Abstract:The assessment of chemical alternatives for hazardous substances is an important prerequisite for avoiding regrettable substitution, and several methods have been developed in the past to perform such an hazard assessment for chemical alternatives. We investigate here whether GreenScreen®, Cradle to Cradle®, Multi Criteria Decision Analysis (MCDA), the Pollution Prevention Options Analysis System, the U.S. EPA Safer Choice Standard and Criteria, and the GHS column model 2020 from IFA use similar criteria for the evaluation of substances as Article 57 of REACH and how suitable these methods are for assessing per- and polyfluoroalkyl substances. MCDA and GreenScreen® were analyzed in detail using two different datasets. The results of the assessments show that none of the investigated hazard assessment methods use the same criteria as described in Article 57 of REACH. It was also not possible to parameterize multi-attribute value theory (MAVT), a commonly used MCDA method, to align with Article 57 of REACH by using the relatively simple objective hierarchy that has been proposed in previous publications. There is therefore an urgent need for a modified/new method that can be used in the future to assess organic substances that are used within the European Economic Area.

Multi-process regulation of novel brominated flame retardants: Environmentally friendly substitute design, screening and environmental risk regulation

Abstract:Novel brominated flame retardants (NBFRs), one of the most widely used synthetic flame-retardant materials, have been considered as a new group of pollutants that potentially affect human health. To overcome the adverse effects of NBFRs, a systematic approach for molecular design, screening, and performance evaluation was developed to generate environmentally friendly NBFR derivatives with unaltered functionality. In the present study, the features of NBFRs (long-distance migration, biotoxicity, bioenrichment, and environmental persistence) were determined and characterized by the multifactor comprehensive characterization method with equal weight addition, and the similarity index analysis (CoMSIA) model was constructed. Based on the three-dimensional equipotential diagram of the target molecule 2-ethylhexyl tetrabromobenzoic acid (TBB), 23 TBB derivatives were designed. Of these, 22 derivatives with decreased environmental impact and unaltered functional properties (i.e., flame retardancy and stability) were selected using 3D-QSAR models and density functional theory methods. The health risks of these derivatives to humans were assessed by toxicokinetic analysis; the results narrowed down the number of candidates to three (Derivative-7, Derivative-10, and Derivative-15). The environmental impact of these candidates was further evaluated and regulated in the real-world environment by using molecular dynamics simulation assisted by the Taguchi experimental design method. The relationship between the binding effects and the nonbonding interaction resultant force (TBB derivatives-receptor proteins) was also studied, and it was found that the larger the modulus of the binding force, the stronger the binding ability of the two. This finding indicated that the environmental impact of the designed NBFR derivatives was decreased. The present study aimed to provide a new idea and method for designing NBFR substitutes and to provide theoretical support for restraining the potential environmental risks of NBFRs.
Modifying multi-criteria decision analysis for assessments of chemical alternatives

Abstract:Effectual assessment of chemical alternatives is necessary to avoid “regrettable” substitution. In a preceding study, an analysis of seven assessment-of-alternatives (AoA) methods found that none of them is fully aligned with Article 57 of the European REACH regulation, indicating a need for a method better reflecting European regulations. This paper presents a modified multi-criteria decision analysis (MCDA) method for the assessment of chemical alternatives (MCDA-ACA). We investigate and recommend MCDA method parameters for the assessment of chemical alternatives, specifically scope of objectives considered, aggregation of objectives, curvature of the value functions, weights, and the introduction of a classification threshold. The MCDAACA method allows for the aggregation of hazards in such a way that poor performance in one hazard cannot be compensated for by good performance in another hazard. The MCDA-ACA method parameters were developed and tested using two datasets with the aim to classify chemical alternatives into acceptable (non-regrettable) and unacceptable (regrettable) alternatives according to the regulations set in Europe. The flexibility of the general method was explored by adapting the method to align with two hazard assessment frameworks, GreenScreen® and Article 57 of REACH alone. The results show that MCDA-ACA can easily be adapted to other jurisdictions as required.

Probing Legacy and Alternative Flame Retardants in the Air of Chinese Cities

Abstract:An increasing number of alternative flame retardants (FRs) are being introduced, following the international bans on the use of polybrominated diphenyl ether (PBDE) commercial mixtures. FRs' production capacity has shifted from developed countries to developing countries, with China being the world's largest producer and consumer of FRs. These chemicals are also imported with e-waste to China. Therefore, it is important to understand the current status of regulated brominated FRs, their phase-out in China, and their replacement by alternatives. In this study, a broad suite of legacy and alternative FRs, including eight PBDEs, six novel brominated FRs (NBFRs), two dechlorane plus variants (DPS), and 12 organophosphate FRs (OPFRs) were evaluated in the air of 10 large Chinese cities in 2018. OPFRs are the most prevalent FRs in China, exhibiting a wide range of 1–612 ng/m3, which is several orders of magnitude higher than PBDEs (1–1827 pg/m3) and NBFRs (1–1428 pg/m3). BDE 209 and DBDPE are the most abundant compounds in brominated FRs (>80%). The North China Plain (NCP, excluding Beijing), Guangzhou, and Lanzhou appear to be three hotspots, although with different FR patterns. From 2013/2014 to 2018, levels of PBDEs, NBFRs, and DPs have significantly decreased, while that of OPFRs has increased by 1 order of magnitude. Gas-particle partitioning analysis showed that FRs could have not reached equilibrium, and the steady-state model is better suited for FRs with a higher log KOA (>13). Abstract:Endocrine disruptors (EDs) are chemical substances that interfere with the endocrine system, adversely affecting human health and environment. Legislation with aim to eliminate and ban EDs have been introduced in EU, but the identification of EDs remains challenging and crucial step towards regulation and risk management. A guidance for ED assessment has been recently established for pesticides and biocides in the EU, which heavily relies on traditional toxicological testing in vivo. Most notably lacking mechanistic methods for some ED modalities and not covering many other modalities that might be affected by EDs. In this project, we focus on the ED assessment according to the valid legislation and explore the possibility to employ alternative methods to bolster the mechanistic understanding of the ED effects and eventually decrease the need for in vivo testing. We selected a well‐studied industrial chemical perfluorooctanesulfonic acid (PFOS) to be a model compound in a case study for ED assessment where the EU criteria were applied in the frame of human health risk assessment with focus on thyroid disruption and developmental neurotoxicity. A systematic literature review has been conducted for these effects (Scopus, Pubmed, Embase), and relevant studies were selected by title/abstract screening (RAYYAN) and full‐text examination. Selected studies were assessed for reliability (SciRAP), and all relevant data were extracted into a database and assessed by Weight of Evidence (WoE) approach. The initial analysis showed potential endocrine adverse effects and endocrine activity, meeting the ED criteria. The use of mechanistic and alternative methods enhanced the outcomes of WoE assessment. Also, the study provides a great hands‐on experience with the most up‐to‐date development in the area of risk assessment and EDs.

Life cycle assessment of flame retardants in an electronics application

Abstract:PurposeFlame retardants are added to plastics and textiles to save lives. However, certain brominated flame retardants (BFRs) form an environmental hazard and should be replaced by less harmful alternatives. In the recently completed European research project ENFIRO, we examined which alternatives are most suitable from a technical and environmental perspective. This study describes the LCA comparison of BFRs and halogen-free flame retardants (HFFRs) in an electronics product, in order to compare their environmental impacts over the whole life cycle and identify where in the life cycle the main impacts occur.MethodsThis cradle to grave LCA used the complete life cycle of a laptop computer as the functional unit. Specific attention was paid to often neglected aspects, including emissions of flame retardants in all life cycle phases, emissions during accidental fire and improper waste treatment. New characterization factors for toxicity of flame retardants were calculated using USES-LCA2 and included in the impact assessment.Results and discussionThe largest differences in impact were found to occur in the waste phase due to an increased dioxin emission formed out of BFRs during improper waste treatment. Minor human toxicity and ecotoxicity impacts of FRs are present due to volatilization in the use phase. FR emissions during accidental fire vary with the FR’s mode of action (active in the gaseous or solid phase). The BFR scenario has a higher impact than the HFFR scenario due to a higher rate of smoke formation and a higher terrestrial ecotoxicity score. In most phases of the life cycle of FRs, fossil energy use related impact categories dominate the LCA score, i.e. climate change, fossil depletion, and particulate matter formation. Over the full life cycle, the BFR scenario has a slightly higher environmental impact than the HFFR scenario, mainly through the contribution of human toxicity in the waste phase.ConclusionsThe study shows that for improvements of the life cycle environmental performance of FRs, the waste treatment phase is critical. Export and improper treatment of WEEE have the highest impact of all waste treatment options for both the BFR and HFFR scenarios, and efforts should be intensified to reduce the amount of WEEE ending up in this scenario. The study further shows that processes which are often ignored in LCA can give relevant insights into the environmental performance of a product. It is therefore recommended to broaden the scope and system boundaries of future LCA studies to include unofficial scenario options (specifically in the end-of-life phase) to provide a more complete description of the full environmental impact of a product’s life cycle and thereby contribute to relevant discussions in society and policy.

A European nano-registry as a reliable database for quantitative risk assessment of nanomaterials? A comparison of national approaches

Abstract:Despite the fact that nanomaterials have been in use for decades and chemicals legislation is largely harmonised within the EU, quantitative and safety-relevant information on nanomaterials is still scarce. In particular, information about production volumes, their unique physicochemical properties (size, specific surface area, etc.) and nanomaterial exposure, which may lead to adverse effects on human health and the environment, is still lacking. While the latest amendments of the REACH Annexes have led to certain improvements, a harmonised EU-wide nano-registry would provide additional quantitative data for risk assessment but is not foreseeable for the near future. Since the European Commission, the European Parliament and some member states take contrasting approaches to the regulation of nanomaterials, France, Belgium, Denmark, Sweden and Norway (as a country of the European Economic Area), launched national mandatory reporting systems to collect quantitative information, thus fostering early risk assessment of nanomaterials. In this study, we compare national registries - based on a literature review and expert interviews - and show differences between the regulations under the respective national laws and REACH regulation. These include, for instance, thresholds for notification and level of detail on the specification of the nanomaterial, mixture and/or product, the definition of exceptions for the requirement to register and the timing of registration. As this heterogenous regulatory framework hinders comparability and potentially creates trade barriers, we argue that a harmonised EU-wide nano-registry would substantially improve the current situation by promoting the safe and sustainable handling of nanomaterials, increasing transparency and trust, and consequently nurturing innovation. Such an EU-wide nano-registry should both cover nanomaterials as substances or mixtures, such as in REACH registration, and the semi-/finished products they will be used in, since the exposure, and thus the hazardous potential of released nanomaterials during their life cycle, depends largely on the scope of application.

Abstract:For many years, efforts have been made to reduce the flammability of unsaturated polyester resins (UPRs), which are often used in the rail, shipbuilding, and construction industries. Without modification, they often fail to meet fire safety standards. Despite a rich history of flame retardants (FRs) applied to UPRs, researchers seek new solutions that will provide lower flammability and smoke density, as well as attaining a lower environmental impact from the composites. The objective of the study is to highlight the most important recent research on promising nano FRs in order to promote their further development. Mechanisms of action of several groups of nano FRs, such as clay-based, carbon-based, transition metal compounds, layered double hydroxides, polyhedral oligomeric silsesquioxanes, and others, including bio-based, have been studied. Particular emphasis has been laid on nano FRs applied to UPRs, and their influences on thermal stability, flammability, and mechanical properties. Moreover, the environmental impact and toxicity of nano FRs have been discussed. Results have proved that nano FRs applied at low loadings may significantly improve thermal stability, with a simultaneous increase or only a slight decrease in mechanical properties. However, attention on related environmental issues has highlighted the necessity of carefully selecting novel nano FRs.

Quantitative Assessment of Whether Phosphorus-Based Flame Retardants Are Optimizing or Degrading the Fire Hazard of Aircraft Carbon Fiber/epoxy Composites

Abstract:Fire hazards are a primary concern in aircraft accidents, and controlling combustible materials is pivotal in managing cabin fire risks. This work aims to elucidate the impact of widespread measures that introduce phosphorus-based flame retardants (PFRs) on the fire safety of carbon fiber epoxy composites (CF/EP). Firstly, the hazardous indicators for flame retardant CF/EP composites exhibit two categories of optimization and degradation, which are caused by the intrinsic feature, pyrolysis characteristics and chemical valence states of PFRs. Secondly, the fire hazards of composites are assessed by fusing the analytic network process and information weights guided by Nash Equilibrium theory. The method combines the merits of subjective and objective weights from dimensions of intrinsic hazards, interrelationships and data dispersion of indicators. Finally, the assessed fire hazard rankings under 35 and 65 kW/m2 both are EP > CF/EP16%DOPO-HQ > CF/EP > CF/EP7%DPO. In conclusion, DOPO-HQ (+1) primarily increases the fire hazard of CF/EP due to gas-phase flame-retardant effects, while DPO (+3) predominantly reduces the fire hazard of CF/EP by contributing to condensed-phase effects. This work highlights the contradictory effects of PFRs on the fire safety of CF/EP composites due to structure and thermal stability. The proposed ANP and IW hybridization method provides a theoretical foundation for selecting flame retardants and assessing material fire hazards comprehensively.

Abstract:The increasing market demand for electronic devices has raised concerns regarding the environmental impact of the flame retardants used in their production. Traditionally, brominated flame retardants have been used for their effectiveness despite their detrimental effects on the environment and hindrance to the principles of a circular economy. Evidently, harmful flame retardants should be replaced with less harmful ones. Hence, the exploration of alternatives, such as organophosphorus flame retardants like aluminum diethyl phosphinate (DEPAL), presents a promising avenue. Therefore, the goal of this study is to investigate the environmental impacts of DEPAL-based flame retardants in polyamides for use in connectors, plugs, and USB-C ports and to compare their impacts with those of their brominated flame-retardant counterparts. The life cycle impact results show that the flame-retardant polyamides using DEPAL have lower environmental impacts than the ones using halogenated flame retardants. Nonetheless, it is important to note that the production of phosphorus still requires large amounts of energy. Hence, switching to renewable energy can significantly lower the footprint of DEPAL. Furthermore, it offers an eco-friendly alternative to traditional flame retardants.

Abstract:Septic shock during pregnancy is very rare but has high mortality. We report a case of septic shock in a pregnant woman with ileus, showing that severe ileus in a pregnant woman could be attributed to life-threatening septic shock and that we should give special attention to a nosocomial infection.

Abstract:Introduction. Replacing hazardous substances with less hazardous ones is a basic principle of any good chemical risk management. At the first stage of scientific research on the development of a national concept for the replacement of highly hazardous chemicals in the composition of products (food, synthetic detergents and household chemicals, pesticides, paints and varnishes, basic chemicals) with safer analogues, the purpose of the research was: •to study international and domestic approaches to the organization of monitoring and regulation of substances highly hazardous to human health and the environment; •analysis, selection and scientific substantiation of criteria for selecting chemicals for their replacement with safer analogues. Materials and methods. materials, used for the analysis were the literature sources from the bibliographic databases Web of Science, MedLine, EMBASE, Global Health, PubMed, Scopus, RSCI. The guidelines and recommendations of the OECD, WHO, ILO, FAO, UNEP on the organization and implementation of monitoring of highly hazardous chemicals in environmental objects have been studied and analyzed. A number of international agreements are considered, which are based on criteria for prohibiting or restricting the use of substances on the market that cause an unacceptable risk. Results. An analysis of international approaches to the sound management of chemicals has shown that the identification of causal relationships between health and/or environmental conditions and exposure to a chemical factor is a trigger for the concept of substitution. In this regard, the tools and databases of socio-hygienic monitoring carried out by Rospotrebnadzor can serve as the basis for identifying highly hazardous substances that require management decisions to be made to ban, restrict circulation and replace them with safe analogues. The study of foreign and domestic materials on the selection of priority criteria for identifying substances of greatest concern for the purpose of risk assessment and further regulation showed that the following indicators are the main ones: •biological activity (carcinogens, mutagens, GHS class 1A and 1B reprotoxicants, endocrine disruptors), •stability in the environment, •bioaccumulative potential (bioconcentration factor BCF >2000, partition coefficient n-octanol/water Log Kow ≥ 4), •the possibility of cross-media transfer (air, water flows), •toxicity to representatives of aquatic biota (acute and chronic toxicity of hazard class 1 in accordance with GHS), •production volumes (volumes of emissions and discharges), •number of contacts.

Abstract:There have been many attempts to compile comprehensive lists of flame retardants. However, this goal has proven challenging due to the heterogeneity of compounds that can be used as flame retardants coupled with changes in formulation chemistry over time. Flame retardants have been the focus of many recent existing hazard, exposure, and risk assessments. These assessments have been class-based or for individual chemical substances. Here, diverse sets of publicly available data sources from governmental organizations and the open literature were compiled to develop an inventory of chemicals used as flame retardants and organohalogen flame retardants. The chemical substances from these data sources were mapped to appropriate chemical identifiers via manual curation and deduplicated. Despite different data sources containing a large number of overlapping chemical substances, compiling information from multiple data sources was found to increase the breadth of potential flame retardant chemistries. The flame retardant and organohalogen flame retardant inventories were developed as a resource for scientists interested in better understanding properties of flame retardant and organohalogen flame retardant classes.

Abstract:Abstract The EU Directive 2010/63/EU on the protection of animals used for scientific purposes and other EU regulations, such as REACH and the Cosmetic Products Regulation advocate for a change in the way toxicity testing is conducted. Whilst the Cosmetic Products Regulation bans animal testing altogether, REACH aims for a progressive shift from in vivo testing towards quantitative in vitro and computational approaches. Several endpoints can already be addressed using non-animal approaches including skin corrosion and irritation, serious eye damage and irritation, skin sensitisation, and mutagenicity and genotoxicity. However, for systemic effects such as acute toxicity, repeated dose toxicity and reproductive and developmental toxicity, evaluation of chemicals under REACH still heavily relies on animal tests. Here we summarise current EU regulatory requirements for the human health assessment of chemicals under REACH and the Cosmetic Products Regulation, considering the more critical endpoints and identifying the main challenges in introducing alternative methods into regulatory testing practice. This supports a recent initiative taken by the International Cooperation on Alternative Test Methods (ICATM) to summarise current regulatory requirements specific for the assessment of chemicals and cosmetic products for several human health-related endpoints, with the aim of comparing different jurisdictions and coordinating the promotion and ultimately the implementation of non-animal approaches worldwide. Recent initiatives undertaken at European level to promote the 3Rs and the use of alternative methods in current regulatory practice are also discussed.

Abstract:Experimentation demonstrates long-range surface plasmon polariton waveguides as a useful biosensor to selectively detect gram negative or gram positive bacteria in human urine having a low concentration of constituents. The biosensor can detect bacteria at concentrations of 10(5) CFU/ml, the internationally recommended threshold for diagnostic of urinary tract infection. Using a negative control urine solution of bacterial concentration 1000☓ higher than the targeted bacteria, we obtain a ratio of 5.4 for the positive to negative signals.
Development of a broad spectrum method for measuring flame retardants - overcoming the challenges of non-invasive human biomonitoring studies

Abstract:Flame retardants (FRs), such as polybrominated diphenyl ethers (PBDEs) and phosphate flame retardants (PFRs), are a diverse group of compounds that are used to improve fire safety in many consumer products, such as furniture, textiles, electronics, etc. As these compounds are potentially harmful for human health, there is a need to better understand human exposure. Exposure to environmental contaminants can be monitored by the measurement of external sources of exposures and also by the determination of contaminant levels in human samples. For ethical and practical reasons, noninvasive matrices, such as hair, are preferred but, unfortunately, not widely used due to methodological limitations. A major challenge is sample availability: only small amounts can be sampled per individual. Multi-residue methods are therefore essential in order to determine multiple compounds in low sample amounts. In the framework of the FP7 project (INFLAME), an analytical method for the simultaneous determination of PBDEs and PFRs in human hair has been optimized and validated. Before extraction, hair samples (200 mg) were denaturated in nitric acid (HNO3) for 25 min at 25 °C. Consecutively, the samples were extracted using a mixture of hexane:dichloromethane, and extracts were further fractionated on Florisil. Fraction A which contained PBDEs was additionally cleaned on acidified silica gel and measured by gas chromatography coupled with electron capture negative ionization mass spectrometry (GC-ECNI-MS), while fraction B containing PFRs was directly analyzed by liquid chromatography tandem mass spectrometry (LC-MS/MS). This approach resulted in recoveries between 81-120% for PBDEs and 75-113% for PFRs (relative standard deviation (RSD) < 16%, n = 9). The optimized multi-residue method has been applied to 20 human hair samples. The obtained results indicated that the levels of PBDEs in hair samples were very low (0.2-12 ng/g) in relation to PBDE levels in human hair samples from other studies and most of the time below the method limit of quantification (LOQm). On the contrary, the PFR levels were relatively high as they were in the range of the levels previously found in dust samples (2-5,032 ng/g hair). We would like to highlight that the contribution of air and dust cannot be neglected (especially in the case of PFRs); therefore, we suggest that hair might be a good indicator of retrospective and integral exposure (which includes atmospheric deposition as well as endogenous mechanisms). Moreover, the aim of our study is focused on exposure assessment and levels detected in hair (independently of whether they come from internal or external exposure) and will significantly contribute to the exposure assessment.

Abstract:Rubbers combine the flexibility with mechanical strength, supporting myriad applications, but suffer from inherent flammability. Formulation and production of flame-retardant rubber composites (FRRCs) have intensively been practiced over years, but not comprehensively reviewed. This necessity has outlined collecting, analyzing, screening, classifying, and interpreting the literature with the aim of classifying the FRRCs. We quantified and visualized the flame retardancy of natural and synthetic FRRCs by grouping FRRCs based on their flame retardants (FRs) among green, mineral, phosphorus-based, nitrogen-based, carbonaceous, and hybrids of two or more types. Available data on cone calorimetry, limited oxygen index (LOI), and UL-94 of FRRCs were carefully extracted and plotted. Flame Retardancy Index (FRI) was used to specify the Poor, Good, or Excellent classes of flame retardancy in association with the chemistry and concentration of FRs to broaden the future innovation avenues. Machine Learning (ML) modeling enabled visualization of flame retardancy landscapes of natural and synthetic rubbers in terms of the chemistry and concentration of FRs. Overall, a downward trend in mechanical properties of FRRCSs against FRs amount was oxplored. This study proposed a general guideline for recognizing gaps in previous investigations and mechanistic interpretations. In conclusion, we highlight that the future FRRCs should take advantage of hybridizing FRs in order to meet the fire safety requirements, which would be possible by the innovative colorful checkered flame retardancy chart presented in this survey.
