Abstract:Chromium (Cr) contamination of agricultural soils is a major threat to human and plant health worldwide and causes reductions in plant growth and crop yields. 24-epibrassinolide (EBL) and nitric oxide (NO) have been shown to ameliorate the reductions in growth caused by the stresses induced by heavy metals; however, the interactions between EBL and NO on the alleviation of Cr-induced phytotoxicity have been poorly studied. Hence, this study was undertaken to examine any beneficial effects of EBL (0.01 µM) and NO (100 µM), applied alone or in combination, on the mitigation of stress induced by Cr (100 µM) in soybean seedlings. Although EBL and NO applied alone reduced the toxic effects of Cr, the combined treatment had the greatest effect. Mitigation of Cr intoxication occurred via reduced Cr uptake and translocation and by ameliorating reductions in water contents, light-harvesting pigments, and other photosynthetic parameters. In addition, the two hormones increased the activity of enzymatic and non-enzymatic defense mechanisms increasing the scavenging of reactive oxygen species, thereby reducing membrane damage and electrolyte leakage. Furthermore, the hormones reduced the accumulation of the toxic compound, methylglyoxal, by amplifying activities of glyoxalase I and glyoxalase II. Thus, applications of NO and EBL can significantly mitigate Cr-phytotoxicity when cultivating soybean plants in Cr-contaminated soils. However, further more-in depth studies including field investigations parallel with calculations of cost to profit ratios and yield losses are requested to validate the effectiveness of NO and/or EBL for remediation agents in Cr-contaminated soils with using key biomarkers (i.e., oxidative stress, antioxidant defense, and osmoprotectants) involved in the uptake, accumulation, and attenuation of Cr toxicity tested in our study.
Implications for Warning Symbol Design

Abstract:IN BRIEF •To investigate age-related differences in warning symbol comprehension, younger and older adults were tested on their comprehension for symbols varying on familiarity, complexity and comprehensibility. •Older adults’ comprehension was poorer than younger adults both before and after training. •More familiar and more comprehensible symbols were more easily understood by both younger and older adults, whereas high complexity symbols were more difficult for older adults. Following training, only comprehensibility showed a relationship with comprehension and only for older adults. •To optimize comprehension by older adults, complexity should be minimized and symbols should have a clear relationship with a real-world referent. Abstract:Chromium (Cr) is one of the top seven toxic heavy metals, being ranked 21st among the abundantly found metals in the earth’s crust. A huge amount of Cr releases from various industries and Cr mines, which is accumulating in the agricultural land, is significantly reducing the crop development, growth, and yield. Chromium mediates phytotoxicity either by direct interaction with different plant parts and metabolic pathways or it generates internal stress by inducing the accumulation of reactive oxygen species (ROS). Thus, the role of Cr-induced ROS in the phytotoxicity is very important. In the current study, we reviewed the most recent publications regarding Cr-induced ROS, Cr-induced alteration in the enzymatic antioxidant system, Cr-induced lipid peroxidation and cell membrane damage, Cr-induced DNA damage and genotoxicity, Cr-induced ultrastructural changes in cell and subcellular level, and Cr-induced alterations in photosynthesis and photosynthetic apparatus. Taken together, we conclude that Cr-induced ROS and the suppression of the enzymatic antioxidant system actually mediate Cr-induced cytotoxic, genotoxic, ultrastructural, and photosynthetic changes in plants.

Abstract:Chromium (Cr) is a familiar heavy element and is mainly available in nature with a couple of stable oxidation states such as Cr +3 and Cr +6 . Its soil–plant transfer is accomplished mostly by sulphate or iron ions which serve as carriers without a transport system. The entry of Cr +6 into the plant cells induces biochemical, molecular, cytotoxic and genotoxic alterations leading to the inhibition of seed germination, seedling growth and subsequent development. The harmful effects of Cr +6 /Cr +3 on plants comprise growth inhibition of roots, shoots and leaves which affect plant biomass and productivity. Cr +6 /Cr +3 also causes detrimental consequences on many plant metabolic processes, viz. absorption of water, mineral uptake, hormonal distribution, flowering, senescence, photosynthesis, respiration, carbohydrate, nitrogen, lipid and stress metabolism. Chromium induces phytotoxicity by the generation of free oxygen radicals, while antioxidant enzyme may alleviate the stress. Basically, plant stress resistance may occur through either an avoidance or tolerance mechanisms. The studies in this review highlight the behaviour of Cr in the soil plant system, Cr-speciation, Cr-phytoavailability and Cr-phytotoxicity with fragmentary information on Cr-induced toxicity effects on plant metabolism. Thus, this comprehensive review addresses available information published during the last 3–4 decades. We focussed on the inhibition of growth and development by ions of chromium, starting from seed germination, and with an emphasis on metabolic processes in plant organs. The issue of plant stress resistance mechanisms was also included. We suggest the need to reduce pollution with chromium compounds for a cleaner environment and ensuring the security of sustainable developments.

Abstract:The ever-growing anthropogenic need for development by industrialization is causing severe environmental pollution. Chromium is mainly found in nature in 2 stable forms where Cr 6+ is more toxic and mutagenic than Cr 3+ . Cr is generally known to induce the production of reactive oxygen species (ROS), which in turn affects various metabolic and physiological processes such as alteration in enzymatic antioxidant system, induction of lipid peroxidation and cell membrane damage, ultrastructural changes in cellular and sub-cellular level, alteration in cellular organelles including plastid and mitochondria and causing DNA damage and genotoxicity. Several studies have been conducted in the recent past regarding, genetic and transcriptional regulations for conferring tolerance to stress. However, plants in response to such damaging stress can generate their defense mechanism which helps them alleviate the stress. Among such defense mechanisms important are restriction uptake, intracellular sequestration, complexation, and chelation as well as induction of the antioxidant system. However, it has been seen that exogenous application of various organic acids, amino acids, and nitrous oxide derivatives has facilitated plant growth as well as helped strengthen its defense mechanism against chromium toxicity. We have, therefore, focused on the interaction of chromium with plants emphasizing photosynthetic metabolism, oxidative stress, plant signaling responses, and genotoxic responses of plants towards chromium toxicity explaining various adaptive responses of plants to cope with the chromium stress.

Brassinosteroid Supplementation Alleviates Chromium Toxicity in Soybean (glycine Max L.) Via Reducing Its Translocation

Abstract:Chromium (Cr) phytotoxicity severely inhibits plant growth and development which makes it a prerequisite to developing techniques that prevent Cr accumulation in food chains. However, little is explored related to the protective role of brassinosteroids (BRs) against Cr-induced stress in soybean plants. Herein, the morpho-physiological, biochemical, and molecular responses of soybean cultivars with/without foliar application of BRs under Cr toxicity were intensely investigated. Our outcomes deliberated that BRs application noticeably reduced Cr-induced phytotoxicity by lowering Cr uptake (37.7/43.63%), accumulation (63.92/81.73%), and translocation (26.23/38.14%) in XD-18/HD-19, plant tissues, respectively; besides, improved seed germination ratio, photosynthetic attributes, plant growth, and biomass, as well as prevented nutrient uptake inhibition under Cr stress, especially in HD-19 cultivar. Furthermore, BRs stimulated antioxidative defense systems, both enzymatic and non-enzymatic, the compartmentalization of ion chelation, diminished extra production of reactive oxygen species (ROS), and electrolyte leakage in response to Cr-induced toxicity, specifically in HD-19. In addition, BRs improved Cr stress tolerance in soybean seedlings by regulating the expression of stress-related genes involved in Cr accumulation, and translocation. Inclusively, by considering the above-mentioned biomarkers, foliar spray of BRs might be considered an effective inhibitor of Cr-induced damages in soybean cultivars, even in Cr polluted soil.

Abstract:Environmental sources of chromium (Cr) such as solid waste, battery chemicals, industrial /waste, automotive exhaust emissions, mineral mining, fertilizers, and pesticides, have detrimental effects on plants. An excessive amount of Cr exposure can lead to toxic accumulations in human, animal, and plant tissues. In plants, diverse signaling molecules like hydrogen sulfide (H2S) and nitric oxide (NO) play multiple roles during Cr stress. Consequently, the molecular mechanisms of Cr toxicity in plants, such as metal binding, modifying enzyme activity, and damaging cells are examined by several studies. The reactive oxygen species (ROS) that are formed when Cr reacts with lipids, membranes, DNA, proteins, and carbohydrates are all responsible for damage caused by Cr. ROS regulate plant growth, programmed cell death (PCD), cell cycle, pathogen defense, systemic communication, abiotic stress responses, and growth. Plants accumulate Cr mostly through the root system, with very little movement to the shoots. The characterization of stress-inducible proteins and metabolites involved in Cr tolerance and cross-talk messengers has been made possible due to recent advances in metabolomics, transcriptomics, and proteomics. This review discusses Cr absorption, translocation, subcellular distribution, and cross-talk between secondary messengers as mechanisms responsible for Cr toxicity and tolerance in plants. To mitigate this problem, soil-plant systems need to be monitored for the biogeochemical behavior of Cr and the identification of secondary messengers in plants.
Effects of Chromium Stress on the Content of Phytochelatins and Antioxidative Characteristics in Brassica chinensis L.

Abstract:The content of phytochelatins and the activities of antioxidative enzymes in varieties of Chunyou 1 and Qingfuling were investigated by hydroponic experiments.The results showed that the activities of antioxidant enzymes and the content of acid soluble SH,phytochelatins(PCs) and reduced glutathione(GSH) increased with the increase of Cr3+ concentration for varieties of Chunyou 1 and Qingfuling(P0.05);Under the concentration of 300 mg·L-1 Cr3+ treatment,the activities of superoxide dismutase(SOD),peroxidase(POD),catalase(CAT) in Qingfuling were 24.2%,41.7% and 21.4% higher than control,and the activities of these enzymes in Chunyou 1 were 37.2%,70.7%,43.0% higher than control,respectively,PCs content was 3.17 times as much as control in Chunyou 1,and it was 1.71 times as much as control in Qingfuling.Chunyou 1 was more Cr-resistance than Qingfuling and appeared higher activities of antioxidative enzymes and content of PCs.The physiological respondence varied significantly between different varieties of Brassica chinensis L.under Cr3+ stress.
Effects of Chromium Stress on the Content of Phytochelatins and the Activities of Key Enzymes of Ascorbate-glutathione Cycle in Brassica Chinensis L.

Abstract:The content of phytochelatins and the activities of key enzymes of Ascorbate-glutathione cycle in varieties of Aikangqing and Lvxiu were studied with different concentrations of Cr3+.The results showed that the contents of phytochelatins and antioxidant increased with the increase of Cr3+ concentration in both varieties.Under high concentration of Cr3+(250 mg.L-1),O 2.contents in leaves of cabbages were significantly increased(P0.05),and the increased amount of Aikangqing was lower than that of Lvxiu.AsA and GSH contents in leaves of Aikangqing were significantly increased compared to contro(lP0.05),while those of in Lvxiu were not significantly increased(P0.05).DHA and GSSG contents in Aikangqing were 1.87 and 1.72 times as much as control and those were 1.98 and 1.94 times as much as control in Lvxiu,respectively.The activities of key enzymes in AsA-GSH increased with the increase of Cr3+ concentration.The content of phytochelatins(PCs),the activity of ascorbte peroxidase(APX),and dehydrateasorbate reductase(DHAR)in Aikangqing were 30.9%,38.9%,45.2% higher than control under the concentration of 250 mg.L-1 Cr3+ treatment,respectively,while those in Lvxiu were 11.5%,30.5%,43.0% higher than control,respectively.In conclusion,Aikangqing had a higher activities of key enzymes of AsA-GSH and phytochelatins under Cr3+ stress,which attribute to its higher Cr3+ tolerance.Our results showed that there were chromium-resistance differences between varieties of cabbages,this provides the theoretical guidance and technical support for people had the choice of low accumulation cabbages varieties.

Abstract:The present study was conducted to examine the effects of increasing concentrations of chromium (Cr6+) (0, 25, 50, 100, and 200 μmol) on rice (Oryza sativa L.) morphological traits, photosynthesis performance, and the activities of antioxidative enzymes. In addition, the ultrastructure of chloroplasts in the leaves of hydroponically cultivated rice (O. sativa L.) seedlings was analyzed. Plant fresh and dry weights, height, root length, and photosynthetic pigments were decreased by Cr-induced toxicity (200 μM), and the growth of rice seedlings was starkly inhibited compared with that of the control. In addition, the decreased maximum quantum yield of primary photochemistry (Fv/Fm) might be ascribed to the decreased the number of active photosystem II reaction centers. These results were confirmed by inhibited photophosphorylation, reduced ATP content and its coupling factor Ca2+–ATPase, and decreased Mg2+–ATPase activities. Furthermore, overtly increased activities of antioxidative enzymes were observed under Cr6+ toxicity. Malondialdehyde and the generation rates of superoxide (O2̄) also increased with Cr6+ concentration, while hydrogen peroxide content first increased at a low Cr6+ concentration of 25 μM and then decreased. Moreover, transmission electron microscopy showed that Cr6+ exposure resulted in significant chloroplast damage. Taken together, these findings indicate that high Cr6+concentrations stimulate the production of toxic reactive oxygen species and promote lipid peroxidation in plants, causing severe damage to cell membranes, degradation of photosynthetic pigments, and inhibition of photosynthesis.

Abstract:Chromium (Cr) is present in our environment as a toxic pollutant, which needs to be removed using phytoremediation technology. In present study, two transgenic cotton cultivars (J208, Z905) and their hybrid line (ZD14) were used to explore their Cr uptake and tolerance potential using multiple biomarkers approach. Four different levels of Cr (CK, 10, 50, and 100 μM) were applied. Cr caused a significant reduction in root/shoot length, number of secondary roots, and root fresh and dry biomasses at 100 μM. Cr accumulated more in roots and was found higher in hybrid line (ZD14) as compared with its parent lines (J208, Z905) at all Cr stress levels (10, 50, and 100 μM). Cr translocation was less than 1 in all cultivars. Ultrastructural studies at 100 μM Cr showed an increase in number of nuclei and vacuoles and presence of Cr dense granules in dead parts of the cell (vacuoles/cell wall). Malondialdehyde (MDA), hydrogen peroxide (H2O2), total soluble proteins, superoxide dismutase (SOD), peroxidase (POD), ascorbate peroxidase (APX), catalase (CAT), and glutathione reductase (GR) as a whole were upregulated with elevated levels of Cr. Higher Cr uptake by roots, accelerated metabolism, and Cr sequestration in dead parts of the cell indicate that these cotton cultivars can be useful for Cr accumulation and tolerance.

Abstract:Chromium-VI (Cr (VI)) is a toxic metal pollutant to both plants and animals. In this study, we cultured two transgenic cotton cultivars (J208 (herbicide-resistant), Z905 (insect-resistant)) and their hybrid line (ZD14) in hydroponic media using 0, 10, 50, 100 µM Cr/L for 7 days. Leaf-related growth parameters such as leaf length and width considerably declined. Decrease in leaves’ fresh biomass was less in Z905 followed by ZD14 and J208, while the leaves dry biomass was less in Z905 followed by J208 and ZD14, respectively. Tolerance potential of plant height, root dry weight and leaf dry weight of both transgenic cotton cultivars significantly decreased at Cr concentration (100 µM) as compared with their hybrid line. Cr accumulation in roots tissues was higher (> 90%) than leaf tissues. Stress biomarkers (i.e., (Malondialdehyde (MDA), hydrogen peroxide (H2O2), total soluble protein (TSP)) as a whole upregulated in leaf tissues of all experimental cultivars. Differential changes in superoxide dismutase (SOD) activity were observed, while peroxidase (POD), catalase (CAT) and glutathione-reductase (GR) activities almost showed increasing trend in all cultivars under exceeding levels of Cr. Correlation studies, in terms of principal component analysis (PCA), revealed that 75.36% of the total variation was present among all the traits. The leaf-based ultrastructural modifications at 100 µM Cr stress level occurred more in Z905 followed by J208 and ZD14. All these changes revealed differential role of experimental cotton cultivars against Cr stress. Hybrid line (ZD14) performed well in comparison with its parental cultivars (J208, Z905) as a whole. Deeper molecular studies like transcriptomic and metabolomic need to be carried out to explore the genetic facts regarding such differential responses among these cotton cultivars under Cr stress.

Abstract:This article presents an overview of the mechanism of chromium (Cr) stress in plants. Toxic effects of Cr on plant growth and development depend primarily on its valence state. Cr(VI) is highly toxic and mobile whereas Cr(III) is less toxic. Cr-induced oxidative stress involves induction of lipid peroxidation in plants that cause severe damage to cell membranes which includes degradation of photosynthetic pigments causing deterioration in growth. The potential of plants with the adequacy to accumulate or to stabilize Cr compounds for bioremediation of Cr contamination has gained engrossment in recent years.

Abstract:Chromium (Cr) is an element naturally occurring in rocky soils and volcanic dust. It has been classified as a carcinogen agent according to the International Agency for Research on Cancer. Therefore, this metal needs an accurate understanding and thorough investigation in soil–plant systems. Due to its high solubility, Cr (VI) is regarded as a hazardous ion, which contaminates groundwater and can be transferred through the food chain. Cr also negatively impacts the growth of plants by impairing their essential metabolic processes. The toxic effects of Cr are correlated with the generation of reactive oxygen species (ROS), which cause oxidative stress in plants. The current review summarizes the understanding of Cr toxicity in plants via discussing the possible mechanisms involved in its uptake, translocation and sub-cellular distribution, along with its interference with the other plant metabolic processes such as chlorophyll biosynthesis, photosynthesis and plant defensive system.

Chromium stress induced oxidative burst in Vigna mungo (L.) Hepper: physio-molecular and antioxidative enzymes regulation in cellular homeostasis

Abstract:Vigna mungo (L.) Hepper commonly known as blackgram is an important legume crop with good quality dietary proteins and vitamins. Low production of blackgram in the chromium rich soil of Odisha is a serious concern against its demand. Chromium (VI) was tested on V. mungo var. B3-8-8 at 100, 150, 200, 250 and 300 µM concentration on growth, anti-oxidative enzymes and chromium content at 15, 30 and 45 d of treatments. Seed germination and growth decreased with increase dose and duration. Cr uptake induced oxidative burst with significant increase of osmolytes was observed in cell at lower doses but failed to adjust homeostasis at higher dose. Increase of GPX and SOD and decrease of CAT was observed as dose dependent. Increased protein content was detected in < 200 µM Cr concentration whereas, significant decrease of protein was noted thereafter. Down regulation of proteins (29.2 kDa and 32.6 kDa) was observed at > 250 µM of Cr. Total Cr uptake was greater in root than in shoot which might be due to poor translocation of heavy metal or detoxification. Thus, blackgram was able to maintain homeostasis at lower concentrations of Cr by activating the cascade of enzymes following cellular detoxification mechanism. Abstract:Chromium contamination of the soil is a major scientific concern with reference to crop productivity and human health. In recent years, several approaches are being employed in managing metal toxicity in crop plants. Here, we have investigated about potential and probable crosstalk of nitric oxide (NO) and hydrogen peroxide (H2O2) in mitigating hexavalent chromium [Cr(VI)] toxicity in wheat seedlings. Cr(VI) toxicity reduced the fresh mass and overall growth due to accumulation of reactive oxygen species (ROS) and decreased efficiency of AsA-GSH cycle and downregulation of high affinity sulfate transporter. However, exogenous treatment of NO and H2O2 significantly alleviated Cr toxicity. Application of NO and ROS scavengers reversed stress mitigating effects of NO and H2O2, respectively suggesting that endogenous NO and H2O2 are necessary for rendering Cr toxicity tolerance. Furthermore, NO rescued negative effect of diphenylene iodonium (DPI, NADPH oxidase inhibitor) and H2O2 reversed the negative effect of c-PTIO suggesting that they exhibit independent signalling in mitigating Cr stress. Altogether, data indicated that NO and H2O2 rendered mitigation of Cr stress by up-regulating enzymes (activity and relative gene expression) and metabolites of AsA-GSH cycle, high affinity sulfate transporter (relative gene expression) and glutathione biosynthesis which collectively controlled occurrence of oxidative stress.
Regulation of chromium translocation to shoot and physiological, metabolomic, and ionomic adjustments confer chromium stress tolerance in the halophyte Suaeda maritima

Abstract:Chromium (Cr) is a highly toxic element adversely affecting the environment, cultivable lands, and human populations. The present study investigated the effects of Cr (VI) (100-400 μM) on plant morphology and growth, photosynthetic pigments, organic osmolytes, ionomics, and metabolomic dynamics of the halophyte Suaeda maritima to decipher the Cr tolerance mechanisms. Cr exposure reduced the growth and biomass in S. maritima. The photosynthetic pigments content significantly declined at higher Cr concentrations (400 μM). However, at lower Cr concentrations (100-300 μM), the photosynthetic pigments remained unaffected or increased. The results suggest that a high concentration of Cr exposure might have adverse effects on PS II in S. maritima. The enhanced uptake of Na+ in S. maritima imposed to Cr stress indicates that Na+ might have a pivotal role in osmotic adjustment, thereby maintaining water status under Cr stress. The proline content was significantly upregulated in Cr-treated plants suggesting its role in maintaining osmotic balance and scavenging ROS. The metabolomic analysis of control and 400 μM Cr treated plants led to the identification of 62 metabolites. The fold chain analysis indicated the upregulation of several metabolites, including phytohormones (SA and GA3), polyphenols (cinnamic acid, sinapic acid, coumaric acid, vanillic acid, and syringic acid), and amino acids (alanine, leucine, proline, methionine, and cysteine) under Cr stress. The upregulation of these metabolites suggests the enhanced metal chelation and sequestration in vacuoles, reducing oxidative stress by scavenging ROS and promoting photosynthesis by maintaining the chloroplast membrane structure and photosynthetic pigments. Furthermore, in S. maritima, Cr tolerance index (Ti) was more than 60% in all the treatments, and Cr bio-concentration factor (BCF) and translocation factor (Tf) values were all greater than 1.0, which clearly indicates the Cr-hyperaccumulator characteristics of this halophyte.

Abstract:Wind generation power output estimation is always associated with some uncertainties as a result of wind speed and other weather parameters intermittency, and accurate short-term predictions are important for their efficient operation. This can greatly help transmission and distribution system operators and schedulers to improve the power network control and management. In this paper, a double stage hierarchical genetic algorithm based adaptive neuro-fuzzy inference system (double-stage hybrid GA-ANFIS) approach is proposed for short-term wind power forecast of a microgrid wind farm in Beijing, China. The approach has two hierarchical stages. The first GA-ANFIS stage utilizes numerical weather prediction (NWP) meteorological parameters to predict wind speed at the wind farm exact site and turbine hub height. The second stage maps the actual wind speed and power relationships. Then, the forecasted next day's wind speed by the first stage is applied to the second stage to predict next day's wind power. The presented approach has achieved considerable prediction accuracy enhancement. The accuracy of the proposed model is compared with other four forecasting methods and resulted in the best accuracy improvement of all.
Chromium (VI)-Induced Alterations in Physio-Chemical Parameters, Yield, and Yield Characteristics in Two Cultivars of Mungbean (Vigna radiata L.)

Abstract:Chromium (Cr) presently used in various major industries and its residues possess a potent environmental threat. Contamination of soil and water resources due to Cr ions and its toxicity has adversely affected plant growth and crop productivity. Here, deleterious effects of different levels of Cr (VI) treatments i.e., 0, 30, 60, 90, and 120 μM on two mungbean cultivars, Pusa Vishal (PV) and Pusa Ratna (PR), in hydroponic and pot conditions were evaluated. Germination, seedling growth, biomass production, antioxidant enzyme, electrolytic leakage, oxidative stress (hydrogen peroxide and malondialdehyde), and proline content were determined to evaluate the performance of both cultivars under hydroponic conditions for 15 days. The hydroponic results were further compared with the growth and seed yield attributes of both the genotypes in pot experiments performed over 2 years. Seedling growth, biomass production, total chlorophyll (Chl), Chl-a, Chl-b, nitrogen content, plant height, seed protein, and seed yield decreased significantly under the 120 μM Cr stress level. Activities of antioxidant enzymes superoxide dismutase, catalase, ascorbate peroxidase and peroxidase increased in the leaves following Cr exposure at 60-90 μM but declined at 120 μM. Cr-induced reductions in growth and seed yield attributes were more in the sensitive than in the tolerant cultivar. Cr accumulation in the roots, stems, leaves, and seeds increased with an increase in Cr concentrations in the pot conditions. Furthermore, for both cultivars, there were significant negative correlations in morpho-physiological characteristics under high Cr concentrations. Overall results suggest that (PR) is more sensitive to Cr stress (PV) at the seedling stage and in pot conditions. Furthermore, (PV) can be utilized to study the mechanisms of Cr tolerance and in breeding programs to develop Cr-resistant varieties.

Abstract:AIM:The aim of this study was to investigate the mechanisms of Cr(6+)-induced red blood cells (RBCs) damage.METHODS:The effect of Cr(6+) exposure on RBCs was evaluated by hemolytic rate and blood gas assays. After exposure to 20 μM Cr(6+), the percentage of phosphatidylserine (PS)-exposing cells, intracellular Ca(2+), reactive oxygen species (ROS), and ATP levels were evaluated, and cell morphology was observed. RBCs were exposed to Cr(6+) in different Ringer solutions to investigate the role of changes of Ca(2+), ROS, and ATP levels in eryptosis and morphology.RESULTS:The Cr(6+)-induced damage of RBCs was dose-dependent. After a 6 h incubation with Cr(6+), RBCs exhibited significant Ca(2+) influx, ROS increase, ATP depletion, and PS exposure, but displayed no obvious change in morphology at this time point. After 24 h Cr(6+) exposure, the RBCs decreased in size, appeared to be spike-like, and had decreased forward scatter. Inhibition of Ca(2+) influx attenuated PS-exposure caused by 6 h Cr(6+) exposure, but did not prevent 24 h Cr(6+) exposure-induced morphological change in RBCs. Inhibition of rapid ATP consumption using adenine significantly ameliorated Cr(6+)-caused PS-exposure at 12 h, 24 h and 48 h, and prevented 24 h Cr(6+) incubation-induced morphological change in RBCs.CONCLUSION:Cr(6+) can lead to eryptosis. Ca(2+) influx, increased ROS levels, and rapid ATP consumption are closely related to Cr(6+)-induced RBCs damage. Ca(2+) influx plays an extremely important role in Cr(6+)-mediated toxicity.
