Abstract:Vital physiological processes that drive the insect molt represent areas of interest for the development of alternative control strategies. The western tarnished plant bug (Lygus hesperus Knight) is a pest of numerous agronomic and horticultural crops but the development of novel control approaches is impeded by limited knowledge of the mechanisms regulating its molt. To address this deficiency, we examined the fundamental relationship underlying the hormonal and molecular components of ecdysis. At 27°C L. hesperus exhibits a temporally controlled nymph-adult molt that occurs about 4 days after the final nymph-nymph molt with ecdysteroid levels peaking 2 days prior to the final molt. Application of exogenous ecdysteroids when endogenous levels had decreased disrupted the nymphal-adult molt, with treated animals exhibiting an inability to escape the old exoskeleton and resulting in mortality compared to controls. Using accessible transcriptomic data, we identified 10 chitinase-like sequences (LhCht), eight of which had protein motifs consistent with chitinases. Phylogenetic analyses revealed orthologous relationships to chitinases critical to molting in other insects. RT-PCR based transcript profiling revealed that expression changes to four of the LhChts was coordinated with the molt period and ecdysteroid levels. Collectively, our results support a role for ecdysteroid regulation of the L. hesperus molt and suggest that cuticle clearance is mediated by LhCht orthologs of chitinases that are essential to the molt process. These results provide the initial hormonal and molecular basis for future studies to investigate the specific roles of these components in molting.

Abstract:Molting is controlled by ecdysteroids, which are synthesized and secreted by the Y-organ in crustaceans. Ecdysone inducible gene, E75, is an early-response gene in the 20-hydroxyecdysone (20E) signaling pathway, with crucial roles in arthropod development. Complementary DNA (cDNA) encoding Penaeus monodon E75 (PmE75) was cloned using RT-PCR and RACE. PmE75 cDNA was 3526 bp long and encoded a 799-amino acid protein. Tissue distribution analysis showed that PmE75 was expressed ubiquitously in selected tissues, and was relatively abundant in the epidermis, muscle, and hepatopancreas. Developmental expression revealed that PmE75 was expressed throughout its life cycle. Silencing PmE75 significantly decreased PmE75 expression. Shrimps injected with PBS and dsGFP started molting on Day 7 and had almost completed molting on Day 9, whereas dsPmE75-injected shrimp presented no signs of molting. These results suggested that PmE75 might be involved in molting. In situ hybridization results support this hypothesis. To explore the role of 20E and eyestalks in the regulation of molting in P. monodon, exogenous 20E injection and eyestalk ablation (ESA) were performed, and showed that 20E can induce the transcription and expression of PmE75 in the hepatopancreas, epidermis, and muscle, which were signi?cantly elevated after ESA. These results provide further insights into our understanding of molting.
Comparative transcriptomic characterization of the eyestalk in Pacific white shrimp (Litopenaeus vannamei) during ovarian maturation

Abstract:In crustaceans, some of fundamental regulatory processes related to a range of physiological functions, including ovarian maturation, molting, glucose homeostasis, osmoregulation, etc., occur in the organs of the eyestalk. Additionally, reproduction is regulated by neuropeptide hormones and other proteins released from secretory sites (X-organ/sinus gland, XO/SG) within the eyestalk. As unilateral eyestalk ablation was the most common method used to artificially induce ovarian maturation for farmed Litopenaeus vannamei, to better understand the reproductive regulation mechanism in L. vannamei, we have investigated the transcriptomes of the eyestalk during five ovary developmental stages with or without eyestalk ablation by high-throughput Illumina sequencing technology. The raw reads were assembled and clustered into 127,031 unigenes. Meanwhile, the differentially expressed genes (DEGs) between ovarian development stages were identified. We examined, through DEG enrichment analysis, eyestalk gene expression patterns for Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways, comparing natural to artificially induced ovarian maturation. We also identified a variety of transcripts that appear to be differentially expressed throughout ovarian maturation. These include transcripts that encode G-protein coupled receptors (GPCRs) and neuropeptides, such as the crustacean hyperglycemic hormone (CHH), molt-inhibiting hormone (MIH), and crustacean female sex hormone (CFSH). Furthermore, numerous exoskeleton formation-related genes were found to be down-regulated during ovarian maturation, including cuticle-like proteins, eclosion hormone (EH), and gastrolith-like proteins, of which the latter are the first reported in L. vannamei. Our work is the first reproduction-related investigation of L. vannamei focusing on the eyestalk at the whole transcriptome level. These findings provide novel insight into the function of the eyestalk in reproduction regulation.
Hormonal control of the crustacean molting gland: Insights from transcriptomics and proteomics

Abstract:Endocrine control of molting in decapod crustaceans involves the eyestalk neurosecretory center (X-organ/sinus gland complex), regenerating limbs, and a pair of Y-organs (YOs), as molting is induced by eyestalk ablation or multiple leg autotomy and suspended in early premolt by limb bud autotomy. Molt-inhibiting hormone (MIH) and crustacean hyperglycemic hormone (CHH), produced in the X-organ/sinus gland complex, inhibit the YO. The YO transitions through four physiological states over the molt cycle: basal in intermolt; activated in early premolt; committed in mid- and late premolt; and repressed in postmolt. We assembled the first comprehensive YO transcriptome over the molt cycle in the land crab, Gecarcinus lateralis, showing that as many as 23 signaling pathways may interact in controlling ecdysteroidogenesis. A proposed model of the MIH/cyclic nucleotide pathway, which maintains the basal YO, consists of cAMP/Ca2+ triggering and nitric oxide (NO)/cGMP summation phases. Mechanistic target of rapamycin (mTOR) signaling is required for YO activation in early premolt and affects the mRNA levels of thousands of genes. Transforming Growth Factor-β (TGFβ)/Activin signaling is required for YO commitment in mid-premolt and high ecdysteroid titers at the end of premolt may trigger YO repression. The G. lateralis YO expresses 99 G protein-coupled receptors, three of which are putative receptors for MIH/CHH. Proteomic analysis shows the importance of radical oxygen species scavenging, cytoskeleton, vesicular secretion, immune response, and protein homeostasis and turnover proteins associated with YO function over the molt cycle. In addition to eyestalk ganglia, MIH mRNA and protein are present in brain, optic nerve, ventral nerve cord, and thoracic ganglion, suggesting that they are secondary sources of MIH. Down-regulation of mTOR signaling genes, in particular Ras homolog enriched in brain or Rheb, compensates for the effects of elevated temperature in the YO, heart, and eyestalk ganglia in juvenile Metacarcinus magister. Rheb expression increases in the activated and committed YO. These data suggest that mTOR plays a central role in mediating molt regulation by physiological and environmental factors.

Abstract:The analysis of growth bands in the eyestalk has been increasingly used for estimating crustacean ageing and molting. In this study, we developed an effective method to process and observe the eyestalk microstructure of the swimming crab (Portunus trituberculatus). We found that dark pigmentation as a result of boiling has an influence on the observation of the eyestalk microstructure. Choosing an unboiled eyestalk, this study compared the cross section and longitudinal section, and concluded that the cross section is suitable for the observation of growth increments with 6.1% CV (coefficient of variation), and growth bands are suitable for the observation of the longitudinal section with 5.4% CV. The width of growth increments near the edge of the endocuticle is small, and the width of growth increments of the middle part of the endocuticle is large. Relationship of number of growth bands to molting time was fitted to a linear function with the slope not significantly different from 1, indicating that growth bands are formed associated with molting. Periodicity of growth increment formation was calculated as 3.7 d, however was not verified. Our results provide a new improved technique for identification of crustacean molting and growth.

Abstract:The crustacean molting process is regulated by an interplay of hormones produced by the eyestalk ganglia and Y-organs (YO). Molt-inhibiting hormone and crustacean hyperglycemic hormone released by the sinus gland of the eyestalk ganglia (EG) inhibit the synthesis and secretion of ecdysteroid by the YO, hence regulating hemolymph levels during the molt cycle. The purpose of this study is to investigate the ecdysteroidogenesis pathway, specifically genes linked to changes in ecdysteroid levels occurring at early premolt (ePM). To this end, a reference transcriptome based on YO, EG, and hepatopancreas was de novo assembled. Two genes (cholesterol 7-desaturase Neverland and cytochrome p450 307a1-like Spook) involved in ecdysteroidogenesis were identified from the YO transcriptome using sequence comparisons and transcript abundance. Two other candidates, Hormone receptor 4 and probable cytochrome p450 49a1 potentially involved in ecdysteroidogenesis were also identified. Since cholesterol is the ecdysteroid precursor, a putative cholesterol carrier (Apolipoprotein D-like) was also examined to understand if cholesterol uptake coincided with the increase in the ecdysteroid levels at the ePM stage. The expression level changes of the five candidate genes in the YO were compared between intermolt (IM) and induced ePM (iePM) stages using transcriptomic analysis. Expression analysis using qPCR were carried out at IM, iePM, and normal ePM. The increase in Spook and Neverland expression in the YO at the ePM was accompanied by a concomitant rise in ecdysteroid levels. The data obtained from iePM stage were congruent with those obtained from the normal ePM stage of intact control animals. The present findings support the role of Halloween genes in the ecdysteroidogenesis and molt cycle in the blue crab, Callinectes sapidus.

Abstract:Background: G-protein coupled receptors (GPCRs) are ancient, ubiquitous, constitute the largest family of transducing cell surface proteins, and are integral to cell communication via an array of ligands/neuropeptides. Molt inhibiting hormone (MIH) is a key neuropeptide that controls growth and reproduction in crustaceans by regulating the molt cycle. It inhibits ecdysone biosynthesis by a pair of endocrine glands (Y-organs; YOs) through binding a yet uncharacterized GPCR, which triggers a signalling cascade, leading to inhibition of the ecdysis sequence. When MIH release stops, ecdysone is synthesized and released to the hemolymph. A peak in ecdysone titer is followed by a molting event. A transcriptome of the blackback land crab Gecarcinus lateralis YOs across molt was utilized in this study to curate the list of GPCRs and their expression in order to better assess which GPCRs are involved in the molt process. Results: Ninety-nine G. lateralis putative GPCRs were obtained by screening the YO transcriptome against the Pfam database. Phylogenetic analysis classified 49 as class A (Rhodopsin-like receptor), 35 as class B (Secretin receptor), and 9 as class C (metabotropic glutamate). Further phylogenetic analysis of class A GPCRs identified neuropeptide GPCRs, including those for Allatostatin A, Allatostatin B, Bursicon, CCHamide, FMRFamide, Proctolin, Corazonin, Relaxin, and the biogenic amine Serotonin. Three GPCRs clustered with recently identified putative CHH receptors (CHHRs), and differential expression over the molt cycle suggests that they are associated with ecdysteroidogenesis regulation. Two putative Corazonin receptors showed much higher expression in the YOs compared with all other GPCRs, suggesting an important role in molt regulation. Conclusions: Molting requires an orchestrated regulation of YO ecdysteroid synthesis by multiple neuropeptides. In this study, we curated a comprehensive list of GPCRs expressed in the YO and followed their expression across the molt cycle. Three putative CHH receptors were identified and could include an MIH receptor whose activation negatively regulates molting. Orthologs of receptors that were found to be involved in molt regulation in insects were also identified, including LGR3 and Corazonin receptor, the latter of which was expressed at much higher level than all other receptors, suggesting a key role in YO regulation.

Abstract:The eyestalks of crustaceans play an essential role in controlling a variety of physiological functions by converting light into hormonal signals. To obtain a more complete description of eyestalk biology in the commercially important Chinese mitten crab (Eriocheir sinensis), we conducted comparative transcriptome analysis of eyestalks during the day and at night using high-throughput sequencing on an Illumina HiSeq 4000 platform. We obtained 47,092 unigenes-including 4771 differentially expressed genes (DEGs)-from eyestalks during the day and at night. We found that 4269 DEGs were upregulated during the day and 502 DEGs were upregulated at night. We identified five DEGs that may contribute to molting, including molt-inhibiting hormone, cuticle, catalase, aquaporin, and ubiquitin-conjugating enzyme; hence, similar to other crustaceans, Eriocheir sinensis may molt at night. We further identified eight DEGs related to behavior regulation, including three glutamate dehydrogenase genes that were upregulated during the day. Thus, changes in the eyestalks may partially compensate for daily changes in illumination in the Chinese mitten crab's normal environment. Our present study is the first genome-wide transcriptome analysis of the eyestalks of Eriocheir sinensis during the day and at night. Our findings provide a valuable insight into the molecular basis of circadian cycle regulation in crustaceans.

Abstract:Considerable differences in weight gain rates (WGRs) after molting is a puzzling phenomenon exhibited by individuals of the Chinese mitten crab Eriocheir sinensis even under the same environmental conditions. However, the mechanisms underlying the control of growth difference during the molting cycle have not been illustrated. In the present study, juvenile E. sinensis individuals were cultured in the same environment during one molting cycle. After molting, the E. sinensis individuals were classified into the high- and low-WGR groups on the basis of WGRs. Hemolymph samples were collected at the intermolt (InM), premolt (PrM) and postmolt (PoM) stages of the molting cycle for metabolomic and transcriptomic analyses. Differential metabolites (DMs) and differentially expressed genes (DEGs) at the InM, PrM, and PoM stages were identified and mapped to Kyoto Encyclopedia of Genes and Genomes pathways. Integrated metabolomic and transcriptomic analyses revealed significantly varied metabolic pathways between different WGR groups at the three molting stages. These pathways involved amino acid metabolism, nucleotide metabolism, and energy metabolism. Results showed that the high-WGR group exhibited more active cellular protein biosynthesis, a robust neuroendocrine system, and higher antioxidant and detoxifying capabilities than the low-WGR group, which may promote weight gain during the molting cycle. This study provided novel insights into the molecular and metabolic mechanisms in controlling the growth of E. sinensis during the molting cycle, and afforded the theoretical basis for further aquaculture and selective breeding practices of crustacean species.

Abstract:Landsat 8 is the first satellite in the Landsat mission to acquire spectral imagery of the Earth using pushbroom sensor instruments. As a result, there are almost 70,000 unique detectors on the Operational Land Imager (OLI) alone to monitor. Due to minute variations in manufacturing and temporal degradation, every detector will exhibit a different behavior when exposed to uniform radiance, causing a noticeable striping artifact in collected imagery. Solar collects using the OLI's on-board solar diffuser panels are the primary method of characterizing detector level non-uniformity. This paper reports on an approach for using a side-slither maneuver to estimate relative detector gains within each individual focal plane module (FPM) in the OLI. A method to characterize cirrus band detector-level non-uniformity using deep convective clouds (DCCs) is also presented. These approaches are discussed, and then, correction results are compared with the diffuser-based method. Detector relative gain stability is assessed using the side-slither technique. Side-slither relative gains were found to correct streaking in test imagery with quality comparable to diffuser-based gains (within 0.005% for VNIR/PAN; 0.01% for SWIR) and identified a 0.5% temporal drift over a year. The DCC technique provided relative gains that visually decreased striping over the operational calibration in many images.

Abstract:Power line based communication systems (PLC) are emerging as strong contenders in the home and personal environments. Their advantages of high data rates, no additional wiring and easy scalability make them very attractive for deployment in local area networks and broadband power line (BPL) Internet access. Effective use of the powerline bandwidth requires robust physical and medium access control (MAC) protocols to mitigate the harsh conditions of the powerline channel, as well as the capability to support prioritized multimedia traffic. Home Plug 1.0 is the most commonly used technology for powerline communication. HomePlug 1.0 uses carrier sense multiple access/collision avoidance (CSMA/CA) to provide a maximum MAC data rate of 8 Mbps. Just as in 802.11, the throughput performance of this scheme becomes critical when the number of users increases, as in BPL. In this paper, a modification in the medium access control protocol of Home Plug 1.0 is proposed, to make it a constant contention window based scheme. This modification is shown to significantly enhance the MAC performance under saturation conditions. An analytical and simulation framework is used to tune the modified protocol for best performance, under the assumption that the number of active stations is known or can be reliably estimated

Abstract:Litopenaeus vannamei, with an annual production of 5–6 million tons and a value of USD 50–60 billion, is a cornerstone of global aquaculture. However, molting-related losses of 5–20% significantly impact this industry, and the physiological mechanisms of molting remain unclear. This study aims to elucidate the role of eclosion hormone (EH) in molting regulation and enhances the understanding of molting physiology in L. vannamei. This study investigated the role of (EH) in L. vannamei molting regulation. Two EH cDNAs, LvEH I and LvEH II, were identified, and their expression patterns across tissues and seven molting stages (A, B, C, D0, D1, D2, and D3) were analyzed. LvEH I was predominantly expressed in the gill, epidermis, and eyestalk, while LvEH II was mainly expressed in the eyestalk and brain. LvEH I was highly expressed in the eyestalk, epidermis, and gills at the D2 and D3 stages of molting, whereas LvEH II was highly expressed in both the D2 (brain) and D3 (eyestalk) stages. RNA interference (RNAi) targeting LvEH I revealed its critical role in molting, as silencing LvEH I disrupted the expression of molting-regulation genes, ETH, CCAP, CHH, EH II, CDA, and bursicon (Burs), significantly delaying the molting process. These findings highlight both LvEH I and LvEH II as indispensable for normal molting in L. vannamei and provide a foundation for developing effective molting management strategies to reduce industry losses.

Abstract:The snow crab, Chionoecetes opilio, is a giant deep-sea brachyuran. While several decapod crustaceans generally continue to molt and grow throughout their lifetime, the snow crab has a fixed number of molts. Adolescent males continue to molt proportionately to their previous size until the terminal molt at which time an allometric increase in chela size occurs and an alteration of behavioral activities occurs, ensuring breeding success. In this study, we investigated the circulating concentrations of methyl farnesoate (an innate juvenile hormone in decapods) (MF) before or after the terminal molt in males. We then conducted eyestalk RNAseq to obtain molecular insight into the regulation of physiological changes after the terminal molt. Our analyses revealed an increase in MF titers after the terminal molt. This MF surge may be caused by suppression of the genes that encode MF-degrading enzymes and mandibular organ-inhibiting hormone that negatively regulates MF biosynthesis. Moreover, our data suggests that behavioral changes after the terminal molt may be driven by the activation of biogenic amine-related pathways. These results are important not only for elucidating the physiological functions of MFs in decapod crustaceans, which are still largely unknown, but also for understanding the reproductive biology of the snow crab.

The role of ETH is well defined in controlling the molt process in insects, but it is yet to be defined in crustaceans. In this study, we investigated the behavioral response of intermolt crayfish to ETH and Corazonin injections as well as the impact of ETH on the molt period using in vivo assays. Injection of Corazonin and ETH resulted in a clear and immediate eye twitching response to these two neuropeptides. The Corazonin injection induced eye twitching in slow and asynchronous manner, while ETH injection caused eye twitching in a relatively fast and synchronous way. A single injection of ETH to crayfish resulted in a remarkable prolong molt period, at twice the normal molting cycle, suggesting that ETH plays a key role in controlling the molt cycle in decapod crustaceans. Given the key significance of ETH in molt regulation and its plausible application in pest control, we characterized ETH across the pancrustacean orders. Bioinformatic analysis shows the mature ETH sequence is identical in all studied decapod species. ETH can be classified into specific groups based on the associated motif in each insect order and shows an insect motif -KxxPRx to be conserved in crustaceans.
Integrative Proteomic and MicroRNA Analysis: Insights Into Mechanisms of Eyestalk Ablation-Induced Ovarian Maturation in the Swimming Crab Portunus trituberculatus

Abstract:Eyestalk ablation is the most common method to induce ovarian maturation in decapod crustacean aquaculture, but it jeopardizes broodstock survival and larvae production. It is important to understand the molecular basis underlying the maturation triggered by ablation and thereby develop an alternative measure for maturation manipulation. In this study, we investigate alterations of ovarian proteome and miRNA profile after ablation in a commercially important marine crab Portunus trituberculatus. Quantitative proteomic analysis using iTRAQ reveals that 163 proteins are differentially expressed following ablation, and modulation of methyl farnesoate metabolism and activation of calcium signaling may play important roles in the ovarian maturation induced by ablation. miRNA expression profiling identifies 31 miRNAs that show statistically significant changes. Integration of miRNA and proteome expression data with miRNA target prediction algorithms generates a potential regulatory network consisting of 26 miRNAs and 30 proteins linked by 71 possible functional associations. The miRNA-protein network analysis suggests that miRNAs are involved in promoting ovarian maturation by controlling expression of proteins related to methyl farnesoate synthesis, calcium signals, and energy metabolism. Experimental validation and temporal expression analysis indicate multiple miRNAs can act synergistically to regulate expression of Farnesoic acid O-methyltransferase and Calmodulin. Our findings provide new insights for elucidating the mechanisms underlying eyestalk ablation-induced ovarian maturation and could be useful for devising an alternative technique for manipulating reproduction in P. trituberculatus and other decapods.

Identification of candidate genes from androgenic gland in Macrobrachium nipponense regulated by eyestalk ablation

Abstract:Abstract The eyestalk of crustaceans, such as Macrobrachium nipponense , contains many neurosecretory hormones affecting the process of reproduction, molting, metabolism of glucose, and other functions. In this study, important metabolic pathways and candidate genes involved in male sexual development were selected from M. nipponense . The methodology involved performing long-read and next generation transcriptome sequencing of genes from the androgenic gland after eyestalk ablation. qPCR analysis revealed that the mRNA expression of Mn-IAG was significantly increased after ablation of both the single-side (SS) and double-side (DS) eyestalk, compared with the control group (CG). The long-read transcriptome generated 49,840 non-redundant transcripts. A total of 1319, 2092 and 4351 differentially expressed genes (DEGs) were identified between CG versus SS, SS versus DS and CG versus DS, respectively. These data indicated that ablation of the double-sided eyestalk played stronger regulatory roles than the single-side ablation on male sexual development in M. nipponense . This was consistent with the qPCR analysis. Cell Cycle, Cellular Senescence, Oxidative Phosphorylation, Glycolysis/Gluconeogenesis and Steroid Hormone Biosynthesis were the primary enriched metabolic pathways in all three comparisons, and the important genes from these metabolic pathways were also selected. qPCR permitted secondary confirmation of ten DEGs identified through RNA-seq. RNAi-mediated silencing analyses of Hydroxysteroid dehydrogenase like 1 ( HSDL1 ) revealed that HSDL1 has a positive regulatory effect on testes development. This study provides valuable insight into male sexual development in M . nipponense , including metabolic pathways and genes, paving the way for advanced studies on male sexual development in this species and in other crustaceans.

Abstract:Eriocheir sinensis is widely appreciated by the surrounding population due to its culinary delicacy and rich nutrients. The E. sinensis breeding industry is very prosperous and molting is one of the important growth characteristics. Research on the regulation of molting in E. sinensis is still in the initial stages. There is currently no relevant information on the regulatory mechanisms of heart development following molting. Comparative transcriptome analysis was used to study developmental regulation mechanisms in the heart of E. sinensis at the post-molt and inter-molt stages. The results indicated that many regulatory pathways and genes involved in regeneration, anti-oxidation, anti-aging and the immune response were significantly upregulated after molting in E. sinensis . Aside from cardiac development, the differentially expressed genes (DEGs) were relevant to myocardial movement and neuronal signal transduction. DEGs were also related to the regulation of glutathione homeostasis and biological rhythms in regard to anti-oxidation and anti-aging, and to the regulation of immune cell development and the immune response. This study provides a theoretical framework for understanding the regulation of molting in E. sinensis and in other economically important crustaceans.
