Abstract:Despite extensive research on the seasonal dynamics of non-structural carbohydrate (NSC) and nitrogen (N) concentrations, the size and relative contributions of NSC and N pools across different tree organs are not well understood. We have measured the changes in NSC and N concentrations in leaves, branches, stems and all root branch orders at monthly intervals in control and girdled trees of larch ( Larix gmelinii ) and ash ( Fraxinus mandshurica ). The biomass of each plant compartment was also determined to calculate the size of the NSC and N pools. In both species, 13–37 % of the NSC and N pools were mobilized at the beginning of the growing season. Among the mobilized pools, stems and non-absorptive roots (branch orders 4–9) acted as the largest NSC sources in larch and ash, respectively, while branches served as the largest N source in both species. After stem girdling, 22 and 50 % of the root NSC stores in larch and ash, respectively, were mobilized to maintain root activities during the growing season. Tree mortality was observed 1 year after girdling, at which time there was still an abundant NSC pool in the roots. We conclude that (1) different storage organs differ in their contribution to new tissue growth at the beginning of the growing season and that those storage organs holding higher fractions of the NSC or N pool are not necessarily those which mobilize more NSC or N; (2) tree growth may not be limited by carbon (C) availability; (3) C storage in non-absorptive roots plays an important role in maintaining tree survival after the termination of photosynthate flow from aboveground sources.

Abstract:Carbon (C) assimilation can be severely impaired during periods of environmental stress like drought or defoliation, making trees heavily dependent on the use of C reserve pools for survival; yet, dynamics of reserve use during periods of reduced C supply are still poorly understood. We used stem girdling in mature poplar trees (Populus tremula L. hybrids), a lipid-storing species, to permanently interrupt phloem C transport and induced C shortage in the isolated stem section below the girdle and monitored metabolic activity during three campaigns in the growing seasons of 2018, 2019, and 2021. We measured respiratory fluxes (CO2 and O2), NSC concentration, the respiratory substrate (based on isotopic analysis and CO2/O2 ratio) and the age of the respiratory substrate (based on radiocarbon analysis). Our study shows that poplar trees can survive long periods of reduced C supply from the canopy by switching in metabolism from recent carbohydrates to older storage pools with a potential mixture of respiratory substrates, including lipids. This mechanism of stress resilience can explain why tree decline may take many years until death occurs.

Abstract:Motivated by high oil prices, several large fleet companies initiated future plans to hybridize their fleets to establish immunity of their optimized business models against severe oil price fluctuations, and adhere to increasing awareness of environmentally friendly solutions. The hybridization projects increased maintenance costs especially for costly and degradable components such as Li-ion batteries. This paper introduces a degradation-based resource allocation policy to optimally utilize batteries on fleet level. The policy, denoted as degradation-based swapping optimization, incorporates optimal implementation of swapping and substitution actions throughout a plan of finite-time horizon to minimize projected maintenance costs. The swapping action refers to the interchange in the placement of two batteries within a fleet. The substitution action refers to the replacement of degraded batteries with new ones. The policy takes advantage of the different degradation rates of the state of health of the batteries because of different loading conditions, achieving optimal placement at different time intervals throughout the plan horizon. A mathematical model for the policy is provided. The optimization of the generated model is studied through several algorithms. Numerical results for sample problems are obtained to illustrate the capability of the proposed policy in establishing substantial savings in the projected maintenance costs compared to other policies.

Abstract:Non-structural carbohydrate reserves of stems and roots underpin overall tree fitness. These reserves also contribute to productivity under short-rotation management practices such as coppicing for bioenergy. While sucrose and starch comprise the predominant carbohydrate reserves of , utilization is understood primarily in terms of starch turnover. The tonoplast sucrose transport protein SUT4 modulates sucrose export from source leaves to distant sinks during photoautotrophic growth, but the possibility of its involvement in remobilizing carbohydrates from sinks during heterotrophic growth has not been explored. Here, we used knockout mutants of (INRA 717-1B4) in winter (cool) and summer (warm) glasshouse coppicing experiments to strain carbon demand and test for SUT4 involvement in reserve utilization. We show that epicormic bud emergence, subsequent growth, and xylem hexose increases were delayed or reduced in mutants following lower temperature ‘winter’ coppicing. Depletion of stem reserves during post-coppice regrowth was not impaired in , but winter maintenance costs may have been higher in metabolic terms. Bark accrual of abundant defense metabolites, including salicinoids, chlorogenic acids, and flavonoid products, was prioritized in the summer, but attenuated in mutants. Summer sprout growth was not reduced in compared to controls. Together, our results point to shifting priorities for SUT4 modulation of seasonal trade-offs between growth and other priorities in the stem and emerging buds during reserve utilization in .

Stem girdling affects the onset of autumn senescence in aspen in interaction with metabolic signals

Abstract:Autumn senescence in aspen (Populus tremula) is precisely timed every year to relocate nutrients from leaves to storage organs before winter. Here we demonstrate how stem girdling, which leads to the accumulation of photosynthates in the crown, influences senescence. Girdling resulted in an early onset of senescence, but the chlorophyll degradation was slower and nitrogen more efficiently resorbed than during normal autumn senescence. Girdled stems accumulated or retained anthocyanins potentially providing photoprotection in senescing leaves. Girdling of one stem in a clonal stand sharing the same root stock did not affect senescence in the others, showing that the stems were autonomous in this respect. One girdled stem with unusually high chlorophyll and nitrogen contents maintained low carbon-to-nitrogen (C/N) ratio and did not show early senescence or depleted chlorophyll level unlike the other girdled stems suggesting that the responses depended on the genotype or its carbon and nitrogen status. Metabolite analysis highlighted that the tricarboxylic acid (TCA) cycle, salicylic acid pathway, and redox homeostasis are involved in the regulation of girdling-induced senescence. We propose that disrupted sink-source relation and C/N status can provide cues through the TCA cycle and phytohormone signaling to override the phenological control of autumn senescence in the girdled stems.

Abstract:Increased risk of soil waterlogging in winter and spring at northern latitudes will potentially affect forest production in the future. We studied gas exchange, chlorophyll content index, chlorophyll fluorescence, nutrient concentration and biomass accumulation in 1-year-old silver (Betula pendula Roth) and pubescent birch (Betula pubescens Ehrh.) seedlings. We hypothesized that B. pubescens has different physiological mechanisms that make it tolerate waterlogging better than B. pendula. The treatments were: (i) no waterlogging throughout the experiment; (ii) 4-week waterlogging during dormancy (dormancy waterlogging 'DW'); (iii) 4-week waterlogging during the early growing season (growth waterlogging 'GW'); and (iv) 4-week DW followed by 4-week GW during the early growing season ('DWGW'). Stomatal conductance and light-saturated net assimilation rate were reduced by GW in both species, and in B. pubescens also by DW. However, recovery was seen during the follow-up growing season. In B. pendula, DW, GW and DWGW temporarily resulted in reduced stem biomass, and GW and DWGW caused reduced leaf biomass. In B. pubescens, the stem biomass was decreased in GW and DWGW. Leaf nitrogen (N) and phosphorus (P) concentrations were generally low, and increased by GW, while potassium, calcium, magnesium and to some extent, boron and zinc concentrations decreased in both species and additionally manganese in B. pendula. The increases in N and P are mostly due to a concentration effect due to smaller leaf biomass, yet suggest that their uptake was not impaired. The decreases in cation concentrations are likely to be connected to impaired root functioning, which was not yet fully recovered from GW. We conclude that morphological acclimation to waterlogging of the leaves and roots rather than photosynthesis explains why B. pubescens is able to grow better in wetter areas than B. pendula.

Unravelling resilience mechanisms in forests: role of non-structural carbohydrates in responding to extreme weather events

Abstract:Extreme weather events are increasing in frequency and intensity due to global climate change. We hypothesized that tree carbon reserves are crucial for resilience of beech, buffering the source-sink imbalance due to late frosts and summer droughts, and that different components of non-structural carbohydrates (NSCs) play specific roles in coping with stressful situations. To assess the compound effects on mature trees of two extreme weather events, first a late frost in spring 2016 and then a drought in summer 2017, we monitored the phenology, radial growth and the dynamics of starch and soluble sugars in a Mediterranean beech forest. A growth reduction of 85% was observed after the spring late frost, yet not after the drought event. We observed a strong impact of late frost on starch, which also affected its dynamic at the beginning of the subsequent vegetative season. In 2017, the increase of soluble sugars, associated with starch hydrolysis, played a crucial role in coping with the severe summer drought. Non-structural carbohydrates helped to counteract the negative effects of both events, supporting plant survival and buffering source-sink imbalances under stressful conditions. Our findings indicate a strong trade-off between growth and NSC storage in trees. Overall, our results highlight the key role of NSCs on beech trees, response to extreme weather events, confirming the resilience of this species to highly stressful events. These insights are useful for assessing how forests may respond to the potential impacts of climate change on ecosystem processes in the Mediterranean area.

Abstract:The effects of short-term extreme events on tree functioning and physiology are still rather elusive. European beech is one of the most sensitive species to late frost and water shortage. We investigated the intra-annual C dynamics in stems under such conditions. Wood formation and stem CO2 efflux were monitored in a Mediterranean beech forest for three years (2015-2017), including a late frost (2016) and a summer drought (2017). The late frost reduced radial growth and, consequently, the amount of carbon fixed in the stem biomass by 80%. Stem carbon dioxide efflux in 2016 was reduced by 25%, which can be attributed to the reduction of effluxes due to growth respiration. Counter to our expectations, we found no effects of the 2017 summer drought on radial growth and stem carbon efflux. The studied extreme weather events had various effects on tree growth. Even though late spring frost had a strong impact on beech radial growth in the current year, trees fully recovered in the following growing season, indicating high resilience of beech to this stressful event. Unexpected phenology and lifespan of shallow and deep fine roots of walnut trees grown in a silvoarable Mediterranean agroforestry system

Abstract:Background and AimsFine roots play a major role in the global carbon cycle through respiration, exudation and decomposition processes, but their dynamics are poorly understood. Current estimates of root dynamics have principally been observed in shallow soil horizons (<1 m), and mainly in forest systems. We studied walnut (Juglans regia × nigra L.) fine root dynamics in an agroforestry system in a Mediterranean climate, with a focus on deep soils (down to 5 m), and root dynamics throughout the year.MethodsSixteen minirhizotron tubes were installed in a soil pit, at depths of 0.0–0.7, 1.0–1.7, 2.5–3.2 and 4.0–4.7 m and at two distances from the nearest trees (2 and 5 m). Fine root (diameter ≤ 2 mm) dynamics were recorded across three diameter classes every 3 weeks for 1 year to determine their phenology and turnover in relation to soil depth, root diameter and distance from the tree row.ResultsDeep (>2.5 m) root growth occurred at two distinct periods, at bud break in spring and throughout the winter i.e., after leaf fall. In contrast, shallow roots grew mainly during the spring-summer period. Maximum root elongation rates ranged from 1 to 2 cm day−1 depending on soil depth. Most root mortality occurred in upper soil layers whereas only 10 % of fine roots below 4 m died over the study period. Fine root lifespan was longer in thicker and in deeper roots with the lifespan of the thinnest roots (0.0–0.5 mm) increasing from 129 days in the topsoil to 190 at depths > 2.5 m.ConclusionsThe unexpected growth of very deep fine roots during the winter months, which is unusual for a deciduous tree species, suggests that deep and shallow roots share different physiological strategies and that current estimates based on the shortest root growth periods (i.e., during spring and summer) may be underestimating root production. Although high fine root turnover rates might partially result from the minirhizotron approach used, our results help gain insight into some of the factors driving soil organic carbon content.

Early Summer Drought Stress During the First Growing Year Stimulates Extra Shoot Growth in Oak Seedlings (Quercus petraea)

Abstract:More severe summer droughts are predicted for mid-latitudes in Europe. To evaluate the impact on forest ecosystems and more specifically on forest regeneration, we studied the response to summer drought in oak seedlings (Quercus petraea). Acorns were collected from different mother trees in three stands in Belgium, sown in pots and grown in non-heated greenhouse conditions. We imposed drought on the seedlings in early summer by first watering the pots to saturation and then stopping any watering. Weight of the pots and stomatal conductance were regularly measured. Re-watering followed this drought period of 5 weeks. Height of the seedlings and apical bud development were observed. Stomatal resistance increased toward the end of the experiment in the drought-treated group and was restored after re-watering. The seedlings from the drought treatment displayed a higher probability to produce additional shoot growth after re-watering (p ≤ 0.05). A higher competition for water (two plants per pot) increased this chance. Although this chance was also higher for smaller seedlings, the actual length of the extra growth after re-watering was higher for larger seedlings (p ≤ 0.01). Both in the drought-treated and in the control group the autochthonous provenance growing on a xeric site produced less extra shoots compared to the two other provenances. Finally, stressed plants showed less developed apical buds compared to the control group after re-watering, suggesting a phenological effect on the growth cycle of oaks (p ≤ 0.0001). The higher chance for an extra shoot growth after the drought period can be considered as a compensation for the induced growth arrest during the drought period.

Abstract:The dependence of trees on carbon and nutrient storage is critical to predicting the forest vulnerability under climate change, but whether evergreen and deciduous species differ in their use and allocation of stored resources during spring phenology is unclear. Using a high temporal resolution, we evaluated the role of spring phenology and shoot growth as determinants of the carbon and nutrient storage dynamics in contrasting leaf habits. We recorded the phenology and shoot elongation and determined the concentrations of total non-structural carbohydrates (NSCs), starch, soluble carbohydrates, nitrogen (N) and phosphorus (P) in buds, expanding shoots and previously formed shoots of two sympatric Nothofagus species with contrasting leaf habit. Species reached similar shoot lengths, though shoot expansion started 35 days earlier and lasted c. 40 days more in the deciduous species. Thus, although the deciduous species had a relatively constant shoot growth rate, the evergreen species experienced a conspicuous growth peak for c. 20 days. In the evergreen species, the greatest decreases in NSC concentrations of previously formed shoots and leaves coincided with the maximum shoot expansion rate and fruit filling, with minimums of 63 and 65% relative to values at bud dormancy, respectively. In contrast, minimum NSC concentrations of the previously formed shoots of the deciduous species were only 73% and occurred prior to the initiation of shoot expansion. Bud N and P concentrations increased during budbreak, whereas previously formed shoots generally did not decrease their nutrient concentrations. Late spring phenology and overlapping of phenophases contributed to the greater dependence on storage of proximal tissues in the studied evergreen compared with deciduous species, suggesting that phenology is a key determinant of the contrasting patterns of storage use in evergreen and deciduous species.

Abstract:Photosynthesis in woody tissues (Pwt ) is less sensitive to water shortage than in leaves, hence, Pwt might be a crucial carbon source to alleviate drought stress. To evaluate the impact of Pwt on tree drought tolerance, woody tissues of 4-m-tall drought-stressed Populus tremula trees were subjected to a light-exclusion treatment across the entire plant to inhibit Pwt . Xylem water potential (Ψxylem ), sap flow (FH2O ), leaf net photosynthesis (Pn,l ), stem diameter variations (ΔD), in vivo acoustic emissions in stems (AEs) and non-structural carbohydrate concentrations ([NSC]) were monitored to comprehensively assess water and carbon relations at whole-tree level. Under well-watered conditions, Pwt kept Ψxylem at a higher level, lowered FH2O and had no effect on [NSC]. Under drought, Ψxylem , FH2O and Pn,l in light-excluded trees rapidly decreased in concert with reductions in branch xylem starch concentration. Moreover, sub-daily patterns of ΔD, FH2O and AEs were strongly related, suggesting that in vivo AEs may inform not only about embolism events, but also about capacitive release and replenishment of stem water pools. Results highlight the importance of Pwt in maintaining xylem hydraulic integrity under drought conditions and in sustaining NSC pools to potentially limit reductions in xylem tension.

Distribution characteristics of photoassimilates in walnut leaves to different organs

Abstract:The understanding of photoassimilate distribution serves as the fundamental basis for scientific regulation of fruit quality. Currently, there is a scarcity of research on whole-plant scale photoassimilate distribution in walnut. In order to clarify the characteristics of leaf photoassimilates translocation to various organs in 5-year-old 'Wen185' (J. regia 'Wen185') walnut during the growing season, this study used the 13C isotope pulse labeling technique to label the whole plant of walnut trees in the growing season, temporal variations of 13C abundance (δ13C), 13C partition rate (R13C), leaf source strength and fruit sink strength were analyzed in various organs at different days after tree flowering. The findings indicated that during the periods of 30-70 days and 90-110 days after flowering, there was a higher distribution of 13C in fruits and vegetative branches. However, at 110-130 days after flowering, the predominant allocation of 13C shifted towards main trunk and roots. In-depth study of source leaves and sink fruits showed that chlorophyll content in leaves increased significantly 30-50 days after anthesis, indicating that they gradually became mature functional leaves. The increase of net photosynthetic rate led to increase of source strength, and the retention of photoassimilates in leaves was higher at this time. From 30 to 70 days after flowering, the fresh weight and volume of fruit increased rapidly, which increased the capacity of the sink and enhanced the competition ability against photoassimilates. The recovery of photosynthetic capacity of leaves from 90 to 110 days promoted the output of photoassimilates. At this time, walnut entered the oil conversion period, and the demand for photoassimilates increased. All these factors jointly promoted the unloading of photoassimilates in fruit. In summary, maintaining adequate material conditions and optimizing tree structure at 30-70d and 90-110d after anthesis are important for more efficient distribution of photoassimilates to fruit.

Abstract:Springtime bud-break and shoot development induces substantial carbon (C) costs in trees. Drought stress during shoot development can impede C uptake and translocation. This is therefore a channel through which water shortage can lead to restricted shoot expansion and physiological capacity, which in turn may impact annual canopy C uptake. We studied effects of drought and re-hydration on early season shoot development, C uptake and partitioning in five individual 10-year old Picea mariana [black spruce] trees to identify and quantify dynamics of key morphological/physiological processes. Trees were subjected to one of two treatments: (i) well-watered control or (ii) drought and rehydration. We monitored changes in morphological [shoot volume, leaf mass area (LMA)], biochemical [osmolality, non-structural carbohydrates (NSC)] and physiological [rates of respiration (R d ) and light-saturated photosynthesis ( A sat )] processes during shoot development. Further, to study functional compartmentalization and use of new assimilates, we 13 C-pulse labeled shoots at multiple development stages, and measured isotopic signatures of leaf respiration, NSC pools and structural biomass. Shoot water potential dropped to a minimum of −2.5 MPa in shoots on the droughted trees. Development of the photosynthetic apparatus was delayed, as shoots on well-watered trees broke-even 14 days prior to shoots from trees exposed to water deficit. R d decreased with shoot maturation as growth respiration declined, and was lower in shoots exposed to drought. We found that shoot development was delayed by drought, and while rehydration resulted in recovery of A sat to similar levels as shoots on the well-watered trees, shoot volume remained lower. Water deficit during shoot expansion resulted in longer, yet more compact (i.e., with greater LMA) shoots with greater needle osmolality. The 12 C: 13 C isotopic patterns indicated that internal C partitioning and use was dependent on foliar developmental and hydration status. Shoots on drought-stressed trees prioritized allocating newly fixed C to respiration over structural components. In conclusion, temporary water deficit delayed new shoot development and resulted in greater LMA in black spruce. Since evergreen species such as black spruce retain active foliage for multiple years, impacts of early season drought on net primary productivity could be carried forward into subsequent years.

Abstract:Summary The 'assimilates inhibition hypothesis' posits that accumulation of nonstructural carbohydrates (NSCs) in leaves reduces leaf net photosynthetic rate, thus internally regulating photosynthesis. Experimental work provides equivocal support mostly under controlled conditions without identifying a particular NSC as involved in the regulation. We combined 3‐yr in situ leaf gas exchange observations (natural dynamics) in the upper crown of mature Betula pendula simultaneously with measurements of concentrations of sucrose, hexoses (glucose and fructose), and starch, and similar measurements during several one‐day shoot girdling (perturbation dynamics). Leaf water potential and water and nitrogen content were measured to account for their possible contribution to photosynthesis regulation. Leaf photosynthetic capacity (A/Ci) was temporally negatively correlated with NSC accumulation under both natural and perturbation states. For developed leaves, leaf hexose concentration explained A/Ci variation better than environmental variables (temperature history and daylength); the opposite was observed for developing leaves. The weaker correlations between NSCs and A/Ci in developing leaves may reflect their strong internal sink strength for carbohydrates. By contrast, the strong decline in photosynthetic capacity with NSCs accumulation in mature leaves, observed most clearly with hexose, and even more tightly with its constituents, provides support for the role of assimilates in regulating photosynthesis under natural conditions.

Abstract:In Uruguay, there are more than a million hectares forested, and Eucalyptus species predominate. Drought events are becoming more frequent and affecting growth of the stands, especially during the early stages of the crop. Thus, the need arises to analyze the effects of such events on processes that determine growth, such as photosynthesis. The effect of water restriction on photosynthetic parameters was analyzed under greenhouse conditions in 6-months-old rooted cuttings of a clone of E. grandis (GG) and two hybrids with red gums: E. grandis × E. camaldulensis (GC) and E. grandis × E. tereticornis (GT). To define irrigation regimes water potential of substrate (MPa) was assessed using samples with increasing water content (%HR, w|w). Data was used to fit a water retention curve. Field’s capacity pot weight was determined on water drained pots. The control condition (WW) was established at Ψw = -0.15 MPa, while the water restricted (WR) was Ψw = -0.30 MPa. Treatments were maintained by manually water feeding for 16 weeks. Physiological parameters and photosynthetic pigments were recorded 6 and 16 weeks after water withholding to simulate short- and long-term water restriction scenarios, respectively. The following measurements were registered on current year fully expanded sun-exposed leaves: stomatal conductance (gs), maximum net assimilation rate (Amax), PSII quantum efficiency (FPSII), and proportion of chlorophyll and carotenoids in a HPLC elution (%Chl, %Carot). In the initial period, GG maintained Amax and gs between conditions. However, despite exhibiting lower gs in the restricted condition, GC maintained their Amax and GT increased in WR. There was no effect of treatment on photosynthetic pigments and energy partitioning varied little. At the end of the 16-week period, GG showed only decreased stomatal conductance values, and did not change photosynthetic pigment content nor Amax despite water restriction. Hybrids showed increasing values of FPSII, %Chl, and Amax. The PCA for long-term water restriction data showed that pigment content and FPSII may explain the increased assimilation rate of the hybrid clones under WR. Changes in water use efficiency due to higher % Chl in the restricted condition for both hybrids are discussed. There are few reports of increments in chlorophyll in Eucalyptus in drought. Recent information suggests that these types of strategies are adaptative responses that allow plants to cope with stress.

Abstract:Shifts in the age or turnover time of non-structural carbohydrates (NSC) may underlie changes in tree growth under long-term increases in drought stress associated with climate change. But NSC responses to drought are challenging to quantify, due in part to large NSC stores in trees and subsequently long response times of NSC to climate variation. We measured NSC age (Δ14 C) along with a suite of ecophysiological metrics in Pinus edulis trees experiencing either extreme short-term drought (-90% ambient precipitation plot, 2020-2021) or a decade of severe drought (-45% plot, 2010-2021). We tested the hypothesis that carbon starvation - consumption exceeding synthesis and storage - increases the age of sapwood NSC. One year of extreme drought had no impact on NSC pool size or age, despite significant reductions in predawn water potential, photosynthetic rates/capacity, and twig and needle growth. By contrast, long-term drought halved the age of the sapwood NSC pool, coupled with reductions in sapwood starch concentrations (-75%), basal area increment (-39%), and bole respiration rates (-28%). Our results suggest carbon starvation takes time, as tree carbon reserves appear resilient to extreme disturbance in the short term. However, after a decade of drought, trees apparently consumed old stored NSC to support metabolism.
