Abstract:Azadirachta indica A. Juss. (neem), native to India, is well known worldwide for its insecticidal and ethanopharmacological properties. Although endophytic microbes are known from this plant as only leaves and stems were the subjects of past reports. Now, a variety of procedures and a number of different media were used to isolate the maximum number of endophytic fungi from unripe fruits and roots. A total of 272 isolates of 29 filamentous fungal taxa were isolated at rate of 68.0% from 400 samples of three different individual trees (at locations-Az1, Az2, Az3). Mycological agar (MCA) medium yielded the highest number of isolates (95, with a 14.50% isolation rate) with the greatest species richness. Mycelia Sterilia (1, 2, 3) accounted for 11.06%, Coelomycetes 7.25%, while Hyphomycetes showed the maximum number of representative isolates (81.69%). Mycelia-Sterilia (1, 2, 3), based on their 5.8S ITS 1, ITS2 and partial 18S and 28S rDNA sequences were identified as Fusarium solani (99%), Chaetomium globosum (93%) and Chaetomium globosum (93%) respectively. Humicola, Drechslera, Colletotrichum, and Scytalidium sp. were some of the peculiar fungal endophytes recovered from this plant.

Abstract:Abstract Nitrogen-fixing bacteria can be isolated from rhizosphere plants, and one of the N 2 -fixing bacteria is Azotobacter. One of the plants that can grow well in dry and barren soil conditions is Neem ( Azadirachta indica A. Juss)(Mimba). The purpose of this study was to determined any Azotobacter isolated from the rhizosphere of the neem plant. The type of research is an experimental study, tracing the diversity of Azotobacter in the rhizosphere of the neem plant. The study was conducted by taking soil samples from the rhizosphere of neem plants on the Bali Island. Morphological characterization based on colony observation in terms of shape, color, margin, and texture. The isolated cells were determined based on Gram staining and determination of the presence of spores. Isolates were characterized by various biochemical methods namely nitrate, reductase, citrate utilization, methyl red, VP test, catalase (method of slip-cover), and oxidase tests. The soil sample was taken from rhizospheres of 5 plants. The results showed that the N2-fixing bacteria isolated in the five soil samples were Azotobacter paspali and Azotobacter vinelandii (Azomonas) groups. Actinomycetes were found when isolating using specific media Azotobacter on plant 2.

Abstract:In our ongoing search for production improvements of bioactive secondary metabolites from marine Streptomyces through the induction of mutations using UV light, out of 145 isolates, mutant 10/14 was able to produce potent antibacterial metabolites other than the parent strain as established by chromatographic analysis. Up-scaling fermentation of mutant 10/14, followed by working up and isolation delivered five metabolites, phenazine, 1-acetyl-β -carboline, perlolyrin and erythromycin A, along with an oily substance. The latter two compounds were responsible for the antibacterial activity of the strain. In this article, we discuss with the mutation of the marine Streptomyces sp. AH2, bioactivity evaluation, fermentation and isolation of the microbial metabolites. Moreover, we study to first time in detail the 1D and 2D NMR and ESI MS data including ESI MS2 and MS3 patterns combined with HRESI MS of erythromycin A.

Abstract:Traditional medicines such as Azadirachta indica (A. indica) possess medicinal properties that should be explored in drug discovery to combat infectious diseases and antimicrobial resistance. This study provided insights into the antibacterial activities of A. indica. This narrative review utilised Google Scholar and PubMed to search for articles that were published from January 2000 to September 2023 on the antibacterial activities of A. indica. The study found that A. indica has activity against Escherichia coli, Staphylococcus aureus, Enterococcus faecalis, and other bacteria. The antibacterial activities of A. indica were reported to be due to its phytochemical content such as phenolics, tannins, saponins, flavonoids, terpenoids, fatty acids, and alkaloids. This review found that A. indica has antimicrobial activity against some Gram-negative and Gram-positive bacteria due to the presence of phytochemicals. There is a need to increase investment in drug discovery and the integration of traditional medicine into the healthcare system.

Abstract:Endophytes are microorganisms that inhabit various plant parts and cause no damage to the host plants. During the last few years, a number of novel endophytic fungi have been isolated and identified from medicinal plants and were found to be utilized as bio-stimulants and bio fertilizers. In lieu of this , the present study aims to isolate and identify endophytic fungi associated with the leaves of Anisomeles indica L. an important medicinal plant of the Terai-Duars region of West Bengal. A total of ten endophytic fungi were isolated from the leaves of A. indica and five were identified using ITS1/ITS4 sequencing based on their ability for plant growth promotion, secondary metabolite production, and extracellular enzyme production. Endophytic fungal isolates were identified as Colletotrichum yulongense Ai1, Colletotrichum cobbittiense Ai2, Colletotrichum alienum Ai2.1, Colletotrichum cobbittiense Ai3, and Fusarium equiseti . Five isolates tested positive for their plant growth promotion potential, while isolates Ai4. Ai1, Ai2, and Ai2.1 showed significant production of secondary metabolites viz. alkaloids, phenolics, flavonoids, saponins, etc. Isolate Ai2 showed maximum total phenolic concentration (25.98 mg g −1 ), while isolate Ai4 showed maximum total flavonoid concentration (20.10 mg g −1 ). Significant results were observed for the production of extracellular enzymes such as cellulases, amylases, laccases, lipases, etc. The isolates significantly influenced the seed germination percentage of tomato seedlings and augmented their growth and development under in vitro assay. The present work comprehensively tested these isolates and ascertained their huge application for the commercial utilization of these isolates both in the agricultural and industrial sectors.

Abstract:Objective: The objective of this study was to evaluate and screen the antibacterial activity of medicinal plant which were most cultivated in ancient time. Study Design: Experimental Study. Setting: Microbiology Section, Pathology Department, Sheikh Zayed Hospital/Medical College Rahim Yar Khan. Period: February 2019 to May 2019. Material & Methods: The fresh aqueous extract of garlic cloves (Allium sativum), onion bulb (Allium cepa) and neem (Azadirachta indica) leaves used against the clinically isolated Staphylococcus aureus(S.aureus), Escherichia coli (E.coli), Klebsiella, Acinetobacter and Pseudomonas species by using agar well diffusion method. Aqueous extract of used plant tested at different concentrations of 100%, 75%, 50%, 25%. Results: All these concentrations showed inhibitory effect towards Gram positive bacteria (S.aureus), while Gram negative bacteria more sensitive to higher concentration. In this analysis, on comparison we observed that fresh garlic juice extract was more efficient on all tested bacteria either Gram positive or Gram negative than other and give maximum zone of inhibition on S.aureus (40mm) due to their high chemistry. Gram negative bacteria mainly E. coli somehow resistant to onion and neem extract because they possess the ability to rapidly mutate their genetic makeup towards phytochemical compounds. Conclusion: The result of study also elicits that efficacy of fresh extract depends upon concentrations levels. The plant showed maximum zone of inhibition at 100% than at 25% because on higher level they contain higher bioactive compounds.

Abstract:Antimicrobial-resistant bacteria pose serious public health risks, necessitating bioprospecting for novel antimicrobial drugs. The endophytic fungi of the mangrove ecosystem are hotspots for discovering new bioactive chemical compounds. In this context, an investigation was designed to determine the isolation of the major endophytic fungi inhabiting the leaves of Acanthus ilicifolius, a mangrove plant with a long history of traditional use in the Chinese and Indian medical systems. Based on the morphological characterizations and molecular analysis of internal transcribed spacer rDNA sequence data, the study identified three unique endophytic fungal species, namely, Periconia macrospinosa, Coprinopsis cinerea, and Alternaria sp. Of these, P. macrospinosa was identified as the most dominant one, with the highest relative frequency (35.22%). The antibacterial activity of P. macrospinosa isolate (CMFRI/fPM-01) was evaluated by the well and diffusion method against six human pathogens, viz., Escherichia coli, Klebsiella pneumoniae, Staphylococcus aureus, Vibrio cholerae, Vibrio parahaemolyticus, and Vibrio vulnificus. The results demonstrated a high and wide spectrum of antimicrobial action of the isolate against all the tested human pathogens, with no significant difference (P > 0.05) in the activity between the pathogens. The antibacterial activity was further confirmed by determining the fungal culture supernatant's minimum inhibitory concentration and minimum bactericidal concentration. Although the studied fungi are known from other sources, this is the first report of P. macrospinosa and C. cinerea as endophytes in A. ilicifolius leaves. The outcomes also showed that the P. macrospinosa isolate could be used to discover effective antibacterial drugs against various human diseases.

Abstract:Azadirachta indica has several medicinal uses, especially its leaves. Over 4000 years ago, Ayurvedic medicine used it for its therapeutic benefits. This study examined the biological activity of Neem crude extracts and green-chemically produced Ag-NPs. TPCs and TFCs were measured for polyphenolic burden in consecutive extracts. DPPH, ABTS, and FRAP experiments measured antioxidant and antimicrobial activity against seven strains of food-borne pathogenic bacteria and eight mycotoxigenic fungi. At 1000 μg/mL, ethanolic and aqueous extracts of Neem leaves had 80.10% and 69.41% in DPPH and 71.42% and 74.61% in ABTS assays for the antioxidant activity, compared to 93.58% for BHT. At 800 μg/mL, both extracts showed antioxidant activity with 57.52 and 57.87 μM in the FRAP assay, compared to 139.97 μM for Ascorbic acid. Both extracts demonstrated antimicrobial activity with 0.02 to 0.35 mg/mL as antibacterials, 0.03 to 2.17 mg/mL as antifungals, and 0.04 to 0.42 mg/mL as antibacterials. Compared to Neem crude extract, Neem Ag-NPs had the lowest MIC values as antibacterials and antifungals at 0.05 to 0.07 mg/mL and 0.07 to 0.20 mg/mL, respectively. Neem Ag-NPs and crude extract boost antioxidant and antibacterial properties.

Abstract:Plant-beneficial microorganisms also known as endophytes colonize the inside healthy tissues of living plants and form mutualistic relationships with them. Endophytes are important components of the plant microbiome and give a variety of benefits to their hosts. Nam-Nam plant (Cynometra cauliflora), is an indigenous tree to Peninsular Malaysia with various medicinal properties. This study aimed to isolate and characterize endophytic bacteria from different parts of Nam-Nam plants such as leaves, stems, and roots. The ethyl acetate extracts from the endophytic bacteria were tested for their antibacterial activity against 7 bacterial pathogens. Plant growth promotion traits including starch hydrolysis, phosphate solubilization, nitrogen fixation, and indole-3 acetic acid (IAA) production were screened among the endophytic bacteria isolates. Molecular identification by 16S rRNA gene sequencing was performed for isolates with good antibacterial activity and plant growth promotion traits. A total of 33 endophytic bacteria comprising 27 Gram-negative and 6 Gram-positive bacteria were isolated. The antibacterial activity was demonstrated by 7 isolates in which R1L3 and TKL2 extracts exhibited significant activity against Bacillus cereus, Escherichia coli, and Proteus vulgaris. Production of IAA was exhibited by 15 isolates wherein R1S4 produced the highest IAA (20.62 μg/mL). Analysis of the 16S gene sequence revealed that RIR2, TKS2/R1L3, and R1S4/R1S5 belonged to Methylobacterium radiotolerans, Mycobacteroides abscessus, and Sphingomonas sp, respectively. The findings from this study showed that Nam-Nam plants harbored endophytic isolates with the potential to be established as a source of natural compounds that can be used to develop new anti-infection agents in the future. This is the first study to report on antibacterial activity and IAA production by endophytic bacteria from the Nam-Nam plants.
Isolation and identification of endophytic fungi from Alhagi sparsifolia Shap. and their antibacterial activity

Abstract:In order to explore the endophytic resources of Alhagi sparsifolia Shap. and identified novel antibacterial substances. Thirty endophytic fungal strains were isolated from the stems and roots of A. sparsifolia Shap. Morphological and molecular biology methods were used to identify ten strains of fungi, including four strains of Aspergillus niger, three strains of Alternaria alternata, two strains of Aspergillus flavus, and one strain of Fusarium incarnatum. All these strains were isolated from A. sparsifolia Shap. for the first time, and of these Aspergillus was the dominant genus. Antibacterial activity of the ten strains against Escherichia coli, Staphylococcus aureus, Candida albicans, and Pseudomonas aeruginosa were evaluated using the disc diffusion method. The results demonstrated that the metabolites from all the strains had inhibitory effects on at least one indicator bacterium. Notably, the endophytic fungi AFJ3 and AFG6 demonstrated strong broad-spectrum antibacterial activity, particularly against E. coli, with inhibition zones measuring 32.0 ± 0.3 and 31.3 ± 0.3 mm, respectively. The three endophytic fungi (AFG1, AFG2, and AFG3) isolated from the roots demonstrated significant antibacterial activity against P. aeruginosa forming an inhibition zone of diameter 31.3 ± 0.1, 25.6 ± 0.2, and 25.6 ± 0.2 mm, respectively. However, the strains of endophytic fungi demonstrated no significant inhibitory effects on C. albicans. Ultra-high performance liquid chromatography with quadrupole time-of-flight mass spectrometry/mass spectrometry (UPLC-QTOF-MS/MS) analysis depicted that the ethyl acetate phase of AFJ3 and AFG6 fermentation broth predominantly contained organic acids, phenolic acids, flavonoids, and fatty acids. These secondary metabolites often exhibited good antibacterial activity. This study broadens our understanding of endophytic fungi in A. sparsifolia Shap. The antibacterial activity of some strains of endophytic fungi was significant, making it worthy of further research on their active material.

Abstract:Bacterial endophytes reside within the tissues of living plant species without causing any harm or disease to their hosts. Bacterial endophytes have produced a variety of bioactive compounds that can be used for different biomedical applications. In the current study, two bacterial endophytes were isolated from healthy Moringa oleifera leaves, and identified genetically as Stenotrophomonas maltophilia and Alcaligenes faecalis. Phytochemical results illustrated that A. faecalis produced phenolics at 547.2 mg/g, tannins at 156.7 μg/g, flavonoids at 32.8 μg/g, and alkaloids at 111.2 μg/g compared to S. maltophilia, which produced phenolics at 299.5 mg/g, tannins at 78.2 μg/g, flavonoids at 12.4 μg/g, and alkaloids at 29.4 μg/g. GC-MS analysis indicated that A. faecalis extract has 24 bioactive compounds, including 9 major compounds, namely octadecanoic acid, hexadecanoic acid, linoleic acid ethyl ester, octadecenoic acid, methyl ester, methyl stearate, nonacosane, indolizine, palmitoleic acid, and heptacosane. On the other hand, S. maltophilia extract has 11 bioactive compounds, including 8 major compounds, namely oleic acid, octadecanoic acid, hexadecanoic acid, cis-2-phenyl-1, 3-dioxolane-4-methyl, ergotamine, diisooctyl phthalate, diethyl phthalate, and pentadecanoic acid. To check the safety of these extracts, the cytotoxicity of Ethyl acetate (EA) extracts of S. maltophilia and A. faecalis were evaluated against the Vero normal cell line, and the results confirmed that these extracts are safe to use. Moreover, results revealed that EA extracts of S. maltophilia and A. faecalis exhibited anticancer activity against the cancerous MCF7 cell line, where IC50 was 202.4 and 119.7 μg/mL, respectively. Furthermore, EA extracts of S. maltophilia had antibacterial and antifungal activity against Gram-positive and Gram-negative bacteria, and unicellular fungi. Likewise, the EA extract of A. faecalis exhibited antibacterial and antifungal activity against Gram-positive bacteria, as well as unicellular fungi, but did not show any activity against Gram-negative bacteria. Also, EA extracts of S. maltophilia and A. faecalis exhibited moderate antioxidant activity where IC50 were 146.2 and 147.6 μg/mL, respectively. In conclusion, the two isolated endophytic bacteria S. maltophilia and A. faecalis have promising bioactive compounds that have antibacterial, antioxidant, and anticancer activities.

Abstract:The study was conducted to assess the phytochemical and antibacterial potentials of Azadirechtin indica against Salmonella typhi, Staphylococcus aureus and Pseudomonas aeruginosa using microbiological techniques. Ethanol and water were used to extract the active components from the Azadirechtin indica leaves. Salmonella typhi showed range of inhibition zone from 11mm to 16mm at 50% to 90% concentrations of the plant ethanolic extract. Staphylococcus aureus had a range of inhibition zone from 10mm to 13mm at 50% to 90 % concentrations of the plant ethanolic extract and Pseudomonas aeruginosa had a range of inhibition zone from 10mm to 12mm at 50% to 90% concentrations of the plant ethanolic extract. Ethanol extract showed greater antibacterial activity against the test organisms when compared with the aqueous extract. Preliminary screening of phytochemicals in the leaf extract of Azadirechtin indica revealed the presence of alkaloids (7.376mg/100g), saponins (12.041 mg/100g), flavonoids (2.641mg/100g), tannins (3.431mg/100g) and cyannogenic glycosides (0.952 mg/100g). This result provides scientific support for Azadirechtin indica to be further investigated and used as drug.

Abstract:Elephantorrhiza elephantina , a wild plant in southern Africa, is utilized in traditional medicine for various ailments, leading to its endangerment and listing on the Red List of South African Plants. To date, there have been no reports on bacterial endophytes from this plant, their classes of secondary metabolites, and potential medicinal properties. This study presents (i) taxonomic characterization of bacterial endophytes in leaf and root tissues using 16S rRNA, (ii) bacterial isolation, morphological, and phylogenetic characterization, (iii) bacterial growth, metabolite extraction, and LC–MS-based metabolite fingerprinting, and (iv) antimicrobial testing of bacterial crude extracts. Next-generation sequencing yielded 693 and 2,459 DNA read counts for the rhizomes and leaves, respectively, detecting phyla including Proteobacteria, Bacteroidota, Gemmatimonadota, Actinobacteriota, Verrucomicrobiota, Dependentiae, Firmicutes, and Armatimonodata. At the genus level, Novosphingobium , Mesorhizobium , Methylobacterium , and Ralstonia were the most dominant in both leaves and rhizomes. From root tissues, four bacterial isolates were selected, and 16S rRNA-based phylogenetic characterization identified two closely related Pseudomonas sp. (strain BNWU4 and 5), Microbacterium oxydans BNWU2, and Stenotrophomonas maltophilia BNWU1. The ethyl acetate:chloroform (1:1 v/v) organic extract from each isolate exhibited antimicrobial activity against all selected bacterial pathogens. Strain BNWU5 displayed the highest activity, with minimum inhibitory concentrations ranging from 62.5 μg/mL to 250 μg/mL against diarrhoeagenic Escherichia coli , Escherichia coli O157:H7 , Salmonella enterica , antibiotic-resistant Vibrio cholerae , Staphylococcus aureus , Bacillus cereus , and Enterococcus durans . LC–MS analysis of the crude extract revealed common antimicrobial metabolites produced by all isolates, including Phenoxomethylpenicilloyl (penicilloyl V), cis-11-Eicosenamide, 3-Hydroxy-3-phenacyloxindole, and 9-Octadecenamide.

Abstract:Azadirachta indica is a plant commonly known as neem. It is native to the Indian subcontinent and most countries in Africa, including Nigeria. The aim of this study was to synthesise silver nanoparticles and examine antimicrobial activities of crude methanolic extract of Azadirachta indica leaves on clinical isolates of Staphylococcus aureus, Escherichia coli and Candida albicans. Methanol was the solvent used for the extraction. The characterization of synthesized AgNPs was carried out using the UV-Visible spectrophotometer. Antimicrobial activity was conducted using disc diffusion method while the Minimum Inhibitory Concentration (MIC), Minimum Bactericidal Concentration (MBC) and Minimum Fungicidal Concentration (MFC) were determined using broth dilution method. The antimicrobial activity of the synthesized Azadirachta indica AgNPs was carried out using standard agar well diffusion method. Qualitative phytochemicals analysis revealed presence of tannins, phenols, alkaloids, phytosteroid, and terpenoid while flavonoids, steroids and saponin were absent. Antibacterial activities of the extracts using the ditch method (concentrations of 100mg/mL, 50mg/mL, 25mg/mL, 12.5mg/mL, and 6.25mg/mL) recorded no zone of inhibition for Staphylococcus aureus and Escherichia coli. Antimicrobial activity of synthesized silver nanoparticles (AgNPs) from methanolic extract showed no zone of inhibition for Staphylococcus aureus. The zone of inhibition against Escherichia coli was 5.33mm while for Candida albicans the zone of inhibition was 15.33mm. Based on the results obtained from this study, it could be said that methanolic extract of Azadirachta indica leaves contains phytochemicals of pharmacological significance which could be exploited as antifungal agent.
Proclaiming Plant Growth-Promoting and Antifungal Properties of Pseudomonas lurida and Bacillus velezensis Isolated from Rhododendrons of Darjeeling Hills

Abstract:Endophytes have drawn attentions due to their effectiveness in providing benefits to host and non-host plants. In this study endophytic bacteria were isolated from stem and leaf samples of medicinally important plants Rhododendron griffithianum Wight and Rhododendron arboreum Smith subsp. cinnamomeum (Wall. ex G. Don) grown at higher altitudes of Darjeeling, India. Two endophytic bacteria, Pseudomonas lurida RGDS03 and Bacillus velezensis RCDL12 were identified based on 16S rRNA gene sequencing analysis. The endophytes exhibited indole acetic acid (IAA), gibberellic acid (GA), siderophore production, phosphate solubilization, nitrogen-fixing abilities, though B. velezensis RCDL12 showed superior production of IAA (126.04 ± 0.40 μg/mL), GA (241.00 ± 0.44 μg/mL), and phosphate (74.4 ± 0.41 μg/mL) solubilization as compared to P. lurida RGDS03. Purity of extracted IAA from these two endophytes was confirmed by HPLC and LC–MS analysis. In this study, P. lurida RGDS03 inhibited mycelial growth of two tested phytopathogens Phytophthora sp. and Pestalotiopsis sp. of broad host range. However, only against Pestalotiopsis sp. did B. velezensis RCDL12 exhibit antifungal activity. Study was conducted on growth promotion capabilities of isolates on rice and mung bean seedlings. P. lurida RGDS03, B. velezensis RCDL12 and consortium of both the strains reported with promising growth promotion on both rice (85–97%) and mung bean (86–99%) in terms of their seed germination, vegetative growth (root and shoot length, fresh and dry weight), and chlorophyll content as compared to the control plants (untreated). This study has emphasized growth-promoting and biocontrol activities of endophytic bacteria from rhododendrons, and application to enhance crop development for sustainable agriculture.

Abstract:To study the diversity, distribution and community structure of endophytic fungi, a medicinal plant Cinnamomum camphora (L.) Presl., was selected from ayurvedic garden of Banaras Hindu University (BHU), Varanasi, India. Differences were observed between the endophytic myco-population of young and mature tissues of leaf, stem and petiole. The leaf segments with midrib (43.93 %, Isolation Frequency) were found to have more endophytic isolates than the vein less foliar segments (21.21 %), however, the segments with lateral veins (34.84 %) are comparatively closer to the midrib segments in endophytic recovery. The mature tissues are found to harbor more endophytic diversity than the young ones. It was observed that some endophytes restrict the probable invasion of other endophytes in their respective tissues, and thus defend their tissue specificity. The endophytic microbes obtained in this study are evaluated for their antimicrobial activity, against an array of human and plant pathogens. Pestalotiopsis sp. showed significant inhibitory activity against Phytophthora cryptogea (57.7 %), Pythium aphanidermatum (54.5 %) and Microsporum nanum (51.4), while Phomopsis sp. inhibited P. aphanidermatum moderately. This preliminary work not only resolutes the understanding about plant-endophytes interactions and distribution within the host, but also provides platform for screening and isolation of novel natural antimicrobial compounds, that may be utilized in designing novel drugs for sustainable relief to the mankind.

Abstract:Pseudomonas aeruginosa is a well-known pathogen for its rapid development of multi-drug antibiotic resistance. This pathogen is responsible for numerous human diseases, particularly affecting immunocompromised and elderly patients. Hence, discovering novel therapeutics has become necessary in the fight against antimicrobial resistance. This study is focused on evaluating the potential inhibitory activity of eleven phytocompounds from Azadirachta indica against the nucleotide-binding site of the FtsZ protein of P. aeruginosa through a cheminformatics approach. FtsZ is an indispensable and highly conserved protein in prokaryotic cell division. Docking studies revealed favourable binding energies (ΔG= − 8.3 to − 5.4 kcal/mol) for all selected phytoconstituents. Finally, we selected Nimbiol (CID 11119228), as a lead compound, exhibiting a binding energy (ΔG= −7.8 kcal/mol) for the target. Based on our findings, Nimbiol shows potential as an anti-FtsZ compound, making it a promising candidate for further in vitro and in vivo investigations to assess its antimicrobial activity.
