Abstract:Continuous cropping and soilborne diseases affect soil properties and soil microbial diversity and structure, which are the main factors posing obstacles to the continuous cropping of ginseng. This study explored the response of the physicochemical properties, enzyme activity, and microbial community of ginseng rhizosphere soil to continuous cropping and soilborne disease (root rot of ginseng). We used woodland soil without ginseng planting as a control to study these changes. The results showed that continuous cropping and soilborne disease significantly affected soil physicochemical properties, enzyme activities, and microbial communities. The levels of total nitrogen, hydrolyzable nitrogen, organic matter, and soil pH decreased significantly, while available phosphorus, available potassium, total phosphorus, and total potassium showed significant accumulation after continuous cropping. The activities of urease, catalase, sucrase, acid phosphatase, alkaline phosphatase, and polyphenol oxidase decreased significantly after continuous cropping. Using MiSeq high-throughput sequencing, we found that the alpha diversity and the number of bacterial and fungal communities significantly changed after continuous cropping and soilborne disease. A redundancy analysis suggests that soil physicochemical properties and enzyme activities also affect soil microbial communities. In summary, this study revealed the effects of continuous cropping and soilborne disease on soil and provides a theoretical basis for alleviating soilborne disease in ginseng.

Abstract:Continuous monoculture can increase plant morbidity and reduce crop yields. However, the Ganoderma lucidum mushroom possesses unique nutrient acquisition methods and displays an altered response to the generation of reactive oxygen species (ROS). The effects of continuous cropping on G. lucidum remain unclear. Here, we investigated the effects of consecutive monoculture soil on the yield, bioactive components, antioxidant properties, nutrient metabolism, and enzyme activity of G. lucidum. The results showed no significant differences in terms of yield between the control check and treatment groups after 2 years. However, the content of polysaccharides and triterpenoids after 2 years of treatment and the content of total phenols and flavonoids after 3 years of treatment significantly increased. Moreover, the antioxidant activity (AOA) of the polysaccharides under consecutive monoculture soil treatments also significantly increased. These differences were caused by changes in nutrient metabolism and the activities of antioxidant and ligninolytic enzymes. Under continuous monoculture soil treatment, the content of reducing sugars, water-soluble sugars, free amino acids, and water-soluble proteins significantly increased. The activities of superoxide dismutase, manganese peroxidase, and lignin peroxidase increased with the time of continuous monoculture, while the activities of peroxidase, catalase, and laccase decreased. The increase in ligninolytic enzymes was postulated to mediate the ROS scavenging ability of G. lucidum. In conclusion, continuous cropping soil could increase the levels of bioactive components and AOAs of G. lucidum.

Abstract:Abstract Panax notoginseng is a highly regarded medicinal plant that has been cultivated for more than 400 years in Southwest China. The obstacles associated with the continuous cropping of P. notoginseng are the greatest issues for the development this plant. In the present study, the micro-ecologies of soils differing in the duration of P. notoginseng planting were compared, the results of which could provide important information to aid in solving the problems associated with the continuous cropping of P. notoginseng . Soils in which P. notoginseng had grown for 1, 3 or 5 years, as well as unplanted or fallow soil, which had a P. notoginseng planting interval of 1, 3, 6 or 9 years, were collected in Yunnan Province, China. The numbers and physiological groups of microorganisms, soil enzyme activities and nutrients present in the soil were analyzed to identify the effects of continuous cropping and determine the influence of crop rotation on the soil. After P. notoginseng was planted, the ecological structure of the soil and the balance of soil nutrients changed. These changes in the soil ecosystem prevented the soil from adapting to the continuous cropping of P. notoginseng , which eventually limited the growth of P. notoginseng and increased the incidence of diseases. After rotation of P. notoginseng , some soil indicators were restored, and some indicators with irregular changes may have been caused by crop rotation and field fertilization management practices. Thus, the selection of suitable crop rotations will facilitate the use of continuous cropping for P. notoginseng .

Abstract:Abstract Background Continuous cropping of ginseng ( Panax ginseng Meyer) cultivated in farmland for an extended period gives rise to soil-borne disease. The change in soil microbial composition is a major cause of soil-borne diseases and an obstacle to continuous cropping. The impact of cultivation modes and ages on the diversity and composition of the P. ginseng rhizosphere microbial community and technology suitable for cropping P. ginseng in farmland are still being explored. Methods Amplicon sequencing of bacterial 16S rRNA genes and fungal ITS regions were analyzed for microbial community composition and diversity. Results The obtained sequencing data were reasonable for estimating soil microbial diversity. We observed significant variations in richness, diversity, and relative abundances of microbial taxa between farmland, deforestation field, and different cultivation years. The bacterial communities of LCK (forest soil where P. ginseng was not grown) had a much higher richness and diversity than those in NCK (farmland soil where P. ginseng was not grown). The increase in cultivation years of P. ginseng in farmland and deforestation field significantly changed the diversity of soil microbial communities. In addition, the accumulation of P. ginseng soil-borne pathogens ( Monographella cucumerina , Ilyonectria mors - panacis , I. robusta , Fusarium solani , and Nectria ramulariae ) varied with the cropping age of P. ginseng . Conclusion Soil microbial diversity and function were significantly poorer in farmland than in the deforestation field and were affected by P. ginseng planting years. The abundance of common soil-borne pathogens of P. ginseng increased with the cultivation age and led to an imbalance in the microbial community.

Abstract:American ginseng is an important herbal medicinal crop in China. In recent years, there has been an increasing market demand for ginseng, but the production area has been shrinking due to problems associated with continuous monocropping. We analyzed the microbiome in bulk soils to assess whether and, if so, what changes in the bulk soil microbiome are associated with continuous American ginseng cropping. The alpha diversity of fungi and bacteria was significantly lower in the soils planted with American ginseng than the virgin (non-planted) land. The relative abundance of Fusarium spp. and Ilyonectria spp., known plant root pathogens, was much higher in the soils cropped with American ginseng than the non-planted. On the other hand, a number of bacteria with biodegradation function, such as Methylibium spp., Sphingomonas spp., Variovorax spp., and Rubrivivax spp., had lower abundance in the soils cropped with American ginseng than the non-cropped. In addition, soil pH was lower in the field planted with American ginseng than the non-planted. Accumulation of fungal root pathogens and reduction of soil pH may, therefore, have contributed to the problems associated with continuous monocropping of American ginseng.

Abstract:ABSTRACT The production of ginseng, an important Chinese medicine crop, has been increasingly challenged by soil degradation and pathogenic disease under continuous cropping in Northeast China. In a field experiment, an Alfisol garden continuously cropped with Chinese ginseng (Panax ginseng C. A. Meyer) was treated with soil amendment at 20 t ha−1 with maize (MB) and wood (WB) biochar, respectively, compared to conventional manure compost (MC). Two years after the amendment, the rooted topsoil and ginseng plants were sampled. The changes in soil fertility and health, particularly in the soil microbial community and root disease incidence, and in ginseng growth and quality were portrayed using soil physico-chemical assays, biochemical assays of extracellular enzyme activities and gene sequencing assays as well as ginsenoside assays. Topsoil fertility was improved by 23% and 39%, ginseng root biomass increased by 25% and 27%, and root quality improved by 6% and 18% with WB and MB, respectively, compared to MC. In the ginseng rhizosphere, fungal abundance increased by 96% and 384%, with a significant and insignificant increase in bacterial abundance, respectively, under WB and MB. Specifically, the abundance of Fusarium spp. was significantly reduced by 19–35%, while that of Burkholderia spp. increased by folds under biochar amendments over MC. Relevantly, there was a significant decrease in the abundance proportion of pathotrophic fungi but a great increase in that of arbuscular mycorrhizal fungi, along with an enhanced microbial community network complexity, especially fungal community complexity, under biochar amendments. Thus, biochar, particularly from maize residue, could promote ginseng quality production while enhancing soil health and ecological services, including carbon sequestration, in continuously cropped fields.

Abstract:Abstract This study aims to verify the time-variant feature of American ginseng (AG) continuous cropping obstacles and to explore the factors impeding continuous cropping. We verified the feature with a plant-soil feedback pot experiment and then investigated the factors by comparing the properties of control soils that had not been previously used for growing ginseng (CS) with those of soils with a 10-year-crop-rotation cycle following the growth of AG (RS). It’s found that the survival rate of AG in RS was lower than that in CS. The RS had lower pH, available potassium content, and urease activity. Additionally, p -coumaric, p -hydroxybenzoic, vanillic, caffeic, and cinnamic acid levels were lower in RS than in CS, but salicylic acid levels showed the opposite pattern. RS had higher Rhodanobacter and lower Acidothermus , Sphingomona s relative abundances in bacterial community. It’s also found that many bacteria were substantially correlated with phenolic acids and soil physiochemical properties. Results indicate that even after 10-year crop rotation, the negative effects of prior continuous cropping of AG has not been eliminated. The growth of AG can be affected negatively with deterioration of soil physicochemical properties and with lower levels of phenolic acids which promote pathogen reproduction. Probiotics reduction also weighs. Moreover, biotic factors are interrelated with abiotic ones. Therefore, it can be inferred that the comprehensive change of soil properties is the main obstacle for continuous cropping.

Abstract:ABSTRACT The replant problem is a widespread challenge in agricultural production and results in significant financial losses. The survival and development of American ginseng ( Panax quinquefolius L.), a well-known perennial medicinal plant, also typically suffer from replant problems. At present, efficient strategies for this issue and the underlying mechanisms are still lacking. Here, a series of in situ field experiments over a 4-year period were conducted to investigate the effectiveness and possible mechanisms of soil fumigation and fungicide in improving American ginseng survival under continuous cropping. During the 4-year ginseng cultivation, the seedling emergence rates in the dazomet fumigation-treated group (DF) decreased from 90.67% to 52.92%, from 78.67% to 0% in the lime sulfur fungicide-treated group (LS), and from 66.67% to 0% in the control. The fresh root weight in DF increased from 0.36 to 28.72 g, from 0.32 to 13.74 g in LS, and from 0.42 to 9.06 g in the control for the first 2 years. However, no ginseng survived during the following 2 years. Moreover, DF significantly decreased root rot disease incidence by 55.55% compared to LS-treated soil and 69.39% compared to non-treated soil. The correlation analysis results showed that both diversity and structure indices of bacteria and fungi were significantly correlated with root biomass and disease incidence in the second year. In summary, DF has presented a significant beneficial effect on American ginseng development. Additionally, the second year is the critical period to shape the soil microbial community, which could have a long-lasting effect on the development of American ginseng. IMPORTANCE Numerous reports of soil fumigants and fungicides on annual crops exist; however, it is unclear whether the single application to perennial plants persistently improves plant growth and controls disease or whether it has a long-lasting impact on soil microbes. We found that soil fumigation enhances ginseng growth and suppresses root rot disease by reshaping the soil microbial community. Our findings benefit the agricultural development of ginseng and provide a theoretical basis for the prevention of ginseng diseases.

Abstract:Continuous cropping obstacles severely hindered the sustained development of the ginseng industry. Among the obstacles, an imbalance of soil microbiome community was considered one of the major culprits. The fungal community is an essential part of the soil microbiome community. Extensive characterization of the fungal community composition and variation during ginseng cultivation will help us understand the mechanism underlying continuous cropping obstacles. By using a high-throughput amplicon sequencing method, the non-rhizospheric fungal community of farmland cultivated ginseng of 2 years old (C2) and 5 years old (C5), understory wild ginseng of 15 years old (W15) and 35 years old (W35), fallow fields which have been abandoned for 10 (F10) years were characterized. Farmland cultivated ginseng and understory wild ginseng harbored distinct non-rhizospheric fungal communities, and extension of cultivation periods enlarged the fungal community difference between two cultivation modes. Extended cultivation periods significantly decreased the OTU richness and PD whole tree indices, and OTU number and cultivation periods were negatively correlated. Extension of cultivation periods led to an increased abundance of pathotrophs. Still, the increased abundance of pathotrophs may not be the leading cause of severe continuous cropping obstacles in farmland cultivated ginseng. Compared with understory wild ginseng, farmland cultivated ginseng had a lower abundance of symbiotrophs and a higher abundance of saprotrophs. This changed symbiotrophs/saprotrophs ratio may have some correlation with the severe continuous cropping obstacles that occurred in farmland cultivated ginseng. Fallowing on the fungal community of the non-rhizosphere soil was generally opposite of that of extension of ginseng cultivation periods. The impacts of farmland cultivation on the fungal community of the non-rhizosphere soil can last for decades, even if the following is practiced.

Abstract:Ginsengs, widely acknowledged for their health-promoting properties, are predominantly grown for their roots, necessitating an extended cultivation period of a minimum of 4 to 6 years for maturation. The prolonged growth duration in a specific location makes ginseng plants susceptible to soil-borne ailments, such as root rot, leading to significant detrimental effects. Focusing on the crucial role of the plant microbial community in maintaining ginseng health, the study reveals that repeated and continuous cultivation leads to the collapse of the initial disease-suppressive rhizosphere community, resulting in severe root rot. The dominance of Pseudomonadaceae in the rhizosphere subsequently reinstates disease suppression, aligning with suppressive soil generation phenomena. The research investigates the applicability of identified patterns to field conditions and demonstrates that rhizosphere samples from the field closely resemble conditions observed in pot-based NH4Cl treatment experiments. These findings emphasize the critical role of the rhizosphere microbial community in ginseng health maintenance during extended cultivation, offering insights into disease prevention strategies. The study also suggests the potential of pot-based experiments in simulating field conditions and informs future approaches for sustainable ginseng cultivation.

The biological characteristics of pathogens, its occurrence and environmental factors affecting the occurrence of diseases, as well as chemical and biological control measures were described. Based on the current problems existing in the Panax notoginseng production, some suggestions to enhance the researches on studying mechanism of disease resistance in Panax notoginseng, breeding and extension of new resistant varieties, work out agricultural, chemical and biological measures for the controlling of Panax notoginseng combining traditional agriculture and biological control were put forward. The present paper illustrates the development of Panax notoginseng with the balance of economic, social and environmental benefits.
Exploring the allelopathic autotoxicity mechanism of ginsenosides accumulation under ginseng decomposition based on integrated analysis of transcriptomics and metabolomics

Abstract:Continuous cropping obstacles seriously constrained the sustainable development of the ginseng industry. The allelopathic autotoxicity of ginsenosides is the key "trigger" of continuous cropping obstacles in ginseng. During harvest, the ginseng plants could be broken and remain in the soil. The decomposition of ginseng residue in soil is one of the important release ways of ginsenosides. Therefore, the allelopathic mechanism of ginsenosides through the decomposed release pathway needs an in-depth study. To investigate this allelopathic regulation mechanism, the integrated analysis of transcriptomics and metabolomics was applied. The prototype ginsenosides in ginseng were detected converse to rare ginsenosides during decomposition. The rare ginsenosides caused more serious damage to ginseng hairy root cells and inhibited the growth of ginseng hairy roots more significantly. By high-throughput RNA sequencing gene transcriptomics study, the significantly differential expressed genes (DEGs) were obtained under prototype and rare ginsenoside interventions. These DEGs were mainly enriched in the biosynthesis of secondary metabolites and metabolic pathways, phytohormone signal transduction, and protein processing in endoplasmic reticulum pathways. Based on the functional enrichment of DEGs, the targeted metabolomics analysis based on UPLC-MS/MS determination was applied to screen endogenous differential metabolized phytohormones (DMPs). The influence of prototype and rare ginsenosides on the accumulation of endogenous phytohormones was studied. These were mainly involved in the biosynthesis of diterpenoid, zeatin, and secondary metabolites, phytohormone signal transduction, and metabolic pathways. After integrating the transcriptomics and metabolomics analysis, ginsenosides could regulate the genes in phytohormone signaling pathways to influence the accumulation of JA, ABA, and SA. The conclusion was that the prototype ginsenosides were converted into rare ginsenosides by ginseng decomposition and released into the soil, which aggravated its allelopathic autotoxicity. The allelopathic mechanism was to intervene in the response regulation of genes related to the metabolic accumulation of endogenous phytohormones in ginseng. This result provides a reference for the in-depth study of continuous cropping obstacles of ginseng.

Abstract:Abstract Background Ginseng red skin root syndrome (GRS) is one of the most common ginseng ( Panax ginseng Meyer) diseases. It leads to a severe decline in P. ginseng quality and seriously affects the P. ginseng industry in China. However, as a root disease, the characteristics of the GRS rhizosphere microbiome are still unclear. Methods The amplicon bacterial 16 S rRNA genes and fungal ITS (Internal Transcribed Spacer) regions Illumina sequencing technology, combined with microbial diversity and composition analysis based on R software, was used to explore the relationship between soil ecological environment and GRS. Results There were significant differences in the diversity and richness of soil microorganisms between the rhizosphere with different degrees of disease, especially between healthy P. ginseng (HG) and heavily diseased groups. The variation characteristics of microbial abundance in different taxa levels were analyzed. The interaction network of rhizosphere microorganisms of P. ginseng under GRS background was established. We also found that different P. ginseng rhizosphere microbial communities have multiple changes in stability and complexity through the established interaction network. Microbes closely related to potential pathogenic fungi were also identified according to the interaction network, which provided clues for looking for biological control agents. Finally, the Distance-based redundancy analysis (dbRDA) results indicated that total phosphorus (TP), available potassium (AK), available phosphorus (AP), catalase (CAT), invertase (INV) are the key factors that influence the microbial communities. Moreover, the content of these key factors in the rhizosphere was negatively correlated with disease degrees. Conclusions In this study, we comprehensively analyzed the rhizosphere characteristics of P. ginseng with different levels of disease, and explored the interaction relationship among microorganisms. These results provide a basis for soil improvement and biological control of field-grown in the future.

Abstract:The changes of microbial communities of rhizospheric soil in different ages are speculated to cause soil-borne diseases and replanting problem in American ginseng (Panax quinquefolius L.) cultivation. This study analyzed the physicochemical properties and microbial communities of rhizospheric soil during the planting of American ginseng in the Wendeng area of Weihai, China. The water content and organic matter content of American ginseng rhizospheric soil decreased year by year. A decline in the diversity of bacteria and fungi was observed in the rhizospheric soils planting American ginseng compared with the traditional crop wheat in the control group. During the later planting stage, the abundances of Proteobacteria, Actinobacteria, and Basidiomycota were lower, whereas that of Acidobacteria, Firmicutes, and Mucoromycota were higher. Through the correlation analysis between environmental factors and microbial community, it was found that the content of soil phosphorus was significantly positively correlated with the root rot pathogen Fusarium. The results of functional prediction showed that the decrease of secondary metabolite synthesis of rhizospheric soil bacteria and the increase of plant pathogenic fungi may be the important reasons for the increase of diseases in the later stage of American ginseng planting. This study revealed the evolution of rhizosphere microbial community and function in the process of American ginseng planting, which is valuable for planting management.

Abstract:Changes in the microbial community are considered important factors affecting the yield and quality of ginseng cultivated in farmland, while the characteristics of the change in the microbial community of the soil before and after cultivation of ginseng in farmland are not well understood. Here, we aimed to fill this research gap by comparing the differences in soil nutrients and microbial communities between continuously cultivated ginseng soils for 1-, 2-, 3- and 5-year-old ginseng soils and corresponding uncultivated ginseng soils. The soil properties (pH, total soil organic carbon (TOC) content and total nitrogen (TN) content) of aged ginseng soil (3- and 5-year-old) were significantly reduced compared with those of the control soil after ginseng was cultivated in farmland. With the exception of 2-year-old ginseng, soil bacterial richness and diversity were higher in different ages of cultivated ginseng soils than in the control soil, while fungal diversity decreased in aged ginseng soil and increased in young ginseng soil (1- and 2-year-old). Ascomycota and Basidiomycota showed a consistent increasing/decreasing trend in cultivated ginseng soils of all ages compared to the control. In addition, ginseng cultivation resulted in a substantial decrease in the complexity of soil bacterial networks but a slight decrease in the complexity of fungal networks. The results from the linear discriminant size effect (LEfSe) analysis showed that beneficial microorganisms were biomarkers of the young ginseng soil, including Nitrosospira , Cephalotrichum and Humicola , while harmful microorganisms were biomarkers of aged ginseng, such as Plectosphaerella , Cladosporium and Alternaria . PICRUSt2 analysis revealed that the increase in the incidence of ginseng disease with continuous cultivation time may be related to the decreases in amino acid synthesis and physiological metabolism and the increase in fungal saprophytes. Soil pH was the most common and best predictor of bacterial and fungal community structure, followed by TOC and ammonium nitrogen (NH 4 + -N) contents. Meanwhile, the decreased of soil pH, TOC and AK may be the driving factors for the growth of ginseng soil pathogens. Overall, this study revealed the changing ginseng rhizosphere soil microbial community at different continuous cultivation times, provided a theoretical basis for ginseng cultivation, and promoted the development of medicinal plants such as ginseng.

Abstract:Abstract Soil physicochemical properties and fungal communities are pivotal factors for continuous cropping of American ginseng ( Panax quinquefolium L.). However, the response of soil physicochemical properties and fungal communities to replant disease of American ginseng has not yet been studied. High-throughput sequencing and soil physicochemical analyses were undertaken to investigate the difference of soil fungal communities and environmental driver factors in new and old ginseng fields; the extent of replant disease in old ginseng fields closely related to changes in soil properties and fungal communities was also determined. Results indicated that fungal communities in an old ginseng field were more sensitive to the soil environment than those in a new ginseng field, and fungal communities were mainly driven by soil organic matter (SOM), soil available phosphorus (AP), and available potassium (AK). Notably, healthy ginseng plants in new and old ginseng fields may influence fungal communities by actively recruiting potential disease suppressive fungal agents such as Amphinema , Cladophialophora , Cadophora , Mortierella, and Wilcoxina . When these key groups and members were depleted, suppressive agents in the soil possibly declined, increasing the abundance of pathogens. Soil used to grow American ginseng in the old ginseng field contained a variety of fungal pathogens, including Alternaria , Armillaria , Aphanoascus , Aspergillus , Setophoma, and Rhexocercosporidium . Additionally, micro-ecological factors affecting disease outbreaks in the old ginseng field included a strengthening in competition relationships, a weakening in cooperation relationships, and a change of trophic strategies among fungal communities.

Abstract:Root rot is a serious soil-borne fungal disease that seriously affects the yield and quality of Panxa ginseng . To develop a sustainable strategy for alleviating ginseng root rot, an herb-based soil amendment is suggested in this study. Mixed powers of medicinal herbs (MP) and corn stalks (CS) were used as soil amendments, respectively, along with a control group (CK) without treatment. The application of MP and CS led to significant relief from ginseng root rot. The disease index (%) represents both the incidence rate and symptom severity of the disease. The disease index of the MP and CS group was 18.52% and 25.93%, respectively, lower than that of CK (40.74%). Correspondingly, three soil enzyme activities improved; the antifungal components in the soil increased; and the relative abundances of root rot pathogens decreased in response to MP Soil enzyme activities were negatively correlated with disease grades. MP group also led to possible interactive changes in the communities of soil fungi and chemical components. In conclusion, our results suggest that the use of herb-based soil amendments has significant potential as an ecological and effective approach to controlling root rot disease of ginseng by the changing rhizosphere fungal community and soil compositions.
