Abstract:Abstract Recurrent spontaneous abortion (RSA) is a multi-factor disease. The mammalian target of the the rapamycin (MTOR) gene has been reported to be involved in mouse embryo development and regulates the proliferation of embryonic stem cells. Our study explored the relationship between the single nucleotide polymorphism (SNP) rs17027478 in the promoter region of MTOR gene and the development of RSA. A total of 306 patients with RSA and 127 healthy females as the controls were recruited in the case-control study. The predesigned TaqMan SNP Genotyping Assay was adopted to analyze the association between rs17027478 and the development of RSA. Quantitative real-time reverse transcription polymerase chain reaction and luciferase reporter assays were conducted to analyze the function of the variant. It was found that a significant association exists between the variant and the risk of RSA among the patients who experienced no less than three spontaneous abortions (p = 0.043). However, the significant difference disappeared among the total samples (p = 0.524). Furthermore, we observed lower MTOR mRNA levels in the blood of RSA patients compared with healthy females (p = 0.020). The luciferase reporter assay showed that the rs17027478A allele significantly reduced the luciferase activity (p = 0.029). The results demonstrated that the variant rs17027478 in the promoter region of MTOR might be a good candidate responsible for the pathogenesis of RSA.

Association of Sun Safety Behaviors and Barriers with Sunburn History in College Students in a Region with High UV Exposure

Abstract:Over five million cases of skin cancer are diagnosed each year in the United States with melanoma the third most common cancer in young adults. While publications have shown that sunburns increase the risk of developing melanoma throughout the lifetime including in adolescence and adulthood showing the importance of altering sun exposing behaviors throughout the lifetime, use of sun protection in college students remails low. In Fall 2019, an online survey of undergraduate students living on campus at a large southwestern university was conducted to determine the frequency of recent sunburns as well as sun protective behaviors and perceived knowledge of and barriers to sun protection. Associations between knowledge, behaviors, and barriers with self-reported sunburn were evaluated using logistic regression. Over 46% of 458 students reported at least one sunburn in the past three months and 21% reported having multiple sunburns in that period. Furthermore, 53% reported that they intentionally tanned their skin outdoors occasionally or more frequently, while 6.4% reported using an indoor tanning bed occasionally or more. Adjusted for skin sensitivity, recent sunburn history was associated with higher tanning activity scores and with high agreement that tanning was attractive (p < 0.01). This information can inform a more targeted series of intervention programming on the university campus.

Abstract:OBJECTIVE: Using the terror management health model (J. L. Goldenberg & J. Arndt, 2008), the authors examined tanning outcomes as a function of priming tanning-relevant standards for attractiveness after reminders of death.DESIGN: Study 1 consisted of 101 female college students recruited from a midwestern university; Study 2 consisted of 53 female participants recruited from a beach in south Florida. In both experiments, participants answered questions about their mortality or a control topic, and were presented with a fashion article that highlighted either the attractiveness of tanned (just in Study 1), pale, or natural-looking skin.MAIN OUTCOME MEASURES: Self-reported suntan intentions (Study 1) and sunscreen intentions (Study 2). Study 2 also assessed participants' interest in various sun protection products.RESULTS AND CONCLUSIONS: Study 1 demonstrated that mortality salience led to higher tanning intentions when the association between tanned skin and physical attractiveness was made salient and reduced intentions when the attractiveness of paler skin was highlighted. In Study 2, beachgoers, after reminders of death, reported greater preference for high sun protection sunscreen after reading an article about the attractiveness of paler skin tones. These findings contribute to an emerging understanding of how mortality concerns can influence health-related judgment and behavior.

Abstract:In a case-control study based in two areas of Kenya, hepatosplenic schistosomiasis mansoni was shown to be linked with low levels of IL-5 and with correspondingly high IFN-gamma, TNF, and circulating soluble TNF receptor I (sTNFR-I), sTNFR-II, and sICAM-1. PBMC from the hepatosplenic cases responded to in vitro Ag stimulation with significantly higher levels of IFN-gamma and TNF, but lower levels of IL-5, compared with nonhepatosplenic controls matched for age and infection intensity. Most of these correlations were confounded by differences between geographical areas. However, principle component analysis identified a high IFN-gamma and TNF, and low IL-5 axis in the data as the first principle component; this was significantly associated with hepatosplenomegaly (p < 0.0005) even after controlling for area. High plasma levels of sTNFR-I (p < 0.001), sTNFR-II, (p < 0.0001), and sICAM-1 (p < 0.009) were also significantly associated with hepatosplenomegaly, independently of area, in the case of the soluble forms of both TNF receptors. These parameters were negatively related to IL-5. These results suggest that proinflammatory cytokines are involved in the hepatosplenic disease process in infected individuals who have low anti-inflammatory Th2 responses and that sTNFR may be a useful circulating marker for this disease process, perhaps reflecting the level of TNF activity in hepatic tissues.

Geography, facilities, and promotional strategies used to encourage indoor tanning in New York City

Abstract:There is emerging evidence for the relationship between indoor tanning and melanoma. Eighty-five indoor tanning facilities in New York City were observed to determine number of tanning machines, pricing, promotions, products, and hours. Census data by zip code was used to determine population density, gender, race, age, percent living in poverty, percent unemployed, and percent college educated of areas in which tanning facilities were located. Pricing varied by the type of machine, number of sessions purchased, and single versus bundled sessions. Facilities were located in areas that had greater population density and slightly greater median age. Compared with the zip code areas with no facilities, those with tanning facilities had a higher proportion of white residents; a lower proportion of residents living in poverty and unemployed; and a higher proportion of residents with a college education. Our data suggest that the strategic location of facilities and promotions used in NYC seek to maximize patronage by those with comparatively high levels of income and education and who may be more influenced by the social desirability of artificial tanning. Long-term interventions aimed at changing social norms regarding tan skin are needed.
Sun exposure concern, sun protection behaviors and physical activity among Australian adults

Abstract:Objective: Sun protective behavior and physical activity are both important for cancer prevention, and are currently promoted separately. Few studies have examined sun protective behaviors, in association with physical activity. This study aimed to explore the interrelationships between sun protective behaviors and physical activity. Method: In an observational epidemiological study, 1,992 adults aged between 20 and 65 years provided responses to self-completion survey items on their concern about sun exposure, sun protection behaviors, physical activity, indoor sedentary behaviors, and their socio-demographic attributes. Results: Men and women, and those who were active and inactive used different sun protection measures. Women reported more frequent use of sunscreen and seeking shade when outdoors, while men reported more hat wearing; women and those who were physically inactive were more concerned about sun exposure and skin damage. After controlling for age, education, and income, concern about sun exposure was associated positively with use of sun protection behaviors; however, those most concerned about sun exposure also tended to be physically inactive and spent more time in indoor sedentary behaviors. Conclusion: To promote physical activity and sun protection simultaneously, it is important to take into account findings on their interrelationships, including the significant gender differences that we have identified.
Body image, self-esteem, and behavioral risk for chronic disease among college students: Additional evidence for integrated prevention

Abstract:Objective: To examine associations between two body image constructs (body appreciation and body satisfaction) and five health behaviors (diet, physical activity, weight management, tobacco exposure, and alcohol intake) associated with risk for chronic disease, controlling for self-esteem. Participants: Three hundred and forty-four college students enrolled at a large, public university in the southeastern US. Methods: Students completed an online survey composed of measures of body appreciation, body satisfaction, self-esteem, and frequency of engagement in preventive health behaviors. Hierarchical linear regression models were used to assess relationships between constructs. Results: Body appreciation, but not body satisfaction or self-esteem, significantly and positively predicted engagement in diet-, physical activity-, and weight-related health behaviors. No associations were found for substance use outcomes. Conclusions: Results illuminate health risks among college students and provide additional evidence to support the development of holistic preventive interventions that simultaneously address aspects of mental and physical health among college students.

Abstract:Dysfunctional appearance beliefs have been identified as important risk factors in the development of body dissatisfaction and disordered eating behaviors among early adolescents. The current two‐wave study among 10–14‐year‐olds (N = 1,597, Mage = 11.25, SD = 1.05) aimed to delineate factors that contribute to the endorsement of such beliefs. Results showed that tweens discussed appearance‐related topics with their friends more often when they frequently watched tween television programs (all time 1). Both media exposure and peer conversations (time 1) were related to attributing benefits to attractiveness (time 1), which, in turn, predicted the internalization of appearance ideals (time 1). The internalization of appearance ideals positively predicted the endorsement of dysfunctional appearance beliefs 6 months later.

Abstract:In an approach to the identification of bile salt-binding carriers, the photoactivable bile acid derivatives A) 3 beta-azido, 7 alpha,12 alpha-dihydroxy-5 beta-cholan-24-oic acid, B) 7,7-azo-3 alpha,12 alpha-dihydroxy-5 beta-cholan-24-oic acid, and C) 11 xi-azido-12-oxo-3 alpha,7 alpha-dihydroxy-5 beta-cholan-24-oic acid were synthesized in unconjugated and taurine-conjugated form. Photolysis of the 3 beta-azido derivatives was studied using a light source with a maximum emission at 300 nm and established a half-life time of 18.5 min. The photochemistry of the 7,7-azo derivatives was investigated using light with a maximum at 350 nm and had a half-life time of 2.2 min. The 11 xi-azido-12-oxo derivatives were photolyzed with light having a maximum at 300 nm resulting in a half-life time of 8.5 min. The suitability of the 7,7-azo derivatives for photoaffinity labeling was demonstrated by photolyses in 14C-labeled methanol and acetonitrile. The generated carbene reacted with the solvents under covalent bond formation of 6 to 12%. The efficiency of all synthesized photolabile derivatives for photoaffinity labeling of bile salt binding proteins was demonstrated.
The Impact of Parental Knowledge and Tanning Attitudes on Sun Protection Practice for Young Children in Germany

Abstract:Public health campaigns have improved knowledge on UVR-associated skin cancer risk and increased sun protection awareness. However, tanned skin is still a common beauty ideal. The relationship between knowledge, attitudes and protective behavior is not fully understood yet. A population-based survey was thus performed in the district of Erlangen involving 2,619 parents of 3- to 6-year old children. By means of a self-administered standardized questionnaire parental knowledge about risk factors for skin cancer, their attitudes towards tanning and details of protective measures taken for their children were assessed. The study analyzed specifically the impact of parental tanning attitudes on sun-protective measures for their children while controlling for parental knowledge about skin cancer risk factors. While parental knowledge was significantly (inversely) associated with agreement to the statement "Tanned skin is healthy skin", this was not the case for "Tanning makes me look better". Overall, tanning affirmative attitudes were inversely associated with protective measures taken for the children, whereas parental knowledge had a positive impact on sun protection at the beach only. Multivariable analyses provided evidence for an effect of parental attitude on protective behavior independent of parental knowledge. Tanning attitudes and tanned skin as the misguided ideal of beauty need to be addressed in future public health campaigns to enhance the effectiveness of preventive activities in changing sun protective behavior.
Adolescents' online appearance preoccupation: A 5-year longitudinal study of the influence of peers, parents, beliefs, and disordered eating

Abstract:Online behaviors, especially those concerning appearance preoccupation, have been related to poorer mental health, but early risk factors for online appearance preoccupation are not well understood. In this study, data were collected in the first years of adolescence (age 10–13 years) to investigate correlates of later online appearance preoccupation (ages 15–18 years). Drawing data from a longitudinal study, 261 Australian adolescents (48% male; M age = 12.0, SD = 0.89) completed surveys to report on social pressures, personal beliefs and attitudes, and body change and eating behaviors at the start of the study (when age 10 to 13). They also reported their online appearance preoccupation (appearance comparison/presentation, and appearance-related activity) five years later at age 15 to 18. Results demonstrated that the most prominent risks for online appearance preoccupation and activity in later adolescence were social in origin. Specifically, appearance-related conversations with friends in early adolescence emerged as the most salient preceding risk factor for boys' and girls' online appearance preoccupation five years later. Also, there were distinct early risks in models estimated separately for boys and girls; these included parents' appearance-related attitudes and behaviors for boys and internalization of appearance ideals and emotional eating for girls.

Indoor Tanning Bed Use and Risk of Food Addiction Based on the Modified Yale Food Addiction Scale.

Abstract:The popularity of indoor tanning may be partly attributed to the addictive characteristics of tanning for some individuals. We aimed to determine the association between frequent indoor tanning, which we view as a surrogate for tanning addiction, and food addiction. A total of 67,910 women were included from the Nurses' Health Study II. In 2005, we collected information on indoor tanning during high school/college and age 25-35 years, and calculated the average use of indoor tanning during these periods. Food addiction was defined as ≥3 clinically significant symptoms plus clinically significant impairment or distress, assessed in 2009 using a modified version of the Yale Food Addiction Scale. Totally 23.3% (15,822) of the participants reported indoor tanning at high school/college or age 25-35 years. A total of 5,557 (8.2%) women met the criteria for food addiction. We observed a dose-response relationship between frequency of indoor tanning and the likelihood of food addiction (Ptrend<0.0001), independent of depression, BMI, and other confounders. Compared with never indoor tanners, the odds ratio (95% confidence interval) of food addiction was 1.07 (0.99-1.17) for average indoor tanning 1-2 times/year, 1.25 (1.09-1.43) for 3-5 times/year, 1.34 (1.14-1.56) for 6-11 times/year, 1.61 (1.35-1.91) for 12-23 times/year, and 2.98 (1.95-4.57) for 24 or more times/year. Frequent indoor tanning before or at early adulthood is associated with prevalence of food addiction at middle age. Our data support the addictive property of frequent indoor tanning, which may guide intervention strategies to curb indoor tanning and prevent skin cancer.

Abstract:Introduction: Perceptions about one’s body can have a significant impact on social activities, quality of life, and overall psychological well-being. Women by far bear the brunt of negative psychological outcomes associated with poor body image. Little is known about the investment in appearance maintenance behaviors that women may engage to attain an idealized body standard. The current study investigated the connection between self-objectification, body image, and the time and money women invest in their appearance. Methods: 450 college-aged (M= 20.23) women completed an online survey that assessed self-objectification, body image, and appearance maintenance (e.g., non-hygienic grooming) behaviors. Results: Higher self-objectification scores were significantly correlated with body dissatisfaction (r = .41, p < .001). Self-objectification was also correlated with the amount of time (r = .17, p < .001) and money (r = .21, p < .001) that women invested in grooming. These associations suggest that women who engage in self-objectification tend to spend more time and money on appearance. There was also a significant association between grooming and body dysphoria (r = .11, p = .039). Conclusions: Overall, these findings provide initial support that women who self-objectify invest more time and money in their appearance and tend to have fewer positive feelings towards their bodies.
Targeted nanoassembly loaded with docetaxel improves intracellular drug delivery and efficacy in murine breast cancer model.

Abstract:Docetaxel is one of the most promising chemotherapeutic agents for the treatment of metastatic breast cancer, but it shows fearful side effects. We hypothesized that a novel targeted nanoassembly (TNA) could provide efficient intracellular drug delivery in breast tumor cells overexpressing epidermal growth factor (EGF) receptor and thus improve the efficacy and reduce the side effects of docetaxel. We prepared the novel docetaxel loaded TNAs formed by polyethylene glycol-distearoylphosphatidylethanolamine (PEG-DSPE) and modified with EGF. Compared with nontargeted nanoassemblies (NNAs), TNAs showed obvious improvement of cell-specific uptake and internalization, and revealed more cytotoxicity against MDA-MB-468 cells by inducing more late apoptosis and subG1 cells at low drug concentration, or more G2/M arrest at high drug concentration than NNAs or Taxotere. In BALB/c mice bearing breast tumor xenografts, TNAs showed stronger inhibition of tumor growth compared with NNAs (relative tumor volume in mice treated with 5 mg/kg TNAs = 0.99 and 10 mg/kg NNAs = 1.71, p < 0.05) or Taxotere (relative tumor volume in mice treated with 5 mg/kg TNAs = 0.99 and 10 mg/kg Taxotere = 4.20, p < 0.01). In particular, tumor disappeared completely in the TNA group at a dose of 10 mg/kg. The maximum tolerated dose (MTD) of TNAs was about four times higher than that of Taxotere. TNAs also demonstrated a much longer circulation time in vivo and more drug accumulation in tumor in a murine breast cancer model than Taxotere. TNA treatment also prolonged survival of mice. These results suggested that TNAs could have more potential as a delivery system for breast cancer chemotherapy.

Abstract:Online appearance preoccupation is a known correlate of poorer body image, social problems, and mental health symptoms among youth. Yet, this study is the first to investigate individual and social risks and resources expected to explain the level of concurrent, as well as change in, online appearance preoccupation across one year. We focused on two risk factors of appearance teasing online by peers and appearance-based sensitivity to rejection, and two positive resources of appearance neutral support from important others and ways of coping with appearance-related stressors. A total of 752 Australian adolescents and emerging adults (14–21 years; M = 17.8 years, SD = 1.8 years; 41% young men; about 95% White or Asian) recruited from secondary schools or university were included. Results of regression analyses suggested that, concurrently, online appearance teasing, appearance rejection sensitivity, and positive coping were uniquely associated with online appearance preoccupation. Further, the positive association of peer appearance teasing with online appearance preoccupation was stronger among young men than young women. Over time and once all risks and resources were considered, positive coping with appearance pressures was associated with a decrease in online appearance preoccupation from T1 to T2. Gender (girl/young woman) and intensity of social media use were notable risks for concurrent online appearance preoccupation and predictors of increases in preoccupation over one year. Findings are discussed in light of theory and provide practical suggestions for future research and interventions among youth.

Abstract:Abstract Holistic emerging approaches allow us to understand that every organism is the result of integration mechanisms observed at every level of nature: integration of DNA from virus and bacteria in metazoans, endosymbiotic relationships and holobionts. Horizontal gene transfer events in Bacteria, Archaea and Eukaryotes have resulted in the chimeric nature of genomes. As a continuity of this genomic landscape, the human body contains more bacterial than human cells. Human microbiome has co-evolved with the human being as a unity called holobiont. The loss of part of our microbiome along evolution can explain the continuous increasing incidence of immune and inflammatory-related diseases. Life is a continuous process in which the organism experiences its environment and this interaction impacts in the epigenetic system and the genomic structure. The emerging perspectives restitute the great importance of Lamarck’s theoretical contributions (the milieu) and Darwin’s pangenesis theory.

Abstract:Background: Theory and research indicate that an appearance focused self-concept (i.e., placing overriding importance on physical appearance for self-definition and self-worth) plays a role in the etiology and maintenance of disordered eating and eating disorders. Although the consequences of an appearance focused self-concept are palpable, less is known about its correlates. Accordingly, we examined a range of factors that may characterize appearance focused people, including personality traits (perfectionism, impulsivity, sensation-seeking, hopelessness, and anxiety sensitivity), self-concept (global self-esteem and self-concept clarity), sociocultural (thin-ideal, muscular-ideal, general attractiveness internalizations, and perceived pressure to be thin), and early life experiences (adverse childhood experiences, attachment styles) factors. Methods: Female undergraduate university students (N = 568; Mage = 19.58, SDage = 4.24) completed a questionnaire battery that included the Beliefs About Appearance Scale, Depressive Experiences Questionnaire-Self-Criticism-6 Scale, Frost-Multidimensional Perfectionism Scale, the Revised Almost Perfect Scale, Substance Use Risk Profile Scale, Self-Concept Clarity Scale, Rosenberg Self-Esteem Scale, Sociocultural Attitudes Towards Appearance Questionnaire-4 Scale, Adverse Childhood Experiences Questionnaire, Experiences in Close Relationships Scale Short Form, and the Dietary Restraint subscale of the Eating Disorders Examination Questionnaire. Results: Multiple regression analyses were conducted for each set of factors separately and together. For personality traits, perfectionism, impulsivity, and anxiety sensitivity were uniquely associated with appearance focused self-concept. For self-concept, global self-esteem and self-concept clarity were uniquely associated with appearance focused self-concept. For sociocultural, general attractiveness internalization, thin-ideal internalization, and perceived pressure to be thin were uniquely associated with appearance focused self-concept. For early life experiences, attachment anxiety and avoidance were uniquely associated with appearance focused self-concept. In the combined analysis, the various factors explained 54% of the variance in appearance focused self-concept. Impulsivity, global self-esteem, general attractiveness internalization, and perceived pressure to be thin were uniquely associated with appearance focused self-concept. Conclusions: Results for global self-esteem were consistent with prior research. Findings for evaluative concerns perfectionism were inconsistent with prior research. We discuss future research directions to examine the link between evaluative concerns perfectionism and appearance focused self-concept. We also discuss how sociocultural factors (general attractiveness internalization and perceived pressure to be thin) and impulsivity may help cultivate an appearance focused self-concept, advancing knowledge on the characteristics of appearance-focused people.

Abstract:Nature-based interventions have demonstrated positive effects on health behaviours and psychological states. Recent research has expanded to investigate the impact of nature exposure on body image, leaving ample room for further exploration into the influence of nature exposure on positive concerns related to appearance management. This paper examined the effect of nature exposure on body appreciation, adaptive appearance management, and the underlying psychological mechanism through an outdoor field study and two psychological experiments. Study 1 demonstrated that natural outdoor recreation increased body appreciation, and this effect was mediated by individuals' level of nature connectedness. Study 2 found that viewing images of natural outdoor environments facilitated greater adaptive appearance management. This relationship was sequentially mediated by both nature connectedness and body appreciation. Study 3 conducted a field study and revealed that viewing images of natural outdoor environments not only fostered body appreciation but also influenced participants’ intention to purchase grooming products, which are regarded as practices related to adaptive appearance management. These findings provide empirical evidence regarding the beneficial outcomes of exposure to natural outdoor environment, thereby contributing to the advancement of knowledge in the domain of body image and appearance management.
