Abstract:The effect of 1-methylcyclopropene (1-MCP) on quality, antioxidant enzymes and glucosinolate contents in broccoli (Brassica oleracea var. italica) florets was investigated in the present study. Broccoli florets were treated with air (control) and 2.5μl/l 1-MCP for 6h at 20°C, and were then stored at 20°C for 5days. 1-MCP treatment markedly extended shelf life, reduced postharvest deterioration, retarded chlorophyll degradation and inhibited the increase of malondialdehyde amount and activities of polyphenol oxidase and lipoxygenase in florets. The activities of superoxide dismutase, peroxidase and catalase in florets treated with 1-MCP were higher than those in control florets. 1-MCP treatment reduced the rate of decrease of total carotenoids, ascorbic acid and glucosinolates in florets when compared to those in the control. These results indicated that 1-MCP treatment could be a good candidate for extending shelf life, maintaining visual quality and reducing loss of health-promoting compounds, particularly glucosinolates in broccoli.

Abstract:Broccoli is a rich source of health-promoting antioxidants and anticarcinogenic glucosinolates, which has long been recognized for their outstanding benefits to human nutrition and plant defense. The composition and content of glucosinolate are closely associated with the flavor and anticancer activity of broccoli. Up to now, broccoli is among a few Brassica vegetables, in which the biosynthetic pathway of glucosinolate has been widely studied and has attracted extensive attention. Recent studies in glucosinolate research have also identified the genetic variations, as well as the functions of individual glucosinolate profiles and their degradation products in broccoli, which provide the basic aims and powerful strategies for breeding of broccoli varieties with optimal glucosinolate composition and content. To fully exploit the potentially beneficial effects of broccoli, it is important to investigate the glucosinolate variation and metabolism across the whole food chain, from preharvest production to post-harvest storage, processing, and cooking. This chapter provides a general overview of glucosinolate biosynthetic pathway, as well as the genetic variation and function of individual glucosinolate profiles in broccoli, highlights the recent advances in glucosinolate accumulation of broccoli upon different preharvest and post-harvest handlings, and discusses their potential application in broccoli breeding, production, storage, processing, and consumption.

Abstract:Influences of storage temperatures (4 and 20C) on weight loss and carotenoids and glucosinolates (GSs) contents in pakchoi leaves were investigated. The weight loss was prevented by low temperature. At 9 days of storage, the contents of chlorophylls, beta-carotene, lutein and GSs at 20C decreased to 52, 48, 26 and 47%, respectively, compared to those at 4C. At 4C, the total GS content was not significantly changed during 3 days, however increased at 5 and 7 days and returned to the beginning level at 9 days. At 20C, total GS was increased at 3 days, and then decreased gradually. The change trend of aliphatic GS was similar to that of total GS. The indolyl and aromatic GS was not significantly affected by storage temperature. The gluconapin, glucobrassicanapin and 4-methoxyglucobrassicin contents were increased and then decreased with the time; however, the decrease processes were delayed by low temperature.PRACTICAL APPLICATIONSPakchoi is a traditional vegetable in China and belongs to the cruciferous family which is considered to protect human against the risk of cancer, and provides benefits from numerous nutrient and phytochemical compounds with anticancer activity, such as carotenoids and glucosinolates. The contents of carotenoids and GSs might be lost due to unsuitable storage conditions before consumption, and temperature is a critical factor in vegetable storage. Hence, it is necessary to investigate the influence of temperatures on phytochemicals as well as visual quality to provide some suggestions for pakchoi storage. Our results imply that low temperature was beneficial to reduce the loss of beta-carotene, lutein and chlorophylls, maintained the visual quality and total GSs content or even slightly increased the contents of glucoalyssin, gluconapin, glucobrassicanapin and 4-methoxyglucobrassicin at the end of experiment. We suggest that pakchoi should be packed with polyethylene bag and stored at low temperature, especially for longer time storage.
Effects of Different Cooking Methods on Health-Promoting Compounds of Broccoli.

Abstract:The effects of five domestic cooking methods, including steaming, microwaving, boiling, stir-frying, and stir-frying followed by boiling (stir-frying/boiling), on the nutrients and health-promoting compounds of broccoli were investigated. The results show that all cooking treatments, except steaming, caused significant losses of chlorophyll and vitamin C and significant decreases of total soluble proteins and soluble sugars. Total aliphatic and indole glucosinolates were significantly modified by all cooking treatments but not by steaming. In general, the steaming led to the lowest loss of total glucosinolates, while stir-frying and stir-frying/boiling presented the highest loss. Stir-frying and stir-frying/boiling, the two most popular methods for most homemade dishes in China, cause great losses of chlorophyll, soluble protein, soluble sugar, vitamin C, and glucosinolates, but the steaming method appears the best in retention of the nutrients in cooking broccoli.

Abstract:Broccoli has been described as one of the most important vegetables worldwide, and it is a rich source of vitamin C and anti-carcinogenic phytochemicals, such as glucosinolates. The edible part of broccoli is floret head, which is high in respiration rate, and easy to be senescent and yellowing. As a kind of highly perishable products, broccoli heads usually undergo losses in nutrients and quality during postharvest period. The researches on the influence of different postharvest handlings in functional components and quality of broccoli are significant to postharvest physiology, but also have an important guiding role for the broccoli industry. Based on recent investigations, the effects of different postharvest handlings, including cooling storage, atmosphere control, modified atmosphere packaging, 1-methylcyclopropene (1-MCP) and light treatment on functional components and quality of broccoli were reviewed, and the related mechanisms were verified by elucidating the biosynthesis and metabolism pathway of glucosinolates. The common postharvest handling methods of broccoli products were summarized and evaluated, and future researches as well as their potential in broccoli industry were prospected.

Abstract:Broccoli florets were treated with 1-methylcyclopropene (1-MCP) at concentrations of 0-5.0 μL/L for 6 h at 20 C followed by storage at 20 ℃. 1-MCP markedly extended the storage life and delayed the degradation of chlorophyll. The beneficial effects were dependent upon concentrations, and 2.5 μL/L 1-MCP was the most effective one among all the treatments, the contents of vitamin C and protein in this treatment were always higher than those in the control during storage at 20 C. SOD and CAT activities were higher while POD activity was lower in the treatment than in the control.

Abstract:Brassica vegetables are rich in glucosinolates, a group of secondary metabolites, which is related to flavor, quality, resistance and anticancer activity. In addition, they also play a vital role in plant defense against pathogens and herbivores. In this review, we briefly elucidated the biosynthetic and degradation pathway of glucosinolate, and highlighted the effects of different postharvest treatments, such as cooling storage, controlled atmosphere storage, modified atmosphere packaging, 1-methylcyclopropene (1-MCP) treatment, as well as processing and cooking methods on compositions and contents of glucosinolate profiles and nutritional quality in Brassica vegetables. Particularly, from the perspective of dynamic equilibrium of glucosinolate metabolism, we analyzed the mechanism of regulating glucosinolate components and contents, and also discussed how to maintain the contents of favorable glucosinolate profiles and nutritional quality in Brassica vegetable products during postharvest period.

Abstract:The effect of 1-methylcyclopropene (1-MCP) on postharvest characters, antioxidants and glucosinolate contents in bolting stems of Chinese kale (Brassica alboglabra Bailey) was investigated. Chinese kale was treated with air (control) and 10μll−1 of 1-MCP for 24h, followed by storage for up to seven days at 20°C. 1-MCP treatment obviously extended the shelf life, delayed weight loss and decreased the rate of softening, chlorophyll degradation and changes in hue angle. In addition, ethylene production was suppressed and the respiration rate declined. Moreover, 1-MCP treatment postponed the decrease of ascorbic acid, carotenoids and glucosinolates. 1-MCP treatment of bolting stems also delayed the accumulation of total phenolics, and maintained a high level of antioxidant capacity. These results demonstrate that 1-MCP treatment is a good practice for extending shelf life, maintaining the appearance and nutrient value, and reducing the loss of health-promoting compounds, particularly antioxidants and glucosinolates in the Chinese kale.

Abstract:Broccoli (Brassica oleracea var. italica) florets were packaged in polyethylene bags with no holes (M0), two microholes (M1), and four macroholes (M2), and then stored at 4 or 20°C. The effects of modified atmosphere packaging (MAP) treatments on visual quality and glucosinolate contents were determined by comparing with non-wrapped florets. The results showed that MAP treatments, especially with M0 and M1, extended the shelf life and reduced the postharvest deterioration of broccoli florets stored at 4 and 20°C. All three MAP treatments reduced the decreasing concentration rates of individual, total aliphatic and indole glucosinolates in broccoli florets when compared to those in the control, with M0 being the most significant, followed by M1 and M2 during 23 days of storage at 4°C or 5 days of storage at 20°C. Broccoli florets with M0 treatment maintained the visual quality and glucosinolate contents for 13 days at 4°C and 3 days at 20°C.

Abstract:The present study investigated the contents of bioactive compounds and nutrients, as well as antioxidant capacities in heads, stems, leaves and roots of mature broccoli (Brassica oleracea var. italica Planck) plants. The results showed that the heads had the highest amounts of aliphatic glucosinolates, indole glucosinolates, total glucosinolates and glucoraphanin, which was one of the most potential anticarcinogenic glucosinolate identified thus far. The contents of gluconasturtiin, glucoerucin and 4-methoxyglucobrassicin in the roots were remarkably higher than those in the other organs. As for the nutrients, the broccoli heads con-tained markedly higher concentrations of soluble proteins, reducing sugars and soluble solids than those in the leaves, roots and stems. The highest contents of total phenols, L-ascorbic acid, chlorophyll and carotenoids,as well as antioxidant capacities were observed in leaves, and the significant differences existed between leaves and other organs. The leaves and roots of broccoli, which are rich sources of bioactive components, have great potential for producing health-promoting products and biological pesticides as raw materials.
Molecular Mechanisms Through Which Short-Term Cold Storage Improves the Nutritional Quality and Sensory Characteristics of Postharvest Sweet Potato Tuberous Roots: A Transcriptomic Study

Abstract:Sweet potato (Ipomoea batatas (L.) Lam.) is a commercially relevant food crop with high demand worldwide. This species belongs to the Convolvulaceae family and is native to tropical and subtropical regions. Storage temperature and time can adversely affect tuberous roots’ quality and nutritional profile. Therefore, this study evaluates the effect of storage parameters using physicochemical and transcriptome analyses. Freshly harvested tuberous roots (Xingxiang) were stored at 13 °C (control) or 5 °C (cold storage, CS) for 21 d. The results from chilling injury (CI) evaluation demonstrated that there was no significant difference in appearance, internal color, weight, and relative conductivity between tuberous roots stored at 13 and 5 °C for 14 d and indicated that short-term CS for 14 d promoted the accumulation of sucrose, chlorogenic acid, and amino acids with no CI symptoms development. This, in turn, improved sweetness, antioxidant capacity, and nutritional value of the tuberous roots. Transcriptome analyses revealed that several key genes associated with sucrose, chlorogenic acid, and amino acid biosynthesis were upregulated during short-term CS, including sucrose synthase, sucrose phosphate synthase, phenylalanine ammonia-lyase, 4-coumarate-CoA ligase, hydroxycinnamoyl-CoA quinate hydroxycinnamoyltransferase, serine hydroxymethyltransferase, alanine aminotransferase, arogenate dehydrogenase, and prephenate dehydratase. These results indicated that storage at 5 °C for 14 d could improve the nutritional quality and palatability of sweet potato tuberous roots without compromising their freshness.
Exogenous Application of Phytosulfokine Α (pskα) Delays Yellowing and Preserves Nutritional Quality of Broccoli Florets During Cold Storage.

Abstract:In this study, we tested the exogenous application of phytosulfokine alpha (PSK alpha) for delaying the yellowing of broccoli florets during cold storage. Our results showed that the lower yellowing in broccoli florets treated with 150 nM PSK alpha was probably due to the higher endogenous accumulation of PSK alpha, leading to the endogenous accumulation of guanosine 3', 5'-cyclic monophosphate (cGMP). Besides, broccoli florets treated with 150 nM PSK alpha exhibited a higher accumulation of phenols and flavonoids by triggering gene expression and activities of phenylalanine ammonia-lyase (PAL) and chalcone synthase (CHS). Moreover, the higher expression of L-galactotno-1,4-lactone dehydrogenase (GLDH) gene and the lower expression of ascorbic acid oxidase (AAO) gene in broccoli florets treated with 150 nM PSK alpha may be the reasons for the higher accumulation of ascorbic acid. In conclusion, the exogenous application of PSK alpha is a promising strategy in delaying the yellowing and preserving the nutritional quality of broccoli florets during cold storage.

Abstract:Broccoli (Brassica oleracea L. var. Italica) is largely consumed all over the world and has a high economic importance. Likewise, broccoli contains high levels of glucosinolates, carotenoids and total phenols, which are related with the prevention of chronic diseases. The present project's objective was to evaluate the effect of industrial freezing on the stability of bioactive molecules in seven commercial broccoli cultivars (Tlaloc®, Endurance®, Florapack®, Domador®, Steel®, Iron Man® and Avenger®). In general, industrial freezing increased the extractability of total glucosinolates, whereas total phenols remained constant in most broccoli cultivars. Likewise, broccoli subjected to industrial freezing showed higher levels of total carotenoids (∼60-300% higher) as compared with fresh broccoli. Results suggest that bioactive compounds in frozen broccoli would be more bioavailable than in raw.

Changes in Glucoraphanin Content and Quinone Reductase Activity in Broccoli (brassica Oleracea Var. Italica) Florets During Cooling and Controlled Atmosphere Storage

Abstract:The effects of cooling and controlled atmosphere (CA) treatments on glucoraphanin content and induction of quinone reductase (QR) activity in broccoli florets were investigated. The results showed that both the glucoraphanin content and QR activity of broccoli florets increased during the initial 6 day of storage, and then decreased markedly at different temperatures of 0, 5, and 10°C. Immediate cooling of broccoli florets at 0 and 5°C could preserve the glucoraphanin content and QR activity for 12 days. Keeping broccoli florets at 20°C for 6h before cooling at 5°C had little effects on shelf-life, glucoraphanin content and QR activity. CA treatments with elevated CO2 (21% O2+10% CO2, and 21% O2+20% CO2) and an air treatment (21% O2) were found to increase the glucoraphanin content and the QR activity over the first 5 days of storage at 5°C, while CA treatments with reduced O2 concentrations (1% O2, 1% O2+10% CO2) led to a steady decrease of glucoraphanin content and the QR activity during 20 days of storage at 5°C. The highest content of glucoraphanin and QR activity was found in florets stored under 21% O2+10% CO2 at 5°C. These conditions were able to maintain the visual quality, glucoraphanin content and QR activity of the broccoli florets for 20 days.

Abstract:The effects of long-term freezer storage and different defrosting methods on the retention of glucosinolates, vitamin C, and total phenols in frozen broccoli florets were investigated in the present study. Frozen broccoli florets were stored in a freezer at −20 °C for 165 days or subjected to defrosting by three different house-hold thawing methods (water, air, and refrigerator defrosting). Results showed that all glucosinolates were well preserved, while vitamin C and total phenols were reduced by less than 12% and 19% of the control, respectively, during the storage. Besides, refrigerator and air defrosting were better than water defrosting in glucosinolates retention, and refrigerator defrosting was the best in vitamin C preservation. No difference was observed in reserving phenolic compounds among the three methods. In conclusion, long-term freezer storage is an excellent way to preserve broccoli florets, and refrigerator defrosting is the best way to maintain the nutritional compounds in frozen broccoli florets.

A New Food Ingredient Rich in Bioaccessible (Poly)Phenols (and Glucosinolates) Obtained from Stabilized Broccoli Stalks

Abstract:Broccoli (Brassica oleracea var. italica) stalks account for up to 35% of the broccoli harvest remains with the concomitant generation of unused waste that needs recovery to contribute to the sustainability of the system. However, due to its phytochemical composition, rich in bioactive (poly)phenols and glucosinolates, as well as other nutrients, the development of valorization alternatives as a source of functional ingredients must be considered. In this situation, the present work aims to develop/obtain a new ingredient rich in bioactive compounds from broccoli, stabilizing them and reducing their degradation to further guarantee a high bioaccessibility, which has also been studied. The phytochemical profile of lyophilized and thermally treated (low-temperature and descending gradient temperature treatments), together with the digested materials (simulated static in vitro digestion) were analysed by HPLC-PDA-ESI-MSn and UHPLC-3Q-MS/MS. Broccoli stalks and co-products were featured by containing phenolic compounds (mainly hydroxycinnamic acid derivatives and glycosylated flavonols) and glucosinolates. The highest content of organosulfur compounds corresponding to the cores of the broccoli stalks treated by applying a drying descendant temperature gradient (aliphatic 18.05 g/kg dw and indolic 1.61 g/kg dw, on average, while the breakdown products were more abundant in the bark ongoing low temperature drying 11.29 g/kg dw, on average). On the other hand, for phenolics, feruloylquinic, and sinapoylquinic acid derivatives of complete broccoli stalk and bark, were more abundant when applying low-temperature drying (14.48 and 28.22 g/kg dw, on average, respectively), while higher concentrations were found in the core treated with decreasing temperature gradients (9.99 and 26.26 g/kg dw, on average, respectively). When analysing the bioaccessibility of these compounds, it was found that low-temperature stabilization of the core samples provided the material with the highest content of bioactives including antioxidant phenolics (13.6 and 33.9 g/kg dw of feruloylquinic and sinapoylquinic acids, on average, respectively) and sulforaphane (4.1 g/kg dw, on average). These processing options enabled us to obtain a new product or ingredient rich in bioactive and bioaccessible compounds based on broccoli stalks with the potential for antioxidant and anti-inflammatory capacities of interest.

Abstract:Glucosinolates (GLS) are bioactive compounds found in Brassicaceae vegetables. Industrial food processing treatments, such as blanching, quick-freezing (QF), vacuum freeze-drying (VFD), vacuum-drying (VD) and oven-drying (OD), significantly affect the degradation and ingestion of GLS. Here, the effects of these treatments, followed by boiling, on the GLS content and mimicking ingestion level of isothiocyanate from Brassicaceae vegetables (broccoli, cauliflower, white and red cabbages, Chinese and baby cabbages, white and red radish roots) were investigated. The results showed that blanching-QF maintained or increased the GLS content as well as preserved the ingestion level of isothiocyanate, an optimum treatment for GLS preservation. Blanching-VFD was recommended for these vegetables, while blanching-VD and blanching-OD caused relatively high GLS losses and low isothiocyanate production. Additionally, stabilities of individual GLS during processing rely on their chemical structures and species. Generally, aliphatic GLS from Brassicaceae showed lower loss than indole GLS, indicating differences in their stabilities during processing.

Abstract:The effects of industrial pre-freezing processing and freezing handling on the contents of glucosinolates and antioxidants (vitamin C, polyphenols, carotenoid and chlorophyll), as well as the antioxidant capacity in broccoli (Brassica oleracea L. var. italica) florets were investigated in the present study. Our results showed that the glucosinolate accumulations were significantly decreased after pre-freezing processing, whereas elevated levels of phenols, carotenoids, chlorophyll, and also antioxidant capacity were observed in frozen broccoli florets. The contents of vitamin C remained constant during above mentioned processing. In conclusion, the current industrial freezing processing method is a good practice for the preservation of main antioxidant nutrients in broccoli florets, although some improvements in pre-freezing processing, such as steam blanching and ice-water cooling, are needed to attenuate the decrease in glucosinolate content.

Abstract:We report a trinuclear Co(II) complex containing bridging dicyanamides and a tris-chelated HAT, which possesses approximately 37% void space. The magnetic exchange pathways appear in the structure as a corner sharing tetrahedral network. The compound crystallizes in the monoclinic space group P2(1)/c [a = 13.655(3) A, b = 15.189(3) A, c = 22.367(4) A, beta = 114.100(2) degrees, V = 4234.5(14) A(3), Z = 4, R(F(o)) = 0.0823]. The magnetic data were fit to an S = 3/2 model for systems dominated by zero-field splitting effects with g = 2.01 and D = 38.9 cm(-1).
