Abstract:The high case-fatality rate--especially among young adults--during the 1918-1919 influenza pandemic is incompletely understood. Although late deaths showed bacterial pneumonia, early deaths exhibited extremely "wet," sometimes hemorrhagic lungs. The hypothesis presented herein is that aspirin contributed to the incidence and severity of viral pathology, bacterial infection, and death, because physicians of the day were unaware that the regimens (8.0-31.2 g per day) produce levels associated with hyperventilation and pulmonary edema in 33% and 3% of recipients, respectively. Recently, pulmonary edema was found at autopsy in 46% of 26 salicylate-intoxicated adults. Experimentally, salicylates increase lung fluid and protein levels and impair mucociliary clearance. In 1918, the US Surgeon General, the US Navy, and the Journal of the American Medical Association recommended use of aspirin just before the October death spike. If these recommendations were followed, and if pulmonary edema occurred in 3% of persons, a significant proportion of the deaths may be attributable to aspirin.
Metabolome and transcriptome analysis of terpene synthase genes and their putative role in floral aroma production in Litchi chinensis

Abstract:Volatile organic compounds (VOCs) are essential traits of flowers since they attract pollinators, aid in seed distribution, protect the plant from internal and external stimuli, and are involved in plant‐plant and plant‐environment interactions. Apart from their role in plants, VOCs are used in pharmaceuticals, fragrances, cosmetics, and flavourings. Litchi chinensis Sonn. is a popular fruit due to its enticing red appearance, exotic taste, and high nutritional qualities. Litchi flowers bloom as inflorescences primarily on the shoot terminals. There are three distinct flower types, two male and one female, all of which are produced on the same panicle and rely on insect pollination. Herein, we used a comprehensive metabolomic approach to examine the volatile profile of litchi fruit (green pericarp, yellow pericarp, and red pericarp) as well as male and female flowers (bud stage, half open and full bloom). From a quantitative examination of the volatiles in L. chinensis, a total of 19, 22, and 21 VOCs were discovered from female flowers, male flowers, and fruits, with the majority of them belonging to sesquiterpenes. Multivariate analysis revealed that the volatile profiles of fruits differ from those of male and female flowers. Three VOCs were unique to male flowers and ten to the fruit, while eight VOCs were shared by both male and female flowers and 11 by both male and female flowers and the fruit. Furthermore, for the first time, we identified and comprehensively studied the TERPENE SYNTHASE genes (TPS) using the litchi genome and transcriptome database, which revealed 38 TPS genes unevenly distributed across the 15 chromosomes. A phylogenetic study showed that LcTPS were grouped into TPS‐b, TPS‐c, TPS‐e, TPS‐f, and TPS‐g subfamilies, with TPS‐b having the most genes. The conserved motifs (RRX8W, NSE/DTE, and DDXXD) were studied in LcTPSs, and significant variation between subfamilies was discovered. Furthermore, after integrating the metabolome and transcriptome datasets, several VOCs were shown to be development‐specific and highly linked with distinct LcTPS genes, making them promising biomarkers. Interestingly, LcTPS17/20/23/24/31 were associated with monoterpene edges, while the rest were connected to sesquiterpene edges, indicating their probable participation in the aroma biosynthesis mechanism of certain compounds. Abstract:The 'Lilly of the Valley' species, Convallaria, is renowned for its fragrant white flowers and distinctive fresh and green floral scent, attributed to a rich composition of volatile organic compounds (VOCs). However, the molecular mechanisms underlying the biosynthesis of this floral scent remain poorly understood due to a lack of transcriptomic data. In this study, we conducted the first comparative transcriptome analysis of C. keiskei, encompassing the leaf, flower, and root tissues. Our aim was to investigate the terpene synthase (TPS) genes and differential gene expression (DEG) patterns associated with essential oil biosynthesis. Through de novo assembly, we generated a substantial number of unigenes, with the highest count in the root (146,550), followed by the flower (116,434) and the leaf (72,044). Among the identified unigenes, we focused on fifteen putative ckTPS genes, which are involved in the synthesis of mono- and sesquiterpenes, the key aromatic compounds responsible for the essential oil biosynthesis in C. keiskei. The expression of these genes was validated using quantitative PCR analysis. Both DEG and qPCR analyses revealed the presence of ckTPS genes in the flower transcriptome, responsible for the synthesis of various compounds such as geraniol, germacrene, kaurene, linalool, nerolidol, trans-ocimene and valencene. The leaf transcriptome exhibited genes related to the biosynthesis of kaurene and trans-ocimene. In the root, the identified unigenes were associated with synthesizing kaurene, trans-ocimene and valencene. Both analyses indicated that the genes involved in mono- and sesquiterpene biosynthesis are more highly expressed in the flower compared to the leaf and root. This comprehensive study provides valuable resources for future investigations aiming to unravel the essential oil-biosynthesis-related genes in the Convallaria genus.

Abstract:Many species of the Dendrobium genus are traditional Chinese herbal medicine and ornamental plants. Flower fragrance is one of the most important horticultural ornamental characters and plays a crucial role in the ecology, economy, and aesthetics of plants. However, the volatile constituents and key regulatory genes related to floral biosynthesis are poorly understood. In this experiment, the flowers from two species of Dendrobium with high-scent smells, Dendrobium moniliforme (L.) Sw. (D. moniliforme), and light-scent smells, Dendrobium nobile “H1” (D. “H1”), were selected. The aim of this study was to explore the key gene expression profiles of floral biosynthesis by combining volatile constituent determination and transcriptome analysis in two different Dendrobium species. Physiological determination results showed that 60 volatile compounds were identified in D. moniliforme and 52 volatile compounds were identified in D. ‘H1’ flowers in four flowering stages, and the full bloom stage was the most complicated stage because there were 41 and 33 volatile compounds, respectively. These compounds belong to terpenes, aromatics, fatty acids, nitrogenous compounds, ketones, alcohols, and alkanes, respectively. The components identified in the gynandrium and petals revealed that the petals were probably the most important sites affecting the release of volatiles. The relative content of terpene compounds was the highest, with 77.25% (D. moniliforme) and 50.38% (D. “H1”), respectively. Transcriptome analysis showed that differentially expressed genes (DEGs) were highly enriched in terpenoid backbone biosynthesis and that four linalool synthetase (LIS) genes were up-regulated in high-scent smell species. This study is helpful to explore the key genes of flower fragrance and provides a theoretical basis for further understanding of the regulatory molecular functions of floral synthesis and release, as well as for the cultivation of new aromatic species.

Abstract:Chrysanthemum indicum var. aromaticum (CIA) is an endemic plant that occurs only in the high mountain areas of the Shennongjia Forest District in China. The whole plant, in particular the flowers of CIA, have intense fragrance, making it a novel resource plant for agricultural, medicinal, and industrial applications. However, the volatile metabolite emissions in relation to CIA flower development and the molecular mechanisms underlying the generation of floral scent remain poorly understood. Here, integrative metabolome and transcriptome analyses were performed to investigate floral scent-related volatile compounds and genes in CIA flowers at three different developmental stages. A total of 370 volatile metabolites, mainly terpenoids and esters, were identified, of which 89 key differential metabolites exhibited variable emitting profiles during flower development. Transcriptome analysis further identified 8,945 differentially expressed genes (DEGs) between these samples derived from different flower developmental stages and KEGG enrichment analyses showed that 45, 93, and 101 candidate DEGs associated with the biosynthesis of phenylpropanoids, esters, and terpenes, respectively. Interestingly, significant DEGs involved into the volatile terpenes are only present in the MEP and its downstream pathways, including those genes encoding ISPE, ISPG, FPPS, GPPS, GERD, ND and TPS14 enzymes. Further analysis showed that 20 transcription factors from MYB, bHLH, AP2/EFR, and WRKY families were potentially key regulators affecting the expressions of floral scent-related genes during the CIA flower development. These findings provide insights into the molecular basis of plant floral scent metabolite biosynthesis and serve as an important data resources for molecular breeding and utilization of CIA plants in the future.

Abstract:Summary Cananga odorata is known as a natural perfume tree of the Annonaceae family in Magnoliales. However, its phylogenetic position and the molecular mechanisms involved in the biosynthesis of the floral volatile organic compounds (VOCs) remain unclear. Here, by combining a variety of sequencing platforms, we present a telomere‐to‐telomere (T2T) genome of C. odorata with 735.83 Mb, which represents the highest integrity and assembly quality of genome in magnoliid plants reported to date. Phylogenetic analysis based on multiple datasets and approaches showed that C. odorata, as a member of magnoliids, is sister to eudicots, after their divergence from monocots. Metabolomic of VOCs in the essential oil and flowers scent showed that sesquiterpenes, especially β‐caryophyllene, were the major compounds. Two CoTPS21 homologues derived from tandem duplication events were highly expressed during flower development and were identified as the key sesquiterpene synthases for the production of β‐caryophyllene. In addition, CoSPL3 and CoSPL9 were considered as potential transcription factors for activating the expression of CoTPS21 homologues. Our results shed light on the molecular mechanisms underlying the biosynthesis of the unique floral fragrance in C. odorata and provide new insights into the phylogenetic position of magnoliids.

Abstract:Flower fragrance is one of the traits that holds important economical values in flowering plants. Extensive attention has converged on fragrance preservation in flower cultivation and breeding. Dendrobium loddigesii is an important species for cultivating aromatic Dendrobium orchid varieties for the long term due to its fragrance. Few studies focus on exploring related genes responsible for the aroma components in D. loddigesii. We analyzed flowers from two aromatic D. loddigesii varieties using high-throughput RNA sequencing and gas chromatography-mass spectrometry (GC-MS). The metabolomics results showed that the main volatile compounds responsible for the aroma formation of D. loddigesii were terpenes, especially monoterpenes. The de novo transcriptome assembly comprised 175,089 unigenes, and 24,570 unigenes of the genes were identified as differential expressed genes (DEGs) between the two varieties. Among these DEGs, 525 genes were mapped into seven pathways that related to the floral scent synthesis. Seventeen key genes were significantly correlated with volatile aroma metabolites, including geraniol, α-pinene, eugenol, and (Z)-3-hexenal. These results provide references for understanding the aroma biosynthesis and perfume formulations of D. loddigesii.
Identification and functional analysis of terpene synthases revealing the secrets of aroma formation in Chrysanthemum aromaticum

Abstract:C. aromaticum is widely cultivated for its aromatic, medicinal, and tea-applicable properties, earning the nickname 'lavender in composite'. Terpenoids are the major compounds of C. aromaticum fragrance. To reveal the molecular mechanisms of terpenoid biosynthesis in C. aromaticum, NGS and SMRT sequencing were employed to identify the key terpene synthase genes. A total of 59,903 non-redundant transcripts were obtained by the transcriptome analysis. Twenty-nine terpene synthase genes (TPSs) were identified, and phylogenetic analysis showed that they belong to four subfamilies of terpene synthases. Five CaTPSs were successfully cloned. Subcellular localization showed they were present in the nucleus and cytosol. Structure models of five terpene synthases were predicted, and molecular docking results showed good binding affinities with FPP/GPP. In vitro enzymatic tests showed that CaTPS7, CaTPS8, CaTPS10 and CaTPS20 could catalyze substrates to produce terpenoids. CaTPS7 and CaTPS20 were both able to effectively convert the precursor FPP into caryophyllene. CaTPS8 could convert FPP to trans-nerolidol and nerolidyl acetate, while CaTPS10 could convert FPP to elemene and aristolochene. This study lays the groundwork for further research to depict the metabolism network of terpenoid in C. aromaticum. These identical terpene synthase genes could be introduced into the cultivated chrysanthemums to enhance their fragrance.
De novo assembly and comparative transcriptome analysis: novel insights into sesquiterpenoid biosynthesis in Matricaria chamomilla L.

Abstract:Matricaria chamomilla , originally native to the temperate zone of Eurasia, has been one of the most popular herbal plants owing to its pharmacologically active sesquiterpenoid metabolites, such as (-)-α-bisabolol and bisabolol oxide A. Although the sesquiterpenoids of M. chamomilla has been well characterized, information on genes responsible for the biosynthesis of these compounds are scarce, and further investigations on the plant have been hindered by the lack of genomic information. In this study, we performed a de novo assembly of the M. chamomilla transcriptome using Illumina HiSeq 2500 high-throughput sequencing. A single run yielded 21 Gb clean reads that were assembled into 83,741 unigenes. A total of 21,626 and 18,929 unigenes were assigned to Gene Ontology categories and the Cluster of Orthologous Groups, respectively. Differentially expressed genes (DEGs) in flowers, stems, roots, and leaves were screened, and 1115 DEG unigenes were assigned to specific secondary metabolites using the Kyoto Encyclopedia of Genes and Genomes (KEGG). We selected a set of putative genes that were most likely involved in the biosynthesis of sesquiterpenoid in M. chamomilla . The genes in the sesquiterpenoid synthesis pathway displayed significant differential expression in different organs. Out of the 83,741 unigenes, 5555 were identified as having simple sequence repeat motifs, with mono-nucleotide repeats being the most frequently found repeat type. Our data provided a comprehensive sequence resource for M. chamomilla belonging to the Asteraceae family, which contains many medicinal herbs. The present mass sequence data are expected to facilitate further study on the regulatory mechanism of the terpenoid biosynthetic pathway in M. chamomilla .
Identification and Validation of Magnolol Biosynthesis Genes in Magnolia officinalis

Abstract:Bacterial infections pose a significant risk to human health. Magnolol, derived from Magnolia officinalis, exhibits potent antibacterial properties. Synthetic biology offers a promising approach to manufacture such natural compounds. However, the plant-based biosynthesis of magnolol remains obscure, and the lack of identification of critical genes hampers its synthetic production. In this study, we have proposed a one-step conversion of magnolol from chavicol using laccase. After leveraging 20 transcriptomes from diverse parts of M. officinalis, transcripts were assembled, enriching genome annotation. Upon integrating this dataset with current genomic information, we could identify 30 laccase enzymes. From two potential gene clusters associated with magnolol production, highly expressed genes were subjected to functional analysis. In vitro experiments confirmed MoLAC14 as a pivotal enzyme in magnolol synthesis. Improvements in the thermal stability of MoLAC14 were achieved through selective mutations, where E345P, G377P, H347F, E346C, and E346F notably enhanced stability. By conducting alanine scanning, the essential residues in MoLAC14 were identified, and the L532A mutation further boosted magnolol production to an unprecedented level of 148.83 mg/L. Our findings not only elucidated the key enzymes for chavicol to magnolol conversion, but also laid the groundwork for synthetic biology-driven magnolol production, thereby providing valuable insights into M. officinalis biology and comparative plant science.
Volatile secondary metabolome and transcriptome analysis reveals distinct regulation mechanism of aroma biosynthesis in Syringa oblata and S. vulgaris

Abstract:Lilacs have high ornamental value due to their strong aroma. However, the molecular regulatory mechanisms of aroma biosynthesis and metabolism in lilac were largely unclear. In this study, two varieties with distinct aroma, Syringa oblata 'Zi Kui' (faint aroma) and Syringa vulgaris 'Li Fei' (strong aroma), were used for exploring the regulation mechanism of aroma difference. Via GC-MS analysis, a total of 43 volatile components were identified. Terpene volatiles was the most abundant volatiles constituting the aroma of two varieties. Notably, 3 volatile secondary metabolites were unique in 'Zi Kui' and 30 volatile secondary metabolites were unique in 'Li Fei'. Then, a transcriptome analysis was performed to clarify the regulation mechanism of aroma metabolism difference between these two varieties, and identified 6411 differentially expressed genes (DEGs). Interestingly, ubiquinone and other terpenoid-quinone biosynthesis genes were significantly enriched in DEGs. We further conducted a correlation analysis between the volatile metabolome and transcriptome and found that TPS, GGPPS, and HMGS genes might be the key contributors to the differences in floral fragrance composition between the two lilac varieties. Our study improves the understanding in the regulation mechanism of Lilac aroma and would help improve the aroma of ornamental crops by metabolic engineering.

[Effectiveness of Formica rufa and autologous blood injection in patients with ankylosing spondylitis: a double-blind randomized study].

Abstract:While complementary medicine and homeopathy is becoming an increasingly prominent part of the health care practices, there is a lack of controlled studies concerning their effectiveness. In our study, we wanted to answer the question whether a combination of Formica rufa D6 and reinjection of the patient's own blood is superior to injection of placebo. 104 patients with ankylosing spondylitis entered a prospective, randomized, double-blind study. During four weeks they received twice weekly either 1 ml of Formica rufa D6 in combination with 0.5 ml of blood or 1.5 ml NaCl intramuscular. Before and after therapy, mobility, thoracic excursion and doctor's overall assessment were measured in addition to patient's health status, using a German version of the Arthritis Impact Measurement Scales, before and after therapy as well as four, twelve, and twenty-four weeks later. We were not able to detect any statistical difference between treatment and placebo group in any of the parameters measured. Therefore, the therapy under study may not be regarded effective.
Analysis of terpenoids and their gene regulatory networks on the basis of the transcriptome and metabolome of Opisthopappus longilobus

Abstract:Opisthopappus longilobus , which is a unique wild plant resource in China, produces leaves and flowers with distinct aromas. However, there have been relatively few molecular studies on its floral aroma, which has hindered the research on this plant species at the molecular level and the breeding of novel varieties. In this study, transcriptome and metabolome analyses were performed using O. longilobus leaves, buds, and inflorescences at the exposure, initial opening, and blooming stages. Using high-quality reads and assembly software, a total of 45,674 unigenes were annotated according to the Nr, Swiss-Prot, KOG, and KEGG databases. Additionally, a GC-MS system and a self-built database were used to detect 1,371 metabolites in the leaves, buds, and inflorescences. Terpene metabolites were the most common compounds (308 in total). We analyzed the gene network regulating terpenoid accumulation in O. longilobus and identified 56 candidate genes related to terpenoid synthesis. The expression of OlPMK2 , OlMVK1 , OlTPS1 , and OlTPS3 may lead to the accumulation of 11 different terpenoids specifically in the inflorescences at the exposure, initial opening, and blooming stages. The generated data may be useful for future research on O. longilobus genetic resources and the molecular mechanism regulating aroma formation in this plant species. The findings of this study may be used to accelerate the breeding of new O. longilobus varieties with enhanced aromatic traits.

Abstract:The fascinating scent of rose (Rosa genus) flowers has captivated human senses for centuries, making them one of the most popular and widely used floral fragrances. Despite much progress over the last decade, many biochemical pathways responsible for rose scents remain unclear. We analyzed the floral scent compositions from various rose varieties and selected the modern cultivar Rosa hybrida 'Double Delight' as a model system to unravel the formation of rose dominant volatile terpenes, which contribute substantially to the rose fragrance. Key genes involved in rose terpene biosynthesis were functionally characterized. Cytosolic geranyl diphosphate (GPP) generated by geranyl/farnesyl diphosphate synthase (G/FPPS1) catalysis, played a pivotal role in rose scent production, and terpene synthases (TPSs) in roses play an important role in the formation of most volatile terpenes, but not for geraniol, citral or β-citronellol. Subsequently, a series of enzymes, including geraniol dehydrogenase (GeDH), geranial reductase (GER), 12-oxophytodienoate reductase (OPR) and citronellal reductase (CAR), were characterized as involved in the transformation of geraniol to β-citronellol in roses through three successive steps. Interestingly, the β-citronellol biosynthesis pathway appears to be conserved in other horticultural plants like Lagerstroemia caudata and Paeonia lactiflora. Our findings provide valuable insights into the biosynthesis of rose volatile terpenoid compounds and offer essential gene resources for future breeding and molecular modification efforts.

Abstract:BackgroundEssential oils (EOs) of Lavandula angustifolia, mainly consist of monoterpenoids and sesquiterpenoids, are of great commercial value. The multi-flower spiciform thyrse of lavender not only determines the output of EOs but also reflects an environmental adaption strategy. With the flower development and blossom in turn, the fluctuation of the volatile terpenoids displayed a regular change at each axis. However, the molecular mechanism underlying the regulation of volatile terpenoids during the process of flowering is poorly understood in lavender. Here, we combine metabolite and RNA-Seq analyses of flowers of five developmental stages at first- and second-axis (FFDSFSA) and initial flower bud (FB0) to discover the active terpenoid biosynthesis as well as flowering-related genes.ResultsA total of 56 mono- and sesquiterpenoids were identified in the EOs of L. angustifolia ‘JX-2’. FB0’ EO consists of 55 compounds and the two highest compounds, β-trans-ocimene (20.57%) and (+)-R-limonene (17.00%), can get rid of 74.71 and 78.41% aphids in Y-tube olfactometer experiments, respectively. With sequential and successive blossoms, temporally regulated volatiles were linked to pollinator attraction in field and olfaction bioassays. In three characteristic compounds of FFDSFSA’ EOs, linalyl acetate (72.73%) and lavandulyl acetate (72.09%) attracted more bees than linalool (45.35%). Many transcripts related to flowering time and volatile terpenoid metabolism expressed differently during the flower development. Similar metabolic and transcriptomic profiles were observed when florets from the two axes were maintained at the same maturity grade. Besides both compounds and differentially expressed genes were rich in FB0, most volatile compounds were significantly correlated with FB0-specific gene module. Most key regulators related to flowering and terpenoid metabolism were interconnected in the subnetwork of FB0-specific module, suggesting the cross-talk between the two biological processes to some degree.ConclusionsCharacteristic compounds and gene expression profile of FB0 exhibit ecological value in pest control. The precise control of each-axis flowering and regular emissions at transcriptional and metabolic level are important to pollinators attraction for lavender. Our study sheds new light on lavender maximizes its fitness from “gene-volatile terpenoid-insect” three layers.

Genome-Wide Characterization of Differentially Expressed Scent Genes in the MEP Control Network of the Flower of Lilium 'Sorbonne'

Abstract:Fragrance is an important feature of ornamental lilies. Components of volatile substances and important genes for monoterpene synthesis in the 2-C-methyl- d -erythritol 4-phosphate (MEP) pathway were examined in this study. Twenty volatile compounds (2 in the budding stage, 3 in the initial flowering stage, 7 in the semi-flowering stage, 17 in the full-flowering stage, and 5 in withering stage) were detected in the Oriental lily 'Sorbonne' using gas chromatography–mass spectrometry. The semi- and full-flowering stages were key periods for volatile substance production and enzyme function. Sequence assembly from samples collected during all flowering stages resulted in the detection of 274,849 genes and 129,017 transcripts. RNA sequencing and heatmapping led to the detection of genes in the MEP monoterpene metabolism pathway. Through gene ontology and Kyoto Encyclopedia of Genes and Genomes pathway analysis, we extracted key genes ( LiDXS2 , LiLIS , and LiMYS ) and transcription factors (in the bHLH , MYB , HD-ZIP , and NAC families) associated with the MEP pathway. Tissue localization revealed that LiDXS2 , LiLIS , and LiMYS were expressed in Lilium 'Sorbonne' petals in the full-flowering stage. Genes regulating the 1-deoxy- d -X-lignone-5-phosphate synthase family of rate-limiting enzymes, involved in the first step of monoterpene synthesis, showed high expression in the semi- and full-flowering stages. LiDXS2 was cloned and localized in chloroplast subcells. The relative expression of terpene-related genes in the MEP and mevalonic acid pathways of wild-type and LiLIS/LiMYS transgenic Arabidopsis thaliana , and changes in chemical composition, confirmed that LiLIS/LiMYS regulates the monoterpene synthesis pathway. The results of this study provide a theoretical basis for the synthesis of lily aromatic substances and the cultivation of new garden flower varieties.

Abstract:Abstract Linalool and ionone are two important aromatic components in sweet osmanthus petals, and the regulatory mechanisms produce these two components remain unclear. In this study, we employed whole-genome methylation sequencing and ATAC-seq technology to analyze the genomic DNA methylation status and chromatin accessibility of sweet osmanthus cultivars “Zaohuang” and “Chenghong Dangui”. Results showed that the promoter region of TPS2, a key gene in the linalool synthesis pathway, was less methylated in “Chenghong Dangui” than in “Zaohuang”. The chromatin was more accessible in “Chenghong Dangui” than in the “Zaohuang” cultivar, which resulted in a much stronger expression of this gene in “Chenghong Dangui” than in “Zaohuang”. This eventually led to a high quantity of linalool and its oxides in the petals of “Chenghong Dangui”, but there were lower levels present in the petals of “Zaohuang”. These results suggest that DNA methylation and chromatin accessibility play major roles in linalool synthesis in sweet osmanthus. The methylation level of the promoter region of CCD4, a key gene for ionone synthesis, was higher in “Zaohuang” than that in “Chenghong Dangui”. The chromatin accessibility was lower in “Zaohuang” than in “Chenghong Dangui”, although the expression of this gene was significantly higher in “Zaohuang” than in “Chenghong Dangui”. ChIP-seq analysis and a series of experiments showed that the differential expression of CCD4 and CCD1 in the two cultivars may predominantly be the result of regulation by ERF2 and other transcription factors. However, a 183 bp deletion involving the CCD4 promoter region in “Chenghong Dangui” may be the main reason for the low expression of this gene in the petals of “Chenghong Dangui”. This study provides an important theoretical basis for improving selective breeding of key floral fragrance components in sweet osmanthus.

Abstract:Dendrobium officinale is a kind of traditional Chinese herbal medicine. Its flowers could be used as health care tea for its aroma flavor and medicinal value. Most recent studies demonstrated that terpenoids are the main components of the aromatic compounds in the flowers, but the biosynthesis of terpenoids is poorly understood in D. officinale . In the experiment, the flowers from two cultivars of D. officinale with different smells were collected. The transcriptome analysis and combined volatile terpenoids determination were performed to identify the genes related to the biosynthesis of the terpenoids. The results showed that the different products of volatile terpenoids are α-thujene, linalool, α-terpineol, α-phellandrene, γ-muurolene, α-patchoulene, and δ-elemene in two cultivar flowers. The transcriptome analysis detected 25,484 genes in the flowers. And 18,650 differentially expressed genes were identified between the two cultivars. Of these genes, 253 genes were mapped to the terpenoid metabolism pathway. Among these genes, 13 terpene synthase (TPS) genes may have correlations with AP2/ERF, WRKY, MYB, bHLH, and bZIP transcription factors by weighted gene co-expression network analysis (WGCNA). The transcription factors have regulatory effects on TPS genes. These results may provide ideas for the terpenoid biosynthesis and regulatory network of D. officinale flowers.
