Abstract:Salt stress is a major abiotic factor that negatively impacts the growth, performance, and secondary metabolite production in pepper (Capsicum annuum L.) plants. Brassinosteroids (BRs) play a crucial role in enhancing plant tolerance to abiotic stress, yet their potential in mitigating salt stress in pepper plants, particularly by promoting sugar and glycine betaine accumulation, remains underexplored. In this study, we investigated the effects of the foliar application of 2,4-epibrassinolide (EBR) on salt-stressed pepper seedlings. Our findings revealed that EBR treatment significantly increased the levels of proline, sugar, and glycine betaine under salt stress compared to untreated controls. Moreover, EBR enhanced the antioxidant defense mechanisms in pepper seedlings by increasing sugar and glycine betaine levels, which contributed to the reduction of reactive oxygen species (ROS) and malondialdehyde (MDA) accumulation.

Abstract:: The present research was performed to assess the effect of 24-epibrassinolide (EBR) on salt-stressed soybean plants. Salt stress suppressed growth, biomass yield, gas exchange parameters, pigment content, and chlorophyll fluorescence, but all these parameters were up-regulated by EBR supply. Moreover, salt stress increased hydrogen peroxide, malondialdehyde, and electrolyte leakage. EBR supplementation reduced the accumulation of oxidative stress biomarkers. The activities of superoxide dismutase and catalase, and the accumulation of proline, glycinebetaine, total phenols, and total flavonoids increased with NaCl stress, but these attributes further increased with EBR supplementation. The activities of enzymes and the levels of non-enzymatic antioxidants involved in the Asc-Glu cycle also increased with NaCl stress, and further enhancement in these attributes was recorded by EBR supplementation. Salinity elevated the methylglyoxal content, but it was decreased by the EBR supplementation accompanying with up-regulation of the glyoxalase cycle (GlyI and GlyII). Salinity enhanced the Na+ uptake in root and shoot coupled with a decrease in uptake of Ca2+, K+, and P. However, EBR supplementation declined Na+ accumulation and promoted the uptake of the aforementioned nutrients. Overall, EBR supplementation regulated the salt tolerance mechanism in soybean plants by modulating osmolytes, activities of key enzymes, and the levels of non-enzymatic antioxidants.

Exogenous Salicylic Acid Optimizes Photosynthesis, Antioxidant Metabolism, and Gene Expression in Perennial Ryegrass Subjected to Salt Stress

Abstract:Salicylic acid (SA) is a plant growth regulator that can enhance the abiotic stress tolerance of plants; however, the physiological mechanisms are not yet fully understood. The objective of this study was to examine whether exogenous SA could enhance the salt tolerance of perennial ryegrass (Loliumperenne L.; PRG) and investigate the physiological underlying mechanisms. SA was applied to the foliage of PRG at five concentrations (0, 0.25, 0.5, and 1 mM). The SA-treated grass was grown under either control (0 mM NaCl) or salt stress (250 mM NaCl) conditions for 24 d. The SA treatments reduced the leaf electrolyte leakage (EL), malonaldehyde (MDA), and hydrogen peroxide (H2O2) content by 36%, 41%, and 40%, respectively, relative to the control under salt stress as measured at 24 d. The SA treatments also alleviated the decline in the leaf photosynthetic rate (Pn), stomatal conductance (gs), nitrate activity (NR), turfgrass quality (TQ) ratings, and chlorophyll (Chl) content under salt stress. In addition, exogenous SA increased the activity of superoxide dismutase (SOD), ascorbate peroxidase (APX), and peroxidase (POD) as well as the expression levels of the Cyt Cu/ZnSOD, FeSOD, APX, CAT, and POD genes under salt stress. The results of this study suggested that the foliar application of SA at 0.25 and 0.5 mM may enhance photosynthesis and antioxidant defense systems and thus improve tolerance to salt stress in perennial ryegrass.

Abstract:Soil salinization is a major problem all over the world. The accumulation of salt in soil reduces the root water uptake and directly affects plant growth and metabolic activities. Brassinosteroid is a plant hormone that plays an important role in regulation of plant growth and physiological process, including promotion of cell expansion and elongation, signal transduction and stress response. Exogenous 24-epibrassinolide (EBL) has been proved to alleviate various environmental stress in plants. However, the role that EBL plays in salt stress response is still unknown in tall fescue ( Festuca arundinacea ). In this study, the physiology and molecular mechanisms regulated by exogenous EBL of salt stress response in tall fescue was investigated. Tall fescue plants were divided into four groups, including control (CK), NaCl solution (SALT), 24-epibrassinolide (EBL), NaCl solution + 24-epibrassinolide (SE). During the growth period of tall fescue, we found that electrolyte leakage (EL) and malondialdehyde (MDA) were decreased, chlorophyll (Chl) content and antioxidant enzyme activity were increased in leaves of tall fescue in SE group compared with SALT group, indicating that EBL improved the salt tolerance in grasses. Transcriptomic profiling analysis showed that after 12 h of treatments, 10,265, 13,830 and 10,537 differential genes were expressed in EBL, SALT, and SE groups compared with control, respectively. These differentially expressed genes (DEGs) mainly focused on binding, catalytic activity, cellular process, metabolic process, cellular anatomical entity. Moreover, most of the differential genes were expressed in the plant hormone signal transduction pathway. These results helped us to better understand the mechanism of exogenous 24-epibrassinolide to improve the salt tolerance of tall fescue.

Abstract:The effect of brassinosteroid (BR) on relative water content (RWC), stomatal conductance (g(s)), net photosynthetic rate (PN), intercellular CO2 concentration (Ci), lipid peroxidation level, activities of antioxidant enzymes and abscisic acid concentration (ABA) in tomato (Lycopersicon esculentum)seedlings under water stress was investigated. Two tomato genotypes, Mill. cv. Ailsa Craig (AC) and its ABA-deficient mutant notabilis (not), were used. Water stress was achieved by withholding water and both the AC and not plants were treated with 1 mu M 24-epibrassinolide (EBR) or distilled water as a control. The RWC, g(s), Ci and PN were significantly decreased under water stress. However, EBR treatment significantly alleviated water stress and increased the RWC and PN. EBR application also markedly increased the activities of antioxidant enzymes (catalase, ascorbate peroxidase and superoxide dismutase) while it decreased g(s). Ci and the contents of H2O2 and malondialdehyde (MDA). Interestingly, ABA concentration in AC and not plants was markedly elevated after EBR treatment although the increasing rate and amplitude of ABA in not plants treated by EBR was significantly lower than those in AC plants. Our study suggested that amelioration of the drought stress of tomato seedlings may be caused by EBR-induced elevation of endogenous ABA concentration and/or the activities of antioxidant enzymes. Enhanced Seedling Growth and Physiological Performances of Melia azedarach L. by Foliar Application of 24-Epibrassinolide under Salt Stress

Abstract:Salinity is a serious environmental problem following a worsening trend. This study investigates the role of 24-epibrassinolide(EBR) in regulating plant growth and physiological performances, particularly in alleviating the negative effects of salt stress. Melia azedarach L. seedlings from two seed sources, Sheyang (SY) and Xiashu (XS), were exposed to sea salt and treated with different concentrations of EBR within a 60-day period. The results demonstrate that appropriate EBR application improved the seedlings' stress tolerance by promoting growth and physiological systems. In terms of the relative increment, it showed that a difference of 1.45% and 1.13% in the SY and XS groups was the positive effect of the highest EBR treatment concentration. As for diameter growth, the difference observed was 2.51% and 1.80% for the SY and XS groups, respectively. In all physiological measurements, including the content of photosynthetic pigments, water relations, membrane stability, osmolytes and antioxidant enzymes, significant changes generally observed between salt stress alone and the highest EBR treatment concentration. A better performance was observed in the SY seed source, which is of a coastal nature. These findings contribute to our understanding of Melia azedarach's adaptation to changing environments and provide potential for further molecular studies as well as valuable insights for forestry, agricultural and ecological research.

Abstract:The brassinosteroids (BRs) constitute a recently defined class of plant hormone that can enhance the resistance of plants to multiple stresses. However, the effects of BRs on salt-stressed woody plants, notably on photosynthesis and chloroplast ultrastructure, have received little attention. Black locust (Robinia pseudoacacia L.) seeds and seedlings were pretreated with 1.04 µmol L− 1 24-epibrassinolide (24-epiBL) by soaking and root dipping, respectively, and grown under non-saline or saline conditions (0, 100, 200 mmol L− 1 NaCl). Salinity stress decreased photosynthesis, chlorophyll concentration, transpiration, and stomatal conductance but also decreased the water-use efficiency, while chlorophyll fluorescence indicated a decrease in photochemical quenching and in maximum potential quantum efficiency. Indicators of oxidative stress (for example, H2O2 and antioxidant enzymes), membrane leakage, and amounts of Na+ ions in leaves and chloroplasts were increased and, at the highest stress, chloroplast ultrastructure was severely disrupted. Exogenous 24-epiBL improved membrane stability and reduced foliar Na+ levels, while substantially alleviating stress-induced changes in photosynthetic gas exchange. Improvements in chlorophyll content and indicators of oxidative stress were not as large but were still highly significant. Thylakoid membrane structure was protected. Both methods of applying 24-epiBL were effective, but root-dipped seedlings performed marginally better. The results suggest that treatment of black locust seedlings with 24-epiBL prior to planting may improve performance and aid establishment on salt-affected soils.
Effects of Brassinostreoids on photosynthesis and ultrastructure of chloroplasts in Robinia pseudoacacia seedlings under salt stress

Abstract:[Objective] This article focus on the effect of the exogenous 24-epibrassinosteroid (BRs),used by roots dipping way,on photosynthesis and ultrastructure of chloroplasts in Robinia pseudoacacia seedling sunder salt stress which results could provide the technical guidance for forestation on saline and alkaline land.[Method] The research was based on the annual R.pseudoacacia seedings used different concentration gradients of BRs(0.1,0.3,0.5 mg/L) by the root treatment under 100 mmol/L NaCl stress for 50 d.CK1 is blank control (0 mmol/L NaCl+0 mg/L BRs),CK2 is stress control (100 mmol/L NaCl+0 mg/L BRs).According to the analysis of multiple comparison and ANOVA((P＜0.05)),it showed some results about the changes of morphological,moisture physilological indexed,chlorophyll,photosynthetic and chloroplast ultrastructure parameters.[Result] We found that the salt stress had a significant reduction of plant height/diameter (H/SBD) and relative water content(RWC)(P＜0.05) of the leaves,a notable increase with specific leaf weight(SLW) and salt injury index(P＜0.05).The chlorophyll a and chlorophyll b also declined significantly(P＜0.05),but the chlorophyll a/b rose slightly under salt stress.The leaf photosynthetic rate (Pn),stomatal conductance (Gs),transpiration rate (Tr),intercellular CO2 concentration (Ci),water use efficiency(WUE)decreased and the chloroplast ultrastructures in the leaves of R.pseudoacacia seedings were damaged by the salinity (100 mmol/L NaC1).Through the usage of exogenous BRs,the results showed that in contrast to CK2,application of BRs improved the values of H/SBD and RWC,SLW decreased besides 0.1 mg/L BRs treatment and the salt injury index had an obvious reduce (P＜0.05).The effective role of BRs played in maintaining the normal photosynthesis of the leaves under salt stress attributed to improvement with the contents of chlorophyll a,chlorophyll b and chlorophyll a/b.As well as,the concentration of 0.5 mg/L BRs increased stomatal conductance (Gs),maintained the transpiration rate (Tr) and intercellular CO2 concentration (Ci),water use efficiency(WUE),kept leaf photosynthesis compared to stress control(CK2).Analysis of ultrastructure of chloroplasts,the plants dealing with BRs can keep the certain numbers of chloroplasts,maintain the structure of chloroplasts completely and tightness of thylakoid lamellas,increase the numbers of eosinophilic osmium particles into leaves,which were advantage for photosynthesis by maintaining the functionality of chloroplasts.[Conclusion]These results suggest that the photosynthesis of the seedings (Robinia L.)is restrained under 100 mmol/L salt stress for 50 d,adding BRs is able to maintain the certain degree of leaf photosynthesis,improves the tolerance of the R.pseudoacacia seedings under the stress (100 mmol/L NaC1) and the optimal concentration of BRs is 0.5 mg/L.
Physiological mechanism of exogenous brassinolide alleviating salt stress injury in rice seedlings

Abstract:Brassinolide (BR) is a sterol compound, which can regulate plant seed germination, flowering, senescence, tropism, photosynthesis, stress resistance, and is closely related to other signaling molecules. This study aimed to evaluate the ability of soaking with BR to regulate growth quality at rice seedling stage under salt stress. Results demonstrated that salt stress increases the contents of ROS, MDA, Na + and ABA, reduces the the SPAD value, net photosynthetic rate (Pn), stomatal conductance (Gs), transpiration rate (Tr), maximum fluorescence (Fm), variable fluorescence (Fv), the effective photochemical efficiency of PSII (Fv/Fo) and the maximum photochemical efficiency of PSII (Fv/Fm), reduces the biomass production and inhabits plant growth. All of these responses were effectively alleviated by BR soaking treatment. Soaking with BR could increase the activities of superoxide dismutase, peroxidase, catalase, ascorbate peroxidase, and the contents of ascorbic acid, glutathione as well as soluble protein and proline, while BR soaking treatment inhibited the accumulation of ROS and reduced the content of MDA. BR soaking significantly reduced the contents of Na + and increased the contents of K + and Ca 2+ , indicating that soaking with BR is beneficial to the excretion of Na + , the absorption of K + and Ca 2+ and the maintenance of ion balance in rice seedlings under salt stress. BR also maintained endogenous hormone balance by increasing the contents of indoleacetic acid (IAA), zeatin (ZT), salicylic acid (SA), and decreasing the ABA content. Soaking with BR significantly increased the SPAD value, Pn and Tr and enhanced the Fm, Fv/Fm and Fv/Fo of rice seedlings under NaCl stress, protected the photosythetic system of plants, and improved their biomass. It is suggested that BR was beneficial to protect membrane lipid peroxidation, the modulation of antioxidant defense systems, ion balance and endogenous hormonal balance with imposition to salt stress.

Effects of Exogenous 24-Epibrassinolide on the Leaf Photosynthetic Characteristics and Polyamines Content of Cucumber Seedlings under Hypoxia Stress

Abstract:A water culture experiment with 1/2 Hoagland solution was carried out to study the effects of exogenous 24-epibrassinolide (EBR) on the leaf photosynthetic characteristics and polyamines (PAs) content of cucumber seedlings under hypoxia stress. Under the stress, the leaf net photosynthetic rate (Pn), stomatal conductance (g(s)), transpiration rate (Tr), and intercellular CO2 concentration (Ci) decreased significantly while the chlorophyll content was in adverse, and the seedling growth was inhibited. Hypoxia stress increased the leaf Put, Spd, and Spm contents and Put/PAs ratio significantly, but decreased the (Spd+Spm)/Put. Under the stress, exogenous EBR not only increased the Pn, g(s), Tr, and chlorophyll content significantly, but also increased the contents of free Spm, conjugated Spd and Spm, and bound Put, Spd, and Spm significantly. Meanwhile, the PAs content and (Spd + Spm)/Put ratio were further increased, and the Put/PAs ratio decreased. The results suggested that exogenous EBR could regulate the formation and form transformation of PAs in cucumber seedling leaf, made the leaf keep a higher photosynthetic performance, promote the increase of leaf area and dry mass, and alleviate the damage of hypoxia stress.
The Positive Effect of Different 24-Epibl Pretreatments on Salinity Tolerance in Robinia Pseudoacacia L. Seedlings

Abstract:As a brassinosteroid (BR), 24-epibrassinolide (24-epiBL) has been widely used to enhance the resistance of plants to multiple stresses, including salinity. Black locust (Robinia pseudoacacia L.) is a common species in degraded soils. In the current study, plants were pretreated with three levels of 24-epiBL (0.21, 0.62, or 1.04 µM) by either soaking seeds during the germination phase (Sew), foliar spraying (Spw), or root dipping (Diw) at the age of 6 months. The plants were exposed to salt stress (100 and 200 mM NaCl) via automatic drip-feeding (water content ~40%) for 45 days after each treatment. Increased salinity resulted in a decrease in net photosynthesis rate (Pn), stomatal conductance (Gs), intercellular:ambient CO2 concentration ratio (Ci/Ca), water-use efficiency (WUEi), and maximum quantum yield of photosystem II (PSII) (Fv/Fm). Non-photochemical quenching (NPQ) and thermal dissipation (Hd) were elevated under stress, which accompanied the reduction in the membrane steady index (MSI), water content (RWC), and pigment concentration (Chl a, Chl b, and Chl). Indicators of oxidative stress (i.e., malondialdehyde (MDA) and antioxidant enzymes (peroxidase (POD) and superoxide dismutase (SOD)) in leaves and Na+ content in chloroplasts increased accompanied by a reduction in chloroplastid K+ and Ca2+. At 200 mM NaCl, the chloroplast and thylakoid ultrastructures were severely disrupted. Exogenous 24-epiBL improved MSI, RWC, K+, and Ca2+ content, reduced Na+ levels, maintained chloroplast and thylakoid membrane structures, and enhanced the antioxidant ability in leaves. 24-epiBL also substantially alleviated stress-induced limitations of photosynthetic ability, reflected by elevated chlorophyll fluorescence, pigment levels, and Pn. The positive effects of alleviating salt stress in R. pseudoacacia seedlings in terms of treatment application was Diw > Sew > Spw, and the most positive impacts were seen with 1.04 µM 24-epiBL. These results provide diverse choice for 24-epiBL usage to defend against NaCl stress of a plant.
Exogenous Brassinosteroid Enhances Zinc tolerance by activating the Phenylpropanoid Biosynthesis pathway in Citrullus lanatus L

Abstract:Zinc (Zn) is an important element in plants, but over-accumulation of Zn is harmful. The phytohormone brassinosteroids (BRs) play a key role in regulating plant growth, development, and response to stress. However, the role of BRs in watermelon ( Citrullus lanatus L.) under Zn stress, one of the most important horticultural crops, remains largely unknown. In this study, we revealed that 24-epibrassinolide (EBR), a bioactive BR enhanced Zn tolerance in watermelon plants, which was related to the EBR-induced increase in the fresh weight, chlorophyll content, and net photosynthetic rate (Pn) and decrease in the content of hydrogen peroxide (H 2 O 2 ), malondialdehyde (MDA), and Zn in watermelon leaves. Through RNA deep sequencing (RNA-seq), 350 different expressed genes (DEG) were found to be involved in the response to Zn stress after EBR treatment, including 175 up-regulated DEGs and 175 down-regulated DEGs. The up-regulated DEGs were significantly enriched in 'phenylpropanoid biosynthesis' pathway (map00940) using KEGG enrichment analysis. The gene expression levels of PAL , 4CL , CCR , and CCoAOMT , key genes involved in phenylpropanoid pathway, were significantly induced after EBR treatment. In addition, compared with Zn stress alone, EBR treatment significantly promoted the activities of PAL, 4CL, and POD by 30.90%, 20.69%, and 47.28%, respectively, and increased the content of total phenolic compounds, total flavonoids, and lignin by 23.02%, 40.37%, and 29.26%, respectively. The present research indicates that EBR plays an active role in strengthening Zn tolerance, thus providing new insights into the mechanism of BRs enhancing heavy metal tolerance.

Abstract:Zinc (Zn) is an important element in plants, but over-accumulation of Zn is harmful. It is well-known that brassinolide (BR) plays a key role in the regulation of abiotic stress responses in plants. However, the effects of brassinolide on alleviating Zn phytotoxicity in watermelon ( Citrullus lanatus L.) seedlings are not clear. The purpose of this study was to study the effect of 24-epibrassinolide (EBR, one of the bioactive BRs) on Zn tolerance of watermelon seedlings and its potential resistance mechanism. Exposure to excessive Zn significantly inhibited shoot and root fresh weight of watermelon, but this could be significantly alleviated by the optimum 0.05 μM EBR. Exogenous spraying EBR increased the pigments and alleviated oxidative damage caused by Zn through reducing Zn accumulation and the levels of reactive oxygen species (ROS) and malonaldehyde (MDA) and increasing the activities of antioxidant enzymes and contents of ascorbic acid (AsA) and glutathione (GSH). Importantly, the relative mRNA levels of antioxidant genesincluding Cu/Zn-superoxidedismutase ( Cu-Zn SOD ), catalase ( CAT ), ascorbic acid peroxidase ( APX ), and glutathione reductase ( GR ) were significantly induced after EBR treatment. In addition, EBR pre-treatment induced lignin accumulation under Zn stress, and the activity of phenylalanine ammonia-lyase (PAL) and 4-coumaric ligase (4CL), two key enzymes regulating lignin synthesis, also tended to be consistent. Collectively, the present research proves the beneficial effects of EBR in response to Zn stress through enhancing antioxidant defense and lignin accumulation and provides a new insight into the mechanism of BR-enhancing heavy metal tolerance.

Abstract:The present study investigates a possible mechanism of 24-Epibrassinolide (EBL) mediated alleviation of salinity stress in Brassica juncea L. seedlings. EBL (10 –6 M and 10 –8 M) pretreated seedlings were grown up to ten-days either in alone and/or combination with defined levels of salinity treatments (100 mM and 200 mM NaCl) under laboratory growth conditions. Results showed that salinity stress significantly ( P ≤ 0.05 ) increased the Na + accumulation in a dose-dependent manner, which aggravated the nutrient(s) homeostasis and pigment contents by inducing oxidative damage through elevated hydrogen peroxide, malondialdehyde, and methyl glyoxal production. EBL mediated alleviation of salinity stress was achieved through decreased nuclear damage and cell death, coupled with increased membrane stability index and nutrients accumulation. The positive roles of EBL were also reflected through maintenance of AsA/DHA and GSH/GSSG redox ratios' that were maximally enhanced by 78% and 28%, respectively under 200 mM NaCl. EBL mediated up-regulation of γ-glutamyl cysteine synthetase activity increased the glutathione pool, which boosted the ascorbate–glutathione cycle and methyl glyoxal detoxification system. The nitrate reductase activity, endogenous nitric oxide and S-nitrosothiols pools were maximally increased by 45, 32, and 33% respectively in 200 mM NaCl treatment, over the salinity alone counterpart. Taken together, our findings suggested that EBL pretreatment alleviates salinity stress by regulating nitric oxide homeostasis, osmolytes synthesis, antioxidant defense systems, and increasing the nutrients and pigments contents in mustard seedlings under salinity stress. Therefore, seed-priming with EBL can be considered as an effective approach for improving the plant productivity under salinity stress.

Abstract:Brassinosteroids (BRs) represent a group of plant signaling molecules with a steroidal skeleton that play an essential role in plant adaptation to different environmental stresses, including drought. In this work, the effect of pretreatment with 0.4 μM 24-epibrassinolide (EBR) on the oxidant/antioxidant system in 4-day-old wheat seedlings (Triticum aestivum L.) was studied under moderate drought stress simulated by 12% polyethylene glycol 6000 (PEG). It was revealed that EBR-pretreatment had a protective effect on wheat plants as evidenced by the maintenance of their growth rate, as well as the reduction in lipid peroxidation and electrolyte leakage from plant tissues under drought conditions. This effect was likely due to the ability of EBR to reduce the stress-induced accumulation of reactive oxygen species (ROS) and modulate the activity of antioxidant enzymes. Meanwhile, EBR pretreatment enhanced proline accumulation and increased the barrier properties of the cell walls in seedlings by accelerating the lignin deposition. Moreover, the ability of EBR to prevent a drought-caused increase in the intensity of the total dark respiration and the capacity of alternative respiration contributes significantly to the antistress action of this hormone.

Abstract:The accumulation of salts within the rhizosphere is a common phenomenon in arid and semi-arid regions where irrigation water is high in salts. A previous study established the ameliorative effect of foliarly applied 24-epibrassinolide (BR) on soybean under salinity stress. As a follow-up to that study, this work evaluated the effects of BR on the electrical conductivity of saturated soil extracts (ECses) under soybean exposed to salt stress. Three salinity levels (3.24, 6.06 and 8.63 dS/m) in a factorial combination with six frequencies of BR application—control, seedling, flowering, podding, seedling + flowering and seedling + flowering + podding—were the treatments, and the rhizospheric ECse was monitored from 3 to 10 weeks after the commencement of irrigation with saline water (WAST). The principal component analysis revealed that samples in saline BR treatments clustered together based on the BR application frequencies. There was a significant increase in ECse with increases in salinity and WAST. The frequent application of BR significantly reduced ECse to 5.07 and 4.83 dS/m relative to the control with 6.91 dS/m, respectively, at week 10. At 8.63 dS/m, the application of BR (seedling + flowering + podding) reduced ECse by 31.96% compared with the control. The underlining mechanism is a subject for further investigation.
24-epibrassinolide improves cadmium tolerance and lateral root growth associated with regulating endogenous auxin and ethylene in Kentucky bluegrass

Abstract:The application of phytohormones is a viable technique to increase the efficiency of phytoremediation in heavy metal-contaminated soils. The objective of this study was to determine how the application of 24-epibrassinolide (EBR), a brassinosteroid analog, could regulate root growth and tolerance to cadmium (Cd) stress in Kentucky bluegrass. As a result, the number of lateral root primordia and total root length in the Cd-treated seedlings decreased by 33.1 % and 56.5 %, respectively. After the application of EBR, Cd accumulation in roots and leaves, and the negative effect of Cd on root growth were reduced under Cd stress. Additionally, the expression of the brassinosteroid signaling gene PpBRI1 was significantly upregulated by exogenous EBR. Moreover, exogenous EBR upregulated the expression of genes encoding antioxidant enzymes and improved the activity of antioxidant enzymes, thereby reduced oxidative stress in roots. Finally, targeted hormonomics analysis highlighted the utility of the application of EBR to alleviate the effect of Cd on the reduction in auxin (IAA) content and the increase in ethylene (ACC) content. These were known to be associated with the upregulation in the expression of auxin biosynthesis gene PpYUCCA1 and downregulation in the expression of ethylene biosynthesis gene PpACO1 in the roots treated with Cd stress. Overall, the application of EBR alleviated Cd-induced oxidative stress in addition to improving root elongation and lateral root growth crosstalk with auxin and ethylene in Kentucky bluegrass subjected to Cd stress. This study further highlights the potential role of brassinosteroids in improving the efficiency of phytoremediation for Cd-contaminated soils.

Abstract:Introduction: High salinity significantly hampers global agricultural productivity. Plants typically undergo lower nitrogen utilization efficiency (NUE) under salt stress. As an active byproduct from brassinolide biosynthesis, 24-epibrassinolide (EBR) is involved in regulating the stress-treated plant N absorption and assimilation. However, the exogenous EBR application effects' on N absorption and assimilation in apple exposed to the salt-stressed condition remains unclear. Methods: We sprayed exogenous EBR (0.2 mg L −1 ) on apple dwarf rootstock (M9T337) seedlings (growing hydroponically) under salt (NaCl) stress in a growth chamber. We analyzed the seedling development, photosynthesis and its-mediated C fixation, N ( NO3− ) absorption and assimilation in reponse to exogenous EBR application under salt stress. Results: The findings demonstrated that NaCl stress greatly hampered seedlings' root growth and that exogenous EBR application obviously alleviated this growth suppression. Exogenous EBR-treated plants under NaCl stress displayed the more ideal root morphology and root activity, stronger salt stress tolerance and photosynthetic capacity as well as higher C- and N-assimilation enzyme activities, NO3− ion flow rate and nitrate transporter gene expression level than did untreated plants. Furthermore, the results of isotope labeling noted that exogenous EBR application also enhanced 13 C-photoassimilate transport from leaves to roots and 15 NO3− transport from roots to leaves under NaCl stress. Conclusion: Our findings imply that exogenous EBR application, through strengthening photosynthesis, C- and N-assimilation enzyme activities, nitrate absorption and transport as well as synchronized optimizing the distribution of seedlings' C and N, has a fundamental role in improving NUE in apple rootstock seedlings under salt stress.

Abstract:Brassinosteroids (BRs), a group of naturally occurring plant steroidal compounds, are essential for plant growth, development and stress tolerance. Recent studies showed that BRs could induce systemic tolerance to biotic and abiotic stresses; however, the molecular mechanisms by which BRs signals lead to responses in the whole plant are largely unknown. In this study, 24-epibrassinosteroid (EBR)-induced systemic tolerance in Cucumis sativus L. cv. Jinyan No. 4 was analyzed through the assessment of symptoms of photooxidative stress by chlorophyll fluorescence imaging pulse amplitude modulation. Expression of defense/stress related genes were induced in both treated local leaves and untreated systemic leaves by local EBR application. With the suppressive subtractive hybridization (SSH) library using cDNA from the phloem sap of EBR-treated plants as the tester and distilled water (DW)-treated plants as the driver, 14 transcripts out of 260 clones were identified. Quantitative Real Time-Polymerase Chain Reaction (RT-qPCR) validated the specific up-regulation of these transcripts. Of the differentially expressed transcripts with known functions, transcripts for the selected four cDNAs, which encode an auxin-responsive protein (IAA14), a putative ankyrin-repeat protein, an F-box protein (PP2), and a major latex, pathogenesis-related (MLP)-like protein, were induced in local leaves, systemic leaves and roots after foliar application of EBR onto mature leaves. Our results demonstrated that EBR-induced systemic tolerance is accompanied with increased transcript of genes in the defense response in other organs. The potential role of phloem mRNAs as signaling components in mediating BR-regulated systemic resistance is discussed.
Exogenous Application of 24-Epibrassinolide Confers Saline Stress and Improves Photosynthetic Capacity, Antioxidant Defense, Mineral Uptake, and Yield in Maize

Abstract:Salinity is one of the major environmental stresses threatening crop production, the natural ecosystem, global food security, and the socioeconomic health of humans. Thus, the development of eco-friendly strategies to mitigate saline stress and/or enhance crop tolerance is an important issue worldwide. Therefore, this study was conducted during the summer of 2022 to investigate the potential of 24-Epibrassinolide (EBL) for mitigating saline stress and improving photosynthetic capacity, antioxidant defense systems, mineral uptake, and yield in maize (Zea mays L.) grown under a controlled hydroponic system. Three saline stress levels—S1 (control/no added NaCl), S2 (60 mM NaCl), and S3 (120 mM NaCl)—were continuously applied with nutrient solution, whereas exogenous EBL (i.e., control, 0.1 μM and 0.2 μM) was applied as exogenous application three times (i.e., 40, 55, 70 days after sowing). The experiment was designed as a split-plot in a randomized complete block design (RCBD) in which saline stress was the main factor and EBL treatment was the sub-factor. Results showed that saline stress significantly affected plant growth, physiological performance, biochemistry, antioxidant activity, and yield attributes. However, the exogenous application of EBL at 0.2 μM significantly mitigated the salt stress and thus improved plant performance even under 120 mM NaCl saline stress. For instance, as compared to untreated plants (control), 0.2 μM EBL application improved plant height (+18%), biomass (+19%), SPAD (+32%), Fv/Fm (+28%), rate of photosynthesis (+11%), carboxylation efficiency (+6%), superoxide dismutase (SOD +14%), catalase (CAT +18%), ascorbate peroxidase (APX +20%), K+ (+24%), 100-grain weight (+11%), and grain yield (+47%) of maize grown under salt stress. Additionally, it resulted in a 23% reduction in Na+ accumulation in leaves and a 25% reduction in for Na+/K+ ratio under saline stress as compared to control. Furthermore, the Pearson's correlation and principal component analysis (PCA) highlighted the significance of exogenous EBL as saline stress mitigator in maize. Overall, our results indicated the protective effects of EBL application to the alleviation of saline stress in crop plants. However, further exploration of its mechanism of action and crop-specific response is suggested prior to commercial use in agriculture.
