Abstract:In 1967 we witnessed an important step towards the purification of neurophysin proteins when Dean et al.’ showed that acid extraction of acetonedried posterior pituitary powder at pH 1.9 (instead of a pH near 4) prevented proteolysis of neurophysins during isolation. Since that time the recommended procedure of isolation has been acid extraction of the powder at low pH, precipitation of crude neurohypophyseal hormone-neurophysin complexes, followed by removal of hormone(s) and fractionation of the neurophysin-like proteins by gel filtration? and, more recently, by an additional step of ionexchange ch r~matography .~*~ While neurophysin preparations thus obtained are adequate for many studies, there has clearly been in recent years a need for neurophysins with a high degree of purity which cannot always be achieved by chromatographic methods alone. The demand for highly purified neurophysin is particularly important when one wishes to determine whether this class of proteins possesses intrinsic “hormonal” activity; neurophysin preparations have been reported to exhibit lipolytic a~ t iv i ty ,~” positive chronotropic effect^,^ and natriuretic activity. l o p l l Furthermore, the noncovalent complexes between neurohypophyseal hormones and neurophysin proteins are a very interesting model for the examination of peptide-protein interactions; the nagging question of whether in solution the proteins have 0ne12913 or moreI4-l7 specific neurohypophyseal hormonebinding sites as well as the accurate determination of appropriate affinity constants may only be resolved with the help of highly purified neurophysin proteins. Moreover, highly purified neurophysins set a favorable stage for the production of specific antibodies with no or minimal crossreactivity. Bovine neurophysin I and I1 (BNP-I and -11) obtained by chromatographic procedures proved inadequate in our hands for determination of their amino acid sequences18 (also Schlesinger, see for citation Reference 19). In this context it is little wonder that researchers began to apply more sophisticated separation techniques to the isolation of neurophysin proteins, such as affinity chromatography,20 isoelectric and polyacrylamide gel electroph~resis.’~~~~-~~ This study illustrates the fractionation of a mixture of bovine neurophysins-I, -C and -11 obtained by gel filtration on Sephadex G 7 5
