Abstract:[Aim] This study aims to investigate the gene controlling the eye color of the white-backed planthopper (WBPH),Sogatella furcifera and to explore the potential influence of eye color mutants on mating capacity.[Methods] An orange-eye mutant was established in our insectary,and the gene symbol related to this mutant was designated as org.The genetic basis of this phenotype was determined by designing eight groups of cross-mating experiment containing three genotypes (+/+,+/org,org/org).Whether the eye color related genes influence the mating capacity in WBPH was tested in a mating competition experiment.[Results]This orange-eye mutant had a pair of bright orange ommatea which was distinctly different from the wild type WBPH.The orange-eye phenotype occurred in all developmental stages of WBPH:the eyespot of eggs (3 d after ovipisition) and the compound eyes of nymphs and adults of both sexes and wing morphs.Reciprocal crosses between homozygous normal-eye WBPH and orange-eye WBPH produced F1 progeny with only normaleye color.The sex ratio in the offspring produced from all groups was not biased to a specific sex based on Chisquare test (α =0.05,x2c =0.19-1.53).The mating competitiveness of the orange-eye mutant was significantly weaker than that of the wild type phenotype in WBPH.[Conclusion] Our results indicate that the inheritance of the orange-eye trait is controlled by an autosomal recessive allele,and eye color related gene has significant impacts on mating capacity in WBPH.
Long-wave opsin involved in body color plastic development in the brown planthopper

Abstract:Abstract Background Opsin is an important molecular basis of the visual system, besides, it was discovered that opsins have also evolved additional functions despite color vision. However, the study of the unconventional function has been limited to a few species. With the increasing number of genome databases, various numbers and types of opsins have been identified in insects due to gene duplications or losses. The brown planthopper ( Nilaparvata lugens , BPH) is a Hemipteran rice pest known for its long-distance migration capability. Opsins in BPH were identified and characterized by analyzing the genome and transcriptome. RNA interference (RNAi) was used to investigate the functions of opsins, and the potential molecular mechanism was studied by the Illumina Novaseq 6000 platform-based transcriptome sequencing. Results Four opsins belonging to G protein-coupled receptors were identified in BPH genome, including one long-sensitive opsin ( Nllw ) and three ultraviolet-sensitive opsins ( NlUV1-3 ). A tandem array of NlUV1/2 on chromosome suggested a gene duplication event, with similar exons distribution. Spatiotemporal expression modes showed that the four opsins were highly expressed in eye with age-different expression levels, indicating functional differentiation. RNAi targeting each of the four opsins did not significantly affect the survival of BPH in phytotron, but silencing of Nllw resulted in melanism of the body color. Further transcriptome analysis revealed up-regulation of a tyrosine hydroxylase gene ( NlTH ) and down-regulation of an arylalkylamine-N-acetyltransferases gene ( NlaaNAT ) in BPH, indicating that Nllw is involved in body color plastic development through the tyrosine-mediated melanism pathway. Conclusions This study provides the first evidence in a Hemipteran insect that an opsin ( Nllw ) takes part in the regulation of cuticle melanization, confirming the visual system cross-talks with the morphological differentiation in insects.

Abstract:Abstract Visual systems adapt to different light environments through several avenues including optical changes to the eye and neurological changes in how light signals are processed and interpreted. Spectral sensitivity can evolve via changes to visual pigments housed in the retinal photoreceptors through gene duplication and loss, differential and coexpression, and sequence evolution. Frogs provide an excellent, yet understudied, system for visual evolution research due to their diversity of ecologies (including biphasic aquatic-terrestrial life cycles) that we hypothesize imposed different selective pressures leading to adaptive evolution of the visual system, notably the opsins that encode the protein component of the visual pigments responsible for the first step in visual perception. Here, we analyze the diversity and evolution of visual opsin genes from 93 new eye transcriptomes plus published data for a combined dataset spanning 122 frog species and 34 families. We find that most species express the four visual opsins previously identified in frogs but show evidence for gene loss in two lineages. Further, we present evidence of positive selection in three opsins and shifts in selective pressures associated with differences in habitat and life history, but not activity pattern. We identify substantial novel variation in the visual opsins and, using microspectrophotometry, find highly variable spectral sensitivities, expanding known ranges for all frog visual pigments. Mutations at spectral-tuning sites only partially account for this variation, suggesting that frogs have used tuning pathways that are unique among vertebrates. These results support the hypothesis of adaptive evolution in photoreceptor physiology across the frog tree of life in response to varying environmental and ecological factors and further our growing understanding of vertebrate visual evolution.

A Thermal Receptor for Nonvisual Sunlight Detection in Myriapods.

Abstract:Organisms from cyanobacteria to humans have evolved a wide array of photoreceptive strategies to detect light. Sunlight avoidance behavior is common in animals without vision or known photosensory genes. While indirect light perception via photothermal conversion is a possible scenario, there is no experimental evidence for this hypothesis. Here, we show a nonvisual and extraocular sunlight detection mechanism by identifying the broad-range thermal receptor 1 (BRTNaC1, temperature range = 33 to 48 °C) in centipede antennae. BRTNaC1, a heat-activated cation-permeable ion channel, is structurally related to members of the epithelial sodium channel family. At the molecular level, heat activation of BRTNaC1 exhibits strong pH dependence controlled by two protonatable sites. Physiologically, temperature-dependent activation of BRTNaC1 upon sunlight exposure comes from a striking photothermal effect on the antennae, where a slightly acidic environment (pH 6.1) of the body fluid leads to the protonation of BRTNaC1 and switches on its high thermal sensitivity. Furthermore, testosterone potently inhibits heat activation of BRTNaC1 and the sunlight avoidance behavior of centipedes. Taken together, our study suggests a sophisticated strategy for nonvisual sunlight detection in myriapods.

Abstract:In this study, the morphology and ultrastructure of the compound eye of Asi. xanthospilota were examined by using scanning electron microscopy (SEM), transmission electron microscopy (TEM), micro-computed tomography (μCT), and 3D reconstruction. Spectral sensitivity was investigated by electroretinogram (ERG) tests and phototropism experiments. The compound eye of Asi. xanthospilota is of the apposition type, consisting of 611.00 ± 17.53 ommatidia in males and 634.8 0 ± 24.73 ommatidia in females. Each ommatidium is composed of a subplano-convex cornea, an acone consisting of four cone cells, eight retinular cells along with the rhabdom, two primary pigment cells, and about 23 secondary pigment cells. The open type of rhabdom in Asi. xanthospilota consists of six peripheral rhabdomeres contributed by the six peripheral retinular cells (R1~R6) and two distally attached rhabdomeric segments generated solely by R7, while R8 do not contribute to the rhabdom. The orientation of microvilli indicates that Asi. xanthospilota is unlikely to be a polarization-sensitive species. ERG testing showed that both males and females reacted to stimuli from red, yellow, green, blue, and ultraviolet light. Both males and females exhibited strong responses to blue and green light but weak responses to red light. The phototropism experiments showed that both males and females exhibited positive phototaxis to all five lights, with blue light significantly stronger than the others.
Color-Pattern Analysis of Eyespots in Butterfly Wings: A Critical Examination of Morphogen Gradient Models

Abstract:Butterfly wing color patterns consist of many color-pattern elements such as eyespots. It is believed that eyespot patterns are determined by a concentration gradient of a single morphogen species released by diffusion from the prospective eyespot focus in conjunction with multiple thresholds in signal-receiving cells. As alternatives to this single-morphogen model, more flexible multiple-morphogen model and induction model can be proposed. However, the relevance of these conceptual models to actual eyespots has not been examined systematically. Here, representative eyespots from nymphalid butterflies were analyzed morphologically to determine if they are consistent with these models. Measurement of ring widths of serial eyespots from a single wing surface showed that the proportion of each ring in an eyespot is quite different among homologous rings of serial eyespots of different sizes. In asymmetric eyespots, each ring is distorted to varying degrees. In extreme cases, only a portion of rings is expressed remotely from the focus. Similarly, there are many eyespots where only certain rings are deleted, added, or expanded. In an unusual case, the central area of an eyespot is composed of multiple "miniature eyespots," but the overall macroscopic eyespot structure is maintained. These results indicate that each eyespot ring has independence and flexibility to a certain degree, which is less consistent with the single-morphogen model. Considering a "periodic eyespot", which has repeats of a set of rings, damage-induced eyespots in mutants, and a scale-size distribution pattern in an eyespot, the induction model is the least incompatible with the actual eyespot diversity.

Opsin gene expression plasticity and spectral sensitivity in male damselflies could mediate female colour morph detection

Abstract:The visual systems of Odonata are characterized by many opsin genes, which form the primary light-sensitive photopigments of the eye. Female-limited colour polymorphisms are also common in Odonata, with one morph typically exhibiting male-like (androchrome) colouration and one or two morphs exhibiting female-specific colouration (gynochromes). These colour polymorphisms are thought to be maintained by frequency-dependent sexual conflict, in which males form search images for certain morphs, causing disproportionate mating harassment. Here, we investigate opsin sensitivity and gene expression plasticity in mate-searching males of the damselfly Ischnura elegans during adult maturation and across populations with different female morph frequencies. We find evidence for opsin-specific plasticity in relative and proportional opsin mRNA expression, suggesting changes in opsin regulation and visual sensitivity during sexual maturation. In particular, expression of the long-wavelength sensitive opsin LWF2 changed over development and varied between populations with different female morph frequencies. UV-Vis analyses indicate that short- and long-wavelength opsins absorb wavelength of light between 350 and 650 nm. Assuming opponency between photoreceptors with distinct short- and long-wavelength sensitivities, these sensitivities suggest male spectral visual discrimination ability of andromorph and gynomorph females. Overall, our results suggest that opsin sensitivity and expression changes contribute to visual tuning that could impact conspecific discrimination.

Local Pharmacological Effects of Tungstate on the Color-Pattern Determination of Butterfly Wings: A Possible Relationship Between the Eyespot and Parafocal Element

Abstract:Butterfly wing color patterns can be changed by the application of a temperature shock or pharmacological agents such as tungstate, producing a distinctive type of elemental modification called the TS (temperature shock) type. Heterochronic uncoupling between the signaling and reception steps during the color-pattern determination process has been proposed as a mechanism for TS-type changes. As an extension of this hypothesis, both the parafocal element (PFE) and the eyespot in the same wing compartment are considered to be determined by morphogenic signal(s) emitted from the same eyespot focus. However, these models need to be examined with additional experimental data. Furthermore, there is controversy as to whether the action of tungstate on wing color patterns is direct or Indirect. Using a species of nymphalid butterfly (Junonia orithya), we have devised a simple method for the local application of pharmacological agents directly on developing wings of pupae. Local tungstate application resulted in reduced eyespots and circular dislocated PFEs in the eyespot-less compartments only on the treated wing, demonstrating that tungstate directly induces color-pattern changes on wings. We further examined the eyespot-PFE relationship in normal and cold-shocked Individuals, showing that an eyespot can be superimposed on a PFE and vice versa, probably depending on the timing of their fate determination. Taken together, we propose a two-morphogen model for the normal color-pattern determination, in which the morphogenic signals for the eyespot and PFE are different from each other despite their Identical origin. This two-morphogen model is compatible with the heterochronic uncoupling model for TS-type changes.

Effect of monochromatic light on light adaptation and opsin expression in Ectropis grisescens

Abstract:Light has a substantial effect on the behaviour and physiology of nocturnal moths. Ectropis grisescens is a major nocturnal tea pest in China, and light traps are commonly used to control geometrid moths because of their positive phototaxis. However, some moths gather around light traps and enter the light adaptation state, which decreases the efficacy of light traps in controlling this pest. We identified opsin genes and the spectral sensitivities of the photoreceptors of E. grisescens moths. We also determined the effects of several monochromatic lights on opsin gene expression and light adaptation. We detected three types of opsin genes and six spectral sensitive peaks (at 370, 390, 480, 530, 550, and 580 nm). We also observed significant changes in the diurnal rhythm of opsin gene expression under different light conditions. When active males were suddenly exposed to different monochromatic lights, they quickly entered the light adaptation state, and the adaptation time was negatively correlated with the light intensity. Males were most sensitive to 390 nm wavelengths, followed by 544 nm, 457 nm, and 593 nm. Red light (627 nm) did not affect the activity of E. grisescens males but had detectable physiological effects.

Evolutionary renovation of L/M opsin polymorphism confers a fruit discrimination advantage to ateline New World monkeys

Abstract:New World monkeys exhibit prominent colour vision variation due to allelic polymorphism of the long-to-middle wavelength (L/M) opsin gene. The known spectral variation of L/M opsins in primates is broadly determined by amino acid composition at three sites: 180, 277 and 285 (the 'three-sites' rule). However, two L/M opsin alleles found in the black-handed spider monkeys (Ateles geoffroyi) are known exceptions, presumably due to novel mutations. The spectral separation of the two L/M photopigments is 1.5 times greater than expected based on the 'three-sites' rule. Yet the consequence of this for the visual ecology of the species is unknown, as is the evolutionary mechanism by which spectral shift was achieved. In this study, we first examine L/M opsins of two other Atelinae species, the long-haired spider monkeys (A. belzebuth) and the common woolly monkeys (Lagothrix lagotricha). By a series of site-directed mutagenesis, we show that a mutation Y213D (tyrosine to aspartic acid at site 213) in the ancestral opsin of the two alleles enabled N294K, which occurred in one allele of the ateline ancestor and increased the spectral separation between the two alleles. Second, by modelling the chromaticity of dietary fruits and background leaves in a natural habitat of spider monkeys, we demonstrate that chromatic discrimination of fruit from leaves is significantly enhanced by these mutations. This evolutionary renovation of L/M opsin polymorphism in atelines illustrates a previously unappreciated dynamism of opsin genes in shaping primate colour vision.
Behavioural red-light sensitivity in fish according to the optomotor response

Abstract:Various procedures have been adopted to investigate spectral sensitivity of animals, e.g. absorption spectra of visual pigments, electroretinography, optokinetic response, optomotor response (OMR) and phototaxis. The use of these techniques has led to various conclusions about animal vision. However, visual sensitivity should be evaluated consistently for a reliable comparison. In this study, we retrieved behavioural data of several fish species using a single OMR procedure and compared their sensitivities to near-infrared light. Besides cavefish that lack eyes, some species were not appropriate for the OMR test because they either stayed still or changed swimming direction frequently. Eight of 13 fish species tested were OMR positive. Detailed analyses using medaka, goldfish, zebrafish, guppy, stickleback and cichlid revealed that all the fish were sensitive to light at a wavelength greater than or equal to 750 nm, where the threshold wavelengths varied from 750 to 880 nm. Fish opsin repertoire affected the perception of red light. By contrast, the copy number of long-wavelength-sensitive ( LWS ) genes did not necessarily improve red-light sensitivity. While the duplication of LWS and other cone opsin genes that has occurred extensively during fish evolution might not aid increasing spectral sensitivity, it may provide some other advantageous ophthalmic function, such as enhanced spectral discrimination.

Abstract:Morphological organization, ultrastructure and adaptational changes under different light intensities (10000, 100, 1, and 0.01 mW/m 2 ) of the compound eye of the oriental fruit moth Grapholita molesta (Busck 1916) were investigated. Its superposition type of eyes consist of approximately 1072 ommatidia in males and 1029 ommatidia in females with ommatidial diameters of around 15 μm. Each ommatidium features a laminated corneal lens densely covered by corneal nipples of 256 nm in height. Crystalline cones are formed by four cone cells, proximally tapering to form a narrow crystalline tract with a diameter of 1.5 μm. Eight retinula cells, two primary and six secondary pigment cells per ommatidium are present. The 62.3 μm long rhabdom is divided into a thin 1.8 μm wide distal and a 5.2 μm wide proximal region. Distally the fused rhabdom consists of the rhabdomeres of seven retinula cells (R1-R7) and connects with the crystalline cone. In the proximal rhabdom region, the pigment-containing retinula cell R8 occupies a position in centre of the rhabdom while R1-R7 cells have taken peripheral positions. At this level each ommatidial group of retinula cells is surrounded by a tracheal tapetum. In response to changes from bright-light to dim-light adaptations, the pigment granules in the secondary pigment cells and retinula cells migrate distally, with a decrease in the length of crystalline tract.

Abstract:The first exploratory study was conducted on the compound eye morphology and spectral characteristics of Agasicleshygrophila (Selman & Vogt, 1971) to clarify its eye structure and its spectral sensitivity. Scanning electron microscopy, paraffin sectioning, and transmission electron microscopy revealed that A.hygrophila has apposition compound eyes with both eucones and open rhabdom. The micro-computed tomography (CT) results after 3D reconstruction demonstrated the precise position of the compound eyes in the insect's head and suggested that the visual range was mainly concentrated in the front and on both sides of the head. The electroretinogram (ERG) experiment showed that red, yellow, green, blue, and ultraviolet light could stimulate the compound eyes of A.hygrophila to produce electrical signals. The behavioural experiment results showed that both males and females had the strongest phototaxis to yellow light and positive phototaxis to red, green, and blue light but negative phototaxis to UV light. This study of the compound eyes of A.hygrophila will be helpful for decoding its visual mechanism in future studies.
Molecular Mechanisms Underlying the Spectral Shift in Zebrafish Cone Opsins

Abstract:Visual pigments are essential for converting light into electrical signals during vision. Composed of an opsin protein and a retinal-based chromophore, pigments in vertebrate rods (Rh1) and cones (Rh2) have different spectral sensitivities, with distinct peak absorption wavelengths determined by the shape and composition of the chromophore binding pocket. Despite advances in understanding Rh1 pigments such as bovine rhodopsin, the molecular basis of spectral shifts in Rh2 cone opsins has been less studied, particularly the E122Q mutation, which accounts for about half of the observed spectral shift in these pigments. In this study, we employed molecular modeling and quantum mechanical techniques to investigate the molecular mechanisms behind the spectral difference in blue-shifted Rh2-1 (absorption peak = 467 nm, 122Q) and green-shifted Rh2-4 (absorption peak = 505 nm, 122E) zebrafish cone opsins. We modeled the pigments 3D structures based on their sequences and conducted all-atom molecular dynamics simulations totaling 2 microseconds. Distance analysis of the trajectories identified three key sites: E113, E181, and E122. The E122Q mutation, previously known, validates our findings, while E181 and E113 are newly identified contributors. Structural analysis revealed key features with differing values that explain the divergent spectral sensitivities of Rh2-1 and Rh2-4: 1) chromophore atom fluctuations and C5-C6 torsion angle, 2) binding pocket volume, 3) hydration patterns, and 4) E113- chromophore interaction stability. Quantum mechanics further confirms the critical role of residue E181 in Rh2-1 and E122 in Rh2-4 for their spectral behavior. Our study provides new insights into the molecular determinants of spectral shifts in cone opsins, and we anticipate that it will serve as a starting point for a broader understanding of the functional diversity of visual pigments.

Physiologically induced color-pattern changes in butterfly wings: Mechanistic and evolutionary implications

Abstract:A mechanistic understanding of the butterfly wing color-pattern determination can be facilitated by experimental pattern changes. Here I review physiologically induced color-pattern changes in nymphalid butterflies and their mechanistic and evolutionary implications. A type of color-pattern change can be elicited by elemental changes in size and position throughout the wing, as suggested by the nymphalid groundplan. These changes of pattern elements are bi-directional and bi-sided dislocation toward or away from eyespot foci and in both proximal and distal sides of the foci. The peripheral elements are dislocated even in the eyespot-less compartments. Anterior spots are more severely modified, suggesting the existence of an anterior-posterior gradient. In one species, eyespots are transformed into white spots with remnant-like orange scales, and such patterns emerge even at the eyespot-less "imaginary" foci. A series of these color-pattern modifications probably reveal "snap-shots" of a dynamic morphogenic signal due to heterochronic uncoupling between the signaling and reception steps. The conventional gradient model can be revised to account for these observed color-pattern changes.

Abstract:Spectral responses from the compound eyes of 35 lepidopteran species representing 14 families were investigated electrophysiologically using ERG recordings. The light-stimuli used overed the range of 383–700 nm wavelengths. All species show three or four maxima in their spectral sensitivity curves. Two of these peaks were usually associated with ultraviolet and blue light (383 and 460 nm, respectively). The other maxima occurred in the 500–620 nm region. In Nymphalidae the highest peak was found in response to 560–580 nm stimuli. Of all wavelengths tested, these are the longest wavelengths to produce principal peak sensitivities.Pieridae and Lycaenidae have maxima in the UV region which represent significantly higher sensitivities than the secondary peaks to stimuli of longer wavelengths.Satyridae, Danaidae, Hesperiidae and diurnal moths except Epicopeia (Epicopeidae) generally have similar sensitivity curves with principal peaks between 500 and 520 nm.In Papilionid species except Graphium (max = 560 nm) high maxima occur in the UV and blue (460 nm) region.Noctural Sphingid moths possess the highest peak sensitivity at 540 nm. All other noctural moths tested have three or four maxima.

Abstract:We studied the anatomy and physiology of photoreceptors and second‐order neurons of a butterfly, Papilio xuthus. Of nine photoreceptors in an ommatidium, two are long visual fibers (LVFs), sensitive to either ultraviolet, violet, or blue, terminating in the medulla. We found the ultraviolet and violet receptors had bushier terminals. The photoreceptors form a module with five second‐order lamina monopolar cells (LMCs) of different morphology projecting from the lamina to the medulla. Spectral sensitivity measurement grouped the LMCs into three classes irrespective of the cells' morphology. The results indicate that the distal medulla is the most plausible region for early visual integration. Extensive analysis of the flower‐visiting behavior of a butterfly, Papilio xuthus, has indicated complex interaction between chromatic, achromatic, and motion cues. Their eyes are spectrally rich with six classes of photoreceptors, respectively sensitive in the ultraviolet, violet, blue, green, red, and broad‐band wavelength regions. Here, we studied the anatomy and physiology of photoreceptors and second‐order neurons of P. xuthus, focusing on their spectral sensitivities and projection terminals to address where the early visual integration takes place. We thus found the ultraviolet, violet, and blue photoreceptors and all second‐order neurons terminate in the distal region of the second optic ganglion, the medulla. We identified five types of second‐order neurons based on the arborization in the first optic ganglion, the lamina, and the shape of the medulla terminals. Their spectral sensitivity is independent of the morphological types but reflects the combination of pre‐synaptic photoreceptors. The results indicate that the distal medulla is the most plausible region for early visual integration.

Abstract:Abstract Background Eye pigmentation genes have been utilized as visible markers for constructing genetic control prototypes in several insect vectors of human disease. Here, orthologs of two ommochrome pathway genes, kynurenine 3-hydroxylase ( kmo ) and cardinal , were investigated in Plutella xylostella , a globally distributed, economically important pest of Brassica crops. Results Both somatic mosaic and germline mutations were efficiently created using the CRISPR/Cas9 system, and null mutant strains of Pxkmo and Pxcardinal were obtained. A frame-shift mutation in Pxkmo caused yellow compound eyes at adult stage while an in-frame mutation lacking two amino acids resulted in a hypomorphic red eye phenotypes. In contrast, Pxcardinal -deficient moths with a frame-shift mutation exhibited yellow eye pigmentation in newly emerged adults which turned to red as the adults aged. Additionally, differences were observed in the coloration of larval ocelli, brains and testes in Pxkmo and Pxcardinal yellow-eye mutant lines. Conclusions Our work identifies the important roles of Pxkmo and Pxcardinal in P. xylostella eye pigmentation and provides tools for future genetic manipulation of this important crop pest.

A simple method for studying the molecular mechanisms of ultraviolet and violet reception in vertebrates

Abstract:Background: Many vertebrate species use ultraviolet (UV) reception for such basic behaviors as foraging and mating, but many others switched to violet reception and improved their visual resolution. The respective phenotypes are regulated by the short wavelength-sensitive (SWS1) pigments that absorb light maximally (λmax) at ~360 and 395-440 nm. Because of strong epistatic interactions, the biological significance of the extensive mutagenesis results on the molecular basis of spectral tuning in SWS1 pigments and the mechanisms of their phenotypic adaptations remains uncertain. Results: The magnitudes of the λmax-shifts caused by mutations in a present-day SWS1 pigment and by the corresponding forward mutations in its ancestral pigment are often dramatically different. To resolve these mutagenesis results, the A/B ratio, in which A and B are the areas formed by amino acids at sites 90, 113 and 118 and by those at sites 86, 90 and 118 and 295, respectively, becomes indispensable. Then, all critical mutations that generated the λmax of a SWS1 pigment can be identified by establishing that 1) the difference between the λmax of the ancestral pigment with these mutations and that of the present-day pigment is small (3 ~ 5 nm, depending on the entire λmax-shift) and 2) the difference between the corresponding A/B ratios is < 0.002. Conclusion: Molecular adaptation has been studied mostly by using comparative sequence analyses. These statistical results provide biological hypotheses and need to be tested using experimental means. This is an opportune time to explore the currently available and new genetic systems and test these statistical hypotheses. Evaluating the λmaxs and A/B ratios of mutagenized present-day and their ancestral pigments, we now have a method to identify all critical mutations that are responsible for phenotypic adaptation of SWS1 pigments. The result also explains spectral tuning of the same pigments, a central unanswered question in phototransduction.
