Abstract:Visible-light-induced regioselective cascade radical cyclization of α-bromocarbonyls for the synthesis of benzazepine derivatives is described. Abstract:A visible‐light‐mediated reaction of N ‐phenylpropynamides with diethyl 2,2‐dibromomalonate for the synthesis of bromo‐containing azaspirotrienediones is described. This method allows the formation of C‐Br, C‐C and C=O bonds in one pot through the radical addition/ipso‐cyclization/oxidation cascade reaction. A possible radical mechanism involving single electron transfer and energy transfer processes is proposed based on the results of cyclic voltammetry, Stern‐Volmer quenching experiments and control experiments.

Abstract:A visible-light photoredox catalyzed radical cascade cyclization of simple ethers with cyanamides is developed at room temperature. This strategy involves sequential inert Csp3-H/Csp2-H functionalizations through intermolecular addition reaction of oxyalkyl radicals to N-cyano groups followed by radical cyclization of iminyl radicals in situ generated with C-2 aryl rings. This method allows for efficient synthesis of tetracyclic benzo[4,5]imidazo[1,2-c]quinazolines. Importantly, this is the first example of an intermolecular-intramolecular radical cascade cyclization reaction of cyanamides.

Abstract:A novel class of alkyne-tethered amides facilitates an unprecedented photoinduced palladium-catalyzed radical relay formal [5 + 2] reaction. This innovative strategy allows for the rapid construction of diverse fused benzoazepine structures, yielding structurally novel and compelling compounds. With a broad substrate scope and excellent functional group tolerance, the methodology synthesizes biologically active compounds. Notably, the resulting tricyclic benzo[b]azepines offer diversification opportunities through simple transformations. DFT calculations elucidate a seven-membered ring closure mechanism involving the alkenyl radical and Pd(I) rebound alongside a concerted metalation-deprotonation (CMD) process.

This method exhibits a broad substrate scope and high functional group tolerance, giving the corresponding bibenzyl products with generally good yields. Abstract:An efficient visible light-induced radical tandem cyclization method for the synthesis of 4-aryl-1,2-dihydronaphthalenes is reported. The target products were obtained by the radical tandem cyclization of different types of monosubstituted aryl acetylenes with simple bromomalonates as radical precursors. Compared to conventional nucleophilic addition reactions, the method was obtained under mild conditions by irradiation with 7 W blue light at room temperature using fac-Ir(ppy)(3) as a photocatalyst. The reaction conditions are mild and the post-treatment is simple. This free radical triggered tandem cyclisation avails readily available bromomalonate esters and aryl alkynes as the starting materials. The method is very suitable for a wide range of substrates with good applicability to different bromomalonates and different alkynes.

Abstract:A visible light-promoted radical relay of N-allylbromodifluoroacetamide with quinoxalin-2(1H)-ones was developed in which 5-exo-trig cyclization and C-C bond formation were involved. This protocol was performed under mild conditions to facilely offer a variety of hybrid molecules bearing both quinoxalin-2(1H)-one and 3,3-difluoro-γ-lactam motifs. These prepared novel skeletons would expand the accessible chemical space for structurally complex heterocycles with potential biological activities.

Abstract:A novel alkoxycarbonyl-radical-triggered cascade cyclization of 1,7-enynes, with alkyloxalyl chlorides as the ester units, for the synthesis of benzo[j]phenanthridines is described. The reaction conditions exhibit excellent compatibility with a broad range of alkoxycarbonyl radical sources and realize the installation of an ester group in the polycyclic compound. This radical cascade cyclization reaction features excellent functional group tolerance, mild reaction conditions, and good to excellent yields.

Abstract:A diverse array of 6‐aroyl benzimidazo[1,2‐c]quinazolines was successfully synthesized using easily obtainable N‐cyanobenzimidazoles and α‐keto acids, facilitated by photoredox catalysis. A mechanistic investigation revealed that this methodology might involve both photoredox and energy transfer catalytic cycles, generating acyl radicals that subsequently promote radical relay cyclization.

Abstract:We have developed a new method for the synthesis of 2-substituted benzimidazoles via amidinyl radicals generated by visible-light-promoted reduction of N-phenyl amidoxime esters in the presence of an iridium photocatalyst. This is the first report of the use of N-phenyl amidoxime esters as amidinyl radical precursors, and the first use of substituted benzene rings as amidinyl radical acceptors. This method widens the application range of substrates and overcomes the shortcomings of the traditional methods for the synthesis of 2-substituted benzimidazoles, which requires harsh reaction conditions, involves difficult-to-prepare substituted o-phenylenediamine substrates, and produces acidic waste.

Abstract:Polycyclic indole scaffolds are ubiquitous in pharmaceuticals and natural products and in materials science. Here, we present a visible-light-initiated intramolecular aryl migration/desulfonylation/cyclization cascade reaction for the synthesis of tetracyclic indolo[2,1-a]isoquinolin-6(5H)-ones. This protocol not only exhibited a wide substrate scope but also provided a mild route to access a variety of tetracyclic N-fused indoles.

Abstract:A visible-light-induced radical cascade cyclization of aryl isonitriles and cyclobutanone oxime esters for the synthesis of cyclopenta[b]quinoxalines has been accomplished for the first time. The key to the success of this process was the integration of the in situ-formed nitrile radical followed by the cascade radical isonitrile/nitrile insertion-cyclization. The easy introduction of substituents for both substrates and the high functional group tolerance of the reaction make it an efficient strategy to give various quinoxaline derivatives in moderate to good yields.

Abstract:A green and highly efficient strategy for the preparation of bridged spirocyclic compounds via visible-light-induced cyclization of 2-(2-(arylethynyl)benzylidene)malononitrile derivatives with 2,6-di(tert-butyl)-4-methylphenol (BHT) at room temperature was developed. Abstract:A light-induced non-activated olefin cascade radical addition/cyclization reaction was proposed, leading to the successful synthesis of 39 quinazolinone derivatives containing amide moieties, with yields up to 88%. The reaction mechanism was studied by radical strapping test and DFT methods, verifying an oxidative-triggered cascade process promoted by the acylamino radicals.
