Abstract:For ethanol production from cellulosic biomass, an effective pretreatment step is required to reduce biomass recalcitrance by altering its chemical and structural features. Sodium hydroxide (NaOH) and lime (CaO) are two alkali reagents which have been extensively studied in chemical pretreatment of lignocellulosic biomass. However, high temperatures and pressures are routinely applied to effectively improve the digestibility of lignocellulose, which lead to the high cost of pretreatment. In previous studies, distinct properties of NaOH and CaO make it possible to manipulate these two alkali reagents to let them work together in lignocellulose pretreatment to reduce temperature, chemical loading, and residence time requirements. Corn Stover is considered as a potential feedstock for sugar production due to its great amount and easy access in the United States. In this research, NaOH and CaO are used together to pretreat corn stover at 50 oC. Three pretreatment parameters are studied: residence time, NaOH concentration, and CaO loading. The pretreatments were evaluated based on the sugar productions in the subsequent enzymatic hydrolysis. At recommended pretreatment conditions (9h, 0.75% NaOH, 0.05 g CaO g-1 raw biomass), the total reducing sugar yields was 544.0 mg g-1 raw biomass, at an overall carbohydrate conversion of 77.0%. However, compared with pretreatment at recommended conditions at room temperature, pretreatment at 50 oC may result in more energy and chemical input.

Abstract:Corn stover pretreatment using a combination of sodium hydroxide (NaOH) and calcium oxide (CaO) at room temperature was investigated for improved cost-effectiveness of biomass-to-sugar conversion in this study. The effects of NaOH loading, CaO loading, and residence time on enzymatic hydrolysis were studied, and the total reducing sugar yield in the enzymatic hydrolysis was used to evaluate the pretreatment conditions, Compared with NaOH pretreatment, pretreatment with the combination of NaOH and CaO resulted in a similar sugar production rate but at a potentially lower cost. The addition of CaO not only increased the alkalinity, which favored biomass digestibility improvement, but also contributed to better biomass preservation in the pretreatment. On the basis of the sugar production rate and cost-benefit considerations, the two recommended pretreatment conditions were 3 h, 0.05 g NaOH g(-1) raw biomass, 0.1 g CaO g(-1) raw biomass and 6 h, 0.05 g NaOH g-1 raw biomass, 0.05 g CaO g-1 raw biomass, at which the total reducing sugar yields were 529.0 and 538.9 mg g(-1) raw biomass, respectively, with overall carbohydrate conversions of 75.6 and 77.0%, respectively.

Abstract:A two-step sequential dilute alkali/acid pretreatment process was developed to improve the breakdown of the lignocellulosic structure of corn cob. Uniform design was used to optimize the hydrolysis process and multiple regression models were developed. Alkali hydrolysis of corn cob at optimum conditions of 1.81% (w/v) NaOH concentration and 5 (w/v) solid:liquid ratio for 90 min resulted in 82.03% lignin removal. The biomass on further dilute acid pretreatment at the optimized conditions of 6% (w/v) H 2 SO 4 and 5 w/v solid:liquid ratio for 15 min resulted in 74% xylose yield. The xylose-rich acid hydrolysate was used as a substrate to produce xylitol by Debaryomyces nepalensis NCYC 3413. It resulted in a xylitol yield of 0.467 g/g of xylose and a xylitol titer of 21.15 g/L. The changes in the physical and chemical structure of the corn cob due to the sequential alkali/acid pretreatment were analyzed by SEM, XRD, FTIR, and TGA.

Abstract:The present study developed three different microwave-assisted alkalic salt pretreatments (Na3PO4·12H2O, Na2CO3 and CH3COONa) to improve sugar yield from corn cob wastes. Process inputs included pretreatment time, alkalic salt concentration and microwave power intensity with the sugar yield as the corresponding output. All three pretreatment models gave high coefficient of determination values (R2> 0.85). The Na3PO4·12H2O treatment (11.55% Na3PO4.12H2O at 700 W for 6 min) led to the maximum sugar yield (0.76 ± 0.01 g/g). All three pretreatment methods displayed key structural changes in the corn cob structure after pretreatment. This study gave an 11% higher sugar yield in contrast to a previous microwave-assisted pretreatment report on corn cobs. The developed pretreatment strategy was effective for improving the sugar recovery from lignocellulosic wastes and can be channelled towards microbial production of fuels and high value commodities.

Abstract:Enzymatic hydrolysis of maize straw polysaccharides was investigated for the production of reducing sugars. After maize straw was pretreated with 2% sodium hydroxide at 80°C for 1h to delignify, the cellulosic residues were hydrolyzed by cellulase from Trichoderma reesei ZU-02 and the hydrolysis yield at 48h was 65.9%. A certain amount of cellobiose was accumulated in the hydrolysate due to low cellobiase activity in T. reesei cellulase. Supplementing cellobiase from Aspergillus niger ZU-07 greatly reduced the inhibitory effect caused by cellobiose, and the hydrolysis yield at 48h was improved to 81.2% with cellobiase activity enhanced to 10CBU/g substrate. The addition of 5g/l Tween-80 improved the enzymatic hydrolysis by increasing the hydrolysis yield of 7.5%. Fed-batch hydrolysis was started with a batch hydrolysis containing 80g/l substrate, with cellulosic residues added at 6 and 12h to get a final substrate concentration of 110g/l. After 72h of hydrolysis, the reducing sugars concentration reached 89.5g/l with a hydrolysis yield of 83.3%. The hydrolysate from fed-batch hydrolysis contained 56.7g/l glucose, 23.6g/l xylose, and 5.7g/l arabinose, which is suitable for subsequent fermentation process.

Abstract:Pretreatment is a key process restricting the development of biorefinery. This work developed a pretreatment process based on an ethanolamine/acetamide alkaline deep eutectic solvent (ADES). Under microwave assistance, pure ADES pretreatment at 100 degrees C for 10 min achieved 95.9 % delignification and 95.2 % hemicellulose removal of bamboo shoot shells (BSS). Further, when 75 % water was added to pure DES to prepare hydrated DES (75 %-HADES), impressive delignification (93.2 %), hemicellulose removal (92.2 %) and cellulose recovery (94.8 %) were still achieved. The cellulose digestibility of the 75 %-HADES pretreated solid residue was significantly increased from 12.2 % (the control) to 91.2 %. Meanwhile, the structural features of hemicellulose and lignin macromolecules fractionated by 75 %-HADES pretreatment were well preserved, offering opportunities for downstream utilization. Overall, this work proposes an effective pretreatment strategy with the potential to enable the utilization of all major components of bamboo shoot shells.

Abstract:The present study optimized two microwave-assisted alkaline (Na3PO4.12H2O and NaOH) pretreatment methods for the enhancement of sugar recovery from different lignocellulosic substrates (bamboo and corn cobs) with varying structural compositions. Subsequently, the chemical recyclability of the optimized alkaline pretreatments were evaluated on corn cobs. Furthermore, kinetics of microbial cell growth and bioethanol production in simultaneous saccharification and fermentation (SSF) processes were assessed. The optimized Na3PO4.12H2O pretreatment conditions resulted in the maximum reducing sugar yield (0.512 g/g) from corn cobs. Interestingly, the recyclability studies demonstrated that the NaOH-based black liquor resulted in a slightly higher average sugar yield (0.37 g/g) compared to the Na3PO4.12H2O (0.31 g/g) over three pretreatment cycles. Kinetics showed a maximum specific growth rate and maximum potential bioethanol concentration of 0.175 h−1 and 16.928 g/L respectively for the optimized NaOH pretreated corn cobs. The findings from this study provide significant knowledge enhancements pertaining to the use of gold standard, well-known NaOH methods, in addition to the emerging Na3PO4.12H2O alkaline catalyst for the pretreatment of lignocellulosic substrates with varying structural compositions. Moreover, recyclability of Na3PO4.12H2O and NaOH black liquor demonstrated the potential to reduce costs associated with the application of chemicals and fresh water in pretreatment systems. Kinetics of cell growth and bioethanol production elucidates noteworthy indices such as the maximum specific growth rate of the cells, lag time and maximum bioethanol production for lignocellulosic process development and scale-up procedures.

Abstract:Na2CO3 was employed as an efficient yet cheap alkaline catalyst for the pretreatment of corn stover. To systematically obtain an optimal condition, the effects of critical pretreatment parameters including Na2CO3 concentration (2-6%), temperature (120-160 °C), and reaction time (10-30 min) on glucose yield were evaluated in lab-scale using response surface methodology. The best conditions were found to be Na2CO3 of 4.1%, temperature of 142.6 °C, and reaction time of 18.0 min, under which glucose yield reached to 267.5 g/kg biomass. Physical properties, based on scanning electron microscopy (SEM) imagery, surface area, pore volume and size, and crystallinity of pretreated corn stover, were examined. The Na2CO3 pretreatment apparently damaged the surface and altered structural features of corn stover, which resulted in the enhancement of enzymatic of hydrolysis. These results evidently support that Na2CO3 is indeed a robust and feasible catalyst for pretreating lignocellulosic biomass.

Abstract:Corncob (CB) is a valuable agricultural waste in high abundance. It is mainly composed of cellulose, hemicellulose and lignin that link in a complex interlocking matrix. The key to its utilization depends on separating cellulose from lignin and hemicellulose using an environment-friendly and effective pretreatment method to improve glucose yield. The study aimed to investigate the effect of sequential pretreatment of CB with ultrasound (US) in conjunction with ternary deep eutectic solvent (TDES). The US, type and molar ratio of TDES, temperature, and time were optimized. The results showed that under the optimum conditions, US (40 +60 kHz, 60 min) + TDES (ChCl-Gly-LA, 1:1:2, 120 degrees C, 3 h), the removal of lignin and hemicellulose were 87.54 % and 67.00 %, respectively, and the recovery of cellulose was 82.18 %. After 72 h of enzymatic hydrolysis, the yield of glucose and xylose were 76.94 % and 41.26 %, respectively. Comparatively, pretreated CB yielded 61.58 % glucose and 26.90 % xylose higher than the untreated. Scanning electron microscopy revealed that the surface of the CB became cracked and rugged. X-ray diffraction showed that the CrI increased significantly from 36 % to 59 %. Fourier transform mid-infrared spectroscopy indicated significant changes in the characteristic functional groups of lignin and hemicellulose. Increases in Tonset, Tendset, Tp, Vmax and decreases in Char at 600 degrees C all corresponded to increases in thermal stability. These results further confirmed that TDES combined with ultrasonic pretreatment effectively removed lignin and hemicellulose. In conclusion, this approach could greenly and efficiently pretreat CB as a new method for biomass pretreatment.
Statistical optimization of aqueous ammonia pretreatment and enzymatic hydrolysis of corn cob powder for enhancing sugars production

Abstract:Response surface methodology (RSM) was applied to systematic optimization of aqueous ammonia pretreatment and enzymatic hydrolysis of corn cob powder (CCP) using self production cellulases from Aspergillus niger HQ-1. Significant variables including aqueous ammonia concentration, temperature and time during pretreatment were screened and optimized. Results showed that CCP with 0.3 mm of particle size should be pretreated using 11.6 wt% aqueous ammonia solution with 20.0% (w/v) of solid loading at 71.3 °C for 8.8 h. The optimized pretreatment conditions could result in 97.1% of cellulose recovery, 66.6% of hemicellulose recovery and 75.7% of lignin removal, respectively. Significant variables including enzyme loading, reaction time and biomass loading during enzymatic hydrolysis were also screened and optimized. Maximal reducing sugar (822.31 mg/g dry substrate, mg/gds) and xylose (316.72 mg/gds) could be obtained while the pretreated CCP was hydrolyzed at 50.0 °C, pH 4.8 for 36.6 h with 10.9 FPU/gds (FPU/g dry substrate) of enzyme loading, 6.95% (w/v) of biomass loading and 0.5% (w/v) of Tween-80. The optimized enzymatic hydrolysis conditions could result in 92.1% of holocellulose conversion and 79.3% of hemicellulose conversion, respectively. In addition, results also indicated that the crude cellulases solution containing xylanases from A. niger HQ-1 could be applied to enzymatic hydrolysis of the pretreated CCP.

Statistical Optimization of Tween-80-Assisted Potassium Hydroxide Pretreatment and Enzymatic Hydrolysis for Enhancing Sugar Yields from Corn Cob

Abstract:With the addition of Tween 80, potassium hydroxide pretreatment and enzymatic hydrolysis were statistically optimized to maximize sugar yields from corn cob (CC). The results indicated that the sugar yields from CC could be influenced significantly by the potassium hydroxide concentration, temperature and time during pretreatment. The optimized pretreatment conditions were as follows: potassium hydroxide, 46 g·L−1; Tween 80, 3.0 g·L−1; solid dose, 200 g·L−1; temperature, 78 °C; and time, 50 min. After optimization, the lignin reduction and recoveries of cellulose and hemicellulose were 89.7%, 97.8% and 68.0%, respectively. In addition, sugar production could also be influenced by the biomass loading, enzyme loading and reaction time. A maximal glucose production (518.48 mg·gds−1, milligrams per gram of dry substrate) and xylose production (351.14 mg·gds−1), 97.2% cellulose conversion and 82.9% hemicellulose conversion from CC could be obtained when the biomass loading was 195 g·L−1 and the enzyme loading was 8.9 FPU·gds−1 (filter paper activity units per gram of dry substrate) and when the Tween 80 concentration was 3.0 g·L−1 at 50 °C for 30.4 h during hydrolysis. This is the first systematic study of combined Tween 80 pretreatment of CC by potassium hydroxide and hydrolysis of CC by cellulase preparation to increase sugar production.

Abstract:In this work, the effectiveness of hydrogen peroxide-assisted sodium carbonate (HSC) pretreatment of corn stover to produce fermentable sugars was investigated. It was found that the addition of H2O2 (a green oxidative agent) in Na2CO3 pretreatment could enhance the lignin removal and increase the hydrophilicity of the residual lignin, leading to the improved enzymatic digestibility and the yield of fermentable sugars. Also, results showed that lignin removal (in the range of 12–65%) was positively and linearly correlated with final total sugar yields. Post PFI refining could further improve the enzyme accessibility by increasing porosity of substrates. After HSC pretreatment (40 wt % Na2CO3 and 15 wt % H2O2, 120 °C for 60 min) and enzymatic hydrolysis, the final total sugar yield could achieve 79%, which was about 10% higher in comparison with the conventional Na2CO3 pretreatment under the same conditions. HSC pretreatment will have less environmental impact and less barriers for large scale production.

Abstract:Sugarcane bagasse (SCB), a widely available lignocellulosic by-product, could be potentially used in macrofungal bioconversion for high-value bioactive compounds. For macrofungus with poor lignin degradation capacity, its recalcitrant lignin structure blocked the release of fermentable sugars for cell growth. Herein, the main objective was to construct an effective delignification pretreatment strategy for SCB biotransformation and synthesis of bioactive substances by Agaricus sinodeliciosus var. Chaidam. Different pretreatments including chemical (alkali and Fenton oxidation) and ultrasound methods were used to intensify the delignification process. Physico-chemical characteristics of native and pretreated SCB were examined via XRD, XPS, FT-IR and C-13 ssNMR to characterize crystallinity index, surface lignin and chemical structure. Lignocellulose-degrading enzymes were also used to assess the ability of the macrofungus to degrade SCB. By comparing morphological structure, chemical composition and fermentation characteristics, a suitable multiple sequential pretreatment method was finally identified. Our findings revealed that Fenton-ultrasound-alkali pre-treatment has an optimum delignification effect on SCB with never-uncovered excellent ability to detoxify harmful substances produced by Fenton oxidation. Under this pretreatment condition, the cellulose content was increased by 106.45%, whereas hemicelluloses and lignin were decreased by 48.34% and 74.15%, respectively. The macrofungal growth, intracellular polysaccharides, total phenols and terpenoids formation were 2.4, 1.46, 1.36 and 1.66 times higher than those of the untreated, respectively. Fermentation kinetic results showed outstanding fit and prediction of macrofungal growth and enzymatic activity secretion on above pretreated SCB. This study shed light on developing a novel pretreatment strategy for low-cost SCB lignocellulose transformation into high-value products.
Effect of steam-assisted alkaline pretreatment plus enzymolysis on converting corn stalk into reducing sugar

Abstract:In order to improve corn stalk conversion into total reducing sugar effectively, steam-assisted alkaline pretreatment and enzymatic hydrolysis was studied. This result showed that cellulose and hemicelluloses contents in corn stalk were decreased to 7.01% and 4.41% (P < 0.05), and total reducing sugar yield was increased to 335.09 mg/g biomass (P < 0.05), when corn stalk was pretreated with steam and alkali (0.8% NaOH + 1.2% CaO, W/V) for 1.0 h at a ratio of alkaline solution to corn stalk of 5:1 (V/W), followed by enzymatic hydrolysis at 22.5 FPU/g biomass cellulase for 48 h. The synthetical enzymatic hydrolysis conditions of pretreated corn stalk were pH 5.0, 50 °C, 60 filter paper unit (FPU)/g biomass and 48 h reaction time, in which total reducing sugar yield was 358.97 mg/g biomass (P < 0.05). The further response-surface methodology results showed that the total reducing sugar reached 493.74 mg/g biomass under the conditions of 52.39 FPU/g biomass cellulase, 6599.99 U/g biomass β-glucanase and 40 h reaction time. Scanning electron microscopy (SEM), X-ray diffraction (XRD) and pore structure analyses indicated that steam-assisted alkaline pretreatment plus enzymatic hydrolysis were effective in breaking the crystal structure of corn stalk for increasing total reducing sugar yield.

Abstract:To enhance fermentable sugar production, sodium hydroxide pretreatment and enzymatic hydrolysis of corn stover powder (CSP) were optimized using response surface methodology (RSM), respectively. It was found that pretreatment parameters including temperature, time, and sodium hydroxide concentration and hydrolysis parameters including reaction time, biomass loading, and enzyme loading had significant effects on sugar yields. After optimization, CSP with particle size 0.3 mm should be pretreated using 2.05% (w/v) sodium hydroxide solution at solid loading of 20.0% (w/v), 89.3 °C for 38.0 min. Maximal yields of glucose (451.58 mg/gds) and xylose (247.63 mg/gds) could be obtained after hydrolysis at 50 °C for 43.4 h with 7.9% (w/v) of biomass loading, 7.6 FPU/gds of enzyme loading, and 0.2% (w/v) of Tween-80, respectively. The optimized pretreatment conditions resulted in 92.5% of cellulose recovery, 54.6% of hemicellulose recovery, and 78.8% of lignin removal, respectively. Furthermore, the corresponding cellulose conversion and hemicellulose conversion during hydrolysis were 97.1% and 93.6%, respectively.

In addition, attempts were made to enhance cellulose fractionation and improve enzymatic saccharification. In this regard, these two wastes were efficiently employed as substrates for bioethanol production. This research demonstrates the effect of disodium hydrogen phosphate (Na2HPO4) and zinc chloride (ZnCl2) (NZ) as a new catalyst on the development of the sequential pretreatment strategy in the noticeable enzymatic hydrolysis. Physico-chemical changes of the native and the pretreated sustainable wastes were evaluated by compositional analysis, scanning electron microscopy (SEM), X-ray diffractometry (XRD), Fourier transform infrared spectroscopy (FTIR), and thermogravimetric analysis (TGA). These investigations showed major structural changes after the optimized sequential pretreatment. This pretreatment not only influences the delignification process, but also affects the functionalization of cellulose chemical structure. NZ released a higher glucose concentration (328.8 and 996.8 mg/dl) than that of ZnCl2 (Z), which released 203.8 and 846.8 mg/dl from CS and TP, respectively. This work led to the production of about 500 mg/dl of ethanol, which is promising and a competitor to other studies. These findings contribute to increasing the versatility in the reuse of agricultural and agro-industrial wastes to promote interaction areas of pollution prevention, industrialization, and clean energy production, to attain the keys of sustainable development goals.

Abstract:The efficient conversion of hemicellulose and cellulose into reducing sugars remains as one major challenge for biorefinery of lignocellulosic biomass. In this work, saccharification of corncob in the aqueous phase was effectively realized via pretreatment by magnetic carbon-based solid acid (MMCSA) catalyst, combined with the subsequent in situ enzymatic hydrolysis (occurring in the same pretreatment system after separation of MMCSA). Through the combined two-step hydrolysis of corncob, the total sugar (xylose and glucose) yield of 90.03% was obtained, including a xylose yield of 86.99% and an enzymatic digestibility of pretreatment residue of 91.24% (cellulase loading of 20 FPU/g, 24 h). Compared with the traditional enzymatic hydrolysis of pretreated residue, the presented in situ enzymatic hydrolysis system can reach a comparable enzymatic digestibility in one-third reacting time with a half cellulase loading and save about 31% water consumption, which provides a more sustainable and low-cost method for the saccharification of lignocellulose.

Abstract:Microwave-Assisted Alkali (MAA) pretreatment of corncob biomass was optimized for maximum fermentable sugar yield using dilute sulfuric acid hydrolysis. A Central Composite Design (CCD) investigated the effects of microwave power (450–9000 W), NaOH concentration (1.5 to 4.5% wt./v), and irradiation time (10–20 min) on lignin removal. The optimal MAA pretreatment conditions were found to be 691.73 W, 3.4% NaOH, and 15.5 min, resulting in the selective removal of 73.36% lignin with high retention of cellulose (71.69%) and hemicellulose (22.14%). Characterization techniques (FTIR, SEM, XRD, TGA) confirmed substantial changes in surface morphology, functional groups, crystallinity, and thermal stability of the pretreated corncobs. Dilute sulfuric acid hydrolysis (0.5–2.5%, 110130 °C, 3060 min) of the optimized sample yielded a maximum reducing sugar output of 537.42 mg/g, achieved at 0.534% H 2 SO 4 , 122.3 °C, for 58 min. Optimizing Microwave-Assisted Alkali pretreatment process for corncobs could boost biofuel production efficiency, sustainably utilize agricultural waste, and advance a more sustainable energy landscape.

Abstract:In this study, the effectiveness of NaOH extraction (AE), Fenton oxidation (FO), and synergistic pretreatment using AE and FO for enzymatic saccharification of corn stalks (CS) was compared-by statistical analyses. The results showed that AE-FO treatment resulted in statistically higher enzymolysis efficiency and glucose yield than single AE and single FO treatments. Compared with single AE and FO treatments, AE-FO resulted in statistically significant higher lignin removal and xylan removal, consequently improving the cellulose accessibility to cellulase. Among various synergistic pretreatment parameters, utilizing Plackett Burman design determined the most effective factor improving cellulose accessibility to cellulase was NaOH loading. The results that we obtained could be extended to help further improve the synergistic pretreatment process by using CS for the production of biofuels and sugar-based chemicals.

Abstract:Xylooligosaccharides (XOS) hold industrial importance owing to their prebiotic, antioxidant, antibiotic, and anti-inflammatory potential. The fungal enzyme xylanase ( Talaromyces thermophilus F1208) is known for its capacity to produce XOS by hydrolytic degradation. This study aims to achieve the maximum xylan yield through experiments by optimizing corn cob pyrolysate's parameters. The xylan in the pyrolysis solution was converted into XOS using the ultrasonic-coupled enzymolysis method and optimizing its parameters. Optimizing pyrolysis conditions employing maleic acid concentration, pyrolysis temperature, and holding time resulted in a maximum xylan yield (20.04%) at 0.2%, 180 °C, and 30 min of the three parameters, respectively. The multivariate response surface methodology (RSM) for pyrolysis statistically validated the results and suggested pyrolysis temperature and holding time to be the most crucial parameters influencing the yield. Similarly, the highest yield of XOS (21.75%) was obtained at 7.5 pH, 4-h reaction time, and 6-U/mL enzyme dosage for the enzyme hydrolysis alone by selecting these three as optimization parameters. In the case of enzyme hydrolysis coupled to ultrasonication, the maximum yield of 27% XOS was achieved at 190 °C pretreatment temperature, 30-min ultrasonication time, 55 °C ultrasonication temperature, and 0.31-W/cm 2 ultrasonication frequency, which was ~25% higher than that obtained using enzymatic hydrolysis alone. Moreover, ultrasonic intervention shortened the reaction time to 12.5% of the original time. The difference in enzymatic hydrolysis of xylan components (H 1 –H 5 ) after alcohol precipitation was also investigated. It was found that the H 2 and H 3 substrates were slow reaction xylans, which were not easy to be enzymatically hydrolyzed.
