Abstract:Adverse effects of manufactured nickel oxide nanoparticles on the microalgae Chlorellavulgaris were determined by algal growth-inhibition test and morphological observation via transmission electron microscopy (TEM). Results showed that the NiO nanoparticles had severe impacts on the algae, with 72 h EC(50) values of 32.28 mg NiOL(-1). Under the stress of NiO nanoparticles, C. vulgaris cells showed plasmolysis, cytomembrane breakage and thylakoids disorder. NiO nanoparticles aggregated and deposited in algal culture media. The presence of algal cells accelerated aggregation of nanoparticles. Moreover, about 0.14% ionic Ni was released when NiO NPs were added into seawater. The attachment of aggregates to algal cell surface and the presence of released ionic Ni were likely responsible for the toxic effects. Interestingly, some NiO nanoparticles were reduced to zero valence nickel as determined by X-ray diffraction (XRD) and X-ray photoelectron spectroscopy (XPS) analysis. The maximum ratios of nickel reduction was achieved at 72 h of exposure, in accordance with the time-course of changes in soluble protein content of treated C. vulgaris, implying that some proteins of algae are involved in the process. Our results indicate that the toxicity and bioavailability of NiO nanoparticles to marine algae are reduced by aggregation and reduction of NiO. Thus, marine algae have the potential for usage in nano-pollution bio-remediation in aquatic system.
Stability of nickel oxide nanoparticles and its influence on toxicity to marine algae Chlorella vulgaris

Abstract:This study considered the stability of nickel oxide nanoparticles (nNiO) in seawater including their ability of aggregation and ion release. Furthermore, the relationship between these properties and their toxicity on marine algae Chlorella vulgaris was investigated. The results showed nNiO inhibited the growth of algal cells and decreased their chlorophyll content, which was due to the shading effects by aggregation of nNiO in seawater. Moreover, the release of Ni2+ depended on concentration of the nNiO solution. About 1.63% Ni2+ (varied from 0.89 to 3.63%) was detected and it may mediate the generation of ROS under both visible light and ultraviolet (UV) irradiation, which resulted in oxidative stress in algae. Therefore, the stability of nNiO in water affected its toxicity, which should be considered when assessing the nano-pollution risks in aquatic ecosystem.

Abstract:Current scientific evidence suggests the frozen elephant trunk (FET) technique plays an important role in modern aortic arch repair operations, both for aneurysmal disease and acute aortic dissection. Its use in extended aneurysm is generally therapeutic, aiming for complete exclusion of the diseased descending thoracic aorta. In acute aortic dissection type A, the application of FET is more prophylactic in nature, with the aim of preventing late dilatation of the proximal descending thoracic aorta. This review will present the journey of the elephant trunk from its conception to the current available technology. By tracing the historical evolution of the FET technique and prosthesis development, we explore the challenges and limitations of evidence-based surgical research. We present data from our growing experience in aortic arch reconstruction, the results from our latest 27 patients undergoing the 4-branch FET indicating substantially reduced morbidity and mortality (0%) in this complex patient cohort.
Toxicity of Oxide Nanoparticles to the Green Algae Chlorella Sp.

Abstract:Increasing studies focus on nanotoxicity, but many unknowns are remaining to be investigated. This study examined toxicities of four varieties of oxide nanoparticles (Al2O3, SiO2, ZnO, and TiO2) to the green algae Chlorella sp. Nanoparticulate Al2O3, SiO2, and TiO2 (DJ3, rutile) had no significant toxicity, whereas nano-ZnO and nano-TiO2 (HR3, anatase) greatly inhibited the algal growth with 6d EC30 of ca. 20 and 30mgL−1, respectively. Comparative experiments showed the algal toxicities decreased with an order of Zn2+>nano-ZnO>bulk-ZnO when the concentration of the test material was lower than 50mgL−1. However, nano-ZnO had higher algal toxicity than Zn2+ ions at concentrations >50mgL−1 and released less Zn2+ ions into the culture media than bulk-ZnO, which suggests that dissolved Zn2+ ions from nano-ZnO were not the dominant mechanism for the algal growth inhibition. Shading effects of nano-ZnO and HR3 were also excluded from the main mechanism of the nanotoxicity by the comparison of the nanotoxicities in the presence and absence of illumination. Large aggregates of nano-ZnO and HR3 entrapping and wrapping the algal cells were observed, which may contribute to the nanotoxicity. These results will be helpful for understanding the toxicity of oxide nanoparticles.
Evaluation of the Toxicity of ZnO Nanoparticles to Chlorella Vulgaris by Use of the Chiral Perturbation Approach

Abstract:The toxicity of ZnO nanoparticles (NPs) has been widely investigated because of their extensive use in consumer products. The mechanism of the toxicity of ZnO NPs to algae is unclear, however, and it is difficult to differentiate between particle-induced toxicity and the effect of dissolved Zn2+. In the work discussed in this paper we investigated particle-induced toxicity and the effects of dissolved Zn2+ by using the chiral perturbation approach with dichlorprop (DCPP) as chiral perturbation factor. The results indicated that intracellular zinc is important in the toxicity of ZnO NPs, and that ZnO NPs cause oxidative damage. According to dose–response curves for DCPP and the combination of ZnO NPs with (R)-DCPP or (S)-DCPP, the toxicity of DCPP was too low to perturb the toxicity of ZnO NPs, so DCPP was suitable for use as chiral perturbation factor. The different glutathione (GSH) content of algal cells exposed to (R)-DCPP or (S)-DCPP correlated well with different production of reactive oxygen species (ROS) after exposure to the two enantiomers. Treatment of algae with ZnO NPs and (R)-DCPP resulted in reduced levels of GSH and the glutathione/oxidized glutathione (GSH/GSSG) ratio in the cells compared with the control. Treatment of algae with ZnO NPs and (S)-DCPP, however, resulted in no significant changes in GSH and GSH/GSSG. Moreover, trends of variation of GSH and GSH/GSSG were different when algae were treated with ZnSO4·7H2O and the two enantiomers. Overall, the chiral perturbation approach revealed that NPs aggravated generation of ROS and that released Zn2+ and NPs both contribute to the toxicity of ZnO NPs.

Abstract:Usage of nanoparticle in various products has increased tremendously in the recent past. Toxicity of these nanoparticles can have a huge impact on aquatic ecosystem. Algae are the ideal organism of the aquatic ecosystem to understand the toxicity impact of nanoparticles. The present study focuses on the toxicity evaluation of zinc oxide (ZnO) and iron oxide (Fe2O3) nanoparticles towards freshwater microalgae, Chlorella vulgaris. The dose dependent growth retardation in Chlorella vulgaris is observed under ZnO and Fe2O3 nanoparticles and nanoform attributed more toxicity than their bulk counterparts. The IC50 values of ZnO and Fe2O3 nanoparticles was reported at 0.258 mg L-1 and 12.99 mg L-1 whereas, for the bulk-form, it was 1.255 mgL-1 and 17.88 mg L-1, respectively. The significant decline in chlorophyll content and increase in proline content, activity of superoxide dismutase and catalase, indicated the stressful physiological state of microalgae. An increased lactate dehydrogenase level in treated samples suggested membrane disintegration by ZnO and Fe2O3 nanoparticles. Compound microscopy, scanning electron microscopy and transmission electron microscopy confirm cell entrapment, deposition of nanoparticles on the cell surface and disintegration of algal cell wall. Higher toxicity of nanoform in comparison to bulk chemistry is a point of concern.

Abstract:The potential toxicity of CoFe2O4 nanobeads (NBs) in Chlorella vulgaris was observed up to 72h. Algal cell morphology, membrane integrity and viability were severely compromised due to adsorption and aggregation of NBs on algal surfaces, release of Fe(3+) and Co(2+) ions and possible mechanical damage by NBs. Interactions with NBs and effective decrease in ions released by aggregation and exudation of algal cells as a self defense mechanism were observed by Fourier transform infrared attenuated total reflectance (FTIR-ATR) and inductively coupled plasma mass spectrometry (ICP-MS). The results corroborated CoFe2O4 NBs induced ROS triggered oxidative stress, leading to a reduction in catalase activity, activation of the mutagenic glutathione s-transferase (mu-GST) and acid phosphatase (AP) antioxidant enzymes, and an increase in genetic aberrations, metabolic and cellular signal transduction dysfunction. Circular dichroism (CD) spectra indicated the weak interactions of NBs with BSA, with slight changes in the α-helix structure of BSA confirming conformational changes in structure, hence the potential for functional interactions with biomolecules. Possible interferences of CoFe2O4 NBs with assay techniques and components indicated CoFe2O4 NBs at lower concentration do not show any significant interference with ROS, catalase, mu-GST and no interference with CD measurements. This study showed ROS production is one of the pathways of toxicity initiated by CoFe2O4 NBs and illustrates the complex processes that may occur between organisms and NBs in natural complex ecosystem.
Inhibitory effects of silver nanoparticles in two green algae, Chlorella vulgaris and Dunaliella tertiolecta

Abstract:Freshwater microalga Chlorella vulgaris and marine microalga Dunaliella tertiolecta were used to investigate toxic effects induced by 50 nm silver nanoparticles (AgNPs). To induce AgNPs effect, we exposed Chlorella vulgaris and Dunaliella tertiolecta for 24h to 0-10 mg/L. We showed that growth media had different effects in AgNPs agglomerates' formation. Cellular viability, reactive oxygen species (ROS) formation and lipids peroxidation were employed to assess the toxic effects of AgNPs. AgNPs were able to interact directly with the Chlorella vulgaris cells surface and large aggregates were observed. AgNPs have a negative effect on Chlorella vulgaris and Dunaliella tertiolecta, as manifested by a strong decrease in chlorophyll content, viable algal cells, increased ROS formation and lipids peroxidation. The variability in sensitivity of both algae towards AgNPs was observed. We conclude that AgNPs have a negative effect on aquatic algae and these alterations might have serious consequences on structure and function of aquatic plant communities.
Toxicity of nanoparticles of CuO, ZnO and TiO2 to microalgae Pseudokirchneriella subcapitata

Abstract:Toxicities of ZnO, TiO2 and CuO nanoparticles to Pseudokirchneriella subcapitata were determined using OECD 201 algal growth inhibition test taking in account potential shading of light. The results showed that the shading effect by nanoparticles was negligible. ZnO nanoparticles were most toxic followed by nano CuO and nano TiO2. The toxicities of bulk and nano ZnO particles were both similar to that of ZnSO4 (72 h EC50 approximately 0.04 mg Zn/l). Thus, in this low concentration range the toxicity was attributed solely to solubilized Zn2+ ions. Bulk TiO2 (EC50=35.9 mg Ti/l) and bulk CuO (EC50=11.55 mg Cu/l) were less toxic than their nano formulations (EC50=5.83 mg Ti/l and 0.71 mg Cu/l). NOEC (no-observed-effect-concentrations) that may be used for risk assessment purposes for bulk and nano ZnO did not differ (approximately 0.02 mg Zn/l). NOEC for nano CuO was 0.42 mg Cu/l and for bulk CuO 8.03 mg Cu/l. For nano TiO2 the NOEC was 0.98 mg Ti/l and for bulk TiO2 10.1 mg Ti/l. Nano TiO2 formed characteristic aggregates entrapping algal cells that may contribute to the toxic effect of nano TiO2 to algae. At 72 h EC50 values of nano CuO and CuO, 25% of copper from nano CuO was bioavailable and only 0.18% of copper from bulk CuO. Thus, according to recombinant bacterial and yeast Cu-sensors, copper from nano CuO was 141-fold more bioavailable than from bulk CuO. Also, toxic effects of Cu oxides to algae were due to bioavailable copper ions. To our knowledge, this is one of the first systematic studies on effects of metal oxide nanoparticles on algal growth and the first describing toxic effects of nano CuO towards algae.
Toxicity of the mixture of nanoparticles to algae–bacteria consortia in OECD media

Abstract:The present study investigated the effect of single as well as a mixture of nanoparticles (ZnO and CuO NPs) on algae–bacteria consortia using the OECD 96-h toxicity test, one of the first efforts as per the authors' knowledge. Microalgae ( Scenedesmus obliquus ) and bacteria ( Escherichia coli ) were used as test organisms in OECD media. Effects of the different concentrations of NPs (0, 0.1 mg L −1 , 1 mg L −1 , 10 mg L −1 , and 100 mg L −1 ) on 3 algae–bacteria ratios (1 : 1, 1 : 100 and 100 : 1) were studied using parameters, such as chlorophyll, biomass, lipid, and protein contents, reactive oxygen species (ROS) generation, and extracellular polymeric substance (EPS) components. At environmentally relevant NP concentration (0.1 mg L −1 ), the order of toxicity of NPs to algae–bacteria consortia was found to be: CuO NPs (highest toxicity) > ZnO + CuO NPs > ZnO NPs (least toxicity). At 100 mg L −1 NP concentration, structural changes and cell leakage in the samples containing NPs with algae–bacteria consortia were observed during TEM analysis. FTIR (Fourier transform infra-red) analysis indicated the addition of bonds and a difference in the peak location and its intensity values. The corresponding metal ions (Zn and Cu ions) resulted in higher toxicity to algae–bacteria consortia than metal oxide NPs. When nanoparticles were interacting with the algae–bacteria consortia in the suspension, it was observed that by absorption and dissolution, nanoparticles would enter inside the algal and bacterial cells simultaneously, altering the surface charge of the cell membrane. Due to the formation of EPSs, some of the nanoparticles would not be able to enter the cell cytoplasm but would interact with the EPS. ROS generation would take place extracellularly as well as intracellularly due to the interaction of nanoparticles. Overall, the mixture of NPs at environmentally relevant concentrations (<1 mg L −1 ) poses lower toxicity to algae–bacteria consortia than individual nanoparticles. More experimental studies need to be performed to understand the effect of NPs on the functioning of algae–bacteria consortia for effective wastewater treatment using algae–bacteria consortia.

Interactions of CuO nanoparticles with the algae Chlorella pyrenoidosa: adhesion, uptake, and toxicity

Abstract:The potential adverse effects of CuO nanoparticles (NPs) have increasingly attracted attention. Combining electron microscopic and toxicological investigations, we determined the adhesion, uptake, and toxicity of CuO NPs to eukaryotic alga Chlorella pyrenoidosa. CuO NPs were toxic to C. pyrenoidosa, with a 72 h EC50 of 45.7 mg/L. Scanning electron microscopy showed that CuO NPs were attached onto the surface of the algal cells and interacted with extracellular polymeric substances (EPS) excreted by the organisms. Transmission electron microscopy (TEM) showed that EPS layer of algae was thickened by nearly 4-fold after CuO NPs exposure, suggesting a possible protective mechanism. In spite of the thickening of EPS layer, CuO NPs were still internalized by endocytosis and were stored in algal vacuoles. TEM and electron diffraction analysis confirmed that the internalized CuO NPs were transformed to Cu2O NPs (d-spacing, ∼0.213 nm) with an average size approximately 5 nm. The toxicity investigation demonstrated that severe membrane damage was observed after attachment of CuO NPs with algae. Reactive oxygen species generation and mitochondrial depolarization were also noted upon exposure to CuO NPs. This work provides useful information on understanding the role of NPs-algae physical interactions in nanotoxicity.
Toxicity of Iron-Based Nanoparticles to Green Algae: Effects of Particle Size, Crystal Phase, Oxidation State and Environmental Aging.

Abstract:With the increasing environmental application and discharge of iron-based nanoparticles (NPs), a comprehensive understanding of their fate and ecotoxicological effect in the aquatic environment is very urgent. In this study, toxicities of 4 zero-valent iron NPs (nZVI) of different sizes, 2 Fe2O3 NPs of different crystal phases, and 1 type of Fe3O4 NPs to a green alga (Chlorella pyrenoidosa) were investigated, with a focus on the effects of particle size, crystal phase, oxidation state, and environmental aging. Results show that the algal growth inhibition of nZVI increased significantly with decreasing particle size; with similar particle sizes (20-30 nm), the algal growth inhibition decreased with oxidation of the NPs with an order of nZVI > Fe3O4 NPs > Fe2O3 NPs, and (z-Fe2O3 NPs presented significantly higher toxicity than y-Fe2O3 NPs. The NP-induced oxidative stress was the main toxic mechanism, which could explain the difference in algal toxicity of the NPs. The NP-cell heteroagglomeration and physical interactions also contributed to the nanotoxicity, whereas the effect of NP dissolution was negligible. The aging in distilled water and 3 surface water samples for 3 months increased surface oxidation of the iron-based NPs especially nZVI, which decreased the toxicity to algae. These findings will be helpful for the understanding of the fate and toxicity of iron-based NPs in the aquatic environment. (C) 2016 Elsevier Ltd. All rights reserved.
Interaction of Fe2O3 nanoparticles with marine microalga Chlorella sorokiniana: Analysis of growth, morphological changes and biochemical composition

Abstract:The wide utilization of iron-based nanoparticles (NPs) based on their preferential properties has led to the discharge and accumulation of these materials into the aquatic environment. In this regard, a comparative study of different concentrations of α-Fe2O3 NPs and their micro form was conducted using microalga Chlorella sorokiniana up to the stationary growth phase. This study revealed that high concentrations of NPs (100 and 200 mg L-1) imposed a stressful condition on algal cells documented by a reduction in microalga growth, including cell number and specific growth rate. The physical contact between the algal cells and NPs resulted in a shading effect as well as morphological changes validated by scanning electron microscope results. The biochemical composition of C. sorokiniana exposed to high levels of Fe2O3 NPs was also evaluated. The increase in total carbohydrate content of algal cells along with a significant reduction in unsaturated fatty acids was found. Moreover, Fe2O3 NPs exposure induced oxidative stress evidenced by an increase in lipid peroxidation. To cope with oxidative stress, superoxide dismutase activity and antioxidant potential of microalga as defensive mechanisms increased in the culture with high concentrations of NPs. Besides, due to the interactions, microalga tended to form a protective layer from further cell-NP interactions through the secretion of extracellular polymeric substances. Nonetheless, the nano form of Fe2O3 was more toxic than its micro form due to its small size. Overall, this trial may provide additional insight into the toxicological mechanism and safety assessments of Fe2O3 NPs in the aquatic environment.
Toxicity evaluation of iron oxide nanoparticles to freshwater cyanobacteria Nostoc ellipsosporum

Abstract:The extensive usage of iron oxide nanoparticles (FeO NPs) in commercial and biomedical applications raises the risk of releasing their remains into the aquatic ecosystems and this could possibly cause cytotoxic effects on aquatic organisms. Thus, the toxicity assessment of FeO NPs on cyanobacteria, which are primary producers at the bottom of food chain in aquatic ecosystems, is essential to gain information about the potential ecotoxicological threat on aquatic biota. The present study investigated the cytotoxic effects of FeO NPs on Nostoc ellipsosporum using different concentrations (0, 10, 25, 50 and 100 mg L −1 ) to track the time-dependent and dose-dependent effects and compared with its bulk equivalent. In addition, the impacts of FeO NPs and bulk counterpart on cyanobacterial cells were assessed under nitrogen as well as nitrogen-deficient conditions, because of ecological role of cyanobacteria in nitrogen fixation. The study revealed that the highest protein content was observed in the control in both types of BG-11 media compared to treatments of nano and bulk particles of Fe 2 O 3 . A 23% reduction in protein in nanoparticle treatment and a 14% reduction in bulk treatment at 100 mg L −1 was observed in BG-11 medium. At same concentration, in BG-11 0 media, this decline was even more intense with 54% reduction in nanoparticle and a 26% reduction in bulk. Catalytic activity of catalase and superoxide dismutase was found to be linearly correlated with the dose concentration for nano and bulk form in BG-11 as well as BG-11 0 media. The increased levels of lactate dehydrogenase act as biomarker of the cytotoxicity brought on by nanoparticles. Optical, scanning electron, and transmission electron microscopy all demonstrated the cell entrapment, nanoparticle deposition on the cell surface, cell wall collapse and membrane degradation. A cause for concern is that nanoform was found to be more hazardous than bulk form.

Abstract:Extensive application of nanomaterials enlarges its concentrations in the aquatic environments and poses a threat to algae. This study comprehensively analyzed the physiological and transcriptional responses of Chlorella sp. after being exposed to chromium (III) oxide nanoparticles (nCr2O3). The nCr2O3 at 0–100 mg/L presented adverse effects on cell growth (96 h EC50 = 16.3 mg/L), decreasing the photosynthetic pigment concentrations and photosynthetic activity. Moreover, more extracellular polymeric substances (EPS), especially polysaccharides in soluble EPS, were produced in algae cell, which mitigated the damage of nCr2O3 to cells. However, with the increase of nCr2O3 doses, the EPS protective responses were exhausted, accompanied by toxicity in the form of organelle damage and metabolic disturbance. The enhanced acute toxicity was closely related to the physical contact of nCr2O3 with cells, oxidative stress, and genotoxicity. Firstly, large amounts of nCr2O3 aggregated around and were attached to cells, causing physical damage. Then, the intracellular reactive oxygen species and malondialdehyde levels were significantly increased that led to lipid peroxidation, especially at 50–100 mg/L nCr2O3. Finally, the transcriptomic analysis further revealed that the transcription of ribosome, glutamine, and thiamine metabolism-related genes were impaired under 20 mg/L nCr2O3, suggesting nCr2O3 inhibited algal cell growth through metabolism, cell defense, and repair, etc.Graphical Abstract

Abstract:The present study investigated the possible toxic effect of ZnO and CuO nanoparticles (NPs) on freshwater microalgae, Scenedesmus obliquus at environmentally- relevant nanoparticle concentration (1 mg/L) and high concentration (10 mg/L) in BG-11 medium under white light LED-illumination over 35 days. The effect of time on the stability of media, nanoparticles, and their relation to toxicity to algae was also studied. The transmission electron microscopy indicated structural damage to algae due to the presence of a mixture of nanoparticles (at 10 mg/L). FTIR (Fourier Transform infrared) analysis of a sample containing a mixture of nanoparticles showed an addition of bonds and a difference in the peak location and its intensity values. The inhibition time for biomass was observed between 14 days and 21 days at 10 mg/L NPs. At 1 mg/L, the order of toxicity of NPs to algae was found to be: CuO NPs (highest toxicity) > ZnO NPs>ZnO + CuO NPs (least toxicity). During exposure of algae cells to a mixture of NPs at 10 mg/L NP concentration, a smaller value of metal deposition was observed than that during exposure to individual NPs. Antagonistic toxic effects of two NPs on dry cell weight of algae was observed at both concentration levels. Future work is needed to understand the steps involved in toxicity due to mixture of NPs to algae so that environmental exposures of algae to NPs can be managed and minimized.

Abstract:This study aims to assess the toxicity of the commonly-spread titanium dioxide nanoparticles (TiO2 NPs) by evaluating the exposure impact of the particles on both freshwater algae Chlorella pyrenoidosa and zebrafish liver cell line (ZFL), the two common in vitro models in toxicological studies. To compare the toxic effects of TiO2 NPs with different physiochemical properties, three types of manufactured TiO2 were used: bulk TiO2, Degussa P25 TiO2, and ultrafine TiO2 NPs. Both short and long-term biological responses of green algae, such as the effect on the cell growth rate, pigment autofluorescence, and esterase activity were investigated. The dosage, physical property of TiO2 particles, and their interactions with algal cells affect cellular growth, especially after short-term exposure. The hydrodynamic size plays a critical role in determining the acute toxicity to C. pyrenoidosa in terms of autofluorescence and esterase activity, while all types of TiO2 NPs show toxic effects after exposure for 14 days. However, this observation is not seen when studying the effect of introduced particles in ZFL, for the precipitated Degussa P25 TiO2 showed the highest cellular inhibition. Interestingly, despite the obvious overall toxicity toward C. pyrenoidosa, the photocatalytical properties of TiO2 NPs may contribute to the enhanced photosynthesis in the low concentration range (< 40 µg mL−1). Overall, we found that the physical interactions between TiO2 particles and the cells, particles' size and dispersibility play critical role in the cytotoxic effect for both algal and ZFL cells, while the photocatalytical properties of TiO2 particles may produce mixed effects on the cytotoxicity of green algae.

Abstract:Evaluation of 50 nm zinc oxide nanoparticles’ (ZnO-NPs) effects on the microalgae Chlorococcum sp. growing in high salt growth medium (HSM) was investigated by using flow cytometry parameters (cell size (FSC), granularity (SSC), chlorophyll a fluorescence (FL3), and formation of reactive oxygen species (ROS)). Algal cells in exponential growth were exposed to 0–100 mg/L of ZnO-NPs and their physiological responses were measured after 24 and 96 h of treatment. Behavior of ZnO-NPs was analyzed in HSM and results indicated that ZnO-NPs formed agglomeration with a large distribution. Total soluble Zn concentration increased when initial ZnO-NP concentration increased. Significant negative effect on algal cells was observed after 96 h exposition and at high ZnO-NP concentration. This negative impact was evaluated by the significant increase in ROS production, inhibition in the photosynthetic electron transport, and reduction in cell growth. In this study, using flow cytometry multi-parameters might help to prevent and evaluate inhibitory effect of oxide nanoparticles on aquatic photosynthetic microorganisms.

Unraveling the risks of nAl2O3 on harmful algal blooms: Insights from paralytic shellfish toxins production of Alexandrium tamarense

Abstract:As one of the commonly used and cost-effective nanomaterials, nanosized aluminum oxide (nAl2O3) posses unique properties and chemical stability. However, its extensive use and resultant dissemination into aquatic ecosystems prompt concerns over the proliferation and repercussions of harmful algal blooms, particularly those caused by dinoflagellates producing toxins. This study investigated the sub-chronic effects of nAl2O3 on growth, physiological activities, and paralytic shellfish toxins (PSTs) production in Alexandrium tamarense. Results showed dose-dependent inhibition in growth (EC50 = 20.6 mg L-1), esterase activity, and photosynthetic efficiency (Fv/Fm) during the sub-chronic exposure (13-day). The internalization of nAl2O3 in microalgal cells and the significant decrease in the total cellular PSTs content were observed under high nAl2O3 concentrations (>40 mg L-1). The study also demonstrated a clear decrease in the content of some derivatives of PSTs (GTX5, C1/2, and GTX2/3) with the increase in nAl2O3 concentrations, accompanied by the induction of an unknown derivative. Excessive ROS production, dissolved Al, and physical inhibition were suggested as potential mechanisms for nAl2O3 toxicity and changes in PSTs toxin profile. Overall, this research enhances our understanding of the potentiated risks and threats on the possible concurrent events of toxic dinoflagellate, such as Alexandrium species and nanoparticles in aquatic environments.
Weekly Docetaxel and Gemcitabine following Docetaxel plus Epirubicin or Vinorelbine as First-Line Treatment of Metastatic Breast Cancer: Results of a Multicenter Phase II Study

Abstract:Aims and Background Sequential docetaxel and gemcitabine following initial docetaxel plus epirubicin or vinorelbine association could be worthwhile as first-line treatment of metastatic breast cancer. Methods Fifty-eight patients entered a phase II study that included two sequential phases. In the first phase, 36 and 22 patients previously unexposed or exposed to adjuvant anthracyclines received the association of docetaxel (75 mg/m2, day 1) with epirubicin (75 mg/m2, day 1) or vinorelbine (20 mg/m2, days 1 and 5), respectively, every 21 days for 4 courses. In the second phase, patients who had a response (R) or stable disease (SD) received docetaxel (35 mg/m2) and gemcitabine (800 mg/m2) on days 1, 8 and 15 every 28 days for 4 courses. Results In the first phase, grade ≥ III neutropenia occurred in 51% and 37% of patients during docetaxel-epirubicin and docetaxel-vinorelbine, respectively. In the second phase, it occurred in the 27% and 15% of patients initially treated with docetaxel-epirubicin and docetaxel-vinorelbine, respectively. On an intention to treat basis, the complete (CR) + partial response (PR) rate to the first phase was 71%, and 22% of patients had SD, without a significant difference between the docetaxel-epirubicin and docetaxel-vinorelbine arms. After the second phase, the CR + PR rate was 65%, and 14% of patients had SD. Median time to progression and survival were 12.1 and 22.0 months, respectively, without a significant difference between patients initially treated with docetaxel-epirubicin and docetaxel-vinorelbine. Conclusions Following an initial docetaxel-based treatment, weekly docetaxel and gemcitabile maintains high percentages of R and SD, with improved toxicity. Survival was similar in patients previously untreated and treated with adjuvant anthracyclines.
