Abstract:One hundred sixty patients with idiopathic scoliosis underwent preoperative and postoperative sagittal plane analysis of the thoracic spine, thoracolumbar junction, and lumbar spine. The data suggest that mild to moderate improvements in thoracic hypokyphosis are possible. When crossing the thoracolumbar junction, reversal of rod bend and reversal of hooks on the derotation rod appears to provide the most physiologic sagittal contour. Cotrel-Dubousset instrumentation to the mid and distal lumbar spine can preserve and, at times, enhance lumbar lordosis.

Abstract:A label-free DNA biosensor based on three-dimensional reduced graphene oxide (3D-rGO) and polyaniline (PANI) nanofibers modified glassy carbon electrode (GCE) was successfully developed for supersensitive detection of breast cancer BRCA1. The results demonstrated that 3D-rGO and PANI nanofibers had synergic effects for reducing the charge transfer resistance (R-ct), meaning a huge enhancement in electrochemical activity of 3D-rGO-PANI/GCE. Probe DNA could be immobilized on 3D-rGO-PANI/GCE for special and sensitive recognition of target DNA (1.0x10(-15)-1.0x10(-7) M) with a theoretical LOD of 3.01x10(-16) M (3S/m). Furthermore, this proposed nano-biosensor could directly detect BRCA1 in real blood samples.

Abstract:The development of high-efficient technologies for cancer biomarkers detection has attracted tremendous research effort for its great clinic significance. In this work, we designed a new type of flexible and robust nanohybrid microelectrode by modifying carbon fiber with dual nanoenzyme, i.e., AuPd alloy nanoparticles (AuPd-ANPs) decorated graphene quantum dots (GQDs) assembly, and explored its practical application in electrochemical sensing system for sensitive detection of cancer biomarker hydrogen peroxide (H2O2) in human breast cancer cells and tissue. For the preparation of dual nanoenzyme modified microelectrode, ionic liquid was used as the electrolyte for the effective electrodeposition of GQDs on carbon fiber substrate to form a close-packed assembly under a very negative potential, then the highly dense AuPd-ANPs were uniformly decorated on GQDs assembly by electrodeposition. In virtue of the structural merits and synergistic contribution of dual nanoenzyme in enhancing the electrocatalytic activity to H2O2, the resultant nanohybrid microelectrode exhibited good sensing performances for electrochemical detection of H2O2, including a high sensitivity of 371 μA cm-2 mM-1, a wide linear range from 1.0 μM to 18.44 mM, a low detection limit of 500 nM (a signal-to-noise ratio of 3:1), as well as good selectivity and biocompatibility, which could be used for real-time tracking H2O2 released from different types of human breast cells and in situ sensitive detection of H2O2 in clinical breast cancer tissue.

Abstract:For real-time evaluation of the cell behavior and function under in vivo-like 3D environment, the 3D functionalized scaffolds simultaneously integrate the function of 3D cell culture, and electrochemical sensing is a convincing candidate. Herein, Fe3O4 nanoparticles as the nanozyme (peroxide oxidase mimics) were modified on graphene foam scaffold to construct a 3D integrated platform. The platform displayed a wide linear range of 100 nM to 20 μM and a high sensitivity of 53.2 nA μM−1 toward detection of hydrogen peroxide (H2O2) under the working potential of + 0.6 V (vs. Ag/AgCl). The obtained 3D scaffold also displayed satisfactory selectivity toward the possible interferents that appeared in the cell culture environment. Furthermore, the cells still maintained high cell viability (almost 100%) after their growth and proliferation on the scaffold for 7 days. With the superior performance on cell culture and electrochemical monitoring, the functions on the 3D culture of MCF-7 or HeLa cells and in situ monitoring of cell-released H2O2 was easily achieved on this 3D platform, which show its great application prospects on further cancer-related disease diagnosis or drug screening.Graphical abstractA nanozyme-based three-dimensional graphene scaffold was successfully constructed for cell culture and identification of cancer cells through in situ electrochemical monitoring of the cell-released H2O2.

Abstract:In situ monitoring of H 2 O 2 in cellular microenvironments plays a critical role in the early diagnosis and pretreatment of cancer, but is limited by the lack of efficient and low-cost strategies for the large-scale preparation of real-time biosensors. Herein, a universal strategy for MXene-based composite inks combined with a scalable screen-printing process is validated in large-scale manufacturing of electrochemical biosensors for in situ detection of H 2 O 2 secreted from live cells. Compositing biocompatible carboxymethyl cellulose (CMCS) with excellent conductive MXene, a water-based ink electrode (MXene/CMCS) with tunable viscosity is efficiently printed with desirable printing accuracy. Subsequently, the MXene/CMCS@HRP electrochemical biosensor exhibits stable electrochemical performance through HRP nanoflower modification, showing rapid electron transport and high electrocatalytic capacity, and demonstrating a low limit of detection (0.29 μM) with a wide linear detection range (0.5 μM–3 mM), superior sensitivity (56.45 μA mM −1 cm −2 ), long-term stability and high anti-interference ability. Moreover, this electrochemical biosensor is effectively employed for in situ detection of H 2 O 2 secreted from HeLa cells, revealing good biocompatibility and outstanding biosensing capability. This proposed strategy not only extends the possibility of low-cost biomedical devices, but also provides a promising approach for early diagnosis and treatment of cancer.

Abstract:In this work, we develop a new type of mesoporous 2D N, B, and P codoped carbon network (NBP-CNW) arranged into high-order 3D nanotube arrays (NTAs), which are wrapped onto a flexible carbon fiber microelectrode, and this microelectrode is employed as a high-performance carbon-based nanocatalyst for electrochemical biosensing. The NBP-CNW-NTAs synthesized by a facile, controllable, ecofriendly and sustainable template strategy using ionic liquids as precursors possess a high structural stability, large surface area, abundant active sites, and effective charge transport pathways, which dramatically improve their electrocatalytic activity and durability in the redox reaction of cancer biomarker H 2 O 2 . Benefiting from these unique structural merits, superb electrochemical activity and good biocompatibility, the NBP-CNW-NTAs-modified microelectrode demonstrates excellent sensing performance toward H 2 O 2 and is embedded in a homemade microfluidic electrochemical biosensor chip for the real-time tracking of H 2 O 2 secreted from different live cancer cells with or without radiotherapy treatment, which provides a new strategy for distinguishing the types of cancer cells and evaluating the radiotherapeutic efficacy of cancer cells. Furthermore, the functional microelectrode is integrated into an implantable probe for the in situ detection of surgically resected human specimens to distinguish cancer tissues from normal tissues. These will be of vital significance for cancer diagnoses and therapy in clinical practice.

Abstract:Let $n$ be a positive integer. Denote by $\mathrm{PG}(n,q)$ the $n$-dimensional projective space over the finite field $\mathbb{F}_q$ of order $q$. A blocking set in $\mathrm{PG}(n,q)$ is a set of points that has non-empty intersection with every hyperplane of $\mathrm{PG}(n,q)$. A blocking set is called minimal if none of its proper subsets are blocking sets. In this note we prove that if $\mathrm{PG}(n_i,q)$ contains a minimal blocking set of size $k_i$ for $i\in\{1,2\}$, then $\mathrm{PG}(n_1+n_2+1,q)$ contains a minimal blocking set of size $k_1+k_2-1$. This result is proved by a result on groups with maximal irredundant covers.

Abstract:We report the design and fabrication of a new type of nanohybrid microelectrode based on a hierarchical nanostructured Au/MnO2/graphene-modified carbon fiber (CF) via in situ electrochemical synthesis, which leads to better structural integration of different building blocks into the CF microelectrode. Our finding demonstrates that wrapping CF with graphene nanosheets has dramatically increased the surface area and electrical conductivity of the CF microelectrode. The subsequent template-free electrodeposition of MnO2 on graphene-wrapped CF gives rise to a porous nanonest architecture built up from twisted and intersectant MnO2 nanowires, which serves as an ideal substrate for the direct growth of Au nanoparticles. Owing to the structural merit and synergy effect between different components, the hierarchical nanostructured noble metal/metal oxide/graphene-coated CF demonstrates dramatically enhanced electrocatalytic activity. When used for nonenzymatic H2O2 sensing, the resultant modified microelectrode exhibits acceptable sensitivity, reproducibility, stability, and selectivity, which enable it to be used for real-time tracking H2O2 secretion in human cervical cancer cells. Abstract:The rational design and construction of high-performance flexible electrochemical sensors based on hierarchical nanostructure functionalized microelectrode systems are of vital importance for sensitive in situ and real-time detection of biomolecules released from living cells. Herein, we report a novel and facile strategy to synthesize a new kind of high-performance microelectrode functionalized by dual nanozyme composed of rime-like Cu 2 (OH) 3 NO 3 wrapped ZnO nanorods assembly [Cu 2 (OH) 3 NO 3 @ZnO], and explore its practical application in electrochemical detection of hydrogen peroxide (H 2 O 2 ) released from living cells. Benefiting from the merits of the unique hierarchical nanohybrid structure and high catalytic activities, the resultant Cu 2 (OH) 3 NO 3 @ZnO-modified AGF microelectrode shows remarkable electrochemical sensing performance towards H 2 O 2 with a low detection limit of 1 μM and a high sensitivity of 272 μA cm −2 mM −1 , as well as good anti-interference capability, long-term stability, and reproducibility. These properties enabled the proposed microelectrode-based electrochemical platform to be applied for in situ amperometric tracking of H 2 O 2 released from different types of human colon cells, thus demonstrating its great prospect as a sensitive cancer cell detection probe for the early diagnosis and management of various cancer diseases.

The C-MOF NSs {i.e., Cu-HHTP (HHTP = 2,3,6,7,10,11-hexahydroxytriphenylene)-NSs} provide large surface area and abundant active open metal sites (Cu–O4), which could improve the catalytic activity of Cu-HHTP-NSs towards H2O2. Moreover, abundant exposed O atoms also serve as anchor sites for the deposition of high-density ultrafine Au-NPs (∼3 nm) without agglomeration. Owing to the synergistic contributions of high catalytic activity of Cu-HHTP-NSs and Au-NPs as well as their unique structural and electrical properties, the as-prepared nanohybrid modified electrode exhibits good sensing performances to H2O2 with an extremely low detection limit of 5.6 nM (3σ rules) and a high sensitivity of 188.1 μA cm−2 mM−1. Abstract:Endothelial dysfunction is a prominent occurrence in coronary arteries after myocardial ischemia and reperfusion. However, this has not been studied in coronary veins. Endothelium-dependent vasorelaxation was studied in cardiac venous rings obtained from dogs subjected to 60 min of coronary artery occlusion followed by 270 min of reperfusion, as well as from dogs subjected to sham ischemia-reperfusion. Myocardial ischemia resulted in a 96 +/- 2% decrease in coronary flow to the ischemic area 60 min after occlusion of the left anterior descending (LAD) coronary artery, which led to a significant degree of cardiac necrosis (i.e., 32.9 +/- 3.9% of area at risk). Cardiac venous rings isolated from sham ischemia-reperfusion dogs relaxed 68 +/- 3% to 200 microM ADP, 65 +/- 3% to 2 microM A23187, and 76 +/- 4% to 200 microM sodium nitrite (NaNO2). Corresponding values for cardiac venous rings isolated from ischemic-reperfused dogs were 32 +/- 3% for 200 microM ADP (P less than 0.01 vs. sham), 31 +/- 3% for 2 microM A23187 (P less than 0.01 vs. sham), and 74 +/- 3% for 200 microM NaNO2 (NS from sham). In rings from control dogs, vasorelaxation to ADP and A23187 was markedly inhibited by 4 mM NG-methyl-L-arginine (L-NMMA) and 10 methylene blue and restored after NG-nitro-L-arginine by 3 mM L-arginine. These results demonstrate that a significant degree of endothelial dysfunction occurred in cardiac venous rings after ischemia and reperfusion.(ABSTRACT TRUNCATED AT 250 WORDS)
Enhanced Nonenzymatic Sensing of Hydrogen Peroxide Released from Living Cells Based on Graphene Aerogel/Platinum Nanoparticle

Abstract:The high levels of H2O2 are closely associated with cancer and progressive neurodegenerative diseases. Therefore, the accurate and sensitive detection of hydrogen peroxide is of great importance. In this study, we report a facile hydrothermal fabrication of graphene aerogel/platinum nanoparticle (GA/Pt NPs) nanocomposites with three-dimensional (3D) interconnected pores. The GA/Pt NPs product were first characterized by scanning electron microscopy (SEM), transmission electron microscopy (TEM), X-ray diffraction (XRD), and X-ray photoelectron spectroscopy (XPS). Cyclic voltammetry (CV) measurements reveal that GA/Pt NPs can directly catalyze the reduction of H2O2 and display enhanced current responses. The biosensor exhibits remarkable sensitivity (138.23 mu A mM(-1) cm(-2)), an approximate linear detection range of H2O2 concentration (1 mu mol . L-1 to 2785 mu mol . L-1), and a detection limit of 0.33 mu mol . L-1. Moreover, good anti-interference property, reproducibility, and long-term stability of the enzymeless sensor were achieved. These results indicate that GA/Pt NPs is a good candidate for H2O2 detection. Due to these remarkable analytical advantages, the as-made sensor was applied to determine the H2O2 released from PC12 cells with satisfactory results. These results demonstrate that this new nanocomposite with the high surface area and electrocatalytic activity is a promising candidate for use as an enhanced electrochemical sensing platform in the design of nonenzymatic biosensors.
3D heteroatom-doped graphene-wrapped flexible carbon fiber microsensor for real-time hydrogen peroxide detection in live cancer cells

Abstract:Balancing electrostatic repulsive force and bonding interactions are the key principles for constructing 3D graphene architecture on single carbon fiber (CF). Herein, we present a simple and facile method to modify 3D porous N, B co-doping graphene on single CF (). In the process, the Ni nanoparticles were deposited on CF and used as a reductant for reducing graphene oxide (GO), promising graphene-wrapped CF at low temperature. Ionic liquids were introduced to prevent re-aggregation of graphene sheets during the reduction process of GO, but also acted as heteroatom sources for doping graphene, which provides high electrical conductivity, numerous doping sites and edges for H 2 O 2 electrocatalysis reduction. It is found that the flexible microelectrodes show a broad linear range from 0.5 μM to 4.25 mM and a low detection limit of 100 nM for H 2 O 2 detection. When applied to track the amount of H 2 O 2 generated from real samples, the sensor exhibits desirable performance in distinguishing cancer cells from normal cells and further assessing the therapeutic effect of antitumor drugs on live cancer cells. Therefore, this study may offer an attractive method for synthesizing advanced graphene-based electrodes with great promise for biosensors.

Abstract:Accurate, in situ and long-term electrically monitoring of cell development plays an important role in cell study, which brings in challenges in terms of biocompatibility, processability, and sensing capability of electrochemical sensors. Based on biocompatible conductive polyaniline (PAni) hydrogels, we constructed a flexible sensor with flexible carbon cloth for electrical analysis of living cells. The carbon fiber substrate modified with conductive PAni hydrogels was selected as the electrode to promote the current collection of the sensor. The three dimensional nanostructured mesoporous matrix of PAni hydrogels is favorable for in situ generation of catalytic Pt nanoparticles and cell growth. With these hierarchically nanostructured features, the hydrogel electrochemical sensor was endowed with high sensitivity and selectivity in the detection of H2O2 (with a low detection limit of 1.6 μM in 0.01 M PBS and a wide linear range from 10 μM to 10 mM), and good biocompatibility for cell growth as long as 5 days. The accurate detection of H2O2 released from cells enabled us to differentiate the physiological states of cells and imitate the different stimuli-responsive behavior, which can provide real-time information on cell biological events. With outstanding biocompatibility, operability and repeatability, this strategy can be expanded to the fields of other biosensor fabrication and cell-related biomarker monitoring, which exhibits a broad application potential in bioanalysis catering to new generation sensors.

Abstract:Breast cancer detection at an early stage significantly increases the chances of successful treatment and survival. This study presents an electrochemical biosensor for detecting breast cancer cells, utilizing silver nanoclusters encapsulated by hemoglobin and Cu (II)-porphyrin-metal organic framework (BioMOF) in a graphene-incorporated nanohybrid probe. This Hb-AgNCs@MOF-G probe demonstrates high electrochemical activity, superior dispersity, porosity, and a large surface area for effective functionalization. Using a green ultrasonic-assisted stirring method, we fabricate ultra-small 5 nm particles that readily immobilize on a glassy carbon electrode, generating a detection signal when interacting with ferricyanide/ferrocyanide redox probes. The resulting immunosensor detects as few as 2 cells/mL using Electrochemical Impedance Spectroscopy (EIS) "signal on" and 16 cells/mL via Square Wave Voltammetry (SWV) "signal off", within a broad range of cell concentrations (10 2 –5 × 10 4 cells/mL). Our designed sensor shows improved selectivity (5- to 16-fold) and robust detection in human blood with a recovery efficiency between 94.8–106% (EIS method) and 95.4–111% (SWV method). This sensor could streamline early cancer diagnosis and monitor patient treatment without requiring labelling or signal amplification. As a pioneering endeavor, we've utilized integrated porous MOFs with Hb-encapsulated silver nanoclusters in cancer detection, where these components collectively enhance the overall functionality.

Abstract:The exploration into nanomaterial-based nonenzymatic biosensors with superb performance in terms of good sensitivity and anti-interference ability in disease marker monitoring has always attained undoubted priority in sensing systems. In this work, we report the design and synthesis of a highly active nanocatalyst, i.e., palladium and platinum nanoparticles (Pt&Pd-NPs) decorated ultrathin nanoporous gold (NPG) film, which is modified on a homemade graphene paper (GP) to develop a high-performance freestanding and flexible nanohybrid electrode. Owing to the structural characteristics the robust GP electrode substrate, and high electrochemically catalytic activities and durability of the permeable NPG support and ultrafine and high-density Pt&Pd-NPs on it, the resultant Pt&Pd-NPs-NPG/GP electrode exhibits excellent sensing performance of low detection limitation, high sensitivity and anti-interference capability, good reproducibility and long-term stability for the detection of small molecular biomarkers hydrogen peroxide (H2O2) and glucose (Glu), and has been applied to the monitoring of H2O2 in different types of live cells and Glu in body fluids such as urine and fingertip blood, which is of great significance for the clinical diagnosis and prognosis in point-of-care testing.

Abstract:Three-dimensional (3D) cell culture can better reproduce the in vivo cell environment and has been extensively used in fields such as tissue engineering, drug screening, and pathological research. Despite the tremendous advancement of 3D cultures, an analysis technique that could collect real-time information of the biological processes therein is sorely lacking. Electrochemical sensing with fast response and high sensitivity has played a vital role in real-time monitoring of living cells, but most current sensors are based on planar electrodes and fail to perfectly match the 3D cell culture matrix. Herein, we developed a robust 3D electrochemical sensor based on functionalized graphene foam (GF), which could be integrated with hydrogels for the 3D culture and in situ monitoring of cells for the first time. Specifically, platinum nanoparticles (Pt NPs) electrodeposited on GF (GF/Pt NPs) conferred the prominent electrochemical sensing performance, and the anti-fouling coating of poly(3,4-ethylenedioxythiophene) (PEDOT) endowed the GF/Pt NPs electrode with greatly improved stability. As a proof of concept, collagen hydrogel with microglia seeded in was filled into the interspace of the 3D GF/Pt NPs/PEDOT sensor to establish an integrated platform, which allowed the successful real-time monitoring of reactive oxygen species released from microglia in the collagen matrix.
