Abstract:Rapid, simple and highly sensitive flow-injection (FI) chemiluminescence (CL) and flow-injection electrogenerated chemiluminescence (ECL) methods were developed for the determination of escitalopram oxalate (ESC), a selective serotonin reuptake inhibitor used as an antidepressant drug. The CL method was based on the CL reaction of ESC with acidic cerium(IV) and tris(2,2'-bipyridyl)ruthenium(II) (Ru(bipy)(3)(2)+). Various experimental parameters affecting CL intensity were carefully studied and optimised. The method enabled the determination of 0.001-50 µg/mL of ESC in bulk form with a correlation coefficient r = 0.9999. The limit of detection (LOD) was 0.01 ng/mL (S/N = 3). The ECL method was based on the ECL reaction of Ru(bipy)(3)(2)+ with the drug in an acidic medium, permitting the determination of ESC in the range of 0.00001-70 µg/mL with r = 0.9999 and LOD of 1 x 10(-4) ng/mL. The proposed methods were applied to the determination of ESC in commercial tablets. The results were compared statistically with those obtained from a published method using t- and F-tests.
Spectrophotometric method for simultaneous estimation of escitalopram oxalate and clonazepam in tablet dosage form

Abstract:A simple, accurate and precise spectrophotometric method has been developed for simultaneous estimation of escitalopram oxalate and clonazepam in combined dosage form. Simultaneous equation method is employed for simultaneous determination of escitalopram oxalate and clonazepam from combined dosage forms. In this method, the absorbance was measured at 238 nm for escitalopram oxalate and 273 nm for clonazepam. Linearity was observed in range of 5-100 mug/ml and 5-50 mug/ml for escitalopram and clonazepam respectively. Recovery studies confirmed the accuracy of proposed method and results were validated as per ICH guidelines. The method can be used for routine quality control of pharmaceutical formulation containing escitalopram and clonazepam.
Expression of MeCP2 in postmitotic neurons rescues Rett syndrome in mice.

Abstract:Mutations in MECP2 are the cause of Rett syndrome (RTT) in humans, a neurodevelopmental disorder that affects mainly girls. MeCP2 is a protein that binds CpG dinucleotides and is thought to act as a global transcriptional repressor. It is highly expressed in neurons, but not in glia, of the postnatal brain. The timing of MeCP2 activation correlates with the maturation of the central nervous system, and recent reports suggest that MeCP2 may be involved in the formation of synaptic contacts and may function in activity-dependent neuronal gene expression. Deletion or targeted mutation of Mecp2 in mice leads to a Rett-like phenotype. Selective mutation of Mecp2 in postnatal neurons leads to a similar, although delayed, phenotype, suggesting that MeCP2 plays a role in postmitotic neurons. Here we test the hypothesis that the symptoms of RTT are exclusively caused by a neuronal MeCP2 deficiency by placing Mecp2 expression under the control of a neuron-specific promoter. Expression of the Mecp2 transgene in postmitotic neurons resulted in symptoms of severe motor dysfunction. Transgene expression in Mecp2 mutant mice, however, rescued the RTT phenotype.

Abstract:Abstract Background Fluconazole, an antifungal drug, prevents fungi growth by inhibiting the formation of a protective covering. Ivermectin has several biological activities, such as antibacterial, antiviral, and anti-cancer characteristics, and offers various therapeutic outcomes. There are several commercial products containing these two drugs. Therefore, developing a method that can allow the simultaneous estimation of Fluconazole and ivermectin is inevitable to monitor them in commercial dosage forms. The hyphenated methodology that combines spectroscopic and chromatographic techniques is gaining high interest in the pharmaceutical industry. Consequently, the objective of present research work was to investigate robust and sensitive LC–MS/MS avenue for simultaneous determination of Fluconazole and ivermectin in pure material and combined dosage form. Results The simultaneous quantification of Fluconazole and ivermectin in tablet dosage form has been developed and validated using a straightforward, sensitive, practical, and repeatable LC–MS/MS approach. The separation was performed using a C 18 (150 × 4.6 mm) column, injection volume of 10 μL, and elution with acetonitrile: formic acid at a ratio of 70:30, with the column temperature at 30 °C, and a flow rate of 4.0 mL/min. The retention times of Ivermectin and Fluconazole were 1.10 min and 1.05 min, respectively. The calibration curves for Fluconazole and ivermectin demonstrated significant linearities indicated by the correlation coefficients (r 2 = 0.999 and r 2 = 0.997) and precision (% R.S.D. of 1.58 and 1.13). The linear correlation between peak area and concentration allowed high percentage recoveries of 98.5%–99.4% and 97.8%–99.3% for Fluconazole and Ivermectin, respectively. The L.O.D.s for Fluconazole and ivermectin were found to be 0.0034 and 0.074 g/mL, respectively. The L.O.Q.s for Fluconazole and ivermectin were 0.010 and 0.225 g/mL, respectively. Conclusion All the analytical parameters were identified and found to be within the acceptable range set forth by the ICH guidelines, demonstrating the devised method's acceptability in the simultaneous detection and estimation of Fluconazole and ivermectin in the commercial dosage forms.
Development and Validation of HPTLC Method for the Estimation of Escitalopram oxalate and Flupentixol Dihydrochloride in Pharmaceutical Formulation

Abstract:A simple, precise, accurate High-performance thin-layer chromatography (HPTLC) method has been developed and validated for the estimation of escitalopram oxalate (ESC) and flupentixol dihydrochloride (FLU) in pharmaceutical formulation. Separation was achieved on an aluminum HPTLC plate coated with a 0.2 mm layer of silica gel 60 F254 with chloroform: methanol 4:6 (v/v) as mobile phase. The densitometry analysis was carried out in absorbance mode at 254 nm. The Rf values of escitalopram and flupentixol were 0.24 and 0.44 respectively. The linearity of the method was achieved in the range 3-7 μg/spot for ESC with correlation coefficient value 0.998 and 0.3-0.7μg/spot for FLU with correlation coefficient value 0.999. The developed method was validated according to ICH guidelines and the results were statistically validated. The limit of detection and quantification for escitalopram oxalate and flupentixol dihydrochloride was found to be 500 ng/spot and 50 ng/spot, 1000 ng/spot, and 100 ng/spot respectively.

Abstract:Objective:To establish a method for simultaneous determination of three consitituents in AEC Tablet containing as- pirin,ethenzamide and caffeine.Method:Inertsil-ODS column is used.The mobile phase is composed of methanol -0.1% di- ethylamine-acetic acid(40:56:4)with flow rate of 1.0mL/min.The detection wavelength is 280nm.Results:The linear re- sponse range were 120-480μg/mL for aspirin(r=0.9998),40-160μg/mL for ethenzamide(r=0.9998),20-80μg/mL for caf- feine(r=0.9997).The mean recoveries of aspirin,ethenzamide,caffeine were 100.3%,100.7%,99.7% respectively.Conclu- sion:The method is simple,quick and suitable for determination of these three constituents in compound preparation simulta- neously.

The accuracy of the methods was assessed by recovery studies and was found tobe99.64%and98.98%forSitagliptinphosphateandMetforminhydrochloride.Thedevelopedmethodwasvalidatedwithrespecttolinearity,accuracy(recovery),precisionandspecificity.Theresultswerevalidatedstatistically as per ICH Q2 R1 guideline and were found to be satisfactory. Abstract:Recently, a new formulation containing Mirabegron (MB) and Solifenacin succinate (SFS) has been approved for the management of over active bladder. However, only one analytical method has been reported for the simultaneous determination of both the analytes. Therefore, the current study was design to develop simple UV derivative spectroscopic and rapid RP-HPLC methods for simultaneous determination of MB and SFS. The chromatographic separation of MB and SFS was performed using Phenomenex Kinetex C18 (150mm × 4.5 mm × 5 μm) analytical column. A mixture of Water: Acetonitrile (20:80%v/v) was consider as mobile phase, at a flow rate of 1ml/min and at detector wavelength 225nm. A linear response was observe over the concentration range 2.5-12.5 μg/ml and 0.5-2.5 μg/ml respectively. The first order derivative method was develop by derivatisation of the zero absorption spectra for the first absorption spectra. The Zero crossing point of MB and SFS at 221 nm and 266 nm was obtain respectively. Beer’s law is obey in the concentration range of 7.5-20 μg/ml and 1.5-4 μg/ml for MB and SFS with correlation coefficient (R2) of 0.9984 and 0.9993 respectively. Both the methods were validated in accordance to guidelines for linearity, precision, repeatability, limit of detection (LOD), Limit of Quantification (LOQ), accuracy and robustness. Further, both the methods were validated and compared statistically using Student’s-t-test and employed for the concurrent estimation of MB and SFS in formulations. The proposed methods were simple, accurate, precise, and rapid. Therefore, they can be use for regular quality control of MB and SFS formulations and dissolution studies as well.
Rapid and sensitive method for simultaneous determination of citalopram with antihistamines by liquid chromatography

Abstract:A highly sensitive liquid chromatographic method with UV detection has been developed for simultaneous determination of citalopram, levocetirizine and loratadine in bulk drug, pharmaceutical formulation and human serum at 230nm employing 80:20 v/v methanol-water as mobile phase with pH3.5, adjusting flow rate of 1.0mL.min-1. Separation was achieved on Shimadzu Shim-pack CLC-ODS (M) 25M column within the linear range of 0.4-12.5, 0.8-25 and 0.8-25μg.mL-1 with R2 >0.998 and detection limit 7.75, 3.35 and 10.26ng.mL-1respectively. ICH guidelines were followed for validation showing 0.22-1.76, 0.06-1.83 and 0.22-2.11% RSD. The recovery of analytes in tablets and serum was found to be in acceptable range. The method was fruitfully employed for the determination of studied analyte in pharmaceutical formulation and human serum.

Abstract:Recently, a brand-new combination of mirabegron and solifenacin succinate received approval for managing the overactive bladder syndrome's bothersome symptoms. Herein, we introduce six validated UV-spectrophotometric approaches relying on simple mathematical calculations for rapid and concurrent analysis of MBR and SOS in their synthetic mixtures, commercial tablets, and human urine without preliminary separation. The proposed approaches were dual-wavelength, induced dual-wavelength, Fourier self-deconvolution, area under the curve, ratio difference, and first derivative ratio methods. These approaches were applied successfully for the concurrent determination of MBR and SOS in their mixtures (suffered from severe spectral overlap) with excellent recoveries without interference from pharmaceutical excipients or biological matrices. The concurrent analysis of MBR and SOS in spiked biological fluids was accomplished for the first time in this study using the Fourier approach. The proposed approaches were linear in ranges of (50–450) μg/mL and (70–1200) μg/mL for MBR and SOS, in order. The proposed approaches' greenness was evaluated using two tools namely; penalty points scoring system and green analytical procedure index (GAPI), revealing their superiority in greenness over the reported LC method. The proposed approaches were compared statistically in terms of t- and F-values with the reported LC method, revealing no discernible differences concerning accuracy or precision at p = 0.05. Accordingly, the proposed approaches are valid for regular analysis of MBR and SOS as affordable substitutes in QC labs lacking expensive chromatographic instruments.

Abstract:The study aimed to develop a new reverse-phase high-performance liquid chromatography (RP-HPLC) method with diode array detection (DAD) detection for simultaneous estimation of escitalopram (EST) and clonazepam (CZP) in tablet dosage forms with a quality by design (QbD) approach. The chromatographic conditions were optimized by Box-Behnken design (BBD) and developed method was validated for the linearity, system suitability, accuracy, precision, robustness, sensitivity, and solution stability according to International Council for Harmonization (ICH) guidelines. EST and CZP standard drugs peaks were separated at retention times of 2.668 and 5.046 min by C-18 column with dimension of 4.6 × 100 mm length and particle size packing 2.5 µm. The mobile phase was methanol: 0.1% orthophosphoric acid (OPA) (25:75, v/v), with a flow rate of 0.7 mL/min at temperature of 26 °C. The sample volume injected was 20 µL and peaks were detected at 239 nm. Using the standard calibration curve, the % assay of marketed tablet was founded 98.89 and 98.76 for EST and CZP, respectively. The proposed RP-HPLC method was able to detect EST and CZP in the presence of their degradation products, indicating the stability-indicating property of the developed RP-HPLC method. The validation parameter's results in terms of linearity, system suitability, accuracy, precision, robustness, sensitivity, and solution stability were in an acceptable range as per the ICH guidelines. The newly developed RP-HPLC method with QbD application is simple, accurate, time-saving, and economic.
Selective LC-MS/MS determination of citalopram enantiomers and application to a pharmacokinetic evaluation of generic and reference formulations

Abstract:Two methods using LC-MS/MS were validated to quantify citalopram (CTP) racemate [(R/S)-CTP] and the enantiomers (R)-CTP and (S)-CTP in human plasma, respectively. Paroxetine hydrochloride was used as the internal standard, and samples were extracted by protein precipitation with acetonitrile. The non-enantioselective method was conducted using a C18 column, and the mobile phase consisted of water for solvent A and acetonitrile for solvent B, both with 0.1% formic acid. For the chiral method, an analytical column Lux Cellulose-1 was used. Mobile phase A was composed of water with 0.025% of formic acid and 0.05% of diethylamine, and mobile phase B consisted of acetonitrile:2-propanol (95:5, v/v). No significant matrix effects were observed at the retention times of analytes and internal standard. The mean recovery was 89%, and the assays were linear in the concentration range of 1-50 and 5-30 ng/mL for the non-enantioselective and enantioselective methods, respectively. The intra- and inter-day precisions of both methods were less than 12.30%, and the accuracies were less than 12.13%. The validated methods were successfully applied to a pharmacokinetic study in which 20-mg CTP tablets were administered to healthy volunteers, and their plasma levels were monitored over time in a bioequivalence study. HIGHLIGHTS: Simple and rapid LC-MS/MS method for the quantification of citalopram and its enantiomers in human plasma. Both methods were demonstrated to be selective, reliable, and sensitive. Both methods have sufficient sensitivity to quantify the steady state through concentrations already reported for citalopram and escitalopram. Validated method presented in this study can be suitably applied to pharmacokinetic studies involving citalopram and escitalopram. Bland-Altman analysis suggested that non-enantioselective and enantioselective methods can be applied in pharmacokinetic studies.

Abstract:The aim of this study is to create and validate a fast, easy-to-use, affordable, sensitive, and accurate method for measuring Escitalopram and Flupentixol in bulk and pharmaceutical products using Reversed-Phase High-Performance Liquid Chromatography (RP-HPLC). Agilent Eclipse XDB C18 column was used with a running phase composed of 0.1% Tri fluoro acetic acid: Acetonitrile (30:70 v/v) at a flow rate of 1.0 ml/min. UV detection was used at a wavelength of 231 nm. Flupentixol and Escitalopram correlation coefficients were found to be 0.999 over a concentration range of 1.25-7.5 μg/ml and 25-150 μg/ml, respectively. Escitalopram and Flupentixol have respective retention times of 3.235 and 3.858 minutes. The run time for separating Flupentixol and Escitalopram peaks was 6 minutes. This method, proposed as a regular analysis and quality control tool for medications that contain these active drugs either individually or in combinatio, was evident to be a suitable one. Facile approaches for determination of Bromhexine Hydrochloride and its active metabolite Ambroxol Hydrochloride using Eosin Y

Abstract:New validated Spectroscopic methods were developed to assay Bromhexine Hydrochloride and its active metabolite Ambroxol Hydrochloride separately in pure form and pharmaceutical formulations. The spectrophotometric assay (method I) shows complex formation between each of the drugs and Eosin Y at 540nm at pH 3.6 and 3.4mL of 4×10-4M Eosin for Bromhexine and Ambroxol. The Spectrofluorimetric assay (method II) depends on quenching eosin native fluorescence by the studied drugs, which measured at 540nm after excitation at 302nm. The spectrophotometric absorbance-concentration plot is rectilinear over the ranges (1.0-5.0) and (1.0-10.0) μg/mL for bromhexine and ambroxol with LOD of 0.31 and 0.14μg/mL and LOQ of 0.94 and 0.42μg/mL for the two drugs respectively. The fluorometric-concentration plot is linear along the range (1.0-5.0) μg/mL and (1-10) μg/mL for the two drugs respectively with LOD of 0.13μg/mL and 0.22μg/mL and LOQ of 0.4μg/mL and 0.65μg/mL for the two drugs, respectively. Developed assays have been validated in agreement with ICH recommendations and they were used in the analysis of commercial drug formulations containing the two mucolytic drugs and the results were matching with those obtained by the comparison method.

Abstract:Rapid, sensitive, reproducible, and cost-effective zero-order and first-order derivative spectrophotometric method has been developed for the quantification of solifenacin succinate in bulk and its tablet formulation. Preliminary spectrophotometric determination of the drug was carried out in 0.1 N HCl, methanol, and in phosphate buffer pH 2.0 with a total of thirty-three parametric variations. The selected method with three parametric variations employing peak–zero (P–0) and peak–peak (P–P) techniques was assessed for stability, indicating a potential for force-degraded solutions. The method was validated in accordance with the ICH guidelines with respect to linearity, accuracy, precision, limit of detection (LOD), limit of quantification (LOQ), and robustness. Excellent linearity was noted in a concentration range of 1.0–80.0 μg/mL for all three method variants. Limits of detection for the proposed method ranged from 0.03–0.16 μg/mL and limits of quantitation were found to be in the range of 0.09–0.48 μg/mL, respectively. Excellent recovery of the drug was obtained from its marketed tablet formulation with the proposed method.

Abstract:An efficient, cost effective, rapid reverse phase high-performance liquid chromatographic method was developed and validated for the determination of Ezetimibe and Simvastatin in pharmaceutical dosage forms. Chromatography was carried out by using X-terra C18, 150 x 4.6mm, 3.5μ or equivalent internal diameter with a mixture of 0.5 mL glacial acetic acid in 2000 mL water as mobile phase A and Acetonitrile as mobile phase B. Analytical method validation parameters such as specificity, linearity, precision, accuracy, solution stability and robustness, limit of detection (LOD) and limit of quantification (LOQ) was done. The correlation coefficient was found to be linear for each analyte in the desired concentration range. The average recovery was found with range of 96.7 and 107.7 for Ezetimibe and Simvastatin and their respective impurities respectively. The proposed method is highly sensitive, precise and accurate, which was evident from the LOD value range of minimum 0.135 ppm for Ezetimibe and 0.356 ppm for Lovastatin & Epilovastatin for Simvastatin impurities. Hence the present method can be applied successfully for the quantification of finished dosage form in the combined formulations of Ezetimibe and Simvastatin.

Abstract:Two simple and sensitive spectrophotometric methods have been developed for the determination of eperisone hydrochloride based on its ability to be detected in UV region (Normal UV) and its oxidation using potassium permanganate in alkaline medium (kinetic spectroscopic). The detection was done at 261.40 nm and 603.5 nm. The different experimental parameters affecting the method development were studied and optimized. The initial rate and fixed time method were utilized to construct calibration graph, and 5 minutes and 3 minutes, respectively, were found suitable for the determination of the concentration of drug. Linearity was found over the concentration range of 2–20 μ g/mL, 15–30 μ g/mL, and 15–35 μ g/mL by UV, initial rate, and fixed time methods, respectively. The results were validated as per the ICH guidelines. RSD values were found to be less than 2%. The methods were applied for estimation of eperisone hydrochloride in RAPISONE (Abbott, Maharashtra). The assay results were found to be 100.4% ± 0.08, 99.93% ± 0.05, and 99.41% ± 0.04 by UV, initial rate, and fixed time method, respectively. Statistical comparison of the proposed methods showed a good agreement indicating no significant difference in accuracy and precision, thus confirming the suitability of UV and kinetic method for the estimation of eperisone hydrochloride in bulk as well as in tablet dosage forms.
