Abstract:Methyl ferulate (MF) exhibited outstanding inhibitory activity against Alternaria alternata, while the specific target and molecular mechanism have not been explored based on omics technology. This study aims to reveal the regulatory molecular mechanism of A. alternata in response to MF by integrating transcriptome, proteome, and metabolome approaches. The results showed that after 200 mg L-1 MF treatment of A. alternata, a total of 909 up-regulated genes, 964 down-regulated genes, 302 up-regulated proteins, and 83 down-regulated proteins were detected. According to bioinformatics analysis, the genes and proteins associated with cell membrane lipid metabolism (including sphingolipid metabolism, steroid biosynthesis, and ether lipid metabolism) were significantly promoted, while genes involved in DNA replication and repair of genetic information processing (including base excision repair and homologous recombination) were notably inhibited. Furthermore, two detoxification-related pathways, ATP binding cassette (ABC) transporters and glutathione metabolism, were also induced. In summary, our results confirmed that the mycelial growth of A. alternata was remarkably suppressed by MF via destroying the cell membrane structure, interfering with membrane lipid metabolism, promoting sphingolipids synthesis, disturbing genetic information processing, and impeding DNA replication and repair. This study proposed the antifungal mechanism of MF against A. alternata at the molecular level and provided a theoretical reference for MF as a new natural antifungal agent applying to the control of postharvest pathogene in fruit and vegetables.

Abstract:Cloves (Syzygium aromaticum) have been used as a traditional Chinese medicinal herb for thousands of years. Cloves possess antiseptic, antibacterial, antifungal, and antiviral properties, but their potential anticancer activity remains unknown. In this study, we investigated the in vitro and in vivo antitumor effects and biological mechanisms of ethyl acetate extract of cloves (EAEC) and the potential bioactive components responsible for its antitumor activity. The effects of EAEC on cell growth, cell cycle distribution, and apoptosis were investigated using human cancer cell lines. The molecular changes associated with the effects of EAEC were analyzed by Western blot and (qRT)-PCR analysis. The in vivo effect of EAEC and its bioactive component was investigated using the HT-29 tumor xenograft model. We identified oleanolic acid (OA) as one of the components of EAEC responsible for its antitumor activity. Both EAEC and OA display cytotoxicity against several human cancer cell lines. Interestingly, EAEC was superior to OA and the chemotherapeutic agent 5-fluorouracil at suppressing growth of colon tumor xenografts. EAEC promoted G0/G1 cell cycle arrest and induced apoptosis in a dose-dependent manner. Treatment with EAEC and OA selectively increased protein expression of p21(WAF1/Cip1) and γ-H2AX and downregulated expression of cell cycle-regulated proteins. Moreover, many of these changes were at the mRNA level, suggesting transcriptional regulation by EAEC treatment. Our results demonstrate that clove extract may represent a novel therapeutic herb for the treatment of colorectal cancer, and OA appears to be one of the bioactive components.
Antimicrobial Activity of Clove Plant Flower Bud Extract (Syzygium aromaticum) on Escherichia coli

Abstract:The continuous use of antibiotics for the treatment of bacterial infections has led to the increase in bacterial resistance to these antibiotics. Many bacteria, including E. coli are currently multi-drug resistant, which poses a great problem to the medical and pharmaceutical industry [1]. This research was conducted to investigate the antimicrobial properties of Syzygium aromaticum against the above-mentioned organisms. The plant extracts were in Hot water, Cold water, and Ethanol, and the susceptibility of the organisms was tested using agar well diffusion and disk diffusion methods. The plant extracts showed antimicrobial activity on all isolates used i.e E.coli. From the result, the minimum inhibitory concentration (MIC) was 0.24mg/ml, and the minimum bactericidal concentration (MBC) was 3.91mg/ml. The diameter zone of Inhibition in (mm) against the plant concentration in (mg/ml) conentrations were 500mg/ml, 250mg/ml, 125mg/ml, 62.50mg/ml, 31.25mg/ml, and 15.63mg/ml. The values of the diameter zone of inhibition against the plant concentrations ranged from 16mm to 0mm while control ranged from 38mm to 10mm. The Phytochemical components determined in Cloves were Alkaloids, Saponin, Phenols, Tannins, Flavonoids, Steroids, Soluble Carbohydrate, Glycosides, Terpenes. This research showed that all the clove extracts against E. coli had the lowest MBC values, which proved to be the most effective and all the clove extracts against E. coli had the lowest MIC value of 0.24mg/ml, which proved to be the most effective of the three.

Abstract:Background: The increasing resistance of microbial pathogens to conventional antibiotics necessitates the exploration of alternative antimicrobial agents. This study aims to evaluate the antimicrobial potential and phytochemical properties of Syzygium aromaticum (clove) and Piper nigrum (black pepper) extracts, both of which are known for their historical use in traditional medicine and culinary applications. Methods: Hydroalcoholic and aqueous extracts of clove and black pepper were prepared. The antimicrobial activity of these extracts was assessed using the disk diffusion method against Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, Candida albicans, and Aspergillus niger. Minimum inhibitory concentration (MIC) was determined using the broth dilution method. Qualitative phytochemical screening identified the presence of key bioactive compounds, while quantitative analysis measured total phenolic and flavonoid contents. LC-HRMS/MS analysis of ethanolic extracts was performed. Results: Both spices extracts exhibited significant antimicrobial activity, with inhibition zones ranging from 14 to 18 mm. clove showed superior antimicrobial efficacy compared to black paper, particularly against fungi. MIC values ranged between 3 mg/mL and 6 mg/mL for both spices. Phytochemical analysis revealed higher total phenolic and flavonoid contents in clove, with hydroalcoholic extracts showing greater concentrations than aqueous extracts. HPLC quantified higher eugenol content in clove extracts and higher piperine content in black pepper extracts. The differences in bioactive compound content were statistically significant (p < 0.05). Conclusion: The study confirms that both spices possess significant antimicrobial properties, attributable to their rich phytochemical composition, particularly phenolics and flavonoids. Clove exhibited slightly superior antimicrobial activity compared to black paper. These findings support the potential use of these spices as complementary antimicrobial agents. Further research should investigate their synergistic effects with conventional antibiotics and explore their applications in food preservation and alternative medicine.
Analysis of the synergistic antifungal mechanism of small molecular combinations of essential oils at the molecular level

Abstract:The bacteriostatic concentration of essential oil (EO) in practical application is usually higher than the flavor threshold, resulting in a lack of balance between bacteriostatic concentration and flavor, which has become the bottleneck of its application in the field of agriculture products. One of the effective ways to solve this problem is the synergistic antibacterial effect of the active components of EO. In this paper, molecular docking , independent gradient model (IGM) and symmetric matching perturbation theory (SAPT) are combined for the first time to investigated the molecular mechanism of eugenol and citral small molecules (E AC ) against Aspergillus niger . The results show that E AC can combine with β-(1,3)-glucan synthase and chitin synthase proteins, and then destroy the integrity of cell wall, in which the trans -citral has the best affinity with these two enzyme proteins. In addition, E AC can bind to tryptophan, tyrosine and phenylalanine in cell membrane. The binding of citral small molecules to these three amino acids is mainly hydrogen bond, while the binding force of eugenol to them is mainly van der Waals force or π-π interaction. Finally, the synergistic antifungal effect of E AC was confirmed by the damage of membrane proteins and the leakage of cell contents. In this combination, citral binds to the cell wall, which promotes the interaction between eugenol and cell membrane, and finally leads to the serious destruction of cell membrane. The above results show that E AC has the potential to be developed into a new type of agriculture products preservative with synergistic antifungal effect.

Abstract:The occurrence of candidiasis, including superficial infections, has recently increased dramatically, especially in immunocompromised patients. Their treatment is often ineffective due to the resistance of yeasts to antimycotics. Therefore, there is a need to search for new antifungals. The aim of this study was to determine the antifungal effect of clove essential oil (CEO) and eugenol (EUG) towards both reference and clinical Candida spp. strains isolated from the oral cavity of patients with hematological malignancies, and to investigate their mode of action and the interactions in combination with the selected antimycotics. These studies were performed using the broth microdilution method, tests with sorbitol and ergosterol, and a checkerboard technique, respectively. The CEO and EUG showed activity against all Candida strains with a minimal inhibitory concentration (MIC) in the range of 0.25–2 mg/mL. It was also found that both natural products bind to ergosterol in the yeast cell membrane. Moreover, the interactions between CEO and EUG with several antimycotics—cetylpyridinium chloride, chlorhexidine, silver nitrate and triclosan—showed synergistic or additive effects in combination, except nystatin. This study confirms that the studied compounds appear to be a very promising group of phytopharmaceuticals used topically in the treatment of superficial candidiasis. However, this requires further studies in vivo.

Syzygium aromaticum essential oil and its major constituents: Assessment of activity against Candida spp. and toxicity

Abstract:Syzigium aromaticum essential oil (EO), eugenol, and β-caryophyllene were evaluated regarding antifungal, antibiofilm, and in vitro toxicity. Additionally, in vivo toxicity of EO was observed. Anti- Candida activity was assessed through broth microdilution assay for all compounds. Time-kill assay (0, 1, 10, 30 min, 1, 2, and 4 h) was used to determine the influence of EO and eugenol on Candida Growth kinetics. Thereafter, both compounds were evaluated regarding their capacity to act on a biofilm formation and on mature biofilm, based on CFU/ml/g of dry weight. Cell Titer Blue Viability Assay was used for in vitro cytotoxicity, using oral epithelial cells (TR146) and human monocytes (THP-1). Lastly, Galleria mellonella model defined the EO in vivo acute toxicity. All compounds, except β-cariofilene (MIC > 8000 μg/ml), presented antifungal activity against Candida strains (MIC 500–1000 μg/ml). The growth kinetics of Candida was affected by the EO (5xMIC 30 min onward; 10xMIC 10 min onward) and eugenol (5xMIC 10 min onward; 10xMIC 1 min onward). Fungal viability was also affected by 5xMIC and 10xMIC of both compounds during biofilm formation and upon mature biofilms. LD 50 was defined for TR146 and THP1 cells at, respectively, 59.37 and 79.54 μg/ml for the EO and 55.35 and 84.16 μg/ml for eugenol. No sign of toxicity was seen in vivo up to 10mg/ml (20 x MIC) for the EO. S . aromaticum and eugenol presented antifungal and antibiofilm activity, with action on cell growth kinetics. In vivo acute toxicity showed a safe parameter for the EO up to 10 mg/ml.

Abstract:Abstract Aims Focusing on phytochemicals to target the virulence factors of Candida albicans is a promising avenue for novel antifungal compounds. Given the limited prior research on essential oil (EO) components and their specific effects on C. albicans virulence, our study aimed to explore their impact and uncover the underlying mechanisms. Methods and results We examined the effects on viability, dimorphic transition, biofilm formation, and changes in the expression of critical virulence-related genes. The results showed that Dehydrocostus lactone, displayed the most potent growth-inhibiting activity with the lowest MIC value, followed by Thymol and Costunolide. A substantial, dose-dependent decrease in germ tube formation occurred after exposure to sub-inhibitory concentrations of the EO components, with Carvacrol, Dehydrocostus Lactone, and Thymol exerting the most potent inhibitory effects. Across sub-inhibitory concentrations, Alpha Bisabolol consistently showcased the most potent antibiofilm activity, followed by lower but significant inhibitory effects with Dehydrocostus Lactone, Thymol, Alpha Pinene, Costunolide, Carvone, and Carvacrol. Alpha Bisabolol, Alpha Pinene, and Dehydrocostus Lactone caused almost total downregulation of ACT1 while minimal changes occurred in expression of HWP1, SAP4, ALS3, and ECE1. Conclusions Considering that actin is essential for various cellular processes, including budding, cell shape maintenance, and the formation of filaments in C. albicans, it is a plausible hypothesis that inhibiting ACT1 or disturbing actin’s normal functioning could potentially affect the fungus’s virulence, which warrants additional research and exploration. This study underscores the potent antifungal and anti-virulence properties of various EO components, which effectively cripple C. albicans and reduce its disease-causing ability. This innovative approach holds promise for effective clinical therapies.

Abstract:Fusarium oxysporum is the main pathogen of Panax notoginseng root rot, and chemical fungicides remain the primary measures to control the disease. Plant essential oil (EO) is a volatile plant secondary metabolic product that does not produce any residue to replace chemical pesticide. To comprehensively understand the antifungal mechanism of Alpinia officinarum Hance EO, the physiological indicators, proteome and metabolome were analyzed using F. oxysporum spores and hyphae treated with different EO concentrations. The cell membrane was damaged after both low and high concentrations of EO treatment, along with leakage of the cell contents. To resist the destruction of membrane structure, fungi can increase the function of steroid biosynthesis and expression of these catalytic enzymes, including squalene monooxygenase (SQLE), sterol 14alpha-demethylase (CYP51, CYP61A), delta14-sterol reductase (TM7SF2, ERG4), methylsterol monooxygenase (MESO1), and sterol 24-C-methyltransferase (SMT1). Furthermore, the tricarboxylic acid cycle (TCA) was influenced by inhibiting the expression of glutamate synthase (GLT1), 4-aminobutyrate aminotransferase (ABAT), and succinate-semialdehyde dehydrogenase (gabD); increasing malate and gamma-aminobutyric acid (GABA); and decreasing citrate content. The spore germination rate and mycelia growth were decreased because the expression of cohesin complex subunit SA-1/2 (IRR1) and cohesion complex subunit (YCS4, BRN1, YCG1) were inhibited. Particularly, under high EO concentrations, cyclin-dependent kinase (CDC28) and DNA replication licensing factor (MCM) were further inhibited to disrupt the cell cycle and meiosis, thus affecting cell division. The results of this study will enrich the understanding of the antifungal mechanism of EOs and provide an important basis to develop new plant-derived fungicides.

Abstract:The essential oil of clove has a wide range of pharmacological and biological activities and is widely used in the medicine, fragrance and flavoring industries. In this work, 22 components of the essential oil obtained from clove buds were identified. Eugenol was the major component (76.23%). The essential oil exhibited strong antibacterial activity against Staphylococcus aureus ATCC 25923 with a minimum inhibitory concentration (MIC) of 0.625 mg/mL, and the antibacterial effects depended on its concentration and action time. Kill-time assays also confirmed the essential oil had a significant effect on the growth rate of surviving S. aureus. We hypothesized that the essential oil may interact with the cell wall and membrane first. On the one hand it destroys cell wall and membranes, next causing the losses of vital intracellular materials, which finally result in the bacterial death. Besides, essential oil penetrates to the cytoplasmic membrane or enters inside the cell after destruction of cell structure, and then inhibits the normal synthesis of DNA and proteins that are required for bacterial growth. These results suggested that the effects of the clove essential oil on the growth inhibition of S. aureus may be at the molecular level rather than only physical damage.

Abstract:The development of natural essential oil as an alternative to synthetic chemicals in the control of postharvest decay is currently in the spotlight. In the present study, the efficacy of seven essential oils in suppressing Collelotrichum gloeosporioides identified from sweet cherry was evaluated in vitro and clove oil was proved to be the most promising inhibitor. Thus, the antifungal properties and potential mechanisms of clove oil in vitro and in vivo by fumigation and contact treatments were intensively investigated. For C. gloeosporioides, the minimal inhibitory concentrations (MIC) of clove oil in air and contact phase were 80 and 300 μL/L in vitro testing, respectively. Based on the radial growth of C. gloeosporioides mycelium in medium, the fumgitoxic ability of essential oil was observed in a dose-dependent manner, which was not as dramatic as that under in vivo conditions. Furthermore, scanning electron microscopy and transmission electron microscopy of C. gloeosporioides exposed to clove oil exhibited obviously deleterious morphological and ultrastructural alterations confirming the disruption of fungal cell wall and endomembrane system, which resulted in increasing in permeability and causing the loss of intracellular constituents. In future, essential oils, combined with nano-emulsification approaches, could be good candidates as safe and effective antifungal agents for fungal spoilage of fresh commodities.

Abstract:The use of natural compounds to prevent and treat infective diseases is increasing its importance, especially in the case of multidrug-resistant (MDR) microorganisms-mediated infections. The drug resistance phenomenon is today a global problem, so it is important to have available substances able to counteract MDR infections. Syzygium aromaticum (L.) Merr. & L.M. Perry (commonly called clove) is a spice characterized by several biological properties. Clove essential oil (EO) consists of numerous active molecules, being eugenol as the principal component; however, other compounds that synergize with each other are responsible for the biological properties of the EO. S. aromaticum is traditionally used for bowel and stomach disorders, cold and flu, oral hygiene, tooth decay, and for its analgesic action. Its EO has shown antioxidant, antimicrobial, anti-inflammatory, neuro-protective, anti-stress, anticancer, and anti-nociceptive activities. This review aims to investigate the role of E. S. aromaticum EO in the counteraction of MDR microorganisms responsible for human disorders, diseases, or infections, such as Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Salmonella typhi, Candida albicans, Giardia lamblia, Streptococcus mutans, Porphyromonas gingivalis, and Klebsiella pneumoniae. This study might orient clinical researchers on future therapeutic uses of S. aromaticum EO in the prevention and treatment of infectious diseases.

Abstract:Using the mycelia growth rate method, inhibition effects of ethanol extracts from 17 Chinese medicine plants were determinated against 8 species of plant pathogenic fungi, including Rhizoctonia cerealis, Fusarium graminearum, Gaeumannomyces graminis, Fusarium oxysporum f. sp. vasinfectum, Valsa mali, Colletotrichum gloeosporioides, Fusarium oxysporum sp. cucumebrium, Colletotrichum lagenarium. The results showed that the ethanol extracts of Syzygium aromaticum has the highest antifungal effect among 17 medicine plants on any tested pathogen, with inhibition rate above 60%, and the inhibition effects on V. mali and F. graminearum were the best, with inhibitory rate 96.3% and 97.4%, respectively. Then, the active substances in S. aromaticum were traced using indicator fungi V. mali and F. graminearum, and 2-methoxy-4-(2-propenyl) phenol (eugenol) was isolated by liquid–liquid extraction and silica gel column chromatography and inferred by 1H-NMR and 13C-NMR analysis. The EC50 values of eugenol were 42.04 mg/L against V. mali and 190.58 mg/L against F. graminearum, showing excellent antifungi activity.

Abstract:<section class="article-section__content"><h3 class="article-section__sub-title section1"> Aims</h3><p>In this study, the antifungal effect of cinnamaldehyde against <i>Fusarium sambucinum</i> and its underlying mechanisms were determined. </p></section><section class="article-section__content"><h3 class="article-section__sub-title section1"> Methods and Results</h3><p>Minimum inhibitory concentration and minimal fungicidal concentration of cinnamaldehyde were 3 and 4 mmol l<sup>‐1</sup> on spore germination and colony development assays <i>in vitro</i>, respectively. Furthermore, the lesion diameter of potato tubers and tuber slices inoculated with <i>F. sambucinum</i> was reduced by 76.9% and 69% after treatment with 4 mmol l<sup>‐1</sup> cinnamaldehyde. Cytometric analyses reveled that cinnamaldehyde significantly affected the integrity of cell membrane firstly, then decreased mitochondrial membrane potential and induced the accumulation of intracellular reactive oxygen species. Meanwhile, High performance liquid chromatography (HPLC) results indicated that 3 mmol l<sup>‐1</sup> cinnamaldehyde could reduce the ergosterol content by 67.94%. This effect was accompanied by a down regulation of <i>ERG11, ERG6</i> and <i>ERG4</i> which were involved in ergosterol biosynthesis. </p></section><section class="article-section__content"><h3 class="article-section__sub-title section1"> Conclusion</h3><p>Theses results suggest that cinnamaldehyde exerts strong antifungal activity against <i>F. sambucinum</i>, probably by affecting the ergosterol biosynthetic processes what leads to the disruption of cell membrane integrity. </p></section><section class="article-section__content"><h3 class="article-section__sub-title section1"> Significance and Impact of the Study</h3><p>Cinnamaldehyde is a predominant constituent and key flavor compound of cinnamon essential oil. It has been used as a food additive and flavorant. It is expected to be a novel and safe fungicide for controlling dry rot in potato tubes.</p></section>

Abstract:The high occurrence and mortality rates related to candidiasis emphasize the urgent need to introduce new therapeutic approaches to treat this infection. Eugenol, the main phenolic component of Clove and Cinnamomum essential oil, has been used to inhibit growth and different virulence factors of Candida , including strains with decreased susceptibility to antifungals, particularly fluconazole. The results showed that this compound could bind to Candida membrane and decrease ergosterol biosynthesis, consequently leading to cell wall and membrane damage. Additionally, eugenol not only reduced germ tube formation, which reduces nutrient absorption from host tissues, but it also increased the levels of lipid peroxidation and reactive oxygen species, which induces oxidative stress and causes high permeability in the fungal cell membrane. Eugenol inhibited Candida cells’ adhesion capacity; additionally, this compound inhibited the formation of biofilms and eliminated established Candida biofilms on a variety of surfaces. Furthermore, by disrupting fungal cell integrity, eugenol could boost the entry of the antifungal drugs into the Candida cell, improving treatment efficacy. Therefore, eugenol could be used in the clinical management of various presentations of candidiasis, especially mucocutaneous presentations such as oral and vulvovaginal infections. However, further investigations, including in vivo and animal studies, toxicology studies and clinical trials, as well as molecular analysis, are needed to improve formulations and develop novel antifungal agents based on eugenol.

Citronellal Exerts Its Antifungal Activity by Targeting Ergosterol Biosynthesis in Penicillium digitatum

Abstract:Ergosterol (ERG) is a potential target for the development of antifungal agents against Penicillium digitatum, the pathogen of green mold in citrus fruits. This study examined the mechanism by which citronellal, a typical terpenoid of Cymbopogon nardus essential oil, acts on ergosterol to exhibit its antifungal activity against P. digitatum. We previously reported that citronellal inhibited the growth of P. digitatum with minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) of 1.36 and 2.72 mg/mL, respectively. In citronellal-treated cells, the membrane integrity and ergosterol contents significantly decreased, whereas lanosterol, which serves as a precursor for ergosterol biosynthesis, massively accumulated. Addition of 150 mg/L of exogenous ergosterol decreased the inhibitory rate of citronellal, restoring the ergosterol content and hence the membrane structure to normal levels, and triggered expression of nearly all ERG genes. Based on our findings, we deduce that citronellal damages the cell membrane integrity of P. digitatum by down-regulating the ERG genes responsible for conversion of lanosterol to ergosterol, the key downregulated gene being ERG3, due to the observed accumulation of ergosta-7,22-dienol.
