Abstract:Pupilloconstriction to light can be mediated in rats through direct illumination of retinae previously transplanted to intracranial locations. Transplant-driven and normal pupillary light responses are stable under optimal testing conditions, and parameters describing the response can be quantified precisely. The present study demonstrates the interaction between transplant-driven and normal pupillary response patterns. When stimuli are presented concurrently to a transplanted retina and to the remaining eye in host rats, a greater degree of pupilloconstriction occurs than when either the transplanted or the host eye is illuminated independently. This suggests that transplant and host retinal inputs act in concert to determine pupil diameter. The second portion of this study investigates the pattern of retinal input to the pretectum to determine if a structural basis for such functional interactions may exist. Crossed and uncrossed retinal projections to the olivary pretectal nucleus occupy non-overlapping regions of this bilaterally represented nucleus in normal rats, with a greater number of optic axons directed to the contralateral olivary pretectal nucleus. Retinae transplanted to the midbrain of neonatal rats, from whom the contralateral eye had been removed, also project to the olivary pretectal nucleus at maturity. By contrast with the normal pattern of segregated retinal inputs, however, the terminal fields of transplant axons were found to overlap extensively with the retinal projection from the remaining host eye. In addition, the relative proportion of transplant axons directed to the ipsilateral and contralateral olivary pretectal nucleus varied among animals. The lack of spatial segregation between inputs from transplant and host sources and the relative proportion of ipsilateral and contralateral transplant axons together may represent a structural basis for the observed functional interactoin of these inputs to the neural circuit subserving pupilloconstriction to light. These features may also relate to the marked improvements in transplant-mediated responses that frequently occur when optic input from the remaining host eye is eliminated. The results presented here, together with our previous transplant studies, show that this preparation can be used to provide insight into more general questions as to the dynamic interactions that occur between converging sensory inputs in the generation of integrated output responses.

Abstract:The major goal of regenerative medicine is to repair damaged tissues and organ systems, thereby restoring their native functions in the host. Control of innervation by re-grown or implanted structures, and integration of the nascent nerves into behavioral/cognitive programs of the host, remains a critical barrier. In the case of sensory organs, this is particularly true, as afferent neurons must form connections with the host to communicate auditory, visual, and tactile information. Xenopus embryos and tadpoles are powerful models for such studies, as grafting techniques allow for the creation of eyes and other sensory structures along the body axis, and the behavior of the resulting organism can be quantitatively analyzed. Previous work has demonstrated that ectopic eyes could be grafted in blinded tadpoles, allowing some of the animals to learn in a simple light-preference assay. Here, we show that it is possible to improve the efficiency of the process in the context of a novel image-forming vision assay, using a drug already approved for human use. Innervation of the host by ectopic eyes can be increased by targeting a serotonergic signaling mechanism: grafts treated with a 5-HT1B/D agonist strongly innervate the recipient compared with untreated grafts, without large-scale disruption of the host nervous system. Blind animals possessing eye grafts with the augmented innervation demonstrate increased performance over untreated siblings in wavelength-based learning assays. Furthermore, treated animals also exhibit enhanced visual pattern recognition, suggesting that the increased innervation in response to 5-HT1B/D activation leads to enhanced functional integration of the ectopic organ with the host central nervous system and behavioral programs. These data establish a model system and reveal a new roadmap using small molecule neurotransmitter drugs to augment innervation, integration, and function of transplanted heterologous organs in regenerative medicine.

Abstract:Background Transplantation of neural cells is a promising stroke recovery therapy requiring animal model research, but rejection of human xenografts is an obstacle. While a variety of immunosupression strategies have been reported, few comparison data are available. We asked if different cyclosporine regimens would affect rat brain responses to human neural cell grafts. Methods Rats received intracerebral grafts of neural progenitors derived from human embryonic stem cells, and 6 rats each were randomized to 1 of 4 immunosupression groups: 1) SC (subcutaneous) group - cyclosporine 10 mg/kg/day SC starting 2 days prior to transplantation; 2) SC/PO (per os) group - the same injections for the first 7 days followed by 100 ug/ml in the drinking water; 3) PO group - only cyclosporine in the drinking water starting 2 days prior to transplantation; 4) Control - no immunosupression. Immunostaining, stereology, and optical densitometry were performed after 2 weeks for quantification of markers of human cells, neural cell types, and immune cells. Results More rats in the SC (6/6) and SC/PO (5/6) groups had surviving graft cells than the PO (1/6) and control (3/6) groups (p<0.05), with a trend toward more surviving graft cells as well. All rats with surviving graft cells showed co-labeling for the neural progenitor marker Nestin, and a minority of graft cells co-labeled for the cell division marker Ki67 and the neuronal marker MAP2. Optical densitometry showed more cellularity (p<0.05) and reactive astrocytosis (p<0.05) in grafted versus contralateral hemispheres, which was similar between groups. Areas of dead graft cell debris were seen in all groups; in these areas the human nuclear antigen was present in the cytoplasm of cells co-labeling for the microglia marker CD11b, suggesting that they had phagocytosed graft cells. Conclusions The survival of human neural cells in rat brains differed with immunosupression regimens, but immune cell infiltration, phagocytosis of graft cells, and reactive astrocytosis occurred in all groups. Frequent injections of laboratory animals is undesirable, and a compromise strategy of peri-transplant injections followed by mixing the drug in the drinking water showed good results to prevent graft cell rejection. Further research is needed to optimize the immunosupression approach for this application.

Abstract:Intravitreal transplantation of allogeneic human retinal progenitor cells (hRPCs) holds promise as a treatment for blinding retinal degenerations. Prior work has shown that neural progenitors are well-tolerated as allografts following single injections; however, sequential delivery of allogeneic cells raises the potential risk of host sensitization with subsequent immune rejection of grafts. The current study was designed to assess whether an immune response would be induced by repeated intravitreal transplants of allogeneic RPCs utilizing the mouse animal model. We injected murine retinal progenitor cells (gmRPCs), originally derived from donors with a C57BL/6 genetic background, into BALB/c recipient mice in order to provide safety data as to what might be expected following repeated treatment of patients with allogeneic human cell product. Immune responses to gmRPCs were mild, consisting of T cells, B cells, neutrophils, and natural killer cells, with macrophages clearly the predominating. Animals treated with repeat doses of gmRPCs did not show evidence of sensitization, nor was there immune-mediated destruction of the grafts. Despite the absence of immunosuppressive treatments, allogeneic gmRPC grafts survived following repeat dosing, thus providing support for the preliminary observation that repeated injection of allogeneic RPCs to the vitreous cavity is tolerated in patients with retinitis pigmentosa.

Still, it is unknown whether olfactory ensheathing glia also have neuroprotective properties. Olfactory ensheathing glia express brain-derived neurotrophic factor, one of the best neuroprotectants for axotomized retinal ganglion cells. Therefore, we aimed to investigate the neuroprotective capacity of olfactory ensheating glia after optic nerve crush. Olfactory ensheathing glia cells from an established rat immortalized clonal cell line, TEG3, were intravitreally injected in intact and axotomized retinas in syngeneic and allogeneic mode with or without microglial inhibition or immunosuppressive treatments. Anatomical and gene expression analyses were performed. Olfactory bulb-derived primary olfactory ensheathing glia and TEG3 express major histocompatibility complex class II molecules. Allogeneically and syngenically transplanted TEG3 cells survived in the vitreous for up to 21 days, forming an epimembrane. In axotomized retinas, only the allogeneic TEG3 transplant rescued retinal ganglion cells at 7 days but not at 21 days. In these retinas, microglial anatomical activation was higher than after optic nerve crush alone. In intact retinas, both transplants activated microglial cells and caused retinal ganglion cell death at 21 days, a loss that was higher after allotransplantation, triggered by pyroptosis and partially rescued by microglial inhibition or immunosuppression. However, neuroprotection of axotomized retinal ganglion cells did not improve with these treatments. The different neuroprotective properties, different toxic effects, and different responses to microglial inhibitory treatments of olfactory ensheathing glia in the retina depending on the type of transplant highlight the importance of thorough preclinical studies to explore these variables.
Host-derived endothelial regeneration of corneal transplants in a rat keratoplasty model

Abstract:Background: It has been observed that formerly rejected opaque corneal transplants can regain clarity in the rat. We hypothesized that graft endothelium is regenerated by the host. Therefore, we used green fluorescent protein (GFP) transgenic rats to assess the origin of cells following keratoplasty. Methods: Allogeneic corneal transplantations were carried out between Fischer strain rats as graft donors and GFP-transgenic Lewis rats as recipients. In a second group, syngeneic transplantations were performed between GFP-negative Lewis donors and GFP-positive Lewis recipients, where endothelial-cell-free grafts after mechanical endothelial debridement were used. All grafts were followed up clinically for signs of opacity and rejection. After 6 weeks, corneal flatmounts counterstained with DAPI were analyzed by confocal microscopy. Results: Syngeneic transplantation of endothelial-cell-free grafts led to medium opacity levels without rejection and to subsequent clearing. All grafts showed a population of GFP-positive, host-derived endothelial cells on the graft after 6 weeks. In the allogeneic transplantation group, all grafts but one were rejected after a median of 17 days. While the graft that was not rejected maintained the GFP-negative transplant endothelium, all formerly rejected grafts showed GFP-positive endothelium on the transplant after 6 weeks, accompanied by clinical clearing of the graft. Conclusion: GPF positivity shows that in both a syngeneic and an allogeneic setting, the host-derived corneal endothelium can compensate for the endothelial cell loss of the graft. Following rejection, the grafts are repopulated by host-derived endothelial cells in the rat. This finding demonstrates a high regenerative capacity of the peripheral corneal endothelium in the rat, which should be considered whenever interpreting rat keratoplasty results.
Sustained neurotrophic factor cotreatment enhances donor and host retinal ganglion cell survival in mice

Abstract:Retinal ganglion cells (RGCs) lack regenerative capacity in mammals, and their degeneration in glaucoma leads to irreversible blindness. The transplantation of stem cell-derived RGCs lacks clinically relevant effect due to insufficient survival and integration of donor cells. We hypothesize that the retinal microenvironment plays a critical role in this process, and we can engineer a more acceptable setting for transplantation. Since the adult mammalian retina does not have regenerative capacity, we turned to the developing human retina to reconstruct cell-cell interactions at a single-cell level. We established a human fetal retina atlas by integrating currently available single-cell RNA sequencing datasets of human fetal retinas into a unified resource. We align RGC transcriptomes in pseudotime to map RGC developmental fate trajectories against the broader timeline of retinal development. Through this analysis, we identified brain-derived neurotrophic factor (BDNF) and glial-derived neurotrophic factor (GDNF) as key factors in RGC survival, highly expressed during fetal development but significantly reduced in adulthood despite the persistence of their receptors. To demonstrate the practical application of these findings, we show that using a slow-release formulation of BDNF and GDNF enhances RGC differentiation, survival, and function in vitro and improves RGC transplantation outcomes in a mouse model. BNDF/GDNF co-treatment not only increased survival and coverage of donor RGCs within the retina but also showed neuroprotective effects on host RGCs, preserving retinal function in a model of optic neuropathy. Altogether, our findings suggest that manipulating the retinal microenvironment with slow-release neurotrophic factors holds promise in regenerative medicine for treating glaucoma and other optic neuropathies. This approach not only improves donor cell survival and integration but also provides a neuroprotective benefit to host cells, indicating a significant advancement for glaucoma therapies.

Interactive events subserving the pupillary light reflex in pigmented and albino rats

Abstract:The consensual pupilloconstrictor response requires the intensity information that is delivered to each eye to be integrated to produce an averaged response such that the pupillary diameter in each eye is equal. Here we have examined how luminance information from each eye is integrated in the rat and where the integration occurs, using a pupillometry system to record pupillary diameter after illumination of one or both eyes. In albino rats the size of the uncrossed optic pathway to the pupilloconstrictor centre, the olivary pretectal nucleus, is reduced three-fold and there is a concomitant but less dramatic reduction in the size of the consensual response. Unilateral lesions of the olivary pretectal nucleus in pigmented rats restrict all optic input to the opposite pretectum. Stimulation of one or both eyes in these animals suggests that integration occurs not only at the level of the olivary pretectal nucleus, but also downstream in the Edinger-Westphal nucleus. Parallels with human studies and the opportunity to perform secondary lesions suggests the rat may provide a good model in which to study the substrates of the pupilloconstrictor response and mechanisms of integration of convergent parallel inputs in general.
Alterations in sympathetic nerve traffic in genetic haemochromatosis before and after iron depletion therapy: a microneurographic study.

Abstract:AIMS Haemochromatosis (HH) displays a number of circulatory alterations concurring at increase cardiovascular risk. Whether these include sympathetic abnormalities in unknown. METHODS AND RESULTS In 18 males with primary HH (age: 42.3 ± 10.4 years, mean ± SD), clinic and beat-to-beat blood pressure (BP, Finapres), heart rate (HR, EKG), and muscle sympathetic nerve activity (MSNA, microneurography) traffic were measured in the iron overload state and after iron depletion therapy. Haemochromatosis patients displayed elevated serum iron indices while other haemodynamic and metabolic variables were superimposable to ones seen in 12 healthy subjects (C). Muscle sympathetic nerve activity was significantly greater in HH than C (64.8 ± 13.3 vs. 37.8 ± 6.7 bs/100 hb, P < 0.01). Iron depletion caused a significant reduction in serum ferritin, transferrin saturation, and MSNA (from 64.8 ± 13.3 to 39.2 ± 9.2 bs/100 hb, P < 0.01) and a significant improvement in baroreflex-MSNA modulation. This was paralleled by a significant increase in the high-frequency HR variability and by a significant reduction in the low-frequency systolic BP variability components. Before after iron depletion therapy, MSNA was significantly and directly related to transferrin saturation, liver iron concentration, and iron removed, while the MSNA reductions observed after the procedure were significantly and inversely related to the baroreflex-MSNA increases detected after iron depletion. In C, all variables remained unchanged following 1 month observation. CONCLUSION These data provide the first evidence that in HH iron overload is associated with an hyperadrenergic state and a baroreflex alteration, which are reversed by iron depletion. These findings underline the importance of iron overload in modulating sympathetic activation, possibly participating at the elevated cardiovascular risk reported in HH.
Modulation of microglia activation after human donor retinal ganglion cell transplantation

Abstract:Stem cell-derived retinal ganglion cell (RGC) transplantation therapy offers a promising avenue for restoring vision in patients with significant RGC loss. The major challenge in this therapeutic approach is ensuring the survival of transplanted human donor RGCs within the host retina. Previously, we demonstrated the pivotal role of host retinal microglia and macrophages in the acceptance and survival of human donor cells. This current study aimed to improve the survival and integration of human donor RGCs through modulation of the host retinal microglia. Pretreatment of human donor RGCs with annexin V and soluble FasL (sFasL) before transplantation significantly enhanced their acceptance and survival rate. Detailed analyses of the host mouse retina post-transplantation revealed morphological changes and activation patterns in microglia and macrophages. These findings indicate that sFasL and annexin V activate pro-survival pathways and foster a supportive environment for the acceptance and survival of transplanted human donor RGCs.

Transplantation of Human Embryonic Stem Cell-Derived Retinal Tissue in the Subretinal Space of the Cat Eye

Abstract:To develop biological approaches to restore vision, we developed a method of transplanting stem cell-derived retinal tissue into the subretinal space of a large eye animal model (cat). Human embryonic stem cells (hESC) were differentiated to retinal organoids in a dish. HESC-derived retinal tissue was introduced into the subretinal space of wild-type cats following a pars plana vitrectomy. The cats were systemically immunosuppressed with either prednisolone or prednisolone plus cyclosporine A. The eyes were examined by fundoscopy and spectral domain-optical coherence tomography (SD-OCT) imaging for adverse effects due to the presence of the subretinal grafts. Immunohistochemistry was done with antibodies to retinal and human markers to delineate graft survival, differentiation and integration into cat retina. We successfully delivered hESC-derived retinal tissue into the subretinal space of the cat eye. We observed strong infiltration of immune cells in the graft and surrounding tissue in the cats treated with prednisolone. In contrast we showed better survival and low immune response to the graft in cats treated with prednisolone plus cyclosporine A. Immunohistochemistry with antibodies (STEM121, CALB2, DCX and SMI-312) revealed large number of graft-derived fibers connecting the graft and the host. We also show presence of human specific synaptophysin puncta in the cat retina. This work demonstrates feasibility of engrafting human embryonic stem cells derived retinal tissue into the subretinal space of large eye animal models. Transplanting retinal tissue in degenerating cat retina will enable rapid development of preclinical in vivo work focused on vision restoration.

Abstract:ObjectivePhotoreceptor cells transplantation was used to determine the localization of the amino acid excitatory neurotransmitter glutamate in Royal College of Surgeons Rat(RCS rat).MethodThe healthy photoreceptor cells of Wistar rats were grafted into the sub-retinal space of the left eye of the RCS rat by outer-path way.2 weeks later,host animals were sacrificed.Both sides of eyes were examined for light microscopic analysis.So did the 74 days in Wistar rats and 74 days in RCS rats.Immunocytochemistry was used to determine the localization of the amino acid neurotransmitters(glutamate).The relative density of the glutamate-like immunoreactivity (IR)neurofibrilla in inner plexiform layer(IPL) and ganglion cell layer(GCL) of the four groups was gauged.ResultsAfter 2 weeks of transplantation,the transplanted photoreceptor cells survived and the outer plexiform layer was rebuilt.There were significant differences in the level of the relative density of the glutamate-like immunoreactivity(IR)in neurofibrilla in inner plexiform layer (IPL) and ganglion cell layer(GCL) of the four groups within the IPL between the grafted eyes and the control eyes of RCS rats.ConclusionThese results suggest that the level of glutamate neurotransmitters in the reconstructed retina is almost normal,which indicate the function of the morbid retina may be resumed by transplanting the photoreceptor cells.
Melanopsin ganglion cells in the mouse retina independently evoke pupillary light reflex

Abstract:Purpose: The pupillary light reflex (PLR) is crucial for protecting the retina from bright light. The intrinsic photosensitive ganglion cells (ipRGCs) in the retina mediate the PLR, which directly sense light and receive inputs from rod/cone photoreceptors. Previous work used genetic knockout mice to reveal that rod/cone photoreceptors drive transient constriction, and ipRGCs drive the sustained component. We acutely ablated photoreceptors by a chemical injection to examine the role of rod and cone photoreceptors in PLR. Methods: PLR and the multiple electrode array (MEA) recording were conducted with C57BL6/J (wildtype: WT) and Cnga3-/-; Gnat1-/- (rod/cone dysfunctional) mice. n-Nitroso-n-methylurea (MNU) was applied to C57 mice by intraperitoneal injection, and PLR was conducted after 5-7 days of injection. Three different light levels (mesopic, low photopic, and high photopic) were tested. Immunohistochemistry was conducted using the anti-Gnat1 and anti-melanopsin antibodies with DAPI. Results: PLR was induced by all light levels we tested, and the level of constriction increased as the light level increased. After the MNU injection, PLR was not induced at mesopic light stimulus, but was fully induced by high light. The level of PLR was identical between WT and MNU mice, suggesting that ipRGCs fully contributed to the PLR at this light level. Immunohistochemistry revealed that photoreceptors were ablated by the MNU injection, but ipRGCs were preserved. The MEA recording revealed that a population of ipRGCs generated fast and robust spikes in MNU-injected retinal tissues in ex vivo. Conclusions: Contrary to previous observations, our results demonstrate that ipRGCs are the major contributor to the PLR induced by high light.

Abstract:The CICERO method of crown fabrication consists of optically digitizing a gypsum die, designing the crown layer buildup, and subsequently pressing, sintering, and milling consecutive layers of a shaded high-strength alumina-based core material, a layer of dentin porcelain, and a final layer of incisal porcelain. Final finishing is performed in the dental laboratory. The CICERO method allows efficient production of all-ceramic restorations without compromising esthetics or function. This article reviews the process involved in the fabrication of a CICERO crown.
Plasma therapy of primary rat mammary carcinoma: dependence of consumption of C3 during absorption of plasma with sepharose derivatives on the anticoagulant.

Abstract:A previous study demonstrated inhibition of growth of primary rat mammary carcinomas after infusion of tumor-bearer plasma absorbed against Sepharose derivatives. In this report we have quantitated changes in individual complement components that occur during absorption of rat plasma with Sepharose derivatives and defined optimal conditions for consumption of the third component of complement (C3) (other complement components defined similarly). The concentration of functionally active C1 to C9 was measured before and after absorption in plasmas from both normal rats and rats with mammary tumors. C3 activity in plasmas from normal and tumor-bearing rats was reduced (consumed) during absorption under appropriate conditions with Sepharose 4B, inactivated CNBr Sepharose, or Protein A-Sepharose. The concentration of functionally active C1 and C4 did not decrease significantly during absorption with Sepharose derivatives. Consumption of C3 in rat plasma was influenced by the anticoagulant and by the time and temperature of incubation with Sepharose derivative. C3 consumption in rat plasma anticoagulated with acid citrate dextrose solution was variable; addition of Mg2+ (5 mM) to plasma anticoagulated with acid citrate dextrose solution augmented C3 consumption. There was no C3 consumption in plasma anticoagulated with ethylenedinitrilotetraacetic acid (a chelator of calcium and magnesium). In contrast, this reduction was observed in plasma anticoagulated with [(ethylenebis(oxyethylenenitrilo)]tetraacetic acid (a chelator of calcium). The results demonstrate optimal conditions for activation of the alternative pathway of complement during absorption of rat plasma with Sepharose derivatives and suggest in vivo experiments to define the role of this pathway in inhibition of growth of mammary tumors.
Differential response of C57BL/6J mouse and DBA/2J mouse to optic nerve crush

Abstract:Abstract Background Retinal ganglion cell (RGC) death is the final consequence of many blinding diseases, where there is considerable variation in the time course and severity of RGC loss. Indeed, this process appears to be influenced by a wide variety of genetic and environmental factors. In this study we explored the genetic basis for differences in ganglion cell death in two inbred strains of mice. Results We found that RGCs are more susceptible to death following optic nerve crush in C57BL/6J mice (54% survival) than in DBA/2J mice (62% survival). Using the Illumina Mouse-6 microarray, we identified 1,580 genes with significant change in expression following optic nerve crush in these two strains of mice. Our analysis of the changes occurring after optic nerve crush demonstrated that the greatest amount of change (44% of the variance) was due to the injury itself. This included changes associated with ganglion cell death, reactive gliosis, and abortive regeneration. The second pattern of gene changes (23% of the variance) was primarily related to differences in gene expressions observed between the C57BL/6J and DBA/2J mouse strains. The remaining changes in gene expression represent interactions between the effects of optic nerve crush and the genetic background of the mouse. We extracted one genetic network from this dataset that appears to be related to tissue remodeling. One of the most intriguing sets of changes included members of the crystallin family of genes, which may represent a signature of pathways modulating the susceptibility of cells to death. Conclusion Differential responses to optic nerve crush between two widely used strains of mice were used to define molecular networks associated with ganglion cell death and reactive gliosis. These results form the basis for our continuing interest in the modifiers of retinal injury.

Abstract:Retinal ganglion cell (RGC) death occurs after optic nerve injury due to acute trauma or chronic degenerative conditions such as optic neuropathies (e.g., glaucoma). Currently, there are no effective therapies to prevent permanent vision loss resulting from RGC death, underlining the need for research on the pathogenesis of RGC disorders. Modeling human RGC/optic nerve diseases in non-human primates is ideal because of their similarity to humans, but has practical limitations including high cost and ethical considerations. In addition, many retinal degenerative disorders are age-related making the study in primate models prohibitively slow. For these reasons, mice and rats are commonly used to model RGC injuries. However, as nocturnal mammals, these rodents have retinal structures that differ from primates - possessing less than one-tenth of the RGCs found in the primate retina. Here we report the diurnal thirteen-lined ground squirrel (TLGS) as an alternative model. Compared to other rodent models, the number and distribution of RGCs in the TLGS retina are closer to primates. The TLGS retina possesses ~600,000 RGCs with the highest density along the equatorial retina matching the location of the highest cone density (visual streak). TLGS and primate retinas also share a similar interlocking pattern between RGC axons and astrocyte processes in the retina nerve fiber layer (RNFL). In addition, using TLGS we establish a new partial optic nerve injury model that precisely controls the extent of injury while sparing a portion of the retina as an ideal internal control for investigating the pathophysiology of axon degeneration and RGC death. Moreover, in vivo optical coherence tomography (OCT) imaging and ex vivo microscopic examinations of the retina in optic nerve injured TLGS confirm RGC loss precedes proximal axon degeneration, recapitulating human pathology. Thus, the TLGS retina is an excellent model, for translational research in neurodegeneration and therapeutic neuroprotection.

Abstract:Background: Brain-derived neurotrophic factor (BDNF) has been demonstrated to play an important role in survival, differentiation, and neurite outgrowth for many types of neurons. This study was designed to identify the role of BDNF during peripheral nerve xenotransplantation. Materials and methods: A peripheral nerve xenotransplantation from rats to mice was performed. Intracellular cytokines were stained for natural killer (NK) cells, natural killer T (NKT) cells, T cells, and B cells and analyzed by flow cytometry in the spleen of the recipient mouse. Serum levels of related cytokines were quantified by cytometric bead array. Results: Splenic NK cells significantly increased in the xenotransplanted mice (8.47 +/- 0.88x10(7) cells/mL) compared to that in the control mice (4.66 +/- 0.78x10(7) cells/mL, P=0.0003), which significantly reduced in the presence of BDNF (4.85 +/- 0.87x10(7) cells/mL, P=0.0004). In contrast, splenic NKT cell number was significantly increased in the mice with xenotransplantation plus BDNF (XT + BDNF) compared to that of control group or of mice receiving xenotransplantation only (XT only). Furthermore, the number of CD3+ T cells, CD3+CD4+ T cells, CD3+CD4- T cells, interferon-gamma-producing CD3+CD4+ T cells, and interleukin (IL)-17-producing CD3+CD4+ T cells, as well as CD3-CD19+ B cells, was significantly higher in the spleen of XT only mice compared to the control mice (P<0.05), which was significantly reduced by BDNF (P<0.05). The number of IL-4-producing CD3+CD4+ T cells and CD3+CD4+CD25+Foxp3+ T cells was significantly higher in the spleen of XT + BDNF mice than that in the spleen of XT only mice (P<0.05). Serum levels of IL-6, TNF-alpha, interferon-gamma, and IL-17 were decreased, while IL-4 and IL-10 were stimulated by BDNF following xenotransplantation. Conclusion: BDNF reduced NK cells but increased NKT cell accumulation in the spleen of xenotransplanted mice. BDNF modulated the number of splenic T cells and its subtype cells in the mice following xenotransplantation. These findings suggest that BDNF inhibits rejection of peripheral nerve following xenotransplantation by regulating innate as well as adaptive immune reaction.

Abstract:Considerable research effort has been invested into the transplantation of mammalian photoreceptors into healthy and degenerating mouse eyes. Several platforms of rod and cone fluorescent reporting have been central to refining the isolation, purification and transplantation of photoreceptors. The tracking of engrafted cells, including identifying the position, morphology and degree of donor cell integration post-transplant is highly dependent on the use of fluorescent protein reporters. Improvements in imaging and analysis of transplant recipients have revealed that donor cell fluorescent reporters can transfer into host tissue though a process termed material exchange (ME). This recent discovery has chaperoned a new era of interpretation when reviewing the field's use of dissociated donor cell preparations, and has prompted scientists to re-examine how we use and interpret the information derived from fluorescence-based tracking tools. In this review, we describe the status of our understanding of ME in photoreceptor transplantation. In addition, we discuss the impact of this discovery on several aspects of historical rod and cone transplantation data, and provide insight into future standards and approaches to advance the field of cell engraftment.
