Abstract:Concern is increasing regarding the environmental impact of the high usage rate and intensive release of antibiotics used for human and animal therapy in major urban areas of China. In the present study, regional environmental distribution simulations and risk assessments for 3 commonly used fluoroquinolones in the Wenyu River catchment were conducted using a typical catchment model widely used in Europe. The fluoroquinolone antibiotics investigated (ofloxacin, norfloxacin, and ciprofloxacin) are consumed at high levels for personal health care in China. These antibiotics were simulated in the aquatic environment of the Wenyu River catchment across the Beijing City area for annual average concentrations, with regional predicted environmental concentrations (PECs) of approximately 711ng/L, 55.3ng/L, and 22.2ng/L and local PECs up to 1.8 mu g/L, 116ng/L, and 43ng/L, respectively. Apart from hydrological conditions, the concentrations of fluoroquinolones were associated closely with the sewage treatment plants (STPs) and their serving population, as well as hospital distributions. The presence of these fluoroquinolones in the catchment area of the present study showed significant characteristics of the occurrence of pharmaceuticals in the aquatic environment in an urban river, with typical down-the-drain chemicals. Significantly high concentrations of specific antibiotics indicated non-negligible risks caused by the intensive use in the local aquatic environment in a metropolitan area, particularly ofloxacin in upstream Shahe Reservoir, middle stream and downstream Qing River, and Liangma River to the Ba River segment. Abstract:Abstract The residual antibiotics in the environment have lately caused widespread concerns. However, little information is available on the antibiotic bioaccumulation and its health risk in drinking water resources of South China. Therefore, the occurrence, distribution, and health risk of four quinolone antibiotics including ofloxacin (OFX), norfloxacin (NOR), ciprofloxacin (CIP), and enrofloxacin (ENR) in the Qingshitan reservoir using high-performance liquid chromatography were investigated. Results revealed that the concentrations in water, sediment, and edible fish ranged from 3.49–660.13 ng/L, 1.03–722.18 μg/kg, and 6.73–968.66 μg/kg, respectively. The ecological risk assessment via the risk quotient (RQ) method showed that the values in sediment were all greater than 1, posing a high risk to the environment. The health risk index of water samples was at the maximum acceptable level, with OFX at the top while the rest were at the medium risk level. The main edible fish kinds of the reservoir had high dietary safety and the highest contaminations were found in carnivorous feeding habits and demersal habitat fishes with OFX as the highest magnitude. Source identification and correlation analysis using SPSS showed significant relationships between NOR with pH and turbidity (in water), as well as total phosphor (TP) and total organic carbon (TOC) in sediment. NOR was the highest in sediment which mostly sourced from livestock wastewater, croplands irrigation drain water, and stormwater. Correlations between CIP and ENR with TP were significant, while OFX was positively associated with total nitrogen (TN) which mainly originated from urban sewage as well as directly dosed drugs in fish farms. In conclusion, our results are of great significance for ensuring the safety of drinking water and aquatic products in this region.

Abstract:Five categories of antibiotics viz., nineteen fluoroquinolones, sixteen sulfonamides, four tetracyclines, three chloramphenicols, four nitroimidazoles and three of their metabolites were measured in 72 water samples from the river near the livestock and poultry farms in Zhejiang province of China.48.6 % water sample was positive with at least one drug residue.The detection frequency ranged from 8.3 % for fluoroquinolones to 37.5 % for chloramphenicols.For all the target veterinary drugs, florfenicol had the highest occurrence of 31.9 % with the mean concentration of 484.3 ng L -1 .Based on the calculation of hazard quotients, the risk to the aquatic organisms was evaluated and it was demonstrated that the occurrence of sulfamethoxazole, sulfamonomethoxine and florfenicol might pose a risk to aquatic organisms in some study areas.
Occurrence and risk assessment of quinolones and sulfonamides in freshwater aquaculture ponds in Northeast Zhejiang, China

Abstract:Antibiotics play an essential role in the aquaculture industry, but their overuse and weak degradability inevitably lead to light to severe residues in natural and aquaculture environments. Most studies were interested in the occurrence, distribution, and ecological risks of a limited number of antibiotics in natural environments (rivers, lakes, and coastal regions) with a minor focus on antibiotic presence in either water, sediments, or organisms in aquaculture environments located in specific regions. In this study, we conducted a comprehensive investigation into the occurrence and distribution of up to 32 antibiotics [including 15 quinolones (QNs) and 17 sulfonamides (SAs)] in organisms and their corresponding environmental matrices from 26 freshwater aquaculture ponds in Northeast Zhejiang, China. A total of 13, 9, 7, and 7 antibiotics were detected in pond water, sediments, feeds, and aquaculture organisms, respectively, with concentration ranges of 0.6-92.2 ng/L, 0.4-1169.3 ng/g dw, <LOD-17.5 ng/g dw, and < LOD-234.3 ng/g ww, respectively, where enrofloxacin (ENR) was the main component in different matrix samples, with a proportion range of 51 %-88 %. There were differences in the predominant antibiotic and concentration abundance among fishponds, shrimp ponds, crab ponds, and turtle ponds. Feed was likely one of the most essential source of antibiotics in freshwater aquaculture ponds. The risk quotient (RQ) results revealed that ciprofloxacin (CIP) and ENR showed high risks to algae in water at the maximum measured concentrations and norfloxacin (NOR), CIP, ENR, and ofloxacin (OFL) exhibited high risks in sediments. However, the joint probability curve (JPC) showed acceptable risk in water. On the basis of the calculated hazard quotients (HQ < 1), the risk of human antibiotic ingestion through aquaculture organisms was acceptable. These results suggest that the rational control of antibiotic use is necessary for human health and ecological security.

Abstract:Antibiotic residues are being highlighted around the world as emerging concerns in aquatic environments. Here, the occurrence of antibiotics in tributaries of the Xiaoqing River, China, as well as the spatial and temporal distributions, were reported. The total concentrations of 19 antibiotics in surface water and sediments ranged from 8.20–809.07 ng L–1 and from 39.57– 2,151.64 μg kg–1 dry weight (dw), respectively. Among all detected antibiotics, ofloxacin showed the highest concentration and detection frequency in both surface water (283.83 ng L–1, 100%) and sediment (1,827.18 μg kg–1 dw, 100%). The pollution levels of antibiotics in different seasons were in the following order: dry season > normal season > wet season, except for clarithromycin, sulfadiazine, sulfamerazine and sulfaquinoxaline. Domestic, aquaculture and livestock waste-water might be the main pollution sources of antibiotics in the tributaries of the Xiaoqing River. Moreover, the Pearson correlation coefficient showed significantly positive correlations between antibiotics and heavy metals (such as Cu, Zn and Pb), indicating that there may be a combination of contaminants. The environmental risk assessment indicated a high environmental risk of ofloxacin, clarithromycin, roxithromycin and sulfamethoxazole.

Abstract:A sensitive liquid chromatography-fluorescence detection method, combined with one-step solid-phase extraction, was established for detecting the residual levels of the four typical fluoroquinolone antibiotics (ofloxacin, norfloxacin, ciprofloxacin, and enrofloxacin) in influent, effluent, and surface waters from Hangzhou, China. For the various environmental water matrices, the overall recoveries were from 76.8 to 122%, and no obvious interferences of matrix effect were observed. The limit of quantitation of this method was estimated to be 17 ng/L for ciprofloxacin and norfloxacin, 20 ng/L for ofloxacin, and 27 ng/L for enrofloxacin. All of the four typical fluoroquinolone antibiotics were found in the wastewaters and surface waters. The residual contents of the four typical fluoroquinolone antibiotics in influent, effluent, and surface water samples are 108-1405, 54-429, and 7.0-51.6 ng/L, respectively. The removal rates of the selected fluoroquinolone antibiotics were 69.5 (ofloxacin), 61.3 (norfloxacin), and 50% (enrofloxacin), indicating that activated sludge treatment is effective except for ciprofloxacin and necessary to remove these fluoroquinolone antibiotics in municipal sewage. The risk to the aquatic environment was estimated by a ratio of measured environmental concentration and predicted no-effect concentration. At the concentrations, these fluoroquinolone antibiotics were found in influent, effluent, and surface waters, and they should not pose a risk for the aquatic environment.

Abstract:The presence of thirteen antibiotics categorized into four different groups (fluoroquinolones, macrolides, sulfonamides and trimethoprim) was investigated in the rivers discharging to the Laizhou Bay and the seawater of the bay, and the impacts of river discharge on the marine environment were assessed. The results revealed that the same antibiotics predominated in both the river water and the seawater. Additionally, the detected antibiotics in the river water were generally higher than those in the inner bay and in the open bay, reflecting the importance of the riverine inputs as a source of antibiotics. Risk assessment based on the calculated risk quotients (RQ) showed that enoxacin, ciprofloxacin, and sulfamethoxazole in the two aquatic environments both posed high ecological risks (RQ>1) to the most sensitive aquatic organisms Vibrio fischeri, Microcystis aeruginosa and Synechococcus leopoliensis, respectively.

Abstract:The malformins constitute a small family of biologically active cyclic pentapeptides produced by the fungus, Aspergillus niger. These plant growth regulators induce malformations in stems and petioles of Phaseolus vulgaris and curvatures in roots of Zea mays (R. W. Curtis, Science 128:661, 1958). Four malformins have been isolated from culture filtrates of A. niger, malformin A1 and A2 from strain 58-883, and B1 and B2 from strain 56-30 (S. Takeuchi, M. Senn, R. W. Curtis, and F. W. McLafferty, Phytochemistry 6:287, 1967). They are most readily isolated as their respective mixtures, malformin A and B. We report here the isolation of malformin A from culture filtrates of A. awamori, and evidence for the presence of compounds with malforminlike activity in the culture filtrates of other fungi including members of the A. niger group of aspergilli. The fungi were grown in shake-flask culture for 7 days on corn steep-glucose medium as described earlier (R. W. Curtis, Plant Physiol. 33:17, 1958), and were then filtered. The filtrates were adjusted to pH 8.3 with NaHCO3, were extracted three times with equal volumes of ethyl ether, and the ether was evaporated on a steam bath. The residues, containing ether-soluble neutral and basic compounds, were treated with water containing Tween 80 (2 drops/100 ml). They were mixed, and the water-soluble compounds were tested for malformin activity by treating the apical buds of seedlings of P. vulgaris var. Black Valentine and the roots of Z. mays, the single-cross WF9 x 38-11, as described in earlier reports. The concentrations of the test solutions ranged from 20 to 1/1,024 times that of the original culture filtrate. Abstract:Although antibiotics are widely detected in river water, their quantitative relationships with influencing factors in rivers remain largely unexplored. Here, 15 widely used antibiotics were comprehensively analyzed in the Dongjiang River of the Pearl River system. The total antibiotic concentration in river water ranged from 13.84 to 475.04 ng/L, with fluoroquinolones increasing from 11 % in the upstream to 38 % in the downstream. The total antibiotic concentration was high downstream and significantly correlated with the spatial distribution of population density, animal production, and land-use type. The total risk quotient of antibiotics for algae was higher than that for crustaceans and fish. Based on the optimized risk quotient method, amoxicillin, ofloxacin, and norfloxacin were identified as priority antibiotics. The key predictors of antibiotic levels were screened through Mantel test, correlation analysis, and multiple regression models. Water physicochemical parameters significantly impacted antibiotics and could be used as easy-to-measure surrogates associated with elevated antibiotics. Cropland negatively affected fluoroquinolones and sulfonamides, whereas urban land exerted positive impacts on fluoroquinolones, β-lactam, and sulfonamides. In the main stream, population, animal production, urbanization status, and economic development had key effects on the distribution of florfenicol, norfloxacin, ofloxacin, and sulfadiazine.

Abstract:Fluoroquinolone antibiotics are pseudo-persistence in aquatic environments as emerging contaminants. In this study, the Shitoukoumen Reservoir and its four inflow rivers were selected as a typical river-reservoir system. The factors, including river input, rainfall, flow regulation, population, antibiotic half-life, and antibiotic emission path were investigated to find out the reasons for antibiotic variations in the river-reservoir system during the reservoir discharge and filling periods. The results showed that ofloxacin was the most prevalent antibiotic in both the reservoir and inflow-river water, with median concentrations of 18.6 ± 2.4 and 39.7 ± 6.2 ng L−1, respectively. The concentrations of enrofloxacin, norfloxacin, and ofloxacin in the reservoir fluctuated during the reservoir discharge and filling periods. Enrofloxacin was only detected in the reservoir during the discharge period due to its input from the inflow rivers. Artificial reservoir discharge decreased the dissolved concentrations of norfloxacin and ofloxacin in the reservoir, whereas reservoir filling increased their accumulation. Occurrence, distribution, and risk assessment of antibiotics in the Songhua River in China

Abstract:The occurrence, distribution, and risk assessment of antibiotics in freshwater systems are receiving global attention, because of their impact on the environment and human health. However, few studies have focused on this topic in Northeast China and its Songhua River, the third-largest river in China. This study investigated the occurrence and distribution of 12 antibiotics, including three cephalosporins (cefazolin, cefmetazole, cefotaxime), three macrolides (azithromycin, clarithromycin, roxithromycin), three fluoroquinolones (ofloxacin, norfloxacin, flumequine), and three sulfonamides (sulfadiazine, sulfapyridine, sulfamethoxazole) in the mainstream and tributaries of the Songhua River. A total of 152 surface water samples were collected in January, May, July, and October 2016. These samples were analyzed using solid-phase extraction (SPE) and ultra-high-performance liquid chromatography coupled with tandem mass spectrometry (UHPLC-MS/MS). The results indicated the wide use of all 12 antibiotics in the mainstream of the Songhua River. Sulfamethoxazole and cefazolin were the dominant antibiotics, with maximum concentrations of 73.1 and 65.4 ng L-1, respectively. Other antibiotics were present at mean concentrations below 15 ng L-1, except cefmetazole, present at a mean concentration of 35.6 ng L-1. The spatial distribution of antibiotics showed that unbalanced regional development may lead to the distribution pattern of the antibiotics in the tributaries and the mainstream. Thus study also assessed the seasonal variation of antibiotics in urban surface water; cephalosporin, sulfonamide, fluoroquinolone, and macrolide concentrations were significantly higher during the icebound season than during non-icebound season. This may be due to the low temperature and water flow of the river in winter. Risk assessment showed that azithromycin, clarithromycin, roxithromycin, flumequine, and sulfamethoxazole posed a low or median risk to the aquatic organisms in the mainstream. The potential risks created by antibiotics to the aquatic environment should not be neglected in the Songhua River. Abstract:Ten antibiotics belonging to three groups (macrolides, fluoroquinolones and sulfonamides) were investigated in riverine runoff of the Pearl River Delta (PRD) and Pearl River Estuary (PRE), South China for assessing the importance of riverine runoff in the transportation of contaminants from terrestrial sources to the open ocean. All antibiotics were detected in the eight outlets with concentrations ranging from 0.7 to 127 ng L−1. The annual mass loadings of antibiotics from the PRD to the PRE and coast were 193 tons with 102 tons from the fluoroquinolone group. It showed that antibiotics decreased from the riverine outlets to the PRE and open ocean. Risk assessment showed that most of these antibiotics showed various ecological risks to the relevant aquatic organisms, in which ofloxacin (OFL), erythromycin (ETM) and ciprofloxacin (CIP) posed high ecological risks to the studied aquatic environments.

Abstract:The identification and source tracing of antibiotic contaminants in rivers has become a research hotspot. In this study, surface water samples were collected from 67 locations in the Yangtze River Basin, China. The detection rate, concentration, and distribution characteristics of various antibiotics in these water samples were analyzed. Based on these data, the sources of these antibiotics were traced and correlated with factors such as urban population size, industrial activity, and wastewater treatment efficiency within the sampling area. A total of 57 antibiotics were detected in the Yangtze River, with detection rates of roxithromycin, 1,7-dimethylxanthine (1,7-DTX), and florfenicol (FF) exceeding 80%. The average total concentration of antibiotics was significantly higher in upstream regions (525.4 ng/L) compared with middle regions (89.5 ng/L) and downstream regions (246.6 ng/L). Furthermore, antibiotic concentrations positively correlated with population density and economic development (R = 0.857, p < 0.05). Based on ecological risk assessment and priority indexes, enoxacin, roxithromycin, sarafloxacin, and clarithromycin (CLR) posed medium risks. This research provides scientific guidance for the periodic monitoring and regulation of antibiotic contaminants in river ecosystems.

Abstract:Three fluoroquinolone antibiotics agents (FQs) in groundwater and reclaimed water have been investigated in Changzhou and Beijing, China. The occurrence of ofloxacin (OFL), enrofloxacin (ENR) and norfloxacin (NOR) is in nanograms per liter and has 100% frequency. The concentration order of FQs in reclaimed water is NOR>OFL>ENR, whilst the order in groundwater is NOR>ENR>OFL. And then the single and mixture adsorption-desorption have been studied and showed that (i) silty clay loam has higher sorption capacity than loamy sand, (ii) competitive adsorption exists when the three selected FQs coexist, (iii) ENR has a significantly priority sorption to NOR, whilst OFL has a least sorption among the mixture, (iv) there is no significant difference between the desorption results of mixture and the indivdual compound in relatively low concentration, (v) the formed chemical bonds and the irreversible combination of adsorption point are the significant influential factors for explaining desorption hysteresis of the selected FQs. Based on the above study, transport model and risk quotient have been performed, and the calculated risk quotient reveals that: (i) the selected FQs risk order in reclaimed water is OFL>ENR>NOR, (ii) in groundwater, OFL and ENR pose a higher risk than NOR no matter whether considering the long time groundwater recharge. This study will help policy makers to decide which FQs need to be covered in the priority substance lists defined in legislative frameworks.
Distribution and risk assessment of antibiotics under water level fluctuation in the riparian zone of the Hanjiang River

Abstract:The riparian zone (RZ) is an important region connecting surface water and groundwater, and it has widely been acknowledged for its pollutant buffering capacity. However, the decontaminating effect of RZ on trace organic compounds such as antibiotics has received little attention. This study explored the distribution of 21 antibiotics and 4 sulfonamide metabolites in river water and groundwater in the lower reaches of the Hanjiang River. The diffusion and exchange of contaminants between the river and riverbanks under the influence of water conservancy projects (Xinglong Dam and the Yangtze-Hanjiang Water Diversion Project) were investigated. Macrolide antibiotics were prevalent in river water (62.5-100%) and groundwater samples (42.9-80.4%). Ofloxacin and chlortetracycline were detected with the highest concentrations in river water (12.2 ng L-1) and groundwater (9.3 ng L-1) respectively. Higher levels of antibiotics were observed in spring and winter than in other seasons. The river-groundwater interaction has a certain interception effect on antibiotics, especially near riverbanks. Redox sensitive element Fe2+ showed significantly positive correlations with some tetracycline and macrolide antibiotics (p < 0.05), and thus the migration mechanism between Fe2+ and antibiotics under the condition of redox change should be investigated further. Environmental risks posed by antibiotics were assessed for algae, daphnids, and fish in surface water and groundwater. Only clarithromycin and chlortetracycline presented a medium risk to algae (0.1 < RQ < 1), and the rest presented low risk (RQ < 0.1). Nevertheless, the risk range may be further extended by interactions between groundwater and surface water. Accurate understanding of antibiotic transport in RZ is critical for developing management strategies aimed at reducing the pollution load on the watershed.

Abstract:Antibiotics have attracted widespread attention around the world because they are ubiquitous in the environment and can lead to antibiotic-resistant microbes developing and pose ecotoxicological risks. In this study, we determined the spatiotemporal distributions of 39 antibiotics in 19 drinking water sources in Jiangsu area of the lower Yangtze River and attempted to identify the sources of the antibiotics and to prioritize the antibiotics. The total antibiotic concentrations in spring and fall were 234.56-6515.99 and 151.12-2562.59 ng/L, respectively. In spring, the total antibiotic concentration gradually increased from upstream to downstream. In fall, the antibiotic concentration did not markedly vary upstream to downstream (total concentrations 151.12-432.17 ng/L) excluding site S9 and S10. Analysis using a positive matrix factorization (PMF) model indicated that the antibiotics had four main sources. Pharmaceutical wastewater was the main source, contributing 34.1% and 41.2% of total antibiotics in spring and fall, respectively, and domestic wastewater was the second most important source, contributing 24.4% and 43% of total antibiotics in spring and fall, respectively. Pharmaceutical wastewater was the main source from midstream to downstream, but the other sources made different contributions in different areas because of the various ranges of human activities. An ecological risk assessment was performed. Stronger risks were posed by antibiotics in spring than fall, and fluoroquinolone antibiotics posed the strongest risks. Optimized risk quotients indicated that norfloxacin was a high-risk contaminant. An assessment of the risk of resistance development indicated that norfloxacin, ciprofloxacin, and enrofloxacin posed moderate to high risks of resistance development and should be prioritized for risk management. The results of this study are important reference data for identifying key sources of antibiotics and developing strategies to manage antibiotic contamination in similar areas.

Abstract:Artificial recharge to groundwater with reclaimed water is considered a promising method to alleviate groundwater depletion and over-exploitation. However, the occurrence of fluoroquinolone antibiotics (FQs) was ubiquitous in wastewater, surface water, groundwater and even drinking water threating human health and ecology. In this study, the occurrence of six selected FQs in reclaimed water effluent and their removal by tertiary treatment units were investigated. The overall removal efficiencies in average of the tertiary treatment processes in Beijing and Changzhou were ranging from 21.2% to 55.2%. Activated carbon exhibited better performance for FQs removal than ozone and biological treatment such as membrane bioreactor, anaerobic-anoxic-oxic and biofilter. The results of two pilot study showed that the impact of reclaimed water to groundwater quality in terms of FQs concentration by direct injection in GBD was stronger than surface spreading in Changzhou, which might be due to the recharge strategy and the physical and chemical characteristics of sediment and aquifer soil. The hazard quotient (HQ) values of ofloxacin (OFL) in reclaimed water was up to 12.54, indicating the extreme eco-toxicological risk, while enrofloxacin (ENR) exhibited medium risk. After recharge with reclaimed water, the HQ values of OFL and ENR in groundwater ranged from low to medium ecological risk to the environment. Thus, the FQs in reclaimed water need to be paid more attention during their reuse for groundwater recharge, especially by direct injection. It is suggested that FQs should be considered in the priority substances lists in standards and guidelines of reclaimed water reuse for groundwater recharge to ensure the safety of groundwater.

Abstract:Antibiotics are causing widespread concern as one of the emerging contaminants. There is the abuse of antibiotics in high-density open aquaculture, and the tailwater is often discharged into surrounding rivers. At the same time, the water replenishment of open aquaculture ponds from nearby rivers containing antibiotic contamination from different sources may result in cross-contamination. However, it is still unclear which pollution intensity is greater in rivers or in open aquaculture. So in this paper, the levels of 20 antibiotics (i.e., Fluoroquinolones (FQs), Sulfonamides (SAs), Tetracyclines (TCs), Macrolides (MLs) and Lincosamides (LCs)) in rivers and high-density open aquaculture ponds were investigated in the Baini River basin in the suburbs of Guangzhou, China. The results showed that norfloxacin (NFX) was the predominant antibiotic in river and aquaculture water, with concentrations ranging from 6.12 to 156.04 ng/L and from 7.47 to 82.62 ng/L in both aquatic systems, respectively. As for the pollution intensity of antibiotics, the annual pollution contribution (28.64 kg/a) of the river water supply to open aquaculture is higher than that (10.81 kg/a) of open aquaculture to the river, which means river pollution has a greater impact on aquaculture ponds. The risk quotient (RQ) showed that the ecological risk of lincomycin (LIN), erythromycin (ERY), sulfamethoxazole (SMX), norfloxacin (NFX), ciprofloxacin (CFX) and chlortetracycline (CTC) in rivers and aquaculture environments had high ecological risks from 1.21 to 1.81. Water interactions with contaminated rivers will result in a corresponding increase in the ecological risk of antibiotics in the aquaculture environment. Overall, according to the results, the risk of polluted rivers to open aquaculture cannot be ignored, and it is recommended that open aquaculture should use these water sources with caution, and that the water quality evaluation of aquaculture water should be increased with monitoring indicators for emerging contaminants such as antibiotics.
