Abstract:In pH 6.6 Britton-Robinson buffer medium, the CdS quantum dots capped by thioglycolic acid could react with aminoglycoside (AGs) antibiotics such as neomycin sulfate (NEO) and streptomycin sulfate (STP) to form the large aggregates by virtue of electrostatic attraction and the hydrophobic force, which resulted in a great enhancement of resonance Rayleigh scattering (RRS) and resonance non-linear scattering such as second-order scattering (SOS) and frequency doubling scattering (FDS). The maximum scattering peak was located at 310 nm for RRS, 568 nm for SOS and 390 nm for FDS, respectively. The enhancements of scattering intensity (DeltaI) were directly proportional to the concentration of AGs in a certain ranges. A new method for the determination of trace NEO and STP using CdS quantum dots probe was developed. The detection limits (3 sigma) were 1.7 ng mL(-1) (NEO) and 4.4 ng mL(-1) (STP) by RRS method, were 5.2 ng mL(-1) (NEO) and 20.9 ng mL(-1) (STP) by SOS method and were 4.4 ng mL(-1) (NEO) and 25.7 ng mL(-1) (STP) by FDS method, respectively. The sensitivity of RRS method was the highest. The optimum conditions and influence factors were investigated. In addition, the reaction mechanism was discussed.
Surface Enhanced Raman Spectra of Cysteamine

Abstract:Raman spectra are usually used to detect substances.However,the intensity of signals is weak,thus the application of the normal Raman spectra in micro detection is limited.Herein Raman spectra of cysteamine by different methods such as the powdered,silver colloid adsorption,nanoscale silver film of cysteamine and nanosilver particles adsorption on slides was attained.Then the silver colloid and silver film were studied by UV-Visible spectrophotometer,scanning electronic microscopy (SEM) and transmission electronic microscopy(TEM).The Raman spectrum of cysteamine was calculated by the Gaussian'03 software.The calculation results are compared with the experimental results.The method was optimized by changing the self-assembly time.

Abstract:A novel method is used for the determination of some aminoglycoside antibiotics (AGs) such as etimicin (ETM), isepamicin (ISP) and amikacin (AMK). It is based on the resonance Rayleigh scattering (RRS) intensities enhanced by AGs-induced CdTe quantum dots aggregation. Under the optimum conditions, the increments in RRS intensity were directly proportional to the concentration of AGs in certain ranges. At the same time, the second-order scattering, the frequency-doubling scattering and the frequency-trebling scattering intensities were also enhanced and their increments were proportional to the concentration of AGs. Among them, the RRS method had the highest sensitivity; the linear ranges and detection limits for ETM, ISP and AMK were 0.085-7.2, 0.0067-1.2, 0.017-6.0 and 0.025, 0.0051, 0.0020 mu g mL(-1). This method was applied to the measurement of AGs in human serum and urine with satisfactory results. In addition, the reaction mechanism and the reasons for the enhancement of RRS are discussed using fluorescence, RRS, transmission electron microscope technology and quantum chemistry method. Resonance Rayleigh Scattering Spectral Method for Determination of Methotrexate and Ceftriaxone with CdTe QDs As the Probe

Abstract:CdTe Quantum Dots(CdTe QDs)were synthesized in an aqueous solution using Cysteamine as a stabilizing agent.The stabilizing agent self-assembled on the surface of CdTe QDs to form supermolecular complex cations with positive charges.In a weak acidic buffer(pH=5.2-5.6),negatively charged methotrexate(MTX)and ceftriaxone(CTRX)could bind with the complex cations to form larger aggregates via electrostatic attraction,resulting consequently increased intensities of resonance rayleigh scattering(RRS)intensity,second-order scattering(SOS)and frequency-doubling scattering(FDS).Under optimum conditions,the increments of RRS intensity(ΔI)were directly proportional to the concentration of MTX and CTRX in certain ranges.The RRS method had the highest sensitivity.The concentrations of MTX and CTRX were found to be in the range from 0.01 to 4.50 μg/mL and 0.03 to 4.20 μg/mL,with a detection limit of 3.06 ng/mL and 8.90 ng/mL,respectively.This method proved to be simple,rapid and sensitive and show a promising prospect of application to the detection of MTX and CTRX in human serum.

Abstract:The widespread usage of antibiotics is leading to an increased negative impact on living organisms due to the consumption of polluted water containing drug residues. The emergence of drug-resistant bacteria has additionally manifested a growing need to identify commonly available medications in water sources. A rapid colorimetric nanosensor is developed in this study to selectively detect Neomycin, an antibiotic of class aminoglycosides. A straightforward reduction method was employed for the formation of Schiff base (4-nitrobenzohydrazide triazole thiazole) capped silver nanoparticles (4NHTS-AgNPs). Upon treatment with Neomycin, the colloidal solution of 4NHTS-AgNPs exhibited an immediate color change from yellow to red while no visible change was observed by addition of other competitive drugs. With an increase in drug concentration, the plasmonic characteristic peak of 4NHTS-AgNPs at 405 nm exhibited a progressive reduction. Furthermore, incubation with NeO resulted in a bathochromic shift and broadening to longer wavelength, specifically to 565 nm. The combined quenching and shift of the plasmonic peak were attributed to the aggregation of 4NHTS-AgNPs induced by the NeO through hydrogen bonding. The 4NHTS-AgNPs based sensor demonstrated a linear dynamic range of 0.02–100 μM, with a limit of detection and limit of quantification of 0.0061 μM and 0.020 μM, respectively. Even in the presence of interfering drugs and for real sample analysis, the selectivity of 4NHTS-AgNPs based sensor towards neomycin remained consistent.

Abstract:Mn-doped ZnS quantum dots (QDs) were prepared successfully by wet chemical route, which were characterized by Dynamic Force Microscope (DFM), X-ray Diffraction (XRD), Inductively Coupled Plasma-Atomic Emission Spectrometry (ICP-AES), Infrared Radiation (IR), Ultraviolet (UV) and Resonance light scattering (RLS). The nanoparticles were water-soluble, chemically stable, non-toxic and biocompatible. Then, a novel method for the determination of Cefotaxime Sodium (CTX) in aqueous solutions has been developed based on the quenching of resonance light scattering (RLS) of ZnS:Mn QDs in the presence of CTX. Under the optimal conditions, with the quenching RLS signals at 604 nm, the linear range of calibration curve was 1 × 10–6–2 × 10–5 mol/L for CTX. The detection limit was 1 × 10–9 mol/L. Compared with other methods, this method was more sensitive, fast and reliable.

Abstract:In a medium of pH=5.6,CdS quantum dots could react with etimicin sulfate to form larger aggregates by virtue of electrostatic attraction and the hydrophobic force,which resulted in a great enhancement in resonance Rayleigh scattering(RRS) and resonance non-linear scattering such as second-order scattering(SOS) and frequency doubling scattering(FDS).The maximum scattering peaks were located at 310 nm for RRS,568 nm for SOS and 320 nm for FDS.The enhancements of scattering intensity(ΔI) were directly proportional to the concentration of etimicin sulfate(ETM) within a certain range.Of the three methods,RRS had the highest sensitivity,its detection limit(3σ) being 1.6 ng/mL.An examination of the effects of coexisting substances showed that the method had good selectivity.Based on these results,a new method,highly sensitive and simple,was developed,which can be used to determinate ETM by CdS-QDs as a probe.The recovery for the determination of ETM in human serum and uric samples was in the range of 94.5%-113.7%.
A SERS-based multiple immuno-nanoprobe for ultrasensitive detection of neomycin and quinolone antibiotics via a lateral flow assay

Abstract:The authors describe an ultrasensitive method for simultaneous detection of neomycin (NEO) and quinolones antibiotics (QNS). It is based on the use of (a) two immuno-nanoprobes (a probe for NEO and a probe for QNS), (b) surface-enhanced Raman scattering (SERS) detection, and (c), a portable lateral flow assay (LFA). The two probes consist of gold nanoparticles (AuNPs) conjugated to the Raman active molecule 4-aminothiophenol (PATP), and to monoclonal antibody against NEO (NEO mAb) or against NOR (NOR mAb). Quantitative detection of NEO and QNS was realized via SERS of the PATP-coated AuNPs captured in the test line of a LFA. Under optimized condition, the visual limits of LFA are 10 ng·mL−1 for NEO and 200 ng·mL−1 for NOR, and with LODs down to 0.37 pg·mL−1 and 0.55 pg·mL−1 by using SERS. The NEO test line is not interfered by the NEO analogues gentamycin, streptomycin and tobramycin, but the NOR test line suffers from different degrees of cross-reactivity (CR) to 12 common other QNS, the CRs ranging from 1.5% to 136%. The recoveries of NEO and NOR from spiked milk samples ranged between 86% and 121%, with relative standard deviations (RSD) from 3% to 6%. The method is highly sensitive, accurate and effective. It may be applied to simultaneous detection of NEO and 8 QNS, including NOR, enoxacin, ciprofloxacin, ofloxacin, fleroxacin, marbofloxacin, enrofloxacin, and pefloxacin.Graphical abstractSchematic of a lateral flow assay (LFA) based on an indirect competitive model. Abstract:In this work, a nanogold surface molecularly imprinted polymer spectral probe (AuNP@MIP) for selectively identifying ferrocyanide was prepared under microwave irradiation using nanogold as the core, ferrocyanide as the template ion, methacrylic acid as the monomer, and ethylene glycol dimethacrylate as the cross-linking agent. AuNP@MIP was found to produce a resonance Rayleigh scattering (RRS) peak at 370 nm. When potassium ferrocyanide (K 4 Fe(CN) 6 ) was present, a AuNP@MIP–Fe(CN) 6 complex was formed, producing RRS-energy transfer (RRS-ET). With an increase in ferrocyanide concentration within a certain range, the RRS intensity at 370 nm decreased linearly, and the detection range was 0.02–0.40 μmol L −1 , with a detection limit as low as 0.006 μmol L −1 ferrocyanide. This new method has the advantages of simplicity, rapidity, and selectivity when applied for the determination of K 4 Fe(CN) 6 in table salt.

Abstract:Selective and sensitive determination of ceftriaxone sodium (CTR) trace residues is of great importance for food safety and environmental protection. Herein, a determination method based on ratiometric fluorescence and colorimetric method with nitrogen-rich carbon dots as fluorophore is reported. The functional surfaces of indole-derived carbon dots (I-CDs) containing nitrogen and carbon groups can be selectively bound to CTR by electrostatic forces, leading to a hindered conjugation system and deprotonation of the amine on the pyrrole ring, resulting in a distinct variety in fluorescence and absorption wavelength and intensity. With the addition of CTR, the fluorescence at 577 nm can be selectively quenched, accompanied by a new emission peak appeared at 507 nm. The limits of determination (LODs) were estimated to be 19.7 nM and 78.0 nM based on the ratiometric fluorescence method and colorimetric method, respectively. Finally, the in situ visual quantitative determination of CTR using this nanosensor was achieved by combining with the color recognizer of a smartphone, and the method was further validated by spike and recovery test in real water samples including milk, seawater, and tap water.

Abstract:The authors describe a new composite based on SERS-active copper nanoparticles (CuNPs; 10 ± 2 nm) incorporated into calcium carbonate microspheres (CaCO3-CuNPs; 3.4 ± 0.3 μm). The CaCO3 coating acts as a temporal protector of CuNPs against oxidation. Incorporated CuNPs have significantly improved stability during storage and a month-long shelf lifetime. The composite was used for SERS detection of rhodamine 6G and two antibacterial drugs (ceftriaxone and sulfadimethoxine). Two analytical formats, one with and one without solid phase extraction, are introduced to demonstrate the flexibility of the method. Both formats imply the dissolution of CaCO3 matrix before SERS analysis to release CuNP used as SERS substrate. The study of the influence of pH value and acid nature on the SERS signal demonstrated that HCl is the most efficient candidate to release the CuNPs. Sensitivity (expressed as LOD) is shown to be improved by more than one order when solid phase extraction is used. The average SERS enhancement factor is 10^7 which makes the material efficiency comparable to the one of silver nanoparticles. The LOD (<5 μM), precision (RSDs between 20 and 24% at LOD levels), and trueness (apparent recoveries 84-113%) for the two antibiotics (ceftriaxone and sulfadimethoxine) make the method quite useful for quantitative analysis and therapeutic drug monitoring at physiologically relevant concentrations. Graphical abstract A composite with temporally stable copper nanoparticles was synthesized, studied, and used for SERS detection of two antibacterial drugs. The analytical efficiency of the composite was found appropriate for quantitative analysis due to Raman enhancement comparable with silver nanostructures.

Abstract:Using PEG-10000 and sodium citrate as stabilizer, and NaBH4 as reducer, a stable nanosilver (AgNPs) sol was prepared. In pH 6.6 phosphate buffer solution containing NaCl, the AgNPs were aggregated to large particles, which lead to resonance Rayleigh scattering (RRS) peak at 350 nm enhancement. Upon addition of cysteine, the peak decreased. The decreased value ΔI is linear to cysteine concentration in the range of 5.0×10-8-6.0×10-7 mol/L. Thus, a new RRS method was proposed for detection of cysteine.

Abstract:In this article, a nanosurface molecularly imprinted polymer (MQD@MIP) resonance Rayleigh scattering (RRS) spectral probe for SCN- was prepared by sol-gel method, using Mxene quantum dot as a matrix, thiocyanate (SCN-) as a template ion, (3-aminopropyl) triethoxysilane (APTES) as a functional monomer, tetraethoxysilane (TEOS) as the cross-linker, and ammonia as the initiator. The probe produced an RRS peak at 370 nm and exhibits a strong RRS energy transfer (RRS-ET) effect when the MQD@MIP probe identifies SCN-. As the concentration of SCN- increased, the RRS-ET was enhanced, and the signal value of the system decreased linearly at 370 nm, with a determination range of 0.87-5.22 μg/L, and a detection limit of 0.37 μg/L SCN-. This detection method has the characteristics of simplicity, sensitivity, and specific recognition. The RRS method was used to determine SCN- in the sample, with relative standard deviation of 1.95-10.98% and recovery of 89.0-102.8%.

Abstract:Surface-enhanced Raman scattering (SERS) technique is widely used for the highly sensitive detection of trace residues due to its unparalleled signal amplification ability and plays an important role in food safety, environmental monitoring, etc. Herein, CdSSe nano-flowers (CdSSe NFs) are synthesized via the chemical vapor deposition (CVD) method. CdSSe NFs thin film is used as a SERS substrate with an ultralow limit of detection (LOD, 10−14 M), high apparent enhancement factor (EF, 3.62 × 109), and excellent SERS stability (relative standard deviation, RSD = 3.05%) for probe molecules of Rh6G. Further, CdSSe NFs substrate is successfully applied in the sensitive, quantitative, and label-free analysis of ciprofloxacin (CIP) and enrofloxacin (ENR) antibiotics, which exhibit LODs of below 0.5 ppb. This excellent SERS platform may be widely utilized for sensitive life science and environmental sensing.

Colorimetric Determination of Neomycin Using Melamine Modified Gold Nanoparticles

Abstract:The colorimetric assay for neomycin presented here is based on melamine-modified gold nanoparticles (mel-AuNPs) and the finding that hydrogen bonding between melamine and neomycin results in the aggregation of mel-AuNPs. This results in a change in the color of the solution from wine red to blue and in a red-shift of the absorption maximum of the mel-AuNPs. The concentration of neomycin can be determined by spectrophotometry. The ratio of absorptions at 680 nm and 520 nm is linearly related to the logarithm of the concentration of neomycin in the 0.1 to 5.0 nM range and in the 5 to 100 nM range, with regression coefficients of 0.997 and 0.999, respectively. The detection limit (at an S/N ratio of 3) is 30 pM. This is far below the usual safety limit. The method was applied to the detection of trace levels of neomycin in milk samples and gave recoveries between 98 and 105 %.

Abstract:This article was used nanogold (AuNPs) as the matrix, malachite green (MG) as the template molecule, 3-aminopropyltriethoxysilane (APTES) as the functional monomer, ethyl orthosilicate (TEOS) as the crosslinking agent, and ammonia water (NH 3 ·H 2 O) as the initiator to prepare a gold nanoparticle surface molecular imprinting silica probe (AuNPs@SiO 2 ) for detecting MG using resonance Rayleigh scattering (RRS) method. The AuNPs@SiO 2 generates a resonance Rayleigh scattering peak at 370 nm, which is associated with MG interacting with AuNPs@SiO 2 , it exhibits the resonance Rayleigh scattering energy transfer (RRS-ET) effect. As the concentration of MG increases, RRS-ET enhances, and the signal value of the system linearly decreases at 370 nm, with a detection range of 0.50 ∼ 10.0 nmol/L and a detection limit of 0.15 nmol/L. Based on this, a simple, sensitive and selective RRS-ET method was established for the determination of MG in food samples.

Abstract:Three functional magnetic nanocatalytic probe, which integrates recognition, catalytic amplification, and separation enrichment, is a new approach to construct a simple, fast, highly selective, and sensitive analytical method. In this article, a new magnetic nanosurface molecularly imprinted polymer nanoprobe (Fe3O4@MIP) with trifunctionality was rapidly prepared using a microwave-assisted method with magnetic Fe3O4 nanoparticles as a substrate, chloramphenicol (CAP) as a template molecule, and methacrylic acid as a functional monomer. The characterized nanoprobe was found that could specifically recognize CAP, strongly catalyze the new indicator nanoreaction of fructose (DF)-HAuCl4. The gold nanoparticles (AuNPs) exhibit strong resonance Rayleigh scattering (RRS) and surface enhanced Raman scattering (SERS) effects. Upon addition of CAP, the SERS/RRS signals were linearly weakened. Accordingly, a new SERS/RRS analysis platform for highly sensitive and selective determination of CAP was constructed. The SERS linear range was 0.0125-0.1 nmol/L, with detection limit (DL) of 0.004 nmol/L CAP. Furthermore, it could be combined with magnet-enriched separation to further improve the sensitivity, with a DL of 0.04 pmol/L CAP. The SERS method has been used for the determination of CAP in real samples, with relative standard deviations of 2.37-9.89 % and the recovery of 95.24-107.1 %.
Copper nanoparticles for SERS-based determination of some cephalosporin antibiotics in spiked human urine

Abstract:Copper nanoparticles (CuNPs) were prepared through a wet chemistry method to be used as substituents for noble-metal-based materials in the determination of cephalosporin antibiotics in urine using surface-enhanced Raman spectroscopy (SERS). The synthesis of the CuNPs was optimized to maximize the analytical signal, and microwave heating was used to increase the reaction rate and improve the homogeneity of the CuNPs. Ceftriaxone (CTR), cefazolin (CZL), and cefoperazone (CPR) were used as the analytes of interest. The determination tests were performed on artificially spiked samples of real human urine with concentrations corresponding to therapeutic drug monitoring (TDM) (50-500 μg mL-1). Urine samples collected in the morning and during the day were used to account for deviations in the urine composition, and the universality of the proposed protocol was ensured by performing sample dilution as a pretreatment. The use of calibration plots in the form of Freundlich adsorption isotherms yielded linear calibration plots. All limits of detection were lower than the minimal concentrations required for TDM, equaling 7.5 (CTR), 8.8 (CZL), and 36 (CPR) μg mL-1. Comparison of CuNPs with Ag and Au nanoparticles (AgNPs and AuNPs, respectively) confirmed that CuNPs offered a competitively high Raman enhancement efficiency (for excitation at 638 nm). Further, although the CuNPs demonstrated poorer temporal stability as compared with the AgNPs and AuNPs, the use of freshly prepared CuNPs resulted in satisfactory accuracy (recovery = 93-107%). Given the short analysis time (<20 min, including the time for the synthesis of the CuNPs and the SERS measurements using a portable Raman spectrometer), low sensitivity to the presence of the primary intrinsic urine components and satisfactory figures of merit of the proposed protocol for the determination of cephalosporin antibiotics in urine, it should be suitable for use in TDM.
Bi2MoO6/MWCNTs-COOH/GCE electrochemical probe for ultrasensitively and selectively detecting antibiotic norfloxacin

Abstract:Antibiotics have emerged as a class of contaminants of concern globally, raising widespread worry and unease, primarily because of their inappropriately use and endless migration in food chains. Electrochemical ultrasensitive and selective determination of norfloxacin (NOR) using bismuth-based bimetallic salt bismuth molybdate (Bi2MoO6) and carboxylated multi-walled carbon nanotube (MWCNTs-COOH) hybrid electrode as sensing platforms is presented hereby. The electrocatalytic ability of the electrodes was verified to be enhanced by the synergy effect of both Bi2MoO6 and MWCNTs-COOH using cyclic voltammetry and electrochemical impedance spectroscopy. Linear scan voltammetry was applied to detect NOR. The oxidation peak current was linearly correlated with NOR concentration of 0.03-10 μM and the low limit detection of the optimized method was 6.7 nM (S/N = 3). In addition, it was verified that Bi2MoO6/MWCNTs-COOH/GCE probe had good stability, reproducibility, repeatability and selectivity. The spiked recovery method for NOR in milk and lake water samples showed recoveries of 94.8 - 96.9% and 90.2 - 98.3%, respectively, which can be used to determine NOR in real samples with high sensitivity. The preparation of Bi2MoO6/MWCNTs-COOH/GCE provides a new prototype for probing NOR detection at nanomolar concentration and safeguarding antibiotic contamination in environmental and food chains.
