Abstract:Four Holstein steers fed with Sorghum silage were used to examine the effect of fumaric acid supplementation (20g/kg. diet dry matter (DM)) on methane production, rumen fermentation, blood metabolism and feed digestibility. The protozoal population in the rumen was unaffected by fumaric acid supplementation. The postprandial ruminal concentration of ammonia-N decreased, and that of total volatile fatty acids tended to be higher with fumaric acid supplementation. The proportion of ruminal acetic acid was unaffected, but that of propionic acid increased and that of butyric acid decreased by fumaric acid. The postprandial blood plasma concentration of glucose was increased, whereas that of urea-N was decreased by fumaric acid. The plasma concentration of most of free amino acids was unaffected. Daily methane production decreased by 23.0% and carbon dioxide production decreased by 20.5% with fumaric acid supplementation. Apparent digestibility of dry matter and of neutral detergent fiber were not influenced by fumaric acid. These results indicated that fumaric acid was converted to propionic acid by rumen microorganisms, and that methane production from the rumen was reduced without lowering the ability to digest dietary fiber. However, some dietary conditions that alter the effectiveness of fumaric acid and the long term effect remain to be examined.
Influence of medium chain fatty acids on selected microbes and on in vitro ruminal fermentation of air-exposed corn silage

Abstract:Several medium chain fatty acids and different chemical forms of these acids were evaluated in vitro as treatments of aerobically-exposed corn silage against spoilage and pathogenic microbes and for effects on rumen fermentation. Treatments were control (no additive), myristate (MY), laurate (LA), monolaurin (MLA), methyl ester laurate (MELA), a blend of mono-, di- and triglycerides of laurate (BLA), and monocaprylate (MC). After 24 h of aerobic incubation (37°C), yeast and mold growth were not influenced (P > 0.05) by treatments, while the net growth of lactic acid bacteria was decreased, albeit slightly, compared to that by untreated controls (P < 0.01) by all treatments of the air-exposed corn silage. Compared with controls, wild-type enterococci were decreased (P < 0.01) in MLA, MELA, and BLA. Staphylococcus aureus was reduced (P < 0.01) with MLA, MELA, BLA, and MC. Total aerobes showed reductions (P < 0.01) with MLA, BLA, and MC. Listeria monocytogenes numbers were reduced (P < 0.01) with MELA. Anaerobic incubation (24 h; 39°C) of ruminal fluid (10 mL) with 0.2 g air-exposed and MCFA-treated corn silage revealed higher hydrogen accumulations (P < 0.01) with MLA and MC over the control treatment. Methane was decreased (P < 0.01) solely by MLA. There was an increase (P < 0.01) of acetate with MELA and MC; of propionate with MELA or by BLA; and of butyrate with MLA, MELA, BLA, and MC. Total VFA, hexose fermented, and ammonia were increased (P < 0.01) with MELA, BLA, and MC. The acetate:propionate ratio was increased (P < 0.01) with MC. The results showed that treatment of air-exposed corn silage with esterified MCFA had no effect on yeasts and molds but prevented propagation or reduced populations of some unwanted and potentially desirable bacteria. Modest methane reduction was seen during in vitro incubation of rumen fluid suspensions with MLA-treated silage and ammonia accumulations were increased in esterified MCFA-treated silage. Little, if any, other detrimental effects on beneficial ruminal fermentation characteristics were observed.

Abstract:Context: Fodder beet (FB) is a popular feed for dairy cows in temperate climates due to its high yields, high digestibility, low nitrogen (N) content in the dry matter (DM) and convenience of feeding (grazing in situ ). However, the risk of ruminal acidosis requires research to design feeding regimes that capture these benefits without compromising animal health. Aims: To understand aspects of rumen function when FB is offered in conditions representative of practical feeding in temperate pastoral systems. Methods: Two indoor experiments were undertaken; one with cows in late lactation fed fresh perennial ryegrass with three proportions of FB (0, 0.23 and 0.45) and another with non-lactating cows fed pasture silage with 0.65 FB or barley straw with 0.86 FB. Measurements included rumen pH, short-chain fatty acid (SCFA) and ammonia concentrations determined at 2-h intervals, as well as daily individual cow intakes, estimates of microbial growth and rumen dynamics. Key results: The inclusion of 0, 0.23 and 0.45 FB with fresh pasture in the did not affect daily DM intakes (~14.6 kg), milk yield (~10.7 kg), microbial synthesis (129 g of N/d) or fractional outflow rates of digesta (0.16/h; 11.2 L/h) of lactating cows. The non-lactating cow ration comprising 0.86 FB with straw was inappropriate and resulted in low intakes and insufficient dietary N. Microbial growth was approximately one-third of that in cows fed pasture silage with 0.65 FB. The ruminal pH reached lower values in all treatments where FB was offered. Rumen ammonia concentrations averaged 4.4 mmol/L in cows fed pasture but was sometimes undetectable in lactating cows fed 0.45 FB and in non-lactating cows. The amount of FB in the diet affected the extent of the circadian changes in molar proportions of SCFA. Conclusions: Based on the results presented here, feeding fresh FB to dairy cows should not exceed ~0.4 of their DMI with pasture (late lactation), or ~0.6 of their intake with silage (non-lactating). Implications: These findings could support evidence-based recommendations for FB use, considering its effects on aspects of rumen function, such as microbial protein synthesis and pH.

Abstract:Eight rumen-fistulated Holstein cows, averaging 77 d postpartum, were used in a replicated 4 X 4 Latin square design with 28-d periods to investigate the effect of ammonium salts of isobutyrate, 2-methylbutyrate, isovalerate, and n-valerate on animal performance and their possible mechanism(s) and site(s) of action. Each cow was fed ad libitum a complete mixed diet of 55% corn silage and 45% concentrate on a dry basis that was supplemented with 1.8 kg of premix daily. Treatments were 1) control, 2) ammonium salts of volatile fatty acids in premix, 3) ammonium salts of volatile fatty acids ruminally infused, or 4) ammonium salts of volatile fatty acids abomasally infused. Mean ruminal fluid pH and concentrations of ammonia and volatile fatty acids for treatment comparisons were not different. Plasma concentrations of isobutyrate, 2-methylbutyrate, and valerate differed among treatments, but there was no significant effect on dry matter intake, milk production, milk composition, or efficiency of feed utilization. Apparent nutrient digestibility; disappearance of dry matter, cellulose, and nitrogen from polyester bags suspended in the rumen; and plasma concentrations of glucose, free fatty acids, and growth hormone also were not significantly affected by treatment.

Abstract:This study was performed to evaluate the effects of rye silage treated with sodium formate (Na-Fa) and lactic acid bacteria (LAB) inoculants on the ruminal fermentation characteristics, methane yield and energy balance in Hanwoo steers. Forage rye was harvested in May 2019 and ensiled without additives (control) or with either a LAB inoculant or Na-Fa. The LAB (Lactobacillus plantarum) were inoculated at 1.5 × 1010 CFU/g fresh matter, and the inoculant was sprayed onto the forage rye during wrapping at a rate of 4 L/ton of fresh rye forage. Sixteen percent of the Na-Fa solution was sprayed at a rate of approximately 6.6 L/ton. Hanwoo steers (body weight 275 ± 8.4 kg (n = 3, group 1); average body weight 360 ± 32.1 kg (n = 3, group 2)) were allocated into two pens equipped with individual feeding gates and used in duplicated 3 × 3 Latin square design. The experimental diet was fed twice daily (09:00 and 18:00) during the experimental period. Each period comprised 10 days for adaptation to the pen and 9 days for measurements in a direct respiratory chamber. The body weights of the steers were measured at the beginning and at the end of the experiment. Feces and urine were collected for 5 days after 1 day of adaptation to the chamber, methane production was measured for 2 days, and ruminal fluid was collected on the final day. In the LAB group, the ratio of acetic acid in the rumen fluid was significantly lower (p = 0.044) and the ratio of propionic acid in the rumen fluid was significantly higher (p = 0.017). Methane production per DDMI of the Na-FA treatment group was lower than that of the other groups (p = 0.052), and methane production per DNDFI of the LAB treatment group was higher than that of the other groups (p = 0.056). The use of an acid-based additive in silage production has a positive effect on net energy and has the potential to reduce enteric methane emissions in ruminants.
Effect of Hydrogen-Consuming Compounds on In Vitro Ruminal Fermentation, Fatty Acids Profile, and Microbial Community in Water Buffalo

Abstract:The present study aimed to investigate the effect of hydrogen-consuming compounds on ruminal methane (CH4) production, in vitro fermentation parameters, fatty acids profile, and microbial community in water buffalo. Different sodium nitrate to disodium fumarate ratios [2:1 (F), 1:1 (S), 1:2 (T)] were studied in vitro by batch culture technique in the presence of linoleic acid. Results revealed that the dominant bacterial communities were not affected with sodium nitrate and disodium fumarate, whereas CH4 production and Verrucomicrobia, Succiniclasticum, norank_f__Muribaculaceae, and Prevotellaceae_UCG-003 were reduced (P < 0.05). However, ruminal pH, unsaturated fatty acids/saturated fatty acids (UFA/SFA) and Campilobacterota, Selenomonas, Succinivibrio, Oribacterium, Christensenellaceae_R-7_group, Campylobacter, Shuttleworthia, Schwartzia, and Prevotellaceae_YAB2003_group were increased (P < 0.05). Total volatile fatty acids (TVFA) and Spirochaetae, Fibrobacterota, Verrucomicrobia, Fibrobacter, Treponema, and Prevotellaceae were decreased in F (P < 0.05), but cis-9, trans-11CLA, acetate/propionate and Proteobacteria, Campilobacterota, Selenomonas, Succinivibrio, and Campylobacter were increased in F (P < 0.05). The highly selected bacterial genera in F were Campylobacter and Succinivibrio. The disodium fumarate, enhanced (P < 0.05) the TVFA, propionate, total bacteria, Butyrivibrio proteoclasticus, and Atypical butyrivibrio. The concentrations of C18:3n3, C20:3n6, C21:0, C22:2n6, and C22:1n9, as well as the populations of total fungi, protozoa, methanogens, Butyrivibrio hungatei in T were higher (P < 0.05). The highly selected bacterial genera in T were Fibrobacter and Treponema. Conclusively, the addition of sodium nitrate and disodium fumarate can reduce the CH4 production and optimize ruminal fatty acid composition. Furthermore, disodium fumarate can alleviate the adverse effect of sodium nitrate on the rumen fermentation.

Abstract:Background: C16:0 and cis-9 C18:1 may have different effects on animal growth and health due to unique metabolism in vivo. This study was investigated to explore the different effects of altering the ratio of C16:0 and cis-9 C18:1 in fat supplements on growth performance, lipid metabolism, intestinal barrier, cecal microbiota, and inflammation in fattening bulls. Thirty finishing Angus bulls (626 ± 69 kg, 21 ± 0.5 months) were divided into 3 treatments according to the randomized block design: (1) control diet without additional fat (CON), (2) CON + 2.5% palmitic acid calcium salt (PA, 90% C16:0), and (3) CON + 2.5% mixed fatty acid calcium salt (MA, 60% C16:0 + 30% cis-9 C18:1). The experiment lasted for 104 d, after which all the bulls were slaughtered and sampled for analysis. Results: MA tended to reduce 0-52 d dry matter intake compared to PA (DMI, P = 0.052). Compared with CON and MA, PA significantly increased 0-52 d average daily gain (ADG, P = 0.027). PA tended to improve the 0-52 d feed conversion rate compared with CON (FCR, P = 0.088). Both PA and MA had no significant effect on 52-104 days of DMI, ADG and FCR (P > 0.05). PA tended to improve plasma triglycerides compared with MA (P = 0.077), significantly increased plasma cholesterol (P = 0.002) and tended to improve subcutaneous adipose weight (P = 0.066) when compared with CON and MA. Both PA and MA increased visceral adipose weight compared with CON (P = 0.021). Only PA increased the colonization of Rikenellaceae, Ruminococcus and Proteobacteria in the cecum, and MA increased Akkermansia abundance (P < 0.05). Compared with CON, both PA and MA down-regulated the mRNA expression of Claudin-1 in the jejunum (P < 0.001), increased plasma diamine oxidase (DAO, P < 0.001) and lipopolysaccharide (LPS, P = 0.045). Compared with CON and MA, PA down-regulated the ZO-1 in the jejunum (P < 0.001) and increased plasma LPS-binding protein (LBP, P < 0.001). Compared with CON, only PA down-regulated the Occludin in the jejunum (P = 0.013). Compared with CON, PA and MA significantly up-regulated the expression of TLR-4 and NF-κB in the visceral adipose (P < 0.001) and increased plasma IL-6 (P < 0.001). Compared with CON, only PA up-regulated the TNF-α in the visceral adipose (P = 0.01). Compared with CON and MA, PA up-regulated IL-6 in the visceral adipose (P < 0.001), increased plasma TNF-α (P < 0.001), and reduced the IgG content in plasma (P = 0.035). Compared with CON, PA and MA increased C16:0 in subcutaneous fat and longissimus dorsi muscle (P < 0.05), while more C16:0 was also deposited by extension and desaturation into C18:0 and cis-9 C18:1. However, neither PA nor MA affected the content of cis-9 C18:1 in longissimus dorsi muscle compared with CON (P > 0.05). Conclusions: MA containing 30% cis-9 C18:1 reduced the risk of high C16:0 dietary fat induced subcutaneous fat obesity, adipose tissue and systemic low-grade inflammation by accelerating fatty acid oxidative utilization, improving colonization of Akkermansia, reducing intestinal barrier damage, and down-regulating NF-κB activation.

Abstract:Abstract The objective of this study was to evaluate effects of feeding supplemental isoacids in a growing diet on parameters of rumen fermentation and in situ DM disappearance. Eight mature ruminally cannulated beef heifers (Angus crossbred; mean BW 618 ± 31 kg) were used in a replicated 4 x 4 Latin square design. Heifers were housed in individual pens and fed the following treatment diets: no supplemental isoacids (ISO) in the basal diet (Control); 7.2 g of 2-methylbutyric acid· animal-1· day-1 (ISO1); 7.2 g of 2-methylbutyric + 7.2 g isobutyric acids· animal-1· day-1 (ISO2); and 7.2 g 2-methylbutyric + 7.2 g isobutyric + 7.2 g isovaleric acid· animal-1· day-1 (ISO3). Basal diet consisted of (DM basis) 26.9% wheat hay, 35.5% steam flaked corn, 22.6% wet corn gluten feed, 12.3% DDG, and 2.7% mineral/vitamin supplement. Each experimental period lasted 28 d, with 21 d for diet adaptation and 7 d for data and sample collection. On day 1 of each sampling period ruminal fluid was collected 2 hours before and 1, 3, 7 and 10 hours after morning feeding. The pH of ruminal contents was immediately measured with an electrode pH meter. Urine samples (approximately 200 mL) were collected on day 2 of each sampling period 4 h after the feeding and filtered to remove particles. Urine was analyzed for creatinine, uric acid, and allantoin. Bags were placed into rumen for 72, 48, 36, 24, 12, 8, 4 and 0 hours and all bags were removed at same time on the end of incubation (hour 0), rinsed with tap water and dried at 55°C. Data were analyzed using the MIXED procedure of SAS. A contrast was observed (P = 0.029) for overall rumen pH between control versus isoacid treatments (6.44 vs 6.31). Uric acid, allantoin, total purines and intestinal flow of microbial nitrogen were not affected by treatments (P ≥ 0.10) with overall means of 10.47, 328.80, 339.27 mmol/d and 246.64 g N/d, respectively. Overall, no effects were observed for in situ dry matter disappearance (P ≥ 0.10). However, at hour 4 differences between ISO2 and ISO3 (P = 0.02; 44.89% vs 38.45%) and ISO2 and ISO4 (P = 0.05; 44.89% vs 39.75%) were observed. No effects of in situ DM disappearance was observed (P ≥ 0.10). Supplementing beef heifers with isoacids may reduce rumen pH as a result of increased ruminal volatile fatty acid production; however, supplemental ISO in this study did not affect microbial nitrogen synthesis or dry matter in situ disappearance. Further analyses are currently in progress.

Abstract:Objective: The objectives of this study were to evaluate the effects of different forage proportions in the fermented total mixed ration (FTMR) on growth performance, muscle fatty acid profile, and rumen microbiota of lambs. Methods: Thirty 6-month-old small tail Han sheep × Ujumqin lambs with initial body weight (BW) of 27.8 ± 0.90 kg were selected for the test and divided into two groups of 15 sheep in each treatment (three pens per treatment and five lambs per pen) according to the principle of homogeneity. Two isoenergetic and isonitrogenous diets were formulated according to the NRC. The diet treatments were designed as (1) OH treatment containing 25% alfalfa hay and 35% oat hay, and (2) AH treatment containing 35% alfalfa hay with 25% oat hay. The forage-to-concentrate ratio for both diets was 65: 35 (DM basis). Three replicates were randomly selected from each treatment to determine growth performance, fatty acid profile and rumen bacterial communities in lambs. Results: Results revealed no statistically significant ( p > 0.05) differences in dry matter intake and average daily gain between the two diet groups. Cholesterol and intramuscular fat were significantly ( p > 0.05) higher in the AH group, while no statistically significant difference ( p > 0.05) was found in pH24 value. The muscle fatty acid compositions of lambs were obviously ( p < 0.05) influenced by the diet treatments. Compared with the OH group, the C16:1, C17:0, and C20:3n6 contents were higher ( p < 0.05) in the AH group, whereas the content of C18:1n9c, C20:1, C18:3n3, and C22:6n3 was obviously ( p < 0.05) increased in the OH group. The monounsaturated fatty acid (MUFA) contents were significantly higher in the OH group, whereas no significant differences ( p > 0.05) were detected in saturated fatty acid (SFA) and polyunsaturated fatty acid (PUFA) contents among the two diet treatments. Bacterial composition was generally separated into two clusters based on principal coordinate analysis, and the OH group had a higher Shannon index. The relative abundance at the genes level of the Rikenellaceae_RC9_gut_group was obviously ( p < 0.05) increased in the AH group and the relative abundances of Prevotella_1, Fibrobacter , and Bacteroidales_UCG_001_unclassified were obviously ( p < 0.05) enriched in the OH group. Integrated correlation analysis also underscored a possible link between the muscle fatty acid compositions and significantly altered rumen microbiota. Conclusion: Overall, oat-based roughage in FTMR could promote a beneficial lipid pattern in the Longissimus lumborum muscles of lambs. These findings provide a potential insight into diet effects on fatty acid profile and the rumen microbiome of lambs, which may help make decisions regarding feeding.

Abstract:The dietary energy level can affect ruminal microbiota, and further can affect rumen fermentation and fatty acid (FA) synthesis. In this study, we investigated the correlations between rumen bacteria and rumen fermentation parameters and intramuscular fat (IMF) FA profiles of Holstein bulls fed different energy diets via using 16S rRNA high-throughput sequencing and gas chromatography. The results showed that the improved dietary energy increased propionate, isobutyrate and isovalerate concentrations, and decreased acetate concentration and the acetate/propionate ratio. Increased dietary energy improved beef IMF content and had no effects on cooking loss, Warner-Bratzler shear force, water holding capacity, or drip loss. Increase dietary energy also decreased C18:0, C18:1 trans, C22:0, C20:3n-3, C22:6n-3, and saturated fatty acids, and increased C18:1 cis-9, C18:2n-6 trans, and monounsaturated fatty acids. 16S rRNA high-throughput sequencing analysis revealed that dietary energy had no impact on alpha diversity or the relative abundance of most of the major phyla and genera in rumen. In all dietary treatment groups, the dominant microbial phyla were Bacteroidetes (54.91%) and Firmicutes (33.60%), and the major microbial genus was Prevotella_1 (21.75%). Improved dietary energy decreased the abundances of Firmicutes and Tenericutes and increased that of Proteobacteria at the phylum level, while decreasing those of RC9_gut_group, and increased Prevotellaceae_UCG-004, Phocaeicola, Acetitomaculum, Lachnoclostridium_1, Prevotellaceae_UCG-003, and Anaerovibrio at the genus level. Spearman correlation analysis showed high correlations between rumen bacteria and fermentation parameters/IMF FA profiles. Collectively, our data indicated that dietary energy affects the ruminal microbiota, and further affects ruminal fermentation and IMF FA composition.

Abstract:The potential of dietary supplements of 2 live yeast strains (Saccharomyces cerevisiae) or camelina oil to lower ruminal methane (CH4) and carbon dioxide (CO2) production and the associated effects on animal performance, rumen fermentation, rumen microbial populations, nutrient metabolism, and milk fatty acid (FA) composition of cows fed grass silage-based diets were examined. Four Finnish Ayrshire cows (53±7 d in milk) fitted with rumen cannula were used in a 4×4 Latin square with four 42-d periods. Cows received a basal total mixed ration (control treatment) with a 50:50 forage-to-concentrate ratio [on a dry matter (DM) basis] containing grass silage, the same basal total mixed ration supplemented with 1 of 2 live yeasts, A or B, administered directly in the rumen at 10(10) cfu/d (treatments A and B), or supplements of 60g of camelina oil/kg of diet DM that replaced concentrate ingredients in the basal total mixed ration (treatment CO). Relative to the control, treatments A and B had no effects on DM intake, rumen fermentation, ruminal gas production, or apparent total-tract nutrient digestibility. In contrast, treatment CO lowered DM intake and ruminal CH4 and CO2 production, responses associated with numerical nonsignificant decreases in total-tract organic matter digestibility, but no alterations in rumen fermentation characteristics or changes in the total numbers of rumen bacteria, methanogens, protozoa, and fungi. Compared with the control, treatment CO decreased the yields of milk, milk fat, lactose, and protein. Relative to treatment B, treatment CO improved nitrogen utilization due to a lower crude protein intake. Treatment A had no influence on milk FA composition, whereas treatment B increased cis-9 10:1 and decreased 11-cyclohexyl 11:0 and 24:0 concentrations. Treatment CO decreased milk fat 8:0 to 16:0 and total saturated FA, and increased 18:0, 18:1, 18:2, conjugated linoleic acid, 18:3n-3, and trans FA concentrations. Decreases in ruminal CH4 production to treatment CO were related, at least in part to lowered DM intake, whereas treatments had no effect on ruminal CH4 emission intensity (g/kg of digestible organic matter intake or milk yield). Results indicated that live yeasts A and B had no influence on animal performance, ruminal gas production, rumen fermentation, or nutrient utilization in cows fed grass silage-based diets. Dietary supplements of camelina oil decreased ruminal CH4 and CO2 production, but also lowered the yields of milk and milk constituents due to an adverse effect on intake.

Abstract:This study evaluated the effects of different proportions of palmitic (C16:0) and oleic (cis-9 C18:1) acids in fat supplements on rumen fermentation, glucose (GLU) and lipid metabolism, antioxidant function, and visceral fat fatty acid (FA) composition in Angus bulls. The design of the experiment was a randomized block design with 3 treatments of 10 animals each. A total of 30 finishing Angus bulls (21 ± 0.5 months) with an initial body weight of 626 ± 69 kg were blocked by weight into 10 blocks, with 3 bulls per block. The bulls in each block were randomly assigned to one of three experimental diets: (1) control diet without additional fat (CON), (2) CON + 2.5% palmitic calcium salt (PA; 90% C16:0), (3) CON + 2.5% mixed FA calcium salts (MA; 60% C16:0 + 30% cis-9 C18:1). Both fat supplements increased C18:0 and cis-9 C18:1 in visceral fat (P < 0.05) and up-regulated the expression of liver FA transport protein 5 (FATP5; P < 0.001). PA increased the insulin concentration (P < 0.001) and aspartate aminotransferase activity (AST; P = 0.030) in bull's blood while reducing the GLU concentration (P = 0.009). PA increased the content of triglycerides (TG; P = 0.014) in the liver, the content of the C16:0 in visceral fat (P = 0.004), and weight gain (P = 0.032), and up-regulated the expression of liver diacylglycerol acyltransferase 2 (DGAT2; P < 0.001) and stearoyl-CoA desaturase 1 (SCD1; P < 0.05). MA increased plasma superoxide dismutase activity (SOD; P = 0.011), reduced the concentration of acetate and total volatile FA (VFA) in rumen fluid (P < 0.05), and tended to increase plasma non-esterified FA (NEFA; P = 0.069) concentrations. Generally, high C16:0 fat supplementation increased weight gain in Angus bulls and triggered the risk of fatty liver, insulin resistance, and reduced antioxidant function. These adverse effects were alleviated by partially replacing C16:0 with cis-9 C18:1.

Abstract:OBJECTIVE: The objective of this experiment was to examine the effects of different amylose/amylopectin ratios on rumen fermentation and development of fattening lambs.METHODS: Forty-eight 7-day-old male Small-tailed Han sheep×Northeast fine wool sheep were randomly assigned to four treatments of dietary amylose/amylopectin ratios (0.12, 0.23, 0.24, and 0.48 in tapioca starch, corn starch, wheat starch and pea starch diets, respectively). Three lambs from each treatment were slaughtered at 21, 35, 56, and 77 days of age to determine the rumen fermentation and development.RESULTS: Compared with tapioca starch diet, the pea starch diet significantly increased the concentration of ammonia nitrogen in the ruminal fluid of lambs but significantly decreased the bacterial protein content. At 56 and 77 d, the rumen propionate concentration tended to be greatest in the tapioca starch group than in other groups. The rumen butyrate concentration was the greatest in lambs fed on pea starch compared with those fed on other starch diets. Furthermore, the pea starch diet significantly stimulated rumen development by increasing the papillae height, width and surface area in the rumen ventral or dorsal locations in lambs. However, different amylose/amylopectin ratios diets did not significantly affect the feed intake, body weight, average daily gain, the relative weight and capacity of the rumen in lambs with increasing length of trial periods.CONCLUSION: Lambs early supplemented with a high amylose/amylopectin ratio diet had favourable morphological development of rumen epithelium, which was not conducive to bacterial protein synthesis.

Abstract:Enteric fermentation is one of the largest contributors of methane release to the environment from the livestock sector. Plant bioactive compounds can modulate rumen fermentation for reduced methanogenesis and fatty acid biohydrogenation. The present study investigates the effects of tannin extract from Ficus glomerata (FG) leaves on the rumen fermentation, methanogenesis, feed digestibility and fatty acid biohydrogenation of a total mixed ration with the aim of developing a feed supplement for enhanced livestock production and product quality with lower methane emission. The tannin extract (70% aqueous acetone extract) of FG leaves in the total mixed ration (oat hay/concentrate mixture; 1:1) was studied at four graded dose regimens (0.0 (control), 0.25 mL (FG-0.25), 0.50 mL (FG-0.50) and 1.0 mL (FG-1.0) per 60 mL of buffered rumen fluid) in three replicates for each treatment in a radio-frequency-based automatic gas production system (ANKOM-RF) at 39 °C for 24 h following the standard in vitro gas production protocol. The total gas production (mL or mL/g incubated dry matter (DM)) was gradually reduced (p 0.05) for FG-0.25 with the control and FG-0.50. Compared to the control, the methane concentration (%) in the head space gas, as well as the total methane production (mL or mL/g DM incubated, or mL/g DM digested), were found to be gradually reduced (p < 0.01) with increasing doses (0.25–1.0 mL) of FG extract. The reduced (p 0.05) effects with the control at a lower level of supplementation (FG-0.25) are suggestive of the dose-responsive detrimental effects of tannins on fibrolytic microbes in the rumen. However, the ammonia concentration decreased (p 0.05) with enhanced (p < 0.05) propionate at a lower dose (FG-0.25); however, a dose-dependent reduction was evident at higher dose levels (FG-0.50 and FG-1.0). The production of stearic acid (C18:0), which is a product of the rumen biohydrogenation process, was reduced (p < 0.05), irrespective of the concentration of the FG extract. Compared to the control, the concentration of t-vaccenic acid (C18:1), which is a precursor of conjugated linoleic acid (CLA) in animal products, was increased in all the FG-extract-supplemented groups. It may be concluded that Ficus glomerata leaf tannins can modulate rumen fermentation for reduced methanogenesis and fatty acid biohydrogenation in a total mixed ration. As a higher level of inclusion negatively affects feed digestibility, a lower dose (0.25 mL FG extract per 60 mL fermentation fluid or 4.17 mL FG extract per L of fermentation fluid) is suggested to achieve desirable effects on methane abatement (30%) and an improvement in fatty acid profiles in animal products.

Abstract:High-concentrate diets can lead to subacute ruminal acidosis and are known to result in changes of the ruminal fermentation pattern and mammary secretion of fatty acids. The objective of this paper is to describe modifications in milk fatty acid proportions, particularly odd- and branched-chain fatty acids and rumen biohydrogenation intermediates, associated with rumen parameters during a 6-wk subacute ruminal acidosis induction protocol with 12 ruminally fistulated multiparous cows. The protocol involved a weekly gradual replacement of a standard dairy concentrate with a wheat-based concentrate (610 g of wheat/kg of concentrate) during the first 5 wk and an increase in the total amount of concentrate in wk 6. Before the end of induction wk 6, cows were switched to a control diet because 7 cows showed signs of sickness. The pH was measured continuously by an indwelling pH probe. Milk and rumen samples were taken on d 2 and 7 of each week. Data were analyzed using a linear mixed model and by principal component analysis. A pH decrease occurred after the first concentrate switch but rumen parameters returned to the original values and remained stable until wk 5. In wk 5 and 6, rumen pH values were indicative of increasing acidotic conditions. After switching to the control diet in wk 6, rumen pH values rapidly achieved normal values. Odd- and branched-chain fatty acids and C18:1 trans-10 increased with increasing amount of concentrate in the diet, whereas C18:1 trans-11 decreased. Four fatty acids [C18:1 trans-10, C15:0 and C17:0+C17:1 cis-9 (negative loadings), and iso C14:0 (positive loading)] largely correlated with the first principal component (PC1), with cows spread along the PC1 axis. The first 4 wk of the induction experiment showed variation across the second principal component (PC2) only, with high loadings of anteiso C13:0 (negative loading) and C18:2 cis-9,trans-11 and C18:1 trans-11 (positive loadings). Weeks 5 and 6 deviated from PC2 and tended toward the negative PC1 axis. A discriminant analysis using a stepwise approach indicated the main fatty acids discriminating between the control and acidotic samples as iso C13:0, iso C16:0, and C18:2 cis-9,trans-11 rather than milk fat content or C18:1 trans-10, which have been used before as indicators of acidosis. This shows that specific milk fatty acids have potential in discriminating acidotic cases.

Abstract:A 48 year old man with normal hepatic function presented with abnormally prolonged bleeding following orally administered warfarin. Warfarin and diphenylhydantoin are known to be hydroxylated in the liver. To investigate the possibility of the patient having a metabolic (hydroxylating) defect, his ability to metabolize diphenylhydantoin was studied, as it would have been unethical to administer warfarin again. Parallel studies were carried out on two healthy subjects as a basis for comparison. On three separate occasions the elimination half-life for diphenylhydantoin in the patient (158, 87, 72 hours) was significantly longer than those obtained in the normal subjects (31 and 32 hours). This difference was most probably due to decreased hydroxylation of diphenylhydantoin to 5-(p-hydroxy-phenyl)-5-phenylhydantoin since his urinary elimination of this substance was demonstrated to be significantly less than that of the normal subjects. The results suggest that this patient had a reduced capacity to hydroxylate diphenylhydantoin, and this defect in liver hydroxylation may explain his increased sensitivity to warfarin.
Effect of acidosis in the late-finishing phase on rumen fermentation in feedlot steers

Abstract:The objective was to determine the effects of induced acidosis in the late-finishing phase on rumen fermentation in feedlot steers. Eleven ruminally cannulated steers (body weight [BW] = 795 kg ± 54) were blocked into two groups based on initial BW. For 195 d prior to the start of the study, cattle were consuming a basal finishing diet (60% dry-rolled corn, 15% modified distillers grains, 15% corn silage, and 10% ground corn-based supplement). Steers were randomly assigned to one of the two treatments: control (CON), or induced acidosis (ACD). Both treatments were fasted for 24 h then fed the basal finishing diet. Steers on the ACD treatment received 0.05% of BW of wheat starch via rumen cannula at 0800 and 2000 hours on day 1 and ad libitum refeeding following the fast. On days 1 and 2, CON steers were provided 25% of allotted feed every 6 h. Rumen fluid was collected every 4 h during the challenge period (hours 0 to 48), and 0, 6, and 12 h after feeding during the recovery period (hours 54 to 96). Rumen fluid was analyzed for pH, ammonia, volatile fatty acids (VFA), and lactate. Fecal grab samples were collected every 8 h to determine fecal pH. A treatment × day interaction (P = 0.03) was observed for dry matter intake during the challenge period with steers on the ACD treatments consuming more on day 1 than CON steers. Intake was not different on day 2 (P = 0.88). A treatment × hour effect (P < 0.01) was observed for ruminal pH during the challenge period with the ACD steers having a lesser pH than CON from hours 12 to 32. Duration of time below a pH of 5.6 during the challenge period was greater (P < 0.01) for ACD steers than CON. During the challenge period, a treatment × time interaction (P = 0.04) was observed for total VFA concentration with ACD steers having greater total VFA concentration from hours 12 to 36. Acetate to propionate ratio (A:P) was affected by treatment × hour (P = 0.04) with CON steers having greater A:P from hours 28 to 48. Rumen ammonia and lactate concentrations did not differ (P ≥ 0.25) between treatments or the interaction with time. Challenge and recovery period fecal pH were not affected (P ≥ 0.13) by treatment, time, or their interaction. Recovery period ruminal pH was not different (P = 0.99) between treatments. For the recovery period, total VFA and ammonia concentration were not affected by treatment, time, or their interaction (P ≥ 0.07). Ruminal pH and VFA were affected in the initial 48 h of induced acidosis in the late-finishing phase.

Abstract:Abstract Introduction The use of probiotics and phytobiotics has attracted interest because of their protective effect against acidosis. Ferula elaeochytris (FE) is considered a good source of bioactive compounds, mainly monoterpene α-pinene. This study aimed to investigate the effect of a direct-fed microbial blend (Pro) and FE on rumen fermentation parameters in vitro under normal and acidosis conditions. Material and Methods An in vitro experiment using the Hohenheimer Futterwerttest (HFT) gas production system was conducted. An acidosis challenge was made to compare the effectiveness of the probiotics blend and FE extract on ruminal pH regulation. To generate different ruminal fermentation parameters, the design of the trial considered the 2 additives (Pro and FE) × 6 incubation times (2, 4, 8, 12, 24 and 48 h) × 2 conditions (acidosis and normal) × 2 incubation runs for each feedstuff (barley, alfalfa and straw). Results An acidosis challenge was successfully induced. The Pro and FE additives had no impact on the observed rumen fermentation parameters such as volatile fatty acid concentration or ammonia (P = 0.001). The acidosis condition decreased total in vitro degradability (IVD) by 3.5% and 21.9% for barley and straw, respectively (P < 0.001). The additives had different significant effects on the IVD of nutrients during both normal and acidosis conditions. In alfalfa samples, FE supplementation significantly decreased the IVD of all observed nutrients under the ruminal acidosis condition, although it had no effect during the normal condition. Conclusion An acidosis challenge was successfully induced and the effect of additives was varied on fermentation parameters and rumen degradability of different feeds either under normal or acidosis conditions.
