity of the separation of low-brominated components, the column separation time was shortened, the response of high-boiling components was improved (BDE-190 and BDE-209), the decomposition of BDE-209 on the chromatographic column was effectively prevented, and the requirement of the simultaneous determination of multiple PBDEs was met. The method demonstrated good linearity in the range of 0.40 to 25 μg/L for BDE-209, and 0.08 to 5 μg/L for the other 13 PBDEs, with correlation coefficients greater than 0.995. The method detection limits (MDLs) were in the range of 0.01 to 0.51 μg/L, and the limits of quantification (LOQs) ranged from 0.04 to 1.70 μg/L. The recoveries of the 14 compounds ranged from 75.5% to 120.7%. The intra-day relative standard deviations (RSDs) were within 3.8%-10.9% (n=6) and the inter-day RSDs were within 4.2% to 12.4% (n=6). This method was successfully applied to the determination of 14 PBDEs in 15 serum samples from an adolescent population in an area. Notably, 1.86 to 4.66 ng/g lipid BDE-47 was detected in the serum samples with a detection frequency of 100%, and the other compounds were not detected. The results imply that the adolescent population in this region was exposed to some PBDE. Compared with the existing methods reported, this method has less sample demand and higher sensitivity and accuracy, can simultaneously determine 14 PBDEs, including BDE-209 in human serum, and effectively improve the efficiency of detection. This study offers a new method for studying the impact of polybrominated diphenyl ethers on population health in China.

Abstract:197 words): Animal experiments and some human studies indicate that PBDEs may adversely affect male reproductive function. We collected three serum samples from 27 healthy adult men at 6-month intervals in the FlaRE study and assessed associations between PBDE exposure and reproductive function tests using mixed effects regression models, adjusting for confounders. Geometric mean concentrations of the five most prevalent PBDE congeners (BDE 28, 47, 99, 100, 153) by sampling round (Winter 2010, Summer 2010, Winter 2011) were 25.5 ng/g lipid, 25.5 ng/g lipid, and 21.1 ng/g lipid, respectively. PentaBDEs were significantly and inversely associated with inhibin-B. Among older men in our cohort (n=14, age range: 40 to 65 years old), increased concentration of BDE-100 was associated with a decrease in inhibin-B (p=0.02), an increase in follicular stimulating hormone (FSH) (p=0.02), and non-significant decreases in the inhibin-B/FSH ratio (p=0.16) and the free androgen index (p=0.07). Although we did not examine sperm counts and testicular volume, these outcomes are typically positively correlated with inhibin-B serum levels. These findings suggest that PBDE exposure may decrease testicular function in older men. This presentation will compare our results with other studies and discuss the possible implications of PBDE exposure for male reproductive health.

Abstract:In this study, levels of dechlorane plus (DP) in breast milk and matched adipose tissue samples were measured from 54 women living in Wenling, China. Polybrominated diphenyl ethers (PBDEs) and polychlorinated biphenyls (PCBs) were measured simultaneously for comparison. The levels of ∑DPs/∑PBDEs varied from less than one to several dozens of ng g-1 lipid weight (lw) in matrices and the levels of ∑PCBs varied between several to hundreds of ng g-1 lw. In the same matrix, ∑DPs and ∑PCBs/∑PBDEs showed a significant relationship (p < 0.05), indicating that they shared common sources. Accordingly, there was a strong association of lipid-adjusted concentrations of individual compounds (BDE-209 excluded) between matrices (p < 0.001), suggesting that breast milk could be a proxy for adipose tissue in human bioburden monitoring of these compounds. The predicted lipid-adjusted milk/adipose ratios varied from 0.62 to 1.5 but showed significant differences (p<0.001) between compounds, suggesting a compound-specific transfer between milk lipids and adipose tissue lipids. Specifically, the milk/adipose ratios for syn-DP and anti-DP (-1.40 and 1.3, respectively) were significantly higher than those of CB congeners and hexa/hepta-BDE congeners (p < 0.05). In addition, unlike PCBs/PBDEs (excluding BDE-209), DP's hydrophobicity might not be responsible for its preferable distribution in milk lipids. Instead, the interaction with nonlipid factors played a key role. The fraction of anti-DP between the two kinds of matrices was not significantly different, suggesting that the biochemical transfer processes may not be efficient enough to distinguish DP isomers. Nevertheless, the congener patterns of PCBs/PBDEs gave a clue about the compound-specific transfer between milk and adipose tissue. To our knowledge, this is the first to report the relationships of DP between adipose tissue and breast milk. These results could provide useful and in-depth information on biomonitoring of DP and facilitate the understanding of the accumulation and excretion potentials of DP and its distribution-related mechanism in humans.

Abstract:This study analyzed skin wipe samples from the forehead, palm, forearm, and lower leg of 120 volunteers across different age groups—preschoolers, thresholders, middle-aged, and elderly—with each group comprising 30 individuals with a balanced sex ratio from a city in South China. The research aimed to assess the occurrence, concentration, and associated health risks of polybrominated diphenyl ethers (PBDEs) and polychlorinated biphenyls (PCBs) adsorbed onto human skin from environmental sources. We identified 13 PBDE congeners and 10 PCB congeners across all samples, with average detection frequencies of 48% for PBDEs and 16% for PCBs, with concentrations ranging from non-detectable (ND) to 4200 and ND to 2300 ng/m 2 , respectively. Skin concentrations of both PBDEs and PCBs decreased in the order of face > hand > arm > shank (ANOVA, P < 0.001), suggesting higher exposure to uncovered skin areas than to those covered by clothing, regardless of age or sex. The daily average dose of dermal (DAD derm ) and oral (DAD oral ) for PBDEs spanned from 7.0 × 10 –4 –0.19 ng/kg/d and ND–15 ng/kg/d, respectively, whereas the PCB exposure doses ranged from ND–7.8 ng/kg/d (DAD derm ) and ND–2.0 ng/kg/d (DAD oral ), respectively. Preschool children displayed notably higher DAD oral levels than the other groups ( P < 0.0001), which was attributed to their more frequent hand-to-mouth activity. Preschool boys exhibiting a higher DAD derm ( P < 0.05) and both preschool boys and university women showing elevated DAD oral levels ( P < 0.01 and P < 0.05, respectively). Health risk assessments concluded that the carcinogenic risks from BDE209 and PCBs were within acceptable limits (10 –4 ) for all sampled populations.

Abstract:This study was undertaken to determine levels of halogenated flame retardants (HFRs) on skin surface to provide preliminary exposure estimates from dermal absorption. Skin wipes of palms, back-of-hands, and forearms were collected from 30 participants by using gauze pads soaked in isopropyl alcohol. Eight polybrominated diphenyl ethers (PBDEs), two novel brominated FRs, and two Dechlorane Plus (DPs) isomers were determined. BDE209, Decabromodiphenylethane (DBDPE), and 1, 2-Bis (2, 4, 6-tribromophenoxy) ethane (BTBPE) were most frequently detected. BDE209 showed the highest median level (1760 ng m-2), followed by DBDPE (277 ng m-2) and anti-DP (64 ng m-2). The comparison of levels on palms and back-of-hands revealed no statistical differences, but both were significantly higher than the levels on forearms. Detections of HFRs on arms suggested that skin areas covered by clothing were exposed to these chemicals likewise. BDE209 and DPs levels from three repeated samples showed moderate to strong reliability over 3 months, while the levels of other compounds were not statistically reliable. Males had significant higher levels than females for most HFRs, while participants' age, time from last hand washing to sampling, and other behaviors contributed limitedly to the variations in levels. Daily uptakes of HFRs through dermal absorption were estimated. The estimated median total exposure from palms, back-of-hands, and forearms were 25.9, 1.6, and 7.2 ng d-1 for ∑PBDEs, ∑DPs, and ∑NBFRs, respectively, which were in the same ranges as those from diet and dust ingestion for adults in China, suggesting that dermal absorption would be an important exposure route for HFRs.

Abstract:Polybrominated diphenyl ethers (PBDEs) have been produced in the south coast area of Laizhou Bay, Shandong Province in China, but little is known about the PBDE exposure level of residents to these compounds. We set out to assess potential health risks of PBDEs in the south coast area of the Laizhou Bay by determining the concentrations of PBDEs in serum and breast milk. We measured concentrations of eight PBDE congeners in serum and breast milk. The arithmetic means of Σ8PBDE in pooled serum and breast milk were 613ng/g lipid and 81.5ng/g lipid, respectively. The highest concentration for Σ8PBDE in all serum pools was 1830ng/g lipid from the 41–50year old female group. BDE-209 was the predominant congener, with the mean concentrations of 403ng/g lipid in serum and 45.6ng/g lipid in breast milk, respectively. BDE-209 averagely accounted for 65.8% and 54.2% of the total PBDEs, respectively. Our results suggest that high exposures to PBDEs have led to very high PBDE concentrations in serum and breast milk from the residents living in the south coast area of Laizhou Bay. High PBDE concentrations in human serum, particularly in women, pose a potential public health threat to local residents.

Abstract:Typical halogenated persistent organic pollutants (Hal-POPs), including polybrominated diphenyl ethers (PBDEs), polybrominated biphenyls (PBBs), polychlorinated biphenyls (PCBs), and dichlorodiphenyltrichloroethane (DDT), are a group of ubiquitous organic pollutants with an endocrine disrupting effect. This study evaluated the accumulation and congener profiles of Hal-POPs in the bodies of men who live/work in areas of South China where electronic wastes are collected and managed, especially in their semen samples. The results show that the detection frequency and serum concentrations of Hal-POP congeners within the high-exposure group (HEG) were higher than those of the low-exposure group (LEG). Furthermore, an identical trend was observed for the seminal plasma concentrations of Hal-POPs. The distribution characteristics, such as their mean, median, and discrete values, of PBDE congeners in serum and semen samples from the same subjects were consistent with each other. However, the distribution characteristics of PCB congeners in serum samples were different from those in semen samples. BDE153 was one of the most abundant congeners found in the serum and semen samples; hence, it can be identified as an indicator PBDE congener. Further research is needed to explore the mechanism of Hal-POPs distribution in human semen and serum samples.
