Abstract:In this work, for the first time, we constructed a novel multi-nanozymes cooperative platform to mimic intracellular antioxidant enzyme-based defense system. V2 O5 nanowire served as a glutathione peroxidase (GPx) mimic while MnO2 nanoparticle was used to mimic superoxide dismutase (SOD) and catalase (CAT). Dopamine was used as a linker to achieve the assembling of the nanomaterials. The obtained V2 O5 @pDA@MnO2 nanocomposite could serve as one multi-nanozyme model to mimic intracellular antioxidant enzyme-based defense procedure in which, for example SOD, CAT, and GPx co-participate. In addition, through assembling with dopamine, the hybrid nanocomposites provided synergistic antioxidative effect. Importantly, both in vitro and in vivo experiments demonstrated that our biocompatible system exhibited excellent intracellular reactive oxygen species (ROS) removal ability to protect cell components against oxidative stress, showing its potential application in inflammation therapy.
Facet‐Dependent Biodegradable Mn 3 O 4 Nanoparticles for Ameliorating Parkinson's Disease

Abstract:Parkinson's disease (PD) is a common neurodegeneration disease. Unfortunately, there are no effective measures to prevent or inhibit this disease. In this study, we prepared biodegradable Mn3O4 nanoparticles (NPs) in different shapes and enclosed them by {100}, {200} and {103} facets that exhibited facet-dependent protection against neurotoxicity induced by oxidative damage in a cell model of PD. Notably, Mn3O4 nanorods enclosed by {103} facets exhibited high levels of enzyme-like activity to eliminate reactive oxygen species (ROS) in vitro. We also determined that the uptake pathway of Mn3O4 NPs into MN9D cells was mediated by caveolin. Our data demonstrated that Mn3O4 nanorods could be taken up by cells effectively and conferred excellent levels of neuroprotection while the biodegradation of Mn3O4 NPs in vivo was confirmed by photoacoustic (PA) image of Mn3O4 NPs in brain at 60 days. Furthermore, the oxygen scavenging effect created by Mn3O4 nanorods was successfully applied to a mouse model of PD; the amount of α-synuclein (α-syn) in the cerebrospinal fluid of PD mice was reduced by 61.2% in two weeks, thus demonstrating the potential application of facet-directed Mn3O4 NPs for the clinical therapy of neurodegenerative disease.This article is protected by copyright. Abstract:Reactive oxygen species (ROS) refer to various partially reduced oxygen moieties that are naturally generated due to biochemical processes. Elevated formation of ROS leads to damage to biomolecules, resulting in oxidative stress and cell death. The increased level of ROS also affects therapeutics based on stem cell transplantation. Nanomaterials-based enzyme mimetics have attracted immense attention, but there are several challenges to be addressed in terms of selectivity, efficiency, and biocompatibility. This highlight focuses on a recent investigation by Cheng and coworkers, who engineered an Mn-superoxide dismutase (Mn-SOD)-inspired material with Mn-N5 sites having an axial ligand and 2D d-π-conjugated network. This engineering approach enhances antioxidase-like function and effectively rescues stem cells from ROS. In addition, it also protects osteogenesis-related gene transcription, ensuring survival rates and osteogenic differentiation of hMSCs under ROS environment. This versatile and robust artificial antioxidase holds promise for stem cell therapies and ROS-originated diseases.

Abstract:By mimicking the catalytic domains of natural antioxidant enzymes, a de novo metal‐organic framework modified with polydopamine is fabricated. It not only possesses superoxide dismutase‐like nanozyme activities but also biologically upregulates two endogenous antioxidant enzymes (HO1 and SOD2) through STAT3 signaling, effectively scavenging excessive reactive oxygen species and thereby powerfully salvaging neurons from ischemic insult poststroke. Following stroke, oxidative stress induced by reactive oxygen species (ROS) aggravates neuronal damage and enlarges ischemic penumbra, which is devastating to stroke patients. Nanozyme‐based antioxidants are emerging to treat stroke through scavenging excessive ROS. However, most of nanozymes cannot efficiently scavenge ROS in neuronal cytosol and mitochondria, due to low‐uptake abilities of neurons and barriers of organelle membranes, significantly limiting nanozymes' neuroprotective effects. To overcome this limitation, a manganese‐organic framework modified with polydopamine (pDA‐MNOF), capable of not only mimicking catalytic activities of natural SOD2's catalytic domain but also upregulating two endogenous antioxidant enzymes in neurons is fabricated. With such a dual anti‐ROS effect, this nanozyme robustly decreases cellular ROS and effectively protects them from ROS‐induced injury. STAT‐3 signaling is found to play a vital role in pDA‐MNOF activating the two antioxidant enzymes, HO1 and SOD2. In vivo pDA‐MNOF treatment significantly improves the survival of middle cerebral artery occlusion (MCAo) mice by reducing infarct volume and more importantly, promotes animal behavioral recovery. Further, pDA‐MNOF activates vascular endothelial growth factor expression, a downstream target of STAT3 signaling, thus enhancing angiogenesis. Taken together, the biochemical, cell‐biological, and animal‐level behavioral data demonstrate the potentiality of pDA‐MNOF as a dual ROS‐scavenging agent for stroke treatment.

Abstract:Nanozymes, nanomaterials with enzyme-mimicking activity, have attracted tremendous interest in recent years owing to their ability to replace natural enzymes in various biomedical applications. Nanozymes that can regulate the cellular redox status by mimicking the antioxidant enzymes in mammalian cells are of great therapeutic significance in oxidative stress-mediated disorders. As the distinction of physiological oxidative stress (oxidative eustress) and pathological oxidative stress (oxidative distress) occurs at a fine borderline, it is a great challenge to design nanozymes that can differentially sense the two extremes in cells, tissues and organs and mediate appropriate redox chemical reactions. In this review, we summarize the advances in the development of redox-active nanozymes and their biomedical applications. We highlight the therapeutic significance of the antioxidant and prooxidant nanozymes in various diseases such as cancer, neurodegeneration, and cardiovascular diseases.

Abstract:Oxidative stress is associated with many acute and chronic inflammatory diseases. Development of nanomaterial-based enzyme mimetics for reactive oxygen species (ROS) scavenging is challenging, but holds great promise for the treatment of inflammatory diseases. Herein, we report the highly ordered manganese dioxide encapsulated selenium-melanin (Se@Me@MnO2) nanozyme with high efficiency for intracellular antioxidation and anti-inflammation. The Se@Me@MnO2 nanozyme is sequentially fabricated through the radical polymerization and the in-situ oxidation-reduction. In vitro experimental results demonstrated that the Se@Me@MnO2 nanozyme exhibits multiple enzyme activities to scavenge ROS, including catalase (CAT), glutathione peroxidase (GPx) and superoxide dismutase (SOD). Mechanism researches illustrated that the Se core possesses GPx-like catalytic activity, the Me and the MnO2 possess both the SOD-like and the CAT-like activities. What’s more, due to the stable unpaired electrons existing in the nanozyme, the Se, Me and MnO2 provide synergistic and fast electron transfer effect to achieve the quickly scavenging of hydrogen peroxide, hydroxyl radical, and superoxide anion. Further in vivo experimental results showed that this biocompatible nanozyme exhibits cytoprotective effects by resisting ROS-mediated damage, thereby alleviating the inflammation. This multienzyme mimetics is believed to be an excellent ROS scavenger and have a good potential in clinical therapy for ROS-related diseases.

Abstract:Designing translational antioxidative agents that could scavenge free radicals produced during reperfusion in brain ischemia stroke and alleviate neurologic damage is the main objective for ischemic stroke treatment. Herein, we explored and simply synthesized a biomimic and translational Mn3O4 nanoenzyme (HSA-Mn3O4) to constrain ischemic stroke reperfusion-induced nervous system injury. This nanosystem exhibits reduced levels of inflammation and prolonged circulation time and potent ROS scavenging activities. As expected, HSA-Mn3O4 effectively inhibits oxygen and glucose deprivation-mediated cell apoptosis and endoplasmic reticulum stress and demonstrates neuroprotective capacity against ischemic stroke and reperfusion injury of brain tissue. Furthermore, HSA-Mn3O4 effectively releases Mn ions and promotes the increase of superoxide dismutase 2 activity. Therefore, HSA-Mn3O4 inhibits brain tissue damage by restraining cell apoptosis and endoplasmic reticulum stress in vivo. Taken together, this study not only sheds light on design of biomimic and translational nanomedicine but also reveals the neuroprotective action mechanisms against ischemic stroke and reperfusion injury.

Abstract:Disordered reactive oxygen/nitrogen species are a common occurrence in various diseases, which usually cause cellular oxidative damage and inflammation. Despite the wide range of applications for biomimetic nanoparticles with antioxidant or anti-inflammatory properties, designs that seamlessly integrate these two abilities with a synergistic effect in a simple manner are seldom reported. In this study, we developed a novel PEI-Mn composite nanoparticle (PM NP) using a chelation method, and the curcumin was loaded onto PM NPs via metal–phenol coordination to form PEI-Mn@curcumin nanoparticles (PMC NPs). PMC NPs possessed excellent dispersibility and cytocompatibility, was engineered to serve as an effective nanozyme, and exhibited specific SOD-like and CAT-like activities. In addition, the incorporation of curcumin granted PMC NPs the ability to effectively suppress the expression of inflammatory cytokines in microglia induced by LPS. As curcumin also has antioxidant properties, it further amplified the synergistic efficiency of ROS scavenging. Significantly, PMC NPs effectively scavenged ROS triggered by H2O2 in SIM-A9 microglia cells and Neuro-2a cells. PMC NPs also considerably mitigated DNA and lipid oxidation in Neuro-2a cells and demonstrated an increase in cell viability under various H2O2 concentrations. These properties suggest that PMC NPs have significant potential in addressing excessive ROS and inflammation related to neural diseases.

Abstract:The endothelium‐derived signaling molecule nitric oxide (NO) in addition to controlling multifarious servo‐regulatory functions in the vasculature, suppresses key processes in vascular lesion formation and prevents atherogenesis and other vascular abnormalities. The conversion of NO into cytotoxic and powerful oxidant peroxynitrite (ONOO−) in a superoxide‐rich environment has emerged as a major reason for the reduced NO levels in the vascular wall, leading to endothelial dysfunction, atherosclerosis, and other cardiovascular complications. However, it is a challenge to design SOD mimetics that can selectively catalyze the dismutation of superoxide in the presence of NO, considering their rapid reaction. Herein, we report that the SOD mimetic CeVO4 nanozymes effectively regulate the bioavailability of both NO and superoxide, the two key constitutive molecules of vascular endothelium, even in the absence of the cellular SOD enzyme. The nanozymes optimally modulate the amount of superoxide in the cells under oxidative stress conditions and improve the endogenously generated NO levels by preventing the formation of peroxynitrite. Also, the nanoparticles internalize into the cells via clathrin‐mediated endocytosis and the cellular uptake depends both on the size‐ and morphology of the nanozymes. When administered intravenously to mice, the CeVO4 nanozymes caused no definite organ toxicity and the haematological and biochemical parameters remained unchanged, indicating their biosafety and potential use in biological applications. Abstract:Despite decades of research, Parkinson's disease is still an idiopathic pathology for which no cure has yet been found. This is partly explained by the multifactorial character of most neurodegenerative syndromes, whose generation involves multiple pathogenic factors. In Parkinson's disease, two of the most important ones are the aggregation of α-synuclein and oxidative stress. In this work, we address both issues by synthesizing a multifunctional nanozyme based on grafting a pyridinophane ligand that can strongly coordinate CuII, onto biodegradable PEGylated polyester nanoparticles. The resulting nanozyme exhibits remarkable superoxide dismutase activity together with the ability to inhibit the self-induced aggregation of α-synuclein into amyloid-type fibrils. Furthermore, the combination of the chelator and the polymer produces a cooperative effect whereby the resulting nanozyme can also halve CuII-induced α-synuclein aggregation.

These nanomaterials can regulate the redox levels of cells by mimicking the catalytic processes of natural enzymes such as peroxidase,I catalase, and superoxide dismutase. This paper describes the intracellular ROS regulation 1 and oxidative stress-related diseases treatment effects of metal compounds nanozymes, noble metal nanozymes amid carbon-based nanozymes from the perspective of their enzyme-like activities. As a new type of enzyme mimic, nanozymes are expected to provide a new strategy for the treatment of diseases in the future biomedical field.

Abstract:A novel dual synergetic nanoreactor is designed for managing Parkinson's disease by regulating inflammation and mitigating oxidative damage by encapsulating dihydroquercetin and Pt nanozymes in mannitol‐targeted PLGA nanoparticles. Glial cell‐dominated inflammatory microenvironment and neuronal damage due to oxidative stress are major impediments to the treatment of central nervous degenerative diseases. Herein, a neuroinflammatory regulatory nanoreactor is developed by encapsulating dihydroquercetin (Que) and Pt nanozymes (Ptzymes) in mannitol modified poly(lactic‐co‐glycolic acid) nanoparticles. The nanoreactor shows enhanced antioxidative activities compared with either Que or Ptzymes alone due to synergetic effects of the incorporated active agents. In cellular and animal models of Parkinson's disease (PD), the nanoreactor traverses the blood–brain barrier and accumulates in neurons and microglia, thereby mitigating oxidative damage of neurons, and promoting the polarization of microglia into the anti‐inflammatory M2‐phenotype to control PD progression. This work demonstrates the gains of combining anti‐inflammation small molecule compound and antioxidant nanozyme in a synergetic nanoreactor to effectively prevent neuronal damage and suppress the inflammatory microenvironment, which holds great potential for managing the advances of PD.

Methionine sulfoxide reductase A protects dopaminergic cells from Parkinson's disease-related insults

Abstract:Parkinson's disease (PD) is a neurologic disorder characterized by dopaminergic cell death in the substantia nigra. PD pathogenesis involves mitochondrial dysfunction, proteasome impairment, and alpha-synuclein aggregation, insults that may be especially toxic to oxidatively stressed cells including dopaminergic neurons. The enzyme methionine sulfoxide reductase A (MsrA) plays a critical role in the antioxidant response by repairing methionine-oxidized proteins and by participating in cycles of methionine oxidation and reduction that have the net effect of consuming reactive oxygen species. Here, we show that MsrA suppresses dopaminergic cell death and protein aggregation induced by the complex I inhibitor rotenone or mutant alpha-synuclein, but not by the proteasome inhibitor MG132. By comparing the effects of MsrA and the small-molecule antioxidants N-acetylcysteine and vitamin E, we provide evidence that MsrA protects against PD-related stresses primarily via methionine sulfoxide repair rather than by scavenging reactive oxygen species. We also demonstrate that MsrA efficiently reduces oxidized methionine residues in recombinant alpha-synuclein. These findings suggest that enhancing MsrA function may be a reasonable therapeutic strategy in PD.

Abstract:Reactive oxygen species (ROS)-induced oxidative stress is linked to various diseases, including cardiovascular disease and cancer. Though highly efficient natural ROS scavenging enzymes have been evolved, they are sensitive to environmental conditions and hard to mass-produce. Therefore, enormous efforts have been devoted to developing artificial enzymes with ROS scavenging activities. Among them, ROS scavenging nanozymes have recently attracted great interest owing to their enhanced stability, multi-functionality, and tunable activity. It has been implicated that Mn-contained nanozymes would possess efficient ROS scavenging activities, however only a few such nanozymes have been reported. To fill this gap, herein we demonstrated that Mn3O4 nanoparticles (NPs) possessed multiple enzyme mimicking activities (i.e., superoxide dismutase and catalase mimicking activities as well as hydroxyl radical scavenging activity). The Mn3O4 nanozymes therefore significantly scavenged superoxide radical as well as hydrogen peroxide and hydroxyl radical. Moreover, they were not only more stable than the corresponding natural enzymes but also superior to CeO2 nanozymes in terms of ROS elimination. We showed that the Mn3O4 NPs not only exhibited excellent ROS removal efficacy in vitro but also effectively protected live mice from ROS-induced ear-inflammation in vivo. These results indicated that Mn3O4 nanozymes are promising therapeutic nanomedicine for treating ROS-related diseases.

Abstract:Proteolytic degradation of amyloid-β (Aβ) aggregates and clearance of Aβ-induced reactive oxygen species (ROS) have received significant attention for the treatment of Alzheimer’s disease (AD). However, it is difficult, and often unfeasible, to directly upregulate or transport intracellular native enzymes. More importantly, penetration of the blood-brain barrier (BBB) has presented a major impediment. Herein, we report on the rational design of a polyoxometalatebased nanozyme with both protease-like activity for depleting Aβ aggregates, and superoxide dismutase (SOD)-like activity for scavenging Aβ-mediated ROS. Furthermore, this nanozyme acts as a metal chelator to remove Cu from Cu-induced Aβ oligomers. More intriguingly, the nanozyme can cross the BBB and exhibits low toxicity. This work provides new insights into the design and synthesis of inorganic nanozymes as multifunctional therapeutic agents in the treatment of AD.
