Abstract:The present study aims to apply an electrostatic dry powder coating process to coat aspirin tablets with polymer blends to produce a single-layer coating film, which potentially allows an advantageous delayed sustained drug release profile. The coating formulations containing polymer blends of enteric polymer (hydroxypropyl methylcellulose phthalate or hydroxypropyl methyl cellulose acetate-succinate) and insoluble polymer (Eudragit® RL) were successfully developed and optimized. Results of scanning electron microscopy and acid resistance test demonstrated that curing conditions had a significant impact on the qualities of the produced film. Drug dissolution experiments revealed that changes in the coating level and the coating formulation with different ratios of the enteric polymer to the insoluble polymer achieved an altered and adjustable drug release profile. All of the results suggested that electrostatic powder coating technology is capable to produce a satisfactory coating film with polymer blends, allowing an advantageous functionality of delayed sustained drug release.

Abstract:In this study, complex drug-cellulose acetate (CA) composite films were designed and fabricated possessing pre-determined grid spacing for inter-connected fibrous films. Ibuprofen (IBU) was selected as the active ingredient and grid spacing was varied between 300 and 500 mu m (fiber diameter similar to 35 lm) for various geometries. Process parameter impact on fiber morphology and deposition was investigated. FTIR confirmed IBU encapsulation and XRD analysis indicated the drug was dispersed (amorphous) in films. Inter-connected grid void geometry was shown to impact water contact behavior, and drug release mechanism was shown to be Fickian diffusion. Furthermore, drug release rate depended on geometry of engineered structures. The findings suggest a spatial design approach for modulated drug release from bespoke drug delivery dosage forms. Molecular characterization of wheat germplasm using microsatellite markers.

Abstract:We investigated the genetic diversity of 63 wheat genotypes, composed of 48 accessions and 15 varieties, using 56 polymorphic simple sequence repeat primers. One hundred and eighty-six loci were found, with a mean of 131.26 alleles per locus. Cluster analysis based on microsatellite allelic diversity discriminated the accessions and varieties into different clusters; genetic diversity was the highest between variety Kohistan-97 and accession number 011512, giving a genetic similarity value of 0.4198. Accession numbers 011484 and 011356 gave a genetic similarity value of 0.9589, indicating that these accessions were 95.89% similar. We found that microsatellite markers could characterize and discriminate all of the genotypes; more primers could be used for saturation of different regions in further studies.

Abstract:THE OBJECTIVES OF THIS STUDY WERE: (i). to use blends of gastrointestinal tract (GIT)-insoluble and enteric polymers (ethyl cellulose and Eudragit L) as coating materials for multiparticulate controlled release dosage forms; (ii). to investigate the effects of the polymer blend ratio and coating level on the resulting drug release patterns; and (iii). to explain the observed phenomena based on the physicochemical properties of the systems. Propranolol HCl-loaded pellets were coated in a fluidized bed coater with organic polymer solutions; thin, drug-containing and drug-free, polymeric films were prepared using a casting knife. In vitro drug release, water uptake and dry weight loss studies were performed in 0.1 M HCl and phosphate buffer pH 7.4, respectively. The apparent drug diffusion coefficients within the polymeric systems were determined using different experimental and theoretical techniques (side-by-side diffusion cells, in vitro drug release from thin films; exact and approximate solutions of Fick's second law of diffusion). A broad range of drug release patterns from coated pellets could be achieved by varying the GIT-insoluble:enteric polymer blend ratio. With increasing relative amounts of Eudragit L, the release rates in both media significantly increased. The increase at low pH could be attributed to an increase in water uptake, as observed with thin films. Interestingly, only partial Eudragit L leaching occurred in phosphate buffer pH 7.4 even at high enteric polymer contents, indicating that the GIT-insoluble polymer effectively hindered the dissolution of the entrapped Eudragit L. At high pH, both polymer leaching and polymer swelling contributed to the control of drug release. The determined apparent drug diffusion coefficients take the two effects adequately into account.
Osmotic tablet coatings: Drying stress, mechanical properties and microstructure

Abstract:The coatings in osmotic tablets play a critical role in controlling the release of active pharmaceutical ingredient. Coatings are formed by spraying dilute polymer solution onto the tablet surface. During drying, the films develop shrinkage stress, which can cause cracking. The coatings are also subject to large tensile stress generated by osmotic pressure during the dissolution process, which may rupture the coating. Despite the role of tensile stress in causing fracture in osmotic tablet coatings, a rigorous quantification of the drying stress, mechanical properties and microstructure is missing. The present work fills this gap via detailed measurement of drying stress, Young's modulus and fracture properties of osmotic tablet coatings of cellulose acetate mixed with two different plasticisers, namely polyethylene glycol and hydroxypropyl cellulose. The measurements were complemented with imaging of the surface and the cross-section of films using scanning electron microscopy so as to relate the drying stress and mechanical properties to the microstructure of the films. The phase separation during the drying process increases the pore size, while simultaneously decreasing the modulus and the peak drying stress of the drying films. The results suggest that films with strong adhesion to the tablet surface will not rupture but if the films delaminate, the drying stress are sufficiently large to cause rupture. The detailed study presented here provides guidelines to the formulator for designing rupture-free osmotic coatings.

Abstract:Amorphous solid dispersion (ASD) in a polymer matrix is a powerful method for enhancing the solubility and bioavailability of otherwise crystalline, poorly water-soluble drugs. 6-Carboxycellulose acetate butyrate (CCAB) is a relatively new commercial cellulose derivative that was introduced for use in waterborne coating applications. As CCAB is an amphiphilic, carboxyl-containing, high glass transition temperature (Tg) polymer, characteristics essential to excellent ASD polymer performance, we chose to explore its ASD potential. Structurally diverse drugs quercetin, ibuprofen, ritonavir, loratadine, and clarithromycin were dispersed in CCAB matrices. We evaluated the ability of CCAB to create ASDs with these drugs and its ability to provide solubility enhancement and effective drug release. CCAB/drug dispersions prepared by spray drying were amorphous up to 25 wt % drug, with loratadine remaining amorphous up to 50% drug. CCAB formulations with 10% drug proved effective at providing in vitro solubility enhancement for the crystalline flavonoid drug quercetin as well as ritonavir, but not for the more soluble APIs ibuprofen and clarithromycin and the more hydrophobic loratadine. CCAB did provide slow and controlled release of ibuprofen, offering a simple and promising Long-duration ibuprofen formulation. Formulation with clarithromycin showed the ability of the polymer to protect against degradation of the drug at stomach pH. Furthermore, CCAB ASDs with both loratadine and ibuprofen could be improved by the addition of the water-soluble polymer poly(vinylpyrrolidone) (PVP), with which CCAB shows good miscibility. CCAB provided solubility enhancement in some cases, and the slower drug release exhibited by CCAB, especially in the stomach, could be especially beneficial, for example, in formulations containing known stomach irritants like ibuprofen.

Abstract:The aim of this study was to investigate the importance of the type of plasticizer in polymer blends used for the coating of solid dosage forms, comparing a lipophilic and a hydrophilic plasticizer (dibutyl sebacate (DBS) and triethyl citrate (TEC)). In vitro drug release from propranolol hydrochloride (propranolol HCl)-loaded pellets coated with blends of ethyl cellulose (EC) and Eudragit L (100:0, 75:25, 50:50, 25:75 and 0:100 w/w) was investigated at low as well as at high pH. To better understand the underlying mass transport mechanisms, the physicochemical properties of the film coatings (e.g. mechanical resistance, water uptake and dry weight loss behavior) were determined. Interestingly, drug release strongly depended on the type of plasticizer. Importantly, not only the slope but also the shape of the release curves was affected, indicating that the chemical nature of the plasticizer plays a major role for the underlying drug release mechanisms. Diffusion through the intact polymer coatings and/or through water-filled cracks was found to be dominating for the control of drug release. The relative importance of these pathways strongly depended on the polymer blend ratio and type of plasticizer. In contrast to DBS, TEC rapidly leached out of the coatings, resulting in decreasing mechanical resistances of the films and, thus, facilitated crack formation. In addition, the hydrophilicity of the plasticizer significantly affected the water uptake behavior of the film coatings and, hence, changes in the coatings' toughness and drug permeability. Also the relative affinity of the plasticizer to the different polymers was found to be of significance. In contrast to TEC, DBS has a higher affinity to EC than to Eudragit L, resulting in potential redistributions of this plasticizer within the polymeric systems and changes in the release profiles during storage. Importantly, these effects could be avoided with appropriate curing conditions and preparation techniques for the coating dispersions.

Abstract:Emamectin benzoate (EB), a widely used pesticide, is prone to decomposition by ultraviolet light and suffers from the corresponding loss of efficacy. The timed release of EB based on microspheres is one of the effective methods to solve this issue. As a non-toxic cellulose ester, cellulose acetate butyrate (CAB) is regarded as one of the best wall-forming materials for microcapsules with a good controlled release performance. Herein, two methods—mechanical activation (MA) technology and a conventional liquid phase (LP) method—were employed to synthesize different CABs, namely CAB-MA and CAB-LP, respectively. The molecular structure, rheological property, and thermal stability of these CABs were investigated. The two CABs were used to prepare microspheres for the loading and release of EB via an o/w (oil-in-water) solvent evaporation method. Moreover, the performances such as drug loading, drug entrapment, and anti-photolysis of the drug for these microspheres were studied. The results showed that both CABs were available as wall materials for loading and releasing EB. Compared with CAB-LP, CAB-MA presented a lower molecular weight and a narrower molecular weight distribution. Moreover, the MA method endowed the CAB with more ester substituent groups and less crystalline structure in comparison to the LP method, which had benefits including pelletizing and drug loading.

Abstract:Capsules have been investigated as a popular oral dosage form among communities due to their simplicity and ease of production. Capsules that are considered in the gastro-resistance category can be very beneficial due to enhanced drug absorption, improved stability, targeted release, etc. This study investigated the effect of enteric polymers on uncoated hard capsule fabrication and dissolution properties. The polymers used in this study included HPMCPh and Eudragit L100-55. Eight different formulations based on HPMCPh, Eudragit, and gelatin were examined to identify the ideal formulation for the product of uncoated enteric hard capsules with preferred physicochemical and gastro-resistance properties. The results reveal that the capsules containing Eudragit (F1), HPMCPh (F2), Eudragit/HPMCPh/gelatin (F3), and Eudragit/gelatin (F4) are steady within the simulated stomach environment, and drug release does not occur for 120 min. The outcomes demonstrate that, among the proposed formulas, the F4 formula is suitable both in terms of capsulation form and delayed-release properties and shows no microbial growth. The properties of the optimized sample were studied by FTIR, FESEM, tensile strength, humidity, and rheology. The results illustrated that gelatin-based hydrogels with Eudragit (F4) are potential candidates for manufacturing uncoated enteric hard capsules that inhibit drug release in a gastric pH medium and act as a pH-sensitive drug-release system.

Abstract:Numerous natural polymers either alone or in combination with other polymers were found effective in controlling the drug release. In this study the attempts were made to combine chitosan (degree of deacetylation 84.14 %) and as hydroxylpropyl methylcellulose (HPMC K 15M) to retard the release of aceclofenac in tablet formulation. The tablets were prepared by wet granulation and evaluated for pre and post- compression parameters. All the pre-compression parameters were found within the limit. Hardness and friability values were found in the range of 4.30-4.89 kg/cm2 and 0.1-0.6% respectively. These results proved the good mechanical strength of the formulations. The drug content was found in the range of 97.56 – 99.10 %. Weight variation was found within the official limit. The percent drug release and swelling index was found to be dependent on the concentration of polymer. With increasing the concentration of both the polymers the swelling index was increased and drug release decrease. Highest concentration of both the polymers was found to retard the drug release up to 8 h. The effect of Chitosan and HPMC on drug release was evaluated by design expert software to achieve the optimized formulation. The response of the drug release after 4h was considered to check the drug release. It was found that the enhanced concentration of both the polymers had negative effective on the drug release. The formulation containing highest concentration of the chitosan and HPMC was found be fit in the limits of optimized formulations. The optimized formulation was found to be stable at accelerated stability storage conditions.

Abstract:BACKGROUND: Colon-specific delivery of drugs can be achieved with dosage forms coated with biopolymers that are metabolized selectively by the colonic microflora and yet resistant to enzymatic digestion in the small intestine.AIM: The aim of this study was to study the influence of formulation factors on the performance of mixed films from high-amylose starches and Surelease((R)), applied using a spray-coating process, as potential colon-specific delivery devices.METHODS: 5-Aminosalicylic acid-loaded pellets were prepared by an extrusion-spheronization process and film coated with mixtures of the starches and Surelease((R)). Optimization of the coating formulation, that is, starch-to-Surelease((R)) ratio, film-coating thickness, and type of starch, was undertaken first in enzyme-free media resembling the conditions in the stomach and small intestine. The effect of curing of the film coating on the drug release profile upon storage was also evaluated. Optimized coating formulations were further assessed for enzymatic digestibility using artificial gastric and intestinal juices containing commercially available pepsin and pancreatin or alpha-amylase from hog pancreas, respectively. Finally, drug release was assessed in fluid-simulating conditions in the colon (SCF) containing Bacillus licheniformis alpha-amylase.RESULTS: Film coatings comprising high-amylose starches and Surelease((R)) in a ratio of 1:2 (w/w) and film thickness of approximately 45 microm were able to withstand the chemical and enzymatic environment of the upper gastrointestinal tract, in particular, resisted degradation by the pancreatic alpha-amylases. Stability of the coatings during storage was achieved with additional curing. In SCF, these coatings were susceptible to enzymatic degradation.CONCLUSIONS: This study showed that high amylose starch-mixed films can be successfully used as colon-specific delivery devices. The preparation of the coating dispersions described is simple and rapid, without the need to extract the amylose component of starch.

Mucoadhesive elementary osmotic pump tablets of trimetazidine for controlled drug delivery and reduced variability in oral bioavailability

Abstract:The objectives of this work was preparation and evaluation of the mucoadhesive elementary osmotic pump tablets of trimetazidine hydrochloride to achieve desired controlled release action and augmentation of oral drug absorption. The drug-loaded core tablets were prepared employing the suitable tableting excipients and coated with polymeric blend of ethyl cellulose and hydroxypropyl methylethylcellulose E5 (4:1). The prepared tablets were characterized for various quality control tests and in vitro drug release. Evaluation of drug release kinetics through model fitting suggested the Fickian mechanism of drug release, which was regulated by osmosis and diffusion as the predominant mechanism. Evaluation of mucoadhesion property using texture analyzer suggested good mucoadhesion potential of the developed osmotic systems. Solid state characterization using Fourier-transform infrared spectroscopy, differential scanning calorimetry and powder X-ray diffraction spectroscopy confirmed the absence of any physiochemical incompatibilities between drug and excipients. Scanning electron microscopy analysis showed the smooth surface appearance of the coated tablets with intact polymeric membrane without any fracture. In vivo pharmacokinetic studies in rabbits revealed 3.01-fold enhancement in the oral bioavailability vis-à-vis the marketed formulation (Vastarel MR®). These studies successfully demonstrate the bioavailability enhancement potential of the mucoadhesive elementary osmotic pumps as novel therapeutic systems for other drugs too.
Effect of lipid and cellulose based matrix former on the release of highly soluble drug from extruded/spheronized, sintered and compacted pellets

Abstract:Background: The study was to develop an extended release (ER) encapsulated and compacted pellets of Atenolol using hydrophobic (wax based and polymeric based) and high viscosity grade hydrophilic matrix formers to control the release of this highly water soluble drug by extrusion/spheronization (ES). Atenolol is used for cardiovascular diseases and available as an immediate release (IR) tablet dosage form. The lipids, Carnauba wax (CW), Glyceryl monostearate (GMS) and cellulose based i.e. Hydroxypropyl methylcellulose (HPMC) and Ethyl cellulose (EC) were used in preparing Atenolol ER pellets. Thermal sintering and compaction techniques were also applied to control the burst release of Atenolol. Method: For this purpose, thirty-six trial formulations (F1-F36) were designed by Response Surface Methodology (RSM), using Design-Expert 10 software, keeping (HPMC K4M, K15 M & K100 M), (EC 7FP, 10FP & 100FP), waxes (GMS, & CW), their combinations, sintering temperature and duration, as input variables. Dissolution studies were performed in pH, 1.2, 4.5 and 6.8 dissolution media. Drug release kinetics using different models such as zero order, first order, Korsmeyer-Peppas, Hixon Crowell, Baker-Lonsdale and Higuchi kinetics were studied with the help of DDsolver, an excel based add-in program. Results: The formulations F35 and F36 showed compliance with Korsmeyer-Peppas Super case II transport model (R2 = 0.975-0.971) in dissolution medium pH 4.5. No drug excipient interaction observed by FTIR. Stereomicroscopy showed that sintered combination pellets, (F35), were highly spherical (AR = 1.061, and sphericity = 0.943). The cross-sectional SEM magnification (at 7000X) of F34 and F35 showed dense cross-linking. The results revealed that the optimized formulations were F35 (sintered pellets) and F36 (compacted pellets) effectively controlling the drug release for 12 h. Conclusion: Extended-release encapsulated, and compacted pellets were successfully prepared after the combination of lipids CW (10%) and GMS (20%) with EC (10FP 20% & 100FP 20%). Sintering and compaction, in addition, stabilized the system and controlled the initial burst release of the drug. Extended release (ER) Atenolol is an effective alternative of IR tablets in controlling hypertension and treating other cardiovascular diseases.

Abstract:Objective The aim of this study is to investigate the effects of AMP metabolism on the physiological function of Torulopsis glabrata. Methods Strain cgade12Δade13Δ was constructed by deleting cgade12 and cgade13 with homologous recombination, and was used to study the effects of AMP metabolism on carbon metabolism by comparing the ATP levels, enzymes activity and inter-metabolite concentrations of carbon metabolism to that of ATCC55. And the effects of AMP on metabolisms on organic acid tolerance were studied by compared the cell growth and intracellular environment of cgade12Δade13Δ to that of ATCC55 under organic acid stress. Results The ATP levels of mutant cgade12Δade13Δ was decreased by 12.50% when compared with that of strain ATCC55. The enzymes activity of citrate synthetase, malate dehydrogenase, isocitrate dehydrogenase and α-ketoglutarate dehydrogenase was increased by 31.26%, 19.45%, 28.96%, 18.36% and the intracellular citric acid, α-ketoglutarate, malic acid, succinic acid contents were increased by 44.11%, 73.60%, 50.00%, 65.68%, respectively, compared with the corresponding value of strain ATCC55. However, the intracellular concentration of pyruvic acid in mutant cgade12Δade13Δ was decreased by 20.00% which led to a 73.11% reduction of pyruvic production in fermentation broth. Compared with strain ATCC55, the cell concentrations of cgade12Δade13Δ were increased by 8.71%, 11.21% and 12.71% grown in YNB with 0.4% pyruvic, 0.6% malic acid and 0.2% acetic acid, respectively. Grown in YNB with 0.2% acetic acid the H+-ATPase activity, cell membrane integrity, cell membrane electric potential of mutant cgade12Δade13Δ was increased by 7.04%, 8.71%, 25.14% than that of strain ATCC55, respectively, while the ROS concentration was decreased by 19.51%. Conclusion The deletion of genes cgade12 and cgade13 resulted in a reduction in ATP level but led to an increase in activity of TCA cycle and organic acid tolerance.

Abstract:The work investigates the effect of various formulation variables like drug-polymer ratio, stirring speed, and surfactant (Span 80) concentration on the properties of ethyl cellulose microparticles containing metformin HCl, prepared by emulsification solvent evaporation technique. The drug entrapment efficiency, particle size, and drug release behaviour of these microparticles were influenced by these formulation variables. The sustained release characteristic of these microparticles was more prominent in pH 6.8 than pH 1.2. The drug release from ethyl cellulose microparticles was found to follow the Fickian (diffusion-controlled) release mechanism. The drug-polymer interaction and surface topography of these microparticles were analyzed by FTIR spectroscopy and SEM, respectively.

Abstract:From 1982 to 1984, we conducted a prospective study to evaluate the usefulness of i.v. renal digital subtraction angiography (DSA) for living-related donor (LRD) evaluation. Twenty-eight LRDs were evaluated with the traditional approach of intravenous pyelography (IVP) and standard catheter arteriography (SCA) (group 1). During the same period, 33 LRDs underwent renal DSA and IVP from a single i.v. contrast injection (group 2). If renal arterial imaging with DSA was considered satisfactory, no further radiographic studies were done (group 2-A, n = 23). If renal arterial imaging with DSA was not satisfactory, SCA was then obtained (group 2-B, n = 10). DSA alone accurately defined the number and location of renal arteries in 21 of 23 patients from group 2-A, and in 5 of 10 patients from group 2-B. The major limitation of DSA was in patients with multiple renal arteries; accurate imaging was obtained in only 7 of these 13 patients (54%). In group 2 overall, preoperative renal imaging was not accurate in 2 of 33 patients (6%); in both cases, an unsuspected polar artery was found at nephrectomy. The mean cost per patient of all radiographic renal imaging studies was $953.00 for group 2 and $1721.00 for group 1. These data suggest that the approach of preferentially evaluating LRDs with DSA-IVP, and obtaining SCA only if DSA yields poor visualization, is more cost-effective but not as accurate as the traditional policy of obtaining SCA and IVP in all cases.
A novel controlled porosity osmotic pump system for sodium ferulate

Abstract:A controlled porosity osmotic pump (CPOP) delivery system for sodium ferulate was prepared with cellulose acetate (CA) as semipermeable membrane, polyethyleneglycol 400 (PEG 400) as channeling agent and dibutylphthalate (DBP) as plasticizer and release controller. Effects of coating levels, PEG and DBP content and amount of sodium chloride on in vitro release were studied. Coating formulations were optimized by a L9 (34) orthogonal array design (OAD) with three factors at three levels using statistical analysis. Controlled porosity osmotic pump tablets of sodium ferulate made with the optimal formulation were found to have good in vitro and in vivo release characteristics.
Impact of dry coating lactose as a brittle excipient on multi-component blend processability

Abstract:Previous work demonstrated the benefits of dry coating fine-grade microcrystalline cellulose (MCC) for enabling direct compression (DC), a favored tablet manufacturing method, due to enhanced flowability while retaining good compactability of placebo and binary blends of cohesive APIs. Here, fine brittle excipients, Pharmatose 450 (P450, 19 μ m) and Pharmatose 350 (P350, 29 μ m), having both poor flowability and compactability are dry coated with silica A200 or R972P to assess DC capability of multi-component cohesive API (coarse acetaminophen, 22 μ m, and ibuprofen50, 47 μ m) blends. Dry coated P450 and P350 not only attained excellent flowability and high bulk density but also heightened tensile strength hence processability, which contrasts with reported reduction for dry coated ductile MCC. Although hydrophobic R972P imparted better flowability, hydrophilic A200 better enhanced tensile strength, hence selected for dry coating P450 in multi-component blends that included fine Avicel PH-105. For coarse acetaminophen blends, substantial bulk density and flowability increase without any detrimental effect on tensile strength were observed; a lesser amount of dry coated P450 was better. Increased flowability, bulk density, and tensile strength, hence enhanced processability by reaching DC capability, were observed for 60 wt% ibuprofen50, using only 18 wt% of the dry coated P450, i.e. 0.18 wt% silica in the blend.
