Abstract:Cross-breeding has been successfully applied in agriculture and aquaculture industries to improve important production traits. In our previous studies, we produced the F-1 hybrids with high heterosis in growth by crossing the bay scallops with the Peruvian scallops and selected a new strain, Bohai Red, from the F-1 hybrids. In this study, we selected another new strain from the backcross (BC1) of the F-1 hybrids with bay scallops. The base cohort (G(0)) was size-selected from the BC1, and the G(1), G(2), G(3) and G(4) cohorts were produced with size-selected brood stocks from the G(0), G(1), G(2) and G(3) cohorts respectively. Our results showed that average shell height, shell length, shell width and whole body weight of the G(4) cohort were increased by 16.8%, 16.8%, 11.5% and 43.6%, respectively, compared with the unselected control group of bay scallop. This new strain has now become one of the major cultured Argopecten scallops in northern China waters and was later named QN-2.

Abstract:In our previous studies, inter-specific hybrid F1 with significant heterosis in growth was produced by hybridizing the bay scallop, Argopecten irradians irradians with the Peruvian scallop, A. purpuratus. However, due to the hermaphroditic nature of the two Argopecten scallops, it is difficult to produce the hybrid seed at large scales. To efficiently utilize the genetic resources of the two scallops, this study aimed to breed a new scallop strain from the hybrids. The key step in the selection was the breeding of the F1 generation from the hybrid cohort, most of which were both male- and female-sterile. A small cohort of F1 with normal male- and female-fertility was produced through mass spawning of a large cohort of hybrids. The F2–F6 cohorts were reproduced with size- and color-selected brood stocks from the F1–F5 cohort, respectively. The results showed that, compared with the control bay scallop cohorts, the average shell height, average whole body weight and average adductor muscle weight were increased by 15.6%, 37.9% and 40.7% in F1, by 14.7%, 33.9% and 43.6% in F2, by 14.1%, 33.1% and 38.4% in F3, by 11.7%, 38.8% and 49.3% in F4, by 12.2%, 37.9% and 53.8% in F5, by 18.5%, 42.6% and 75% in F6, respectively. After six generations of selection, the median lethal temperature range was expanded from 3.0–29.0°C in F1 to 0°C–29.1°C in F6, suggesting that the selected strain can be reared in most waters of China, especially in Bohai Sea and Yellow Sea. Thus, through continuous selection on size and color, we have obtained a superior strain from the hybrids between the bay scallop and the Peruvian scallop, which is named ‘Bohai Red’.

Abstract:Two populations of Longidorus henanus Xu & Cheng, 1992 with all four developmental stages were found. Description and morphometrics of all these stages are provided. The morphological and molecular variations among intraspecies of L. henanus are discussed and given. The polytomous key codes of Chen et al. (1997) for L. henanus are modified in parentheses as follows: A34, B23, C34, D2, E3, F(2)3, G(1)23, H12, I1. Furthermore, the phylogenetic trees of 18S rRNA gene and D2-D3 expansion region of 28S rRNA gene supported L. henanus as a unique and valid species with respect to previously sequenced Longidorus species.

Abstract:The present paper deals with the larval and juvenile fish of Lateolabrax japonicus. The morphological characteristics and the ecological habits were observed under the condition of artificial seed breeding in the process of its larval and juvenile fish development. The results show that, about ninth days after hatching, the yolk sac disappeared. About thirty sixth days after hatching, median fin rays were differentiated. About fifty sixth days after hatching, the scales weve formed. About eighty fifth days after hatching, the body was covered completely by scales. With the development of its morphology, its ecological habits were also changed accordingly.

The known causes of pulmonary eosinophilia include fungi – in particular Aspergillus fumigatus, parasites, various toxins and drugs. The pulmonary eosinophilias of unknown cause include Loffler’s syndrome, acute and chronic eosinophilic pneumonia, the hypereosinophilic syndrome, bronchocentric granulomatosis and pulmonary eosinophilias associated with vasculitis. These candidate disorders were all excluded in our case. Controlling the eosinophilia in blood and lung is important in order to prevent tissue damage caused by eosinophils. Although the number of reported cases is small, we want to recognize that pulmonary eosinophilia in our case may be an indication of immune dysfunction accompanied with EPF. Close follow-up of this patient is needed to see whether systemic involvement occurs in the future. Furthermore a better understanding of eosinophil biology may shed light on the pathophysiology of such associations.

Abstract:Twentyfour hours after the intraperitoneal injection of a single dose of allylisopropylacetamide (AIA) in amounts of 100 mg/kg or more to 30-day old male Wistar rats, the livers of most of the animals showed an irregular yellow coloration and a fragile consistency. No macroscopic changes were detected following AIA doses of 25 or 50 mg/kg. Bromsulphthalein retention was not significantly increased after the administration of 25 mg/kg AIA, but distinctly enhanced after 400 mg/kg. Succinate-dehydrogenase-activity in liver homogenate was not altered after 25 mg/kg, but significantly decreased after 400 mg/kg. Ethylmorphine-N-demethylation activity was enhanced after small doses without increase of cytochrome P-450 (cyt. P-450) concentration and decreased after higher doses, accompanied by a remarkable cyt. P-450 loss. GPT activity in serum and liver was not altered after both 25 and 400 mg/kg AIA. GOT activity was slightly but significantly enhanced both in serum and liver after the high dose of 400 mg/kg. Thus in addition to the well-known cyt. P-450 destruction other signs of hepatotoxicity could be demonstrated.

Abstract:The sex determination system of largemouth bass ( Micropterus salmoides , LMB) is XX/XY; however, the underlying molecular mechanisms involved in early sex differentiation, gonadal development, and exogenous hormone-induced sex reversal remain unknown. In this study, LMB at 15 days post-hatching (dph) were fed diets containing 20 mg/kg of 17α-methyltestosterone (17α-MT) or 30 mg/kg of 17β-estradiol (17β-E 2 ) for 60 days, respectively. Serum steroid levels, histological observations of the gonads, and identification of sex-specific markers were employed to screen the gonads of 60-day-old normal female fish (XX-F), normal male fish (XY-M), 17β-E 2 induced pseudo-female fish (XY-F), and 17α-MT-induced pseudo-male fish (XX-M) for transcriptome sequencing in order to uncover genes and pathway involved in the process of sexual reversal. The results from histology and serum sex steroid hormone analysis showed that both 17α-MT and 17β-E2 were capable of inducing sex reversal of LMB at 15 dph. Transcriptome results revealed a total of 2,753 genes exhibiting differential expression, and the expression pattern of these genes in the gonads of XX-M or XY-F resembled that of normal females or males. The male sex-biased genes that are upregulated in XX-M and downregulated in XY-F are referred to as key genes for male reversal, while the female sex-biased genes that are upregulated in XY-F and downregulated in XX-M are referred to as key genes for female reversal. Finally, 12 differentially expressed genes (DEGs) related to male sex reversal were screened, including star2 , cyp17a , cyp11b1 , dmrt1 , amh , sox9a , katnal1 , spata4 , spata6l , spata7 , spata18 and foxl3 . 2 DEGs ( foxl2a and cyp19a1b ) were found to be associated with female sex reversal. The changes in these genes collectively influence the direction of sex differentiation of LMB. Among them, star2 , dmrt1 and cyp19a1b with significantly altered expression levels may play potentially crucial role in the process of gender reversal. The expression patterns of 21 randomly selected genes were verified using qRT-PCR which confirmed the reliability and accuracy of the RNA-seq results. These findings not only enhance our understanding of the molecular basis underlying sex reversal but also provide crucial data support for future breeding research on unisexual LMB.

Abstract:With the development of the laver industry, germplasm depression has become a serious issue, and current cultivars cannot adapt to different aquaculture regions. In order to increasing the genetic diversity and developing more germplasm sources, it is urgent and reasonable to construct a mutant library with more new germplasms. In this research, a mutant library was constructed by ethyl methanesulfonate (EMS)-mutagenized archeospores, and the most optimal treatment procedure was determined by performing different mutagen concentrations (2.25%) and treatment times (30 min). A total of 1860 haploid thalli were produced as the M1 mutant population and further cultured into conchocelis clones for the reservation of germplasm resources. Among these, 667 individual thalli were evaluated for their phenotypic traits, including thallus length, thallus width, length/width, thallus shape, photosynthesis ability, thallus color, thallus margin, and specific growth speed. The mutation frequency of the length/width ratio was 17.39%, Fv/Fm and NPQ were 21.84% and 29.35%, respectively, and SGR was 13.59%. The mutation frequency of thallus color was 0.91%. This work may not only provide a basic practical reference guide for EMS-based mutant library construction for other seaweeds but, more importantly, also serve as a valuable resource for functional genomics research and laver breeding.

Abstract:Abstract Previous work on community structure of the invertebrate fauna of Japanese wetlands indicates the presence of intraguild predation between the heteropterans Laccotrephes japonensis (Nepidae) and Kirkaldyia deyrolli (Belostomatidae). We designed a series of experiments to understand the biotic interactions of several species of sympatric heteroptera and their shared prey. Adult Laccotrephes japonensis (Nepidae), first‐instar nymph of Kirkaldyia (=Lethocerus) deyrolli (Belostomatidae), Hyla japonica tadpole, and fourth‐instar nymph of Appasus japonicus (Belostomatidae) have been shown to be intraguild predator, intraguild prey, common prey, and prey of L. japonensis , respectively. To further understand the factors affecting prey preference by L. japonensis , we also examined the comparison of swimming speed in the three prey animals ( K . deyrolli first‐instar nymph, A . japonicus fourth‐instar nymph, and H . japonica tadpole), and effects of prey animals on weight gain of L. japonensis adult. Despite there being no significant difference in weight gain or swimming speed of the three prey species, L. japonensis exhibited a strong preference for the 1st‐instar nymph of K. deyrolli . We suggested that this may be evidence for one of elimination of a potential competitor, K . deyrolli , by L . japonensis through intraguild predation.

Abstract:Salinity , being an indispensable abiotic factor crucial for the survival of marine organisms, has demonstrated diverse alterations globally in response to the current trend of global warming. In this study, the effect of chronic low salinity stress on teleosts' sex differentiation was investigated using Cynoglossus semilaevis , an economically important fish with both genetic and environmental sex determination system. The cultivation experiment was conducted employing artificially simulated seawater of 20 ppt and ambient sea water of 30 ppt to rear juveniles C. semilaevis . Throughout the experiment, the growth performance was assessed and the histology of gonadal development was examined, a significantly lower masculinization rate was observed in LS group. To gain further insights, transcriptome analysis was conducted using raw reads obtained from 53 libraries derived from gonads of 55 days post fertilization (dpf) and 100 dpf juveniles in both LS and CT groups. GO/KEGG enrichment were further proceeded, Terms and pathways involved in reproduction ability, germ cell proliferation , immune function, steroid metabolism etc., were illuminated and a possible crosstalk between HPI and HPG axis was proposed. WGCNA was conducted and two hub genes, hspb8-like and Histone H2A.V were exhibited to be of great significance in the changes of masculinization rate. Our findings provided solid reference for sex differentiation study of GSD + ESD species in a constantly changing ocean environment, as well as practice guiding significance for the environmental management for the culture of C. semilaevis .

Abstract:BackgroundThe freshwater snail Lymnaea stagnalis (L. stagnalis) has served as a successful model for studies in the field of Neuroscience. However, a serious drawback in the molecular analysis of the nervous system of L. stagnalis has been the lack of large-scale genomic or neuronal transcriptome information, thereby limiting the use of this unique model.ResultsIn this study, we report 7,712 distinct EST sequences (median length: 847 nucleotides) of a normalized L. stagnalis central nervous system (CNS) cDNA library, resulting in the largest collection of L. stagnalis neuronal transcriptome data currently available. Approximately 42% of the cDNAs can be translated into more than 100 consecutive amino acids, indicating the high quality of the library. The annotated sequences contribute 12% of the predicted transcriptome size of 20,000. Surprisingly, approximately 37% of the L. stagnalis sequences only have a tBLASTx hit in the EST library of another snail species Aplysia californica (A. californica) even using a low stringency e-value cutoff at 0.01. Using the same cutoff, approximately 67% of the cDNAs have a BLAST hit in the NCBI non-redundant protein and nucleotide sequence databases (nr and nt), suggesting that one third of the sequences may be unique to L. stagnalis. Finally, using the same cutoff (0.01), more than half of the cDNA sequences (54%) do not have a hit in nematode, fruitfly or human genome data, suggesting that the L. stagnalis transcriptome is significantly different from these species as well. The cDNA sequences are enriched in the following gene ontology functional categories: protein binding, hydrolase, transferase, and catalytic enzymes.ConclusionThis study provides novel molecular insights into the transcriptome of an important molluscan model organism. Our findings will contribute to functional analyses in neurobiology, and comparative evolutionary biology. The L. stagnalis CNS EST database is available at http://www.Lymnaea.org/.

Phylogeography of the Intertidal Limpet Lottia dorsuosa (Mollusca: Gastropoda: Lottiidae)

Abstract:We investigated geographic, genetic, and morphological variations in the intertidal limpet Lottia dorsuosa, a species with a broad distribution across Japan. A total of 98 specimens were obtained from 36 locations spanning the entire Japanese Archipelago, encompassing both the northern and southern regions. Examination of mitochondrial COI sequences revealed a distinct geographic genetic structure, delineating three prominent clades: (1) the Kuroshio Current, (2) the Tsushima Current, and (3) the Ryukyu-China groups. The observed geographical structuring between the Pacific and Sea of Japan side mirror patterns was noted in previously studied species, such as Turbo sazae and Batillaria attramentaria. Within L. dorsuosa, the Kuroshio Current group exhibited a substantial genetic distance, reaching a maximum of 12.35%, when compared to the more closely linked Tsushima Current and Ryukyu-China groups. Compared to other groups, the Ryukyu and China groups represented relatively recent differentiations, and the former group is currently facing a heightened risk of extinction owing to its sparse population density. The emergence of intraspecific geographic structures is likely attributable to the comparatively brief planktonic larval stage, which is a common characteristic of patellogastropod limpets.
Establishment of Parentage Identification Method for Sea Urchin Strongylocentrotus intermedius Based on SSR-seq Technology

Abstract:To establish a parentage identification method for Strongylocentrotus intermedius, 15 microsatellite loci and simple sequence repeat sequencing (SSR-seq) technology were used to perform SSR sequencing and typing of the validation population with known pedigree information and the simulation population. Cervus v3.0 was used for gene frequency statistics, simulated analysis, and parentage identification analysis. The results showed that, in validation population, using 15 microsatellite loci, the highest success rate of parent pairs identification was 86%, the highest success rate of female parent identification was 93%, and the highest success rate of male parent identification was 90%. The simulated population was analyzed using 12–15 loci, and the identification rate was up to 90%. In cases where accurate parentage was not achieved, individuals could exhibit genetic similarities with 1–3 male or female parents. Individuals identified as lacking a genetic relationship can be selected as parents to prevent inbreeding. This study shows that parent pairs or single parents of most offspring can be identified successfully using these 15 selected loci. The results lay a foundation for the establishment of a parentage identification method for S. intermedius.

Abstract:In 2011,seven batches of Litopenaeus vannamei,introduced from America and Singapore were collected to evaluate the growth performance.The nest design was adopted and 130 full-sib families(17 hybridized combinations and 7 inbred combinations)were obtained by artificial insemination.Offspring of the 130 families were cultured separately through the larval and juvenile stages,and were fluorescence-tagged when they reached a body length of 3 cm.The shrimps were tagged with a unique family code by injecting different colours of Visible Implant Fluorescent Elastomers(VIE)before being stocked in one grow-out pond.All families were randomly divided into two groups and then cultured in both Huanghua,Hebei and Aoshan,Qingdao,simultaneously.Body weight(g)of these shrimps was measured after 153 d.Growth performance of every population and combination was evaluated by analyzing the least squares means(LSM)and heterosis for body weight in L.vannamei.The results showed that,on the 153rd day,the variation coefficient of body weight of the L.vannamei stocks was 13%-26%.The LSM for body weight of the UA5 batch,UA4 batch and SIN batch were higher than average by 5.34%,2.51% and 1.67%,respectively.So these 3 batches could be used as parents for high quality cultivars.A growth performance analysis showed the average LSM of hybridized combination(18.14 g)was higher than the average LSM of inbred combinations(17.17 g).The highest LSM occurred in the cross between the UA4 batch and the UA5 batch,with 7.61 percent higher than average.Heterosis analysis showed,they ranged from a low of-1.77% to a high of 11.72%,with a mean of 5.45%,positive heterotic vigor was detected in most hybridized combination.The highest heterosis occurred in the cross between the UA1 batch and the UA2 batch.Our study provides insight into methods for improving the accuracy of genetic selection for growth traits in L.vannamei,and for genetics and breeding research into quantitative traits in other aquatic animals.

Abstract:Simultaneous or functional hermaphrodites possessing both ovary and testis at the same time are good materials for studying sexual development. However, previous research on sex determination and differentiation was mainly conducted in gonochoristic species and studies on simultaneous hermaphrodites are still limited. In this study, we conducted a combined morphological, endocrine and molecular study on the gonadal development of a hermaphroditic scallop Argopecten irradians aged 2–10 month old. Morphological analysis showed that sex differentiation occurred at 6 months of age. By examining the dynamic changes of progesterone, testosterone and estradiol, we found testosterone and estradiol were significantly different between the ovaries and testes almost throughout the whole process, suggesting the two hormones may be involved in scallop sex differentiation. In addition, we identified two critical sex-related genes FoxL2 and Dmrt1L, and investigated their spatiotemporal expression patterns. Results showed that FoxL2 and Dmrt1L were female- and male-biased, respectively, and mainly localized in the germ cells and follicular cells, indicating their feasibility as molecular markers for early identification of sex. Further analysis on the changes of FoxL2 and Dmrt1L expression in juveniles showed that significant sexual dimorphic expression of FoxL2 occurred at 2 months of age, earlier than that of Dmrt1L . Moreover, FoxL2 expression was significantly correlated with estradiol/testosterone ratio (E 2 /T). All these results indicated that molecular sex differentiation occurs earlier than morphological sex differentiation, and FoxL2 may be a key driver that functions through regulating sex steroid hormones in the scallop. This study will deepen our understanding of the molecular mechanism underlying sex differentiation and development in spiralians.

Abstract:<p>Largemouth bass (LMB; <i>Micropterus salmoides</i>) has been an economically important fish in North America, Europe, and China. This study obtained a chromosome‐level genome assembly of LMB using PacBio and Hi‐C sequencing. The final assembled genome is 964 Mb, with contig N50 and scaffold N50 values of 1.23 Mb and 36.48 Mb, respectively. Combining with RNA sequencing data, we annotated a total of 23,701 genes. Chromosomal assembly and syntenic analysis proved that, unlike most Perciformes with the popular haploid chromosome number of 24, LMB has only 23 chromosomes (Chr), among which the Chr1 seems to be resulted from a chromosomal fusion event. LMB is phylogenetically closely related to European seabass and spotted seabass, diverging 64.1 mya from the two seabass species. Eight gene families comprising 294 genes associated with ionic regulation were identified through positive selection, transcriptome and genome comparisons. These genes involved in iron facilitated diffusion (such as claudin, aquaporins, sodium channel protein and so on) and others related to ion active transport (such as sodium/potassium‐transporting ATPase and sodium/calcium exchanger). The claudin gene family, which is critical for regulating cell tight junctions and osmotic homeostasis, showed a significant expansion in LMB with 27 family members and 68 copies for salinity adaptation. In summary, we reported the first high‐quality LMB genome, and provided insights into the molecular mechanisms of LMB adaptation to fresh and brackish water. The chromosome‐level LMB genome will also be a valuable genomic resource for in‐depth biological and evolutionary studies, germplasm conservation and genetic breeding of LMB.</p>

Genetic sex determination in three closely related hydrothermal vent gastropods, including one species with intersex individuals

Abstract:Molluscs have a wide variety of sexual systems and have undergone many transitions from separate sexes to hermaphroditism or vice versa, which is of interest for studying the evolution of sex determination and differentiation. Following the serendipitous observation that sex was the primary driver of genetic structure in the hydrothermal vent gastropod , we investigated sexual systems and sex determination in this species and two others of the same genus. We combined genome-wide multi-locus genotypes obtained from RAD sequencing with anatomical observations of the gonads of the three species occurring in the southwest Pacific Ocean: ( =199), ( =41 ind.) and ( =246). In two of the species ( and ), the sexes are separate and genetically determined by a male-heterogametic (XY) system. External observation of the gonads in the third species ( ) also suggested that the sexes were separate, but histological analyses revealed that 76% of the individuals classified as females from external observation of the gonads presented a mosaic of male and female reproductive tissue. Empirical analyses and simulations showed nonetheless that 14 RAD loci were sex-linked with an XY signature in (as compared with 64 in and 373 in ). Comparison across species and mapping of RAD loci to a non-contiguous reference genome assembly of the related species showed that all sex-linked loci identified in are located on five scaffolds that also contain 15 and 67 sex-linked RAD loci in the other two species, respectively. These results suggest that all three species share the same XY sex determination system, but that the gonad of XX individuals are invaded by a variable proportion of male reproductive tissue. It remains to be seen whether the male tissue in these intersex individuals is functional or not. The identification of Y-specific RAD loci (found only in ) and the phylogenetic analysis of three sex-linked loci shared by all species suggested that X-Y recombination has evolved differently within each species. This situation of three species showing variation in gonadal development around a probably common sex determination system provides new insights into the reproductive mode of poorly known deep-sea species and questions the evolution of gametogenetic polymorphism in these species.

Abstract:The body-length of Lophius litulon mature female was longer than 293 mm which the age was 1.5 years old.The spawning season was from February to May,with April and May as the reproduction peak period.The GSI of female attained highest during April and then during May,while the male attained highest during May.The absolute fecundity of L.litulon was from(3.11-14.1)×105 eggs,with an average of(6.74± 2.87)×105 eggs.The relative fecundity of L.litulon(F/L) was 883-215 3 eggs/mm,with an average of 1 521±581 eggs/mm,and the relative fecundity of L.litulon(F/W) was 181-462 eggs/g,with an average of(264± 76) eggs/g.The diameters of 132 oocytes sampled from a mature female of L.litulon,it ranged from 1.32-1.76 mm,with an average of(1.59±0.19) mm and 33.3% of oocytes ranged between 1.575 to 1.625 mm.It was inferred from the egg-diameter distribution that L.litulon should be regarded as total spawning.
