Abstract:In recent years, deep learning models have been applied to neuroimaging data for early diagnosis of Alzheimer's disease (AD). Structural magnetic resonance imaging (sMRI) and positron emission tomography (PET) images provide structural and functional information about the brain, respectively. Combining these features leads to improved performance than using a single modality alone in building predictive models for AD diagnosis. However, current multi-modal approaches in deep learning, based on sMRI and PET, are mostly limited to convolutional neural networks, which do not facilitate integration of both image and phenotypic information of subjects. We propose to use graph neural networks (GNN) that are designed to deal with problems in non-Euclidean domains. In this study, we demonstrate how brain networks can be created from sMRI or PET images and be used in a population graph framework that can combine phenotypic information with imaging features of these brain networks. Then, we present a multi-modal GNN framework where each modality has its own branch of GNN and a technique is proposed to combine the multi-modal data at both the level of node vectors and adjacency matrices. Finally, we perform late fusion to combine the preliminary decisions made in each branch and produce a final prediction. As multi-modality data becomes available, multi-source and multi-modal is the trend of AD diagnosis. We conducted explorative experiments based on multi-modal imaging data combined with non-imaging phenotypic information for AD diagnosis and analyzed the impact of phenotypic information on diagnostic performance. Results from experiments demonstrated that our proposed multi-modal approach improves performance for AD diagnosis, and this study also provides technical reference and support the need for multivariate multi-modal diagnosis methods.

Abstract:Large-scale population-based studies in medicine are a key resource towards better diagnosis, monitoring, and treatment of diseases. They also serve as enablers of clinical decision support systems, in particular computer-aided diagnosis (CADx) using machine learning (ML). Numerous ML approaches for CADx have been proposed in literature. However, these approaches assume feature-complete data, which is often not the case in clinical data. To account for missing data, incomplete data samples are either removed or imputed, which could lead to data bias and may negatively affect classification performance. As a solution, we propose an end-to-end learning of imputation and disease prediction of incomplete medical datasets via Multi-graph Geometric Matrix Completion (MGMC). MGMC uses multiple recurrent graph convolutional networks, where each graph represents an independent population model based on a key clinical meta-feature like age, sex, or cognitive function. Graph signal aggregation from local patient neighborhoods, combined with multi-graph signal fusion via self-attention, has a regularizing effect on both matrix reconstruction and classification performance. Our proposed approach is able to impute class relevant features as well as perform accurate and robust classification on two publicly available medical datasets. We empirically show the superiority of our proposed approach in terms of classification and imputation performance when compared with state-of-the-art approaches. MGMC enables disease prediction in multimodal and incomplete medical datasets. These findings could serve as baseline for future CADx approaches which utilize incomplete datasets.

Abstract:Both neuroimaging and genomics datasets are often gathered for the detection of neurodegenerative diseases. Huge dimensionalities of neuroimaging data as well as omics data pose tremendous challenge for methods integrating multiple modalities. There are few existing solutions that can combine both multi-modal imaging and multi-omics datasets to derive neurological insights. We propose a deep neural network architecture that combines both structural and functional connectome data with multi-omics data for disease classification. A graph convolution layer is used to model functional magnetic resonance imaging (fMRI) and diffusion tensor imaging (DTI) data simultaneously to learn compact representations of the connectome. A separate set of graph convolution layers are then used to model multi-omics datasets, expressed in the form of population graphs, and combine them with latent representations of the connectome. An attention mechanism is used to fuse these outputs and provide insights on which omics data contributed most to the model's classification decision. We demonstrate our methods for Parkinson's disease (PD) classification by using datasets from the Parkinson's Progression Markers Initiative (PPMI). PD has been shown to be associated with changes in the human connectome and it is also known to be influenced by genetic factors. We combine DTI and fMRI data with multi-omics data from RNA Expression, Single Nucleotide Polymorphism (SNP), DNA Methylation and non-coding RNA experiments. A Matthew Correlation Coefficient of greater than 0.8 over many combinations of multi-modal imaging data and multi-omics data was achieved with our proposed architecture. To address the paucity of paired multi-modal imaging data and the problem of imbalanced data in the PPMI dataset, we compared the use of oversampling against using CycleGAN on structural and functional connectomes to generate missing imaging modalities. Furthermore, we performed ablation studies that offer insights into the importance of each imaging and omics modality for the prediction of PD. Analysis of the generated attention matrices revealed that DNA Methylation and SNP data were the most important omics modalities out of all the omics datasets considered. Our work motivates further research into imaging genetics and the creation of more multi-modal imaging and multi-omics datasets to study PD and other complex neurodegenerative diseases.

Classification of Neurological Gait Disorders Using Multi-task Feature Learning

Abstract:As our population ages, neurological impairments and degeneration of the musculoskeletal system yield gait abnormalities, which can significantly reduce quality of life. Gait rehabilitative therapy has been widely adopted to help patients maximize community participation and living independence. To further improve the precision and efficiency of rehabilitative therapy, more objective methods need to be developed based on sensory data. In this paper, an algorithmic framework is proposed to provide classification of gait disorders caused by two common neurological diseases, stroke and Parkinson's Disease (PD), from ground contact force (GCF) data. An advanced machine learning method, multi-task feature learning (MTFL), is used to jointly train classification models of a subject's gait in three classes, post-stroke, PD and healthy gait. Gait parameters related to mobility, balance, strength and rhythm are used as features for the classification. Out of all the features used, the MTFL models capture the more important ones per disease, which will help provide better objective assessment and therapy progress tracking. To evaluate the proposed methodology we use data from a human participant study, which includes five PD patients, three post-stroke patients, and three healthy subjects. Despite the diversity of abnormalities, the evaluation shows that the proposed approach can successfully distinguish post-stroke and PD gait from healthy gait, as well as post-stroke from PD gait, with Area Under the Curve (AUC) score of at least 0.96. Moreover, the methodology helps select important gait features to better understand the key characteristics that distinguish abnormal gaits and design personalized treatment.
MM-GTUNets: Unified Multi-Modal Graph Deep Learning for Brain Disorders Prediction

Abstract:Graph deep learning (GDL) has demonstrated impressive performance in predicting population-based brain disorders (BDs) through the integration of both imaging and non-imaging data. However, the effectiveness of GDL based methods heavily depends on the quality of modeling the multi-modal population graphs and tends to degrade as the graph scale increases. Furthermore, these methods often constrain interactions between imaging and non-imaging data to node-edge interactions within the graph, overlooking complex inter-modal correlations, leading to suboptimal outcomes. To overcome these challenges, we propose MM-GTUNets, an end-to-end graph transformer based multi-modal graph deep learning (MMGDL) framework designed for brain disorders prediction at large scale. Specifically, to effectively leverage rich multi-modal information related to diseases, we introduce Modality Reward Representation Learning (MRRL) which adaptively constructs population graphs using a reward system. Additionally, we employ variational autoencoder to reconstruct latent representations of non-imaging features aligned with imaging features. Based on this, we propose Adaptive Cross-Modal Graph Learning (ACMGL), which captures critical modality-specific and modality-shared features through a unified GTUNet encoder taking advantages of Graph UNet and Graph Transformer, and feature fusion module. We validated our method on two public multi-modal datasets ABIDE and ADHD-200, demonstrating its superior performance in diagnosing BDs. Abstract:Identification of brain regions related to the specific neurological disorders are of great importance for biomarker and diagnostic studies. In this paper, we propose an interpretable Graph Convolutional Network (GCN) framework for the identification and classification of Alzheimer’s disease (AD) using multi-modality brain imaging data. Specifically, we extended the Gradient Class Activation Mapping (Grad-CAM) technique to quantify the most discriminative features identified by GCN from brain connectivity patterns. We then utilized them to find signature regions of interest (ROIs) by detecting the difference of features between regions in healthy control (HC), mild cognitive impairment (MCI), and AD groups. We conducted the experiments on the ADNI database with imaging data from three modalities, including VBM-MRI, FDG-PET, and AV45-PET, and showed that the ROI features learned by our method were effective for enhancing the performances of both clinical score prediction and disease status identification. It also successfully identified biomarkers associated with AD and MCI.
Modeling Alzheimers' Disease Progression from Multi-task and Self-supervised Learning Perspective with Brain Networks

Abstract:Alzheimer's disease (AD) is a common irreversible neurodegenerative disease among elderlies. Establishing relationships between brain networks and cognitive scores plays a vital role in identifying the progression of AD. However, most of the previous works focus on a single time point, without modeling the disease progression with longitudinal brain networks data. Besides, the longitudinal data is insufficient for sufficiently modeling the predictive models. To address these issues, we propose a Self-supervised Multi-Task learning Progression model SMP-Net for modeling the relationship between longitudinal brain networks and cognitive scores. Specifically, the proposed model is trained in a self-supervised way by designing a masked graph auto-encoder and a temporal contrastive learning that simultaneously learn the structural and evolutional features from the longitudinal brain networks. Furthermore, we propose a temporal multi-task learning paradigm to model the relationship among multiple cognitive scores prediction tasks. Experiments on the Alzheimer's Disease Neuroimaging Initiative (ADNI) dataset show the effectiveness of our method and achieve consistent improvements over state-of-the-art methods in terms of Mean Absolute Error (MAE), Pearson Correlation Coefficient (PCC) and Concordance Correlation Coefficient (CCC). Multi-Modal Diagnosis of Alzheimer's Disease using Interpretable Graph Convolutional Networks

Abstract:The interconnection between brain regions in neurological disease encodes vital information for the advancement of biomarkers and diagnostics. Although graph convolutional networks are widely applied for discovering brain connection patterns that point to disease conditions, the potential of connection patterns that arise from multiple imaging modalities has yet to be fully realized. In this paper, we propose a multi-modal sparse interpretable GCN framework (SGCN) for the detection of Alzheimer's disease (AD) and its prodromal stage, known as mild cognitive impairment (MCI). In our experimentation, SGCN learned the sparse regional importance probability to find signature regions of interest (ROIs), and the connective importance probability to reveal disease-specific brain network connections. We evaluated SGCN on the Alzheimer's Disease Neuroimaging Initiative database with multi-modal brain images and demonstrated that the ROI features learned by SGCN were effective for enhancing AD status identification. The identified abnormalities were significantly correlated with AD-related clinical symptoms. We further interpreted the identified brain dysfunctions at the level of large-scale neural systems and sex-related connectivity abnormalities in AD/MCI. The salient ROIs and the prominent brain connectivity abnormalities interpreted by SGCN are considerably important for developing novel biomarkers. These findings contribute to a better understanding of the network-based disorder via multi-modal diagnosis and offer the potential for precision diagnostics. Beyond the Snapshot: Brain Tokenized Graph Transformer for Longitudinal Brain Functional Connectome Embedding

Abstract:Under the framework of network-based neurodegeneration, brain functional connectome (FC)-based Graph Neural Networks (GNN) have emerged as a valuable tool for the diagnosis and prognosis of neurodegenerative diseases such as Alzheimer's disease (AD). However, these models are tailored for brain FC at a single time point instead of characterizing FC trajectory. Discerning how FC evolves with disease progression, particularly at the predementia stages such as cognitively normal individuals with amyloid deposition or individuals with mild cognitive impairment (MCI), is crucial for delineating disease spreading patterns and developing effective strategies to slow down or even halt disease advancement. In this work, we proposed the first interpretable framework for brain FC trajectory embedding with application to neurodegenerative disease diagnosis and prognosis, namely Brain Tokenized Graph Transformer (Brain TokenGT). It consists of two modules: 1) Graph Invariant and Variant Embedding (GIVE) for generation of node and spatio-temporal edge embeddings, which were tokenized for downstream processing; 2) Brain Informed Graph Transformer Readout (BIGTR) which augments previous tokens with trainable type identifiers and non-trainable node identifiers and feeds them into a standard transformer encoder to readout. We conducted extensive experiments on two public longitudinal fMRI datasets of the AD continuum for three tasks, including differentiating MCI from controls, predicting dementia conversion in MCI, and classification of amyloid positive or negative cognitively normal individuals. Based on brain FC trajectory, the proposed Brain TokenGT approach outperformed all the other benchmark models and at the same time provided excellent interpretability. Dementia classification using a graph neural network on imaging of effective brain connectivity

Abstract:Alzheimer's disease (AD) and Parkinson's disease (PD) are two of the most common forms of neurodegenerative diseases. The literature suggests that effective brain connectivity (EBC) has the potential to track differences between AD, PD and healthy controls (HC). However, how to effectively use EBC estimations for the research of disease diagnosis remains an open problem. To deal with complex brain networks, graph neural network (GNN) has been increasingly popular in very recent years and the effectiveness of combining EBC and GNN techniques has been unexplored in the field of dementia diagnosis. In this study, a novel directed structure learning GNN (DSL-GNN) was developed and performed on the imaging of EBC estimations and power spectrum density (PSD) features. In comparison to the previous studies on GNN, our proposed approach enhanced the functionality for processing directional information, which builds the basis for more efficiently performing GNN on EBC. Another contribution of this study is the creation of a new framework for applying univariate and multivariate features simultaneously in a classification task. The proposed framework and DSL-GNN are validated in four discrimination tasks and our approach exhibited the best performance, against the existing methods, with the highest accuracy of 94.0% (AD vs. HC), 94.2% (PD vs. HC), 97.4% (AD vs. PD) and 93.0% (AD vs. PD vs. HC). In a word, this research provides a robust analytical framework to deal with complex brain networks containing causal directional information and implies promising potential in the diagnosis of two of the most common neurodegenerative conditions.

Abstract:BACKGROUND AND OBJECTIVE:For early identification of Alzheimer's disease (AD) based on multi-modal magnetic resonance imaging (MRI) data, it is important to make comprehensive use of image features and non-image information to analyze the gray matter atrophy and the structural/functional connectivity abnormalities for different courses of AD.METHODS:In this study, we propose an extensible hierarchical graph convolutional network (EH-GCN) for early AD identification. Based on the extracted image features from multi-modal MRI data using the presented multi-branch residual network (ResNet), the brain regions-of-interests (ROIs) based GCN is built to extract structural and functional connectivity features between different ROIs of the brain. In order to further improve the performance of AD identification, an optimized spatial GCN is proposed as convolution operator in the population-based GCN to avoid rebuilding the graph network and take advantage of relationships between subjects. Finally, the proposed EH-GCN is built by embedding the image features and internal brain connectivity features into the spatial population-based GCN, which provides an extensible way to improve early AD identification performance by adding imaging features and non-image information from multi-modal data.RESULTS:Experiments are performed on two datasets, which illustrate the effectiveness of the extracted structural/functional connectivity features and the high computational efficiency of the proposed method. The classification accuracy of AD vs NC, AD vs MCI and MCI vs NC classification tasks reaches 88.71%, 82.71% and 79.68% respectively. The extracted connectivity features between ROIs indicate that functional abnormalities are earlier than gray matter atrophy and abnormalities of structural connections, which is consistent with the clinical manifestations. The proposed method allows for the addition of other modal image features and non-image information from multi-modal data to continuously improve the performance of clinical data analysis.CONCLUSIONS:The proposed method can help us comprehensively analyze the role of gray matter atrophy, the damage of white matter nerve fiber tracts and the degradation of functional connectivity for different courses of AD, which could be useful for further extraction of clinical biomarkers for early AD identification.

Abstract:Alzheimer's disease (AD) is a severe neurodegenerative disorder that usually starts slowly and progressively worsens. Predicting the progression of Alzheimer's disease with longitudinal analysis on the time series data has recently received increasing attention. However, training an accurate progression model for brain network faces two major challenges: missing features, and the small sample size during the follow-up study. According to our analysis on the AD progression task, we thoroughly analyze the correlation among the multiple predictive tasks of AD progression at multiple time points. Thus, we propose a multi-task learning framework that can adaptively impute missing values and predict future progression over time from a subject's historical measurements. Progression is measured in terms of MRI volumetric measurements, trajectories of a cognitive score and clinical status. To this end, we propose a new perspective of predicting the AD progression with a multi-task learning paradigm. In our multi-task learning paradigm, we hypothesize that the inherent correlations exist among: (i). the prediction tasks of clinical diagnosis, cognition and ventricular volume at each time point; (ii). the tasks of imputation and prediction; and (iii). the prediction tasks at multiple future time points. According to our findings of the task correlation, we develop an end-to-end deep multi-task learning method to jointly improve the performance of assigning missing value and prediction. We design a balanced multi-task dynamic weight optimization. With in-depth analysis and empirical evidence on Alzheimer's Disease Neuroimaging Initiative (ADNI), we show the benefits and flexibility of the proposed multi-task learning model, especially for the prediction at the M60 time point. The proposed approach achieves 5.6%, 5.7%, 4.0% and 11.8% improvement with respect to mAUC, BCA and MAE (ADAS-Cog13 and Ventricles), respectively.

A Self-guided Multimodal Approach to Enhancing Graph Representation Learning for Alzheimer's Diseases

Abstract:Graph neural networks (GNNs) are powerful machine learning models designed to handle irregularly structured data. However, their generic design often proves inadequate for analyzing brain connectomes in Alzheimer's Disease (AD), highlighting the need to incorporate domain knowledge for optimal performance. Infusing AD-related knowledge into GNNs is a complicated task. Existing methods typically rely on collaboration between computer scientists and domain experts, which can be both time-intensive and resource-demanding. To address these limitations, this paper presents a novel self-guided, knowledge-infused multimodal GNN that autonomously incorporates domain knowledge into the model development process. Our approach conceptualizes domain knowledge as natural language and introduces a specialized multimodal GNN capable of leveraging this uncurated knowledge to guide the learning process of the GNN, such that it can improve the model performance and strengthen the interpretability of the predictions. To evaluate our framework, we curated a comprehensive dataset of recent peer-reviewed papers on AD and integrated it with multiple real-world AD datasets. Experimental results demonstrate the ability of our method to extract relevant domain knowledge, provide graph-based explanations for AD diagnosis, and improve the overall performance of the GNN. This approach provides a more scalable and efficient alternative to inject domain knowledge for AD compared with the manual design from the domain expert, advancing both prediction accuracy and interpretability in AD diagnosis.

Abstract:Alzheimer's disease (AD) is a progressive and irreversible neurodegenerative disease. To distinguish the stage of the disease, AD classification technology challenge has been proposed in Pattern Recognition and Computer Vision 2021 (PRCV 2021) which provides the gray volume and average cortical thickness data extracted in multiple atlases from magnetic resonance imaging (MRI). Traditional methods either train with convolutional neural network (CNN) by MRI data to adapt the spatial features of images or train with recurrent neural network (RNN) by temporal features to predict the next stage. However, the morphological features from the challenge have been extracted into discrete values. We present a multi-atlases multi-layer perceptron (MAMLP) approach to deal with the relationship between morphological features and the stage of the disease. The model consists of multiple multi-layer perceptron (MLP) modules, and morphological features extracted from different atlases will be classified by different MLP modules. The final vote of all classification results obtains the predicted disease stage. Firstly, to preserve the diversity of brain features, the most representative atlases are chosen from groups of similar atlases, and one atlas is selected in each group. Secondly, each atlas is fed into one MLP to fetch the score of the classification. Thirdly, to obtain more stable results, scores from different atlases are combined to vote the result of the classification. Based on this approach, we rank 10th among 373 teams in the challenge. The results of the experiment indicate as follows: (1) Group selection of atlas reduces the number of features required without reducing the accuracy of the model; (2) The MLP architecture achieves better performance than CNN and RNN networks in morphological features; and (3) Compared with other networks, the combination of multiple MLP networks has faster convergence of about 40% and makes the classification more stable.

Abstract:Background: Persistently infected cells by herpesvirus (HSV, CMV, EBV) and their dysfunction are associated with different disorders. The immune and endocrine systems are involved in pathogenesis of herpesvirus infection. Changes of IFN-γ, IL-4, DHEAS and cortisol level are a part of the regulatory mechanisms of the immune and endocrine system responses in persistent herpesvirus infection. Objectives: The aim of this study was to identify immunoregulatory mechanisms (clinical manifestations, cytokines level-IFN-γ, et IL-4, hormones level-DHEAS, cortisol, parameters of oxidative metabolism, enzymes of cell injury) and their consequences concerning clinical disease expression (low level of hemoglobin (Hb), leucocitopenia, high level of monocytes, hyper gamaglobulins, low level of NBT-test). Study design: 40 children (1-16 years old) with herpesvirus infection were analyzed. In patients routine laboratory investigations were performed: white blood cells count, lactate dehydrogenize-LDH, creatinin-kinaza- CPK, oxidative metabolism of the blood phagocytes, the ability of phagocytes nitro-blue tetrazolium-NBT reduction and immunoglobulin serum level. Serum levels of IFN-γ, IL-4, DHEAS and cortisol were measured by ELISA test. Normal healthy controls had negative ELISA tests for CMV, HSV and EBV Results: The high level of LDH, CPK, low ability of NBT reduction and hypergammaglobulins were detected. The increases of IFN-γ, IL-4 level, as well as, the decrease of DHEAS and cortisol level were present. The patients with the high level of IFN-γ, had the high level of LDH, CPK, GOT, GPT, the low level of Hb, leucocitopenia and monocytes. Conclusions: Two groups of patients were presented. First with the high level of IFN-γ (70%) and second with the high level of IL-4.The patients in the first group, with the high level of IFN-γ, had anemia, respiratory infections, BHR, labial HSV, stomatitis aphtosa. Vasculitis, anginae recidivans, was clinical manifestation characteristics for in patients in the second group with the high level of IL-4. The high level of LDH, CPK, the low ability of NBT reduction and hypergammaglobulins were detected. The patients with the high levels of IFN-γ, had the high level of LDH, CPK, GOT, GPT, the low level of Hb, leukopenia and monocytosis. The decreased level of DHEAS (50%) and the normal level of cortisol (76%) were present. The low levels of DHEAS followed the IgG high values while leucopenia (78%), was followed by high values of DHEAS. The patients with normal cortisol values had the significant monocytosis (80%) while patients with the low cortisol level had the high IgG (85%) level.
Exploiting Task Relationships for Alzheimer’s Disease Cognitive Score Prediction Via Multi-Task Learning

Abstract:Alzheimer’s disease (AD) is highly prevalent and a significant cause of dementia and death in elderly individuals. Motivated by breakthroughs of multi-task learning (MTL), efforts have been made to extend MTL to improve the Alzheimer’s disease cognitive score prediction by exploiting structure correlation. Though important and well-studied, three key aspects are yet to be fully handled in an unified framework: (i) appropriately modeling the inherent task relationship; (ii) fully exploiting the task relatedness by considering the underlying feature structure. (iii) automatically determining the weight of each task. To this end, we present the Bi-Graph guided self-Paced Multi-Task Feature Learning (BGP-MTFL) framework for exploring the relationship among multiple tasks to improve overall learning performance of cognitive score prediction. The framework consists of the two correlation regularization for features and tasks, ℓ2,1 regularization and self-paced learning scheme. Moreover, we design an efficient optimization method to solve the non-smooth objective function of our approach based on the Alternating Direction Method of Multipliers (ADMM) combined with accelerated proximal gradient (APG). The proposed model is comprehensively evaluated on the Alzheimer’s disease neuroimaging initiative (ADNI) datasets. Overall, the proposed algorithm achieves an nMSE (normalized Mean Squared Error) of 3.923 and an wR (weighted R-value) of 0.416 for predicting eighteen cognitive scores, respectively. The empirical study demonstrates that the proposed BGP-MTFL model outperforms the state-of-the-art AD prediction approaches and enables identifying more stable biomarkers.

Abstract:Imaging-based early diagnosis of Alzheimer Disease (AD) has become an effective approach, especially by using nuclear medicine imaging techniques such as Positron Emission Topography (PET). In various literature it has been found that PET images can be better modeled as signals (e.g. uptake of florbetapir) defined on a network (non-Euclidean) structure which is governed by its underlying graph patterns of pathological progression and metabolic connectivity. In order to effectively apply deep learning framework for PET image analysis to overcome its limitation on Euclidean grid, we develop a solution for 3D PET image representation and analysis under a generalized, graph-based CNN architecture (PETNet), which analyzes PET signals defined on a group-wise inferred graph structure. Computations in PETNet are defined in non-Euclidean, graph (network) domain, as it performs feature extraction by convolution operations on spectral-filtered signals on the graph and pooling operations based on hierarchical graph clustering. Effectiveness of the PETNet is evaluated on the Alzheimer's Disease Neuroimaging Initiative (ADNI) dataset, which shows improved performance over both deep learning and other machine learning-based methods.

Abstract:Alzheimer’s disease (AD) is a chronic neurodegenerative disease. Early diagnosis are very important to timely treatment and delay the progression of the disease. In the past decade, many computer-aided diagnostic (CAD) algorithms have been proposed for classification of AD. In this paper, we propose a novel graph neural network method, termed Brain Graph Attention Network (BGAN) for classification of AD. First, brain graph data are used to model classification of AD as a graph classification task. Second, a local attention layer is designed to capture and aggregate messages of interactions between node neighbors. And, a global attention layer is introduced to obtain the contribution of each node for graph representation. Finally, using the BGAN to implement AD classification. We train and test on two open public databases for AD classification task. Compared to classic models, the experimental results show that our model is superior to six classic models. We demonstrate that BGAN is a powerful classification model for AD. In addition, our model can provide an analysis of brain regions in order to judge which regions are related to AD disease and which regions are related to AD progression.
