Abstract:Objectives: The aim of this study is to examine resolution, identification, and characterization of forced degradation products of netarsudil by liquid chromatography-tandem mass spectrometry by validating a simple and sensitive high-performance liquid chromatography method for the resolution, identification, and quantification of two process-related impurities in netarsudil. Materials and methods: Chromatographic separation was accomplished on a ZORBAX Eclipse XDB C18 (250 x 4.6 mm; 5 µ id) column at room temperature as the stationary phase and 257 nm as the detector wavelength with the mobile phase consisting of acetonitrile, methanol, and pH 4.6 phosphate buffer in 45:35:20 (v/v) at 1.0 mL/min flow rate in isocratic elution. Results: The method reported very sensitive detection limits of 0.008 µg/mL for impurity 1 and 0.003 µg/mL for impurity 1. The method produces a calibration curve linear in the concentration level of 25-200 for netarsudil and 0.025-0.2 µg/mL for impurities. The proposed method gives acceptable results for other validation parameters such as accuracy, precision, ruggedness, and robustness. The drug was subjected to various stress conditions such as acid, base, peroxide, and thermal and ultraviolet light to investigate the stability-indicating ability of the method. Considerable degradation was observed in stress studies, and the degradation products were well resolved from process-related impurities. The characterization of degradation products was performed on the basis of collision-induced dissociation mass spectral data, and the possible structures of the six degradation compounds of netarsudil were proposed. Conclusion: The outcomes of other validation studies were likewise satisfactory and proven adequate for the regular analysis of netarsudil and its process-related impurities in bulk drug and pharmaceutical dosage forms and can also be applied for the evaluation of the stress degradation mechanism of netarsudil.
Effects of project versus parallel groups on social interaction and affective responses in senior citizens.

Abstract:Using activity analysis, occupational therapists structure group activities to meet the needs and goals of group members. This study compared the effects of project group structure (subjects participate together on a common task) with parallel group structure (subjects each have a task) in a sample of healthy seniors (32 women and 9 men). Subjects randomly assigned to either experimental condition made creative and imitative collages in a counter-balanced order. Time-sampled observations revealed that project group subjects talked and looked at others significantly more than parallel group subjects. On-task and laughing behaviors were also measured, but results were inconclusive. Project group subjects rated their activities higher on the action factor of the Osgood semantic differential (OSD) than did parallel group subjects, but there were no other differences on the OSD or on MacKenzie's Group Climate Questionnaire. Nor were there significant main effects between the creative collage and the imitative collage. Results are discussed both in terms of group task structure as an occupational therapy tool and in terms of methodologies for future activity analysis research.

Abstract:The present study focused on the forced degradation behavior of sertraline hydrochloride (SRT), selective serotonin reuptake inhibitor (SSRI). The drug was exposed to different stressed conditions (hydrolytic, oxidative, thermal and photolytic) according to ICH Q1A (R2) guidelines. The study revealed that SRT was stable in hydrolytic (acidic, basic and neutral) and thermal degradation conditions. In contrast, five degradation products (DPs) were formed under oxidative and photolytic degradation conditions. The chromatographic separation of drug substance and its DPs was achieved on an Acquity HSS T3 column (100 × 2.1 mm, 1.7 μ) using 0.1% formic acid and acetonitrile as the mobile phase in gradient mode using a UHPLC-DAD system. The DPs were identified and characterized by high-resolution LC/MS and LC/MS/MS in ESI positive mode. Two DPs (DP-I and DP-II) were formed when SRT was exposed to oxidative degradation conditions. Three DPs formed (DP III-V) when exposed to photolytic degradation conditions. All the five major DPs were isolated using Preparative HPLC. The structures of major DPs formed were further confirmed using NMR technique (1D and 2D). The proposed mechanism for the formation of the SRT DPs via the photolytic/oxidative stress degradation pathway are discussed and outlined.

Abstract:INTRODUCTION:Chinese herbal medicines contain complex ingredients that are unstable during the manufacturing process. Current off-line methods are usually time-consuming and inaccurate.OBJECTIVE:The aim of this study was to develop a continuous-flow-based on-line spectroscopic system for monitoring the degradation of unstable compounds in Chinese herbal medicines.METHODOLOGY:An on-line spectroscopic analysis system in the evaluation of the degradation of unstable compounds was implemented, using salvianolic acid B from Danshen (Radix Salviae Miltiorrhizae) as a model compound. Resolution of the mixture spectra obtained during the reaction was performed with multivariate curve resolution (MCR), and the pure spectra and kinetic profiles of reactant species were reconstructed.RESULTS:The degradation process of salvianolic acid B is probably a consecutive two-step reaction of the form A → B → C, where A is salvianolic acid B, and B and C are the intermediate and final products, respectively. The intermediate and final products were suggested to be a mixture of lithospermic acid and danshensu [3-(3, 4-dihydroxyphenyl) lactic acid], and a mixture of danshensu and salvianolic acid A, respectively.CONCLUSION:The findings suggested that the combined use of on-line spectroscopic analysis and MCR might be a cheap, readily available, sensitive tool to monitor the transformation of unstable components of Chinese herbal medicines.

Abstract:The current work identifies and characterises the stress degradation products of the anti-hypertensive drug sacubitril. The stability profile of sacubitril was evaluated under several stress settings including hydrolytic, oxidative, thermal and photolytic conditions in accordance with ICH recommendations. The degradation products of sacubitril were separated using reverse phase liquid chromatography and structural characterization was carried out using HRMS. The chromatographic separation was carried out by Reverse Phase-High Performance Liquid Chromatography (RP-HPLC) using a Fortis C18 column (150 mm X 4.6 mm; 5μm particle size) with mobile phase A (consisting of 0.1% TFA in Milli-Q water) and mobile phase B (100% Acetonitrile) in gradient mode. With a sample injection volume of 10μL, the mobile phase flow rate was 0.7 mL/min. A quadrupole time-of-flight (Q-TOF) mass spectrometer was used for the LC-MS analysis. Sacubitril is relatively stable in oxidative, thermal and photolytic conditions; however, considerable degradation was observed in acid, base and neutral hydrolysis. Three degradation products were identified and the structural characterization of these impurities was performed using the HRMS data and mass fragmentation. The proposed RP-HPLC method was robust, precise and specific in determining the impurity profile of sacubitril under different stress conditions.

Abstract:The aim of this study is identification, isolation, structural characterization and qualification of degradation products DP-1, DP-2, DP-3 and DP-4. These impurities are major acid hydrolysis degradation products identified i n Osimertinib M esylate ( OSM) Tablets. T he O SM i s o bserved s table u nder thermal, photolytic and neutral conditions. The degradation of drug is observed under acidic, basic and oxidative conditions. Basic and oxidative degradation products already been reported, however, four novel degradation products are formed in acid hydrolysis which are not reported in the literature. Therefore, Using HPLC/PDA gradient program with waters X-bridge (250 mm × 4.6 mm, 5 µm) C18 column, the four acid hydrolysis degradants were identified and further characterized using UHPLC-IMS-QTOF-MS (ion mobility quadrupole time-of-flight m ass spectrometer) a nd N MR. T he s tructures o f D P-1, D P-2, D P-3 a nd D P-4 w ere further elucidated as hydroxy OSM Impurity, Des-acryl OSM impurity, Chloro OSM impurity and OSM dimer impurity, respectively. The structural characterization was followed by qualification of Impurities DP-1, DP-2, DP-3 and DP-4 using several different in-silico methodologies. From the results obtained, it can be concluded that no new structural alerts have been generated for DP-1 and DP-4 relative to the parent compound OSM. However, the degradation products, DP-2 and DP-3 shows structural alert positive for genotoxicity, which were both potential genotoxic substances. The current study presents separation, isolation, characterization and in-silico toxicity study for major acid degradants of OSM. The OSM generates DP-1 around 3.1%, DP-2 around 14.1%, DP-3 around 18.3% and DP-4 around 2.8% under acid hydrolysis on HPLC. The stress study was carried out in order to properly define the quality specifications for OSM drug products.

Abstract:This work presents the development and optimization of the stability-indicating method for solifenacin succinate and its main degradation product, formed through oxidation, using the high-performance liquid chromatography technique with diode array detection using the quality by design approach. In this work, a forced degradation study was performed to evaluate the potential degradation profile of the active ingredient, solifenacin succinate. Here, multiple optimizations of the extraction procedure are proposed. Often, the prescribed procedure focuses on recovering the active pharmacological ingredient while neglecting the recovery of impurities, a critical step in the development of stability-indicating methods. In this step, an experimental design of mixtures using the simplex model was used in order to optimize the recovery of the active ingredient and its main degradation product. Mixture solvents were evaluated by combining water (0–40%), acetonitrile (0–100%), and ethanol (0–100%) using the ultrasound-assisted extraction technique, with evaluation times set at 10 and 30 min. The highest values were obtained using a solvent mixture of acetonitrile and ethanol in a ratio of 90 : 10 (v/v), along with a 30-minute ultrasound-assisted extraction. These recovery values exceeded those of the control condition. The development of the chromatographic method was carried out from a screening step based on the conditions reported in the literature. Subsequently, it was submitted to an optimization step where the possible analysis time was significantly reduced without loss of chromatographic efficiency. The optimal condition obtained was on the Xbridge chromatographic column with dimensions of 250 × 4.6 mm and 5 μm particle size. The system operated in gradient mode using a mobile phase containing a phosphate buffer (pH 6.7), with 0.5% diethylamine and acetonitrile in a ratio of 85 : 15 (v/v). Another mobile phase contained a phosphate buffer (pH 6.7) with 0.5% diethylamine and acetonitrile in a ratio of 20 : 80 (v/v). The temperature of the chromatographic column was maintained at 40°C, with a mobile phase flow rate of 1.1 mL/min and an injection volume of 10 μL in a gradient elution mode. The developed method was applied to evaluate the degradation profile of commercially purchased drugs.

Abstract:Abstract Background In the dapagliflozin (DPF) synthesis, 5-Bromo-2-chlorobenzoic acid (5-BC impurity) and 4-Bromo-1-chloro-2-(4-ethoxybenzyl) benzene (4-BC impurity) are used as starting and reagent sources, respectively. The presence of 5-BC and 4-BC impurities in DPF could potentially affect the effectiveness of the final DPF product. The purpose of this investigation was to develop a stability indicating HPLC methodology for the separation of DPF, its process-related impurities and degradants. The method of analysis was developed on Xbridge Phenyl C18 column of dimensions, 250 × 4.6 mm, 5 μm with gradient elution using mobile phase made up of 0.05% aqueous trifluoroacetic acid and acetonitrile (AcN). Results The method proposed indicates a good linearity ( R 2 = 0.9996 and 0.9993), good system precision (RSD ≤ 2%), good method precision (RSD ≤ 2%), accuracy (50–150%), LOD (0.000053 ppm and 0.0000165 ppm) and LOQ (0.00016 ppm and 0.00005 ppm) for 4-BC impurity and 5-BC impurity, respectively. LC–MS was used to detect and characterise degradants that were obtained in acid and base condition were identified and characterised. A comparison of the fragmentation pattern of the [M + H] + ions of DPF and its degradation products revealed the most likely processes for the generation of degradation products. Conclusion DPF sample quality can be evaluated using the suggested method for the presence of 4-BC impurity, 5-BC impurity and 2-(3-(4-ethoxy benzyl)-4-chloro phenyl)-tetrahydro-6-(hydroxy methyl)-2H-pyran-3,4,5-triol.
Isolation and characterization of novel degradation products in Fenoverine by LC-QTOF-MS/MS, LC-MS n and 2D-NMR

Abstract:This study investigates the degradation profile of Fenoverine (FEN), an antispasmodic drug, following the International Conference on Harmonization (ICH) Q1A (R2) guidelines. FEN is stable in the presence of light, humidity, and thermal stress; however, it is highly susceptible to oxidative stress (0.1% H2O2), alkaline stress (0.1 N NaOH), and low susceptibility to acid (1N HCl). Seven degradation products (DPs) were identified using a mass-compatible ultra-high-performance liquid chromatography (UPLC) method: five oxidative degradation products (FOD1 to FOD5) and two alkaline degradation products (FAD1 and FAD2). All DPs, except FOD4, were isolated through preparative HPLC and structurally characterized using high-resolution mass fragmentation, multistage fragmentation, and multidimensional nuclear magnetic resonance (NMR) spectroscopy. Notably, FOD3, FOD4, and FOD5 were identified as isomeric oxidative degradation products (M + 16 Da). Atmospheric pressure chemical ionization (APCI) mass spectrometry and NMR studies pinpointed the oxidation sites, distinguishing FOD3 as FEN's N-oxide, FOD4 as its hydroxyl product, and FOD5 as the sulfoxide. To the best of our knowledge, the discussion of FOD1 to FOD5 and FAD1 in the context of this study appears to be unique and not documented in prior literature. These degradation products were successfully separated from FEN using an Acquity UPLC BEH300 C18 column with 1.7 μm particles as the stationary phase and a mobile phase of 10 mM ammonium acetate in water (pH adjusted to 9.0), with acetonitrile and methanol (80:20, v/v) as co-solvents. In-silico toxicity predictions were conducted, which revealed no structural alerts indicative of mutagenicity for any of the seven degradation products (FOD1 to FOD5, FAD1, and FAD2).

Abstract:Duvelisib (DUV) was first approved globally in 2018. An extensive literature search revealed that the differential role of a potential degradation medium in altering the shelf-life of DUV due to its exposure during storage has not been identified till date. Moreover, its degradation impurities and degradation mechanism are not known. In addition, no analytical method has been reported for the quantification of DUV in the presence of its degradation impurities. Therefore, the aim of this study was to identify the impact of different potential degradation media on the stability of DUV, establish the degradation mechanism, and identify its major degradation impurities. The aim was also to establish a stability-indicating analytical method for the quantification of DUV in the presence of its degradation impurities. This study is the first to report the structure of degradation impurities and the step-by-step degradation mechanism of DUV. This information will be useful for the scientific community and manufacturers in optimizing the formulation parameters and/or storage conditions. The validated method can be employed for analysis of stability study and routine quality control samples of newer DUV formulations in pharmaceutical industries. The identified impurities may serve as impurity standards for specifying their limits in the drug after required qualification studies.

Abstract:Rationale Azelastine HCl is a second‐generation H 1 ‐receptor antagonist approved by the US Food and Drug Administration (US FDA) for treating seasonal allergic rhinitis and non‐allergic vasomotor rhinitis. This study encompasses the validation of a liquid chromatography‐ultra violet photo diode array (LC‐UV/PDA) method for the drug and its extension to liquid chromatography/quadrupole time‐of‐flight mass spectrometry (LC‐Q/TOF‐MS) studies for identification and characterization of various stress degradation products of the drug. Methods Stress degradation of azelastine HCl was undertaken under the International Council for Harmonisation of Technical Requirements for Registration of Pharmaceuticals for Human Use (ICH) prescribed conditions of hydrolytic, photolytic, oxidative, and thermal stress. The degraded drug solutions were analyzed using Ultra Performance Liquid Chromatography (UPLC) employing a C18 (100 × 4.6 mm; 2.6 μ, Kinetex) column by isocratic elution. Detection wavelength was 241 nm. The degradation products were identified and characterized using UPLC‐MS/TOF studies, and an attempt was made to isolate one of the degradation products by solvent extraction. Results The drug was found to significantly degrade under acidic/alkaline/neutral photolytic, oxidative, and alkaline hydrolytic conditions. Six degradation products ( I–VI ) were identified through LC‐Q/TOF‐MS studies that were adequately resolved from the drug with the developed UPLC method. All degradation products ( I–VI ) were ionized in the total ion chromatogram (TIC) in the LC‐MS studies, and these were identified and characterized, and the degradation pathway of the drug was postulated. One of the oxidation products isolated from the degraded drug solution was characterized through differential scanning calorimetry, Fourier‐transform infrared spectroscopy, and nuclear magnetic resonance spectral data. Conclusions Six degradation products generated from stress degradation studies on azelastine HCl were adequately resolved through LC‐UV/PDA studies followed by method validation. These were successfully identified and characterized through LC‐Q/TOF‐MS studies, and the degradation pathways for the generation of these products from the drug have been postulated.

Development of a Stability-Indicating Purity Method for Ubrogepant through Stress Degradation Analysis, Extraction, and Characterization of Unidentified Degradation Products Using Flash Chromatography, NMR, IR, and LC-MS

Abstract:Background: Ubrogepant is a prescription medication used to prevent migraine headaches. It is currently available in tablet form. Objective: The goal of this work is to investigate the drug degradation profile of Ubrogepant, as well as to isolate and characterize undiscovered Ubrogepant degradation products by utilizing LC-MS, NMR, and IR spectroscopic analytical techniques. Furthermore, to develop a high resolution, sensitive, stability-indicating analytical approach for detecting and quantifying Ubrogepant degradation products in its pharmaceutical formulation. Method: To identify and quantify the degradation products of Ubrogepant in pharmaceutical products, a novel gradient reverse-phase HPLC (RP-HPLC) technique with a PDA detector was developed by utilizing a C18 stationary phase column. The eluent comprised a mixture of acetonitrile and water with 0.1% v/v ortho-phosphoric acid. To establish the intrinsic stability of Ubrogepant pharmaceutical product, it was stress tested under various degrading circumstances, including water, alkaline, acid hydrolysis, photolytic, oxidative, and thermal. Flash chromatography was used to isolate the two major degradants, and the structures were determined using NMR (1H , 13C, DEPT 135), IR, and LC-MS methods. Results: The Ubrogepant medication was relatively more degradable in alkaline and acidic circumstances, and two unique degradation products were discovered. Based on spectroscopic and chromatographic evidence, it was conclusively demonstrated that these unique compounds were Ubrogepant hydrolysis products. All degradation products have been separated with a resolution greater than 2.0. The peak purity data shown that the Ubrogepant peak in all of the stress samples examined was pure. Under all stress environments, Ubrogepant achieved a minimum mass balance of 95%. The validated developed approach was sensitive enough to quantify Ubrogepant degradation products at 0.03% of the Ubrogepant test concentration. Conclusions: The proposed method was found to be stability-indicating since it fits all of the regulatory authorities' typical requirements. This method is highly efficient for detecting and quantifying impurities in Ubrogepant drug substances and drug products in QC laboratories. Highlights: Two new degradation products of Ubrogepant were successfully extracted and characterized using NMR, IR, and LC-MS spectroscopic methods. The proposed HPLC method can accurately quantify the degradation products of Ubrogepant in pharmaceutical products and is sensitive enough to detect degradation products of Ubrogepant as low as 0.17 µg/mL.

Abstract:Methods for determination of bisoprolol and related substances mostly use UV detection and a phosphate buffer and are not suitable for MS detection. In this study, LC?UV?MS/MS for separation and characterization of bisoprolol related substances was developed, validated and applied for studying the degradation products of bisoprolol when exposed to hydrolytic stress, heat and light. The method uses a C18 column, formic acid in water and acetonitrile as mobile phases, gradient elution and UV and MS detection. Forced degradation revealed that acid hydrolysis produces the most intensive transformation of bisoprolol to its impurity A, along with impurities L and D. Alkaline hydrolysis produced impurities A, L, Q, G and K; oxidative and thermal degradation produced impurities A, L and K, while photodegradation produced impurities A, L, G and K, all characterized by their mass spectral data. The developed method using two detection systems was demonstrated as efficient since mass spectra allowed identification of the related substances of bisoprolol and quantification was possible using absorbance measurements at 270 nm. The obtained results will fill in the lack of data on the fragmentation patterns of bisoprolol and related substances that could be used by researchers and practitioners in research and quality control laboratories.

Abstract:This work presented the integration of the Analytical Quality by Design concept in developing a UPLC method for the quantification of saponins and phenolic acids in two Compound Danshen Dripping Pills intermediates of a concentrated extract and a supernatant. Prior knowledge and systematic risk analysis were conducted to identify potential critical method parameters (CMPs) that affected critical quality attributes (CQAs). A definitive screening design was employed to investigate the quantitative relationship among the intermediate type, CPPs, and CQAs. Intermediate type was added in the experimental design table as a categorical variable. After the models were built, the method operable design region (MODR) was calculated with an exhaustive search-Monte Carlo method. The CPP combinations with a 95 % or higher probability of reaching the preset standard were collected in MODR. The MODR robustness test was conducted with a Plackett-Burman design. The separation of analytes was achieved with a Waters Acquity HSS T3 (100 x 2.1 mm, 1.8 mu m) column at 30 degrees C, using 0.05 % phosphoric acid and acetonitrile as the mobile phases in gradient elution at a flow rate of 0.40 mL/min. The elution was monitored at UV detection wavelengths of 280 nm and 203 nm. The optimized method was also validated. Good robustness, accuracy, precision, stability and repeatability, and linear regression (R > 0.9997) were obtained. All recoveries of the 9 compounds ranged from 97.8 % to 101.7 %, with an RSD of less than 3.49 %. Nine batches of Danshensu, protocatechualdehyde, rosmarinic acid, salvianolic acid B, salvianolic acid A, notoginsenoside R1, ginsenoside Rg1, ginsenoside Re, and ginsenoside Rb1 were analysed. Danshensu was found to be the compound with the highest content.

Abstract:Abstract Background Flibanserin (FLB) was firstly synthesized as an antidepressant drug; however, due to its enhancing effects on sexual activity, it was approved for treatment of hypoactive sexual desire disorder in women in 2015. Objective The aim of this study was to develop a new and fully validated HPLC method for analysis of FLB in pharmaceutical formulations besides its degradation products, and identification of possible formation mechanisms by using HPLC-DAD-ESI-IT-TOF-MSn. Methods The HPLC separation was achieved in an Supelco Ascentis® Express series phenyl hexyl column (100 × 4.6 mm, ID 2.7μm). The mobile phase was acetonitrile-ammonium acetate solution (50:50, v/v, 10 mM, pH 5.4) mixture, which was pumped at the rate of 0.5 mL/min. Chromatography, detection and structural identification was performed by using LCMS-IT-TOF instrument (Shimadzu, Japan). Results 1-(2-(4-(3-hydroxy-5-(trifluoromethyl)phenyl)piperazine-1-yl)ethyl)-1,3-dihydro-2H-benzo[d]imidazol-2-one is proposed as a novel degradation product, with a mass of 407.1695, and a formula of C20H21F3N4O2 with a margin of error about 0.001 ppm. The developed method is applicable with 98% accuracy within 2.5-50.0 μg/mL range. The LOD and LOQ was about 500 ng/mL and 1.50 μg/mL, respectively. The transferability and variation between laboratories were tested by inter-laboratory comparison and evaluated with one-way analysis of variance. Conclusions A novel FLB degradation product, which was produced under oxidative forced degradation condition was observed and identified for the first time; in addition, the formation kinetics of the degradation product, besides decomposition of FLB was studied. Furthermore, an inter-laboratory comparison was carried out and application of the proposed method on pseudo Addyi® sample was tested using both instrument configurations. Highlights A novel stability indicating assay method was developed and fully validated according to ICH (Q2)R1 for the analysis of FLB in the pharmaceutical preparations. A new degradation product was identified in the oxidative forced degradation condition and characterized using HPLC–DAD–ESI-IT-TOF-MS3. Moreover, the possible mechanism and the formation kinetic of the degradation product were revealed. In addition, the developed method was transferred to another LC-PDA instrument for inter-laboratory comparison. Finally, the current method was applied to pseudo formulation of Addy® in both instruments and ANOVA was applied for evaluation.

Abstract:Notoginsenoside Fc, a protopanaxadiol-type saponin, shows multi-pharmacological activities. Chemical stability evaluation plays a crucial role in drug development. In this study, the forced degradation behavior of Notoginsenoside Fc was investigated under hydrolytic and oxidative conditions. A specific ultra high performance liquid chromatography with quadrupole time-of-flight mass spectrometry was developed for the separation, identification, and characterization of the degradation products of Notoginsenoside Fc. Fifty potential degradation products were formed via deglycosylation, dehydration, hydration, isomerization, side-chain cleaving, oxidation, and superoxidation. Notoginsenoside Fc was subjected to different pH solutions, temperatures, and time periods to assess its stability. A sensitive ultra high performance liquid chromatography-tandem mass spectrometry was developed for the quantification of Notoginsenoside Fc, notoginsenoside ST-4, notoginsenoside Ft1, and relative quantification of notoginsenoside Ft2, 20(R)-notoginsenoside Ft2, notoginsenoside SFt3, and notoginsenoside SFt4. The assay was linear over the concentration range (R-2 > 0.997) with the lowest limit of quantification of 0.02 mu g/mL for Notoginsenoside Fc, Notoginsenoside ST-4, and Notoginsenoside Ft1. The intra-day precision, inter-day precision, and accuracy of the three analytes were within accepted levels. The degradation kinetics of Notoginsenoside Fc in pH 1 and 3 solutions fits to first-and second-order kinetics, respectively. The degradation of Notoginsenoside Fc is pH-, temperature-, and time-dependent.

Abstract:Elagolix sodium (ELS) is a marketed product using to release moderate to severe endometriosis-associated pain. It contains functional groups such as carboxyl group, secondary amino group, 2,4-dioxo pyrimidinyl and several benzyl or benzyl-like position hydrogen atom that are susceptive to occur stress degradation. Forced degradation studies of ELS reveal different degradation profiles of the drug substance which are conducted under photo, thermal, acidic, neutral, alkaline and hydrogen peroxide oxidative conditions under the direction of the ICH guidances. With structural elucidation of LC-PDA/UV-MS n and NMR, the degradants were identified, and seven new degradants are reported in this study. It is confirmed that most of the degradation behaviors of ELS are related to the carboxyl group and secondary amino group in the 3-carboxyl propylamine side chain. Under the oxidative condition using hydrogen peroxide as the oxidant, the secondary amine was oxidized to form an N -hydrogen amine degradant and two further degradants of amine and carbonyl analogs were generated. Under the alkaline degradation condition, the ELS is proven to be stable and no obvious degradants are produced. On the other hand, under the acidic and neutral degradation condition, the 2,4-dioxo pyrimidinyl core of elagolix sodium is stable but the carboxyl group and secondary amine will occur ring cyclization to form the δ-lactam analogs of elagolix sodium. The plausible mechanisms for the degradation of acidic, thermal, photo-degradative and hydrogen peroxide mediated oxidative of elagolix sodium are proposed. It is worth to note that DP-3~4 are the potential degradants which are only found in the solution degradation and are not the real impurities of elagolix sodium.

Abstract:ABSTRACT This research presents the development and validation of chromatographic method for analyzing sorafenib and its pharmacopeial impurities, with a focus on stability studies and degradation product (DP) characterization. Initial method optimization involved exploring various column and buffer combinations, ultimately achieving optimal separation and peak symmetry using an ODS‐AQ YMC (150 mm) column with 0.6 mL/min gradient flow of 10 mM ammonium formate buffer adjusted to pH 3.4 with formic acid as solvent A, and ethanol as solvent B as mobile phase and 246 nm wavelength. Method exhibits calibration curve linear in 50–300 μg/mL for sorafenib and 0.050–0.30 μg/mL for impurities with a detection limit of 0.015 μg/mL for impurities. A structural elucidation of DPs was performed using LC–MS/MS, providing valuable insights into their molecular compositions, and was characterized as 4‐[4‐(carboxyamino)phenoxy]pyridine‐2‐carboxylic acid (DP 1) and 4‐(4‐aminophenoxy)pyridine‐2‐carboxamide (DP 2). Using AGREE and GAPI metrics, evaluation highlighted method sustainability through ethanol–water solvents and shorter column to reduce energy consumption. Toxicity assessments revealed differences in environmental impact and toxicological profiles of DPs, emphasizing importance of managing safety considerations for sorafenib and its DPs. This research offers novel insights into sorafenib analysis by addressing pharmacopeial impurities, characterizing DPs, and evaluating method sustainability and safety.

Abstract:Rationale Enasidenib (EDB) is an orally active selective mutant isocitrate dehydrogenase‐2 enzyme inhibitor approved by the U.S. Food and Drug Administration to treat acute myeloid leukemia. It lacks a reported forced degradation study and a stability‐indicating assay method (SIAM). This study addresses this gap by establishing a degradation profile in accordance with the International Council for Harmonisation Q1A and Q1B (R2) guidelines and developing a validated SIAM for EDB. Methods EDB was exposed to forced degradation under various conditions (hydrolytic, photolytic, oxidative, and thermal stress). Degradation samples were analyzed using high‐performance liquid chromatography on an Agilent ZORBAX Eclipse Plus C18 column with a mobile phase consisting of 0.1% formic acid in Milli‐Q water and acetonitrile at a flow rate of 1 mL/min and detection at 270 nm. Liquid chromatography‐quadrupole time‐of‐flight‐high‐resolution mass spectrometry (LC/Q‐TOF HRMS) was used for the identification and characterization of degradation products. Nitrosamine risk assessment was conducted using a modified nitrosation assay procedure (NAP) test due to the presence of a secondary amine group in the drug, which is liable to forming nitrosamine drug substance–related impurities (NDSRI). Results The drug exhibited significant degradation under acidic, basic, photolytic, and oxidative conditions in the solution state. A total of nine degradation products (DP) were formed (DP‐I, DP‐III, and DP‐IV: acidic conditions; DP‐I and DP‐III: basic conditions; DP‐II, DP‐V, DP‐VI, and DP‐VII: oxidative stress; and DP‐VII, DP‐VIII, and DP‐IX: photolytic conditions), which were separated and identified using reversed‐phase high‐performance liquid chromatography and characterized using liquid chromatography–tandem mass spectrometry. The mechanism behind the formation of EDB degradation products has been discussed, and this study was the first to develop a degradation pathway for EDB. In addition, the possibilities of NDSRI formation for EDB were studied using a modified NAP test, which can contribute to the risk assessment of the drug. Conclusions Forced degradation studies were conducted by establishing a SIAM for EDB. All the degradation products were characterized by mass spectral data obtained using LC/Q‐TOF‐HRMS.
