Abstract:We report on the synthesis and evaluation of biopolymeric spheres of poly(lactide-co-glycolide) containing different amounts of magnetite nanoparticles and Ibuprofen (PLGA-Fe3O4-IBUP), but also chitosan (PLGA-CS-Fe3O4-IBUP), to be considered as drug delivery systems. Besides morphological, structural, and compositional characterizations, the PLGA-Fe3O4-IBUP composite microspheres were subjected to drug release studies, performed both under biomimetically-simulated dynamic conditions and under external radiofrequency magnetic fields. The experimental data resulted by performing the drug release studies evidenced that PLGA-Fe3O4-IBUP microspheres with the lowest contents of Fe3O4 nanoparticles are optimal candidates for triggered drug release under external stimulation related to hyperthermia effect. The as-selected microspheres and their chitosan-containing counterparts were biologically assessed on macrophage cultures, being evaluated as biocompatible and bioactive materials that are able to promote cellular adhesion and proliferation. The composite biopolymeric spheres resulted in inhibited microbial growth and biofilm formation, as assessed against Staphylococcus aureus, Pseudomonas aeruginosa, and Candida albicans microbial strains. Significantly improved antimicrobial effects were reported in the case of chitosan-containing biomaterials, regardless of the microorganisms' type. The nanostructured composite biopolymeric spheres evidenced proper characteristics as prolonged and controlled drug release platforms for multipurpose biomedical applications.
Preparation, characterization and in vitro release properties of ibuprofen-loaded microspheres based on polylactide, poly(ε-caprolactone) and their copolymers

Abstract:In this paper, ibuprofen was encapsulated into microspheres by oil-in-water (o/w) emulsion solvent evaporation method. Biodegradable polymers with certain compositions and characteristics such as polylactide (PLA), poly(epsilon-caprolactone) (PCL) and their block copolymer were used to prepare the microspheres. The results indicate that, under the same processing conditions, the drug entrapment efficiency was similar (similar to 80%) for microspheres prepared with PLA and P(LA-b-CL) (78.7/21.3 by mole), but it was only 25.4% for PCL microspheres. The in vitro drug release rate decreased in the order of PCL, P(LA-b-CL) (78.7/21.3 by mole) and PLA microspheres. PCL microspheres showed more serious burst release during the first day (almost 80%) than P(LA-b-CL) (50%) and PLA microspheres (18%). The complete ibuprofen release duration from the last two kinds of microspheres exceeded I month. Characterization of the microspheres by differential scanning calorimetry (DSC), scanning electron microscopy (SEM) and polarized optical microscope (POM) revealed that ibuprofen was amorphous in PCL microspheres and partially crystalline in P(LA-h-CL) and PI-A microspheres. The different release behaviour of ibuprofen from the three kinds of microspheres could be attributed to the different crystallinity of the studied polymers and drug dispersion state in polymer matrices. All the above results suggest that the copolymer with a certain ratio of lactide to epsilon-caprolactone could have potential applications for long-term ibuprofen release.

And the design of double-walled microspheres also has been investigated. Moreover, encapsulation efficiency and the release profiles can be improved by different modification methods. And it is extensively used in drug deliver, gene therapy and imaging diagnosis, especially in the fields of tissue engineering.

Abstract:Microspheres are multiparticulate drug delivery systems that are designed to deliver drugs to a particular location at a fixed rate. Microspheres are free-flowing powders made up of biodegradable proteins or synthetic polymers with particle sizes ranging from 1 to 1000µm. Benefits of the use of microspheres in fields such as drug delivery, bone tissue manufacturing, and the absorption and desorption of contaminants by regeneration. The study shows the method of planning and measurement of microsphere parameters. Microspheres are complex, such as bioadhesive microspheres, polymeric microspheres, magnetic microspheres, floating microspheres, radioactive microspheres. Microspheres may be used in various fields such as cosmetics, oral drug delivery, target drug delivery, ophthalmic drug delivery, gene delivery, and others listed in the study. In order to achieve optimal therapeutic effectiveness, it is important to deliver the agent to the target tissue at an optimum level within the right timeframe, resulting in little toxicity and minimal side effects. There are different approaches to supplying the medicinal drug to the target site in a continuous managed manner. One such strategy is the use of microspheres as drug carriers. In this article, the value of the microsphere is seen as a novel drug delivery carrier to achieve site-specific drug delivery was discussed. Abstract:Gellan gum is a biocompatible and easily accessible polysaccharide with excellent properties to produce microparticles as drug delivery systems. However, the production methods often fail in reproducibility, compromising the translational potential of such systems. In this work, the production of gellan gum-based microparticles was optimized using the coaxial air flow method, and an inexpensive and reproducible production method. A design of experiments was used to identify the main parameters that affect microparticle production and optimization, focusing on diameter and dispersibility. Airflow was the most significant factor for both parameters. Pump flow affected the diameter, while the gellan gum/alginate ratio affected dispersibility. Microparticles were revealed to be sensitive to pH with swelling, degradation, and encapsulation efficiency affected by pH. Using methylene blue as a model drug, higher encapsulation, and swelling indexes were obtained at pH 7.4, while a more pronounced release occurred at pH 6.5. Within PBs solutions, the microparticles endured up to two months. The microparticle release profiles were studied using well-known models, showing a Fickian-type release, but with no alteration by pH. The developed microparticles showed promising results as drug-delivery vehicles sensitive to pH.

Abstract:Biodegradable amphiphilic graft copolymers with different compositions were synthesized by grafting poly(l-lactide) (PLLA) sequences onto a water-soluble poly-α,β-[N-(2-hydroxyethyl)-l-aspartamide] (PHEA) backbone. The critical micelle concentration (CMC) of the graft polymers was determined by fluorescence probe technique. Using the MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay, the graft polymers were proved to have low cytotoxicity. Based on the specific physicochemical property of the graft copolymers, submicron sized microsphere drug delivery systems were prepared by a very convenient “ultrasonic dispersion method”, which did not involve toxic organic solvents. The drug-loaded microspheres had a regular spherical shape with a narrow size distribution. A hydrophobic drug, prednisone acetate, was encapsulated into polymeric microspheres and the in vitro drug release was studied.
Formation of alginate microspheres prepared by optimized microfluidics parameters for high encapsulation of bioactive molecules

Abstract:Drug delivery systems such as microspheres have shown potential in releasing biologicals effectively for tissue engineering applications. Microfluidic systems are especially attractive for generating microspheres as they produce microspheres of controlled-size and in low volumes, using micro-emulsion processes. However, the flow rate dependency on the encapsulation of molecules at a microscale is poorly understood. In particular, the flow rate and pressure parameters might influence the droplet formation and drug encapsulation efficiency. We evaluated the parameters within a two-reagent flow focusing microfluidic chip under continuous formation of hydrogel particles using a flourinated oil and an ionic crosslinkable alginate hydrogel. Fluorescein isothiocyanate-dextran sulfate (FITC-dextran sulfate MW: 40 kDa) was used to evaluate the variation of the encapsulation efficiency with the flow parameters, optimizing droplets and microsphere formation. The ideal flow rates allowing for maximum encapsulation efficiency, were utilised to form bioactive microspheres by delivering transforming growth factor beta-3 (TGFβ-3) in cell culture media. Finally, we evaluated the potential of microfluidic-formed microspheres to be included within biological environments. The biocompatibility of the microspheres was tested over 28 days using adult human mesenchymal stem cells (hMSCs). The release profile of the growth factors from microspheres showed a sustained release in media, after an initial burst, up to 30 days. The metabolic activity of the cells cultured in the presence of the microspheres was similar to controls, supporting the biocompatibility of this approach. The fine-tuned parameters for alginate hydrogel to form microspheres have potential in encapsulating and preserving functional structure of bioactive agents for future tissue engineering applications.

Abstract:Droplet microfluidics offers precise and simultaneous control of multiple fluids at microscale, which enables synthesis of novel microparticles with compositional and structural diversity in a controllable way. The morphology and functionality of generated microparticles can be well designed by modulating the hydrodynamic profile as well as geometric structures. The synergistic combination of droplet microfluidics with biodegradable materials makes it possible to encapsulate actives/drugs inside microparticles at high efficiency for drug delivery. The utilization of these microfluidic‐generated microparticles with the characteristics of easy biodegradability and good biocompatibility in the field of drug delivery has made considerable progress in recent years. In this review, the commonly used structures of microchannel and methods to generate microparticles with droplet microfluidics are introduced. In addition, recent advances of biodegradable microparticles in the application of drug delivery are discussed and summarized with the focus on two kinds of biopolymers for preparing biodegradable microspheres, natural biopolymers, and synthetic biopolymers. Next, environment‐sensing microencapsulation systems have been discussed because of their ability to release drug upon external stimulation, thereby allowing on‐demand drug delivery. Finally, current challenges of utilizing microparticles in drug delivery are pointed out and some perspectives for the future direction in research and applications are provided. The progress of microfluidic technique and biodegradable microparticles in the application of drug release has been systematically summarized.

Abstract:Monodisperse biodegradable polymer microspheres show broad applications in drug delivery and other fields. In this study, we developed an effective method that combines microfluidics with interfacial instability to prepare monodispersed poly(lactic-co-glycolic acid)-b-polyethylene glycol (PLGA-PEG)/poly(lactic-co-glycolic acid) (PLGA) microspheres with tailored surface morphology. By adjusting the mass ratio of PLGA-PEG to PLGA, the concentration of stabilizers and the type of PLGA, we generated microspheres with various unique folded morphologies, such as "fishtail-like", "lace-like" and "sponge-like" porous structures. Additionally, we demonstrated that risperidone-loaded PLGA-PEG/PLGA microspheres with these folded morphologies significantly enhanced drug release, particularly in the initial stage, by exhibiting a logarithmic release profile. This feature could potentially address the issue of delayed release commonly observed in sustained-release formulations. This study presents a straightforward yet effective approach to construct precisely engineered microspheres offering enhanced control over drug release dynamics.

Abstract:The broad diversity of structures and the presence of numerous functional groups available for chemical modifications represent an enormous advantage for the development of safe, non-toxic, and cost-effective micellar drug delivery systems (DDS) based on natural biopolymers, such as polysaccharides, proteins, and peptides. Different drug-loading methods are used for the preparation of these micellar systems, but it appeared that dialysis is generally recommended, as it avoids the formation of large micellar aggregates. Moreover, the preparation method has an important influence on micellar size, morphology, and drug loading efficiency. The small size allows the passive accumulation of these micellar systems via the permeability and retention effect. Natural biopolymer-based micellar DDS are high-value biomaterials characterized by good compatibility, biodegradability, long blood circulation time, non-toxicity, non-immunogenicity, and high drug loading, and they are biodegraded to non-toxic products that are easily assimilated by the human body. Even if some recent studies reported better antitumoral effects for the micellar DDS based on polysaccharides than for commercial formulations, their clinical use is not yet generalized. This review is focused on the studies from the last decade concerning the preparation as well as the colloidal and biological characterization of micellar DDS based on natural biopolymers.

Abstract:The biocompatible poly(N-vinylpyrrolidinone) (PNVP) microgels were synthesized via surfactant free emulsion polymerization with N-vinylpyrrolidinone (NVP) as the monomer and ethylene glycol dimethacrylate (EGDMA) as the cross-linker at 60 °C. The obtained PNVP microgels are spherical in shape with hydrodynamic diameter of approximately 200 nm and narrow size distribution. The PNVP microgels show rough surfaces due to the different reaction rates of monomer NVP and cross-linker EGDMA. The obtained PNVP microgels could well disperse in phosphate-buffered saline (PBS) solution with long-term stability, which make them candidates for bioapplications. The results of 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyltetrazolium bromide (MTT) tests indicated that the PNVP microgels are biocompatible with low toxicity even at a concentration of 1000 μg/mL. By labeling the PNVP microgels with fluorescein comonomer, it was demonstrated that the PNVP microgels could be uptaken by human embryonic kidney 293 (HEK-293) cells. The experimental results indicated that the release of isoniazid (INH, the hydrophilic model drug) could be well described by a Fickian release, whereas the microgels exhibited burst release for 5-fluorouracil (5-fu, the hydrophobic model drug).
Hyaluronan-based microspheres as tools for drug delivery: a comparative study

Abstract:The present paper describes the production of biodegradable microparticles using different hyaluronan polymers, such as native hyaluronan, the esterified derivative of hyaluronan Hyaff 11p50 (where 50% of the carboxy groups of hyaluronic acid are esterified with benzyl alcohol) and the autocross-linked polymer (ACP) internally esterified derivative of hyaluronan, by solvent evaporation and spray-drying methods. As model drugs cromolyn sodium salt, metronidazole and prednisolone hemisuccinate sodium salt were employed. The influence of polymer and preparation procedure has been evaluated on microparticle characteristics (i.e. morphology and encapsulation yield) and on the drug release profiles. The use of solvent evaporation method, a polymeric matrix constituted of Hyaff 11p50 3% (w/v), a dispersing phase constituted of 80 g of mineral oil (w/o ratio: 0.1), Span 85 0.1% (w/w) as stabilizer, and a stirring speed of 700 rpm resulted in the production of microspheres characterized by spherical shape, absence of aggregates, a mean diameter of 6.4 microm and a recovery of 90% (w/w). The production of drug containing microspheres led to an increase of mean diameter of microspheres and to high encapsulation yields. Moreover in vitro models have demonstrated that in all cases drugs were released from Hyaff 11p50 microspheres in a controlled fashion. Finally mathematical analysis of the drug release modalities has evidenced that drug release from Hyaff 11p50 microspheres is more consistent with kinetics of the diffusion rather than of the dissolution type.

Abstract:Polymeric micelles are built from amphiphilic polymers through self-assembly effects. Due to their unique core shell structure, small size and modifiable surface, polymeric micelles have been widely investigated as nanoscale drug delivery carriers. Such systems may increase drug solubility and have possible applications in tumour targeting and gene therapy. These biomedical applications require that polymeric micelles are biocompatible, have prolonged blood circulation and possess high drug-loading efficiency. In addition, tumour targeting and smart drug release behaviour need special modification towards micelles with multiplicate functional substances. This review focuses on the present progress of polymeric micelles and highlights some critical issues for their application in drug delivery systems. Composition and micellisation procedures are also briefly discussed.
Optimization and characterization of chemically modified polymer microspheres and their effect on cell behavior

Abstract:Porous microspheres composed of poly (l-lactic-co-glycolic acid) (PLGA), poly (l-lactic acid) (PLA), and poly (l-lactide–co-caprolactone) (PLCL) were fabricated. Based on the hydrophilic properties and porous microstructures, PLGA (100kDa Mw and 1.5% concentration) microspheres were selected and cross-linked with gelatin using two different chemical modification methods: ethanediamine/glutaraldehyde (EN/GA) and N-(3-dimethylaminopropyl)-N′-ethyl-carbodiimide/N-hydroxysuccinimide (EDC/NHS). The effect of chemical modification on the surface topography, porosity, wettability, and biocompatibility was evaluated. Porous microspheres modified with gelatin through ethanediamine/glutaraldehyde cross-linking improved the hydrophilic properties of PLGA and generated an open pore microstructure. A significant increase in the cell metabolic activity was recorded after 5 days in culture, suggesting that ethanediamine/glutaraldehyde may be a suitable cross-linking agent for cell culture applications.

Abstract:Polymeric drug delivery technology, which allows for medicinal ingredients to enter a cell more easily, has advanced considerably in recent decades. Innovative medication delivery strategies use biodegradable and bio-reducible polymers, and progress in the field has been accelerated by future possible research applications. Natural polymers utilized in polymeric drug delivery systems include arginine, chitosan, dextrin, polysaccharides, poly(glycolic acid), poly(lactic acid), and hyaluronic acid. Additionally, poly(2-hydroxyethyl methacrylate), poly(N-isopropyl acrylamide), poly(ethylenimine), dendritic polymers, biodegradable polymers, and bioabsorbable polymers as well as biomimetic and bio-related polymeric systems and drug-free macromolecular therapies have been employed in polymeric drug delivery. Different synthetic and natural biomaterials are in the clinical phase to mitigate different diseases. Drug delivery methods using natural and synthetic polymers are becoming increasingly common in the pharmaceutical industry, with biocompatible and bio-related copolymers and dendrimers having helped cure cancer as drug delivery systems. This review discusses all the above components and how, by combining synthetic and biological approaches, micro- and nano-drug delivery systems can result in revolutionary polymeric drug and gene delivery devices.

Abstract:The aim of this paper was to investigate the influence of preparation parameters on gelatin microspheres production, chemico-physical characteristics and drug encapsulation. In particular, we focussed our attention on the manufacturing parameters such as amount of polymer, stirring speed, presence and concentration of surfactants. As model drugs, TAPP-Br, clonidine hydrochloride and bromocriptine mesylate were chosen in order to compare their encapsulation and release characteristics with microspheres. In the second part of this work, a study of the influence of thermal treatment on the microspheres is reported, performed with the aim to possibly modify gelatin dissolution and drug release. In particular, the effect of this treatment was evaluated on microsphere characteristics such as swelling, porosity and dissolution, and finally on the release profiles of the encapsulated drugs.
Novel Biomimetic Polymersomes As Polymer Therapeutics for Drug Delivery

Abstract:Novel amphiphilic diblock copolymers, cholesterol-end-capped poly(2-methacryloyloxyethyl phosphorylcholine) (CMPC), which have poly(2-methacryloyloxyethyl phosphorylcholine) (poly(MPC)) as hydrophilic segment and cholesterol as hydrophobic segment, was specially designed as drug delivery systems. Fluorescence probe technique and transmission electron microscope (TEM) characterizations indicated that this novel amphiphilic copolymer formed micelles structure in water and the critical micelle concentration (CMC) was determined to be 1.57×10−7 mol/l. A commercial obtained polymeric amphiphiles, Cholesterol end capped PEO (CPEO), which had a similar structure with CMPC, was used as a control in the cytotoxicity test. While CPEO showed obvious cytotoxicity, cytotoxicity of this novel amphiphiles was not observed as indicated by cell culture. Anti-cancer drug adriamycin (ADR) was incorporated into the micelles by oil-in-water method. The size of the drug-containing micelles was less than 200 nm, and the size distribution of the drug-containing micelles showed a narrow and monodisperse unimodal pattern. The release rate of ADR from the nanosphere was slow and the release continued over 7 days and the release rate decreased with the increase of molecular weights of the copolymer and the amount of the drug entrapped. These experimental results suggested that the nanoparticles prepared from CMPC block copolymers could be a good candidate for injectable drug delivery carrier.

Advances in Colloid and Interface Science

Abstract:Polymer composite microspheres offer several advantages including highly designable structural properties, adjustable micro-nano particle size distribution, easy surface modification, large specific surface area, and high stability. These features make them valuable in various fields such as medicine, sensing, optics, and display technologies, with significant applications in clinical diagnostics, pathological imaging, and drug delivery in the medical field. Currently, microspheres are primarily used in biomedical research as long-acting controlledrelease agents and targeted delivery systems, and are widely applied in bone tissue repair, cancer treatment, and wound healing. Different types of polymer microspheres offer distinct advantages and application prospects. Efforts are ongoing to transition successful experimental research to industrial production by expanding various fabrication technologies. This article provides an overview of materials used in microsphere manufacturing, different fabrication methods, modification techniques to enhance their properties and applications, and discusses the role of microspheres in drug delivery engineering.
Investigation of size, surface charge, PEGylation degree and concentration on the cellular uptake of polymer nanoparticles

Abstract:In this work a large number of polymer nanoparticles (NPs) with different features have been synthesized through emulsion polymerization-based methods. Poly(methyl methacrylate) (PMMA), poly-ɛ-caprolactone (PCL), and poly(lactic acid) (PLA) based NPs with different size, hydrophobicity, surface charge, PEGylation degree, type of emulsifier and ζ potential have been produced and characterized. All the different NPs have been adopted for cellular uptake studies, leading to a precise quantification of the number of internalized NPs into a selected tumor cell line. The experiments summarize, emphasize and improve the comprehension of the influence of NPs features on the uptake efficiency. In detail, a linear relationship between uptake and both size and NP concentration independently upon other NP characteristics was found. Moreover, it was confirmed that cells are able to internalize and retain for a long time preferentially positively charged NPs. Finally, by coupling results of uptake studies with cell viability measurements, an easy and fast check to control the effectiveness of a selected polymer as drug carrier has been proposed. In particular, we observed that biodegradable PLA-based NPs with high molecular weight, non-PEGylated and positively charged PCL NPs are the better choice to maximize the uptake and minimize side effect against cells.

Polymers for Advanced Technologies

Abstract:Novel stimuli-responsive hydrophilic microspheres were prepared by free radical polymerization of hydroxyethyl methacrylate (HEMA) and methacrylic acid (MA), as hydrophilic monomers, and N-isopropylacrylamide (NIPAAm) and N,N'-ethylenebisacrylamide (EBA), as thermo-sensitive monomer and crosslinker, respectively. Hydrophilic comonomers were introduced in the macromolecular network to synthesize materials with tunable thermal behavior. In addition, by introducing in the polymerization feed both a hydrophilic and a pH-sensitive monomer, such as MA, dual stimuli-responsive (pH and temperature) hydrogels were synthesized. The incorporation of monomers in the network was confirmed by infrared spectroscopy, while the network density and the shape of hydrogels was found to strictly depend on the concentration of monomers in the polymerization feed. Thermal analyses showed negative thermo-responsive behavior with pronounced water affinity of microspheres at a temperature lower than lower critical solution temperature (LCST). In our experiment, the LCST values of the hydrogels were in the range 34.6-37.5 degrees C, close to the body temperature, and the amount of hydrophilic moieties in the polymeric network allows to collect shrinking/swelling transition temperatures higher than the LCST of NIPAAm homopolymers. In order to test the preformed materials as drug carriers, diclofenac diethylammonium salt (DDA) was chosen and drug entrapment percent was determined. Drug release profiles, in media at different temperature and pH, depend on hydrogels crosslinking degree and drug-bead interactions.
