Abstract:The debate surrounding nature versus nurture remains a central question in neuroscience, psychology, and in psychiatry, holding implications for both aging processes and the etiology of mental illness. Epigenetics can serve as a bridge between genetic predisposition and environmental influences, thus offering a potential avenue for addressing these questions. Epigenetic clocks, in particular, offer a theoretical framework for measuring biological age based on DNA methylation signatures, enabling the identification of disparities between biological and chronological age. This structured review seeks to consolidate current knowledge regarding the relationship between mental disorders and epigenetic age within the brain. Through a comprehensive literature search encompassing databases such as EBSCO, PubMed, and ClinicalTrials.gov, relevant studies were identified and analyzed. Studies that met inclusion criteria were scrutinized, focusing on those with large sample sizes, analyses of both brain tissue and blood samples, investigation of frontal cortex markers, and a specific emphasis on schizophrenia and depressive disorders. Our review revealed a paucity of significant findings, yet notable insights emerged from studies meeting specific criteria. Studies characterized by extensive sample sizes, analysis of brain tissue and blood samples, assessment of frontal cortex markers, and a focus on schizophrenia and depressive disorders yielded particularly noteworthy results. Despite the limited number of significant findings, these studies shed light on the complex interplay between epigenetic aging and mental illness. While the current body of literature on epigenetic aging in mental disorders presents limited significant findings, it underscores the importance of further research in this area. Future studies should prioritize large sample sizes, comprehensive analyses of brain tissue and blood samples, exploration of specific brain regions such as the frontal cortex, and a focus on key mental disorders. Such endeavors will contribute to a deeper understanding of the relationship between epigenetic aging and mental illness, potentially informing novel diagnostic and therapeutic approaches.

Abstract:Epigenetic modifications happen in a dynamic and continuous fashion during life. These include molecular modifications of DNA, RNA, and proteins (histones) during cellular renewal and differentiation, and organ development throughout development and life. It is not surprising that deregulation of such fundamental mechanisms could lead to cellular abnormalities and organ disfunction, causing human disease. While all body parts and organs have their own regulatory processes, the mammalian brain and central nervous system appear to be the most complex part of the body. Accordingly, neurons that are the nerve cells in the central and peripheral nervous system have distinct characteristics that makes them especially important. Of note, change in epigenetic modifications in the central or peripheral nervous system may lead to neurological disorders. This special topic on "Epigenetic modification in neurological diseases" includes four peer-reviewed articles published in Frontiers in Neuroscience.Focusing on the role of non-coding regulatory RNA molecules in Alzheimer's disease (AD), Canoy and collogues discuss recent advances in AD pathology [1]. In this systemic review, the authors cover a range of topics on long non-coding RNA (lncRNA) molecules, microRNAs (miRNA), circular RNAs (circRNA), as well as piwi-interacting RNAs (piRNA). In addition to the role of these regulatory RNAs in AD pathology, their potential application as diagnostic markers as well as possible therapeutic targets are also discussed. In this regard, the authors cover implication of these regulators RNAs in different cellular processes including cell propagation and cell death, apoptosis, autophagy, tau phosphorylation, amyloid-beta aggregation, oxidative stress, and neuroinflammation. The authors further explore the workflow of their approach in screening literature and databases that is clearly demonstrated in the first Figure of this article. Additional illustrations are used to present early onset versus late onset, and not-defined cases of Alzheimer's disease along with extra information within the three Figures and one Table of this comprehensive review [1].The article by Tian et al., presents an interesting concept on the impact of elevated hemoglobin A1c (HbA1c) with increased risk for impaired cognition in a sex-dependent manner [2]. While the link between epigenetic modifications such as DNA methylation with that of HbA1c in type 1 diabetes has been reported [3], this study highlights the potential effects of higher levels of HbA1c in cognitive impairment. The authors explain how they have analyzed independent datasets from UK Biobank cohort with and without neuroimaging information, as the gene-outcome and gene-exposure groups, respectively. The authors describe certain types of analysis to include HbA1c levels, brain age gap, and fractional anisotropy. The detailed Figure 1 of this article captures the systemic analysis and study design of the authors and is complemented with two Tables and additional Figures to represent the results [2].A systemic review by Yuan et al., provides engaging contents on the link between epigenetics and depression [4]. This study benefits from the Web of Science core dataset that covers depression epigenetics published studies between 01/2002-to-06/2023. The authors used certain key words in their literature search, highlighting that the link between epigenetics and adolescent depression requires further investigations. The authors discuss the nature of depression, their approach and strategy in dataset screen and analysis, and present their results. Multiple Figures and Tables are The authors make a case by evidence from epidemiological studies to support such association, while acknowledging the need for molecular investigations. The authors explain how certain neurodegenerative diseases such as AD and Parkinson's disease (PD) may have epigenetic components. In the other hand the authors explain that individuals suffering from TBI may show short-term or long-term change of epigenetic mechanisms in the brain cells that could potentially be a link to the risk of experiencing AD, PD, and/or dementia. In this article, the authors discuss epigenetic modifications such as DNA methylation and histone post-translational modification(s) like acetylation. The authors further discuss the link between TBI pathology and epigenetic modifications, presenting their hypothesis that these could potentially be considered a risk for neurodegenerative disorders. They also provide insightful opinion on how certain interventions could prevent the development of neurodegenerative disorders. Their discussions of such therapeutic intermediation include the use of methyl donors, inhibition of chronic neuroinflammation, as well as the application of inhibitors for histone acetylation or methylation. * Comparison of Two Moldable Calcium Phosphate-Based Bone Graft Materials in a Noninstrumented Canine Interspinous Implantation Model.

Abstract:There is a continuing search for novel synthetic materials as an alternative to autologous bone grafting. Different technologies are explored to promote bone formation, which include the addition of BioGlass™ particles in calcium phosphate (CaP)-based materials and the use of surface modification in the form of submicron surface topographies. In this work, we aimed at comparing the bone formation in a noninstrumented canine interspinous model of moldable formulations of a submicron-surface structured tricalcium phosphate/alkylene oxide copolymer (CaP/AOC) or a tricalcium phosphate/BioGlass/collagen (CaP/BG/C) bone graft material. Intramuscular implantation was carried out as well to evaluate soft tissue responses. Eight mature male mongrel dogs underwent single-level, noninstrumented interspinous implantation, where the bone graft materials were implanted at either side of the spinous processes (L3-L4), with separation by the interspinous ligament ensuring comparison of both materials in each animal (n = 8 per material). The materials were also implanted in paraspinal muscle pouches. Animals were euthanized 12 weeks after surgery and the lumbar spines excised and intramuscular implants retrieved. Undecalcified sections were prepared for histological evaluation and histomorphometry was performed to quantify bone formation and material resorption. After 12 weeks, all submicron structured CaP/AOC implants showed abundant bone formation in the (L3-L4) interspinous space (20.8% ± 6.8%), whereas bone was not found in the CaP/BG/C implants (0% ± 0%). Intramuscularly, the CaP/AOC material triggered significant bone formation (12.0% ± 7.8%), whereas CaP/BG/C did not form any bone. In both the spinal and muscular sites, resorption of the CaP/AOC material was evident by a decrease in Feret diameter of the CaP granules as well as in their histological surface compared with the starting material, whereas CaP/BG/C material had a milder resorption. This study shows that a submicron-surface structured CaP/AOC bone graft material has superior bone-forming properties in both an interspinous implantation model and intramuscularly, as compared with a CaP/BG/C bone graft material.

Abstract:Neurodegenerative disorders are major health concerns globally, especially in aging societies. The exploration of brain epigenomes, which consist of multiple forms of DNA methylation and covalent histone modifications, offers new and unanticipated perspective into the mechanisms of aging and neurodegenerative diseases. Initially, chromatin defects in the brain were thought to be static abnormalities from early development associated with rare genetic syndromes. However, it is now evident that mutations and the dysregulation of the epigenetic machinery extend across a broader spectrum, encompassing adult-onset neurodegenerative diseases. Hence, it is crucial to develop methodologies that can enhance epigenetic research. Several approaches have been created to investigate alterations in epigenetics on a spectrum of scales—ranging from low to high—with a particular focus on detecting DNA methylation and histone modifications. This article explores the burgeoning realm of neuroepigenetics, emphasizing its role in enhancing our mechanistic comprehension of neurodegenerative disorders and elucidating the predominant techniques employed for detecting modifications in the epigenome. Additionally, we ponder the potential influence of these advancements on shaping future therapeutic approaches.

Abstract:Psychiatric disorders are complex multifactorial illnesses involving chronic alterations in neural circuit structure and function. While genetic factors are important in the etiology of disorders such as depression and addiction, relatively high rates of discordance among identical twins clearly indicate the importance of additional mechanisms. Environmental factors such as stress or prior drug exposure are known to play a role in the onset of these illnesses. Such exposure to environmental insults induces stable changes in gene expression, neural circuit function, and ultimately behavior, and these maladaptations appear distinct between developmental and adult exposures. Increasing evidence indicates that these sustained abnormalities are maintained by epigenetic modifications in specific brain regions. Indeed, transcriptional dysregulation and associated aberrant epigenetic regulation is a unifying theme in psychiatric disorders. Aspects of depression and addiction can be modeled in animals by inducing disease-like states through environmental manipulations (e.g., chronic stress, drug administration). Understanding how environmental factors recruit the epigenetic machinery in animal models reveals new insight into disease mechanisms in humans.

Abstract:Background: Psychiatric disorders are complex, multifactorial illnesses with a demonstrated biological component in their etiopathogenesis. Epigenetic modifications, through the modulation of DNA methylation, histone modifications and RNA interference, tune tissue-specific gene expression patterns and play a relevant role in the etiology of psychiatric illnesses. Objective: This review aims to discuss the epigenetic mechanisms involved in psychiatric disorders, their modulation by environmental factors and their interactions with genetic variants, in order to provide a comprehensive picture of their mutual crosstalk. Methods: In accordance with the PRISMA guidelines, systematic searches of Medline, EMBASE, PsycINFO, Web of Science, Scopus, and the Cochrane Library were conducted. Results: Exposure to environmental factors, such as poor socio-economic status, obstetric complications, migration, and early life stressors, may lead to stable changes in gene expression and neural circuit function, playing a role in the risk of psychiatric diseases. The most replicated genes involved by studies using different techniques are discussed. Increasing evidence indicates that these sustained abnormalities are maintained by epigenetic modifications in specific brain regions and they interact with genetic variants in determining the risk of psychiatric disorders. Conclusion: An increasing amount of evidence suggests that epigenetics plays a pivotal role in the etiopathogenesis of psychiatric disorders. New therapeutic approaches may work by reversing detrimental epigenetic changes that occurred during the lifespan.

Abstract:Key PointsStriking new evidence implicates the dysregulation of epigenetic mechanisms in neurodegenerative disorders and diseases.Histone acetylation is the modification that is best understood and most tightly associated with synaptic plasticity and memory formation. The dysregulation of histone acetylation has been linked to the memory impairments that are associated with neurodegenerative diseases.Emerging evidence indicates that changes in the DNA methylation status of synaptic plasticity-associated and memory-associated genes can be rapid and reversible. Members of the DNA methyltransferase (DNMT) family promote the rapid methylation and silencing of these genes, whereas the ten-eleven translocation (TET) proteins (TET1, TET2 and TET3) promote the demethylation and activation of genes that are crucial for synaptic plasticity, memory acquisition and storage.Although neurodegenerative disorders differ in their underlying causes and pathophysiologies, many involve the dysregulation of restrictive element 1-silencing transcription factor (REST), which acts via epigenetic mechanisms to regulate gene expression.Polycomb proteins orchestrate the epigenetic remodelling and silencing of genes that are involved in neuronal death, thereby enabling neurons to survive the hypoxia that is associated with ischaemic stroke.Emerging evidence implicates non-coding RNAs, such as microRNAs and long non-coding RNAs, in the epigenetic remodelling of target genes that are involved in neurodegeneration.Although the past decade has witnessed the development of new chromatin-modifying drugs and their advance into clinical trials in patients with brain disorders, there remains the persisting challenge of developing drugs that can penetrate the blood–brain barrier and ameliorate neurodegeneration and cognitive deficits with high specificity and minimal toxicity.

Abstract:Immune dysfunction is implicated in the aetiology of psychiatric, neurodevelopmental, and neurodegenerative conditions, but the issue of causality remains unclear impeding attempts to develop new interventions. We have tested evidence for causality for 735 immune response-related biomarkers on 7 neuropsychiatric conditions, using cutting-edge genomic causal inference methods (Mendelian randomization and genetic colocalization) applied to genomic data on protein and gene expression across blood and brain. We provide robust evidence of causality for 21 biomarkers, including two previously unreported ( , and ), confirming a role of both brain specific and systemic immune response in the pathogenesis of several neuropsychiatric conditions especially schizophrenia, Alzheimer’s disease, depression, and bipolar disorder. Furthermore, 18 of the identified biomarkers are therapeutically tractable, including , , and with drugs approved or in advanced clinical trials, offering an opportunity for repurposing existing drugs for neuropsychiatric indications.

Abstract:Overall Abstract: Interaction between genetic risks and environmental adversities such as childhood trauma may underlie the etiology of psychiatric disorders. Mechanisms underpinning these interactions are poorly understood. This panel will present new data supporting a model in which epigenetic modifications and increased oxidative stress lead to altered trajectories and patients’ stratification. Helen Fisher will introduce the field, how exposure to severe stress may have immediate as well as long-lasting damaging effects on learning, behaviour, and health, and has been implicated in the development of psychosis. She will present epigenome-wide analyses of poly-victimisation across childhood and adolescence, utilising data from the Environmental Risk (E-Risk) Longitudinal Twin Study, an epidemiological study of 2,232 children (1,116 twin pairs) born in 1994–1995 in England and Wales and followed to 18 years of age. Results related to associations with DNA methylation in peripheral blood at age 18 years, specific forms of victimization and candidate genes in the stress response will be discussed. Luis Alameda will show that childhood trauma (sexual, physical and emotional abuse) engages oxidative stress in a cohort of early psychosis patients (n=118). Patients with high peripheral oxidation status were found to have smaller hippocampal volumes and more severe clinical symptoms, while those with lower oxidation status evidenced a compensatory antioxidant regulation and better cognition. Linear discrimination analysis highlights blood oxidation status together with childhood trauma as markers for poorer psychopathological profile, allowing patients’ stratification. Oussama Kebir will present the first longitudinal prospective study of genomic DNA methylation during psychotic transition in help-seeking young individuals (14 converters, 25 non converters). Alterations in gene promoters and pathways relevant for psychosis, including oxidative stress regulation, axon guidance and inflammatory pathways were observed. In particularly, antioxidant defense gene GTSTM5 is differentially methylated through time and two other genes of GST family might be differentially methylated after conversion to psychosis. These findings suggest that conversion to psychosis may depend on the specific control of oxidative metabolism and regulation between these genes. Darina Czamara will report on how cord blood methylation is affected by genetic and prenatal environment. Analysis on SNP (G) and environmental (E) effects as well as on GxE and G+E were performed using Illumina’s Human Omni Express Exom as well as the 450k DNA methylation array in a cohort of 817 Finnish newborns. G and the combination of G and E explained DNA methylation best, environment alone was almost never the best predictor. Furthermore, epigenetic gestational age was associated with prenatal environment as well as with childhood psychiatric problem at age 3 indicating that it might be used as a potential biomarker. Overall, these new results suggest new biomarkers of environmental and genetic adversity that are related to pathogenic mechanisms, including epigenetic, oxidative stress and structural abnormalities, in epidemiological studies and psychiatric disorders. These biomarkers offer the potential for individualized early intervention and preventive strategies.

Abstract:Summary.The neurotrophin hypothesis proposes that repetitive neuronal activity enhances the expression, secretion and actions of neurotrophins to modify synaptic transmission and connectivity thereby providing a connection between neuronal activity and synaptic plasticity. Moreover, there is ample evidence that neurotrophins have numerous neuroprotective effects under pathological conditions, which might be important in particular for neurodegenerative diseases such as Alzheimer’ disease. Current research postulates that effects during brain development lead to defective neural connectivity and altered biochemical functioning resulting in cognitive, emotional and intentional dysfunction later in life. This implicates a possible role in most psychiatric diseases including affective and schizophrenic disorders. This hypothesis is mainly based on new experimental evidence showing that psychiatric disorders are associated with neuronal atrophy and cell loss, impairments of structural plasticity and cellular resilience due to neurodevelopmental disturbances and morphological abnormalities of the brain. Thus, the potential role of neurotrophins in psychiatric disorders has been studied in different ways. Animal studies indicate the involvement of neurotrophins in psychopharmacological therapies and they show that gene expression of cerebral neurotrophins is changed in animal models of several psychiatric disorders. Whether such alterations are causatively associated with increased neural plasticity, improved cognitive function and decreased depressive mood states remains to be elucidated in further studies including man (e.g. in postmortem studies of patients). Association studies tried to link different variants in genes coding for neurotrophins, they have not been conclusive however. They partially allow to separate different subgroups of patients with differing therapy response profiles or indicate an increased vulnerability for a specific disorder. Finally, neurotrophin serum changes have been observed in most psychiatric disorders. The question remains though whether these alterations represent primary-causal or secondary-reactive changes.In conclusion, the issue of neuroprotection and neurotrophins is recognised as an important new lead in the quest for a deeper understanding of psychiatric disorders and the mechanisms of action of psychopharmacological interventions.

Clinical potential of epigenetic and microRNA biomarkers in PTSD

Abstract:Molecular studies identifying alterations associated with PTSD have predominantly focused on candidate genes or conducted genome-wide analyses, often encountering issues with replicability. This review aims to identify robust bi-directional epigenetic and microRNA (miRNA) regulators focusing on their functional impacts on post-traumatic stress disorder (PTSD) and their utility in clinical diagnosis, whilst examining knowledge gaps in the existing research. A systematic search was conducted across multiple databases, including Web of Science, Scopus, Global Health (CABI), and PubMed, augmented by grey literature, yielding 3465 potential articles. Ultimately, 92 studies met the inclusion criteria and were analysed to pinpoint significant epigenetic changes with clinically relevant potential in PTSD. The selected studies explored histone modifications, CpG sites, single nucleotide polymorphisms (SNPs), and miRNA biomarkers. Specifically, nine studies examined epigenetic markers, detailing the influence of methylation on chromatin accessibility at histone positions H3K4, H3K9, and H3K36 within a PTSD context. Seventy-three studies investigated DNA methylation, identifying 20 hypermethylated and five hypomethylated CpG islands consistently observed in PTSD participants. Nineteen studies linked 88 SNPs to PTSD, with only one SNP replicated within these studies. Furthermore, sixteen studies focused on miRNAs, with findings indicating 194 downregulated and 24 upregulated miRNAs were associated with PTSD. Although there are epigenetic mechanisms that are significantly affected by PTSD, a granular deconstruction of these mechanisms elucidates the need to incorporate more nuanced approaches to identifying the factors that contribute to PTSD. Technological advances in diagnostic tools are driving the need to integrate detailed participant characteristics, trauma type, genetic susceptibilities, and best practices for robust reporting. This comprehensive approach will be crucial for enhancing the translational potential of PTSD research for clinical application.

Abstract:Neurological and psychiatric disorders have overlapped phenotypic profiles, but the underlying tissue-specific functional processes remain largely unknown. In this study, we explore the shared tissue-specificity among 14 neuropsychiatric disorders through the disrupted long-range gene regulations by GWAS-identified regulatory SNPs. Through Hi-C interactions, averagely 38.0% and 17.2% of the intergenic regulatory SNPs can be linked to target protein-coding genes in brain and non-brain tissues, respectively. Interestingly, while the regulatory target genes in the brain tend to enrich in nervous system development related processes, those in the non-brain tissues are inclined to interfere with synapse and neuroinflammation related processes. Compared to psychiatric disorders, neurological disorders present more prominently the neuroinflammatory processes in both brain and non-brain tissues, indicating an intrinsic difference in mechanisms. Through tissue-specific gene regulatory networks, we then constructed disorder similarity networks in two brain and three non-brain tissues, highlighting both known disorder clusters (e.g. the neurodevelopmental disorders) and unexpected disorder clusters (e.g. Parkinson’s disease is consistently grouped with psychiatric disorders). We showcase the potential pharmaceutical applications of the small bowel and its disorder clusters, illustrated by the known drug targets NR1I3 and NFACT1, and their small bowel-specific regulatory modules. In conclusion, disrupted long-range gene regulations in both brain and non-brain tissues contribute to the similarity among distinct clusters of neuropsychiatric disorders, and the tissue-specifically shared functions and regulators for disease clusters may provide insights for future therapeutic investigations.

Abstract:Psychiatric disorders are complex multifactorial disorders involving chronic alterations in neural circuit structure and function. While genetic factors play a role in the etiology of disorders such as depression, addiction, and schizophrenia, relatively high rates of discordance among identical twins clearly point to the importance of additional factors. Environmental factors, such as stress, play a major role in the psychiatric disorders by inducing stable changes in gene expression, neural circuit function, and ultimately behavior. Insults at the developmental stage and in adulthood appear to induce distinct maladaptations. Increasing evidence indicates that these sustained abnormalities are maintained by epigenetic modifications in specific brain regions. Indeed, transcriptional dysregulation and associated aberrant epigenetic regulation is a unifying theme in psychiatric disorders. Aspects of depression can be modeled in animals by inducing disease-like states through environmental manipulations, and these studies can provide a more general understanding of epigenetic mechanisms in psychiatric disorders. Understanding how environmental factors recruit the epigenetic machinery in animal models is providing new insights into disease mechanisms in humans.

Abstract:DNA methylation, which is modulated by both genetic factors and environmental exposures, may offer a unique opportunity to discover novel biomarkers of disease-related brain phenotypes, even when measured in other tissues than brain, such as blood. A few studies of small sample sizes have revealed associations between blood DNA methylation and neuropsychopathology, however, large-scale epigenome-wide association studies (EWAS) are needed to investigate the utility of DNA methylation profiling as a peripheral marker for the brain. Here, in an analysis of eleven international cohorts, totalling 3337 individuals, we report epigenome-wide meta-analyses of blood DNA methylation with volumes of the hippocampus, thalamus and nucleus accumbens (NAcc)-three subcortical regions selected for their associations with disease and heritability and volumetric variability. Analyses of individual CpGs revealed genome-wide significant associations with hippocampal volume at two loci. No significant associations were found for analyses of thalamus and nucleus accumbens volumes. Cluster-based analyses revealed additional differentially methylated regions (DMRs) associated with hippocampal volume. DNA methylation at these loci affected expression of proximal genes involved in learning and memory, stem cell maintenance and differentiation, fatty acid metabolism and type-2 diabetes. These DNA methylation marks, their interaction with genetic variants and their impact on gene expression offer new insights into the relationship between epigenetic variation and brain structure and may provide the basis for biomarker discovery in neurodegeneration and neuropsychiatric conditions.

Abstract:Recent genomics and epigenetic advances have empowered the exploration of DNA/RNA methylation and histone modifications crucial for gene expression in response to stress, aging and disease. Interest in understanding neuronal plasticity's epigenetic mechanisms, influencing brain rewiring amid development, aging and neurodegenerative disorders, continues to grow. Histone acetylation dysregulation, a commonality in diverse brain disorders, has become a therapeutic focus. Histone acetyltransferases and histone deacetylases have emerged as promising targets for neurodegenerative disorder treatment. This review delves into histone acetylation regulation, potential therapies and future perspectives for disorders like Alzheimer's, Parkinson's and Huntington's. Exploring genetic–environmental interplay through models and studies reveals molecular changes, behavioral insights and early intervention possibilities targeting the epigenome in at-risk individuals. Scientists have made progress in understanding how our genes and their chemical modifications play a role in how our brains respond to stress, age and diseases. They are particularly interested in how these processes affect the flexibility of our brain circuits, which is important during growth and aging and in conditions like Alzheimer's and Parkinson's. One key area of focus is controlling a specific chemical change called histone acetylation, which tends to go awry in various brain disorders. Researchers are looking at potential treatments that target specific proteins related to this process. This review explores how these chemical changes might be regulated, potential treatments and the future for disorders like Alzheimer's, Parkinson's and Huntington's. Abstract:Advances achieved with molecular biology and genomics technologies have permitted investigators to discover epigenetic mechanisms, such as DNA methylation and histone posttranslational modifications, which are critical for gene expression in almost all tissues and in brain health and disease. These advances have influenced much interest in understanding the dysregulation of epigenetic mechanisms in neurodegenerative disorders. Although these disorders diverge in their fundamental causes and pathophysiology, several involve the dysregulation of histone methylation-mediated gene expression. Interestingly, epigenetic remodeling via histone methylation in specific brain regions has been suggested to play a critical function in the neurobiology of psychiatric disorders, including that related to neurodegenerative diseases. Prominently, epigenetic dysregulation currently brings considerable interest as an essential player in neurodegenerative disorders, such as Alzheimer’s disease (AD), Parkinson’s disease (PD), Huntington’s disease (HD), Amyotrophic lateral sclerosis (ALS) and drugs of abuse, including alcohol abuse disorder, where it may facilitate connections between genetic and environmental risk factors or directly influence disease-specific pathological factors. We have discussed the current state of histone methylation, therapeutic strategies, and future perspectives for these disorders. While not somatically heritable, the enzymes responsible for histone methylation regulation, such as histone methyltransferases and demethylases in neurons, are dynamic and reversible. They have become promising potential therapeutic targets to treat or prevent several neurodegenerative disorders. These findings, along with clinical data, may provide links between molecular-level changes and behavioral differences and provide novel avenues through which the epigenome may be targeted early on in people at risk for neurodegenerative disorders.

Abstract:In this paper, we discuss the application of epigenetics to research in child and adolescent mental health. We look back at lessons learned, highlight current developments in the field and point to areas of priority for future research. We also reflect on why epigenetic research on child and adolescent mental health currently lags behind other areas of epigenetic research and what we can do to overcome existing barriers, before concluding with a view on potential translational applications that could emerge as the field matures. Background Epigenetic processes are fast emerging as a promising molecular system in the search for both biomarkers and mechanisms underlying human health and disease risk, including psychopathology. Methods In this review, we discuss the application of epigenetics (specifically DNA methylation) to research in child and adolescent mental health, with a focus on the use of developmentally sensitive datasets, such as prospective, population‐based cohorts. We look back at lessons learned to date, highlight current developments in the field and areas of priority for future research. We also reflect on why epigenetic research on child and adolescent mental health currently lags behind other areas of epigenetic research and what we can do to overcome existing barriers. Results To move the field forward, we advocate for the need of large‐scale, harmonized, collaborative efforts that explicitly account for the time‐varying nature of epigenetic and mental health data across development. Conclusion We conclude with a perspective on what the future may hold in terms of translational applications as more robust signals emerge from epigenetic research on child and adolescent mental health.

Abstract:In oncology, comprehensive omics and functional enrichment studies have led to an extensive profiling of (epi)genetic and neurobiological alterations that can be mapped onto a single tumor's clinical phenotype and divergent clinical phenotypes expressing common pathophysiological pathways. Consequently, molecular pathway-based therapeutic interventions for different cancer typologies, namely tumor type- and site-agnostic treatments, have been developed, encouraging the real-world implementation of a paradigm shift in medicine. Given the breakthrough nature of the new-generation translational research and drug development in oncology, there is an increasing rationale to transfertilize this blueprint to other medical fields, including psychiatry and neurology. In order to illustrate the emerging paradigm shift in neuroscience, we provide a state-of-the-art review of translational studies on the β-site amyloid precursor protein cleaving enzyme (BACE) and its most studied downstream effector, neuregulin, which are molecular orchestrators of distinct biological pathways involved in several neurological and psychiatric diseases. This body of data aligns with the evidence of a shared genetic/biological architecture among Alzheimer's disease, schizoaffective disorder, and autism spectrum disorders. To facilitate a forward-looking discussion about a potential first step towards the adoption of biological pathway-based, clinical symptom-agnostic, categorization models in clinical neurology and psychiatry for precision medicine solutions, we engage in a speculative intellectual exercise gravitating around BACE-related science, which is used as a paradigmatic case here. We draw a perspective whereby pathway-based therapeutic strategies could be catalyzed by highthroughput techniques embedded in systems-scaled biology, neuroscience, and pharmacology approaches that will help overcome the constraints of traditional descriptive clinical symptom and syndrome-focused constructs in neurology and psychiatry.
