Abstract:Rapid transformation of natural ecosystems changes ecological conditions for important human disease vector species; therefore, an essential task is to identify and understand the variables that shape distributions of these species to optimize efforts toward control and mitigation. Ecological niche modeling was used to estimate the potential distribution and to assess hypotheses of niche similarity among the three main malaria vector species in northern Colombia: Anopheles nuneztovari, An. albimanus, and An. darlingi. Georeferenced point collection data and remotely sensed, fine-resolution satellite imagery were integrated across the Urabá -Bajo Cauca-Alto Sinú malaria endemic area using a maximum entropy algorithm. Results showed that An. nuneztovari has the widest geographic distribution, occupying almost the entire study region; this niche breadth is probably related to the ability of this species to colonize both, natural and disturbed environments. The model for An. darlingi showed that most suitable localities for this species in Bajo Cauca were along the Cauca and Nechí river. The riparian ecosystems in this region and the potential for rapid adaptation by this species to novel environments, may favor the establishment of populations of this species. Apparently, the three main Colombian Anopheles vector species in this endemic area do not occupy environments either with high seasonality, or with low seasonality and high NDVI values. Estimated overlap in geographic space between An. nuneztovari and An. albimanus indicated broad spatial and environmental similarity between these species. An. nuneztovari has a broader niche and potential distribution. Dispersal ability of these species and their ability to occupy diverse environmental situations may facilitate sympatry across many environmental and geographic contexts. These model results may be useful for the design and implementation of malaria species-specific vector control interventions optimized for this important malaria region.

Abstract:Mosquitoes are vectors of many human diseases. In particular, Aedes ægypti (Linnaeus) is the main vector for Chikungunya, Dengue, and Zika viruses in Latin America and it represents a global threat. Public health policies that aim at combating this vector require dependable and timely information, which is usually expensive to obtain with field campaigns. For this reason, several efforts have been done to use remote sensing due to its reduced cost. The present work includes the temporal modeling of the oviposition activity (measured weekly on 50 ovitraps in a north Argentinean city) of Aedes ægypti (Linnaeus), based on time series of data extracted from operational earth observation satellite images. We use are NDVI, NDWI, LST night, LST day and TRMM-GPM rain from 2012 to 2016 as predictive variables. In contrast to previous works which use linear models, we employ Machine Learning techniques using completely accessible open source toolkits. These models have the advantages of being non-parametric and capable of describing nonlinear relationships between variables. Specifically, in addition to two linear approaches, we assess a Support Vector Machine, an Artificial Neural Networks, a K-nearest neighbors and a Decision Tree Regressor. Considerations are made on parameter tuning and the validation and training approach. The results are compared to linear models used in previous works with similar data sets for generating temporal predictive models. These new tools perform better than linear approaches, in particular Nearest Neighbor Regression (KNNR) performs the best. These results provide better alternatives to be implemented operatively on the Argentine geospatial Risk system that is running since 2012.

Abstract:Over 50% of the world population is at risk of mosquito-borne diseases. Female Ae. aegypti mosquito species transmit Zika, Dengue, and Chikungunya. The spread of these diseases correlate positively with the vector population, and this population depends on biotic and abiotic environmental factors including temperature, vegetation condition, humidity and precipitation. To combat virus outbreaks, information about vector population is required. To this aim, Earth observation (EO) data provide fast, efficient and economically viable means to estimate environmental features of interest. In this work, we present a temporal distribution model for adult female Ae. aegypti mosquitoes based on the joint use of the Normalized Difference Vegetation Index, the Normalized Difference Water Index, the Land Surface Temperature (both at day and night time), along with the precipitation information, extracted from EO data. The model was applied separately to data obtained during three different vector control and field data collection condition regimes, and used to explain the differences in environmental variable contributions across these regimes. To this aim, a random forest (RF) regression technique and its nonlinear features importance ranking based on mean decrease impurity (MDI) were implemented. To prove the robustness of the proposed model, we compared it to other machine learning techniques and statistical models. Our results show that machine learning techniques perform better than linear statistical models for the task at hand, and RF performs best. By ranking the importance of all features based on MDI in RF and selecting the subset comprising the most informative ones, a more parsimonious but equally effective and explainable model can be obtained...

Abstract:Mosquitoes are important vectors of disease pathogens and multiple species are undergoing geographical shifts due to global changes. As such, there is a growing need for accurate distribution predictions. Ecological niche modelling (ENM) is an effective tool to assess mosquito distribution patterns and link these to underlying environmental preferences. Typically, macroclimatic variables are used as primary predictors of mosquito distributions. However, they likely undervalue local conditions and intraspecific variation in environmental preferences. This is problematic, as mosquito control takes place at the local scale. Utilising high-resolution (10 × 10 m) Maxent ENMs on the island of Bonaire as model system, we explore the influence of local environmental variables on mosquito distributions. Our results show a distinct set of environmental variables shape distribution patterns across ecologically-distinct species, with urban variables strongly associated with introduced species like Aedes aegypti and Culex quinquefasciatus, while native species show habitat preferences for either mangroves, forests, or ephemeral water habitats. These findings underscore the importance of distinct local environmental factors in shaping distributions of different mosquitoes, even on a small island. As such, these findings warrant further studies aimed at predicting high-resolution mosquito distributions, opening avenues for preventative management of vector-borne disease risks amidst ongoing global change and ecosystem degradation.

Abstract:In recent decades, field and semi-field studies of malaria transmission have gathered geographic-specific information about mosquito ecology, behaviour and their sensitivity to interventions. Mathematical models of malaria transmission can incorporate such data to infer the likely impact of vector control interventions and hence guide malaria control strategies in various geographies. To facilitate this process and make model predictions of intervention impact available for different geographical regions, we developed AnophelesModel. AnophelesModel is an online, open-access R package that quantifies the impact of vector control interventions depending on mosquito species and location-specific characteristics. In addition, it includes a previously published, comprehensive, curated database of field entomological data from over 50 Anopheles species, field data on mosquito and human behaviour, and estimates of vector control effectiveness. Using the input data, the package parameterizes a discrete-time, state transition model of the mosquito oviposition cycle and infers species-specific impacts of various interventions on vectorial capacity. In addition, it offers formatted outputs ready to use in downstream analyses and by other models of malaria transmission for accurate representation of the vector-specific components. Using AnophelesModel, we show how the key implications for intervention impact change for various vectors and locations. The package facilitates quantitative comparisons of likely intervention impacts in different geographical settings varying in vector compositions, and can thus guide towards more robust and efficient malaria control recommendations. Abstract:Malaria constitutes an important cause of human mortality. After 2009 Greece experienced a resurgence of malaria. Here, we develop a modelbased framework that integrates entomological, geographical, social and environmental evidence in order to guide the mosquito control efforts and apply this framework to data from an entomological survey study conducted in Central Greece. Our results indicate that malaria transmission risk in Greece is potentially substantial. In addition, specific districts such as seaside, lakeside and rice field regions appear to represent potential malaria hotspots in Central Greece. We found that appropriate maps depicting the basic reproduction number, R0 , are useful tools for informing policy makers on the risk of malaria resurgence and can serve as a guide to inform recommendations regarding control measures.

Abstract:Anopheles funestus is a major malaria vector in Eastern and Southern Africa and is currently the dominant malaria-transmitting vector in many parts of Tanzania. Previous research has identified its preference for specific aquatic habitats, especially those that persist in dry months. This observation suggests the potential for targeted control through precise habitat mapping and characterization. In this study, we investigated the influence of habitat characteristics, land cover and human population densities on An. funestus distribution during dry seasons. Based on the results, we developed a habitat suitability model for this vector species in south-eastern Tanzania.

Abstract:Vector-borne diseases emergence, particularly malaria, present a significant public health challenge worldwide. Anophelines are predominant malaria vectors, with varied distribution, and influenced by environment and climate. This study, in Ghana, modelled environmental suitability for Anopheles stephensi , a potential vector that may threaten advances in malaria and vector control. Understanding this vector's distribution and dynamics ensures effective malaria and vector control programmes implementation. We explored the MaxEnt ecological modelling method to forecast An. stephensi 's potential hotspots and niches. We analysed environmental and climatic variables to predict spatial distribution and ecological niches of An. stephensi with a spatial resolution of approximately 5 km 2 . Analysing geospatial and species occurrence data, we identified optimal environmental conditions and important factors for its presence. The model's most important variables guided hotspot prediction across several ecological zones aside from urban and peri-urban regions. Considering the vector's complex bionomics, these areas provide varying and adaptable conditions for the vector to colonise and establish. This is shown by the AUC = 0.943 prediction accuracy of the model, which is considered excellent. Based on our predictions, this vector species would thrive in the Greater Accra, Ashanti Central, Upper East, Northern, and North East regions. Forecasting its environmental suitability by ecological niche modelling supports proactive surveillance and focused malaria management strategies. Public health officials can act to reduce the risk of malaria transmission by identifying areas where mosquitoes may breed, which will ultimately improve health outcomes and disease control.

Exploring fine-scale urban landscapes using satellite data to predict the distribution of Aedes mosquito breeding sites

Abstract:Background: The spread of mosquito-transmitted diseases such as dengue is a major public health issue worldwide. The Aedes aegypti mosquito, a primary vector for dengue, thrives in urban environments and breeds mainly in artificial or natural water containers. While the relationship between urban landscapes and potential breeding sites remains poorly understood, such a knowledge could help mitigate the risks associated with these diseases. This study aimed to analyze the relationships between urban landscape characteristics and potential breeding site abundance and type in cities of French Guiana (South America), and to evaluate the potential of such variables to be used in predictive models. Methods: We use Multifactorial Analysis to explore the relationship between urban landscape characteristics derived from very high resolution satellite imagery, and potential breeding sites recorded from in-situ surveys. We then applied Random Forest models with different sets of urban variables to predict the number of potential breeding sites where entomological data are not available. Results: Landscape analyses applied to satellite images showed that urban types can be clearly identified using texture indices. The Multiple Factor Analysis helped identify variables related to the distribution of potential breeding sites, such as buildings class area, landscape shape index, building number, and the first component of texture indices. Models predicting the number of potential breeding sites using the entire dataset provided an R² of 0.90, possibly influenced by overfitting, but allowing the prediction over all the study sites. Predictions of potential breeding sites varied highly depending on their type, with better results on breeding sites types commonly found in urban landscapes, such as containers of less than 200 L, large volumes and barrels. The study also outlined the limitation offered by the entomological data, whose sampling was not specifically designed for this study. Model outputs could be used as input to a mosquito dynamics model when no accurate field data are available. Conclusion: This study offers a first use of routinely collected data on potential breeding sites in a research study. It highlights the potential benefits of including satellite-based characterizations of the urban environment to improve vector control strategies.

Abstract:French Guiana lacks a dedicated model for developing an early warning system tailored to its entomological contexts. We employed a spatiotemporal modeling approach to predict the risk of Aedes aegypti larvae presence in local households in French Guiana. The model integrated field data on larvae, environmental data obtained from very high-spatial-resolution Pleiades imagery, and meteorological data collected from September 2011 to February 2013 in an urban area of French Guiana. The identified environmental and meteorological factors were used to generate dynamic maps with high spatial and temporal resolution. The study collected larval data from 261 different surveyed houses, with each house being surveyed between one and three times. Of the observations, 41% were positive for the presence of Aedes aegypti larvae. We modeled the Aedes larvae risk within a radius of 50 to 200 m around houses using six explanatory variables and extrapolated the findings to other urban municipalities during the 2020 dengue epidemic in French Guiana. This study highlights the potential of spatiotemporal modeling approaches to predict and monitor the evolution of vector-borne disease transmission risk, representing a major opportunity to monitor the evolution of vector risk and provide valuable information for public health authorities.

Abstract:Though malaria control initiatives have markedly reduced malaria prevalence in recent decades, global eradication is far from actuality. Recent studies show that environmental and social heterogeneities in low-transmission settings have an increased weight in shaping malaria micro-epidemiology. New integrated and more localized control strategies should be developed and tested. Here we present a set of agent-based models designed to study the influence of local scale human movements on local scale malaria transmission in a typical Amazon environment, where malaria is transmission is low and strongly connected with seasonal riverine flooding. The agent-based simulations show that the overall malaria incidence is essentially not influenced by local scale human movements. In contrast, the locations of malaria high risk spatial hotspots heavily depend on human movements because simulated malaria hotspots are mainly centered on farms, were laborers work during the day. The agent-based models are then used to test the effectiveness of two different malaria control strategies both designed to reduce local scale malaria incidence by targeting hotspots. The first control scenario consists in treat against mosquito bites people that, during the simulation, enter at least once inside hotspots revealed considering the actual sites where human individuals were infected. The second scenario involves the treatment of people entering in hotspots calculated assuming that the infection sites of every infected individual is located in the household where the individual lives. Simulations show that both considered scenarios perform better in controlling malaria than a randomized treatment, although targeting household hotspots shows slightly better performance.

Abstract:The influence of climate on mosquito-borne diseases like dengue and chikungunya is well-established, but comprehensively tracking long-term spatial and temporal trends across large areas has been hindered by fragmented data and limited analysis tools. This study presents an unprecedented analysis, in terms of breadth, estimating the SIR transmission parameters from incidence data in all 5,570 municipalities in Brazil over 14 years (2010-2023) for both dengue and chikungunya. We describe the Episcanner computational pipeline, developed to estimate these parameters, producing a reusable dataset describing all dengue and chikungunya epidemics that have taken place in this period, in Brazil. The analysis reveals new insights into the climate-epidemic nexus: We identify distinct geographical and temporal patterns of arbovirus disease incidence across Brazil, highlighting how climatic factors like temperature and precipitation influence the timing and intensity of dengue and chikungunya epidemics. The innovative Episcanner tool empowers researchers and public health officials to explore these patterns in detail, facilitating targeted interventions and risk assessments. This research offers a new perspective on the long-term dynamics of climate-driven mosquito-borne diseases and their geographical specificities linked to the effects of global temperature fluctuations such as those captured by the ENSO index.

Abstract:Insecticide resistance and behavioural adaptation of malaria mosquitoes affect the efficacy of long-lasting insecticide nets - currently the main tool for malaria vector control. To develop and deploy complementary, efficient and cost-effective control interventions, a good understanding of the drivers of these physiological and behavioural traits is needed. In this data-mining exercise, we modelled a set of indicators of physiological resistance to insecticide (prevalence of three target-site mutations) and behavioural resistance phenotypes (early- and late-biting, exophagy) of anopheles mosquitoes in two rural areas of West-Africa, located in Burkina Faso and Cote d Ivoire. To this aim, we used mosquito field collections along with heterogeneous, multi-source and multi-scale environmental data. The objectives were i) to assess the small-scale spatial and temporal heterogeneity of physiological resistance to insecticide and behavioural resistance phenotypes, ii) to better understand their drivers, and iii) to assess their spatio-temporal predictability, at scales that are consistent with operational action. The explanatory variables covered a wide range of potential environmental determinants of vector resistance to insecticide or behavioural resistance phenotypes : vector control, human availability and nocturnal behaviour, macro and micro-climatic conditions, landscape, etc. The resulting models revealed many statistically significant associations, although their predictive powers were overall weak. We interpreted and discussed these associations in light of several topics of interest, such as : respective contribution of public health and agriculture in the selection of physiological resistances, biological costs associated with physiological resistances, biological mechanisms underlying biting behaviour, and impact of micro-climatic conditions on the time or place of biting. To our knowledge, our work is the first modeling insecticide resistance and feeding behaviour of malaria vectors at such fine spatial scale with such a large dataset of both mosquito and environmental data.

Abstract:Recent outbreaks of the West Nile virus have been reported in southern Spain, a region with important wetland habitats for migratory birds. Here, we analyzed the role of species association and abiotic parameters on the abundance of seven mosquito species in the Doñana National Park, Spain. We applied the Joint Species Distribution Models (JSDM), using the Hierarchical Modeling of Species Communities approach to simultaneously model the effect of habitat type, normalized difference vegetation index, hydroperiod, distance to rivers, land surface temperature, and the abundance of seven mosquito species. We created three models with varied parameters and evaluated the effects of abiotic parameters and species-to-species associations, which served as proxies for species interactions. Our models estimated the species-specific responses of the mosquito community to abiotic parameters. A positive association was identified within the Culex community with potential biotic interactions. However, Ochlerotatus caspius, Ochlerotatus detritus, and Anopheles atroparvus had no statistically significant association with each other and with the Culex species after accounting for the effect of abiotic parameters. We also found that the potential species associations estimated at particular sites and abiotic parameters influenced the model projection for the average abundance of mosquito species. The JSDM will allow spatial projection of the abundance of each mosquito species, which is an important parameter for epidemiological models. The JSDM inference of species association is important because predation, competition, and facilitation affect the distribution and abundance of different species.

Long-term predictive models for mosquito borne diseases: a narrative review

Abstract:In face of climate change and increasing urbanization, the predictive mosquito-borne diseases (MBD) transmission models require constant updates. Thus, is urgent to comprehend the driving forces of this non stationary behavior, observed through spatial and incidence expansion. We observed that temperature is a critical driver in predictive models for MBD transmission, also being consistently used in multiple reviewed papers with considerable incidence predictive capacity. Rainfall, however, have more subtle importance as moderate precipitation creates breeding sites for mosquitoes, but excessive rainfall can reduce larvae populations. We highlight the frequent use of mechanistic models, particularly those that integrate temperature-dependent biological parameters of disease transmission in incidence proxies as the Vectorial Capacity (VC) and temperature-based basic reproduction number $R_0(t)$, for example. These models show the importance of climate variables, but the socio-demographic factors are often not considered. This gap is a significant opportunity for future research to incorporate socio-demographic data into long-term predictive models for more comprehensive and reliable forecasts. With this survey, we outline the most promising paths to be followed by long-term MBD transmission research and highlighting the potential facing challenges. Thus, we offer a valuable foundation for enhancing disease forecasting models and supporting more effective public health interventions, specially in the long term.

Exploring Dengue Dynamics: A Multi-Scale Analysis of Spatio-Temporal Trends in Ibagué, Colombia

Abstract:Our study examines how dengue fever incidence is associated with spatial (demographic and socioeconomic) alongside temporal (environmental) factors at multiple scales in the city of Ibagué, located in the Andean region of Colombia. We used the dengue incidence in Ibagué from 2013 to 2018 to examine the associations with climate, socioeconomic, and demographic factors from the national census and satellite imagery at four levels of local spatial aggregation. We used geographically weighted regression (GWR) to identify the relevant socioeconomic and demographic predictors, and we then integrated them with environmental variables into hierarchical models using integrated nested Laplace approximation (INLA) to analyze the spatio-temporal interactions. Our findings show a significant effect of spatial variables across the different levels of aggregation, including human population density, gas and sewage connection, percentage of woman and children, and percentage of population with a higher education degree. Lagged temporal variables displayed consistent patterns across all levels of spatial aggregation, with higher temperatures and lower precipitation at short lags showing an increase in the relative risk (RR). A comparative evaluation of the models at different levels of aggregation revealed that, while higher aggregation levels often yield a better overall model fit, finer levels offer more detailed insights into the localized impacts of socioeconomic and demographic variables on dengue incidence. Our results underscore the importance of considering macro and micro-level factors in epidemiological modeling, and they highlight the potential for targeted public health interventions based on localized risk factor analyses. Notably, the intermediate levels emerged as the most informative, thereby balancing spatial heterogeneity and case distribution density, as well as providing a robust framework for understanding the spatial determinants of dengue.

Abstract:Use has been made of the mechanism of DNA deoxyribose damage by the ene-diyne-containing chromophore of the antitumor antibiotic neocarzinostatin to provide chemical evidence for the structure of the activated drug-DNA complex. Radical centers at C-2 and C-6 of the diradical form of the glutathione-activated chromophore abstract hydrogen atoms from C-1' of the C residue and C-5' of the T residue in AGC.GCT to generate a bistranded lesion consisting of an abasic site at C and a strand break at T. This laboratory has proposed a molecular model for the drug-DNA interaction in which the naphthoate moiety of the chromophore intercalates between A.T and G.C, placing the diradical core in the minor groove, so that the radical centers at C-6 and C-2 are close to C-5' of T and C-1' of C, respectively. To determine which radical center abstracts one of the hydrogen atoms from C-5', the self-complementary oligodeoxynucleotide GCAGCGCTGC was synthesized with 2H at both 5' positions of the T residue and treated with glutathione-activated chromophore. Sequencing-gel electrophoresis showed that drug attack was limited to the T and C residues and that abstraction of 2H from C-5' exhibited a small isotope selection effect of 1.25. 1H NMR spectroscopic examination of the reacted chromophore, isolated by HPLC, indicated that 2H was selectively abstracted by C-6, providing experimental corroboration of the model and further elucidating the chemical mechanism. Since direct strand breakage at the T residue exceeds (44% more) abasic site formation at the C residue, other models of drug-DNA interaction leading to only single-strand breaks are also considered.

Abstract:Mosquitoes are responsible for the transmission of major pathogens worldwide. Modelling their population dynamics and mapping their distribution can contribute effectively to disease surveillance and control systems. Two main approaches are classically used to understand and predict mosquito abundance in space and time, namely empirical (or statistical) and process-based models. In this work, we used both approaches to model the population dynamics in Reunion Island of the 'Tiger mosquito', Aedes albopictus, a vector of dengue and chikungunya viruses, using rainfall and temperature data. We aimed to i) evaluate and compare the two types of models, and ii) develop an operational tool that could be used by public health authorities and vector control services. Our results showed that Ae. albopictus dynamics in Reunion Island are driven by both rainfall and temperature with a non-linear relationship. The predictions of the two approaches were consistent with the observed abundances of Ae. albopictus aquatic stages. An operational tool with a user-friendly interface was developed, allowing the creation of maps of Ae. albopictus densities over the whole territory using meteorological data collected from a network of weather stations. It is now routinely used by the services in charge of vector control in Reunion Island.

Abstract:The protective effectiveness of vector control in malaria relies on how the implemented tools overlap with mosquito species-specific compositions and bionomic traits. In Ethiopia, targeted entomological data enabling strategic decision-making are lacking around high-risk migrant worker camps in the lowlands and resident communities in the highlands—resulting in suboptimal malaria control strategies for both populations. This study investigates spatial and temporal mosquito behavior, generating baseline evidence that will improve malaria control for both migrant workers in the lowlands and their home communities in the highlands.
