Abstract:Pollution by antibiotics and antibiotic resistance genes has become a major health concern. A microbial fuel cell coupled constructed wetland (CW-MFC) is a new installation to simultaneously treat wastewater and produce energy. This study was conducted to assess the removal efficiency of tetracycline (TC) and sulfamethoxazole (SMX) and the development of TC and SMX resistance genes (tetA, tetC, tetO, tetQ, tetW, sulI and sulII) in the CW-MFCs. At the same time, electricity was also produced during the co-metabolism of the antibiotics and glucose. The results indicated that the CW-MFCs could significantly reduce the concentration of TC and SMX in wastewater. Accumulation of antibiotics and the relative abundances of the tet and sul genes in different layers of the CW-MFCs were: anode layer > cathode layer > middle layer > effluent. The relative abundances of ARGs in the substrates ranged from 1.40 × 10−4 to 6.30 × 10−1 for tet genes and 5.58 × 10−3 to 9.27 × 10−2 for sul genes. The relative abundances of the tetC, tetQ, TetW, sulI and sulII genes were significantly affected by the antibiotics in effluent of the CW-MFCs. In addition, the highest power density obtained under the external resistance of 1000 Ω was 0.0578 W m−2. However, an obvious drop of power density was observed when the antibiotic concentrations increased from 400 to 1600 μg L−1.

Effects of Advanced Treatment Systems on the Removal of Antibiotic Resistance Genes in Wastewater Treatment Plants from Hangzhou, China.

Abstract:This study aimed at quantifying the concentration and removal of antibiotic resistance genes (ARGs) in three municipal wastewater treatment plants (WWTPs) employing different advanced treatment systems [biological aerated filter, constructed wetland, and ultraviolet (UV) disinfection]. The concentrations of tetM, tetO, tetQ, tetW, sulI, sulII, intI1, and 16S rDNA genes were examined in wastewater and biosolid samples. In municipal WWTPs, ARG reductions of 1-3 orders of magnitude were observed, and no difference was found among the three municipal WWTPs with different treatment processes (p > 0.05). In advanced treatment systems, 1-3 orders of magnitude of reductions in ARGs were observed in constructed wetlands, 0.6-1.2 orders of magnitude of reductions in ARGs were observed in the biological aerated filter, but no apparent decrease by UV disinfection was observed. A significant difference was found between constructed wetlands and biological filter (p < 0.05) and between constructed wetlands and UV disinfection (p < 0.05). In the constructed wetlands, significant correlations were observed in the removal of ARGs and 16S rDNA genes (R(2) = 0.391-0.866; p < 0.05). Constructed wetlands not only have the comparable ARG removal values with WWTP (p > 0.05) but also have the advantage in ARG relative abundance removal, and it should be given priority to be an advanced treatment system for further ARG attenuation from WWTP.
Fate of Sulfadiazine and Its Corresponding Resistance Genes in Up-Flow Microbial Fuel Cell Coupled Constructed Wetlands: Effects of Circuit Operation Mode and Hydraulic Retention Time

Abstract:Few studies have evaluated sulfadiazine (SDZ) removal and the potential risks of antibiotic resistance gene (ARG) accumulation in up-flow microbial fuel cell coupled constructed wetlands (UCW-MFCs). This study aims to investigate the removal of SDZ, accumulation of ARGs (sulI, sulII, intI), and bacterial community changes in four UCW-MFC reactors with two hydraulic retention times (HRTs) and two circuit operation modes. The results indicated that effluent from a closed circuit UCW-MFC had a lower SDZ concentration due to a higher electrode absorption capacity for SDZ and the dehydrogenase activity of the bacteria. Additionally, the ARG abundance in closed circuit system was higher than that in open circuit system. A lower HRT was correlated with a higher SDZ concentration accumulation on the electrode, which led to a higher ARG abundance. Also, an obvious increase in the ARG abundance was observed during treatment periods. The relative abundance of target ARGs in the anode was higher than in the cathode and the bottom. A cluster analysis of bacterial communities at the phyla level showed they were more significantly affected by HRT. Furthermore, a closed circuit system and a high HRT contributed to high microbial diversity. In addition, five phyla of bacteria might be potential hosts for ARGs. Enrichment of ARG potential hosts induced by SDZ may be responsible for ARG abundance increase. Electricity production characteristics of the UCW-MFCs were also affected by HRT.
Occurrence and Removal of Antibiotic Resistance Genes in Municipal Wastewater and Rural Domestic Sewage Treatment Systems in Eastern China.

Abstract:Antibiotic resistance genes (ARGs) are emerging environmental contaminants and pose a threat to public health. In this study, four tetracycline resistance genes (tetM, tetO, tetQ and tetW) and two sulfonamide resistance genes (sulI and sulII) were evaluated in 4 municipal wastewater and 8 rural domestic sewage treatment systems with different wastewater handling abilities and treatment processes using quantitative polymerase chain reaction (qPCR). In the influents, the relative abundance of different ARGs showed significant variations among the sampling sites. In addition, significant correlations (tetQ: R(2)=0.712, P<0.05; tetO: R(2)=0.394, P<0.05) between the gene copy numbers and wastewater-receiving capacity were observed. Statistical analysis revealed a positive correlation (R(2)=0.756, P<0.05) between the gene copy numbers of sulI and intI1, whereas the gene numbers of tetM and sulI were strongly correlated with 16S rDNA. Significant reductions (1-3 orders of magnitude) in ARGs were observed in municipal wastewater treatment systems, but a smaller reduction was found in the rural domestic sewage treatment systems. These results provide insights into the occurrence and removal of ARGs in wastewater treatment systems in both rural and urban areas in eastern China.

Abstract:Wastewater treatment plants (WWTPs) contain diverse antibiotic resistance genes (ARGs), and thus are considered as a major pathway for the dissemination of these genes into the environments. However, comprehensive evaluations of ARGs dynamic during wastewater treatment process lack extensive investigations on a broad spectrum of ARGs. Here, we investigated the dynamics of ARGs and bacterial community structures in 114 samples from eleven Chinese WWTPs using high-throughput quantitative PCR and 16S rRNA-based Illumina sequencing analysis. Significant shift of ARGs profiles was observed and wastewater treatment process could significantly reduce the abundance and diversity of ARGs, with the removal of ARGs concentration by 1-2 orders of magnitude. Whereas, a considerable number of ARGs were detected and enriched in effluents compared with influents. In particular, seven ARGs mainly conferring resistance to beta-lactams and aminoglycosides and three mobile genetic elements persisted in all WWTPs samples after wastewater treatment. ARGs profiles varied with wastewater treatment processes, seasons and regions. This study tracked the footprint of ARGs during wastewater treatment process, which would support the assessment on the spread of ARGs from WWTPs and provide data for identifying management options to improve ARG mitigation in WWTPs.

Abstract:In coastal eco-industrial zones, wastewater treatment plants (WWTPs) and constructed wetlands (CWs) can alleviate the challenge of water shortage and the negative effect of sewage discharge, while the problems of antibiotic resistance genes (ARGs) have not attracted enough attention. In this research, the Wafergen SmartChip system was adopted to investigate the ARG profiles in a coupled system combined WWTPs and CWs in a coastal industrial park. Potential risks of antibiotic resistance in chemical industrial wastewater were confirmed due to the higher abundance of target ARGs (> 107 copies/mL). General decline with partial enrichment in absolute and relative abundance of ARGs from the WWTPs to CWs revealed the effective removal of ARGs in the coupled system, while the fate of different ARG types varied greatly. Aminoglycoside and sulfonamide ARGs were detected with higher abundance (up to 5.34 ×107 and 3.61 ×107 copies/mL), especially aac(6')-Ib and sul1. Denitrification, secondary sedimentation, and acid hydrolysis contributed to the removal of aminoglycoside, sulfonamide, β-lactamase, chloramphenicol, and multidrug ARGs. Catalytic ozonation contributed to the removal of tetracycline and MLSB ARGs. Subsurface CWs worked effectively for the removal of sulfonamide, tetracycline, and multidrug ARGs, especially tetX, cphA, tetG, and strB. Close correlations between ARGs and MGEs emphasized the vital roles of anthropogenic pollutants and horizontal gene transfer on the diffusion of ARGs. Actinobacteria, Bacteroidota, and Cyanobacteria were dominant in the CWs, while Proteobacteria, Firmicutes, and Planctomycetota were prevalent in the WWTPs. Redundancy analysis and variance partitioning analysis indicated that transposase and water quality posed greater influences on the distribution of ARGs. Co-occurrence network revealed that potential multiple antibiotic resistant pathogenic bacteria decreased in the CWs. The coupled system has a limited effect on the reduction of ARGs and potential ARG hosts, providing a comprehensive insight into the fate of ARGs in conventional water-processing systems.

Abstract:For revealing the characteristics of antibiotic resistance genes (ARGs) in wastewater treatment systems, real-time PCR was adopted to investigate the variation of abundances of cell-associated ARGs and cell-free ARGs, in a municipal wastewater treatment system (M for short) and a coking wastewater treatment system (C for short). In system M, the absolute abundances of the cell-associated ARGs, sul Ⅱ,tetC,blaPSE-1, and ermB, were much higher than those of the cell-free fractions in the influent. The biological treatment process did not enrich antibiotic resistance bacteria (ARBs) and membrane filtration of the MBR effectively reduced both cell-associated and cell-free DNA in water. The total ARGs removal was 2.54-4.95 logs. In system C, the biological treatment process enriched the sul Ⅱ -carried ARBs; however, the relative and absolute abundances of cell-free sul Ⅱ were decreased. The succeeding process, coagulation-sand filtration, decreased the absolute abundance of cell-associated sul Ⅱ, but increased the absolute abundance of cell-free sul Ⅱ in water. The proportion of cell-free sul Ⅱ in total sul Ⅱ gene increased from 0.05% in the biological treatment effluent to 1.33% in the sand filtration effluent and further increased to 9.31% after the effluent was kept at 25℃ and at dark for five days. The ratio of cell-free ARGs to total ARGs increased with deep removal of ARBs and lysis of residual cells. The risk of ARG proliferation by cell-free DNA in the effluent needs further evaluation.

Abstract:The intensive use of antibiotics results in their continuous release into the environment and the subsequent widespread dissemination of antibiotic resistance genes (ARGs), thus posing potential risks for public health. Although vertical up-flow constructed wetlands (VUF-CWs) have been widely used to treat wastewater in remote or rural regions, few studies have assessed the potential risks of ARG dissemination when VUF-CWs are applied to treat wastewaters containing antibiotics. In this study, the removal performance of two typical antibiotics (sulfamethoxazole (SMX) and tetracycline (TC)) and the fate of ARGs were evaluated in three lab-scale VUF-CWs. The results indicated that high removal efficiencies (>98%) could be achieved for both SMX and TC. However, the exposure of antibiotics resulted in harboring abundant ARGs (mainly sul- and tet-related genes), even with increasing abundances with operation time. The abundances of ARGs had a positive correlation with the accumulation of SMX and TC in different layers of VUF-CWs, where the tet and sul genes have the highest abundance in the bottom layer due to the highest antibiotic exposure concentration. Positive correlations were observed between the abundance of tet gene and antibiotic concentration in effluent. Although the effluent had lower abundances of the ARGs than that in the wetland media, the occurrence of ARGs in effluent might still pose risk for public health. Further studies are required to explore effective control strategies to eliminate ARGs from VUF-CWs.

Abstract:The effects of the continuous accumulation of Zinc (Zn) on the fate of antibiotic resistance genes (ARGs) in constructed wetland-microbial fuel cells (CW-MFCs) remain unclear. In this study, the impacts of Zn addition and a circuit mode on antibiotic removal, occurrence of ARGs, the bacterial community, and bacterial functions were investigated in three groups of CW-MFCs. The results showed that continuous Zn exposure enriched the target ARGs during the initial stage, while excessive Zn accumulation decreased antibiotic removal and the abundance of ARGs. A principal component analysis demonstrated that ARGs and the bacterial community distribution characteristics were significantly impacted by the mass accumulation of antibiotics and Zn, as well as the circuit mode. A redundancy analysis, partial least squares path modeling, and Procrustes analysis revealed that the accumulation of antibiotics and Zn, the composition of the bacterial community, the circuit mode, and the abundance of intI associated with horizontal gene transfer jointly contributed to the distributions of ARGs in the electrodes and effluent. Moreover, continuous exposure to Zn decreased the bacterial diversity and changed the composition and function of the bacterial community predicted using PICRUSt tool. The co-occurrence of ARGs, their potential hosts and bacterial functions were further revealed using a network analysis. A variation partition analysis also showed that the accumulation of target pollutants and the circuit mode had a significant impact on the bacterial community composition and functions. Therefore, the interaction among ARGs, the bacterial community, bacterial functions, and pollutant accumulations in the CW-MFC was complex. This study provides useful implications for the application of CW-MFCs for the treatment of wastewater contaminated with antibiotics and heavy metals.

Abstract:The fate and removal efficiency of antibiotic resistance genes (ARGs) and mobile genetic elements (MGEs) in livestock wastewater by microbial fuel cell (MFC) was evaluated by High-throughput quantitative PCR. The results showed that 137 ARGs and 9 MGEs were detected in untreated livestock wastewater. The ARG number of macrolide-lincosamide-streptogramin group B (MLSB), tetracycline and sulfonamide were relatively higher. Throughout the treatment process, the number and abundance of ARGs and MGEs significantly decreased. The relative abundance of tetracycline, sulfonamide and chloramphenicol resistance genes showed the most obvious decreasing trend, and the relative abundance of MGEs decreased by 75% (from 0.012 copies/16S rRNA copies to 0.003 copies/16S rRNA copies). However, the absolute abundance of beta-lactamase resistance genes slightly increased. The operation process of MFC produces selective pressure on microorganisms, and Actinobacteria were predominant and had the ability to decompose antibiotics. The COD removal rate and TN removal rate of livestock wastewater were 67.81% and 62.09%, and the maximum power density and coulomb efficiency (CE) reached 11.49% and 38.40% respectively. This study demonstrated that although the removal of COD and TN by MFC was limited, MFC was quite effective in reducing the risk of antibiotic toxicity and horizontal gene transfer.

Prevalence and Proliferation of Antibiotic Resistance Genes in Two Municipal Wastewater Treatment Plants.

Abstract:The propagation of antibiotic resistance genes (ARGs) is an emerging health concern worldwide. Thus, it is important to understand and mitigate their occurrence in different systems. In this study, 30 ARGs that confer resistance to tetracyclines, sulfonamides, quinolones or macrolides were detected in two activated sludge wastewater treatment plants (WWTPs) in northern China. Bacteria harboring ARGs persisted through all treatment units, and survived disinfection by chlorination in greater percentages than total Bacteria (assessed by 16S rRNA genes). Although the absolute abundances of ARGs were reduced from the raw influent to the effluent by 89.0%-99.8%, considerable ARG levels [(1.0 ± 0.2) × 10(3) to (9.5 ± 1.8) × 10(5) copies/mL)] were found in WWTP effluent samples. ARGs were concentrated in the waste sludge (through settling of bacteria and sludge dewatering) at (1.5 ± 2.3) × 10(9) to (2.2 ± 2.8) × 10(11) copies/g dry weight. Twelve ARGs (tetA, tetB, tetE, tetG, tetH, tetS, tetT, tetX, sul1, sul2, qnrB, ermC) were discharged through the dewatered sludge and plant effluent at higher rates than influent values, indicating overall proliferation of resistant bacteria. Significant antibiotic concentrations (2%-50% of raw influent concentrations) remained throughout all treatment units. This apparently contributed selective pressure for ARG replication since the relative abundance of resistant bacteria (assessed by ARG/16S rRNA gene ratios) was significantly correlated to the corresponding effluent antibiotic concentrations. Similarly, the concentrations of various heavy metals (which induce a similar bacterial resistance mechanism as antibiotics - efflux pumps) were also correlated to the enrichment of some ARGs. Thus, curtailing the release of antibiotics and heavy metals to sewage systems (or enhancing their removal in pre-treatment units) may alleviate their selective pressure and mitigate ARG proliferation in WWTPs.

Abstract:The variation of five antibiotic resistance genes (ARGs)—tetG, tetW, tetX, sul1, and intI1—in a full-scale municipal wastewater treatment plant with A2O-MBR system was studied. The concentrations of five resistance genes both in influent and in membrane bioreactor (MBR) effluent decreased as sul1 > intI1 > tetX > tetG > tetW, and an abundance of sul1 was statistically higher than three other tetracycline resistance genes (tetG, tetW, and tetX) (p < 0.05). The concentrations of five ARGs in the influent were all higher in spring (median 105.81–107.32 copies mL−1) than they were in other seasons, and tetW, tetX, and sul1 reached its lowest concentration in autumn (104.61–106.81 copies mL−1). The concentration of ARGs in wastewater decreased in the anaerobic effluent and anoxic effluent, but increased in the aerobic effluent, and then sharply declined in the MBR effluent. The reduction of tetW, intI1, and sul1 was all significantly positively correlated with the reduction of 16S ribosomal DNA (rDNA) in the wastewater treatment process (p < 0.01). The concentration of ARGs (copies mg−1) in sludge samples increased along the treatment process, but the abundance of five ARGs (ratio of ARGs to 16S rDNA) remained the same from anaerobic to anoxic to aerobic basins, while an increment ratio in MBR was observed for all ARGs.

Abstract:The nucleotide sequence of a 1980-base-pair segment of DNA, containing the lpd gene encoding the lipoamide dehydrogenase component (E3) of the pyruvate dehydrogenase complex of Escherichia coli K12, has been determined by the dideoxy chain-termination method. The lpd structural gene comprises 1419 base pairs (473 codons, excluding the initiating AUG codon). It is preceded by a good promoter and an excellent ribosome binding site and it ends with a typical rho-independent terminator sequence. The results confirm that the lpd gene is an independent gene linked to, but not part of, the ace operon that encodes the E1 and E2 components of the pyruvate dehydrogenase complex. The location and transcriptional polarity of the lpd gene relative to the restriction map of the corresponding region of DNA, are completely consistent with previous genetic and post-infection labelling studies. The composition, Mr (50554 or 51274 if the FAD cofactor is included), amino-terminal sequence and carboxy-terminal sequence predicted from the nucleotide sequence are in excellent agreement with previous studies on the purified enzyme. The enzyme also exhibits a remarkable degree of sequence homology with peptides of the pig heart enzyme and with other pyridine nucleotide disulphide oxidoreductases whose sequences have been defined: human erythrocyte glutathione reductase and plasmid-encoded mercuric reductase.

Abstract:Constructed wetland (CW) is considered a promising technology for the removal of emerging contaminants. However, its removal performance for antibiotic resistance genes (ARGs) is not efficient and influence of virulence factor genes (VFGs) have not been elucidated. Here, removal of intracellular and extracellular ARGs as well as VFGs by electricity-intensified CWs was comprehensively evaluated. The two electrolysis-intensified CWs can improve the removal of intracellular ARGs and MGEs to 0.96- and 0.85-logs, respectively. But cell-free extracellular ARGs (CF-eARGs) were significantly enriched with 1.8-logs in the electrolysis-intensified CW. Interestingly, adding Fe-C microelectrolysis to the electrolysis-intensified CW is conducive to the reduction of CF-eARGs. However, the detected number and relative abundances of intracellular and extracellular VFGs were increased in all of the three CWs. The biofilms attached onto the substrates and rhizosphere are also hotspots of both intracellular and particle-associated extracellular ARGs and VFGs. Structural equation models and correlation analysis indicated that ARGs and VFGs were significantly cooccurred, suggesting that VFGs may affect the dynamics of ARGs. The phenotypes of VFGs, such as biofilm, may act as protective matrix for ARGs, hindering the removal of resistance genes. Our results provide novel insights into the ecological remediation technologies to enhance the removal of ARGs.

Abstract:Wastewater treatment plants (WWTPs) have been regarded as important point-sources of antibiotic resistance genes (ARGs) in aquatic environments. To investigate the distribution and removal of ARGs in WWTPs, a pharmaceutical wastewater treatment plant (PWWTP) and an integrated wastewater treatment plant (IWWTP) in a fine-chemical industrial park were chosen, and polymerase chain reaction (PCR) and real-time PCR techniques were used to determine the occurrence and abundances of ARGs along the treatment processes. Ten and fifteen ARGs were detected initially in the influents of PWWTP and IWWTP respectively, in which tetracycline and sulfonamide resistance genes were frequently reported, while dfrA13 was first reported in WWTPs. The most abundant ARGs in the influents were sul Ⅰ and sul Ⅱ, followed by dfrA13, tetQ, floR, tetO, and tetW. The total ARGs increased by 0.21 log after the treatment by PWWTP, whose effluent contributed 0.87% to the inflow yet 5.05% to the total ARGs of IWWTP. Finally the total ARGs removed by IWWTP was 1.03 log, with the remaining ARGs then transported within the final effluent to the nearby coastal area. The authors concluded that the environmental and other impacts from the spread of ARGs on the microbial communities of the coastal environment needed further study.

Abstract:This study comprehensively analyzed the fates of antibiotics, antibiotic resistance genes (ARGs) and microbial community during upgrading process of municipal wastewater treatment plant (MWTP) of Anaerobic/Anoxic/Oxic-membrane bioreactor (A2/O-MBR) from regulation to stable operation phase. The factors most affecting ARGs profile were investigated, and the potential hosts for ARGs were further determined. Results showed that the removal rate of total antibiotics and the reduction of ARGs increased by 10.49% and 1.37 logs in A2/O-MBR during upgrading process, respectively. The most prevalent ARG was sul1 in both water and sludge phase. Mobile genetic elements (MGEs) and microbial community were the most significant factors affecting ARGs distribution in water phase and sludge, respectively. Notably, the abundance of potential antibiotic resistance bacteria (ARB) was significantly decreased during upgrading process. Therefore, sul1and intI1 proliferation as well as microbial community succession should be emphasized in ARGs pollution management and control. This study expected to provide a theoretical basis for the application of MBR process to remove antibiotics and ARGs in upgrading of MWTPs.

Abstract:Accumulation and transmission of antibiotic resistance genes (ARGs) in greywater treatment systems present risks for its reuse. In this study, a gravity flow self-supplying oxygen (O2) bio-enhanced granular activated carbon dynamic biofilm reactor (BhGAC-DBfR) was developed to treat greywater. Maximum removal efficiencies were achieved at saturated/unsaturated ratios (RSt/Ust) of 1:1.1 for chemical oxygen demand (97.6 ± 1.5%), linear alkylbenzene sulfonates (LAS) (99.2 ± 0.5%), NH4+-N (99.3 ± 0.7%) and total nitrogen (85.3 ± 3.2%). Microbial communities were significantly different at various RSt/Ust and reactor positions (P < 0.05). The unsaturated zone with low RSt/Ust showed more abundant microorganisms than the saturated zone with high RSt/Ust. The reactor-top community was predominant by aerobic nitrification (Nitrospira) and LAS biodegradation (Pseudomonas, Rhodobacter and Hydrogenophaga) related genera; but reactor-bottom community was predominant by anaerobic denitrification and organics removal related genera (Dechloromonas and Desulfovibrio). Most of the ARGs (e.g., intI-1, sul1, sul2 and korB) were accumulated in the biofilm, which were closely associated with microbial communities at reactor top and stratification. The saturated zone can achieve over 80% removal of the tested ARGs at all operation Phases. Results suggested that BhGAC-DBfR can provide assistance in blocking the environment dissemination of ARGs during greywater treatment.

Abstract:Municipal sewage treatment plants (MSTPs) are environmental pools for antibiotic resistant bacteria (ARB) and antibiotic resistance genes (ARGs), which is cause for growing environmental-health concerns. In this study, the effects of different wastewater treatment processes on microbial antibiotic resistance in four MSTPs were investigated. PCR, q-PCR, and molecular cloning integrally indicated that the tetracycline resistance (tet) genes significantly reduced after activated-sludge treatment. Illumina high-throughput sequencing revealed that the broad-spectrum profile of ARGs and mobile element genes (MGEs) were also greatly decreased by one order of magnitude via activated sludge treatment and were closely associated with each other. Correlations between ARGs and bacterial communities showed that potential ARB, such as Acinetobacter, Bacteroides, and Cloaibacterium, were removed by the activated-sludge process. Sedimentation processes cannot significantly affect the bacterial structure, resulting in the relative abundance of ARGs, MGEs, and ARB in second-clarifier effluent water being similar to activated sludge. A comprehensive study of ARGs associated with MGEs and bacterial structure might be technologically guided for activated sludge design and operation in the MSTPs, to purposefully control ARGs carried by pathogenic hosts and mobility.
