Abstract:The vacuolar NHX-type Na(+)/H(+) exchangers play a key role in salt tolerance in plants. However, little is known about the Na(+)/H(+) exchangers in the salt-resistant tree, Populus euphratica. In this study, we identified six putative vacuolar Na(+)/H(+) exchanger genes from P. euphratica, designated as PeNHX1-6. Real-time polymerase chain reaction indicated that the PeNHX1/3/6 transcripts were abundant compared with the other three PeNHX genes in the three tissues (roots, stems and leaves) examined. After NaCl treatment for 6 h, the transcript levels of PeNHX1-6 were upregulated in the roots. To address the function of PeNHX1-6, complementation studies were performed with the salt-sensitive yeast mutant strain R100, which lacks activity of the endosomal Na(+)/H(+) antiporter NHX1. The results showed that PeNHX1-6 compensates, at least in part, for the function of yeast NHX1. Moreover, PeNHX3 was targeted to the tonoplast when transiently expressed in onion. Together, these results suggest that PeNHX1-6 function as vacuolar Na(+)/H(+) exchangers and that PeNHX products play an important role in the salt resistance of P. euphratica.
Functional Characterization of PsnNAC036 under Salinity and High Temperature Stresses

Abstract:Plant growth and development are challenged by biotic and abiotic stresses including salinity and heat stresses. For Populus simonii × P. nigra as an important greening and economic tree species in China, increasing soil salinization and global warming have become major environmental challenges. We aim to unravel the molecular mechanisms underlying tree tolerance to salt stress and high temprerature (HT) stress conditions. Transcriptomics revealed that a PsnNAC036 transcription factor (TF) was significantly induced by salt stress in P. simonii × P. nigra. This study focuses on addressing the biological functions of PsnNAC036. The gene was cloned, and its temporal and spatial expression was analyzed under different stresses. PsnNAC036 was significantly upregulated under 150 mM NaCl and 37 °C for 12 h. The result is consistent with the presence of stress responsive cis-elements in the PsnNAC036 promoter. Subcellular localization analysis showed that PsnNAC036 was targeted to the nucleus. Additionally, PsnNAC036 was highly expressed in the leaves and roots. To investigate the core activation region of PsnNAC036 protein and its potential regulatory factors and targets, we conducted trans-activation analysis and the result indicates that the C-terminal region of 191–343 amino acids of the PsnNAC036 was a potent activation domain. Furthermore, overexpression of PsnNAC036 stimulated plant growth and enhanced salinity and HT tolerance. Moreover, 14 stress-related genes upregulated in the transgenic plants under high salt and HT conditions may be potential targets of the PsnNAC036. All the results demonstrate that PsnNAC036 plays an important role in salt and HT stress tolerance.

Transcriptome Analysis of Poplar Under Salt Stress and Over-Expression of Transcription Factor NAC57 Gene Confers Salt Tolerance in Transgenic Arabidopsis

Abstract:NAC domain genes belong to a large plant-specific transcription factor family, which is well-known to be associated with multiple stress responses and plant developmental processes. In this study, we screened differentially expressed genes (DEGs) and detected mRNA abundance of NAC family by RNA-Seq in the poplar leaves under salt stress condition. A total of 276 up-regulated DEGs and 159 down-regulated DEGs were identified to be shared in Populus alba × Populus glandulosa and Populus simonii × Populus nigra. Among 170 NAC members, NAC57 gene was significantly up-regulated in response to salt stress in the two species. Tissue-specific and salt-responsive analyses indicated the expression pattern of NAC57 gene was spatial and temporal in poplar under salt stress. Particle bombardment results showed subcellular localization of NAC57 was not solely nucleus-targeted. Full-length cDNA sequence of the NAC57 gene was cloned from P. alba × P. glandulosa and transformed into Arabidopsis thaliana. Under salt stress, transgenic Arabidopsis overexpressing NAC57 showed higher seed germination rate, root length, and fresh weight than wild type plants. In addition, the transgenic plants displayed higher superoxide dismutase activity and peroxidase activity, and lower malondialdehyde content and relative electrical conductivity than the wild type under salt stress condition. Furthermore, histochemical staining indicated reactive oxygen species accumulation was lower in the transgenic plants than that in the wild type under salt stress. All the results indicated that the NAC57 gene plays an important role in salt stress responses.

Abstract:BACKGROUND:NAC (NAM, ATAF, and CUC) transcription factors play important roles in plant biological processes, including phytohormone homeostasis, plant development, and in responses to various environmental stresses.METHODS:TaNAC29 was introduced into Arabidopsis using the Agrobacterium tumefaciens-mediated floral dipping method. TaNAC29-overexpression plants were subjected to salt and drought stresses for examining gene functions. To investigate tolerant mechanisms involved in the salt and drought responses, expression of related marker genes analyses were conducted, and related physiological indices were also measured. Expressions of genes were analyzed by quantitative real-time polymerase chain reaction (qRT-PCR).RESULTS:A novel NAC transcription factor gene, designated TaNAC29, was isolated from bread wheat (Triticum aestivum). Sequence alignment suggested that TaNAC29 might be located on chromosome 2BS. TaNAC29 was localized to the nucleus in wheat protoplasts, and proved to have transcriptional activation activities in yeast. TaNAC29 was expressed at a higher level in the leaves, and expression levels were much higher in senescent leaves, indicating that TaNAC29 might be involved in the senescence process. TaNAC29 transcripts were increased following treatments with salt, PEG6000, H2O2, and abscisic acid (ABA). To examine TaNAC29 function, transgenic Arabidopsis plants overexpressing TaNAC29 were generated. Germination and root length assays of transgenic plants demonstrated that TaNAC29 overexpression plants had enhanced tolerances to high salinity and dehydration, and exhibited an ABA-hypersensitive response. When grown in the greenhouse, TaNAC29-overexpression plants showed the same tolerance response to salt and drought stresses at both the vegetative and reproductive period, and had delayed bolting and flowering in the reproductive period. Moreover, TaNAC29 overexpression plants accumulated lesser malondialdehyde (MDA), H2O2, while had higher superoxide dismutase (SOD) and catalase (CAT) activities under high salinity and/or dehydration stress.CONCLUSIONS:Our results demonstrate that TaNAC29 plays important roles in the senescence process and response to salt and drought stresses. ABA signal pathway and antioxidant enzyme systems are involved in TaNAC29-mediated stress tolerance mechanisms.
Picea wilsonii Transcription Factor PwNAC38 Promotes Drought and Salt Tolerance in Transgenic Arabidopsis and Apple Calli

Abstract:The mechanism of Picea wilsonii adapting to abiotic stress remains largely unknown. NAC (NAM, ATAF and CUC) transcription factors play significant roles in plant response to adverse environments. In this study, based on our previous RNA-seq, we analyzed the expression patterns of PwNAC38, and revealed its functions in the process of PwNAC38-mediated stress responses. An open-reading frame, encoding PwNAC38 protein with 330 amino acids, was isolated from the cDNA library, a process which can be induced by drought, salt and ABA treatment. Subcellular localization and yeast experiments showed that PwNAC38 was a nuclear-localized transcription factor, and could form homodimers. The full length of PwNAC38 showed transcriptional activity, while the truncated segments, C-PwNAC38 (156–330 aa) and N-PwNAC38 (1–156 aa), did not. The constitutive expression of PwNAC38 (OE lines) in Arabidopsis did not exert influence on the growth of transgenic plants under normal conditions, whereas transgenic seedlings showed higher survival rates, and the seeds had stronger vigor and a higher germination rate under drought and salt stress. The seed germination and root growth of PwNAC38 OE lines were significantly inhibited in the presence of ABA, suggesting the hypersensitivity of PwNAC38 to ABA treatment. Physiological assays showed that the activity of antioxidant enzymes, such as SOD and POD, increased, and that the accumulation of superoxide anion decreased, in OE lines under stress conditions. Moreover, overexpression of PwNAC38 significantly improved drought and salt tolerance in apple calli. A qRT-PCR assay showed that overexpression of PwNAC38 in Arabidopsis promoted the expression of drought or ABA-responsive genes ATHB-7, ANAC019, ERD1, DREB2A, RD29A, ABI5 and NCED3. Taken together, our results revealed that PwNAC38 is positively involved in plants' response to drought and salt stress by enhancing ROS scavenging efficiency, and is partially dependent on the ABA signaling pathway.

Abstract:The NAC (NAM, ATAF1/2 and CUC2) is a large gene family of plant-specific transcription factors that play a pivotal role in various physiological processes and abiotic stresses. Due to the lack of genome-wide characterization, intraspecific and interspecific synteny, and drought-responsive expression pattern of NAC genes in poplar, the functional characterization of drought-related NAC genes have been scarcely reported in Populus species. Here, we identified a total of 170 NAC domain-containing genes in the P. trichocarpa genome, 169 of which were unevenly distributed on its nineteen chromosomes. These NAC genes were phylogenetically divided into twenty subgroups, some of which exhibited a similar pattern of exon–intron architecture. The synteny and Ka/Ks analysis indicated that the expansion of NAC genes in poplar was mainly due to gene duplication events occurring before and after the divergence of Populus and Salix. Ten PdNAC (P. deltoids × P. euramericana cv.'Nanlin895') genes were randomly selected and cloned. Their drought-responsive expression profiles showed a tissue-specific pattern. The transcription factor PdNAC013 was verified to be localized in the nucleus. Our research results provide genomic information for the expansion of NAC genes in the poplar genome, and for further characterizing putative poplar NAC genes associated with water-deficit.

Abstract:Identification of NAC transcription factor members, gene family analysis, and key NAC transcription factor screening in response to salt stress in Populus tomentosa. The NAC transcription factor family is one of the largest families of TFs in plants, and members of NAC gene family play important roles in plant growth and stress response. Recent release of the haplotype‐resolved genome assembly of P. tomentosa provide a platform for NAC protein genome‐wide analysis. A total of 270 NAC genes were identified and a comprehensive overview of the PtoNAC gene family is presented, including gene promoter, structure and conserved motif analyses, chromosome localization and collinearity analysis, protein phylogeny, expression pattern, and interaction analysis. The results indicate that protein length, molecular weight, and theoretical isoelectric points of the NAC TF family vary, while gene structure and motif are relatively conserved. Chromosome mapping analysis showed that the P. tomentosa NAC genes are unevenly distributed on 19 chromosomes. The interchromosomal evolutionary results indicate 12 pairs of tandem and 280 segmental duplications. Segmental duplication is possibly related to amplification of P. tomentosa NAC gene family. Expression patterns of 35 PtoNAC genes from P. tomentosa subgroup were analysed under high salinity, and seven NAC genes were induced by this treatment. Promoter and protein interaction network analyses showed that PtoNAC genes are closely associated with growth, development, and abiotic and biotic stress, especially salt stress. These results provide a meaningful reference for follow‐up studies of the functional characteristics of NAC genes in the mechanism of stress response and their potential roles in development of P. tomentosa.

Abstract:The NAC gene family is of great value for plant stress resistance and development. In this study, five NAC genes with a typical NAM domain were isolated from Salix psammophila, which is a stress-resistant willow endemic to western China. Two hundred sixty-two NAC genes from Salix psammophila, Salix purpurea, and Arabidopsis were used to construct the phylogenetic tree to examine the phylogenetic relationship. Five NAC genes in Salix psammophila were the focus of bioinformatics analysis and conserved structural domain analysis. The SpsNAC005 gene was overexpressed in Populus hopeiensis, and the transgenic lines were subjected to salt and simulated drought stress to analyze their phenotype changes and tolerance to stress. The results showed that transgenic poplar height and leaf area increased by 29.73% and 76.36%, respectively, compared with those of wild-type plants. Under stress treatment, the height growth rates and ground diameter growth rates of the transgenic lines were significantly higher than those of the wild-type, whereas their fresh weight and dry weight were decreased compared to those of the wild-type. The SOD activities, POD activities, and Pro contents of the transgenic plants were significantly increased, and the accumulation of MDA was significantly lower than that in the wild-type, and the transgenic lines showed clear tolerance to salt and drought. The expressions of the SOS1, MPK6, HKT1, and P5CS1 genes were downregulated in the transgenic lines. The expression of the PRODH1 gene was downregulated in the transgenic lines. These results indicate that overexpression of the SpsNAC005 gene in transgenic plants can promote plant growth and development and improve tolerance to salt and drought.

Abstract:NAC (NAM, ATAF1/2, and CUC2) transcription factors regulate plant growth and development and response to various stresses. However, there is still limited insight into the NAC family in Chimonanthus praecox. This study performed a genome-wide characterization of the NAC transcription factor family members in C. praecox. A total of 105 NAC family members were identified from the C. praecox genome. The phylogenetic tree categorized the CpNACs into nine groups and the accuracy of this classification was confirmed by the analysis results of conserved motifs, conserved domain, and gene structure. Cis-acting element analysis revealed that the promoters of CpNACs were abundant in elements responsive to various hormones and stresses, implying the functional diversity and complexity of CpNACs. Furthermore, we investigated the function of the CpNAC30. The expression level of CpNAC30 could be significantly induced by abiotic stress and the CpNAC30 was the highest expressed in mature leaves of C. praecox. Overexpression of CpNAC30 reduced salt stress tolerance of transgenic Arabidopsis. Nevertheless, the drought stress tolerance of transgenic plants was enhanced. This study lays a foundation for further understanding the function of CpNACs genes and provides insights for abiotic stress tolerance breeding of C. praecox and other woody plants.

Ectopic Expression of PsnNAC090 Enhances Salt and Osmotic Tolerance in Transgenic Tobacco

Abstract:The NAC transcription factor family is well known to play vital roles in plant development and stress responses. For this research, a salt-inducible NAC gene, PsnNAC090 (Po-tri.016G076100.1), was successfully isolated from Populus simonii × Populus nigra. PsnNAC090 contains the same motifs at the N-terminal end of the highly conserved NAM structural domain. The promoter region of this gene is rich in phytohormone-related and stress response elements. Transient transformation of the gene in the epidermal cells of both tobacco and onion showed that the protein was targeted to the whole cell including the cell membrane, cytoplasm and nucleus. A yeast two-hybrid assay demonstrated that PsnNAC090 has transcriptional activation activity with the activation structural domain located at 167–256aa. A yeast one-hybrid experiment showed that PsnNAC090 protein can bind to ABA-responsive elements (ABREs). The spatial and temporal expression patterns of PsnNAC090 under salt and osmotic stresses indicated that the gene was tissue-specific, with the highest expression level in the roots of Populus simonii × Populus nigra. We successfully obtained a total of six transgenic tobacco lines overexpressing PsnNAC090. The physiological indicators including peroxidase (POD) activity, superoxide dismutase (SOD) activity, chlorophyll content, proline content, malondialdehyde (MDA) content and hydrogen peroxide (H2O2) content were measured in three transgenic tobacco lines under NaCl and polyethylene glycol (PEG) 6000 stresses. The findings reveal that PsnNAC090 improves salt and osmotic tolerance by enhancing reactive oxygen species (ROS) scavenging and reducing membrane lipid peroxide content in transgenic tobacco. All the results suggest that the PsnNAC090 gene is a potential candidate gene playing an important role in stress response.

Abstract:Abiotic stresses such as drought and high salinity inhibit plant growth and productivity. NAC transcription factors play a variety of important roles in plant development and abiotic stress responses. To date, the transformation of NAC genes into poplar plants has not been reported. In this study, we introduced the CarNAC3 and CarNAC6 salinity and drought tolerant genes from Cicer arietinum (chickpea) into Populus deltoides × Populus euramericana ‘Nanlin895’ (poplar) plants using Agrobacterium tumefaciens-mediated transformation. We verified the integration of the two genes into the poplar genome using polymerase chain reaction (PCR) and their stable expression was confirmed at the transcript level using real-time (RT)-PCR. The growth rates of roots and shoots and the clawed root rate increased in transformed plants grown in agar medium or in soil and increased in response to drought and salt stress conditions. Proline and photoprotectant pigment accumulation and antioxidant enzyme activities increased in response to abiotic stress, but the accumulation of photosynthetic pigments and malondialdehyde (MDA) decreased in transgenic lines relative to wild-type control plants. The CarNAC3 transgene was expressed at higher levels than the CarNAC6 transgene. Our results showed that the CarNAC3 and CarNAC6 genes enhanced the capacity for osmotic adjustment and increased antioxidant enzyme activity and suggest that these genes could play a significant role in improving drought and salt tolerance when expressed in poplar plants.
Overexpression of PeNAC122 gene promotes wood formation and tolerance to osmotic stress in poplars

Abstract:Finding the adequate balance between wood formation and abiotic stress resistance is still an important challenge for industrial woody crops. In this study, PeNAC122, a member of the NAC transcription factor family highly expressed in xylem, was cloned from Populus euphratica. Tissue expression and β‐glucuronidase (GUS) staining showed that PeNAC122 was exclusively expressed in phloem fibre and secondary xylem of stems. Subcellular and yeast transactivation assays confirmed that PeNAC122 protein existed in the nucleus and did not have transcriptional activation and inhibitory activity. Overexpression of PeNAC122 poplar lines exhibited reduced plant height, thickened xylem and accumulated lignin content in stems, and also upregulates the expression of secondary cell wall (SCW) biosynthetic genes. Moreover, overexpression of PeNAC122 lines displayed more tolerance to PEG6000‐induced osmotic stress, with stronger photosynthetic performance, higher antioxidant enzyme activity, less accumulation of reactive oxygen species (ROS) in leaves, and higher expression levels of stress response genes DREB2A, RD29 and NCED3. These results indicate that PeNAC122 plays a crucial role in wood formation and abiotic stress tolerance, which, in addition to potential use in improving wood quality, provides further insight into the role of NAC family transcription factors in balancing wood development and abiotic stress resistance. Comparative study of phenol and thiophenol adsorption on Cu(110).

Abstract:Adsorption of phenol and thiophenol (benzenethiol) on Cu(110) is investigated by a scanning tunneling microscope and electron energy loss spectroscopy. Phenol adsorbs intact and forms a cyclic trimer at 78 K. It is dehydrogenated to yield a phenoxy (C6H5O) group at 300 K. On the other hand, thiophenol is dehydrogenated to a thiophenoxy (C6H5S) group even at 78 K. Both products are bonded via chalcogen atom to the short-bridge site with the phenyl ring oriented nearly parallel to the surface. The C6H5O and C6H5S groups are preferentially assembled into the chains along the [001] and [112] directions, respectively. Dipole-dipole interaction is responsible for the chain growth, while the chain direction is ruled by the steric repulsion between chalcogen atoms and adjacent phenyl ring. This work demonstrates a crucial role of chalcogen atom of phenol species in their overlayer growth on the surface.

Abstract:Populus euphratica is mainly distributed in desert environments with dry and hot climate in summer and cold in winter. Compared with other poplars, P. euphratica is more resistant to salt stress. It is critical to investigate the transcriptome and molecular basis of salt tolerance in order to uncover stress-related genes. In this study, salt-tolerant treatment of P. euphratica resulted in an increase in osmo-regulatory substances and recovery of antioxidant enzymes. To improve the mining efficiency of candidate genes, the analysis combining both the transcriptome WGCNA and the former GWAS results was selected, and a range of key regulatory factors with salt resistance were found. The PeERF1 gene was highly connected in the turquoise modules with significant differences in salt stress traits, and the expression levels were significantly different in each treatment. For further functional verification of PeERF1, we obtained stable overexpression and dominant suppression transgenic lines by transforming into Populus alba × Populusglandulosa. The growth and physiological characteristics of the PeERF1 overexpressed plants were better than that of the wild type under salt stress. Transcriptome analysis of leaves of transgenic lines and WT revealed that highly enriched GO terms in DEGs were associated with stress responses, including abiotic stimuli responses, chemical responses, and oxidative stress responses. The result is helpful for in-depth analysis of the salt tolerance mechanism of poplar. This work provides important genes for poplar breeding with salt tolerance.

Genome-Wide Identification of NAC Gene Family Members of Tree Peony (Paeonia suffruticosa Andrews) and Their Expression under Heat and Waterlogging Stress

Abstract:An important family of transcription factors (TFs) in plants known as NAC (NAM, ATAF1/2, and CUC2) is crucial for the responses of plants to environmental stressors. In this study, we mined the NAC TF family members of tree peony (Paeonia suffruticosa Andrews) from genome-wide data and analyzed their response to heat and waterlogging stresses in conjunction with transcriptome data. Based on tree peony's genomic information, a total of 48 PsNAC genes were discovered. Based on how similar their protein sequences were, these PsNAC genes were divided into 14 branches. While the gene structures and conserved protein motifs of the PsNAC genes within each branch were largely the same, the cis-acting elements in the promoter region varied significantly. Transcriptome data revealed the presence of five PsNAC genes (PsNAC06, PsNAC23, PsNAC38, PsNAC41, PsNAC47) and one PsNAC gene (PsNAC37) in response to heat and waterlogging stresses, respectively. qRT-PCR analysis reconfirmed the response of these five PsNAC genes to heat stress and one PsNAC gene to waterlogging stress. This study lays a foundation for the study of the functions and regulatory mechanisms of NAC TFs in tree peony. Meanwhile, the NAC TFs of tree peony in response to heat and waterlogging stress were excavated, which is of great significance for the selection and breeding of new tree peony varieties with strong heat and waterlogging tolerance.

(1,3;1,4)-β-d-glucan in Gramineae is the key component in response to various environmental signals, and it is the most important functional ingredient in oat grain. The NAC transcription factors are important candidate genes used in genetic engineering to improve plant abiotic stress tolerance. In this study, we introduced Populus euphratica PeNAC1, controlled by its own promoter, into hexaploid cultivated oat and produced six transgenic lines. Compared to the non-transgenic control, the expression of PeNAC1 significantly improved the seed germination rate, seedling survival rate, and leaf chlorophyll content in the transgenic plants under salt stress. These physiological changes increased the spikelet number and grain number per spike in the transgenic oat under salinity conditions and reduced the yield loss per plant. The results indicated that the heterologous expression of PeNAC1 plays an effective role in improving the salt tolerance in transgenic oat. In addition, overexpressing PeNAC1 significantly increased the (1,3;1,4)-β-d-glucan content as well as the expression level of the (1,3;1,4)-β-d-glucan biosynthetic genes AsCslF3, AsCslF6, and AsCslF9 in the transgenic lines under salt stress, which suggested that PeNAC1 regulates the synthesis of (1,3;1,4)-β-d-glucan. Our research should assist in the discovery of the diverse action modes of NAC proteins, while PeNAC1 will be useful for improving the salt tolerance and quality of oat through molecular breeding.

Abstract:Following the experimental program initiated in Martinique and Guadeloupe since 1983, a neonatal screening for sickle cell disease was extended to metropolitan France in 1990. It is presently applied to at risk newborns based on the parents ethnic origin, and associated with the screening for phenylketonuria, hypothyroidism and congenital adrenal hyperplaxia, in three regions. The laboratory method is reliable with a very limited number of false positive or false negative results. The cost effectiveness ratio appears very reasonable. All the children detected so far are correctly managed and have a normal development. This screening should be extended to the rest of France in the coming years.

Abstract:Adverse environmental conditions such as high temperature and drought stress greatly limit the growth and production of crops worldwide. Several NAC (NAM, ATAF1/2, and CUC2) proteins have been documented as important regulators in stress responses, but the molecular mechanisms are largely unknown. Here, a stress-responsive NAC gene, SNAC3 (ONAC003, LOC_Os01g09550), conferring drought and heat tolerance in rice is reported. SNAC3 was ubiquitously expressed and its transcript level was induced by drought, high temperature, salinity stress, and abscisic acid (ABA) treatment. Overexpression (OE) of SNAC3 in rice resulted in enhanced tolerance to high temperature, drought, and oxidative stress caused by methyl viologen (MV), whereas suppression of SNAC3 by RNAi resulted in increased sensitivity to these stresses. The SNAC3-OE transgenic plants exhibited significantly lower levels of H2O2, malondiadehyde (MDA), and relative electrolyte leakage than the wild-type control under heat stress conditions, implying that SNAC3 may confer stress tolerance by modulating reactive oxygen species (ROS) homeostasis. Quantitative PCR experiments showed that the expression of a large number of ROS-scavenging genes was dramatically increased in the SNAC3-OE plants, but significantly decreased in the SNAC3-RNAi transgenic plants. Five ROS-associated genes which were up-regulated in SNAC3-OE plants showed co-expression patterns with SNAC3, and three of the co-expressed ROS-associated enzyme genes were verified to be direct target genes of SNAC3. These results suggest that SNAC3 plays important roles in stress responses, and it is likely to be useful for engineering crops with improved tolerance to heat and drought stress.

Abstract:CarNAC3 contains 285 amino acids and a conserved NAC domain. NAC genes, including NAM, ATAF1, ATAF2, and CUC2, are members of one of the largest transcription factor families in plants. CarNAC3, a member of the NAP group, plays an important role in plant development and responses to abiotic stresses. In this study, CarNAC3 was transformed into hybrid poplar plants (Populus deltoides × P. euramericana ‘Nanlin895’) using Agrobacterium tumefaciens. PCR analysis confirmed integration of the introduced T-DNA into the target genome. Reverse transcription PCR confirmed the transformation, and Southern and northern blotting verified the transgene copy number and gene expression, respectively. Fourteen lines of positive transformants were transplanted into a greenhouse to verify their drought and salt tolerances. Under normal conditions, transgenic poplar plants were shorter than the wild-type, but under drought and salt stresses, they maintained their normal rooting and stem growth rates, while those of the wild-type plants were suppressed. Under stress conditions, CarNAC3 expression caused increases in proline and photosynthetic pigment levels and in antioxidant enzyme activities. Furthermore, the expression of CarNAC3 lowered malondialdehyde concentrations compared with the wild-type control. Overall, our results indicated that the CarNAC3 transgene enhanced drought and salt tolerance in transgenic poplar plants.

Abstract:NAC (NAM, ATAF1/2 and CUC2) proteins are key regulators of various plant stress tolerances. However, knowledge of NAC genes remains largely unknown in Miscanthus. Here, we characterized a novel NAC gene MlNAC10 from M. lutarioriparius than plays a role in abiotic stress tolerance. MlNAC10 encodes a nuclear-localized protein with a C-terminal transactivation domain, and has a specific binding affinity to the NAC recognition sequence (NACRS). Ectopic expression of MlNAC10 in Arabidopsis led to increased sensitivity to abscisic acid (ABA) at early seedling growth stages. In addition, the proline content was significantly increased and the reactive oxygen species (ROS) scavenging capability was significantly enhanced in MlNAC10 overexpression lines under ABA treatment. Moreover, the drought and salt stress tolerance was significantly improved in MlNAC10 overexpression lines. Consistently, the activities of three antioxidant enzymes, namely catalase (CAT), peroxidase (POD), and superoxide dismutase (SOD), were dramatically stimulated in the overexpression lines compared to the wild type (WT). Correspondingly, the accumulation of ROS was dramatically decreased and malondialdehyde (MDA) was accumulated at a much lower level in the transgenic lines. Meanwhile, the expression of six abiotic stress-related genes was dramatically stimulated in the overexpression lines in comparison to the WT. Together, our results demonstrated that MlNAC10 acts as an important regulator of drought and salinity stress tolerance by stimulating antioxidant enzymes and alleviating ROS damage via the ABA signaling pathway.
