Abstract:A Lactobacillus brevis CGMCC 1306 isolated from fresh milk without pasteurization was found to have higher glutamate decarboxylase (GAD) activity. An effective isolation and purification procedure of GAD from a cell-free extract of Lactobacillus brevis was developed, and the procedure included four steps: 30%—90% saturation (NH4)2SO4 fractional precipitation, Q sepharose FF anion-exchange chromatography, sephacryl S-200 gel filtration, and resource Q anion-exchange chromatography. Using this protocol, the purified GAD was demonstrated to possess electrophoretic homogeneity via SDS-PAGE. The purification fold and activity recovery of GAD were 43.78 and 16.95%, respectively. The molecular weight of the purified GAD was estimated to be approximately 62 kDa via SDS-PAGE. The optimum pH and temperature of the purified GAD were 4.4 and 37°C, respectively. The purified GAD had a half-life of 50min at 45°C and the Km value of the enzyme from Lineweaver-Burk plot was found to be 8.22. 5′-pyridoxal phosphate (PLP) had little effect on the regulation of its activity.
Overexpression, purification and characterization of phospholipase C from Acinetobacter calcoaceticus

Abstract:In this study, we constructed two recombinant Escherichia coli strains to produce phospholipase C (PLC) from Acinetobacter calcoaceticus. The recombinant enzymes were purified to homogeneity and characterized. [Methods] We cloned the PLC encoding gene plc1, plc2 from genome DNA of A. calcoaceticus ATCC17902. The amplified fragments were inserted into pET28a(+ to obtain expression plasmids. E. coli BL21 (DE3) harboring the above plasmids were cultivated and induced with isopropyl-beta-D-thiogalactopyranoside to express PLCs. The recombinant PLCs were purified by affinity chromatography and their catalytic properties were characterized.Two PLCs from A. calcoaceticus were cloned and functional expressed in E. coli. The recombinant enzymes have activities of 31,160 +/- 418 U/mg for PLC1 and 13640 +/- 354 U/mg for PLC2, when using p-nitrophenyl phosphorycholine as substrate. The purified PLC1 and PLC2 exhibited optimum temperature at 65 degrees C and 50 degrees C, respectively. Their optimal pH were 8 and 7.5, respectively. PLC2 was stable under 40 degrees C and pH at 8, whereas the residual activity of PLC1 was less than 25% in the same condition. Mg2+ and Ca2+ stimulated two enzymes activity, whereas Zn2. stimulated PLC1 and inhibited PLC2. PLC1 and PLC2 hydrolyzed phosphatidylinositol.It is the first time to express and characterize the PLC gene from A. calcoaceticus ATCC17902. These research results provide reference for the study of food-safety microbiological PLC.
Remote sensing methods for surveying and extracting abandoned farmlands

Abstract:The increase of agricultural land abandonment is one of the main changes in land use in China over the recent years. In China, the abandonment phenomenon has become one of the most serious problems in the last 20 years. Abandoned farmlands make the land in a vicious circle. Preventing the farmland from abandonment is an urgent issue for China. Thus, abandoned farmland survey is very important; it can provide scientific evidence for food security, land use planning and economic development of China. The conventional investigation approach of abandonment status is field survey. It cannot meet the demand of large area because of the lack of efficiency and timeliness. Remote sensing technology has advantages of large spatial observing area, high real-time, wide spectrum ranges and all weather imaging capability. It has worked successfully in many fields, but was rarely applied in terms of investigation and assessment of arable land abandonment. In this work, Xinyang City in Henan Province was selected as a study area. SPOT-5 images and MODIS NDVI data were used for surveying and extracting the abandoned farmland. This paper introduces the theories, processes and results of three methods for abstracting abandoned farmland on the basic of remote sensing, assesses the efficiency and accuracy of them, and compares the merits and demerits of them, sums up the workable technique for the study area.
Purification and characterization of an extracellular lipase from Geotrichum marinum

Abstract:An extracellular lipase (EC 3.1.1.3) from Geotrichum marinum was purified 76-fold with 46% recovery using Octyl Sepharose 4 Fast Flow and Bio-Gel A 1.5 m chromatography. The purified enzyme showed a prominent band on SDS-PAGE and a single band on native PAGE based on the activity staining. The molecular mass of the lipase was estimated to be 62 kDa using SDS-PAGE and Bio-Gel A chromatography, indicating that the lipase likely functions as a monomer. The pl of the lipase was determined to be 4.54. The apparent V(max) and Km were 1000 micromol/min/mg protein and 11.5 mM, respectively, using olive oil emulsified with taurocholic acid as substrate. The lipase demonstrated a pH optimum at pH 8.0 and a temperature optimum at 40 degrees C. At 6 mM, Na+, K+, Ca2+, and Mg2+ stimulated activity, but Na+ and K+ at 500 mM and Fe2+ and Mn2+ at 6 mM reduced lipase activity. The anionic surfactant, taurocholic acid, and the zwitterionic surfactant, 3-[(3-cholamidopropyl)dimethylammonio]-1-propanesulfonate, enhanced the activity at 0.1 mM. Other anionic surfactants such as SDS and sodium dioctyl sulfosuccinate, the cationic surfactants methylbenzethonium bromide and cetyltriethylammonium bromide, and the nonionic surfactants Tween-20 and Triton X-100 inhibited the lipase activity to different extents. The lipase was found to have a preference for TG containing cis double bonds in their FA side chains, and the reaction rate increased with an increasing number of double bonds in the side chain. The lipase had a preference for ester bonds at the sn-1 and sn-3 positions over the ester bond at the sn-2 position.
Cloning, Expression, and Characterization of a Wide-pH-Range Stable Phosphite Dehydrogenase from Pseudomonas sp. K in Escherichia coli

Abstract:A phosphite dehydrogenase gene ( ptdh K) consisting of 1,011-bp nucleotides which encoding a peptide of 336 amino acid residues was cloned from Pseudomonas sp. K. gene ptdh K was expressed in Escherichia coli BL21 (DE3) and the corresponding recombinant enzyme was purified by metal affinity chromatography. The recombinant protein is a homodimer with a monomeric molecular mass of 37.2 kDa. The specific activity of PTDH-K was 3.49 U mg −1 at 25 °C. The recombinant PTDH-K exhibited maximum activity at pH 3.0 and at 40 °C and displayed high stability within a wide range of pHs (5.0 to 10.5). PTDH-K had a high affinity to its natural substrates, with K m values for sodium phosphite and NAD of 0.475 ± 0.073 and 0.022 ± 0.007 mM, respectively. The activity of PTDH-K was enhanced by Na + , NH 4 + , Mg 2+ , Fe 2+ , Fe 3+ , Co 2+ , and EDTA, and PTDH-K exhibited different tolerance to various organic solvents.
Purification and characterization of a phospholipid-hydrolyzing phosphoesterase produced by Pediococcus acidilactici isolated from Gouda cheese

Abstract:Lipolysis occurs during ripening of dairy products as a result of esterase or lipase activity. Lactic acid bacteria (LAB) are considered to be weakly lipolytic bacteria compared with other species. In cheeses with extended ripening periods, lipolytic LAB may have several advantages. Pediococcus acidilactici is a LAB frequently found in fermented dairy products, but no previous reports exist on their production of esterases or lipases. Our interest in the relationship of LAB and enzymatic characterization is due to the multiple reports of the benefits of LAB in the gut microbiome, particularly at the intestinal membrane. Pediococci have been characterized as probiotic and especially active in membrane interactions. The aim of this project was to purify, characterize, and identify the phosphoesterase produced by P. acidilactici originally isolated from Gouda cheese and determine its phospholipid (PL) hydrolysis profile, with a focus on increased absorption of these compounds in the human gut. Native zymograms were performed to identify a protein with lipolytic activity in the intracellular fraction of P. acidilactici. The enzyme was purified via size-exclusion HPLC, concentrated via ultrafiltration, and identified using sequence analysis in liquid chromatography (LC)-MS/MS. The purified fraction was subjected to biochemical characterization as a function of pH, temperature, ion concentration, hydrolysis of different substrates, and PL. A single protein with a molecular weight of 86 kDa and esterase activity was detected by zymography. Analysis of the LC-MS/MS results identified a putative metallophosphoesterase with a calculated molecular weight of 45.5 kDa, suggesting that this protein is active as a homodimer. The pure protein showed an optimal activity between pH 8.0 to 9.0. The optimal temperature for activity was 37°C, and the enzyme lost 15% of activity after incubation at 90°C for 1 h. This enzyme showed activity on short-chain fatty acids and exhibited high hydrolysis of phosphatidylinositol. It also hydrolyzed phosphatidylserine, phosphatidylcholine, and sphingomyelin. Phosphatidylethanolamine was hydrolyzed but with less efficiency. The characteristics and lipolytic actions exerted by this protein obtained from LAB hold promise for a potential strain of esterase or lipase that may exert human health benefits through increased digestibility and absorption of nutrients found in dairy products.
Cloning and Heterologous Expression in E.coli of Phospholipase C Gene from Bacillus Cereus

Abstract:As phospholipase C(PLC) could hydrolyze egg yolk and produce a zone of opalescence surrounding colonies on an egg yolk agar plate,one phospholipase C producing strain of Bacillus cereus 12 was isolated from 600 strains preserved in our laoratory.Phosphatidylcholine-preferring phospholipase C(PC-PLC) gene pcplc1 was amplified from the isolated strain.Recombinant plasmid pET28a(+)-pcplc1 was constructed and transformed into E.coli BL21(DE3).After subsequent indution by isopropyl β-D-1-thiogalactopyranoside(IPTG),an approximately 33 kD protein was detected in the cell lysate supernatant by SDS-PAGE.Meanwhile,the recombinant protein showed significant PLC activity on egg yolk agar plate.Induction time,induction temperature and IPTG concentration were investigated and optimized induction conditions for pcplc1 expression were obtained as follows: 4% of inoculum amount,1.5 h of initial culture,4 h of induced culture in the presence of 0.2 mmol/L of IPTG,and 25 ℃ of culture temperature.Unde the optimized conditions,maximum PLC activity in the supernatant was observed to be(30.24 ± 0.18) U/mL(crude enzyme activity).
Quantitative Analysis on Inactivation and Reactivation of Recombinant Glycerol Dehydratase from Klebsiella Pneumoniae Xjpd-Li

Abstract:The genes encoding glycerol dehydratase were cloned and characterized by genomic DNA from Klebsiella pneumoniae XJPD-Li, and the assigned accession number EF634063 was available from the GenBank database. The DNA sequence analysis showed that the clone included three ORFs (dhaB, dhaC and dhaE, encoding α, β and γ subunit of glycerol dehydratase, respectively). Among three subunits of glycerol dehydratase, amino acid residues H13, S193, N359, E407, and M515 of α subunit, N47, L150, V189 of β subunit are different with what had been reported. Subsequently, the expression vector was constructed and transformed into E. coli BL21, and the colony carried genes of glycerol dehydratase were selected. SDS-PAGE examination showed that the three subunits were well expressed. The specific activity of recombined glycerol dehydratase reached to 0.299Umg−1, which was about 3 times comparing with that of the wild strain. The research also displayed that both glycerol and O2 could inactive the glycerol dehydratase expressed in E. coli quickly in 10min. The inactivated glycerol dehydratase could be effectively reactivated under the system as follows: the concentration of ATP, Mg2+ and coenzyme B12 were 50mM, 10mM and 3μM, respectively, when the ratio (W/W) of glycerol dehydratase to reactivation factor was 4:1. The O2-inactivated and glycerol-inactivated dehydratase could be reactivated to 97.3% and 98.9% of initial activity in 10min in above-mentioned conditions, respectively. The reactivation factor together with ATP was considered as the “ON/OFF” reactivating condition.
Purification and Characterization of a Xyloglucanase from Paenibacillus xylanilyticus

Abstract:Xyloglucanase had been extracted from the culture medium fermented by Paenibacillus xylanilyticus.Isolated and purified by methods of ammonium sulfate precipitation,HiPrep26/10 Desalting gel layer chromatography,HiPrepDEAE FF16/10 anion exchange column,HiPrep 16/60 Sephacryl S-100 gel layer chromatography and HiPrep 16/10 Source 30S cation exchange column.The molecular weight of xyloglucanase was about 39 kD,identified by SDS-PAGE.The optimal reaction temperature of xyloglucanase was 50℃,and there was good stabi-lity under 60℃.The optimal reaction pH of xyloglucanase was 7.0,and there was good stability between pH5.0 and pH10.0.The dynamic study demonstrated that the Km value of xyloglucanase was 65 g/L,the maximum reaction velocity(Vmax) was 6.49 μmol/min,and the kcat was 10.86 s-1.The substrate specificity of the xyloglucanase indicated that the xyloglucan had much higher specific activity than other substrates.The purified enzyme was analyzed by mass spectrometry,and the result showed that the purified enzyme had high homology with xyloglucanase from Paenibacillus pabuli.It was the first report about xyloglucanase product from Paenibacillus xylanilyticus.
The conformational analysis of substance P analogs using high-field NMR techniques.

Abstract:High-field nuclear magnetic resonance measurements were carried out on substance P fragments SP4-11' [pGlu5]-SP5-11 and [pGlu6]SP6-11 both at 400 and at 500 MHz. A spectral simulation was carried out on two of these peptides and the coupling constants were interpreted in terms of the conformations. The JNH-CHa coupling constants are all approximately 8 Hz, with the exception of glycine, indicating no preferred conformation for the backbone. For the amino acids other than p-Glu, a comparison of the coupling constant data suggests the same relative rotamer populations for the side chains. Proton longitudinal relaxation time data were measured for all three peptides and support the above conclusions.
Cloning and Expression in E. Coli of an Organic Solvent-Tolerant and Alkali-Resistant Glucose 1-Dehydrogenase from Lysinibacillus Sphaericus G10

Abstract:The gene gdh encoding an organic solvent-tolerant and alkaline-resistant NAD(P)-dependent glucose 1-dehydrogenase (LsGDH) was cloned from Lysinibacillus sphaericus G10 and expressed in Escherichia coli. The recombinant LsGDH exhibited maximum activity at pH 9.5 and 50°C. LsGDH displayed high stability at a wide pH ranging from 6.5 to 10.0 and was stable after incubation at 30°C for 1week in 25mM sodium phosphate buffer (pH 6.5) in the absence or presence of NaCl. The activity of LsGDH was enhanced by Li+, Na+, K+, NH4+, Mg2+, and EDTA at pH 8.0. LsGDH exhibited high tolerance to 60% DMSO, 30% acetone, 30% methanol, 30% ethanol, 10% n-propanol, 30% isopropanol, 60% n-hexanol and 30% n-hexane. The relationship between stability and chain length of the alcohols fit a Gaussian distribution model (R2⩾0.94), and demonstrated lowest enzyme stability in C4-alcohol. The results suggested that LsGDH was potentially useful for coenzyme regeneration in organic solvents or under alkaline conditions.

Abstract:Reductive photodehalogenation of chloroaromatics is assumed to proceed from the triplet excited state, although its energy is often insufficient to promote a clean homolytic C-Cl cleavage. A clear-cut experimental proof is provided that correlates self-quenching of the directly observed triplet excited states of chlorocarbazole-based dyads 1 and 2 with photoreactivity via intramolecular charge transfer.

Abstract:The enzyme α-Galactosidase (α-D-galactoside galactohydrolase [EC 3.2.1.22]) is an exoglycosidase that hydrolyzes the terminal α-galactosyl moieties of glycolipids and glycoproteins. It is ubiquitous in nature and possesses extensive applications in the food, pharma, and biotechnology industries. The present study aimed to purify α-galactosidase from Klebsiella pneumoniae , a bacterium isolated from the human oral cavity. The purification steps involved ammonium sulfate precipitation (70 %), dialysis, ion exchange chromatography using a DEAE-cellulose column, and affinity monolith chromatography. The sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) analysis was used to determine the molecular weight of the purified enzyme. The kinetic constants, Michaelis constant (Km) and maximal velocity (Vmax), for this enzyme were determined by using p -nitrophenyl-α-D-galactopyranoside as substrate. The results showed that the purification fold, specific activity, and yield were 126.52, 138.58 units/mg, and 21.5 %, respectively. The SDS-PAGE showed that the molecular weight of the purified enzyme was 75 kDa. The optimum pH and temperature of the purified α-galactosidase were detected at pH 6.0 and 50 °C, respectively. The kinetic constants, Michaelis constant (Km) and maximal velocity (Vmax), for this enzyme were 4.6 mM and 769.23 U/ml, respectively. α-galactosidase from Klebsiella pneumoniae was purified and characterized. (SDS-PAGE) analysis showed that the purified enzyme appeared as single band with a molecular weight of 75 kDa.

The influence of alendronate on bone formation and resorption in a rat ectopic bone development model.

Abstract:BACKGROUND Most bone grafting techniques that include bone marrow, alloplastic materials, and extracellular bone matrix produce new bone mass, filling bone defects unpredictably. In most cases, the new bone undergoes resorption due to low local strains, resulting in significant bone loss. Recently, it was shown that alendronate and other bisphosphonates reduce bone loss when administered systemically or locally. The aim of this study was to investigate whether alendronate is effective on bone formation or bone resorption. METHODS A total of 64 rats were divided into 2 main groups. In all the rats, fresh bone marrow removed from DA young rats was placed into demineralized rat femur cylinders (DBMC) and implanted into subcutaneous sites of host DA rats, to form new bone. Group A served as an alendronate treatment group, and group B served as a non-treated control. Group A received 100 microl of 1.5 mg/ml alendronate solution at 1, 2, and 3 weeks (group A1) and at 3, 4, and 5 weeks (group A2). At designated times, the rats were sacrificed, and the implanted DBMC was dissected out of the thorax and processed for histological and microradiography image analysis. RESULTS Alendronate given at 1, 2, and 3 weeks (during the bone formation phase) did not increase the amount of bone or the visual bone density in comparison to the time-matched control, after 4 and 8 weeks. When alendronate was injected at 3, 4, and 5 weeks, the bone mass increased by 70% and by 166% after 6 and 10 weeks, respectively, in comparison to the untreated control. The visual bone density in group A2 was maintained at the level of 140 +/- 15 at 6 weeks and 152 +/- 15 at 10 weeks. The matched, non-treated control group B2 was significantly lower, 106 +/- 20 and 108 +/- 15, respectively. The histological sections showed that alendronate treatment at 3, 4, and 5 weeks maintained the normal appearance of the ossicle at 6 and 10 weeks in comparison to the osteopenic bone appearance in the matched controls. CONCLUSIONS This study suggests that alendronate is effective in inhibiting bone loss, but ineffective during the bone formation phase. We suggest, therefore, that alendronate should be administered in procedures where bone resorption is expected.
Phospholipase C (heat-labile hemolysin) of Pseudomonas aeruginosa: purification and preliminary characterization

Abstract:Phospholipase C (heat-labile hemolysin) was purified from Pseudomonas aeruginosa culture supernatants to near homogeneity by ammonium sulfate precipitation followed by a novel application of DEAE-Sephacel chromatography. Enzymatic activity remained associated with DEAE-Sephacel even in the presence of 1 M NaCl, but was eluted with a linear gradient of 0 to 5% tetradecyltrimethylammonium bromide. Elution from DEAE-Sephacel was also obtained with 2% lysophosphatidylcholine, and to a lesser extent with 2% phosphorylcholine, but not at all with choline. The enzyme was highly active toward phospholipids possessing substituted ammonium groups (e.g., phosphatidycholine, lysophosphatidylcholine, and sphingomyelin); however, it had little if any activity toward phospholipids lacking substituted ammonium groups (e.g., phosphatidylethanolamine, phosphatidylserine, and phosphaditylglycerol). Collectively, these data suggest that phospholipase C from P. aeruginosa exhibits high affinity for substituted ammonium groups, but requires an additional hydrophobic moiety for optimum binding. The specific activity of the purified enzyme preparation increased 1,900-fold compared with that of culture supernatants. The molecular weight of the phospholipase C was estimated to be 78,000 by both sodium dodecyl sulfate-polyacrylamide gel electrophoresis and Sephacryl S-200 column chromatography and was 76,000 by high-performance size exclusion chromatography. The isoelectric point was 5.5. Amino acid analysis showed that phospholipase C was rich in glycine, serine, threonine, aspartyl, glutamyl, and aromatic amino acids, but was cystine free.
All MOS transistors bandgap reference using chopper stabilization technique

Abstract:A 0.6-V, 8-μW bandgap reference without BJTs is realized in the standard CMOS 0.13μm technology. All MOS transistors bandgap reference circuit with very low supply voltage 0.6V is designed. The chopper stabilization technique is used to improve the accuracy of the bandgap reference voltage. The measurement results have confirmed that the chopper stabilization technique reduces bandgap voltage error from 100mV to 30mV comparing to the one without chopper stabilization technique.

Abstract:We used the sex ratio of neonate Daphnia, as well as the more standard endpoints of adult survivorship and fecundity and neonatal morphology, as an assay for detecting the effects of the insecticides endosulfan and dieldrin. Dieldrin caused a decrease in sex ratio (number of males/number of males plus females); we observed no endosulfan effect. We estimated (by extrapolation) that the sex ratio was reduced by dieldrin from concentrations of about 30 ppb and higher, based on a linear decrease in sex ratio with log dieldrin concentrations from 50 to 600 ppb. Neither insecticide significantly affected adult survival or clutch size. Because sex ratio changed but total neonate production did not change, the data suggest that the effect of dieldrin was on the sex-determining system during embryogenesis. Neither insecticide caused morphological abnormalities. Mixtures of the two pesticides produced only additive effects.

Abstract:A thermostable GDSL family esterase-encoding gene, EstL5, was directly obtained from the genomic DNA of Geobacillus thermodenitrificans T2. Recombinant hexahistidine-tagged EstL5 was overexpressed, purified, and its biochemical properties were partially characterized. EstL5 was observed to be active within the temperature range of 0-80 degrees C, having maximal activity at 60 degrees C. Unlike most other thermostable enzymes, EstL5 displayed 24% of its highest activity at 0 degrees C. EstL5 exhibited a high level of activity within a pH range of 6.0-11.0, showing the highest activity at pH 8.0. EstL5 also retained 100% of its activity after a 12-h incubation at 55 degrees C. Furthermore, this enzyme was observed to be strongly inhibited by 10% (w/v) SDS and 0.1 mM PMSF. (C) 2012, The Society for Biotechnology, Japan. All rights reserved.

Abstract:The gdh and gdhr genes, encoding B-12-dependent glycerol dehydratase (GDH) and glycerol dehydratase reactivase (GDHR), respectively, in Klebsiella pneumoniae, were cloned and expressed in E. coli. Part of the beta-subunit was lost during GDH purification when co-expressing alpha, beta and gamma subunit. This was overcome by fusing the beta-subunit to alpha- or gamma-subunit with/without the insertion of a linker peptide between the fusion moieties. The kinetic properties of the fusion enzymes were characterized and compared with wild type enzyme. The results demonstrated that the fusion protein GDHALB/C, constructed by linking the N-terminal of beta-subunit to the C-terminal of alpha subunit through a (Gly(4)Ser)(4) linker peptide, had the greatest catalytic activity. Similar to the wild-type enzyme, GDHALB/C underwent mechanism-based inactivation by glycerol during catalysis and could be reactivated by GDHR.

Abstract:GK1506 from the thermophilic bacterium Geobacillus kaustophilus is a member of the phosphotriesterase-like lactonases, which can catalyze the hydrolysis of a broad range of compounds with different chemical properties. It is of particular interest because of its high thermostability and its dual activity towards organophosphate compounds and some lactones. These properties make GK1506 an attractive target for future enzyme engineering and use in practical applications. In order to resolve the crystal structure of GK1506 and to gain a better understanding of its biological function, recombinant GK1506 was expressed, purified and crystallized using 0.1 M HEPES pH 7.6, 12%(w/v) PEG 8000, 8%(v/v) ethylene glycol at 291 K. A 2.6 angstrom resolution native data set was collected from a single flash-cooled crystal (100 K) using 10%(v/v) glycerol as a cryoprotectant. These crystals belonged to space group P2(1), with unit-cell parameters a = 51.444, b = 80.453, c = 92.615 angstrom, beta = 99.29 degrees. Two molecules were assumed to be present per asymmetric unit, which gives a Matthews coefficient of 2.7 angstrom(3) Da(-1)
